Objective To explore the clinical significance of long non-coding RNA(lncRNA) VIM-AS5 expression in human breast cancer tissues and its regulatory mechanism involved in cancer cell proliferation and migration. Methods The Lnc2Cancer 3.0 database was used to analyze the expression of VIM-AS5 in breast cancer tissues and its correlation with the clinical stage and survival time of breast cancer patients. RT-qPCR was used to detect the expression of VIM-AS5 in breast cancer cell lines BT-549, MDA-MB-435, MDA-MB-231 and CAL-51. Plasmid with VIM-AS5 overexpression and negative control were all transfected into CAL-51 cells through liposome recorded as VIM-AS5 group and NC group, respectively. The proliferation and migration of CAL-51 cells were detected by colony formation assay and scratch healing method, respectively. Dual-luciferase reporter gene experiment verified the targeting relationship between VIM-AS5 and miR-500a. RT-qPCR was used to detect the expression of miR-500a in CAL-51 cells. Western blot was used to detect the expression of JAK/STAT3 pathway in CAL-51 cells. Results The expression of VIM-AS5 in breast cancer tissues was significantly lower than that in adjacent tissues(P＜0.01). VIM-AS5 expression was negatively correlated with the clinical stage of breast cancer patients(P＜0.01). The survival time of breast cancer patients with low VIM-AS5 expression was significantly shorter than that of breast cancer patients with high VIM-AS5 expression(P＜0.01). Compared with mammary epithelial cell line MCF-10A cells, VIM-AS5 expression was significantly reduced in breast cancer cells(P＜0.01). The counting number of colony formed in the VIM-AS5 group was significantly lower than that in the NC group(P＜0.01). The cell migration rate in the VIM-AS5 group was significantly lower than that in the NC group(P＜0.01). Dual-luciferase reporter gene experiment confirmed that miR-500a was the target gene of VIM-AS5(P＜0.01). VIM-AS5 can negatively regulate the expression of miR-500a(P＜0.01). Compared with the NC group, the expression of JAK/STAT3 pathway proteins JAK, p-STAT3, c-Myc, Bcl-2, and CDK3 in CAL-51 cells of the VIM-AS5 group were significantly decreased. Conclusions VIM-AS5 is low-expressed in breast cancer cells, and up-regulation of VIM-AS5 may inhibit the proliferation and migration of breast cancer cells CAL-51 by targeting at miR-500a/JAK/STAT3 pathway.

The severe mining pressure of surrounding rock along goaf retaining roadway has become the bottleneck of safe and efficient production of coal mine. As one of the effective measures to effectively reduce roadway pressure, roof cutting and pressure relief has received more and more attention. Taking the wind tunnel of 91-101 working face of Wangzhuang Coal Mine as the engineering background, the variation rules of vertical displacement of surrounding rock, the distribution characteristics of stress and plastic zone along gob retaining roadway under pressure of new roof cutting are studied by combining theoretical analysis and numerical simulation, and the influence law of roof cutting height on surrounding rock deformation is revealed. The research results show that the effect of the new roof cutting technology is remarkable, the side displacement of roadway goaf is reduced by 46.7% compared with that of unbroken roof, and the optimal roof cutting height is 19 m. The average displacement of roof, floor and two sides of gob-side entry retaining is 312 mm and 128 mm respectively, and the deformation of entry retaining is effectively controlled.

In the noisy intermediate-scale quantum (NISQ) era, traditional initial mapping strategies overlook potential adjacency issues in subsequent operations. To address this challenge, a multi- factor interaction cost function is devised, considering the distance between qubits, the interaction time, and the gate operations error rate. Additionally, the comprehensive SWAP optimization strategy (CSOS) is proposed to address the adjacency problem in quantum circuits, including the optimal SWAP selection and the SWAP- based batch update strategy (BUS) to optimize the local qubit mapping in quantum circuits. Optimal SWAP selection involves SWAP operations choices with the highest benefit by comparing advantages. The BUS considers upcoming quantum operation sequences during the mapping phase and proactively performs SWAP operations batches. These approaches reduce the SWAP operation numbers during the entire circuit execution process, minimizing mapping overhead to the greatest extent. Experimental results demonstrate that the CSOS reduce the average number of inserted SWAP gates by 38.1% and lower hardware gate count expenses by approximately 12%. [ABSTRACT FROM AUTHOR]

In order to solve the problems of high cost of microemulsion use and the limitation of microe- mulsion application in high temperature and high salt strata, this paper conducted a systematic research on the development of chemically driven microemulsions and the unique properties of nanoparticles, and summarized the synergistic effect between nanoparticles and microemulsions, the effect of nanoparticle incorporation on the applicability of salinity to the microemulsions as well as the influence on the rheological properties. The current status of nanoparticle synergistic microemulsion research at home and abroad is summarized and reviewed, and the synergistic effect between nanoparticles and microemulsions is summarized to produce mechanisms such as low interfacial tension, change of wettability, reduction of chemical adsorption, enhancement of stability, and improvement of rheological ratio. Objective To investigate the current status of anticoagulation therapy for the elderly patients aged 75 years or over with non-valvular atrial fibrillation in secondary and tertiary hospitals in Chongqing. Methods A total of 3 821 patients aged 75 years old or over with non-valvular atrial fibrillation from 19 hospitals (6 secondary hospitals and 13 tertiary hospitals) in Chongqing between January 1, 2018 and December 31, 2018 were enrolled in this study. The clinical data of patients were collected by electronic medical record system. Their baseline data, scores of CHA2DS2-VASc and HAS-BLED, anticoagulant rate and anticoagulant drugs were compared and analyzed. Results All the patients had their CHA2DS2-VASc score ≥2, and those with HAS-BLED score ≥3 accounted for 25.0%. The overall anticoagulant rate was 36.2%. And of these patients, 10.8% and 25.4% were treated with new oral anticoagulants (NOAC) and warfarin respectively, 43.3% were treated with antiplatelet alone without anticoagulant, and 20.5% were not treated with any antithrombotic therapy. In order to improve the feeding efficiency of the push-rod device of the pre-filled syringe, the simulation analysis was carried out based on the JKR model of EDEM software. With the push-rod as the research object and the discharge number in specified time period as the evaluation index, the influences of the inner tray rotation speed, the outer tray rotation speed, the tilt angle of the inner rotation tray, the angle of the cone tray and the outer rotation radius on the discharge efficiency of the push-rod were investigated. Firstly, single factor analysis was carried out to determine the influence of each single factor and screen out the four factors that with greater influence. Then the orthogonal design of four factors and three levels was carried out to determine the optimal parameter combination and the order of the influence degree of each factor on the discharge efficiency. The discharge stability was studied and the best feeding method was obtained. Finally, the experimental validation was carried out. The results show that the maximum error of simulation and test is 9.6%, which indicates that the simulation analysis of the model is reliable. In order to meet the requirements of heterogeneous protocol integration and interconnection in software-defined interconnection systems, a two-stage multi-protocol switching circuit was proposed. This circuit accommodated the forwarding needs of various heterogeneous protocols by integrating shared memory and Crossbar architectures. In addition, a multi-stage arbitrary scheduling scheme based on time slot was introduced to decouple time and space during the scheduling. Simulation results demonstrate that this switching circuit can dynamically adapt to changes in switching scale, port memory, and port bandwidth requirements resulting from binding mode changes. Compared to traditional combined input and output queued (CIOQ) architecture, the utilization of memory is increased by up to 87.5%, and the forwarding delay is as low as tens of nanoseconds, making it suitable for RapidIO, fibre channel (FC), Ethernet, peripheral component interconnect express (PCIe) single protocol switching as well as hybrid protocol switching combining these protocols. Objective To analyze the effect of continuous intravenous injection of different doses on dexmedetomidine on the recovery period of general anesthesia in patients undergoing thyroid surgery. Methods A total of 120 patients with thyroidectomy admitted to the First Affiliated Hospital of Army Medical University from November 2017 to November 2019 were randomly divided into a control group (60 patients) with continuous intravenous injection of 0.8 μg/kg dexmedetomidine. The observation group (60 patients) were given 0.4 μg/kg dexmedetomidine. The SBP, DBP, HR, Ramsay, VAS score, arousal index and adverse reactions were compared between the two groups at different time periods. Results The scores of SBP, DBP, HR and VAS, the incidence of nausea, vomiting, laryngospasm and choking cough in the observation group were not different from those in the control group at different time periods. The Ramsay scores of the observation group at different time periods were significantly lower than those of the control group(P＜0.05). The time of eye opening, extubation and directional force recovery in the observation group were significantly shorter than those in the control group (P＜0.05). The incidence of bradycardia in the observation group was 1.67%, which was significantly lower than that in the control group(6.68%) (P＜0.05). Conclusions 0.4 and 0.8 μg/kg dexmedetomidine can maintain hemodynamic stability in the recovery period of general anesthesia in patients undergoing thyroid surgery, but 0.8 μg/kg has more sedative effect and higher incidence of bradycardia, so it is recommended to use 0.4 μg/kg dexmedetomidine.

The singularity and statics performance of a 6-DOF mobile redundant parallel mechanism and its real-time optimization design are studied. The statics Jacobian matrix of this parallel mechanism is established,and the singularity and statics performance are analyzed. The statics transfer performance of parallel mechanism is improved by adjusting the leg position of the parallel mechanism. The performance spectrum based on the Jacobian matrix is drawn,the transfer performance of the statics force and moment of the parallel mechanism in different positions is observed intuitively. By above analysis shows that the redundant parallel mechanism can eliminate the singularity,while adjusting the leg position also greatly improves the static transfer performance of parallel mechanism.

In the algorithm of using convolutional neural network to identify verification codes, the traditional algorithm randomly initializes the convolutional kernel parameters, which leads to slow convergence speed, long training time and low recognition accuracy. To solve this problem, this study proposed a verification code recognition method based on principal component analysis and convolutional neural network. The algorithm first used the drop-fall algorithm to segment the preprocessed verification code image, and then lused PCA learning for the segmented image, and took the learned feature vector as the initialization parameter of the convolution core of the corresponding convolution layer. Nanjing 5718 is a rice variety with good eating quality approved by Jiangsu province in 2019 and was confirmed as super rice by the Ministry of Agriculture and Rural affairs in 2021. Studying its yield composition characteristics, photosynthetic characteristics, and physiological basis can be helpful to investigate photosynthetic traits of super rice variety, improve its photo- synthetic performance through genetic improvement, and provide the theoretical basis for high-yield breeding and cultivation of japonica rice. In this study, the parents of Nanjing 5718 and Huaidao 5, a widely popularized rice variety with the same growth period type, as the control, dry weight of aboveground of plant, chlorophyll contents, photosynthetic rates, and photochemical characteristics of photosystem II (PSII), core antenna protein expression, photosynthetic enzyme activities, and chloroplast ultrastructure were investigated in Nanjing 5718 from full expansion of flag leaves at the beginning of booting to 35 days after flowering. The results showed the pigment contents of the leaves in Nanjing 5718 were higher, which were close to male parent Yanjing 608. The net photosynthetic rates, dry weight, and PSII electron transfer activities of flag leaves at the late growth stage were significantly higher in Nanjing 5718 than those in parents and Huaidao 5. The light energy conversion performance of PSII was superior. The core antenna protein CP43 and CP47 were more stable under strong light and high temperature, and their adjustment ability of Nanjing 5718 was better than that of parents and Huaidao 5. In addition, the activities of RuBP carboxylase in flag leaves were higher, the contents of photosynthetic products were higher, the chloroplast structure was more stable, the senescence characteristics appeared later, and the chloroplast decay rates were slower in Nanjing 5718, compared with parents and Huaidao 5. The results indicated that high photosynthetic performance of flag leaves was the basis for the formation of high yield characteristics of super rice Nanjing 5718 with large panicles. The great activity and function of photosystem proteins were main factors of improving leaf photosynthesis, and the stability of chloroplast structure could provide powerful support for high photosynthetic efficiency of leaves. Surface crack identification with visual images is a kind of non-contact detection solution, which is not limited by the material of the tested object and is easy to achieve online automation. Therefore, it has the advantages of fast speed, low cost and high precision. Firstly, the public data sets of typical pavement crack are comprehensively collected, and the characteristics of sample images and the random variable factors are summarized. Subsequently, the advantages and disadvantages of hand-crafted feature engineering, machine learning, and deep learning crack identification methods are compared. Finally, from the aspects of network architecture, testing performance and predicting effectiveness, this paper reviews the development and progress of typical deep learning algorithms such as self-built CNN, transfer learning (TL) and encoder-decoder (ED) that can be easily trained and deployed. We can see the obvious improvement of performance and effect because of algorithm optimization and computing power enhancement. Cashmere and wool fibers are relatively similar in appearance as well as in physical and chemical properties, and it has been a challenge for the textile industry to identify these two fibers quickly and accurately. The identification methods of cashmere and wool fibers are reviewed in this paper. The related researches of identification methods such as microscopy, infrared spectroscopy, proteomics analysis, DNA analysis, image processing and computer vision are mainly introduced. Golden pomelo is delicious and nutritious and has gained great popularity. Golden pomelo will generate bitter taste with the extension of storage time, which seriously affect the development of the related industry. To explore the reason behind, the present work used Meizhou's golden pomelos the research object to investigate the source of bitterness. After grading, the content of titratable acid, soluble solids content, maximum adhesion force, limonin content, nomilin content, naringin content were simultaneously determined by methods such as the random forest method. The results showed that the soluble solids content was extremely significant and positive correlated (R=0.78) with sensory scores. Nomilin (R=-0.69) content and naringin (R=-0.59) content were significantly and negatively correlated with sensory scores. The bitterness source of the golden pomelo after a long-term storage was closely related to its soluble solids content. This study can provide a certain theoretical basis for the research on debittering of golden pomelo and rapid assessment of the fruit quality. [ABSTRACT FROM AUTHOR]

According to geographic origin and amylose content, 87 investigated japonica rice cultivars were divided into three types: Jiangsu common japonica rice, Northeast japonica rice and Jiangsu semi-waxy japonica rice. In order to explore the eating quality characteristics of the three types of japonica rice and their molecular basis, the physicochemical properties and the sequence of major genes Wx and OsSSIIa for starch synthesis were analyzed. Compared with Northeast japonica rice, Jiangsu common japonica rice had lower amylose content, higher protein content and gelatinization temperature, and similar gel consistency and RVA profiles. Compared with Northeast japonica rice and Jiangsu common japonica rice, Jiangsu semi-waxy japonica rice contained the lowest amylose content, the highest gel consistency and unique RVA profile. Variation coefficients of all characters except for setback value were small in three types of rice varieties, indicating that there was a great range of retrogradation within all the cultivars. The correlation coefficients among traits were different among three types of japonica rice, which indicated that the correlations were affected by the ecological conditions of different regions and properties of varieties. By cluster analysis, 87 japonica rice varieties (lines) were divided into four groups. Jiangsu common and Northeast japonica rice varieties (lines) were intersected in group I and group II. Group III was composed of two lines of Jiangsu common japonica rice and one semi-waxy variety (Xudao 9), and the most of their character values were ranged between those common japonica rice and semi-waxy japonica rice. The Jiangsu semi-waxy rice varieties (lines) with unique physicochemical characteristics were clustered into group IV. By sequence analysis, there was no difference in Wxb gene between Jiangsu common japonica rice and Northeast japonica rice, but in Jiangsu semi-waxy japonica rice G/A substitution occurred in Exon 4 of Wxb, which is the Wxmp allele. Objecfive: Constructing the specific overexpression of Nmnat gene in the heart of fruit flies using UAS/ hand-gal4 system to study the effect of the overexpression of Nmnat gene in the heart combined with endurance exercise on lipid toxicity induced by high-fat diet, and to analyze the molecular mechanism. Methods: Male w1118 and Nmnat-UAS fruit flies were respectively hybridized with female hand-gal4. Collecting the normal expression of heart Nmnat (hand>w1118) and the overexpression (hand>Nmnat) of F1 generation female flies. After feeding for 15 days, they began to perform endurance exercise (E) and high-fat diet (HFD). They were divided into hand>w1118, hand>w1118+E, hand>w1118+HFD. hand>w1118+HFD+E, hand>Nmnat, hand>Nmnat +E, hand>Nmnat+HFD, hand>Nmnat+HFD+E, 410 flies in each group. The levels of cardiac TAG, NAD+, SIR2 protein deacetylation, SOD activity and MDA were detected by ELISA. The expression of cardiac related genes by RT-PCR; M-mode detects cardiac function. Results: the expression of Nmnat, dFoxo, bmm, NAD+ level, SIR2 deacetylation, SOD activity and shortened fraction of hand>w1118+HFD+E group were significantly higher than that of hand>w1118+HFD group (P<0.05 or P<0.01), but the expression of dFAS, Col4al, TAG and MDA level, heart rate and fibrillation were significantly lower than that of hand>w"18+HFD group (P<0.05 or P<0.01). There was no significant difference between the heart hand>Nmnat group and hand>Nmnat+HFD group (P>0.05). The expression of Nmnat, dFoxo, bmm, NAD+, SIR2 deacetylation, SOD activity and shortened heart fraction in the hand>Nmnat+HFD+E group were significantly higher than those in the hand>Nmnat+HFD group (P<0.05 or P<0.01), but the expression of dFAS, Col4al, TAG and MDA levels, heart rate and fibrillation were significantly lower than those in the hand>Nmnat+HFD group (P<0.05 or P<0.01). Conclusion: both overexpression of cardiac Nmnat and endurance exercise can prevent excessive accumulation of cardiac lipids, oxidative damage, decreased cardiac contractility and increased fibrillation induced by high-fat diet. The molecular mechanism is related to the fact that both of them can up-regulate the activity of cardiac Nm-nat/NAD+/SIR2/dFoxo pathway. The overexpression of cardiac Nmnat gene combined with endurance exercise can better reduce the occurrence of lipid toxic cardiomyopathy, which is related to the superposition effect of the combination on the up-regulation of cardiac Nmnat/NAD+/SIR2/dFoxo pathway activity. Corrosion is one of the important reasons for durability degradation of hydraulic concrete structures. A simulated dissolution test device was designed to study the change law of material properties before and after the corrosion of the reactive powder concreter under ammonium chloride solution and fresh water environment by 90 d simulation test method. The results show that: Under NH4CI solution environment, the dissolution depth is about 10.65 mm after 90 days of dissolution, and almost 0mm under fresh water condition. Under NH4CI solution environment, the amount of calcium leaching is about 3.15 g/L, and under fresh water condition, the amount of calcium leaching is about 0.56g/L. The corrosion resistance of RPC is about 6 times of ordinary concrete. For RPC specimens, the dissolution acceleration factor of 6 mol/L NH4CI solution is about 52 times that of fresh water. Compared with the active powder concrete without corrosion, The compressive strength decreases by 23% after 90days of corrosion. It can be seen from this paper that 6 mol/L NH4CI solution can significantly improve the accelerated test effect, shorten the test cycle, and ensure the safety and reliability of the test environment in laboratory accelerated test research. Reactive powder concrete has almost no corrosion phenomenon in freshwater environment, so it is more suitable for popularization and application in concrete structures with corrosion risk. Objective To determine the reliability of hand-held dynamometry (HHD) for lower limb isometric muscle strength measurement in children with Duchenne and Becker muscular dystrophy (DMD/ BMD). Methods A total of 21 children [20 males and one female; mean age was (7.88 ± 2.87) years, ranging between 3.96-14.09 years; mean age at diagnosis was (5.88 ± 2.88) years, ranging between 1.35-12.89 years; mean height was (120.64 ± 16.30) cm, ranging between 97-153 cm; mean body weight was (24.62 ± 9.05) kg, ranging between 14-50 kg] with DMD (19/21) and BMD (2/21) were involved from Rehabilitation Center of Children's Hospital of Fudan University. The muscle strength of hip, knee and ankle was measured by HHD under standardized test methods. The test-retest results were compared to determine the inter-test reliability, and the results among testers were compared to determine the inter-tester reliability. Results HHD showed fine inter-tester reliability (ICC = 0.762-0.978) and inter-test reliability (ICC = 0.690-0.938) in measuring lower limb muscle strength of children with DMD/BMD. Results also showed relatively poor reliability in distal muscle groups (foot plantar flexion and dorsiflexion). Conclusions HHD, showing fine inter-tester and inter-test reliability in measuring the lower limb muscle strength of children with DMD/BMD, can be used in monitoring muscle strength changing and assessing effects of clinical interventions. Appropriate schemes for classification of freshwater phytoplankton are prerequisites and important tools for revealing phytoplanktonic succession and studying freshwater ecosystems. An alternative approach, functional group of freshwater phytoplankton, has been proposed and developed due to the deficiencies of Linnaean and molecular identification in ecological applications. The functional group of phytoplankton is a classification scheme based on autoecology. In this study, the theoretical basis and classification criterion of functional group (FG), morpho-functional group (MFG) and morphology-based functional group (MBFG) were summarized, as well as their merits and demerits. FG was considered as the optimal classification approach for the aquatic ecology research and aquatic environment evaluation. The application status of FG was introduced, with the evaluation standards and problems of two approaches to assess water quality on the basis of FG, index methods of Q and QR, being briefly discussed. In GSM based passive bistatic radar, the antenna not only receives the direct signal and multipath signals from the host station, but also receives the signals from other base stations with the same frequency. When the data-independent beamformer is used to acquire the direct signal of the host station, it may be polluted by other signals. The direction of the direct signal of the host station can be first measured by a sub-space method. Then the direct signal is acquired by the signal blocking based adaptive beamformer. The availability of proposed method is proved by simulation data and compared in the practical data. Based on Koch's neural architecture and the philosophy of structural similarity, a novel visual salient region detection method is proposed. This method constructs a multi-feature visual saliency model by using SSIM's high abstraction for HVS, and it uses a new center-surround operator to measure the saliency degree of the natural images. The experimental results show that our method can detect the salient region effectively. In comparison to Itti's method, our method overcomes the weakness of Mosaic caused by neighboring interpolation, and it shows better performance on contour detection and anti-noise. Saliency detection is an important step image processing with computer, which includes the target recognition, image segmentation, and it is widely used in many fields such as network graph'ics and fingerprint recognition. In this paper, we proposed a global approach of structural similarity measure to significant target detection: using structural similarity to the human visual system's high- level abstraction, and taking into account the human visual characteristics and psychological feelings. We used the theory of structural similarity to measure the significant level of the target object, and get the saliency map after weight averaging the results. At last, the method of threshold was used to extract the salient target. Compared with the classic Itti algorithm, our approach can not only overcome the mo'saic and take into account the global spatial information on the impact of structural similarity, but also select the parameters according to computing power and precision. Atrial Ca 2+ handling abnormalities, mainly involving the dysfunction of ryanodine receptor (RyR) and sarcoplasmic reticulum Ca 2+ -ATPase (SERCA), play a role in the pathogenesis of atrial fibrillation (AF). Previously, we found that the expression and function of transient receptor potential vanilloid subtype 4 (TRPV4) are upregulated in a sterile pericarditis (SP) rat model of AF, and oral administration of TRPV4 inhibitor GSK2193874 alleviates AF in this animal model. The aim of this study was to investigate whether oral administration of GSK2193874 could alleviate atrial Ca 2+ handling abnormalities in SP rats. A SP rat model of AF was established by daubing sterile talcum powder on both atria of Sprague-Dawley (SD) rats after a pericardiotomy, to simulate the pathogenesis of postoperative atrial fibrillation (POAF). On the 3rd postoperative day, Ca 2+ signals of atria were collected in isolated perfused hearts by optical mapping. Ca 2+ transient duration (CaD), alternan, and the recovery properties of Ca 2+ transient (CaT) were quantified and analyzed. GSK2193874 treatment reversed the abnormal prolongation of time to peak (determined mainly by RyR activity) and CaD (determined mainly by SERCA activity), as well as the regional heterogeneity of CaD in SP rats. Furthermore, GSK2193874 treatment relieved alternan in SP rats, and reduced its incidence of discordant alternan (DIS-ALT). More importantly, GSK2193874 treatment prevented the reduction of the S2/S1 CaT ratio (determined mainly by RyR refractoriness) in SP rats, and decreased its regional heterogeneity. Taken together, oral administration of TRPV4 inhibitor alleviates Ca 2+ handling abnormalities in SP rats primarily by blocking the TRPV4-Ca 2+ -RyR pathway, and thus exerts therapeutic effect on POAF.

Objective To investigate the effects of self-made carriers on the cryopreservation of ovarian tissue of sheep. Methods Thirty-two ovaries were randomly assigned to fresh group,programmed freezing group,self-made carrier I vitrification group,and self-made carrier Ⅱ vitrification group.The morphology,proliferation,apoptosis,and estrogen level of the ovarian tissue in each group were observed. Results After cryopreservation,the morphology normal rate of the primordial follicles in programmed freezing group,self-made carrier I vitrification group,and self-made carrier Ⅱ vitrification group were 74.2%,72.8%,and 72.3%,respectively,lower than that(83.7%)in the fresh group(χ 2 =13.079,P=0.004).The percentage of normal primary follicles in programmed freezing group was lower than that in the fresh group(χ 2 =12.486,P=0.000).The percentage of normal primary follicles showed no significant difference between vitrification groups and fresh group(P=1.000,P=0.972).There was no significant difference in estrogen level or the positive expression rate of PCNA among the 4 groups(F=0.363,P=0.780;χ 2 =0.359,P=0.949).The number of apoptotic cells in cryopreservation groups was significantly higher than that in the fresh group(F=37.584,P=0.000),and it was significantly higher in the programmed freezing group than in the two vitrification groups(F=18.992,P=0.000). Conclusion Compared with slow programmed freezing,the vitrification with self-made carriers could well preserve the activity of cells in large sheep ovarian tissue blocks.

Objective: To investigate the regulatory effect of the transcription factor NF-kB1 on the expression of miR-195 in prostate cancer (PCa)., Methods: We analyzed the possibility of NF-kB1 binding to the miR-195 promoter and the expression of NF-kB1 in PCa using the JASPAR and Oncomine databases, respectively, and determined the expressions of NF-kB1 and miR-195 in PCa cells by real-time quantitative PCR after inhibiting the former by interfering RNA targeting NF-kB1. We detected the activity of the luciferase reporter gene after constructing its gene plasmid in the miR-195 promoter region and having it co-transfected with the NF-kB1 plasmid. Then we analyzed the correlation between the expressions of miR-195 and NF-kB1 in the prostate tissue., Results: NF-kB1 was overexpressed in PCa. After inhibition of the expression of NF-kB1, that of miR-195 was increased in PC-3 and DU-145 cell lines, with a negative correlation between the NF-kB1 and miR-195 expressions in the PCa tissue. The results of luciferase reporter gene assay showed direct binding of NF-kB1 to the miR-195 promoter zone., Conclusions: NF-kB1 regulates the expression of miR-195 in prostate cancer.

The succession of phytoplankton communities is affected by pure environmental factors as well as the interaction of various factors. Phytoplankton communities with eleven aquatic abiotic factors and four biotic factors were investigated in the Siminghu Reservoir, and the seasonal succession of phytoplankton FGs related to the biotic and abiotic factors was analyzed. The results suggested that a total of 22 phytoplankton functional groups were identified across the samples, and they were all affiliated with Chlorophyta and Bacillariophyta. An obvious seasonal succession was discovered by NMDS, which were present as D+Lo in summer, D+P+Lo in autumn and winter, and X2+P+MP in spring. In particular, we screened 10 functional groups with biomass significantly associated with the seasonal change in the phytoplankton community. A variance partitioning analysis (VPA) revealed that water temperature-zooplankton covariation and water temperature explained the variation in the phytoplankton functional groups throughout the year. A canonical correspondence analysis (CCA) showed that water temperature, transparency, nitrate nitrogen, and the biomass of zooplankton were the most critical factors determining the community dynamics of phytoplankton.

Objective: To investigate the nursing care of prostate cancer (PCa) patients againstradioactive proctitisinduced byCyberKnifetreatment., Methods: Sixty-eightPCapatients undergoingCyberKnife treatment in the observation group receivedspecialnursing care againstradioactive proctitis. The nursing measures includedthoserelevant toCyberKnife treatment, prevention ofradioactive proctitis, skin care, and discharge guidance. Meanwhile, another 54 prostate cancer patients received traditional nursing care as controls. We compared the incidence rate and severity of radioactive proctitis between the two groups of patients., Results: The incidence rate of radioactive proctitiswas markedly lower in the observation group than in the control (2.9% vs 13.0%, P<0.05), but no statistically significant difference was observed in the severity of radioactive proctitis between the two groups of patients., Conclusions: The special nursing care againstCyberKnife-induced radioactiveproctitiscan significantlyreduce the incidence of radioactive proctitis andimprove the effect of CyberKnife treatment of prostate cancer, which therefore deserves wide clinical application.

Objective: To explore the expression of long noncoding RNA (lncRNA) LINC01358 in prostate cancer (PCa) and its effect on the proliferation and migration of PCa cells., Methods: The lncRNA array was used to screen differentially expressed lncRNAs in PCa and the corresponding carcinoma-adjacent normal tissues from 3 patients. The expressions of LINC01358 in the primary PCa, metastatic PCa, and carcinoma-adjacent tissues were compared using the PCa dataset of the Memorial Sloan Kettering Cancer Center (MSKCC). The data obtained were validated by determining the expression of LINC01358 in the PCa and carcinoma-adjacent tissues of another 10 patients by quantitative real time PCR (qRT-PCR). The effects of lncRNA LINC01358 on the proliferation of DU145 cells and migration of PCa cells were detected by MTT and Transwell assay, respectively., Results: Totally, 79 differentially expressed lncRNAs in the lncRNA array, 36 highly and the other 43 lowly expressed in the PCa tissue. LINC01358 was up-regulated in the cancerous tissue. According to the MSKCC data, the LINC01358 expression was markedly higher in metastatic PCa (5.81±0.19, n = 19) and primary PCa (5.47±0.04, n = 131) than in the PCa-adjacent tissue (5.15±0.07, n = 29) and significantly correlated with postoperative biochemical relapse of the malignancy (P<0.05). qRT-PCR indicated a remarkably higher expression of LINC01358 in the PCa than in the carcinoma-adjacent tissue (6.02±1.12 vs 3.21±0.21, P<0.05). Transfection of the DU145 cells with siRNA significantly decreased the level of LINC01358 and inhibited the proliferation and migration of the PCa cells., Conclusions: LINC01358 is highly expressed in the PCa tissue and knockdown of LINC01358 may inhibit the proliferation and migration of PCa cells. LncRNA LINC01358 may be involved in the development and progression of PCa and become an index for the early diagnosis as well as a new target for the gene therapy of the malignancy.

"M+N"theory can be used as a method to improve the prediction accuracy in spectral analysis.The measured component,M kinds of non-measurement component,and N kinds of outside interference are induced into the entire measuring system,with the impact of"M"factors and "N"factors on the measurement accuracy considered systematically and comprehensively.Our human experiment system testing blood oxygen saturation based on "M+N"theory has been established.Dynamic spectrum method was used to eliminate the effects of different persons and different measuring parts which belonged to the system error of"N"factors.And then the D-value estimation was used to eliminate the effects of motion pseudo signal which belonged to the random error of"M"factors.Sixty two groups of valid data were obtained.The prediction model of blood oxygen saturation was built based on partial least squares regression method.The correlation coefficient and relative error were 0.796 8and ±0.026 6,while the result of oximeter was 0.595 7and relative error was±0.076 0,respectively.The results show that the prediction accuracy of the measurement method based on the"M+N"theory is much higher than that of the oximeter.

Objective: To explore if the new onset hypertension was related with metabolic risk factors and lifestyle in Beijing community residents to guide the targeted prevention of hypertension., Methods: A population-based survey with a randomly cluster sampling was made to observe the relationship between metabolic risk factors and lifestyle with new onset hypertension by multivariate and logistic regression., Results: A total of 4 034 subjects with normal blood pressure at baseline were included and 978 (24.2%) residents developed hypertension two years later. Multiple logistic analysis demonstrated that risk factors that contributed to new onset hypertension were prehypertension, overweight, obesity, frequent alcohol drinking, metabolic syndrome (MS), age and male gender, the respective Odds Ratios(OR) and 95% confidence interval (95%CI) are as follows: 3.512 (2.965-4.168), 1.654 (1.423-1.918), 2.264 (1.633-2.951), 1.493 (1.231-1.775), 1.329 (1.227-1.414), 1.226 (1.115-1.335), 1.135(1.018-1.221). College education serves as the protective factor, and the OR (95%CI) was 0.692 (0.476, 0.897)., Conclusions: New onset hypertension in Beijing community residents is mainly related with prehypertension, overweight or obesity, MS, frequent alcohol drinking and low education level. We should pay more attention to modulating metabolic risk factors and lifestyle to reduce the new onset hypertension in China.

Objective: To study the value of amino-terminal pro-brain natriuretic peptide (NT-proBNP) in predicting symptomatic patent ductus arteriosus (sPDA) in preterm infants., Methods: Preterm infants born at a gestational age (GA) of ≤ 32 weeks and diagnosed with patent ductus arteriosus (PDA) by echocardiography within 48 hours after birth between June 2014 and April 2015 were selected as subjects. Their clinical manifestations were observed, and serum NT-proBNP levels were measured and echocardiography was performed at 3 and 5 days after birth. The infants were divided into sPDA group and asymptomatic PDA (asPDA) group based on their clinical manifestations and the results of echocardiography. The correlations between serum NT-proBNP level and echocardiographic indices were analyzed. Serum NT-proBNP levels were compared between the two groups. The receiver operator characteristic (ROC) curve was applied to determine the sensitivity and specificity of serum NT-proBNP in the prediction of sPDA., Results: A total of 69 preterm infants were enrolled in this study, with 13 infants in the sPDA group and 56 infants in the asPDA group. Serum NT-proBNP level was positively correlated with the diameter of the arterial duct (r=0.856; P<0.05)and the ratio of left atrial diameter to aortic root diameter (LA/AO) (r=0.713; P<0.05). At 3 and 5 days after birth, the serum NT-proBNP levels in the sPDA group were significantly higher than those in the asPDA group (P<0.05). The area under the ROC curve (AUC) for the prediction of sPDA by NT-proBNP levels at 3 days after birth was 0.949 (95% CI: 0.892-1.000; P<0.001), with a cut-off value of 27 035 pg/mL (sensitivity: 92.3%; specificity: 94.6%); the AUC for the prediction of sPDA by NT-proBNP levels at 5 days after birth was 0.924 (95% CI: 0.848-1.000; P<0.001), with a cut-off value of 6 411 pg/mL (sensitivity: 92.3%; specificity: 92.9%)., Conclusions: NT-proBNP may be a quantitative index for shunt volume. The measurement of serum NT-proBNP levels on 3 and 5 days after birth may be useful to predict sPDA in preterm infants.

Objective: To explore the effect of Rhodiola on the expression of iNOS mRNA in severe acute pancreatitis (SAP) associated renal injury rats., Methods: A total of 72 healthy rats were randomly divided into the sham-operated group (S), the SAP associated renal injury group (M), and the Rhodiola-treated group (RHO), 24 in each group. Rats in S and M groups were peritoneally injected with 10 mL/kg saline 3h before modeling, while rats in the RHO group were peritoneally injected with 10 mL/kg Rhodiola Injection 3 h before modeling. The peripheral ligament of pancreas was bluntly dissociated in rats of M and RHO groups. The head of pancreas was occlused by nontraumatic blood vessel forceps 3 h later to establish the model. Eight rats were randomly selected from each group at 12, 24, and 36 h after modeling to detect levels of serum amylase, creatinine, and blood urea nitrogen. Serum levels of interleukin 1β (IL-1β) and interleukin 10 (IL-10) were detected by enzyme-linked immunosorbent assay (ELISA). Pathological changes of the left kidney were observed under light microscope. The expression of inducible nitric oxide synthase (iNOS) mRNA in the right kidney was detected with real time polymerase chain reaction (RT-PCR)., Results: Compared with the S group, serum levels of amylase, creatinine (Cr), blood urea nitrogen (BUN), IL-1β, IL-10, and iNOS mRNA expression significantly increased in the M group (P < 0.01). The function of kidney and pancreas were obviously improved in the RHO group than in the M group. Levels of IL-1β and iNOS significantly decreased, but IL-10 levels significantly increased in the RHO group with statistical difference (P < 0.05)., Conclusion: Rhodiola had better protective effect on SAP associated renal injury, which might be achieved through inhibiting the expression of IL-1β, stimulating the expression of IL-10, down-regulating iNOS mRNA expression, reducing the generation of oxygen free radicals and NO damage to cells, and improving hypoxia tolerance capabilities of the kidney.

Objective: To analyze the value of 2- and 9-question patient health questionnaires (PHQ-2 and PHQ-9) for screening status of depression in cardiovascular outpatients., Methods: From June 2013 to January 2014, a total of 201 outpatients from psycho-cardiological outpatients departments were consecutively enrolled into this study. All patients were asked to complete PHQ-9 and the mental psychological assessment by qualified researchers trained by psychiatry according to the composite international diagnostic interviews (CIDI), 50 cases were retested to assess the retest reliability after one week. The PHQ-2 and PHQ-9 were validated with the CIDI as the reference criteria., Results: According to the CIDI, 42 (21.3%) out of the 201 outpatients were diagnosed as depression. For PHQ-9 scale, a cutoff value of 10 presented satisfactory results with 87.1% sensitivity, 83.5% specificity, 58.7% positive predicting value, 95.6% negative predicting value and the area under the ROC curve (AUC) was 0.877 (SE = 0.032, 95% CI: 0.813-0.938). The Cronbach's alpha coefficients and test-retest reliability of the PHQ-9 were 0.809 and 0.882, respectively. Compared with CIDI, the cutoff value of the PHQ-2 was 3 with 85.7% sensitivity, 69.2% specificity, 57.1% positive predicting value, 93.6% negative predicting value, and the AUC was 0.806 (SE = 0.042, 95% CI: 0.774-0.889). The Cronbach's alpha coefficients and test-retest reliability of the PHQ-2 were 0.785 and 0.813, respectively., Conclusion: The PHQ-2 and PHQ-9 are reliable and efficient instruments for screening and especially excluding depression in cardiovascular outpatients.

The high-density sampling of eutrophic water can help to reveal the general rules of phytoplankton community succession and the relationship with environmental factors. The species and abundance of plankton and the physical-chemical factors were tracked for 30 weeks from March to October in a eutrophicated small pond. The relationships between the phytoplankton community dynamic change and the physical-chemical factors as well as the zooplankton in the water body community were studied by the multivariate statistical analysis with PRIMER. A total of 54 phytoplankton and 55 zooplankton species were identified, and the abundances of plankton varied from 0.28 x 10(8) - 6.11 x 10(8) cells x L(-1) for phytoplankton and 26-2.5 x 10(5) ind x L(-1) for zooplankton. The dominant species of phytoplankton were Cyanophyta and Chlorophyta, and an obvious succession process was showed with the seasonal change, which could be roughly divided into three stages: Chlorophyta-Cryptophyta type, Chlorophyta-Cyanophyta type and Cyanophyta type. BIO-ENV showed that pH, water temperature, transparency, total phosphorus and the abundances of Rotifera and Copepoda were the most critical influencing factors on the community dynamics of phytoplankton, and compared to zooplankton, the physical-chemical factors might have a greater influence.

To study the lead excretion effect of the chelator Zn-DTPA on the lead intoxication mice, inductively coupled plasma mass spectrometry (ICP-MS) was applied to detect the lead content of biological samples. The acute lead intoxication mice model was established by injecting lead acetate intraperitoneally with the dose of 1 mg. Zn-DTPA was administered intraperitoneally to mice once daily for five consecutive days 4 h after intoxication. Control group, model group, combination of Zn-DTPA and Ca-DTPA group were evaluated at the same time. The urine was collected every day. The mice were sacrificed in batches in the 2rd, 4th, 6th day. Biological samples including urine, whole blood, femur and brain were prepared and nitrated. Lead concentration was detected by ICP-MS. The result showed that Zn-DTPA could increase lead content in urine markedly and reduce lead content in blood, femur and brain.

Objective: To evaluate the effects of continuous paravertebral nerve block after nephrectomy., Methods: 60 cases receiving simple nephrectomy,American Society of Anesthesiologists(ASA) grading status I or II, aged from 30 to 65 years, weighing from 45 to 80 kg, male or female, were randomly divided into 2 groups: control group (CEA group ) and continuous paravertebral nerve block group (PVB group ), 30 patients in each one. Both groups were performed with 0.2% ropivacaine 250 ml in continuous infusion, assisted with intravenous morphine patient-controlled analgesia. The blood gas was examined in 6, 12, 24 and 48 h postoperatively, and the VAS scores of quiet state and motion state were assessed.In 48 h postoperation, the total amount of morphine was calculated and the complications cases were recorded., Results: Compared with the CEA group, PaO2 in PVB group increased and PaCO2 declined significantly. There was no difference of VAS score under quiet state, but under the motion state the VAS scores declined significantly, the total amount of morphine reduced[ (5.2 ± 1.2) vs (13.9 ± 1.6) mg) ], the cases of nausea, vomiting, pruritus , orthostatic hypotension decreased and the postoperative first anal exhaust time interval shortened (P < 0.05)., Conclusion: Compared with CEA, postoperative administration of PVB for open nephrectomy results in lowering morphine dose as well as side effect related with the use of morphine.

Appropriate schemes for classification of freshwater phytoplankton are prerequisites and important tools for revealing phytoplanktonic succession and studying freshwater ecosystems. An alternative approach, functional group of freshwater phytoplankton, has been proposed and developed due to the deficiencies of Linnaean and molecular identification in ecological applications. The functional group of phytoplankton is a classification scheme based on autoecology. In this study, the theoretical basis and classification criterion of functional group (FG), morpho-functional group (MFG) and morphology-based functional group (MBFG) were summarized, as well as their merits and demerits. FG was considered as the optimal classification approach for the aquatic ecology research and aquatic environment evaluation. The application status of FG was introduced, with the evaluation standards and problems of two approaches to assess water quality on the basis of FG, index methods of Q and QR, being briefly discussed.

Objective: To study the effect of chronic poisoning of ketamine on brain cell apoptosis in adult mouse under different duration and doses., Methods: The mouse model of chronic poisoning of ketamine was established on adult mouse by tail vein injection of ketamine twice every week with different doses (4, 10, 20 and 30 mg/kg). The mice were sacrificed after continuous injection of ketamine of 1, 2, 4, 8 and 12 weeks. The qualitative assessment of apoptosis was made by transmission electron microscope and the quantitative assessment was made by Caspase-3 immumofluorescence staining method and terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL) to estimate the time point of apoptosis. All the experimental results were statistically analyzed., Results: The neuron apoptosis was observed in hippocampus and corpus striatum by transmission electron microscope one week after administration, and continued for eight weeks. High level of Caspase-3 expression was observed one week after administration, but with a low level expression after 4 weeks. The number of TUNEL positive cells obviously increased one week after administration and maintained in a high number at 4 weeks., Conclusion: Ketamine by tail vein injection could induce neuron apoptosis in adult mouse.

Based on the characteristic of high speed line scanning for CCD in transient spectrum detection, a method of transient spectrum detection with array CCD is presented. The high speed line scanning with array CCD was realized by changing the mode of charge transfer. In order to explore the feasibility of this method, a fast detection system of single point based on linear CCD was designed and fabricated. Seven different LED pulses were measured when the system worked at fast detection mode of single point and normal mode respectively. The results demonstrate that the method of fast detection of single point based on linear CCD is feasible, and the rate of single point detection reaches up to 20 MHz. Thus, in theory, it was proved that transient spectrum detection with array CCD by changing the mode of charge transfer is also feasible.

Objective: To explore the feasibility of using enriched bone marrow (BM) compound with fibrin glue (FG) in repairing old radial bone defect., Methods: Totally 36 New Zealand rabbits were equally randomized into three groups: simple FG group, BM+FG group, and enriched BM+FG group. A 1.5-cm segmental bone defect was made at the left radial in each animal. After one month, the defect was implanted with the engineered bone. Before implantation, a compound of enriched BM with FG underwent electron microscopy, long-term culture, and bacteriological culture. Four, 8, and 12 weeks after operations, the osteogenetic effect was evaluated using X-ray observation, HE staining, or Van Gieson staining, and a semi-quantitative analysis was performed., Results: Electron microscopy showed enriched BM were compatible well with FG. No bacterial contamination or oncogenicity was observed after long-term culture. X-ray showed the repair effectiveness was significantly higher in BM+FG group and enriched BM+FG group than in simple FG group. Eight and 12 weeks after surgery, the Yang scores were significantly higher in enriched BM+FG group than in BM+FG group [(9.348±0.364évs.(7.984±0.229éìF=40.167ìP=0.001; (12.664±0.388)vs. (10.584±0.836é, F=20.3647ìP=0.004]. In addition, the Yang's scores at bone defects in BM+FG group and enriched BM+FG group were higher at the 12(th) week than in the 8(th) week. (F=36.004ìP=0.001; F=155.141ìP=0.000; respectively)The bone defects were repaired at varied degrees were histologically observed in BM+FG group and enriched BM+FG group during the observations., Conclusion: Implantation of BM+FG or enriched BM+FG are effective in repairing old radial bone defects, while simple FG shows not such effect.

Objective: To observe the effects of lithium chloride combined with human umbilical cord blood mesenchymal stem cell (hUCB-SCs) transplantation in the treatment of spinal cord injury in rats., Methods: Eighty female SD rats with complete T9 spinal cord transaction were randomized into 4 groups (n=20), namely the control group (group A), lithium chloride group (group B), hUCB-SCs group (group C) and hUCB-SCs(+) lithium chloride group (group D). On days 1 and 3 and the last days of the following weeks postoperatively, the motor function of the hindlimb of the rats were evaluated according to the BBB scores. At 8 weeks, all the rats were sacrificed and the spinal cords were taken for morphological observation. The spinal cord tissues at the injury site were observed with Brdu nuclear labeling to identify the survival and migration of the transplanted SCs. The regeneration and distribution of the spinal nerve fibers were observed with fluorescent-gold (FG) spinal cord retrograde tracing., Results: Brdu labeling showed that the transplanted hUCB-SCs survived and migrated in the spinal cord 8 weeks postoperatively in groups C and D. FG retrograde tracing identified a small amount of pyramidal cells that migrated across the injury site in groups C and D. The BBB scores of the hindlimb motor function 8 weeks postoperatively were 4.11∓0.14, 4.50∓0.15, 8.31∓0.11 and 11.15∓0.18 in groups A, B, C and D, respectively., Conclusion: Lithium chloride can promote the survival and differentiation of hUCB-SCs into neural cells at the injury site. Lithium chloride combined with hUCB-SCs transplantation may accelerate functional recovery of the hindlimbs in rats with complete transection of the spinal cord.

Novel hydroxyl-terminated ionic liquids of 1-(6-hydroxyhexyl)-3-butyl imidazolium bis(trifluoromethyl) sulfonylimide (HHBIM-NTf2) and 1-(8-hydroxyoctyl) -3-butyl imidazolium bis(trifluoromethyl) sulfonylimide (HOBIM-NTf2) were synthesized as stationary phases for capillary gas chromatography (GC). Besides, 1-octyl-3-butyl imidazolium bis(trifluoromethyl) sulfonylimide (OBIM-NTf2) was used as the reference. Selectivities and thermal stabilities of HHBIM-NTf2, HOBIM-NTf2 and OBIM-NTf2 were evaluated by means of the separation of Grob test mixture and positional isomers after the columns were conditioned up to 160 degrees C, 250 degrees C and 300 degrees C, respectively. As a result, baseline separations of Grob test mixture and alcohol mixture and better peak shapes were observed with HHBIM-NTf2 and HOBIM-NTf2. Importantly, baseline separation of dimethoxybenzene isomers was also achieved with HHBIM-NTf2 and HOBIM-NTf2. Satisfactory selectivity of HHBIM-NTf2 still remained even after conditioned at 300 degrees C for 8 h. The results for thermal stability showed that as GC stationary phases, HHBIM-NTf2 and HOBIM-NTf2 were stable at least up to 300 degrees C and 250 degrees C, respectively. The hydroxyl-terminated monocationic ionic liquids possess excellent selectivity and thermal stability and are alternative candidates for GC stationary phases.

Objective: To investigate suitable treatment method for contracture of inframammary scars., Methods: Nine female patients with contracture of inframammary sear hospitalized in our hospital from July 2000 to July 2007 were subjected to skin expansion around the breast. The sites of incisions were mainly located on the inframammary scars. The expanders were placed around the breast and middle chest near the sternum. On the lateral side of chest, the expander should be inserted at the site parallel to upper level of the breast. The expanders should be placed under deep fascia and superficial to the gland. At II stage of operation, the scars were excised and the subcutaneous tissues should be thoroughly loosened to assure that the soft tissue and mammary gland would be restored to its anatomical position. Expanded skin was then designed as advancement or transposition flaps to repair the defects, or effects were closed with suturing., Results: Blood circulation disturbance occurred at the tip of a flap in one patient, with the size of 4.0 cm x 3.0 cm, and the resulting wound healed after skin grafting. Flaps in the other 8 patients survived, and the wounds healed satisfactorily. Nipples and mammary areola were successfully restored to the anatomical positions. Three patients were followed up for 6 months to 2 years, and the result was satisfactory., Conclusions: Expanded flap is feasible for repairing contracture of inframammary scar and with good result.

Objective: To approach the effect of the donor antigenic specificity CD4+CD25+ regulatory T cell (Treg) on cellular immune tolerance function in rat composite tissue allotransplantation (CTA)., Methods: Use the method of immunomagnetic beads to separate CD4+CD25+ Treg, (1 x 10(6))CD4+CD25+ Treg was transfused to rat CTA model. Collected peripheral blood 30 days after operation, and used nylon wool column to separate B cell and T cell. With the stimulation of IgM, detected B cell proliferation and the level of IgG and IgA in serum. Observed the effect of CD4+CD25+ Treg on B cell and T cell function and the survival of allotransplants, and analyzed the data by statistics., Results: The purity of separated CD4+CD25+ Treg was 95.6%. The CPM of T cell of normal control group, topical intervention group, systemic intervention group and non-intervention group were (2436 +/- 358), (2273 +/- 136), (2338 +/- 228) and (3749 +/- 245). The CPM of B cells of normal control group, topical intervention group, systemic intervention group and non-intervention group were (2418 +/- 348), (2252 +/- 127), (2315 +/- 218) and (3720 +/- 224), there was a significant difference in these groups (P < 0.01). The serum level of IgG and IgA of topical intervention group and systemic intervention group were (12.56 +/- 1.30), (2.38 +/- 0.21), (13.48 +/- 1.23) and (2.86 +/- 0.24) g/L, and of normal control group was (12.35 +/- 1.28), (2.36 +/- 0.12) g/L, had no significant difference (P > 0.05). But Treg of non-intervention group was (16.58 +/- 1.12), (3.75 +/- 0.37) g/L, there was a significant difference in the non-intervention group and the three above groups (P < 0.01). The survival time of CTA in intervention of local and systemic groups were (97 +/- 13) and (63 +/- 10) d, which were significant longer than the non-intervention group [(22 +/- 8) d, P < 0.01]., Conclusions: Donor antigen specific CD4+CD25+ Treg has significantly inhibited B cell and T cell function. It can induce immune tolerance and extend the survival time of CTA; as well local application is better than systemic.
