Objective: To analyze the characteristics of skeletal muscle cells gene markers in septic patients by using bioinformatics., Methods: The differential gene expression of marker microarrays (GSE13205) in skeletal muscle tissue of patients with sepsis was obtained from gene expression omnibus (GEO) database of National Center for Biotechnology Information (NCBI). Gene differential expression analysis was carried out using online GEO2R provided by NCBI. Data processing, analysis and mapping were carried out using online bioinformatics array research tool (BART) and Cytoscpe software, the software of the national resource for network biology. Functional annotation and pathway analysis of differential expression genes were performed using Kyoto encyclopedia of genes and genomes (KEGG) and gene ontology (GO) provided by the database for annotation, visualization and integrated discovery (DAVID), and protein interaction analysis was further performed in search tool for the retrieve of interacting genes/proteins (STRING-DB)., Results: The TOP250 genes were extracted from the GSE13205 dataset. A total of 242 differentially expressed genes were included in the analysis. Among them, 78 up-regulated genes and 164 down-regulated genes were identified. After extensive data analysis, these differentially expressed genes were enriched into different biological processes or subsets of molecular functions, mainly enriched in the positive and negative regulation of growth, mineral absorption and other pathways. The 14 most closely related genes among differentially expressed genes were identified from the protein interaction network., Conclusions: The differential expression genes in patients with sepsis were mainly concentrated on cell growth and apoptosis and mediating tumor-related immune function regulation.

Objective: To understand the dietary patterns of the middle-aged and elderly in Bengbu City and to analyze the associations of dietary patterns with dyslipidemia and hypertension., Methods: During September of 2014 to September of 2015, the cluster random sampling method was used to select 860 residents aged ≥45 years old( 45-76 years old) from 6 communities of Bengbu City. They answered a well-designed questionnaire( age, gender, history of chronic disease), received physical examinations ( height, weight, waist circumference, hip circumference) and dietary survey, measured the blood lipids and blood pressure. Factor analysis was applied to determine major dietary patterns in order to analyze the associations between dyslipidemia and hypertension., Results: ( 1) Four dietary patterns was identified, namely "rice and vegetables" pattern, "animal food" pattern, "fruits and milk" pattern, "drinking" pattern. ( 2) There were significant difference of BMI between the "rice and vegetables" pattern and "animal food" pattern( P < 0. 01). There were significant difference of WC between the "rice and vegetables"pattern, "animal food" pattern and "drinking" pattern( P < 0. 01). There were significant difference of SBP between the "rice and vegetables " pattern and "drinking" pattern( P < 0. 01). ( 3) The Q4 group of "rice and vegetables" pattern and "animal food" pattern had higher proportion of protein that contributed to total energy than the Q1 group( 17. 44% vs. 13. 63%, 20. 52% vs. 17. 55%, P < 0. 05). The Q4 group of "rice and vegetables" pattern had higher energy intake of 1. 75-fold than the Q1 group. ( 4) Whether or not controlling the confounding factors of gender, age, degree of education, physical activity and BMI, a positively association was found between "animal food" pattern, "drinking" pattern and dyslipidemia by logistic regression analysis between the Q4 group with the Q1 group( unadjustment OR = 2. 665, OR = 1. 926, P < 0. 01; after adjustment OR = 1. 835, OR = 1. 447, P < 0. 05)., Conclusion: "Animal food" pattern and "drinking" pattern may be risk factors for dyslipidemia and hypertension, reducing the intake of meat, oil in our meals may play an important role in the prevention and control of chronic diseases.

Objective: To evaluate the diet quality and diet structure of community residents in Bengbu City by Chinese Diet Balance Index-07( DBI-07), in order to provide a scientific basis for dietary guidance and nutrition intervention., Methods: A stratified cluster random sampling was used in our design, a total of 772 community residents were selected from 8 communities in Bengbu City in 2015. Food intakes were collected by a continuous 3d 24 h dietary survey, and the dietary quality was evaluated by an adjusted DBI-07 scoring and evaluation system., Results: The overall dietary quality of Bengbu community residents was somewhat imbalance( DQD = 31. 6). Insufficient intake( LBS =26. 1) and excessive intake( HBS = 9. 0) were both coexisted in the residents of Bengbu. Male had a higher level of excessive intake and imbalance than female. Rural areas were more severe in insufficient and imbalance than those of the city. Group age from 45 to 55 has the highest level of dietary imbalance( DQD = 35. 2). From the distribution of DBI scores, intake of vegetables, fruits, milk and beans were insufficient, the intake of vegetables and fruits, milk and beans approaching and reaching RNI accounted for 20. 1%and 9. 3%, respectively. Major dietary pattern of residents is the Pattern A( 27. 9%), B( 32. 3%) and E( 21. 0%)., Conclusion: The dietary quality of residents in Bengbu should be improved. Men, rural residents and 45- 55 age group are the main intervention groups. It is necessary to intensify the dietary guidelines and food guide pagoda, in order to improve the consciousness of residents about reasonable, adequate diet and low salt, low oil.

A method for the determination of sulfonamide potentiators, trimethoprim (TMP), diaveridine (DVD) and ormetoprin (OMP), in different animal origin food matrices (including chicken muscle, fish muscle, chicken liver, egg and milk) has been developed by ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS). The sample was extracted by formic acid-acetonitrile (1:9, v/v), cleaned-up by hexane, separated on an Acquity UPLC BEH C18 column (50 mm x 2.1 mm, 1.7 microm) with gradient elution. The determination was carried out with electrospray ion source under the positive mode and multiple reaction monitoring (MRM) mode. The extraction recoveries of three extraction solvents were observed. The purification condition and concentration condition were optimized. In addition, the mobile phase, column temperature and solid phase extraction column were studied. The calibration curves showed a good linearity in the range of 1.25-30.0 microg/L, and the correlation coefficients (r) were higher than 0.99. The limits of quantification (LOQ, S/N = 10) of the three potentiators were 5.0 microg/kg. At the spiked levels of 5.0, 10.0 and 20.0 microg/kg, the recoveries of the three potentiators were ranged from 61.2% to 108.5%, and the relative standard deviations (RSD, n = 6) ranged from 1.1% to 9.8%. The results indicate that the method is simple, rapid, sensitive and suitable for the qualitative and quantitative analysis of the three potentiators in multiple matrices.

A method for the determination of dicyandiamide (DCD), melamine (MEL) and cyanuric acid (CY) in milk and milk powder has been developed by ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS). After extracted by 5% (mass fraction) trichloroacetic acid, the samples were loaded onto an Acquity UPLC HSS T3 column (50 mm x 2.1 mm, 1.8 microm) and separated using acetonitrile and ammonium acetate as mobile phases in a gradient elution mode. The electrospray was operated in the positive mode and negative mode, and monitored by the multiple reaction monitoring (MRM) mode. The calibration curves showed a good linearity in the range of 5.0 - 200.0 microg/L, and the correlation coefficients (r) were not less than 0.995. When the spiked levels were 0.02, 0.10 and 0.20 mg/kg, the recoveries of DCD, MEL and CY in milk ranged from 60.0%. to 105.8%, with the relative standard deviations (RSDs) of 4.2% - 13.6%; while the spiked levels were 0.05, 0.10 and 0.20 mg/kg in milk powder, the recoveries ranged from 78.0% to 115.0% with the RSDs of 2.7% - 7.5%. The limits of quantification (LOQs, S/N = 10) were 0.02 mg/kg for milk samples and 0.05 mg/kg for milk powder samples. The results indicate that the method is simple, rapid, sensitive and suitable for the qualitative and quantitative analyses of DCD, MEL and CY in milk and dairy product samples.

A method for the determination of bisphenol A in the plastic parts of small household appliances by liquid chromatography-tandem mass spectrometry (LC-MS/MS) was developed. The sample was extracted with accelerated solvent extraction (ASE) and purified by Sep-Pak C18 solid phase extraction. Bisphenol A was separated and detected using LC-MS/MS in negative ion mode with the mobile phases of methanol and water (containing 0.05% ammonia water). The linearity of the method was good in the range of 5 μg/L to 100 μg/L. The recoveries for the spiked sample were from 95.2% to 109.7% at the three levels, 10, 25 and 75 μg/kg. The relative standard deviations were less than 3.8%. The limit of detection was 10 μg/kg. The method is easy-handling, time-saving, sensitive and suitable for the determination of the residual bisphenol A in the plastic parts of household appliances.

An ultra performance liquid chromatography-tandem mass spectrometric method (UPLC-MS/MS) has been developed for the simultaneous determination of eight pesticide residues from seed coating in fruits, vegetable and grain. The sample was extracted by methanol-water (1:1, v/v) and determined by ultra performance liquid chromatography-electrospray ionization tandem mass spectrometry in positive mode (ESI+) and multiple reaction monitoring (MRM) mode. The UPLC analyses were performed on an Acquity UPLC C18 column with gradient eluation. The utility of the method was demonstrated by the analysis of crude extracts, with no sample clean up, from soybean. The linear range was 1 - 200 microg/L. The correlation coefficients (r) were under 0.997. The average recoveries of eight pesticides in samples (from 0.006 to 1.2 mg/kg) ranged from 60% to 110%, and the relative standard deviations (RSDs) were less than 10%. The results indicate that the method is easier, faster, more sensitive, and suitable for the qualitative and quantitative confirmation of pesticide residues from seed coating reagent in fruit, vegetable and grain samples.
