ObjectiveTo determine the human papillomavirus (HPV) infection status and possible influencing factors among healthy women aged 18‒45 years, and to provide a scientific evidence for the prevention of HPV infection and cervical cancer.MethodsA total of 23 HPV types were examined by PCR-reverse dot blot hybridization in 1 210 healthy women who voluntarily participated in the study. Infection status of each HPV genotype and possible factors associated with the infection were determined, including age, ethnicity, marital history, pregnancy history, number of sexual partners, and age at first sexual intercourse.ResultsA total of 1 210 healthy women were examined, of which 267 tested positive for HPV, with a prevalence of 22.07%. The prevalence did not differ significantly across age groups or ethnicities (all P>0.05). Objective To explore the regulative mechanism of hypoxia/reoxygen (H/R) for invasion ability of extravillous trophoblast (EVT). Methods Immortalized human EVT cell line HTR-8/SVneo in early pregnancy, primary EVT cells isolated from the villi of placenta in early pregnancy, and villi explants were cultured respectively. A simulation model of H/R environment in EVT cells invading the uterus was established under the conversion conditions of 1% O2 for 24 h to 20% O2 for another 24 h. Western blotting and immunofluorescence assay were used to detect the expression of key factors such as cell invasion, and Transwell chamber test was employed to observe cell migration and invasion. Results After H/R induction, the migration and invasion abilities of HTR-8/SVneo cells were weakened, and the epitaxial ability of cultured villi explants and the migration and invasion abilities of isolated primary EVT cells were also weakened. Under H/R conditions, the expression levels of MMP-2 and MMP-9, integrin-β1 and integrin-α5 were decreased in HTR-8/SVneo cells (P < 0.05), and the protein expression of DNA methyltransferase (DNMTs) was also down-regulated (P < 0.01). In the primary EVT cells under H/R condition, the expression levels of epithelial-mesenchymal transition (EMT) positive regulatory factors, Slug and Snails, were inhibited (P < 0.05), that of epithelial cell marker E-cadherin was increased (P < 0.01), protein levels of MMP-2 and MMP-9, integrin-β1 and integrin-α5 were decreased (P < 0.05), and cell invasion ability was reduced along with decreased DNMT1 and DNMT3A expression (P < 0.05). However, overexpression of DNMT1, DNMT3A and DNMT3B in HTR-8/SVneo cells promoted the up-regulation of interstitial markers N-cadherin, and MMP-2 and MMP-9 (P < 0.05). Conclusion H/R regulates the expression of EMT-related factors such as MMP-2/-9 through DNMTs, and thereby reduces the migration and invasion abilities of EVT cells.

In view of problem that traditional wire temperature measurement method cannot be applied in high-voltage switchgear because of high-voltage insulation, a wireless temperature measurement system of high-voltage switchgear was designed. Designs of hardware and software of the system were introduced in details, existing problems of the system in test process were analyzed and corresponding optimal measures were given. The test result showed that the system can steadily run in working environment and meet requirement of free maintenance in 2 a with once installation.

In order to access IED which do not support IEC61850 standard into integrated automation system of substation with IEC61850 communication system, the paper introduced overall design schemes of software and hardware of a communication manager of substation based on analysis of function requirements and application mode in communication system of substation of the communication manager. The communication manager is designed by use of Atom 230 microprocessor and Linux embedded operation system, which has multi network ports and multi serial ports for accessing all kinds of IEDs, and has function of converting traditional communication protocols into IEC61850 protocol.

In order to explore the influence of different working conditions on the mechanical properties of dovetail joints, in this paper, the "Qing Dynasty Engineering Rules" of the Ministry of Qing Dynasty and "Wood Construction Technology of Ancient Chinese Buildings" were used as the weighing standards, using the results of wood properties tests conducted by other scholars as a reference, the numerical simulation of pseudo-static load was carried out on the dovetail joints of Qing Dynasty based on the finite element software ABAQUS. The influence of different angles outside the plane of tenon, different materials, different axial loads, and different load positions on the mechanical properties of dovetail tenon joints was investigated. The results showed that: 1) The hysteretic curves of dovetail mortise and tenon are all inverted z-shape. 2) The maximum bending moments at dovetail joints were 0.896, 0.914 kN·m and 0.979 kN·m when the lateral angles of the Fangs were 0°, 1.41° and 2.82°, respectively. When the axial load at the top of the column increases from 0 to 50 kN, the two-way maximum bending moments of dovetail joints change from 0.896 kN·m and 0.523 kN·m to 2.680 kN·m and 5.129 kN·m respectively. The two-way yield moment changed from 0.823 kN·m and 0.473 kN·m to 2.366 kN·m and 4.172 kN·m respectively. When the axial load at the top of the column increases from 50 kN to 100 kN, the changes of the two-way maximum bending moment and yield bending moment do not exceed 1%. 3) The peak viscous damping coefficients of dovetail joints corresponding to horizontal displacements of 0, 25 mm, and 50 mm applied on the beam side are 22.05%, 28.50%, and 29.38%, respectively. ［Objective］In order to further explore the production operation and business management model in line with the actual operation of urban rail transit, ensuring the scientific process and optimization of the station management mechanism and management capacity, it is necessary to study the smart station management model under fully automatic operation (FAO) conditions. ［Method］Taking into account the current planning and construction situations for FAO and smart stations, a smart station management model under FAO conditions is discussed. According to the characteristics of FAO system and smart station, the management model of smart station under FAO is deeply analyzed; by comparing and analyzing the management models of traditional station and FAO smart station, suggestions for the FAO smart station management model are proposed. ［Result & Conclusion］Under FAO conditions, the regional management of smart stations is better than the natural station management of traditional stations, and can effectively strengthen the on-site business guidance. In addition, it is necessary to combine the new technology features of FAO system and smart stations to comprehensively improve the staff's capabilities in station service management, safety management, emergency management, organization management and personnel management, effectively ensuring the operational service quality.

The objects in remote sensing images have the characteristics of arbitrary direction and dense arrangement, and thus objects can be located and separated more precisely by using inclined bounding boxes in object detection task. Nowadays, oriented object detection in remote sensing images has been widely applied in both civil and military defense fields, which shows great significance in the research and application, and it has gradually become a research hotspot. This paper provides a systematic summary of oriented object detection methods in remote sensing images. Firstly, three widely-used representations of inclined bounding boxes are summarized. Then, the main challenges faced in supervised learning are elaborated from four aspects: feature misalignment, boundary discontinuity, inconsistency between metric and loss and oriented object location. Next, according to the motivations and improved strategies of different methods, the main ideas and advantages and disadvantages of each algorithm are analyzed in detail, and the overall framework of oriented object detection in remote sensing images is summarized. Furthermore, the commonly used oriented object detection datasets in remote sensing field are introduced. Experimental results of classical methods on different datasets are given, and the performance of different methods is evaluated. Finally, according to the challenges of deep learning applied to oriented object detection in remote sensing images tasks, the future research trend in this direction is prospected.

The purpose of the experiment was to study the effects of three unconventional fermented feeds on pig growth, serum biochemical indexes and antibody levels. The experiment divided 48 pigs into four groups, with 12 replicates in each group and one pigs in each replicate. The pigs in control group (group A) were fed the basal diet, and 30% unconventional fermented feeds B (3% soybean meal +10% corn +10% fish sauce +3% garlic skin +4% premix), C (3% soybean meal + 11% corn + 12% mango peel + 4% premix) and D (15% corn + 11% mango peel + 4% premix) were added to the basal diet in group B, group C and group D, respectively. The experimental period was 30 d. The results showed that compared with control group, the feed intake of group B and group D was significantly increased ( P<0.05), and the weight gain was extremely decreased ( P<0.01). The feed weight gain cost of group B and group D was 20.37% and 42.40% higher than that of control group, and the feed weight gain cost of group C was 3.17% lower than that of control group. Compared with control group, the serum TP, TC and TG content of pigs in group B were significantly increased (P<0.05), LDH activity and LDL-C content were extremely increased (P<0.01). The content of HDL-C in group D was significantly decreased ( P<0.05), and the content of LDL-C was significantly increased (P<0.05). Compared with control group, the serum PR-gB antibody level in group D was significantly increased ( P<0.05) and reached a positive level. The serum PR-gE antibody, PRRS antibody and PCVD antibody levels in group B were the lowest among all groups, and PR-gE antibody and PCVD antibody level was extremely lower than that of control group ( P<0.01). The serum PR-gE antibody level of group B was extremely lower than that of group C and group D ( P<0.01), and the level of PRRS antibody was extremely significantly lower than that of group D ( P<0.01). The study indicates that group C had the best production effect and cost-effectiveness, group B had the best disease resistance effect, and unconventional fermented feed C could be used as a substitute for the basal diet. This study proposes a finite element model for submerged water jet excavation using a cylindrical nozzle with an aim to determine the depth and diameter of jet orifices during water jet excavation. The model's accuracy was verified through indoor experiments, employing the Lagrangian-Eulerian fluid-solid coupling algorithm. The response surface method was utilized to establish a predictive model for the depth and diameter of jet orifices, which includes the impact of nozzle diameter, jet standoff distance, jet pressure, and their interactions. The construction parameters for jet excavation were optimized using a satisfaction function. Results indicated that for excavation depths of 10 cm, 15 cm, and 20 cm, keeping the stability of the jet orifices to the maximum required higher jet pressure(7.2 MPa), smaller jet standoff distance(1 cm), and an appropriate nozzle diameter(0.928 mm, 1.164 mm, and 1.345 mm). After optimization, the predicted values of jet orifice depth and diameter for the specific target excavation depths showed an error of less than 15 % compared to the experimental values, demonstrating the credibility and reliability of the predictions.

To standardize the market conduct of electricity sales companies and elevate the power market management level， it is essential to conduct a credit evaluation of these companies. Therefore， based on Box-plot and orthogonalization methods， a credit evaluation model based on an enhanced discrete Hopfield neural network （DHNN） is proposed. Firstly， factors influencing the credit levels of electricity sales companies are analyzed， and a credit evaluation index system， which includes 11 indicators such as basic information， foundational management， contract management， and transaction management， is established. The weights for these indicators are determined using the Delphi method. Secondly， outliers in the indicators of electricity sales companies are addressed to derive optimal credit scores， enabling an objective assessment of their credit levels. Finally， the feasibility of the proposed model is verified through case studies. The results indicate that the model can objectively and accurately evaluate the credit levels of electricity sales companies.

Objective To explore the value of gadolinium ethoxybenzyl diethylenetriamine pentaacetic acid （Gd-EOB-DTPA） enhanced MRI in evaluating the degree of esophageal varices. Methods Clinical data of 114 patients diagnosed with hepatitis B cirrhosis and Gd-EOB-DTPA enhanced MRI examination through clinical comprehensive diagnosis at Jiangsu Provincical Hospital of Chinese Medicine from September 2021 to June 2022 was retrospective analyzed. The patients were divided into non-varicose vein group, small varicose vein group and large varicose vein group based on the diagnostic threshold of 3 mm esophageal vein width. The signal intensity (SI) of liver, spleen and erector spinae muscles were measured in each group during the plain scan and hepatobiliary phase (20 minutes after enhancement). The relative liver enhancement (RLE), contrast uptake index （CUI）, and liver-to-spleen contrast index (LSI) were calculated. The differences in indicators between each group were compared, and the receiver operating characteristic curve (ROC) were drawn. The area under the curve (AUC) was used to evaluate the diagnostic efficacy of each indicator in diagnosing varicose veins. Results There was a significant difference in RLE, CUI, and LSI between the non-varicose vein group and the large varicose vein group (P＜0.05), while there was no significant difference in RLE, CUI, and LSI between the small varicose vein group, as well as between small varicose vein group and large varicose vein group (P>0.05). The AUC of RLE, CUI and LSI for diagnosing large varicose veins was 0.668 (95%CI: 0.558-0.793), 0.676 (95%CI: 0.567-0.804) and 0.685 (95%CI: 0.550-0.780), respectively. Conclusion Gd-EOB-DTPA enhanced MRI has certain value in evaluating the degree of esophageal varices.

ObjectiveTo systematically evaluate the efficacy and safety of Tianmeng Oral Liquid in the treatment of hyperprolactinemia caused by antipsychotics in female patients with schizophrenia， and to provide references for intervention of antipsychotic-induced hyperprolactinemia patients.MethodsEnglish databases （PubMed， Cochrane Library， PsycINFO and Embase） and Chinese databases （CNKI and Wanfang） were systematically searched， with a search time frame of the period from database creation to September 16， 2022. Randomized controlled studies （RCTs） on adjunctive Tianmeng Oral Liquid in the treatment of hyperprolactinemia caused by antipsychotics in female patients with schizophrenia were included. Three investigators independently screened the literatures and extracted data according to PICOS principles and evaluated the quality of including literatures， and the Meta-analysis was performed by using RevMan 5.3.ResultsA total of 3 articles including 256 female schizophrenic patients with hyperprolactinemia were included. Meta-analysis results showed that at the treatment endpoint， the improvement of hyperprolactinemia symptoms reported a significant superiority of the intervention group than the control group （RR=1.73， 95% CI： 1.07~2.79， P˂0.05）. Similarly， the significant superiority of the intervention group than the control group were also found in reducing serum prolactin levels （WMD=-55.17， 95% CI： -68.16~-42.18， P

ObjectiveA sensitive and rapid method was developed for the simultaneous determination of L-carnitine and D-carnitine in infant formula by pre-column derivation high performance liquid chromatography， and the key points were explored.MethodsCarnitine in sample was extracted with hydrochloric acid， saponified with potassium hydroxide， purified by acetonitrile and solid phase extraction column （SPE） and derivated. The derivative was separated by the C18 chromatography column， detected by UV detector， and quantified by external standard curve.ResultsThe linear ranges of L-carnitine and D-carnitine were 0.2-10 μg/mL with correlation coefficients of 0.999 9 and 0.999 4. The average recoveries of carnitine were 87.14%-96.96% under the 3 spiked concentrations. The limits of detection （LODs） were 0.6 mg/100 g， and the limit of quantification （LOQ） was 2 mg/100 g. The concentration of potassium hydroxide， purification process with SPE， solvent of derivatization reagent and operation in derivation were the key points in this method.ConclusionThis method is simple， accurate and is suitable for simultaneous detection of L-carnitine and D-carnitine in infant formula. And there is no significant difference between this method and enzymatic spectrophotometry.

Islands are not only important strategic space for China’s social and economic development, but also important barrier for ecological security. Evaluating the ecological status of islands is the basis for rational utilization, protection and management. In this paper, 77 islands were selected and the ecological status of each island in 2019 was evaluated according to the evaluation method of Island Ecological Index. The results showed that the majority of evaluated islands were of good ecological status, the ecological status of township-level islands with residents and non-resident islands was significantly better than that of village-level islands with residents, the distribution of Island Ecological Index in the Bohai Sea, the Yellow Sea and the East China Sea was similar, and the proportion of islands of good ecological status was higher than that of islands in the South China Sea. Vegetation coverage, natural shoreline retention rate and sewage treatment rate were the main factors affecting the ecological status of islands in 2019. Effective measures should be taken to help improve the ecological status of islands and promote the realization of sustainable development of islands, such as strengthening the investment in ecological protection and construction of village-level islands with residents, rationalizing the use of island resources, optimizing the pattern of protection and development, retaining natural shorelines, improving the sewage treatment capacity and so on.

Traditional wireless positioning technology has low accuracy, and UWB positioning technology can achieve centimeter-level high-precision positioning to meet the accuracy requirements of the fully mechanized working face positioning systems. However, the existing researches ignore the impact of position change of the base station installed on the hydraulic support on the positioning effect. In order to solve the above problems, laser ranging technology is applied to the mine environment and a positioning system of fully mechanized working face based on UWB and laser ranging is designed. The positioning base station is installed on the hydraulic support, and the position of the base station changes when the position of the hydraulic support changes. The mine intrinsically safe laser ranging sensor is used to measure the distance between the positioning base stations. The external antenna of the positioning base station transmits fixed-power UWB wireless signals. The positioning base station communicates with the positioning identification card worn on the personnel or installed on the equipment, and determines the positioning information of the moving target through the time-of-flight ranging method. The positioning base station uploads the positioning information data to the ground data center through the mine ring network. The system test is carried out on the E2308 working face of Gaohe Coal Mine, and the result show that the positioning accuracy of the system reaches 0.193 m and the positioning accuracy is high. Moreover, the system base station position calibration accuracy is about 0.011 m, which solves the problem of base station position calibration effectively.

ObjectiveTo explore the differences of cognitive function in patients with treatment-resistant depression and drug-naive first-episode major depressive disorder， and to examine the relationship between severity of clinical symptoms and cognitive function， so as to provide references for prognosis improvement.MethodsFrom November 2016 to December 2019， 119 patients with drug-naive first-episode major depressive disorder and 82 patients with treatment-resistant depression in a hospital in Guangzhou were enrolled， meantime， another 71 healthy individuals recruited from the community were set as healthy control group. Clinical symptoms were assessed using Hamilton Depression Scale-17 item （HAMD-17） and Hamilton Anxiety Scale （HAMA）. Cognitive domains， including speed of processing， working memory， verbal learning and memory， and visual learning and memory were measured with the MATRICS Consensus Cognitive Battery （MCCB）. Multiple covariance analysis was used to compare the differences in cognitive function among three groups. Thereafter， partial correlation analysis was performed within patient groups to explore the relationship of HAMD-17/HAMA score with the four dimensions of MCCB.ResultsThe speed of processing， visual learning and memory scores of treatment-resistant depression group and drug-naive first-episode depression group were lower than those of healthy control group， and the working memory score of the treatment-resistant depression group was lower than that of the healthy control group， with statistical significance （P0.05）.ConclusionCompared with drug-naive first-episode major depressive disorder patients and healthy controls， the impairments of speed of processing， visual learning and memory are more severe in patients with treatment-resistant depression. Moreover， the cognitive function impairment in patients with drug-naive first-episode major depressive disorder and treatment-resistant depression has no correlation with the severity of depressive and anxious symptoms.

In view of problem that theoretical modeling and simulation experiment are mostly used in research of underground path loss characteristics of high -precision ultra wide band (UWB) signal, actual measurement research of UWB signal is less. The path loss measurement and analysis of UWB signal are carried out in two typical scenarios of running roadway of overhead passenger device (monkey car) with relatively common environmental conditions such as roadway size, roadway wall material and pipe facilities，and heading face with bad environment, high coal dust concentration and serious water accumulation. The results show that: ① The path loss index of UWB signal in two kinds of roadway environments is less than 2, and wave guide effect of roadway leads to better propagation characteristics of UWB signal than that of free space. The base station deployment interval of the UWB positioning system in roadway is larger than that of the ground free space propagation scene.② In the same mine, the roadway environment is different, and the path loss characteristics are also different. The path loss index and shadow fading variance of heading face are significantly different from those of monkey car running roadway. ③ From comparison of single test point in roadway of heading face, the path loss in NLOS environment is larger than that in LOS environment. More intensive positioning base stations need to be deployed in roadway in NLOS environment.The results can be used to guide deployment scheme design of mine UWB precise positioning system.

To explore the effects of different oil producing processes on the quality and oxidation stability of Paeonia ludlowii seed oil. The physical and chemical characteristics, oxidation stability and fatty acid composition of Paeonia ludlowii seed oil were tested, and the effects of different refining processes on the quality and oxidation stability of Paeonia ludlowii seed oil were analyzed. The results showed that the iodine value after decolorization was 140 g I/100 g, the acid value after deacidification was 0.36 mgKOH/g, the peroxide value was 1.89 mmol/kg and 0.76 mmol/kg after deacidification and decolorization respectively,and the saponification value was 182 mg/g after deodorization, all of them had significant differences(P＜0.05), while the refractive index did not change significantly and had no significant influence. The order of oil oxidation stability is: degumming oil ＞ crude oil ＞ deodorization oil ＞ deacidification oil ＞decolorization oil; Degluing and deodorization had significant effects on the stearic acid and palmytic acid components in Paeonia ludlowii seed oil respectively (P＜0.05), while there was no significant difference in other components.

Objective To develop a method for objective quantitative analysis of microtubule structure based on image processing and analysis of the pixel ratio and particle size. Methods Human umbilical vein endothelial cells (HUVECs) cultured for 3 d were divided into control group, depolymerization group and polymerization group. Western blotting was performed to detect the changes in the content of acetylated α-tubulin in the cells. Immunofluorescence staining was used to observe the microtubule morphology under a laser confocal microscope, and the acquired images were analyzed using Matlab 7.0 for image segmentation, enhancement, and extraction of the feature parameters. Results ① The content of acetylated α-tubulin was significantly lower in cells with depolymerization and significantly higher in cells with polymerization than in the control cells (both P < 0.05). ②The control cells showed bundles of intact microtubules around the cell nuclei, with a few small fluorescent spots in the cytoplasm. In depolymerization group, the cells presented with broken microtubule fibers with numerous small fluorescent spots distributed diffusely in the cytoplasm. In the polymerization group, a large number of thick microtubule fibers were observed, showing multiple branches with an irregular and roughly radial alignment, and many large fluorescent spots were seen in the cytoplasm. ③The parameters for pixel ratio and particle size analysis, including the average particle size, sorting degree, skewness and kurtosis, all differed significantly among the 3 groups (P < 0.05). ④Based on quantification of these characteristic parameters of the microtubules, the accuracy of Fisher's multivariate discriminant analysis could reach 80.0%. Conclusion The pixel ratio and the characteristic parameters for particle size analysis can be used as objective indicators for quantitative analysis of the microtubes in fluorescence images of cells.

Objective To investigate the application of superb micro-vascular imaging(SMI) technology in accurately targeted biopsy of prostate cancer. Methods One month after 87 patients with clinically suspected prostate cancer received the first systematic 12-points puncture, 48 patients received system 12-points puncture, 39 patients received SMI technology to accurately target abnormal areas replacing local system puncture. According to histo pathological confirmation of the first puncture, the results of two methods in prostate cancer biopsy were compared and analyzed. Results There was no significant difference in prostate volumes between patients with malignant and benign prostate diseases, and the difference in serum PSA levels was statistically significant(t=2.045, P=0.04). In the SMI group, 20 cases were malignant, puncture positive rate was 51.28%, and the total numbers of puncture were 456 needle points; in the control group, 14 cases were malignant, puncture positive rate was 29.17%, and the total numbers of puncture were 576 needle points; there were significant differences in puncture positive rate and the average numbers of puncture point between the two groups (χ2=4.420, P=0.036; t=13.18, P=0.000). There were 79 cases of malignant pathological tissues and the highest Gleason scores were 7.2±0.4 in the SMI group; in the control group, there were 64 cases of malignant pathological tissues, and the highest Gleason scores were 6.5±0.4; there were significant differences between the two groups in the positive rate of single sampling and the highest Gleason score (χ2=8.232, P=0.004; t=5.940, P=0.000). Conclusion SMI technology to guide prostate cancer biopsy could improve the positive rate of draw materials.

Nanosized FeNi particles were loaded along multiwalled carbon nanotubes (MWCNT) to achieve high performance to attenuate electromagnetic (EM) waves of 2-18GHz and power frequency (50Hz). The MWCNT/Ni was produced through the electroless plating method, then followed by a polyol approach to produce MWCNT/FeNi composite nanowires. The microstructure of MWCNT/FeNi composite nanowires was characterized by transmission electron microscopy, scanning electron microscope, X-ray diffraction and vector network analysis. BackgroundDue to the special physical properties of gallium, gallium is considered to be used as reactor coolant in the future. Therefore, a full set of gallium neutron data is needed. However, the experimental data of gallium in EXFOR (Nuclear reaction experimental data) database is rare. There are also some discrepancies among the evaluation data.PurposeThis study aims to obtain elastic scatting differential cross section for natural gallium at 8.0 MeV incident neutrons, so as to fill the blanks of the neutron data of gallium in the database and clarify the discrepancy among the evaluation data.MethodsFirst of all, the elastic scatting differential cross section data of natural gallium was measured at 21 different laboratory angles from 20° to 140° by using the multi-detector fast neutron time-of-flight (NTOF) spectrometer on HI-13 tandem acceleration at the China Institution of Atomic Energy (CIAE). The cross sections of gallium were normalized by n-p elastic scatting cross section, and the quality of data was improved by many methods applied to the measurement and data analysis. Then, the Monte-Carlo code STREUER was employed to rectify the neutron flux attenuation, multiple scattering and the finite geometry of the experiment. Finally, the measured TOF spectra were compared with that of simulated to determine experimental scatting differential cross section of 8.0 MeV incident neutrons.Results & ConclusionsThe results of cross section of secondary neutron for natural gallium are the world's first experimental data in the 8.0 MeV energy region. Compared with the evaluated data, the measured data is generally in good agreement with CENDL-3.2 data, but inconsistent with ENDF/B-8.0 data and JENDL-4.0 data. The divarication among these three data bases has been clarified by the experimental result, hence of great significance to data evaluation and theoretical model improvement.

To attain the relation information of organizations all of which have the same attribute, an organization-relation analysis procedure based is proposed on SNA. The approach as: a social network is constructed that take academies of social sciences organizations as actors. In quantitative and qualitative analyses, the subject-characteristic of actors are shown, organization-relations results from subject-cooccurence, and hierarchical clustering bring on distribution of organization-relations. This paper establishes the hydrodynamic model and mathematical pollutant transport and diffusion model of long distance river channel and cascade reservoir and verifies the models by analytic solutions of pollutant transport and diffusion, and sets up eleven dispatching modes of single reservoir and conducts simulation according to the sudden water pollution accident occurred at different locations at the upper or lower reaches of a reservoir. The results show that: when a sudden water pollution accident occurred at the lower reach, if the pollutant concentration is the sensitive factor of targeted lower reach, a dispatching mode with large flow and short duration is more appropriate for a short-distance reservoir at the upper reach. However, if the reservoir is too far away from the targeted reach, it is difficult to control the pollutant concentration in the targeted reach with the reservoir dispatching mode. If the out-of-limit time of pollutants is a sensitive factor of the targeted reach, a dispatching model with moderate flow and short duration is suitable for a reservoir within a proper distance from the targeted reach.When a sudden pollution accident occurred within a reservoir, if the reservoir area is an important functional area, the discharge facilities of the reservoir should be turned on as soon as possible to reduce the concentration and retention time of the pollutant in the reservoir. However, if the targeted lower reach is an important functional section, it is necessary to delay the turning on of the reservoir and reduce the discharge flow as much as possible to reduce the impact on the targeted lower reach.

Cross section microstructure and fractography of a Auto high strength assembling bolt of 35 steel were observed by optical microscope（ OM） and electron scanning microscope（ SEM）. The hardness of the bolt was measured by HR-150A rockwell hardness tester. The fracture failure of the assembling bolt was analyzed. The results indicate that quenching and tempering treatment on the assembling bolt meet the national standard,but the strength was decreased by the quenching crack generated during quenching. The extension of quenching crack that due to the external force result in the fracture failure.

Background Neuropsychiatric systemic lupus erythematosus (NP-SLE) presents with a wide variety of clinical manifestations, which is often difficult to diagnose with a high mortality. This study aims to investigate the clinical features of NP-SLE involving the central nervous system (CNS) and the differential diagnoses between CNS NP-SLE and intracranial infections. Methods The clinical manifestations, serum immunological features, cerebrospinal fluid (CSF) examinations (including intracranial pressure, leukocyte count, protein, glucose and chloride), CT and (or) MRI and electroencephalogram (EEG) data of 23 NP-SLE patients with CNS involved were retrospectively reviewed. Results Nine patients presented with diffuse manifestations, while 14 patients presented with focal manifestations. Serum analysis showed the positive rates of immunoglobulins anti-nuclear antibody (ANA), anti-double stranded DNA antibody (dsDNA), anti-Sm, anti-ribosmal P protein, anti-SSA and anti-SSB antibodies were 21/22, 7/22, 1/14, 2/14, 9/14 and 3/14 respectively. Patients with decreased serum C3 accounted for 14/20 while patients with decreased serum C4 accounted for 5/20. Besides, patients with increased CSF leukocyte count and microalbumin took up 5/12 and 7/12, while patients with decreased glucose and chloride levels took up 5/12 and 6/12. All 23 patients presented abnormal CT and (or) MRI and 6 patients presented abnormal EEG. Conclusion Serum immunological levels, CT and (or) MRI and EEG examinations contributed to the diagnosis of NP-SLE involving CNS. Although CSF analyses were slightly abnormal, the increase of leukocyte count and average microalbumin was not obvious, and the mean values of glucose and chloride were in the normal range, suggesting that the CSF examinations were helpful for the differential diagnoses from intracranial infections. Glucocorticoids and immunosuppressive drugs were remarkably effective for CNS NP-SLE patients.

In order to reduce hardware cost of embedded wireless mobile communication system and improve developing efficiency of embedded network protocol, the embedded wireless mobile communication terminal based on 8-bit basic MCU was proposed. Furthermore, according to software engineering principle, the general frame was designed to gener- ate embedded network protocol by UML state machine technology. The frame converted UML state figure into imple- menting codes of embedded network protocol. The experiment result shows that embedded wireless mobile communica- tion system not only reduces hardware expense but also incarnates standard and efficiency for development of embedded system software by UML state machine technology.

The adsorption characteristics of functionalized polystyrene resin for acetaldehyde and furfural in Baijiu were evaluated in comparison with those of other sorbents including activated carbon, diatomite, and AB-8 macroporous resin. The adsorption capacity of the modified polystyrene resin was 60.19 and 47.37 mg/g for acetaldehyde and furfural at 240 min and 323 K, respectively, greatly exceeding that of the other sorbents. Adsorption experiments were conducted on three major commercial flavor types of Baijiu, revealing less than 1% change in the total ester content of Baijiu before and after adsorption, so selective adsorption could be achieved using this sorbent. Its structure was characterized through elemental analysis (EA), Fourier transform infrared spectroscopy (FTIR), Brunauer-Emmett-Teller (BET) surface area and pore structure analysis, thermogravimetric analysis (TGA), and scanning electron microscopy (SEM). The adsorption mechanism of aldehydes and furfural on the functionalized resin was investigated using adsorption kinetics, isotherms, and thermodynamics. The findings indicated that the adsorption process fitted well with the pseudo-second-order model and the Freundlich model. Based on the results of ΔG0 > 0, ΔH0 > 0, and ΔS > 0, the adsorption was a non-spontaneous endothermic process. As the adsorption process proceeded, the solution became less orderly, and the adsorption process was primarily governed by chemisorption. This research serves as a reference for the quality and health improvement of Baijiu.

Objective: To evaluate the red meat intake of pregnant women in Chengdu area and explore the effect of red meat intake in the first and second trimesters on gestational diabetes mellitus(GDM)., Methods: From February to July 2017, a prospective study was conducted among healthy singleton pregnant women within 8-14 weeks of gestation in a prenatal clinic of maternal-and-child medical institution in Chengdu City through purposive sampling. Data on maternal demographic characteristics was collected through questionnaire in early pregnancy. 3-day 24 hour dietary recall was used to collect dietary intake information in the first and second trimesters. The 75 g oral glucose tolerance test(OGTT)was conducted at 24 to 28 weeks of gestation, and GDM was diagnosed according to the Guidelines for the Diagnosis and Treatment of Pregnancy Diabetes in China(2014). Multivariate unconditional Logistic regression model was used to explore the association between tertiles of red meat intake and the risk of GDM in the first and second trimesters., Results: A total of 985 valid samples were followed up. The mean intake of total meat and red meat in middle pregnancy(102. 9 g/d and 74. 6 g/d) was higher than that in early pregnancy(70. 7 g/d and 52. 0 g/d). The difference was statistically significant(P&lt;0. 05). Red meat accounted for 73. 6% and 72. 5% of total meat intake in early and middle pregnancy, respectively. After adjusting the confounding factors, multivariate unconditional Logistic regression analysis showed the risk of GDM was 1. 499 times(95%CI 1. 028-2. 185) in the highest red meat intake(&gt;86 g/d)women compared with the lowest red meat intake(&lt;44 g/d) women in the second trimester. No association was found between red meat intake in the first trimester and the occurrence of GDM(OR=1. 029, 95%CI 0. 716-1. 481)., Conclusion: Pregnant women in Chengdu area have excessive intake of red meat in the second trimester. Higher intake of red meat in the second trimester(&gt;86 g/d) may increase the risk of gestational diabetes.

Objective: To study dietary patterns during the second trimester of pregnancy and to investigate the relationship between dietary patterns and gestational weight gain (GWG)., Methods: A prospective cohort study was conducted to select healthy singleton pregnant women at 8-14 weeks of gestation in a maternal and child health care institution in Chengdu city. Food items and quantities were collected at 8-14, 24-28, 32-36 weeks of gestation by using the 3-day 24-hour dietary recall and energy intakes were calculated. Dietary patterns during the second trimester were established by factor analysis and factor scores were calculated. The weight of pregnant women was measured at 8-14, 24-28 weeks of gestation and 1 week before delivery, and the total GWG and the GWG rates in the second and third trimesters were calculated. Multiple linear regression analyses were used to analyze the association between dietary patterns and GWG., Results: A total of 1 004 samples were included. Three dietary patterns were identified: Milk-egg-whole grain pattern, Beverage-dessert pattern and Traditional pattern. The average total GWG was (13.2±4.5) kg. The average weight gain rate was (0.4±0.2) kg/week in the second trimester. The average weight gain rate was (0.5±0.3) kg/week in the third trimester. After adjusting for confounding factors including maternal age, body mass index before pregnancy, dietary energy intake, physical activity, multiple linear regression analysis showed that the factor score of Beverage-dessert pattern was positively associated with the total GWG and the weight gain rate in the third trimester ( β =0.370, 95% confidence interval ( CI ): (0.103, 0.636), P =0.007; β =0.014, 95% CI : (0.000, 0.027), P =0.049, respectively), and the factor score of Traditional pattern was negatively associated with the total GWG ( β =-0.285, 95% CI : (-0.555, -0.015), P =0.039). There was no association between the Milk-egg-whole grain pattern and GWG., Conclusion: Dietary patterns during the second trimester of pregnancy are associated with GWG. The Beverage-dessert pattern may increase the total GWG and weight gain rate in the third trimester. Objective: To investigate the dairy product intake during pregnancy in Southwest China and to explore its relationship with neonatal birth body mass., Methods: A prospective study was conducted to select healthy singleton pregnant women at 8-14 weeks of gestation in a maternal and fetal health care institution in Chengdu City. Dairy product consumption during the first, second, third trimester of pregnancy were collected by 24-hour dietary recalls at 8-14 weeks, 24-28 weeks and 32-36 weeks of pregnancy, respectively, and the total milk intake and milk consumption rate were calculated. According to the dietary guidelines for Chinese pregnant women (2016), the recommended amount of milk (300 g/d) was used as the standard to calculate the compliance rate. The respondents were divided into three groups: no dairy consumption group, insufficient dairy consumption group and suitable dairy consumption group. The gestational age at delivery and neonatal birth body mass were collected by the hospital information system. Logistic regression model was used to analyze the association between milk intake during pregnancy and neonatal birth body mass., Results: A total of 962 pregnant women were included. The average milk intake in the first, second and third trimester of pregnancy were 125.0 (0, 236.1) g/d, 208.3 (0, 284.7) g/d and 250.0 (150.0, 416.7) g/d, respectively, with the compliance rates of 12.6%, 33.2% and 48.4%, respectively. The average neonatal birth body mass was (3 225.0±399.8) g. The incidence of small for gestational age (SGA) and large for gestational age (LGA) was 8.3% and 3.9%, respectively. Compared with no dairy consumption group in the second trimester of pregnancy, the risk of SGA was lower in suitable dairy consumption group (odds ratio ( OR )=0.786, 95% confidence interval ( CI ): 0.385-0.976). Compared with no dairy consumption group in the third trimester of pregnancy, the risk of SGA was lower in insufficient dairy consumption group and suitable dairy consumption group ( OR =0.672, 95% CI : 0.477-0.821 and OR =0.497, 95% CI : 0.116-0.807, respectively). No association was observed between milk intake in the first trimester and neonatal birth body mass, and milk intake in the second and third trimester of pregnancy was not associated with the risk of LGA., Conclusion: Insufficient milk intake of pregnant women is a significant problem in southwest China and needs to be improved. Milk intake during pregnancy is associated with neonatal birth body mass, and increased milk intake in the second and third trimester of pregnancy may reduce the risk of SGA., (Copyright© by Editorial Board of Journal of Sichuan University (Medical Science Edition).)

Objective: To investigate the effects of anxiety and depression on gestational diabetes mellitus(GDM) in early pregnancy., Methods: A prospective study was conducted in 2017 to select 1426 single-child healthy pregnant women of 8-14 weeks from the Sichuan Provincial Hospital for Women and Children through a deliberate sampling method. The age was(28. 6±4. 0) years old. Basic information such as age, pre-pregnancy weight, parity, and gravidity of pregnant women was collected through questionnaire survey. The anxiety self-rating scale(SAS) and depression self-rating scale(SDS) were used to collect information of anxiety and depression in pregnant women, and their anxiety and depressive symptoms were evaluated according to the result of Chinese norm. At 24 to 28 weeks of gestation, the oral glucose tolerance test(OGTT) was conducted. GDM was diagnosed according to the Guidelines for the Diagnosis and Treatment of Pregnancy Diabetes in China(2014). Multivariate unconditional Logistic regression was adopted to analyze the effects of anxiety and depression on GDM in pregnant women., Results: The incidence of GDM in early pregnancy anxiety group and depression group was 41. 8% and 33. 6%, respectively. The incidence of anxiety, depression, and anxiety combined with depression were 7. 7%, 10. 5% and 4. 8%, respectively. The incidence of anxiety and depression in the group of pregnant women younger than 30 years old(9. 0% and 11. 7%) was higher than that in the group of pregnant women older than 30 years old(5. 3% and 8. 1%). The prevalence of anxiety and depression in the group of nulliparous women(8. 8% and 11. 9%) was higher than that in the group of multiparous women(5. 4% and 6. 4%). The difference was statistically significant(P&lt;0. 05). After adjusting the confounding factors such as age, pre-pregnancy body mass index, family history of diabetes, gravidity, parity, energy intake, conception, education, occupation, smoking and drinking, the result showed that the risk of GDM in anxious pregnant women was increased, compared with that in non-anxious pregnant women in early pregnancy(OR=1. 556, 95% CI 1. 014-2. 387). However, the association between early pregnancy depression and the occurrence of GDM was not found(P&gt;0. 05). Compared with the non-anxiety group in the early pregnancy, the risk of GDM in the anxiety group was increased both in pregnant women under 30 years old(OR=1. 654, 95% CI 1. 004-2. 726) and nulliparous women(OR=1. 633, 95% CI 1. 013-2. 634). No correlation between anxiety and risk of GDM was observed in pregnant women over 30 years old and multiparous women(P&gt;0. 05)., Conclusion: Anxiety in early pregnancy increases the risk of GDM. Pregnant women under 30 years old and and nulliparous women are at high risk of anxiety.

Objective: To explore the effect of iron supplement intake on gestational diabetes mellitus(GDM) in early and middle pregnancy., Methods: From February to April 2017, a prospective study was conducted among 807 early pregnant women in a prenatal clinic of a maternal and child medical institution in Chengdu City through purposive sampling. Data on maternal demographic characteristics was collected through questionnaire in early pregnancy. In early and middle pregnancy, the information of iron supplement intake were collected with questionnaire, 3-day 24 hour dietary recall method was used to assess maternal diet. According to the WHO recommendation, 60 mg/d iron supplementation during pregnancy was used as the dividing point, &lt;60 mg/d iron supplementation was used as the low level group, and ≥60 mg/d iron supplementation was used as the high level group. At the 24 th to 28 th pregnant week, the oral glucose tolerance test(OGTT) was conducted, and GDM was diagnosed according to the Guidelines for the Diagnosis and Treatment of Pregnancy Diabetes in China(2014). Multivariate unconditional Logistic regression model was used to analyze the effect of iron supplement intake on gestational diabetes mellitus(GDM) in early and middle pregnancy., Results: A total of 739 valid samples were followed up, the age was(28. 22±3. 75) years old. In early and middle pregnancy, the rate of taking iron supplementation was 5. 0% and 67. 9%, 3. 8% and 47. 1% of them iron supplement intake was more than 60 mg/d. After adjustmenting for body mass index, age, dietary iron, etc. Multivariate unconditional Logistic regression analysis showed that there was a positive correlation between the average intake of iron supplement and the occurrence of GDM in women during the second trimester of pregnancy(OR=1. 059, 95%CI 1. 016-1. 104). Compared with the lower iron supplement intake(&lt;60 mg/d) women in midpregnancy, the risk of GDM was 1. 406 times(95%CI 1. 019-1. 939)in the higher iron supplement intake(≥60 mg/d) women. No correlation was found between iron intake in early pregnancy and the occurrence of GDM., Conclusion: Iron supplement intake during pregnancy may increase the risk of GDM. Appropriate intake of iron supplement for pregnant women is worth discussing.

Objective: To explore the effects of total gestational weight gain( GWG)and gestational weight gain rate( GWGR) per trimester on the neonatal birth weight., Methods: A prospective cohort study was conducted among 549 healthy pregnant women at the( 12 ± 1) th pregnant week from Chengdu City, China who delivered a singleton. Basic information about age, pre-gestational weight and so on was collected by questionnaire, while the height and weight were measured at the first time. Weight at the( 28 ± 1) th and last week before delivery was measured respectively, while the total GWG and the different trimester GWGR were calculated and assessed by Institute of Medicine( IOM) criteria( 2009). The gestational week, neonatal weight and other information were collected after delivery. Multiple non-conditional Logistic regression models were used to test the effect of the total GWG/GWGR per trimester on neonatal birth weight., Results: The total GWG was( 16. 2 ± 4. 6) kg. The prevalence of abnormal total GWG was 59. 1%, the excessive GWG was 44. 3%. The prevalence of insufficient GWGR in the first trimester was 44. 3%. In the second, third, the second and third trimester, the prevalence of excessive GWGR were 63. 6%, 55. 7% and 65. 8%, respectively. After adjusting the age at delivery, pre-pregnancy body mass index, gestational week and other confounding factors, the result showed that compared with adequate GWG group, insufficient GWG group was increased risk of small for gestational age( SGA)( OR =2. 51, 95% CI 1. 08-5. 82), excessive total GWG group was increased risk of large for gestational age( LGA)( OR = 2. 54, 95% CI 1. 20-5. 36). Compared with adequate GWGR group in the second trimester, excessive GWGR group was decreased risk of SGA( OR = 0. 27, 95% CI 0. 13-0. 60). Compared with adequate GWGR group in the second and third trimester, excessive GWGR group was decreased risk of SGA( OR = 0. 28, 95%CI 0. 13-0. 59)., Conclusion: Abnormal total GWG among pregnant women in Chengdu City is a big problem that both excessive and insufficient GWG existed. Excessive total GWG is the independent risk factor for LGA. GWGR in the second and the second and third trimester are significantly associated with SGA.

Objective: To determine the level of moderate-to-vigorous physical activities (MVPA) and its relationship with gestational weight gains (GWG) in the second and the last trimesters of pregnancy., Methods: A prospective study was conducted in Chengdu on 362 healthy pregnant women at the 24-28 gestation weeks who delivered a singleton. Demographic data and pre-pregnancy body mass were collected using a questionnaire. Weight gains at the gestation weeks of 24-28 and 32-36 were measured for the first two trimesters and the last trimester of pregnancy. The Denmark self-reported physical activity scale was used for measuring the duration and intensity of physical activities. Multiple linear regression models were established to determine the relationship between MVPA and GWG., Results: The last trimester had lower average daily MVPA 〔(0.76±0.93) h〕 compared with the second trimester 〔(1.61±1.61) h, t =9.056, P <0.001〕. About 74.6% of the participants met the PA recommendations for the second trimester, compared with 60.5% for the last trimester ( χ 2 =16.387, P <0.001). The participants experienced an average GWG of (7.36±3.78) kg during the first two trimesters, and (5.80±2.57) kg during the last trimester, corresponding to a growth rate of (0.30±0.15) kg/week for the first two trimesters and (0.51±0.22) kg/week for the last trimester. Compared with the most inactive group, the participants with medium PA experienced less GWG 〔(5.34±2.91) kg vs.(6.26±2.54) kg, P <0.05〕 and a lower GWG rate 〔(0.48±0.26) kg/week vs.(0.56±0.20) kg/week, P <0.05〕 during the last trimester. Age, gestational week, ethnicity, pre-pregnant BMI, GDM, pre-pregnant smoking and employment were associated with GWG and the GWG rates during the first two trimesters and the third trimester ( P <0.05). Compared with the most inactive group, low 〔-0.358(-0.691--0.026)〕 and medium 〔-0.762(-1.486- -0.037)〕 PA were associated with lower GWG during the last trimester. Moderate PA was associated with a lower GWG rate 〔-0.071(-0.133--0.008)〕 after adjustment for gestational age, energy intake, pre-pregnancy BMI and other potential confounders., Conclusions: Insufficient physical activities are a serious problem in the pregnant women of Chengdu over the last two trimesters. Objective: To explore the association between gestational weight gain rate( GWGR) and pregnancy outcomes among pregnant women with normal pre-pregnancy body mass index( BMI)., Methods: In this prospective study, 389 healthy pregnant women at the( 12 ± 1) th with normal pre-pregnancy BMI from Chengdu City who delivered a singleton were included in 2013. The basic information such as age, pre-pregnant weight was obtained by questionnaire, height and weight was measured. Then the weight was measured at the( 28 ± 1) th and last week before delivery, respectively, the different trimester GWGR was calculated and classified by IOM criteria. Gestational age, gestational diabetes mellitus( GDM) and other pregnancy outcomes were collected by questionnaire. Multiple non-conditional Logistic regression models were used to test the association between GWGR and pregnancy outcomes., Results: The proportion of insufficient GWGR and excessive GWGR in first trimester was 50. 7% and 31. 8%, respectively, the proportion of excessive GWGR in the second and third trimester was68. 5% and 57. 8%, respectively. After adjusting the age, education, gravidity and family disease history and other confounding factors, multiple non-conditional logistic regression showed: compared with adequate GWGR in the third trimester, insufficient and excessive GWGR were associated with increased risk of cesarean delivery( OR = 2. 48, 95% CI 1. 18-5. 23 and OR = 1. 76, 95% CI 1. 02-3. 03, respectively). Compared with adequate GWGR in the first trimester, excessive GWGR were associated with increased risk of GDM( OR = 2. 55, 95% CI 1. 03-6. 28)., Conclusion: The abnormal of GWGR would increase the risk of cesarean delivery and GDM.

Objective: To examine the relation between energy intake and its sources and gestational weight gain( GWG)., Methods: In this prospective cohort study, 585 healthy women with singleton pregnancy were selected in 2013. The 24-hour recalls was used to collect food intake at the first, second and last trimesters, and energy intake and percentages of energy from macronutrient were calculated. Weight measuring were administered at( 12 ± 1) th, ( 28 ± 1) th weeks' gestation and last week before delivery, and the GWG rate was calculated. Multivariable linear regression was used to analyze therelationship between energy and GWG., Results: The mean energy intake for the first, second and last trimesters were 1718. 6 kcal/d, 2202. 9 kcal/d and 2313. 4 kcal/d, respectively, the average percentages of energy from carbohydrate were 62. 6%, 59. 5%and 59. 1%, respectively, the average percentages of energy from fat were 21. 8% 、23. 9% and 24. 3%, respectively, the average percentages of energy from protein were15. 6% 、16. 5% and 16. 7%, respectively. Multiple linear regression analysis showed that energy intake during pregnancy was positively correlated with GWG during pregnancy( P < 0. 05). The adjusted weight gain for each trimester was 37. 3 g/week( 95% CI 22. 8 to 51. 8, P < 0. 05), 16. 8 g/week( 95% CI 4. 8 to 28. 8, P < 0. 05), and 25. 8 g/week( 95% CI 10. 6 to 41. 1, P < 0. 05) higher in the highest( Q5) versus lowest( Q1)quintile of energy intake. The adjusted weight gain in last trimester was 19. 1 g/week higher( 95% CI 3. 1 to 35. 1, P < 0. 05) in the highest( Q5) versus lowest( Q1) quintile of percentages of energy from carbohydrate. The adjusted weight gain in last trimester was16. 2 g/week lower( 95% CI-3. 1 to-0. 9, P < 0. 05) in the highest( Q5) versus lowest( Q1) quintile of percentages of energy from fat., Conclusion: A positive correlation is found between dietary energy intake and GWG. Percentages of energy from carbohydrate and fat in the last trimester have a significant effect on GWG.

Objective: To investigate the regulatory effect of ATP?binding cassette transporter A1 (ABCA1) knockdown on inflammatory response induced by Pam3CSK4 in mouse mononuclear macrophage RAW264.7 cell line., Methods: A mouse mononuclear macrophage RAW264.7 cell line with stable ABCA1 knockdown was constructed and stimulated with Toll?like receptor 2 (TLR2) ligand Pam3CSK4, and the changes in the transcriptional levels of the proinflammatory and anti-inflammatory cytokines were analyzed in this cell model., Results: In RAW264.7 cells, ABCA1 knockdown significantly up-regulated Pam3CSK4 stimulation?induced expressions of IL?1β, TNF?α and IL?6 and also enhanced the expression of transcription factor cAMP?dependent transcription factor 3 (ATF3) without obviously affecting the expressions of the transcription factors ATF1, ATF2, ATF4 or ATF5., Conclusion: ABCA1 knockdown in macrophages may have both proinflammatory and anti?inflammatory effects. ABCA1 knockdown up?regulates the transcription of ATF3 possibly through a mechanism that is different from that for the other members of the ATF protein family.

Objective: To investigate the relationship between miR-144 and Toll-like receptor 2 (TLR2)., Methods: RT-qPCR was used to determine the expression of TLR2 and its downstream inflammatory cytokine TNF-α in rat macrophage cell line NR8383 transfected by a mimic miR-144 or miR-144 inhibitor. The fragments of 3'UTR region of rat TLR2 mRNA including wild or mutant miR-144 binding site obtained by PCR using rat liver cDNA were ligated to pmirGLO report gene vector digested with SacI and XbaI to construct the recombinant vectors of pmir-TLR2-3'UTR and pmir-mutant-TLR2-3'UTR. The miR-144 targeting TLR2 was further determined by dual luciferase reporter assay and miR-144 mimics., Results: TLR2 and TNF-α in NR8383 cells were decreased after transfection with 100 nmol/L mimic miR-144 for 24 h and increased after transfection with 100 nmol/L miR-144 inhibitor. PCR and double-enzyme digestion with SacI and XbaI confirmed successful insertion of the target fragments. Dual luciferase reporter assay suggested the binding of miR-144 to the 3'UTR of rat TLR2 mRNA., Conclusion: miR-144 negatively regulates the expression of TLR2 and its down-stream cytokine TNF-α by targeting TLR2 in NR8383 cells.

Objective: To prepare the polyclonal antibody of the extracellular domain (405T-573Q) of rat Toll-like receptor 2 (TLR2) and identify its activity., Methods: Firstly, the extracellular domain (507 bp) of rat TLR2 was amplified from rat hepatocytes by reverse transcription PCR, and the recombinant plasmid pET28a-TLR2ex was constructed. Secondly, the fusion protein TLR2ex-6×His was expressed in E.coli BL21(DE3) and purified by affinity chromatography. And then, the polyclonal antibody was prepared by immunizing the BALB/c mice with the purified fusion protein. Finally, the titer and specificity of the polyclonal antibody were analyzed by ELISA and Western blotting, respectively., Results: The recombinant plasmid pET28a-TLR2ex was successfully constructed, and the TLR2ex-6×His fusion protein was successfully expressed and purified. Western blotting demonstrated that the antibody from the TLR2ex-6×His-immunized BALB/c mice specifically and selectively bound with rat TLR2. ELISA also indicated that the titer of polyclonal antibody reached 1:76 800., Conclusion: The polyclonal antibody against the extracellular domain (405T-573Q) of rat TLR2 has been successfully prepared.

Objective: To observe and compare the effects of grain-bean package, dietary fiber (DF) extracted from grain-bean package, and DF from grain corn on the blood lipids and fatty acid synthase (FAS) activity in high-fat, high-cholesterol feeding induced dyslipidemia rats, and observe its effects on regulation of sterol regulatory element protein-1c (SREBP-1c) mRNA expression in rat liver., Methods: Consolidation 50 SD rats of clean grade feeding adaptation for one week, randomly assigned into normal control group, hyperlipidemia model group, grain-bean package group, grain-bean package DF group and grain corn group. Feed with corresponding diets for 8 weeks, and measure the total cholesterol (TC), triglyceridaemia (TG), high density lipoprotein cholesterol (HDL-C), fasting blood glucose (FBG), FAS, SREBP-1c mRNA of all groups., Result: Compared with control group, TC, TG, FBG levels of hyperlipidemia model group were significantly increased (P < 0.05). Compared with model group, TC, TG, FBG levels of grain-bean package group, grain-bean package DF group were significantly decreased, HDL-C levels significantly increased, and activity of FAS, regulation of SREBP-1c were significantly decreased (P < 0.05)., Conclusion: The Grain-bean package dietary fiber can improve blood lipids levels of dyslipidemia rats, and decrease FAS activity and SREBP-1c mRNA expression.

Objective: To study the effects of CD133 suppression by lentivirus-mediated RNA interference (RNAi) on the proliferation and chemosensitivity of CD133(+) cancer stem cells (CSCs) sorted from HepG2 cell line., Methods: CD133(+) and CD133- cells were sorted from HepG2 cell line by flow cytometry, and the expression of CD133 before and after cell sorting were detected. The stem cell property of sorted CD133(+) cells were validated by sphere-forming assay in vitro and xenograft experiments in vivo. Lentivirus-mediated short hairpin RNA (shRNA) targeting CD133 were transfected into CD133(+) cells, and CD133 mRNA and protein expressions of the transfected cells were detected by RT-PCR and Western blotting, respectively. Before and after the transfection, the proliferative ability of CD133(+) cells was evaluated by colony formation assay, and the cell growth inhibition rate and apoptosis following cisplatin exposure were detected using CCK-8 assay and flow cytometry., Results: The sorted CD133(+) cells showed a high purity of (88.74∓3.19)%, as compared with the purity of (3.36∓1.80)% before cell sorting. CD133(+) cells showed a high tumor sphere formation ability and tumorigenesis capacity compared with CD133- cells. CD133 shRNA transfection significantly inhibited CD133 mRNA and protein expressions in CD133(+) cells (P<0.01), resulting also in a significantly lowered cell proliferative ability (P<0.01) and an increased growth inhibition rate (P<0.01) and obviously increased cell apoptosis (P<0.05) after cisplatin exposure., Conclusion: Lentivirus-mediated RNAi for CD133 suppression inhibits the proliferation of CD133(+) liver cancer stem cells and increases their chemosensitivity to cisplatin.
