In this study, antibacterial substances were extracted and identified from the mycelia of Eurotium cristatum FS-1, and evaluated for antibacterial activity against Pseudomonas putida LP-3. The underlying mechanism was investigated by microscopic observations, LP-3 cell content analysis, and metabolomic analysis. The results showed that the half maximal inhibitory concentration (IC50) of the ethyl acetate extract (FSEAE) from the mycelia of Eurotium cristatum FS-1 on P. putida LP-3 (1 × 104 CFU/mL) was 8 mg/mL. As observed by scanning electron microscopy (SEM), FSEAE destroyed the cell membrane of P. putida LP-3. The K+ content in the culture supernatant of P. putida, the activity of oxidative stress enzymes in the cell metabolic pathway (superoxide dismutase, glutathione peroxidase, and catalase) and the content of malondialdehyde (MDA) were measured, showing that the antibacterial effect of FSEAE mainly in the logarithmic growth period of P. putida LP-3 (8­–16 h). Non-targeted metabonomics was used to analyze FSEAE treated samples and control samples (treated by dimethyl sulfoxide), and a total of 124 metabolites were obtained, of which 30 metabolites were significantly different between the two groups. Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis showed that FSEAE affected the growth and metabolism of P. putida mainly by interfering with the tricarboxylic acid cycle. The results from this study can provide a theoretical reference for the development and utilization of Saccharomyces coronatus and its metabolites.

In order to explore the preparation process of liquid organic bacterial fertilizer, the liquid organic fertilizer and plant rhizosphere growth-promoting bacteria Pseudomonas putida Rs-198 were compounded, and the preparation technology and stability of Rs-198 liquid organic bacterial fertilizer and its growth promotion effect on the radish seedlings were studied. The results showed that the optimal dilution ratio of liquid organic fertilizer was 5 times; the optimal culture conditions of Rs-198 in diluted liquid organic fertilizer were 30 ℃, 140 r/min, and the medium volume was 100 mL; the maximum bacterial density can reach 4.6×109 CFU/mL; the prepared Rs-198 liquid organic fertilizer was stored at room temperature for 28 days, and the effective viable count was 2×107 CFU/mL. In addition, bentonite as its best nutritional supplement can increase the number of effective viable bacteria by 60.2%. The pot experiment showed that the liquid organic bacterial fertilizer could significantly increase the biomass of radish seedlings in which the plant height, fresh weight and dry weight increased by 19.6%, 22.2% and 17.6%, respectively. The content of soluble sugar and soluble protein increased by 49.4 % and 12.4 %, respectively. Therefore, Rs-198 liquid organic bacterial fertilizer has a good application prospect. Pulsed electric field ( PEF) technology was applied to aerobic denitrification treatment of nitrate wastewater. By comparing the changes of growth metabolism of aerobic denitrifiers under different PEF strength, frequency, electrode distance and inoculation volume, the optimal process parameters of PEF treatment were determined. The results indicated that under the conditions of PEF strength 0. 8 V · cm - I, frequency 1 000 Hz, electrode distance 5 cm and inoculation volume 5%, the optical density at 600 nm wavelengths ( OD600 ), removal efficiency of COD and NO,- - N of aerobic denitrifiers Pseudomonas putida W207-14 treated with PEF can reach the maximum value in 24 h, which were 1. 926 ± 0. 04, (97. 67 ± 1. 12)% and(90. 34 ± 0. 73 )%, respectively. Compared with the untreated aerobic denitrifiers, the growth time of the treated aerobic denitrifiers is significantly reduced, and the nitrate removal rate is increased by 115. 76%. Phenol is one of the major aromatic pollutants among variety of toxic compounds. Phenol is a man-made as well as a naturally occurring aromatic compound and an important intermediate in the biodegradation of natural and industrial aromatic compounds. To clean up the aromatic contaminated sites a bioremediation method is considered as an economical and safe approach for the environment. In this study, 10 phenol-degrading bacterial strains were isolated from three sites at Midok copper mine in Shahrbabak. These sites include: Magnetit site (WG), Hematit site (WH) and Near Hematit site (NH). Total heterotrophic and phenol degrading bacteria were quantified with most probable number (MPN) and colony forming unit (CFU) methods. The results of this study show that the (WG) site of the mine have the highest phenol degrading bacteria. Totally 10 phenol degrading bacteria were isolated from three sites in Midok copper mine. Then, five bacterial strains were selected according to high growth rate and phenol degradation. Finally two strains named isolates P62 and 69P were selected for analysis of 16S rRNA sequences. Strain P62 belongs to Pseudomonas putidaAHBP62 and strain P69 is related to Arthrobacter scleromaeAHB69P that has capability degradation of 600 (ppm) phenols in 7 days. Due to the low nitrogen removal efficiency, lengthy procedure, and vulnerability to high loads of ammonium of the traditional sewage treatment process, the physiological characteristics, nitrogen removal characteristics, influencing factors, and kinetic analysis were systematically studied for the heterotrophic nitrifier Pseudomonas putida YH, which has efficient nitrogen removal ability. The results showed that strain YH exhibited efficient heterotrophic nitrification ability with a low accumulation of nitrification products. The maximum ammonium removal rate was 99.1%, and nearly 53% of the removed TN was converted to intracellular nitrogen. In addition, nitrite and nitrate could also be metabolized by strain YH under aerobic conditions, with a maximum removal rate of 99.8% and 99.5%, respectively. Additionally, the successful PCR amplification of the napA and nirK genes further improved the aerobic denitrification characteristics of strain YH. The bacterial growth process of strain YH matched the Logistic model ( R 2 >0.99), and the nitrogen degradation conformed to the Compertz model ( R 2 >0.99). The order of the maximum conversion rates of nitrogen ( R m ) was ammonia > nitrate > nitrite, and that of the lag time ( t 0 ) was nitrate > nitrite > ammonia. The optimal conditions for heterotrophic ammonium oxidation with strain YH were succinate as the carbon source, C/N=10, T =30℃, r =160-200 r·min -1 , and pH=7.0. Under the optimal conditions, the average ammonia oxidation rate and R m were 8.35 and 16.71 mg·(L·h) -1 , respectively. Strain YH could adapt to a broad range of ammonium loads. A high ammonium removal rate was observed under high ammonium concentrations (1000 mg·L -1 ), indicating the high potential of strain YH for application in high-strength ammonium wastewater treatment.

The biodegradation of gas-phase mixtrue of dimethyl sulfide (DMS) and 1-propanethiol (PT) was examined in a biotrickling filter (BTF), inoculated with a microbial consortium composed of activated sewage sludge, and pure strains of Alcaligenes sp. SY1 and Pseudomonas putida. S-1. BTF could be successfully started up within only 11 days when the inlet concentrations of DMS and PT were both 50 mg·m -3 and EBRT was 30 s, with 90% removal efficiency (RE) of DMS and 100% RE of PT. In the steady state, the maximum elimination capacities of DMS and PT were 8.7 g·(m 3 ·h) -1 and 12.4 g·(m 3 ·h) -1 , respectively. The presence of PT with a concentration up to 51 mg·m -3 showed an antagonistic removal pattern for DMS, but the opposite did not occur. Meanwhile, the BTF showed high efficiency in the biodegradation of H 2 S. When the concentration of H 2 S was as high as 230 mg·m -3 , the RE of H 2 S could reach 98%. However, H 2 S showed a declining effect on the removal of DMS when the concentration exceeded 115 mg·m -3 .
