Objective Analyze the current situation, hotspots and development trends of sarcopenia animal model to provide research direction and basic information for sarcopenia animal model research.Methods English literature of research on animal models of sarcopenia was retrieved from the Web of Science core data (WOS) set from 1900-01-01 to 2022-12-31. Chinese literature related to animal models of sarcopenia was retrieved from CNKI database between 1915 and 2022. The bibliometric analysis software VOSviewer was used to explore the countries, orgonizations, authors, hotspots and frontier directions in these studies.Results A total of 2 819 articles on animal models of sarcopenia were retrieved from WOS core database. The first paper was published in 1995. The United States has the largest number of animal model studies of sarcopenia with 1 105 articles. The institution with the most published articles is the University of Florida in the United States, with 69 articles. The University of Hong Kong has the highest number of publications in China, with 20 articles. American author Van Remmen H, with 50 publications, is the author of the most articles. The journal with the largest number of articles published on animal models of sarcopenia is the American journal called FASEB Journal, with 196 articles. In total, 423 articles on animal models of sarcopenia were retrieved from the CNKI database. Author LI Zhuyi has published 19 articles, and is the author of the most articles in China. The keyword co-occurrence clustering analysis of WOS literature search found that the research focus on animal model of sarcopenia can be summarized as the correlation between sarcopenia and metabolism, cytology and regenerative medicine of sarcopenia animal models, the study of sarcopenia animal models in bone, muscle, nerve and exercise therapy. The retrieval results of CNKI database revealed that the most extensive research was about on the model of denervated sarcopenia, and researches on the effects of Chinese medicine on sarcopenia were also widely reported. Through reading the full articles or abstracts of the literature, the animal models of sacopenia mainly include natural aging model, genetic modification model, high-fat diet induction model, disuse model, hormone induction model and complex sarcopenia models of other diseases.Conclusion In recent years, the study on animal model of sarcopenia has become a hotspot at home and abroad.The bibliometric analysis provides a basis for the research of animal models of sarcopenia in terms of research direction, hotspots, model animal selection, animal model making, and domestic and international communication and cooperation.

Thyrotoxicosis during pregnancy mainly includes gestational transient thyrotoxicosis (GTT) and Graves’ disease (GD). It is important to distinguish between GTT and GD because these two diseases are treated differently. At present, gold standard is lacking for the differential diagnosis of two diseases. Detection of thyroid-related antibodies combined with medical history and clinical manifestations are the most common differential methods. In recent years, researchers at home and abroad have proposed alternative valuable differential indexes, such as thyroid-related antibody, superior thyroid artery peak systolic velocity of color Doppler ultrasound of the thyroid, thyroid hormone and human chorionic gonadotropin, etc. In this article, the differential diagnosis significance of each index was summarized, aiming to provide better reference for the differential diagnosis of GTT in pregnancy and GD.

Objective To evaluate the teaching effect of stepped training model in transrectal ultrasound-guided transperineal prostate biopsy(PB). Methods Forty doctor trainees in the department of urology of Peking Union Medical College Hospital were randomly divided into two groups: experimental group and control group, with 20 trainees in each group. The experimental group received the stepped teaching model; The control group received the traditional training model. Both groups participated in the training of transperineal PB guided by transrectal ultrasound. One week later, all the members were tested for manipulation and surveyed by the teaching satisfaction questionnaires. Results Eighteen trainees in the experimental group could successfully perform transperineal PB independently, while only twelve trainees in the control group could operate independently, and the completion time of the experimental group was less than that of the control group(P＜0.01). The questionnaire survey showed that the teaching satisfaction score of trainees in the experimental group was higher than that in the control group(P＜0.01). Conclusions The stepped teaching model of transperineal PB guided by transrectal ultrasound is effective in clinical training.

Aiming at the problem that underwater acoustic channel is vulnerable to various threats and attacks due to the uncertainty of marine environment,a key agreement scheme based on ocean acoustic channel is proposed.Firstly,the uncertainty of marine environment is modeled,and the expressions of calculated noise,multipath and Doppler parameter expressions are constructed,and the concept of interference factor of underwater acoustic channel based on Rényi entropy is proposed.Secondly,a Hash function based on Twisted Edwards elliptic curve equation is constructed for conducting identity authentication and extracting the initial key.Then,the typical sequence of piecewise initial keys is used as initial seed to generate piecewise Toeplitz matrix,and the matrix multiplication of Toeplitz matrix and the initial key are used to generate the label by piecewise operation,and securely transfer the initial key.Finally,the initial key is hashed again for privacy amplification and a final secure key generated.The correctness,robustness and confidentiality of the scheme are proved by the information theory,and the upper bound of the probabi-lity of success of the active attack is obtained.Simulation results demonstrate that when the initial information amount is 50 000 bit,the upper bound of the success rate of adversary’s active attack is 4.3×10－23,and the key generation rate is 631 bit/s.Compared with existing schemes,the proposed scheme has obvious advantages in key generation rate and bit error rate.

Objective To evaluate the effect of the Jigsaw method in training of prostate targeted biopsy. Methods Thirty urologists who participated in the prostate biopsy training course in Peking Union Medical College Hospital in December 2020 were selected as the objects of study. They were randomly divided into two groups. The control group was trained with traditional teaching method and the observation group was trained by a Jigsaw method. Both groups of members were tested by same standard on theory and operation skills and surveyed for their by satisfaction by questionnaires. Results The results of examination of theory and operation skill in the observation group were significantly higher than those in the control group(P＜0.05). The questionnaire survey showed that the trainees in observation group were significantly more satisfied with the method than those in the control group(P＜0.05). Conclusions The Jigsaw method applied in the training of prostate targeted biopsy can stimulate trainees'learning enthusiasm and get remarkable learning outcomes so is worthy to be popularized.

Objective To analyze the biological processes regulated by protein kinase WNK2 in ovarian cancer cells based on protein phosphorylation modified mass spectrometry. Methods Ovarian cancer cell line SKOV3 was transfected with lentiviral vector to knockdown WNK2 and the obtained cells were further screened by drug screening. Then the total protein was extracted for protein phosphorylation modification mass spectrometry analysis. Bioinformatics analysis was adopted to detect the phosphorylation changes before and after WNK2 knockdown, and differential proteins were screened out. Gene Ontology (GO) analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis were employed to explore the functions of differentially phosphorylated proteins, and experiments were performed to verify the biological processes regulated by WNK2. Results Phosphorylation modified mass spectrometry indicated that WNK2 knockdown significantly decreased the phosphorylation of 745 proteins with 1 078 altered protein sites, and increased the phosphorylation of 328 proteins with 415 altered protein sites. KEGG and GO enrichment analyses showed WNK2 influences biological processes such as cytoskeleton organization, signal transduction, cell composition, and regulation of kinase activity. Phalloidin staining indicated after WNK2 knockdown resulted in broken and disordered cytoskeleton, while its overexpression promoted cytoskeleton remodeling. Conclusion WNK2 is involved in the cytoskeleton formation in ovarian cancer cells by regulating the phosphorylation of vimentin and other proteins.

Objective To investigate the expression of folate receptor 1(FOLR1) and mesothelin (MSLN) in epithelial ovarian cancer(EOC) and explore their clinical significance. Methods The data of clinical samples and transcriptome sequencing expression matrix of EOC were obtained from TCGA and GEO database(Ovarian Cancer TCGA, Provisional; GSE6008; GSE66957; GSE49997) to analyze the expressions of FOLR1 and MSLN in different tissues and their association with tumor staging and the prognosis of EOC. We also collected paraffin-embedded tissue samples of EOC(105 cases), ovarian borderline tumor(11 cases), and normal ovarian tissue(30 cases) from patients undergoing surgeries in our hospital between 2018 and 2019. The expression levels of FOLR1 and MSLN were detected by immunohistochemistry and their correlation with the clinicopathological characteristics of the patients was analyzed. Results Analysis of TCGA and GEO databases showed that the mRNA levels of FOLR1 and MSLN were significantly higher in EOC tissues than in normal ovarian tissues(P < 0.05), and was also higher in stage Ⅲ~Ⅳ than in stage Ⅰ~Ⅱ EOC(P < 0.05). Survival analysis showed that the patients with a high expression of FOLR1 had significantly longer overall survival(P < 0.05) and disease-free survival(P < 0.05), while the patients with a high expression of MSLN had significantly shortened overall survival(P < 0.05) and disease-free survival(P < 0.05). Immunohistochemical analysis showed that FOLR1 and MSLN proteins were lowly expressed in normal ovarian tissues, and their expressions increased moderately in borderline ovarian tumor and significantly in EOC tissues. The positive expression rates of FOLR1 and MSLN in EOC were 80.95% and 53.33%, respectively. The levels were of FOLR1 and MSLN differed significantly in different tissue types(P < 0.001), and their positive rates were significantly higher in stage Ⅲ~Ⅳ than in stage Ⅰ~Ⅱ EOC(P < 0.05). The positive rate of MSLN was significantly increased in patients with lymph node metastasis(P < 0.05). Subgroup survival analysis of TCGA database showed that patients with a low MSLN expression and a high FOLR1 expression(MLFH) at the mRNA level had the best prognosis, while those with a high MSLN expression and a low FOLR1 expression(MHFL) had the worst prognosis(median survival time 70.57 vs 25.85 months, P < 0.000 1); compared with the MLFL patients, the MHFL patients in the early stage(< 50 months) had a better prognosis, while beyond 50 months, the survival rate of the patients decreased, but the difference was not statistically significant. Conclusion In patients with EOC, a high expression of FOLR1 indicates a more favorable prognosis while a high expression of MSLN is associated with a poor prognosis. These 2 proteins may participate in the process of EOC, and their combined detection may help in prognostic evaluation of EOC.

Based on full-bridge (FB) and half-bridge (HB) sub-modules (SMs), the conventional hybrid modular multievel converter(MMC) is a typical topology of MMC with DC fault clearing ability. However, the proportion of FB SMs in conventional hybrid MMC is usually greater than 50%, so as to increase the cost and losses. To solve this mentioned issue, a DC fault commutated circuit based on thyristor-switched capacitor (TSC) structure in the hybrid MMC was integrated to reduce the number of FBs, the improved TSC-hybrid MMC was proposed in follows. In TSC-hybrid MMC, HB-MMCs and FB-MMCs were both retained their complete commutated body, and meanwhile, they were connected by a three-winding transformer in AC side and in series with DC side. In particularly, a thyristor-switched capacitor structure was integrated in the DC side of TSC-hybrid MMC to clear the DC fault. According to the analysis, the proportion of FB SMs in TSC-hybrid MMC can be reduced to 10%~25% or even lower (the AC-side voltage quality is a main limiting condition), the cost is reduced as well as the on-state losses. To solve the breeding problem of the serious shortage of female cassava flowers, cassava variety 'Xinxianxuan 048' was used as the experimental material. Transcriptional sequencing technology was used to analyze the biological information of differentially expressed genes in female and male flowers during the critical period of cassava flower sex differentiation, explore the functions of differentially expressed genes and possible regulatory pathways involved, excavate candidate genes related to female differentiation, and verify the sequencing results by qRT-PCR method. The results showed that: There were 545 differentially expressed genes between male and female cassava flowers at the critical stage of gender differentiation. Among them, 48.63% of the differential genes were enriched in GO pathway of floral organ morphogenesis and development, and the genes enriched in plant phenylpropanol biosynthesis and plant hormone signal transduction were the most. AGL11, YABBY4, CRC, SUP, and other flower sex differentiation genes were significantly up-regulated in female flowers. Four genes of the cytokinin signaling pathway, including HK4, HPt4, ARR8, and ARR12, were significantly up-regulated in female flowers, while IAA14, GH3, SAUR22, and SAURR20, the early auxin response genes, were significantly down-regulated in female flowers. Therefore, the sexual differentiation of cassava flowers mainly involved the biological pathways of pollen, gametophyte, floral organ morphogenesis and development, and two metabolic pathways of plant phenylpropanoid biosynthesis and plant hormone signal transduction. Cytokinin and auxin might be the main hormones in cassava female flower differentiation. Under the background of building a new power system based on new energy, the traditional decentralized small hydropower controller is more and more difficult to meet the construction needs of distributed small hydropower. In this paper, a distributed small hydropower controller based on ARM+DSP dual-core heterogeneous chip AM2732 is developed, which uses ARM to realize small hydropower control management, fast network transmission and integrity certification of transmission information, and uses DSP to realize small hydropower data acquisition and analyze the processing. According to design requirements, this paper introduces the overall design and hardware and software structure of distributed small hydropower controller. In particular, it focuses on the two key technologies of dual-core information exchange and remote communication integrity certification based on national secret algorithm. Finally, the field application of distributed small hydro-power controller shows that it not only has good functions of small hydropower regulation and communication, it can also support the development needs of new power grids such as small hydropower micro-grid. Capillary gravity circulation cabinet is a new type of terminal equipment dominated by natural convection for cooling in summer and heating in winter. In order to study its operation characteristics, taking an office at the air conditioning end of a capillary gravity circulation cabinet in Shanghai as an example, thermocouple and data acquisition instrument were used to continuously measure the temperature and wind speed at various indoor heights for 14 days. The characteristics of indoor flow field in summer was studied, and the thermal comfort and energy saving were evaluated. The results show that: the performance of gravity circulation cabinet air conditioning is good adaptabliity with outdoor temperature changes. The wind speed at the outlet of gravity circulation cabinet is 0.41 m/s, and that at the return air outlet is 0.20 m/s, the overall wind speed of the room 0.5 m away from the cabinet is less than 0. 10 m/s. The average temperature of indoor working area is 22.5 - 25.4°C, which meets the requirements of thermal comfort, and the temperature non-uniformity coefficient is small. The temperature at the ankle is low due to the cold air inking characteristics of gravity circulation cabinet, and the temperature non-uniformity coefficient is large. A hybrid system model consisting of a phosphoric acid fuel cell (PAFC), a regenerator, and a two-stage parallel thermoelectric generator (TPTEG) was established to harvest the waste heat during the operation of PAFC for performance enhancement. Taking account of the over-potential loss, heat recovery loss, heat leakage of PAFC and the main irreversible loss of the semiconductor, the relationship between the operating current density of PAFC and TPTEG was obtained through numerical analysis, the operating current density interval of TPTEG in the hybrid system was determined, the power and efficiency expressions of the hybrid system were derived, and the impact of TPTEG in parallel on the waste heat recovery of PAFC was analyzed .The results show that the performance of the PAFC-TPTEG hybrid system is better than that of a single PAFC system. The effects of operating temperature and pressure of the fuel cell, number of thermoelectric elements, thermal conductivity coefficient, and figure of mention on the hybrid system were studied. In order to convert RF energy in the environment into DC power for low-power equipment, a dual band energy harvester is designed, which consists of a wideband coplanar waveguide antenna and a new matching network. The broadband antenna's bandwidth is 0.8-2.8 GHz, which can collect radio frequency energy in the environment at 0.915 GHz and 2.4 GHz. Compared with microstrip antennas, the size of the broadband antenna is reduced by nearly 1/3. The new matching network can be matched at both the 0.915 GHz and 2.4 GHz and the matching bandwidth reaches 30 MHz and 100 MHz respectively. The experimental results show that when the system is about 50 cm apart from the reader of 0.1 W, it can effectively collect 201 mV DC power. When the system is about 10 cm apart from the router less than 0.1 W, it can effectively collect 33 mV DC power. This shows that the dual-band rectifier system designed can effectively collect energy in the environment at the ISM frequency bands (0.915 GHz and 2.4 GHz). [ABSTRACT FROM AUTHOR]

In order to reveal the phenotypic variation of Lilium brownii in natural populations native to Qinba moun-tainous area, and to discuss the relationship between phenotypic variation of natural populations, a systematic investi-gation was carried out among 17 natural populations in this area, containing 21 quantitative traits and 8 qualitative traits of both the flowers and the leaves.The results were as follows:with a strong adaptability, L .brownii had a wide distribution range of this area, most of them lived in mountains at an altitude height varying from 705 m to 1 913 m;Rich variation took place in the phenotypic traits.The length of the top leaves had a notably positive correlation with latitude, the width of the stigmas and the length of the top leaves had a notably positive correlation with altitude, the bottom leaf spacing had a notably positive correlation with the annual precipitation, while the length of pedicel and the length of the top leaves had a significantly negative correlation with the mean annual temperature;L .brownii n-ative to Qinba mountainous area could be divided into 2 groups:Group Qinling and Group Bashan, according to the re-sults of UPGMA cluster analysis.These findings may offer basic data for further studies on genetic breeding and con-servation biology of L .brownii . In this study, the leaf litters of Populus simonii and other 11 tree species were put into soil separately or in mixture after grinding, and incubated in laboratory to analyze the effects of their decomposition on soil properties and the interactions between the litters decomposition. The decomposition of each kind of the leaf litters in soil increased the soil urease, dehydrogenase, and phosphatase activities and the soil organic matter and available N contents markedly, but had greater differences in the effects on the soil available P content and CEC. The decomposition of the leaf litters of Caragana microphylla and of Amorpha fruticosa showed obvious effects in improving soil properties. The decomposition of the mixed leaf litters of P. simonii and Pinus tabulaeformis, Platycladus orientalis, Robinia pseudoacacia, or Ulmus pumila showed interactive promotion effects on the abundance of soil microbes, and that of the mixed leaf litters of P. simonii and P. orientalis or C. microphylla showed interactive promotion effects on the soil organic matter, available P, and available K contents and soil CEC but interactive inhibition effects on the activities of most of the soil enzymes tested. The decomposition of the mixed leaf litters of P. simonii and Larix principisrupprechtii showed interactive promotion effects on the activities of most of the soil enzymes and soil nutrient contents, while that of the mixed leaf litters of P. simonii and P. sylvestris var. mongolica showed interactive inhibition effects. Overall, the decomposition of the mixed leaf litters of P. simo-nii and U. pumila, P. tabulaeformis, L. principis-rupprechtii, or R. pseudoacacia could improve soil quality, but the mixed leaf litters of P. simonii and P. orientalis, C. microphylla, P. sylvestris var. mongolica, Hippophae rhamnoides, or A. fruticosa showed an interactive inhibition effect during their decomposition. Steel fiber reinforced self-stressing concrete(SFRSC) was used as strengthening material, and fatigue properties of damaged simple-support beams strengthened to continuous beams were studied. SFRSC, steel fiber concrete and ordinary concrete were used as wet joints and composite layer materials to strengthen simple-support beams respectively. Through fatigue tests, stress levels of reinforcements in hogging moment area and mid-span, crack widths and mid-span deflection were comparatively analyzed. The experimental results show that SFRSC strengthening simple-support beams to continuous beams can reduce stress amplitude of reinforcements in hogging moment area and mid-span, crack widths and mid-span deflection, and can improve structure fatigue life and normal use performance. Compared with beams strengthened by ordinary concrete, the maximum crack width in hogging moment area of beams strengthened by SFRSC can be decreased 25. 0%-32. 0% after 100 thousand fatigue cycles. Protein phosphorylation is one of the most common and important post-translational modifications that regulates almost all life processes. In particular, protein phosphorylation regulates the development of major diseases such as tumors, neurodegenerative diseases, and diabetes. For example, excessive phosphorylation of Tau protein can cause neurofibrillary tangles, leading to Alzheimer's disease. Therefore, large-scale methods for identifying protein phosphorylation must be developed. Rapid developmentin efficient enrichment methods and biological mass spectrometry technologies have enabled the large-scale identification of low-abundance protein O -phosphorylation modifications in, allowing for a more thorough study of their biological functions. The N -phosphorylation modifications that occur on the side-chain amino groups of histidine, arginine, and lysine have recently received increased attention. For example, the biological function of histidine phosphorylation in prokaryotes has been well studied; this type of modification regulates signal transduction and sugar metabolism. Two mammalian pHis kinases (NME1 and NME2) and three pHis phosphatases (PHPT1, LHPP, and PGAM5) have been successfully identified using various biological methods. N -Phosphorylation is involved in multiple biological processes, and its functions cannot be ignored. However, N -phosphorylation is unstable under acidic and thermal conditions owing to the poor chemical stability of the P-N bond. Unfortunately, the current O -phosphorylation enrichment method, which relies on acidic conditions, is unsuitable for N -phosphorylation enrichment, resulting in a serious lag in the large-scale identification of protein N -phosphorylation. The lack of enrichment methods has also seriously hindered studies on the biological functions of N -phosphorylation. Therefore, the development of efficient enrichment methods that target protein N -phosphorylation is an urgent undertaking. Research on N -phosphorylation proteome enrichment methods is limited, hindering functional research. Thus, summarizing such methods is necessary to promote further functional research. This article introduces the structural characteristics and reported biological functions of protein N -phosphorylation, reviews the protein N -phosphorylation modification enrichment methods developed over the past two decades, and analyzes the advantages and disadvantages of each method. In this study, both antibody-based and nonantibody-dependent methods are described in detail. Owing to the stability of the molecular structure of histidine, the antibody method is currently limited to histidine phosphorylation enrichment research. Future studies will focus on the development of new enrichment ligands. Moreover, research on ligands will promote studies on other nonconventional phosphorylation targets, such as two acyl-phosphates (pAsp, pGlu) and S -phosphate (pCys). In summary, this review provides a detailed analysis of the history and development directions of N -phosphorylation enrichment methods.

The discipline development is the pillar for the development of traditional Chinese medicine( TCM). The academic progress in TCM is the commanding height of the discipline development of TCM. To lead and promote the development and academic progress of TCM, the China Association of Chinese Medicine has summarized the Top Ten Academic Achievements in Traditional Chinese Medicine during 2020-2022, the Major Scientific Problems, Engineering Technical Problems, and Industrial Technical Problems in Traditional Chinese Medicine during 2019-2023, and the Remarkable Research Achievements of Traditional Chinese Medicine during 2012-2022. Based on the above research reports and the research achievements awarded the national science and technology prizes in TCM in the last 20 years and according to the current situation and layout of TCM discipline development, this paper reviews the major research achievements of TCM in the last two decades and the latest research progress in TCM during 2020-2023. The major scientific, engineering technical, and industrial technical problems in TCM are analyzed and the emerging trends of TCM are prospected in accordance with the development laws and characteristics of TCM. This review provides new ideas and reference for the high-quality development of TCM in the new era.

Objective: To comprehensively investigate and analyze the distribution characteristics of high altitude deterioration (HADT) among people working in 7 cities of Tibet Autonomous Region, to examine the relevant influencing factors, and to provide baseline survey data for further research on HADT., Methods: A self-designed questionnaire was used to conduct a self-administered survey among employees in seven prefectures or cities (Lhasa City, Qamdo City, Shigatse City, Nyingchi City, Shannan City, Naqu City, and Ngari Prefecture) in Tibet. The respondents were selected through random cluster sampling. The survey covered 21 symptoms involving 4 systems of the human body, including the respiratory, nervous, circulatory, and digestive systems. The distributive characteristics of HADT (as manifested by maladaptation to high altitude) were described and Spearman's correlation was used to examine the influencing factors of maladaptation to high altitude., Results: A total of 3 901 respondents were included in the sample analyzed in the study, including 2 107 (54%) native Tibetans and 1 794 (46%) immigrant Han people. There were 1 994 males (51%) and 1 907 females (49%). Their age ranged from 20 to 57 years, averaging (34.45±8.11) years. The subjects lived at a high altitude for a duration of 0.5 to 54 years, averaging (19.51±13.84) years. The overall rates of maladaptation for the 21 symptoms among native Tibetans and immigrant Han people were 60.10% (26 578/44 247) and 73.20% (27 565/37 674), respectively. The maladaptation rates of the native Tibetan population for the respiratory, nervous, circulatory, and digestive systems of the human body were all lower than those of the immigrant Han population ( P <0.001). There were no significant differences in the maladaptation rates of employees from different regions of Tibet (66.21% for Ngari Prefecture, 65.02% for Qamdo City, 66.67% for Lhasa City, 62.29% for Shigatse City, 65.03% for Shannan City, 64.42% for Nyingchi City, and 61.65% for Naqu City). The type of high-altitude residents (i.e., being native Tibetan or immigrant Han) was the main influencing factor for high-altitude maladaptation of the respiratory, nervous, circulatory, and digestive systems ( P <0.001). According to the findings of the correlation analysis, age, type of high-altitude residents, and the duration of residence at a high altitude were associated with high-altitude maladaptation of the respiratory system, while type of high-altitude residents was the only factor associated with maladaptation of the nervous system, circulatory system, and digestive systems. Age and duration of living at at high altitude had significant effect on self-perceived dyspnea, a type of maladaptation of the respiratory system ( P <0.001). Duration of high-altitude residence had significant effect on cyanotic lips or redness in the cheeks, a type of maladaptation of the circulatory system, and self-perceived loss of appetite, a type of maladaptation of the digestive system ( P <0.05)., Conclusion: More attention should be given to the HADT among employees of public institutions and enterprises who are living in Tibet Autonomous Region and immigrant Han people, in particular, should pay special attention to the protection of their respiratory, nervous, circulatory, and digestive systems., Competing Interests: 利益冲突　所有作者均声明不存在利益冲突, (© 2023《四川大学学报（医学版）》编辑部 版权所有Copyright ©2023 Editorial Board of Journal of Sichuan University (Medical Sciences).)

Targeted temperature management (TTM) has been partially applied in patients with restoration of spontaneous circulation (ROSC) after cardiac arrest (CA). In the 2020 American Heart Association (AHA) cardiopulmonary resuscitation (CPR) guidelines, TTM is used as advanced life support after ROSC for the treatment of patients with CPR. TTM has a protective effect on cardiac function after CA, but the specific mechanism of its protective effect on cardiac function remains unclear. In this paper, the basic experimental progress, clinical trial progress and development prospect of TTM on the protective mechanism of cardiac function after CA are reviewed.

In order to judge the future development trend of science and technology, plan ahead and lay out the frontier technology fields and directions, China Association of Chinese Medicine(CACM) has launched consultation projects for collecting &quot;major scienti-fic issues and engineering technology difficulties in traditional Chinese medicine(TCM)&quot; for the industry for three consecutive years since 2019. Up to now, 18 projects have been selected as major issues for research, and some experience and achievements have been made. These projects have been applied in important scientific and technological work such as scientific and technological planning and deployment at all levels of national, local, and scientific research institutions, the selection and cultivation of major national scientific and technological projects, and the construction of innovation bases, giving full play to the role of the think tank advisory committee of CACM. This study reviewed the selection of major issues for the first time, systematically combed its application in the national layout of science and technology, and put forward the existing problems and improvement suggestions, aiming to provide new ideas for further improving the selection of major issues and research direction, providing a theoretical basis and decision support for the national scientific and technological layout in the field of TCM, and promoting scientific and technological innovation to facilitate the high quality development of TCM.

Objective: To observe the effect of electroacupuncture on sexual development and ovarian estrogen receptor β(ER-β) expression in female adolescent obese rats induced by high-fat diet, so as to explore its underlying mechanisms of improving adolescent obesity., Methods: Female SD rats (age of 21 days) were randomly divided into control, model and acupuncture groups, with 6 rats in each group. The obese model was established by feeding high-fat diet for 6 weeks. Rats of the acupuncture group received electroacupuncture(2 Hz, 0.5-1.2 mA)stimulation at bilateral "Sanyinjiao"(SP6), "Fenglong"(ST40) and "Zusanli"(ST36) for 30 min, once a day for 14 days. The body mass and abdominal circumference of rats were measured before and after treatment. The contents of serum follicle stimulating hormone (FSH), luteinizing hormone (LH) and estradiol (E 2 ) were detected by ELISA. The number of corpus luteum and follicle were observed by HE staining. The expression levels of ER-β mRNA and protein in ovary were detected by fluorescence quantitative PCR and Western blot, respectively., Results: Compared with the control group, the body mass and abdominal circumference, the contents of serum FSH and E 2 , and the expression levels of ER-β mRNA and protein in ovary were significantly increased ( P <0.05)in the model group, while the number of mature follicles and corpus luteum increased significantly. Compared with the model group, the body mass and abdominal circumference, the contents of serum FSH and E 2 , and the expression levels of ER-β mRNA and protein in ovary were significantly decreased ( P <0.05) in the acupuncture group, while the number of mature follicles and corpus luteum decreased significantly., Conclusion: Electroacupuncture can effectively improve the levels of sex hormone and the development of ovary, down-regulate the expression levels of ER-β mRNA and protein in ovary, so as to regulate the process of sexual development of female adolescent obese rats induced by high-fat diet.

The rapid global spread of coronavirus disease 2019 (COVID-19), caused by the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), has introduced various challenges in global public health systems. The poor applicability and sensitivity of the reverse transcription-quantitative polymerase chain reaction (RT-qPCR) and antigen-based tests, as well as the persistent emergence of SARS-CoV-2 variants with different mutations hinder satisfactory epidemic prevention and control. Therefore, there is an urgent need for diagnostic technologies capable of distinguishing SARS-CoV-2 variants with high sensitivity and low (or no) equipment dependence. Diagnosis based on clustered regularly interspaced short palindromic repeats (CRISPR) has low equipment requirements and is programmable, sensitive, and easy to use. Various nucleic acid detection tools with great clinical potential have been developed for the diagnosis of infectious diseases. Therefore, this review focuses on the reported state-of-the-art CRISPR diagnostic technologies developed for the detection and differentiation of SARS-CoV-2 variants, summarizes their characteristics and provides an outlook for their development.

Advances in science and technology promote the rapid development of toxicological detection technologies. However, there is still a lack of decision-making tools for toxicological risk assessment, such as the lack of transparent schemes to evaluate current toxicological research and practice and the lag of toxicological testing tools to evaluate toxicity, resulting in difficulties in toxicity verification and hindering the transformation of toxicological research paradigm. Some scholars have proposed to integrate the concept of evidence-based medicine with the toxicological practice to improve the technical methods of toxicological research concept and risk assessment decision-making. With the promotion of relevant scholars and academic organizations, the concept and connotation of evidence-based toxicology have gradually become clear and a framework for research and practice has been initially formed. Although there are still many challenges, it also provides a new idea for the toxicity risk assessment and safe medication decision-making of traditional Chinese medicine(TCM). The era of digital intelligence has brought new opportunities and broad space for the development of TCM evidence-based toxicology. The exploration of TCM evidence-based toxicology from concept to method is an important embodiment of the development of TCM evidence-based toxicology, and will also promote the continuous enrichment and improvement of the research and practice system of TCM evidence-based toxicology.

Traditional Chinese medicine(TCM) carries the experience and theoretical knowledge of the ancients, and the use of &quot;toxic&quot; Chinese medicines is a major feature and advantage of TCM. &quot;Toxic&quot; Chinese medicines have unique clinical value and certain medication risk under the guidance of TCM theories such as compatibility for detoxification and treatment based on syndrome differentiation. In recent years, the safety events of Chinese medicines have occurred frequently, which has made the safety of Chinese medicine a public concern in China and abroad. However, limited by conventional cognitive laws and technical methods, basic research on toxicity of Chinese medicines fails to be combined with the clinical application. As a result, it is difficult to identify the clinical characteristics of, predict toxic and side effects of, or form a universal precise medication regimen for &quot;toxic&quot; Chinese medicines, which restricts the clinical application of them. In view of the problem that the toxicity of &quot;toxic&quot; Chinese medicines is difficult to be predicted and restricts the clinical application, the evidence-based research concept will provide new ideas for safe applcation of them in clinical practice. The integrated development of multiple disciplines and techniques in the field of big data and artificial intelligence will also promote the renewal and development of the research models for &quot;toxic&quot; Chinese medicines. Our team tried to propose the academic concept of evidence-based Chinese medicine toxicology and establish the data-intelligence research mode for &quot;toxic&quot; Chinese medicines and the intelligent risk prediction method for medicinal combination in the early stage, which provided methodological supports for solving the above problem. Thus, on the basis of summarizing the research status and problems of the clinical medication regimen of &quot;toxic&quot; Chinese medicines, our team took the evidence-based toxicology of TCM as the core concept, and tried to construct the multiple-evidence integrated evaluation and prediction method for &quot;toxic&quot; Chinese medicine, so as to guide the establishment of the non-toxic medication regimen of &quot;toxic&quot; Chinese medicines. Specifically, through the analysis of multivariate data obtained from the basic research, the evidence-based toxicology database of Chinese medicines and the individualized &quot;toxicity-effect&quot; intelligent prediction platform were built based on the disease-syndrome virtual patients, so as to identify the clinical characteristics and risks of &quot;toxic&quot; Chinese medicines and develop individualized medication regime. This study is expected to provide a methodological reference for the establishment of medication regimen and risk prevention strategy for &quot;toxic&quot; Chinese medicines. The method established in this study will bridge clinical research and basic research, enhance the transformation of the scientific connotation of attenuated compatibility, promote the development of evidence-based Chinese medicine toxicology, and ensure the clinical safety of &quot;toxic&quot; Chinese medicines.

Protein complexes are involved in a variety of biological activities. Accurate and comprehensive characterization of the structures and interactions of protein complexes is crucial in determining their biological functions. Chemical cross-linking coupled with mass spectrometry (CXMS) is an emerging investigative technique for protein complexes. CXMS enables the sensitive high-throughput analysis of protein complexes without the requirements of molecular weight and purification. These attributes have spurred the increased use of CXMS for the structure and interaction characterization of purified protein complexes and complicated cell lysate samples. CXMS utilizes chemical cross-linking reagents to covalently connect two reactive amino acids in or between proteins that are spatially close to each other. Subsequently, the cross-linked proteins are digested into cross-linked peptides, followed by LC-MS/MS analysis, as well as database searching to provide cross-linking information for the composition, interaction, and structural site distance restrictions of protein complex identification. Therefore, identification of cross-linked sites has a decisive influence on the characterization of protein complexes. This identification is limited by the unsatisfactory quality of the cross-linked peptide spectrum. Insufficient b/y fragment ions and poor continuity of amino acid sequence matching lead to low coverage and accuracy of cross-linked site identification. Based on the complementary feature of mirror-cutting digestion, an orthogonal digestion strategy based on LysargiNase combined with trypsin was developed in this study. Trypsin is the most commonly utilized digestion enzyme in proteomics, with extremely high enzyme activity and specificity. Trypsin generates C-terminally charged peptides after lysine (K) and arginine (R). LysargiNase is a mirror protease complementary to trypsin that cleaves before the K and R residues. This generates peptides with an N-terminal positively-charged residue. Owing to the different physical and chemical micro-environments of the cross-linked peptides digested by LysargiNase and trypsin, the behavior of their detection ability in MS analysis is diverse. Using the orthogonal digestion strategy, both simple and complicated cross-linked samples were analyzed in this study. For the analysis of bovine serum albumin (BSA), 291 pairs of non-redundant cross-linked sites were obtained, of which 216 pairs of cross-linked sites were provided by trypsin digestion, whereas 75 pairs of cross-linked sites were exclusively supplied by LysargiNase digestion. Except for the 35% increase in the number of identified cross-linked sites, 32% of the spectra of the commonly identified cross-linked peptides have better quality with more b-type fragment ions and consecutive sequence matching. Furthermore, for the Escherichia coli sample, 726 pairs of cross-linked sites were obtained in total, among which, 624 and 274 pairs were identified from trypsin and LysargiNase digestion, respectively. LysargiNase digestion yielded 120 individual cross-linked sites, which resulted in a 16% increase in single trypsin digestion. Consistent with the BSA sample, the quality was improved in 35% of the spectra of commonly identified cross-linked peptides. Corresponding to the identified cross-linked peptides, 242 structural constraints with 607 pairs of intra-cross-linked sites and 29 sets of protein-protein interactions with 119 pairs of inter-cross-linked sites were obtained. The collective results demonstrated that, mirror cutting-assisted orthogonal digestion strategy could significantly increase the number of identified fragment ions and amino acid sequences matching the continuity of the spectra by contributing b-and y-type ions, respectively. This improved the accuracy and coverage of cross-linked peptide identification. The findings additionally demonstrate the superiority of our method in the accurate identification of the cross-linked peptide spectra and the increased number of identified cross-linked sites. In a word, this method is expected to provide new insights for the large-scale and highly accurate characterization of protein complexes.

Background: Malignant plural effusion (MPE) is one of the most common specimen for liquid biopsy gene detection. This study aims to explore a method for isolating tumor cells from large volume of MPE and evaluate its efficacy and application prospect in gene detection., Methods: Pleural effusions (>500 mL) from 20 advanced lung cancer patients were obtained by effusion drainage and used to isolate tumor cells with cell separation media Percoll and Ficoll. Cell number and purity were calculated. DNA was extracted from the supernatant (etDNA), total cells and isolated tumor cells of pleural effusion (ETC-DNA) to detect the mutation of tumor-related genes by next-generation sequencing., Results: The median number of cells isolated from malignant pleural effusion was 8.50×10⁴ (interquel range: 9.25×10³-3.75×10⁵), 85.50%±5.80% of the cells were identified as tumor cells. The detection rates of epidermal growth factor receptor (EGFR) gene mutation of etDNA, total cell DNA and ETC-DNA were 70.00%, 50.00% and 70.00%, reseparately, while the median EGFR mutation abundance in 3 components was 16.05% (4.78%-43.06%), 1.09% (0.00%-2.39%), and 33.02% (18.50%-76.70%), respectively. ETC-DNA had good consistency with tissue DNA (P>0.999, kappa=1.000) and etDNA (P>0.999, kappa=1.000). ETC-DNA inclined to have higher EGFR mutation than etDNA, but the result was not statistically significant., Conclusions: Our method can isolate large amount of tumor cells from a large volume of malignant pleural effusion with high purity. Using ETC-DNA as specimen improves the efficacy of gene detection, thus is worth further study.

We investigated the characteristics of nitrogen uptake, distribution, and utilization in the three-year-old bearing Huangguan pear trees following 15 N-urea application in early spring. The results showed that the growth of pear trees was mainly depended on vegetative organs such as shoots and leaves at the stage from budbreak to shoot growth arrest, but mainly on storage organs (roots) and supplemented by the formation of fruit yield and quality at the stage from shoot growth arrest stage to fruit harvest. Meanwhile, tree biomass, especially that storage organs, substantially increased. All organs, especially newly developed shoots and leaves, acquired more N in shoot growth arrest stage due to vigorous growth, with relatively higher N derived from fertilizer (Ndff). Ndff of each organ except for root was lower at fruit maturity stage than that at shoot growth stage. Most of the labeled nitrogen was distributed in the newly developed organs (shoots and leaves) from budbreak to shoot growth arrest stage, but in the storage organs during shoot growth arrest stage to fruit maturity stage. Labeled fertilizer nitrogen was mainly distributed in the storage organs, followed by the vegetative organs. Reproductive organs had the lowest allocation in the experimental stage. For the three-years-old pear trees, the ratio of absorbed N from fertilizer was responsible for 31.1% and 21.0% of total absorbed nitrogen from budbreak to shoot growth arrest stage and from shoot growth arrest stage to fruit maturity stage, respectively, with the remaining N (68.9% and 79.0% of total) being absorbed from soil N.

This study was aimed to investigate the mechanism of Danzhi Jiangtang Capsules( DJC) in the treatment of diabetic macrovascular disease in Goto-Kakizaki( GK) rats. The diabetic macrovascular disease rat model was induced by feeding high-fat and high-sugar combined with endothelial nitric oxide synthase( NOS) inhibitor N-nitro-L-arginine methyl ester( L-NAME)( 0. 1 g·L -1 ·d -1 ). According to the random array table,the model rats were randomly divided into the model group,DJC groups( 1 260,630,320 mg·kg-1),atorvastatin group( 105 mg·kg -1 ) and metformin group( 10 mg·kg -1 ),with 12 rats in each group. The rats received gavage administration for 8 weeks. Twelve Wistar rats were selected as the normal control group. The changes of body weight,water intake,blood glucose,plasma total cholesterol( TC),triglyceride( TG),high density lipoprotein( HDL-C),low density lipoprotein( LDL-C),interleukin( IL-1β),IL-6,tumor necrosis factor( TNF-α),nitric oxide( NO),endothelin( ET-1) were observed in these rats. Aortic tissue was taken and the pathological changes were observed by HE staining. RT-PCR was used to detect the mRNA levels of IL-1β,IL-6,and TNF-α in rat aorta. RT-PCR of the stem loop was used to detect the levels of miRNA-126,miRNA-155,miRNA-146 a,and miRNA-21 in rat plasma and aortic tissue. The canonical correlation between miRNAs and inflammatory factors was then analyzed. The results showed that DJC increased the rat body weight,lowered water intake,reduced the random blood glucose,reversed the rat aorta tissue damage,reduced serum TC,TG,LDL-C,ET-1,IL-1β,IL-6,TNF-α,as well as miRNA-155,miRNA-146 a and miRNA-21 levels in serum,elevated plasma HDL-C,NO content,reduced the aorta mRNA of IL-1β,IL-6,TNF-α,and the miRNA-155,miRNA-146 a and miRNA-21,elevated miRNA-126 expression in aorta. Aortic miRNA-126,miRNA-155,miRNA-146 a and miRNA-21 expression levels were typically correlated with the expression of inflammatory factors,among which miRNA-126 was negatively correlated,miRNA-155,miRNA-146 a and miRNA-21 were positively correlated with the factors. These results suggested that DJC had therapeutic effects on diabetic macrovascular diseases,and the mechanism of action may be related to the regulation of miRNA-126,miRNA-155,miRNA-146 a and miRNA-21 levels,as well as the reduction of inflammatory factors and vascular inflammatory response.

Protein persulfidation is an important oxidative translational modification which plays vital roles in many important processes including cellular senescence, endoplasmic reticulum stress, vasorelaxation, and apoptosis. The proteome-wide analysis of persulfidation is of great importance; therefore, this study combines filter-aided sample preparation with an iodoacetic acid functionalized polyamidoamine dendrimer to enrich persulfidated peptides (denoted as filter-aided dendrimer enrichment strategy, FADE). To evaluate the performance of this strategy, the synthetic persulfidated standard peptide was spiked into bovine serum albumin (BSA) digests at a mass ratio of 1:100, and was successfully identified by FADE. Moreover, in combination with stable isotope labelling by amino acids in cell culture technology, the FADE strategy was applied to enrich persulfidated peptides from NaHS-stimulated SHSY5Y cells over a concentration gradient, resulting in the identification of 163 persulfidated peptides. Bioinformatic analysis indicated that persulfidation might play important roles in the central nervous system.

The field of circular non-coding RNAs have been gradually attracted wide attention with the developments of high-throughput sequencing. In this review, we systematically summarize three driving models for circRNAs biogenesis: intron-pairing-driven, RNA binding protein-driven and lariat-driven. In addition, we also briefly introduce the current research methods of circRNAs, which include high-throughput library construction methods, identification through bioinformatics and common experimental verification. Here, we also systematically summarize the functions of circRNAs, including microRNA (miRNA) or protein sponges, regulating the alternative splicing (AS) and expression of host genes, and extensive translation. Finally, we provide a systematic characterization and the latest research progress of circRNAs, which provide a new perspective for further studies of circRNAs in plants.

Since circulating tumor cells (CTCs) in the blood carry the genetic and phenotypic information of tumor tissues, they are regarded as important markers for liquid biopsy, especially for the early diagnosis of cancer, determination of treatment options and evaluation of prognosis. However, the capture and detection of CTCs is challenging because of their extremely low concentration in blood. In this review, recent advances in capture methods of CTCs based on their biophysical properties, affinity, and artificial antibodies are summarized. The advantages and drawbacks of these methods are discussed from the aspects of capture efficiency, capture purity and maintenance of activity after release. Future trends in CTCs capture are also discussed.

Background: Currently, there is no consensus on the follow-up strategy (follow-up time interval and content) of non-small cell lung cancer (NSCLC) in the world, and the relevant clinical evidence is also very limited. In this study, we aimed to summarize the recurrence/metastasis sites and timings of stage I NSCLC patients based on their follow-up data, aiming to provide a basis of follow-up time interval and content for this group of patients., Methods: We retrospectively analyzed the 416 stage I NSCLC patients that underwent continuous anatomic lobectomy between Jan. 2000 to Oct. 2013 in our prospective lung cancer database. According to the recurrence/metastasis sites and timings, the long term follow-up time interval and content were explored., Results: The 5-yr disease free survival (DFS) and overall survival (OS) in the whole group were 82.4% and 85.4%, respectively. There were 76 cases (18.3%) had recurrence/metastasis during follow-up, among which the most frequent site was pulmonary metastasis (21 cases, 5.0%), followed by brain metastasis (20 cases, 4.8%), bone metastasis (12 cases, 2.9%), and mediastinal lymph node metastasis (12 cases, 2.9%). Among the factors that could influence recurrence/metastasis, patients with pT2a suffered from a higher recurrence/metastasis rate compared to patients with pT1 (P=0.006), with 5-yr DFS being 73.8% and 87.3%, respectively (P=0.002), and the 5-yr OS being 77.7% and 90.3%, respectively (P=0.011)., Conclusions: The commonest recurrence/metastasis sites of stage I NSCLC after anatomic lobectomy are lung, brain and mediastinal lymph nodes, the risk of recurrence/metastasis within 2 years were equal to that between 3 years and 5 years. The follow-up frequencies and content within 2 years could be adjusted according to T stages.

Background: Thoracoscopic surgery has gradually become the major procedure for lung cancer surgery in our department. Its characteristics are minimal trauma and quick recovery, which make approximately 90% of patients discharge from the hospital after surgery. However, the postoperative complications still happen now and then. We analyzed the patients who had been hospitalized for longer than 7 days after thoracoscopic lung cancer surgery, aiming to summarize the types and risk factors of complications, and improve postoperative safety of patients., Methods: The data were come from the prospective database of Thoracic Surgery Unit One in Peking Cancer Hospital, and patients that underwent thoracoscopic pulmonary surgery between Jan. 2010 and Dec. 2014 with length of stay more than 7 days were included in the study. The classifications of the complications were investigated and graded as mild or severe complications according to modified Claviengrading, the relationship between clinical factors and degrees of complications was also analyzed., Results: The hospitalization of 115 cases were longer than 7 days after surgery, accounting for 10.3% (115/1,112) of the whole patients that underwent surgery during the same period. Eighty-one cases had mild complications, accounting for 7.3% (81/1,112) of the whole cases that underwent surgery during the same period and 70.4% (81/115) of the cases with prolonged length of stay; the proportions of severe complications in both groups were 3.1% (34/1,112) and 29.6% (34/115), respectively; and the proportions of complications that caused perioperative deaths were 0.18% (2/1112) and 1.7% (2/115), respectively. Among all the postoperative complications, the most common was air leakage for more than 5 days after surgery, with a total of 20 cases (1.8% and 17.4%). The other common complications were: atelectasis (19 cases, 1.7% and 16.5%), pulmonary infection (18 cases, 1.6% and 15.7%), etc. The less common complications was bronchopleural fistula (4 cases, 0.36% and 3.5%) with very high risk, and 2 cases died perioperatively due to the combination of acute respiratory distresssyndrome (ARDS). In the clinical factors, only preoperative low pulmonary function (FEV1%<70%) was the potential risk factor for postoperative severe complications (45.8% vs 23.6%, P=0.038). There was no significant difference either regarding the 5 year disease free survival or the 5 year overall survival between mild complication group and severe complication group, with 5 year DFS being 52.2% and 51.9%, respectively (P=0.894) , and 5 year overall survival being 64.0% and 53.5%, respectively (P=0.673)., Conclusions: Continuous postoperative air leakage, atelectasis and pulmonary infections were the major causes for prolonged hospitalization after thoracoscopic surgery for lung cancer, and bronchopleural fistula was the most perilous complications. Patients with low preoperative pulmonary function were more likely to have severe postoperative complication, however, this would not influence the long term survival of the patients.

Diabetes has become a global public health problem that seriously threatens human health. Traditional Chinese medicine, the characteristics of the role of multiple targets, has a unique advantage in the prevention and treatment of diabetes mellitus and its complications. Astragaloside-Ⅳ (AS-Ⅳ), one of the main activities of Astragalus membranaceus, has a series of pharmacological effects including improvement in the function of endothelial cells and neovascularization, anti-inflammatory, antioxidant, regulating energy metabolism, protectionnervous, anti-cancer and so on. In this paper, AS-Ⅳ to prevent and treat diabetes and its complications has been reviewed, which has effect on lowering blood sugar, lowering blood pressure, improving insulin resistance, inhibiting inflammatory reaction and oxidative stress. Additionally, it also can improve the diabetic animal and cell model of diabetic vascular disease, diabetic nephropathy, diabetic retinopathy, diabetic peripheral neuropathy, diabetic cardiomyopathy and other pathological damages. A novel method of the simultaneous detection of multiple kinds of allergenic proteins in infant food with parallel reaction monitoring (PRM) mode using liquid chromatography-tandem mass spectrometry (LC-MS/MS) was established. In this method, unique peptides with good stability and high sensibility were used to quantify the corresponding allergenic proteins. Furthermore, multiple kinds of allergenic proteins are inspected simultaneously with high sensitivity. In addition, such method was successfully used for the detection of multiple allergenic proteins in infant food. As for the sample preparation for infant food, compared with the traditional acetone precipitation strategy, the protein extraction efficiency and capacity of resisting disturbance are both higher with in-situ filter-aided sample pretreatment (i-FASP) method. All allergenic proteins gave a good linear response with the correlation coefficients ( R 2 ) ≥ 0.99, and the largest concentration range of the allergenic proteins could be four orders of magnitude, and the lowest detection limit was 0.028 mg/L, which was better than that reported in references. Finally, the method was conveniently used to detect the allergens from four imported infant food real samples. All the results demonstrate that this novel strategy is of great significance for providing a rapid and reliable analytical technique for allergen proteomics.

Objective: To investigate the clinical effects of aripiprazole on sexual dysfunction induced by amisulpride or risperidone in male patients with schizophrenia., Methods: This study included 75 male patients with drug-induced secondary sexual dysfunction after treated with amisulpride or risperidone for first-episode schizophrenia between October 2014 and October 2016. We substituted aripiprazole for amisulpride or risperidone, gradually increased the dose from 10 to 30 mg/d within 2 weeks, and maintained 30 mg/d from the 3rd week. At 4 and 8 weeks after medication, we evaluated the sexual function of the patients, measured the levels of serum prolactin (PRL) and testosterone (T), obtained the scores of the Positive and Negative Symptoms Scale (PANSS), recorded adverse reactions, and compared the parameters with those before aripiprazole administration., Results: Compared with pre-aripiprazole administration, the patients showed significant increases after 4 weeks of medication in the sexual function score (24.3 ± 2.1 vs 32.6 ± 3.6, P <0.05) and T level (［13.3 ± 2.7］ vs ［17.4±3.0］ mmol/L, P <0.05) but a decreased level of PRL (［38.5 ± 10.5］ vs ［27.9 ± 8.2］ ng/ml, P <0.05). At 8 weeks, the sexual function score and serum PRL were both restored to the baseline levels at admission, and the erectile function score, ejaculation score, total score, and serum T level even exceeded the baseline, though with no statistically significant differences (P >0.05). In comparison with pre-aripiprazole administration, the PANSS score was significantly decreased at 4 weeks after medication (62.1 ± 4.9 vs 57.2 ± 5.5, P <0.05) and even lower at 8 weeks (51.2 ± 5.2) (P <0.05). The incidence rates of medication-related excitation, dizziness, insomnia, and loss of appetite were 6.7%, 5.3%, 4.0% and 1.3% respectively, and no other serious adverse reactions were observed., Conclusions: Aripiprazole is effective for the treatment of drug-induced sexual dysfunction in schizophrenic men by continuously alleviating their positive and negative symptoms and meanwhile improving their sexual function and restoring their sexual hormone levels.

Avalanche photodiode is widely used in laser rangefinder due to high gain characteristics, but introduces highly additive noise during the time of current’s multiplication that makes laser rangefinder’s SNR meet bottleneck. This paper proposes a method of designing a high SNR’s graphene photodetector based on microcavity. The graphene’s unique optoelectronic properties make it an ideal platform for a variety of photonic applications, such as fast lasers, optical modulators, transparent electrodes, and ultrafast photodetectors. It has been recognized internationally to have dominant advantages in photodetectors due to its high carrier mobility, gapless spectrum, and frequency-independent absorption coefficient. With the wavelength of 1.06 μm, the mechanism of light waves’ transmission in the cavity and the graphehne’s absorption are studied by using optical transmission matrix method and scattering matrix method; the light absorption model of the graphene photodetector based on microcavity is established. Device’s final quantum efficiency reaches 91.2%, respectively reaches 0.778 A·W(-1), its full width at half maximum (FWHM) reaches 6 nm; the influence between graphene’s position in the microcavity and device’s absorption shows that device’s absorption’s peak value changes periodically with graphene’s position under resonant condition, and the variety of length of microcavity does not have any influence on the peak value, but changes the graphene’s position when absorption reaches peak value, on the condition that the length of microcavity is n times of half of wavelength, the number of device’s absorption peak value is 2n with the variety of graphene’s position, and all the peak values are symmetrical with respect to the center of microcavity, the final graphene’s position is 0.402 8 mm away from the top mirror of microcavity, and the absorption reaches 94%, Compared with single layer graphene, the absorption rate increases 16 dB; By solving SNR equation of the graphene photodetector based on microcavity and SNR equation of the avalanche photodiode, eventually finds that the SNR of the graphene photodetector based on microcavity is 90.3, which raises 10 dB compared with the avalanche photodiode’s. Theoretical analysis shows the graphene photodetector based on microcavity has high absorption rate, high quantum efficiency, and high SNR. In this paper, the research achievements provide a theoretical reference to update and design higher SNR photodetector used in laser rangefinder’s receiving system.

Objective: To compare the safety, feasibility, and efficacy of a completely nonfluoroscopic approach to radiofrequency catheter ablation (RFCA) using CARTO3 and ablation with conventional fluoroscopic guidance for treatment of idiopathic premature ventricular contractions from the aortic sinus cusp (ASC-PVCs)., Methods: From April 2013 to October 2015, we prospectively enrolled 52 consecutive patients with ASC-PVCs scheduled for either CARTO3 mapping-guided zero-fluoroscopy ablation (group A, n=23) or conventional fluoroscopic ablation (group B, n=29). The success rates, rates of complications, rates of recurrences, number of radiofrequency applications, procedure time, mapping time and fluoroscopy time were compared between the 2 groups., Results: s No significant differences were found in the success rates between the 2 groups [22/23 (96%) vs 24/29 (83%), P=0.21]. No major complications occurred during the procedures in either group. There was no significant difference with regard to the procedure time between the two groups (79.6∓8.8 vs 77.4∓7.2 min, P=0.332). The procedure was completed without any fluoroscopy use in group A, while the mean fluoroscopy time in group B was 23.1∓6.0 min. Group A showed a shorter mapping time than group B (4.3∓1.7 vs 7.8∓2.6 min, P<0.01) with significantly fewer radiofrequency applications (4.8∓1.1 vs 7.9∓3.2, P<0.01). The recurrence rates were comparable between the two groups over a follow-up period of 5 to 20 months., Conclusion: Compared with the conventional fluoroscopic technique, the zero-fluoroscopy approach can shorten the total procedure time and the ablation time with significantly reduced RF applications to eliminate ionizing radiation exposure in RFCA. RFCA guided by CARTO3 system without fluoroscopy is feasible, safe, and effective for treatment of ASC-PVCs.

Background: The bone and soft tissue sarcoma can metastasize to distant sites, most commonly the lungs. Some cases can be cured by radical metastasectomy, but its role, indication and prognostic factors remains controversial. The rarity of the disease combined with the diverse number of subtypes can make bone and soft tissue sarcomas very difficult to study. There are few randomized control studies or international high volume results, and such reports in China are seldom seen. The aim of this study is to investigate surgical treatment outcomes and prognostic factors of pulmonary metastatic bone and soft tissue sarcoma patients., Methods: From January 2007 to December 2015, patients with bone and soft tissue sarcoma who underwent multimodality therapy including definitive surgery for the primary lesion and at least one pulmonary metastasectomy were enrolled in the retrospective study. All the relevant clinical variables were collected, and then statistically analyzed and interpreted with the aid of univariate and multivariate Cox proportional hazard regression method., Results: Totally 155 pulmonary metastasectomies in 144 patients were analyzed. Incomplete R0 resection, a less than 1-year interval from a previous surgery, more than three detected nodules; and the summed maximum diameter of more than 45 mm for pulmonary metastases were independent prognostic indicators by multivariate analysis., Conclusions: We suggest that metastatic bone and soft tissue sarcoma patients can benefit most from aggressive surgical intervention of pulmonary metastasectomy. Its prognostic factors include R0 resection, a longer interval from a previous surgery, smaller total number and total size of pulmonary metastases.

Computational fluid dynamics was used to investigate the effect of the pathogenesis of membranous obstruction of inferior vena cava of Budd-Chiari syndrome with various angles between right hepatic vein and inferior vena cava.Mimics software was used to reconstruct the models from magnetic resonance imaging(MRI)angiograms of inferior vena cava,right hepatic vein,middle hepatic vein and left hepatic vein,and 3DMAX was used to construct the models of 30°,60°,90°and 120°angles between right hepatic vein and inferior vena cava,which was based on the reconstructed models.The model was conducted with clinical parameters in terms of wall shear stress distribution,static pressure distribution and blood velocity.The results demonstrated that the differences between wall shear stress and static pressure had statistical significance with various angles between right hepatic vein and inferior vena cava by SPSS.The pathogenesis of membranous obstruction of inferior vena cava had a correlation with the angles between right hepatic vein and inferior vena cava.

Objective: To elucidate the molecular and electrophysiological mechanisms of Brugada syndrome through functional analysis of a novel SCN5A gene mutation G1712C., Methods: A recombinant plasmid pRc+ channel β1-subunit, and the positive colonies were selected by screening with G418.The standard liposome method was used to transiently transfect HEK293 cells with either the wild-type or mutant Na + channel subunits (hH1 and mhH1, respectively), and the macroscopic Na + currents were recorded using whole-cell patch-clamp technique. Data acquisition and analysis, generation of voltage commands and curve fitting were accomplished with EPC-10, PatchMaster and IGOR Pro 6.0., Results: An HEK293 cell line that stably expressed Na + channel β1-subunit was successfully established. After transient transfection with the WT subunit, large Na + currents were recorded from the stable β1-cell line. Transient transfection with the G1712C subunit, however, did not elicit a Na + current in the cells., Conclusion: Compared with normal Na + channel, the wild-type channel exhibits a similar sodium current. The characteristic kinetics of sodium channel of WT-hH1 was identical to that in normal cardiac muscle cell, and the missense mutation (G1712C) in the P-loop region of the domain IV may have caused the failure of sodium channel expression.

Objective: To explore the effects of different analgesia methods after UPPP., Method: Ninety cases of patients uvulopalatopharyngoplasty were divided into 3 groups randomly, and 30 cases in each group. The group A was the blank control group without any analgesia measures. The cases in group B were treated with intramuscular injection of parecoxib sodium 40 mg after surgery immediately, and continued injecting 40 mg after 12 hours, 24 hours and 36 hours respectively. 100 mg tramadol replaced 40 mg parecoxib sodium in group C. The VAS scoring was performed after surgery 12, 24, 36, 48, 72, 96 hours in 3 groups, and we observed adverse reaction such as lethargy, nausea, vomiting, dizziness, skin rash and so on., Result: The group B and C reduced the pain significantly compared with blank control group. The pain scores in group B were significantly decreased than that in group C (P<. 05)., Conclusion: The analgesic effect of parecoxib sodium after UPPP is significant and better than tramadol. It is worthy to use widely in clinical due to its better effect and less side effect.

A kind of novel epitope surface imprinted particles was prepared by the reversible addition fragmentation chain transfer (RAFT) strategy. The epitope of transferrin, N-terminal peptide of the protein with nine amino acid residues, was chosen as the template and immobi- lized with covalent interaction on the surface of silica particles through the truss arm glutaraldehyde. The living/controlled polymerization was initialed by 2,2'-azobisisobutyronitrile (AIBN) at 70 °C in the solution of N,N-dimethylformamide, with the regulation by triothioester agent 2-(dodecylthiocarbonothioylthio)-2-methylpropanoic acid. Methacrylic acid and 2-hydroxyethyl methacrylate were chosen as the functional monomers and N, N-methylenebisacrylamide was chosen as the cross-linker in this polymerization. For this material, the binding capacity of the nine residue peptide could reach 2.36 mg/g with the imprinting factor (IF) of 1.89, while that for transferrin could reach 4.98 mg/g with IF of 1.61. The equilibrium could be achieved in 120 min for the transferrin recognition. In multi-protein competitive recognition, the imprinted factor of transferrin was the highest in the mixture of transferrin and other competitive proteins, such as cytochrome C and β-lactoglobulin. The results indicated that these epitope surface imprinted particles with RAFT strategy could recognize not only the nine residue peptide but also the transferrin with good selectivity, high binding capacity and fast mass transfer.

Objective: To study the safety, effectiveness and feasibility of suprapubis-assisted umbilical laparoendoscopic mini-dual-site surgery (SAU-LEMDS) in the treatment of varicocele., Methods: This study included 80 varicocele patients aged 24 - 44 (mean 28.5 +/- 2.6) years, 25 cases of grade I, 45 cases of grade II and 10 cases of grade III, 58 cases in the left side, 6 in the right and 16 in both sides, and all with asthenospermia. The patients were treated by SAU-LEMDS under subarachnoid anesthesia combined with general anesthesia in a supine position with a head-down-feet-up slope of 15 degrees. Two 5 mm trocars were inserted bilaterally at the umbilical edge, one with a 5 mm 30 degrees laparoscope placed in it, and another into the abdominal cavity below the pubic hairline with a 5 mm laparoendoscopic clipper placed in it. The operation procedure was similar to that of standard laparoscopic ligation of spermatic veins, with reservation of the spermatic artery and double-ligation of spermatic veins. And the procedure was repeated for the contralateral lesion in the bilateral cases. Postoperative follow-up was conducted for the incidences of orchiatrophy and testicular hydrocele and changes of seminal parameters., Results: All the operations were successful, with the mean operation time of (10 +/- 5.0) min (range 8 to 25 min) for the unilateral cases and (18 +/- 6.5) min (range 15 to 30 min) for the bilateral cases, the mean blood loss of (1.5 +/- 0.5) ml (range 1 to 2 ml), and the mean postoperative hospital stay of (2 +/- 0.5) d (range 1.5 to 3 d). The patients were followed up for 6 -24 (12 +/- 2.5) months, which showed significant improvement in sperm motility as compared with the baseline ([28.53 +/- 5.21] vs [19.62 +/- 3.56]%, P < 0.05), with 28 cases (35.0%) restored to normal. Recurrence was found in 4 cases (5.0%). Testicular hydrocele occurred in 7 cases (8.75%), but orchiatrophy in none. The scars in the umbilicus and suprapubis were invisible because of the wrinkles and pubic hair., Conclusion: SAU-LEMDS is safe, effective and feasible for the treatment of varicocele. It is superior to umbilical laparoendoscopic single-site surgery (U-LESS) for its less invasiveness, simpler operation, and better cosmetic appearance.

Objective: To investigate the cause and the management of treatment and prevention of reoperation following esophagectomy., Methods: Clinical data of 946 cases with esophageal cancer undergoing esophagectomy from January 2000 to December 2012 by the same surgical team in the Beijing Cancer Hospital were retrospectively analyzed. Among them, 19 patients underwent reoperation after esophagectomy because of serious complications. Clinical features and treatment course of these 19 cases were summarized., Results: The indications and procedures of reoperation included thoracotomy for hemorrhage (n=4), diaphragmatic hernia repair (n=4), thoracic duct ligation for chylothorax (n= 4), re-suturing for incision dehiscence (n=4), re-laparotomy and re-thoracotomy for drainage of traumatic pancreatitis (n=1), re-laparotomy for intestinal obstruction (n=1), and tracheotomy for bilateral recurrent laryngeal nerve paralysis (n=1). All the 19 patients were successfully cured without perioperative deaths and further complications., Conclusions: The indications of reoperation following esophagectomy include postoperative bleeding, diaphragmatic hernia, chylothorax and abdominal incision dehiscence.

Objective: To investigate the effects and regulatory mechanism of ILK on angiogenesis in hypertrophic scar., Methods: The human scar microvascular endothelial cells (HSMECs) were isolated from 6 patients' hypertrophic scar in vitro. The HSMECs with good condition in 2nd to 4th generation were selected as experimental objectives. (1) HSMECs were divided into the blank control group (treated with routine culture), negative control group (treated with only Lipofectamine 2000), LY294002 group (incubated with 50 nmol/L LY294002), ILK siRNA group (incubated with 20 nmol/L ILK siRNA). RT-PCR and Western Blot were used to detect the expression of ILK mRNA and its protein after transfecion for 48 h. (2) The digested HSMECs of four groups were resuspended with DMEM without serum and then seeded onto the upper compartment of transwell insert which contained complete medium in its lower compartment. The cell migration experiment was stopped in 10 h and then the migrated cells were counted to analyze the effects of different interventions on the migration ability of HSMECs. (3) The thawed ECMatrix was put into each well of pre-colled 48-well tissue culture plate, and then the plate was put into the incubator at 37 degrees C to make it to become gel. The HSMECs of four groups were seeded onto the surface of the ECMatrix gel and were put into incubator. Eight random view-fields per well should be valued by the sheet of pattern recognition about angiogenesis after 8 hours to evaluate the ability of angiogenesis in vitro between four groups., Results: (1) The expression of ILK mRNA (ILK mRNA = 0.829 +/- 0.109, t = 13.151, P = 0.006) and protein (ILK protein = 0.096 +/- 0.049, t = 36.656, P = 0.000) were both inhibited obviously in ILK siRNA group compared with the blank control group (ILK mRNA = 0.829 +/- 0.109, ILK protein = 1). And, the expression of ILK in LY294002 group was slightly lower than that of black control group, but there was no statistical difference. (2) The number of migrated cells in ILK siRNA group (88.111 +/- 3.079) and LY294002 group (138. 667 +/- 2.404) were respectively lower than that in blank control group (322.333 +/- 3.712, P < 0. 05) in 10th hour. (3) Compared to blank control group (4.333 +/- 0.191), the ability of angiogenesis in vitro decreased significantly ILK siRNA group (2.625 +/- 0.125) and LY294002 group (3.125 +/- 0.250), in which, the vascular network structures were not formed perfectly in 8th hour (P < 0.05)., Conclusions: The ability of HSMECs' migration and angiogenesis in vitro are inhibited significantly when the expression of ILK is down-regulated. It reveals that ILK may play an role in the regulation of scar angiogenesis.
