Objective: To investigate the effect of restraint stress on masseter mechanical hyperalgesia and the activity of neurons and astrocytes in the spinal trigeminal nucleus caudalis (Vc). Methods: The animals were randomly divided into the control group, 1-, 3-, 5-, 7-, 9-, 11-and 14-day stress groups, with 10 rats in each group. The body weight increase and behavior tests were used to testify the animal model. The mechanical sensitivity of masseter of the rat before and after the stress was measured with Von Frey filaments. Histological examinations were used to evaluate the expression of neuronal c-fos and astrocytic glial fibrillary acidic protein (GFAP). Results: Restraint stress resulted in remarkable mechanical allodynia in the masseter muscle. The head withdrawal threshold was significantly lower in the 7-, 9-, 11-and 14-day stress groups ([0.071±0.011], [0.059±0.020], [0.052±0.011], [0.033±0.011] N) than that in the control group ([0.120±0.025] N) ( P< 0.05). Compared to the control group, the rats in the 1-day stress group showed a significant increase of c-fos in neurons of the Vc and then declined to normal level after 1 week gradually. The GFAP expression in astrocytes of the Vc was significantly increased in the 7-, 9-, 11-and 14-day stress groups (4.3±1.0, 4.5±0.6, 4.6±0.5, 4.8±1.3) compared with the control group (2.0±0.8) ( P< 0.05). Conclusions: Chronic restraint stress could lower the threshold of mechanical allodynia in the masseter muscle and activate the neurons and astrocytes in Vc. The activation of neurons and astrocytes plays an important role in the masseter hyperalgesia induced by restraint stress in rats.

Objective: To investigate the effect of occlusal interference on the energy metabolism of masticatory muscle by studying the changes of adenosine triphosphate (ATP), adenosine diphosphate (ADP), inosine monophosphate (IMP), phosphocreatine, creatine, lactate and pH level in masseter muscles of rats after occlusal interference., Methods: Fifty male Sprague-Dawley rats were randomly assigned into experimental group (n=40) and control group (n=10). In experimental group, 0.4 mm thick metal crown was cemented to the upper right first molar of the rat, and maintained for 3, 7, 10, 14 d separately (n=10 for each time point). No occlusal interference was applied for control group. Bilateral masseter muscles of all the rats were acquired under general anesthesia. The samples of 5 rats in each group were fully homogenized with 0.4 mol/L perchlorate (10 mL/g). The homogenates were centrifuged, filtered and analyzed for ATP, ADP, IMP, phosphocreatine, creatine and lactate content by high performance liquid chromatography. The other samples in each group were mixed with homogenates containing 5 mmol/L sodium iodoacetate (10 mL/g), then homogenized and measured for pH value by pH meter in thermostatic water bathunder 37 degrees centigrade., Results: Compared with control group, ATP content in bilateral masseter of the rats increased 3 d after occlusal interference [right side:(5.36±0.13) μmol/g,left side:(5.77±0.25) μmol/g] (P<0.05), and back to normal on 7, 10 and 14 d. There was an increase in IMP [right side:(0.21±0.03) μmol/g,left side:(0.19±0.03) μmol/g]and creatine content [right side:(24.76±2.94) μmol/g,left side:(27.75±2.23) μmol/g]in bilateral masseter of the rats 7 d after occlusal interference (P<0.05) and no difference was detected on 3, 10, and 14. Phosphocreatine content in bilateral masseter started to decline 7 d after occlusal interference and maintained the low level on 10 and 14 d [right side:(10.70±0.71) μmol/g, (11.57±0.52) μmol/g, (10.74±1.39) μmol/g, left side:(10.05±0.57) μmol/g, (10.75±1.12)μmol/g, (10.61±1.15) μmol/g](P<0.05). No change of ADP, lactate or pH level in bilateral muscles of the rats after occlusal interference was observed (P>0.05)., Conclusion: Occlusal interference influences the content of energy metabolites in masticatory muscle of rats, which may be related to the pathological process of masticatory muscles induced by occlusal interference, such as muscle pain, dysfunction and altered fiber architecture.

Objective: To investigate the relationship between the existence of occlusal interference and masticatory muscle hyperalgesia by exploring the stimulus-response relationship between the duration of occlusal interference and masticatory muscle mechanical withdrawal threshold., Methods: Occlusal interference with 0.4 mm-thick crowns on rat molars was removed under anaesthesia at 2, 3, 4, 5, and 6 d after wear, and masticatory muscle mechanical withdrawal threshold was tested at 1, 3, 5, 7, 10, 14, 21 and 28 d., Results: Decreased mechanical withdrawal thresholds were detected in temporal muscles and masseter muscles on both sides following occlusal interference (P < 0.05). After removal of crowns at 2, 3, 4 and 5 d, rats exhibited the similar head withdrawal thresholds as the sham-application control in masticatory muscles on both sides at 10, 14, 14 and 21 d (P > 0.05). No significant differences were detected between the contralateral side with the ipsilateral side (P < 0.05). After removal of crowns at 6 d, rats still exhibited significantly decreased head withdrawal thresholds[right temporal muscle: (1.365±0.018) N; right masseter: (1.437 ± 0.024) N] in masticatory muscles on both sides until the last day of the experiment[the sham-application control: right temporal muscle: (1.554±0.040) N, P < 0.001; right masseter: (1.546±0.019) N, P < 0.001]., Conclusions: The mechanical hyperalgesia can disappear after removal of the occlusal interference at 5 d, and the existence of the occlusal interference is positively correlated with the duration of the mechanical hyperalgesia.

Objective: To determine the classification of masticatory myospasm by analyzing characteristics of clinical appearances., Methods: Thirty-six cases of masticatory myospasm from 2000 to 2010 were included. The clinical data of these patients were analyzed, including patient information, patient history, clinical characteristics, severity and the frequency of myospasmodic movement, electromyogram (EMG), and the efficacy of botulinum toxin injection treatment., Results: There were 11 males and 25 females, aged from 15 to 71. According to the clinical manifestation and EMG findings, patients could be divided into two groups: 18 cases were classified as jaw closing type which involved masseter and/or temporalis muscles presenting as trismus and acute pain, the other 18 cases were jaw opening type which involved lateral pterygoid muscles complaining difficulty in jaw closing and teeth clenching. The jaw closing type was often seen in patients of 20 to 50 years old, the jaw opening was frequently seen in patients over 50 years old. Jaw closing type was attacked intermittently and unilaterally, but jaw opening was often attacked continually and bilaterally. The rating scale of the severity of spasmodic movement was not different between the two types, but the frequency of spasmodic attack was much higher for jaw opening type (P < 0.05). The EMG of jaw closing type was classified into persistent, rhythmic and irregular type. The EMG of jaw opening type was classified into spontaneous and exercise-induced type. Twelve cases were treated by botulinum toxin injection that could significantly relieve symptoms., Conclusions: Masticatory myospasm can be classified into jaw closing and jaw opening types. Jaw closing type involves masseter and/or temporalis muscles and jaw opening type involves lateral pterygoid muscles. Botulinum toxin injection was the most effective therapy for the masticatory myospasm.

Objective: To investigate the changes in electromyographic activities of the temporal and masseter muscles at different positions of the mandible., Methods: Twenty orthodontic patients with Angle Class II malocclusion and mandibular retrusion (ANB<6°) aged 10-14 years were enrolled in this study. All the patients were treated with Forsus fixed functional appliance combined with MBT straight-wire appliance. The electromyographic activities of the temporal (T) and masseter (M) muscles before, during and after functional treatment were evaluated by assessing the average integrated electromyogram (EMG) and T/M ratio at clenching status in different mandibular positions., Results: After functional forward positioning of the mandible, the electromyographic activities of the temporal and masseter muscles decreased and T/M ratio increased significance at the clenching status (P<0.05)., Conclusion: The appliance insertion and activation is associated with a decreased EMG activity of the temporal and masseter muscles, and the T/M ratio is correlated to the position of the mandible.

Objective: To study the effect of energy therapy on Ca2+ concentration and Ca2+ -ATP enzyme activity in rat master muscle after unilateral chew, and to discuss the protective action of the exogenous creatine phosphate on rat masseter muscle after unilateral chew., Methods: The 20 rats were randomly divided into 4 groups, A: Creatine phosphate normal control group; B: Creatine phosphate experimental group; C: Saline normal control group; D: Saline experimental group. The Ca2+ concentration were determined by atomic absorption spectrophotometry, the activity of the Ca2+ -ATP enzyme were determined by super-micro volume Ca2+ -ATP enzyme kit., Results: (1) The Ca2+ concentration of the extraction side of group D which received the saline injection had significant difference compared with the non-extraction side (P = 0.007), the group C (P = 0.009) and the extraction side of group B (P = 0.01); (2) Ca2+ -ATP enzyme activity of group D were higher than its non-extraction side (P = 0.001), group C (P = 0.003) and the extraction side of group B (P = 0.001); (3) The ultrastructural changes of the rat masseter muscle under transmission electron microscope were as follows: The extraction side of group D have more severe pathological manifestations than non-extraction side. Both the extraction side and the non-extraction side of group B had a similar manifestation to the normal control group., Conclusion: Exogenous energy material, creatine phosphate, may have certain degree of protective effect on rat masseter muscles after unilateral chew. And it may become a possible way to improve the injury of the masseter muscle.

Objective: To study gender-related factors in the painful sites and degree of pains in the trigger points of the sternocleidomastoid muscle., Methods: The trigger points of the sternocleidomastoid muscle of healthy volunteers of either gender with different ages were examined by measurable pressure using the right thumb, and the pain degree was assessed with visual analog scale (VAS)., Result: The VAS score was 5.226+/-0.331 in senior male subjects and 5.531+/-0.379 in senior female subjects, without significant differences between them (P>0.05). Nor was significant difference detected between the scores of young male and female subjects (2.940+/-0.331 vs 3.982+/-0.405, P>0.05). In terms of the pain severity, the difference was significant between the medial and anterior areas and between the medial and posterior areas of the mastoid process (P<0.05), but not between the anterior and posterior areas of the mastoid process (P>0.05)., Conclusion: There is no significant gender-related difference in the pain in the trigger points of the sternocleidomastoid muscle elicited by compression with the right thumb. The anterior and posterior areas of the mastoid process are relatively sensitive to compression pain.

The aim of this investigation was to study the EMG of masseter of edentulous patients in chewing cycle. 18 subjects were examined by Disa-2000M Neuro-electromyograph. It was found that the period of chewing cycle of the patients with complete denture was distinctly longer than that of normal people, and the change of the duration prolongs the time phases. The velocity was relatively slow when trying to keep the stability of dentures. The amplitude of potential in masseter was lower in chewing cycle, but it did not affect the function of mastication. The abnormal pause after occlusal contact of denture had an influence on the function of mastication.
