Objective Palaeomagnetic study of the early Palaeozoic North Qiangtang Terrane can quantitatively constrain its palaeogeographic location since the Palaeozoic, which serves as an important foundation and key constraint for studying of the palaeotectonic pattern of the Qinghai-Tibet Plateau, the evolution of the Phanerozoic Tethys and palaeogeographic reconstruction. Methods Prior to palaeomagnetic study, it is essential to investigate the magnetic properties of rocks to identify the types and assemblages of magnetic carriers present in rocks, which will facilitate the selection of demagnetization experimental protocols and discussions on the primary nature of remanent magnetization. This paper focuses on the magnetic properties of limestone and sandstone from the Late Silurian Longmu Co Upper Formation in the North Qiangtang Terrane, through analyses of isothermal remanence acquisition curves, magnetic susceptibility with temperature(χ-T) curves, triaxial thermal demagnetization experiments, low-temperature magnetic properties tests, scanning electron microscopy (SEM) and energy dispersive spectral (EDS) analysis. Results Results show that magnetite is the predominant magnetic carrier in the limestone samples of the Longmu Co Upper Fm, with small amounts of pyrrhotite. The magnetic assemblage in the sandstone samples is more complex, dominated by magnetite, possibly with other magnetic minerals such as pyrrhotite. Conclusion Moreover, it demonstrates that stable high-temperature remanent magnetization components can be isolated from the limestone samples in the Longmu Co Upper Formation, making them suitable for further palaeomagnetic researches.

In recent years, due to the influence of human activities and global warming, the coral reef ecosystem in the South China Sea (SCS) has degraded, and bleaching occurs. Utilizing the remote sensing method to monitor and understand the bleaching status of coral reefs is of great value to the protection and management of the ecological environment in the SCS. This study first obtains coral reef bleaching alert areas (BAAs) through multiperiod sea surface temperature data (SST) and selects the Antelope Reef in the Yongle Atoll of the Paracel Islands as the research area. Then, a new coral reef bleaching monitoring model is proposed, which uses water depth correction, coral reef classification, reflectance adjustment and threshold selection to carry out multitime series coral reef bleaching monitoring research with Landsat-8 remote sensing images from 2013 to 2018. The results show that the model can effectively obtain coral reef bleaching areas and provide a feasible approach for the monitoring of coral reef bleaching in the South China Sea.

[Introduction] In order to solve the problems of poor stable combustion, unstable hydrodynamics, and too low SCR inlet flue gas temperature during the deep peak shaving process of power station boilers, it is imperative to reform the boiler to stabilize combustion. [Method] In this paper, taking the flexibility transformation of a 660 MW thermal power unit boiler as an example, two effective stable combustion transformation schemes were proposed, and the effects of the combustion and pulverizing system transformation on the furnace and NOx emissions were analyzed. [Result] Both retrofit schemes can improve the ultra-low load and stable combustion characteristics of the boiler. The retrofit of the combustion and pulverizing system has little impact on the furnace and NOx emissions and is comparable in terms of technical reliability, construction difficulty, and workload. In contrast, scheme 1 is slightly better. [Conclusion] The two transformation schemes have been theoretically verified and feasible, and can provide technical reference for flexible transformation of the same type of units.

Background: The Ministries of Justice and Education have implemented adolescent substance abuse prevention programs for many years. Rates of substance abuse among high school students continue to rise in spite of such programs. Purpose: This study investigates adolescents' substance abuse-related knowledge and attitudes and self-confidence to resist substance use in order to identify predictive factors of poor adolescent resistance to substance use. Methods: A cross-sectional study survey gathered data on substance abuse-related knowledge and attitudes and self-confidence to resist substance use from 243 second-year senior high school students studying at two schools in northern Taiwan. Results: Participants were most knowledgeable about tobacco (80.2%), followed by alcohol (72.0%), ecstasy (56.0%), and marijuana (30.0%). Only 19.3% demonstrated an understanding of the harmful effects of using Ketamine. A 10-point Likert scale measured participant substance use attitudes. Alcohol was the substance participants were most willing to use (2.18 ± 3.27), followed by tobacco (0.66 ± 2.19), ecstasy (0.45 ± 1.88), Ketamine (0.43 ± 1.93), and marijuana (0.38 ± 1.83). Participants had higher awareness of the harmful effects of Ketamine (t = -2.37, p = .018), marijuana (t = -2.33, p = .021), and tobacco (t = -2.02, p = .044), with participants reporting greater self-confidence to resist using these three substances. Multiple regression analysis found the three most important factors affecting participant self-confidence to resist substance use to be gender (ß = .26, p < .001), knowledge about the substance (ß = .15, p = .028), and attitude toward substance use (ß = -.20, p < .001). Conclusions / Implications for Practice: Level of harmful effects knowledge and attitudes toward use varied among the various substances considered in this paper. Being female, having strong knowledge about the substance, and negative attitude toward substance use correlated with higher levels of self-confidence to resist substance use. Study results will be used in ongoing research designed to establish an empirical basis for adolescent substance use prevention and research, the results of which may be used directly by school nurses and military training education nurses. PURPOSE: To evaluate the bond strength of 3 resin cements on ceramic-dentin. METHODS: Thirty sound human posterior teeth were selected for this study and the flat dentin surfaces were prepared. The specimens were divided into 3 groups (n=10) randomly, which were bonded to the CAD/CAM ceramics with RelyX ARC (etch-and-rinse adhesive), Clearfil DC Bond (self-etching resin cement) and RelyX Unicem (self-adhesive resin cement). The bond strengths were tested and the bond surfaces were observed under stereoscopic microscope. SPSS 11.0 software package was used for statistical analysis. RESULTS: The shear bond strength of RelyX ARC and Clearfil DC Bond was (15.90±6.15)MPa and (14.41±5.07)MPa, respectively. The shear strength of RelyX Unicem was (23.29±7.39)MPa, significantly higher than the others. The shear strength between the three resin cements was significantly different (P<0.05). CONCLUSION: Clearfil DC Bond and RelyX Unicem can achieve a successful bond on CAD/CAM ceramic crowns. Supported by Research Fund of Shanghai Municipal Bureau of Health (2009074). Obesity is often caused by an unhealthy lifestyle, which is a composite of various individual behaviors. Nurses may assist obese patients to lose weight and avoid chronic disease by identifying risky lifestyle behaviors and helping to develop improvement strategies. This article describes the nursing experience of the authors in caring for an obese patient who had made several unsuccessful attempts to reduce weight. An intervention approach was used to review the patient's lifestyle. Using self-regulation theory, the authors identified that the patient's fat-related daily behavior included: lack of exercise, high-fat diet, and daily snacks consumed even late at night. The authors also helped the patient discover the reasons underlying his fat- related behavior and his previous failed attempts to lose weight and to develop a feasible improved approach that considered such. After six weeks of care, the patient's body weight had reduced and body fat and body mass index had decreased with no relapse. The patient further lost significant weight and body fat during the three-month follow up period. The authors would like to share with nursing professionals this approach to weight loss, with the hope that this case study can contribute to medical efforts to help obese patients not only lose weight but also prevent chronic illnesses. A vertical transect investigation on Picea schrenkiana var. tianschanica forests was conducted at five different longitudinal sites (Zhaosu, Gongliu, Wusu, Urumqi, and Hami) in Tianshan Mountains, and the distribution pattern of P. schrenkiana var. tianschanica population at each site was analyzed based on theoretical distribution model and aggregation intensity index. On the whole, the P. schrenkiana var. tianschanica population in Tianshan Mountains presented a clumped distribution, and the distribution pattern and clustering intensity were affected by the developmental stages of stem and the ranges of altitude to some degree. Background Health literacy is closely associated with health status, most domestic studies tend to use multidimensional and multi-item tools for assessing health literacy, lacking simple and effective assessment methods. Objective To simplify health literacy scale and conduct psychometric test within the Chinese population. Methods Adults aged 18 years and above were selected from the "China Family Health Index Survey (2021) " for this investigation. Based on inclusion and exclusion criteria, a total of 7 449 participants were selected and randomly divided into two sample sets, including 3 680 cases in sample set 1 and 3 769 cases in sample set 2. The general information questionnaire, Short-form Health Literacy Questionnaire (HLS-SF12), Perception Social Support Scale (PSSS), and Family Health Scale Short-form (FHS-SF) were administered to the respondents. Classical test theory (CTT) and the Mokken model in item response theory (IRT) were used to screen the original items, and validation analyses such as reliability and validity of the simplified scales were also conducted. Results A 9-item version (HLS-SF9) and a 4-item version (HLS-SF4) were simplified by using CTT and the Mokken model, respectively. Both the HLS-SF9 and the HLS-SF4 had no ceiling effect or floor effect, and the Cronbach's α coefficients for both were 0.913 and 0.842, with split-half reliabilities of 0.871 and 0.815, respectively. The exploratory factor analysis of HLS-SF4 revealed one common factor, accounting for 67.813% of the cumulative variance, with factor loadings exceeding 0.81 for each item. The confirmatory factor analysis of HLS-SF9 showed that χ2/df was 10.844, goodness of fit index (GFI) was 0.985, adjusted goodness of fit index (AGFI) was 0.971, normative fit index (NFI) was 0.986, comparative fit index (CFI) was 0.987, and root mean squared error of approximation (RMSEA) was 0.051. In order to reveal the differences in the biosynthesis and metabolism of the major bioactive composition in sea buckthorn berry pulp during ripening, non-targeted metabolomics based on ultra-high performance liquid chromatography coupled to mass spectrometry (UPLC-MS) was used to analyze the metabolites of sea buckthorn berry pulp from two strains at different developmental stages (green and orange pulp). The results showed that the four sample groups had significant metabolic differences. A total of 124 significantly differential metabolites were detected among the four groups based on variable importance in the projection (VIP) > 1 and t-test P < 0.05, including organic acids, carbohydrates, lipids, flavonoids and secondary metabolites. According to Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis, 15, 7 and 4 significantly differential metabolites associated with lipid, flavonoid and serotonin metabolism were identified, respectively, and eight significantly differential metabolites involved in anticancer function were selected. The significant up-regulation of phosphatidylcholine (PC) (16:0/16:0) and glycerophosphocholine could be the reason for the high accumulation of lipids in sea buckthorn berry pulp. The significant upregulation of isorhamnetin was key for the high flavonoid content in sea buckthorn berry pulp. Relatively high contents of N-acetylserotonin, capecitabine, γ-aminobutyric acid (GABA) and other bioactive composition involved in anticancer function were shown in sea buckthorn berry pulp during its development. These results provide a scientific basis for revealing the metabolic mechanism of bioactive composition of sea buckthorn berries and developing related functional products.

Objective To investigate the effect of recombinant Treponema pallidum protein Tp47 (rTpp47) on the expression of uPA by THP-1 macrophages and on the permeability of human umbilical vein/vascular endothelial cells(HUVECs). Methods THP-1 cells were stimulated with rTpp47 for 24 hours and then the culture supernatant and THP-1 cells were collected respectively. The expression of urokinase-type plasminogen activator (uPA) in THP-1 cells was detected by ELISA and Western blot. After stimulation of HUVECs monolayers by THP-1 cell culture supernatant, FITC-dextran was used to evaluate the permeability of HUVECs. Western blot method was used to observe the effect of uPA on the expression of tight junction protein claudin-5 of HUVECs and the protein kinase C(PKC) signaling pathway of THP-1 cells stimulated by rTpp47. Results The synthesis and secretion of uPA of THP-1 cells stimulated by rTpp47 were significantly higher than those in the control group(P＜0.05, P＜0.001). When HUVECs were stimulated by the THP-1 cell supernatant for 12 hours and 24 hours, the relative permeability of vascular endothelial cells in the experimental group was significantly higher than the control group (P＜0.05 for 12 hours and P＜0.000 1 for 24 hours). Amiloride, the synthetic inhibitor of uPA, significantly inhibited the uPA expression and secretion of THP-1 cells stimulated by rTpp47 (P＜0.0001), alleviated the inhibition effect of rTpp47 on claudin-5 expression of HUVECs (P＜0.05), and significantly suppressed PKC phosphorylation(P＜0.05) in THP-1 cells induced by rTpp47. Conclusions The rTpp47 can stimulate the synthesis and secretion of uPA in THP-1 cells through a potential mechanism of activating PKC signaling pathway and uPA may increase the permeability of vascular endothelial cells.

Pavement cracks constitute a major potential threat to driving safety.Previous manual detection methods are highly subjective and inefficient.Current computer vision methods have limited applications in crack detection.Existing models have poor generalization capabilities and limited detection effects.To address this problem,a dense network structure of pavement crack detection,called Crack U-Net,is proposed to improve the model generalization capabilities and detection accuracy.Firstly,the dense connection structure of Crack U-Net adopts the network design from the encoder-decoder backbone network U-Net.Similar to the encoder-decoder backbone network,this structure of Crack U-Net is able to improve the utilization of feature information and to enhance the robustness of the model,as well.Secondly,the Crack U-block composed of residual blocks and mini-U is proposed as the basic convolution module of the network,which can extract more abundant crack features compared with the traditional dou-ble-layer convolution layer.Finally,dilated convolution is used in the middle layer of up sampling and down sampling in the network to fully capture the crack features,which is at the edge if the image.Crack U-Net runs on public fracture dataset and produces a series of experimental results.The experimental results show that the AIU value of this method on the dataset is 2.2% higher than the previous method,and it is better than the existing fracture segmentation accuracy and generalization.The experimental results also show that Crack U-Net model can be pruned,and the pruned model is suitable for loading to mobile devices for road crack detection.

This study aimed to identify the direct pharmacological targets of Jingfang Granules in treating infectious pneumonia via &quot;target fishing&quot; strategy. Moreover, the molecular mechanism of Jingfang Granules in treating infectious pneumonia was also investigated based on target-related pharmacological signaling pathways. First, the Jingfang Granules extract-bound magnetic nanoparticles were prepared, which were incubated with lipopolysaccharide(LPS)-induced mouse pneumonia tissue lysates. The captured proteins were analyzed by high-resolution mass spectrometry(HRMS), and the target groups with specific binding to the Jingfang Granules extract were screened out. Kyoto Encyclopedia of Genes and Genomes(KEGG) enrichment analysis was used to identify the target protein-associated signaling pathways. On this basis, the LPS-induced mouse model of infectious pneumonia was established. The possible biological functions of target proteins were verified by hematoxylin-eosin(HE) staining and immunohistochemical assay. A total of 186 Jingfang Granules-specific binding proteins were identified from lung tissues. KEGG pathway enrichment analysis showed that the target protein-associated signaling pathways mainly included Salmonella infection, vascular and pulmonary epithelial adherens junction, ribosomal viral replication, viral endocytosis, and fatty acid degradation. The target functions of Jingfang Granules were related to pulmonary inflammation and immunity, pulmonary energy metabolism, pulmonary microcirculation, and viral infection. Based on the in vivo inflammation model, Jingfang Granules significantly improved the alveolar structure of the LPS-induced mouse model of infectious pneumonia and down-regulated the expressions of tumor necrosis factor-α(TNF-α) and interleukin-6(IL-6). Meanwhile, Jingfang Gra-nules significantly up-regulated the expressions of key proteins of mitochondrial function COX Ⅳ and ATP, microcirculation-related proteins CD31 and Occludin, and proteins associated with viral infection DDX21 and DDX3. These results suggest that Jingfang Gra-nules can inhibit lung inflammation, improve lung energy metabolism and pulmonary microcirculation, resist virus infection, thus playing a protective role in the lung. This study systematically explains the molecular mechanism of Jingfang Granules in the treatment of respiratory inflammation from the perspective of target-signaling pathway-pharmacological efficacy, thereby providing key information for clinical rational use of Jingfang Granules and expanding potential pharmacological application.

This study identified the anti-depression targets of Kaixin San(KXS) in the brain tissue with &quot;target fishing&quot; strategy, and explored the target-associated pharmacological signaling pathways to reveal the anti-depression molecular mechanism of KXS. The Balb/c mouse model of depression was established by chronic unpredictable mild stress(CUMS) and the anti-depression effect of KXS was evaluated by forced swimming test and sucrose preference test. KXS active components were bonded to the benzophenone-modified magnetic nanoparticles by photocrosslinking reaction for capturing target proteins from cortex, thalamus and hippocampus of depressive mice. The target proteins were identified by liquid chromatography-mass spectrometry/mass spectrometry(LC-MS/MS). The enrichment analysis on signaling pathways was performed by Cytoscape. The potential biological functions of targets were verified by immunohistochemistry and Western blot assay. The results showed that KXS significantly improved the behavioral indexes. There were 64, 91, and 44 potential targets of KXS identified in cortex, thalamus, and hippocampus, respectively, according to the target identification experiment. The functions of these targets were mainly associated with vasopressin-regulated water reabsorption, salmonella infection, thyroid hormone synthesis, and other signaling pathways. Besides, the results of immunohistochemistry and Western blot showed that KXS up-regulated the expressions of argipressine(AVP) in the cortex, heat shock protein 60(HSP60), cytochrome C oxidase 4(COX4), and thyrotropin-releasing hormone(TRH) in the thalamus, and down-regulated the expressions of tumor necrosis factor-α(TNF-α) and nuclear factor kappa B(NF-κB) p65 in the thalamus. Therefore, KXS may exert anti-depression effect through regulating vasopressin signaling pathway in the cortex and inflammation, energy metabolism, and thyroid hormone signaling pathways in the thalamus, and the effect of KXS on hippocampus is not significant.

This study aims to identify the anti-pneumonia targets of Xiaoer Xiaoji Zhike Oral Liquid(XXZL) with &quot;target fishing&quot; strategy and investigate the related signaling pathways, thereby clarifying the anti-pneumonia mechanism of XXZL. To be specific, the magnetic nanoparticles cross-linked with XXZL extract were prepared based on the photochemical activity of benzophenone, which were then used to capture the target proteins from the lysate of tissue with lipopolysaccharide(LPS)-induced pneumonia in mice. Then, the target proteins were identified by liquid chromatography-tandem mass spectrometry(LC-MS/MS). The signaling pathways and interactions of target proteins were explored with KEGG and STRING analysis on Cytoscape, and the possible biological functions of the target proteins were verified by immunohistochemistry(IHC) and RT-PCR. The result showed that LC-MS/MS identified 62 potential anti-pneumonia targets of XXZL in the lungs. The targets were involved in Ras signaling pathway, mitophagy, leukocyte transendothelial migration, mitogen-activated protein kinase(MAPK) signaling pathway, platelet activation, and actomyosin structure organization, which were closely related to inflammation, pulmonary microcirculation, pulmonary fibrosis, and energy metabolism. XXZL up-regulated the content of CD31, and heat shock protein 60(HSP60) and ATP5 b mRNA expression, down-regulated interleukin-6(IL-6), tumor necrosis factor-α(TNF-α), COL1 A1 content, and alleviated fibrosis, which suggested the obvious effects of XXZL such as anti-inflammation, pulmonary microcirculation improvement, pulmonary fibrosis inhibition, and energy metabolism regulation. This study explained the anti-pneumonia mechanism of XXZL from targets, which can serve as a reference for the clinical application of the prescription.

&quot;Target fishing&quot; strategy was used to investigate the direct targets and mechanism of Shouhui Tongbian Capsules on relaxing bowel. Magnetic beads cross-linked with the chemical constituents from Shouhui Tongbian Capsules were prepared. The potential target proteins were captured from the total protein lysates of rat intestine using the beads. The captured proteins were further identified by LC-MS/MS, and the associated pathways were analyzed by Cytoscape. RESULTS:: showed that 138 potential target proteins were identified, which were involved in eight signaling pathways, including tricarboxylic acid cycle, pyrimidine metabolism, sulfur metabolism, fatty acid degradation, alanine/aspartate/glutamate metabolism, arginine/proline metabolism, valine/leucine/isoleucine degradation, and β-alanine metabolism. Taken together, Shouhui Tongbian Capsules may exert relaxing bowel effect by acting on multiple signaling pathways to promote intestinal gurgling, inhibit inflammation, as well as improve intestinal barrier function, intestinal water secretion, and intestinal flora.

Mechanism study was performed to explore how Shouhui Tongbian Capsules promotes energy metabolism of gastrointestinal stromal cells. In this study, gastrointestinal stromal cells line GIST-882 was used as the model to explore energy metabolism regulation effects of Shouhui Tongbian Capsules extract(10, 20, 50 and 100 μg·mL~(-1)) by measuring the cell proliferation, ATP level, mitochondrial membrane potential, and mitochondrial isocitrate dehydrogenase activity. Meanwhile, Western blot was used to detect the proteins expression of SCF/c-Kit and CDK2/cyclin A signaling pathways. Our results showed that Shouhui Tongbian Capsules promoted cell proliferation and increased ATP level of gastrointestinal stromal cells. In addition, Shouhui Tongbian Capsules obviously improved mitochondrial structural integrity, and increased mitochondrial membrane potential in GIST-882 cells. Mechanism study revealed that Shouhui Tongbian Capsules increased mitochondrial isocitrate dehydrogenase activity and up-regulated the proteins expression of SCF/c-Kit and CDK2/cyclin A signaling pathways. Collectively, our study indicated that Shouhui Tongbian Capsules promoted the energy metabolism for gastrointestinal stromal cells proliferation by activating mitochondrial isocitrate dehydrogenase to induce ATP production, as well as activating SCF/c-Kit and CDK2/cyclin A signaling pathways.

Shouhui Tongbian Capsules was used to explore the therapeutic effect and potential mechanism on slow transit constipation model mice induced by loperamide hydrochloride. In the experiment, loperamide hydrochloride-induced ICR mice were used as the model of slow transit constipation. Fifty ICR mice were divided into the blank group, model group and high, medium and low dose groups of Shouhui Tongbian Capsules extract(100, 200 and 400 mg·kg~(-1)). The model group and the administration groups were then modeled using loperamide hydrochloride intragastrically to obtain slow transit constipation. After successful modeling, high, medium and low doses of drugs were given to each drug group by intragastric administration. After 14 days of administration, the first defecation time, 6 h defecation grain number, 6 h defecation wet weight and dry weight, black feces discharged within 6 h and the fecal water content were measured. Intestinal tissues were taken for c-Kit and SCF immunohistochemical sections to detect the expression of c-Kit and SCF in the blank group, model group and high, medium and low dose groups of the medicinal extract of Shouhui Tongbian Capsules. The tissue changes in the intestinal wall of mice were detected by HE staining. At the same time, partial intestinal tissues were taken to test the activity of ATP synthase and isocitrate dehydrogenase in intestinal tissues of mice. RESULTS:: showed that Shouhui Tongbian Capsules effectively improved the symptoms of slow transit constipation in ICR mice and promoted intestinal movement. Shouhui Tongbian Capsules obviously shortened the time of discharging black stool for the first time, improved the intestinal propulsion rate, increased the water content and amount of feces, and improved the constipation symptoms. Mechanism study revealed that Shouhui Tongbian Capsules increased ATP synthase activity and mitochondrial isocitrate dehydrogenase activity in intestinal tissue, and up-regulated c-Kit/SCF signaling pathway to promote interstitial Cajal cells proliferation, intestinal nerve transmission, intestinal motility and transport capacity.

To investigate the inhibitory effects and mechanism of Cistanche tubulosa ethanol extract( CTEE) against oxygen-glucose deprivation/reperfusion( OGD/R)-induced PC12 cells neuronal injury. In this study,OGD/R-induced PC12 cells were used to explore the neuroprotective effects of CTEE( 12. 5,25,50 mg·L-1) by detecting cell viability with MTT assay,apoptosis with AO/EB and Hoechst 33258,mitochondrial membrane potential changes with JC-1 staining,mitochondrial oxidative stress with MitoSOX staining,as well as the apoptosis-related protein expression( PARP,cleaved PARP,caspase-3,cleaved caspase-3,Bax,Bcl-2) with Western blot. RESULTS:: showed that CTEE effectively protected OGD/R-induced neuronal injury and increased the survival rate of PC12 cells.AO/EB and Hoechst 33258 staining showed that CTEE could effectively inhibit apoptosis. Moreover,JC-1 and MitoSOX staining results showed that CTEE decreased mitochondrial stress and mitochondrial membrane potential imbalance in PC12 cells in a concentration-dependent manner. Meanwhile,CTEE could obviously suppress the activation of key proteins in mitochondrial apoptosis pathway such as caspase-3 and PARP,and significantly inhibit the rise of Bax and down-regulation of Bcl-2. In conclusion,CTEE has obvious protective effects on OGD/R-induced PC12 cells neuronal injury,potentially via inhibiting mitochondrial oxidative stress and apoptosis-related signaling pathway.

Target identification is an important prerequisite for the study of medicine action mechanism. Currently,drug target identification is mostly based on various cell models in vitro. However,the growth microenvironment,nutrition metabolism,biological properties as well as functions are quite different between in vitro cell culture and physiological environment in vivo; wherefore,it is a challenging scientific issue to establish an effective method for identifying drug targets in vivo condition. In this study,we successfully prepared a kind of magnetic nanoparticles( MNPs) which can be chemically modified by the hydroxyl structure of natural bioactive compound echinacoside( ECH) via the epoxy group label on the surface of MNPs. Therefore,organ-selective and recoverable nanoscale target-recognizing particles were prepared. We then intravenously injected the ECH-binding MNPs into rats and distributed them to specific organs in vivo. After cell endocytosis,ECH-binding MNPs captured target proteins in situ for further analysis. Based on this method,we discovered several potential target proteins in the spleen lysates for ECH,and preliminarily clarified the immuno-regulation mechanism of ECH. Collectively,our strategy developed a proof-of-concept technology using nanoparticles for in vivo target identification,and also provided a feasible approach for drug target prediction and pharmacological mechanism exploration.

Traditional Chinese medicine(TCM) is a research area with highly original innovation features,and is also a Chinese name card to the world. However,TCM owns a unique theoretical system which is quite different from western modern medicine,leading to an awkward situation of deficient modern social identity as well as poor international spread. Therefore,how to establish a research strategy in line with the characteristics of TCM itself to systematically interpret the unique scientific connotation of TCM is always a public hot topic. Based on persistent practical exploration and scientific consideration in TCM,our group firstly promoted the concept of traditional Chinese medicine chemical biology(TCM chemical biology,TCMCB). The major idea of TCMCB is to clarify the nature of TCM regulating life progress to link TCM to modern medicine by using TCM components as chemical tools. Notably,TCMCB mainly focuses on TCM target identification and TCM-guided disease molecular mechanism exploration,further to clarify the basic law of TCM mediating disease process. Finally,TCMCB-guided scientific studies can help explain TCM theory and promote the developmentof modern innovative drugs based on identified targets using TCM active components. Moreover,TCMCB is of vital importance for investigating the scientific nature of biological progress and the pattern of disease occurrence and development,indicating a key significance for modern life science and medicine. This review introduces the definition of TCMCB as well as its academic thought,research method,technology system and scientific significance,for providing new research ideas and scientific thoughts for TCM development.

To investigate the immune activation effect and mechanism of low molecular weight saccharides from Cistanche deserticola(LMSC) on mouse peritoneal macrophages, RAW264.7 cells. The RAW264.7 cells were divided into the normal control group, LPS positive control group, and LMSC treatment groups. The RAW264.7 cells were treated with various concentrations of LMSC from 3.91 to 62.5 g•L ⁻¹. The neutral red assay was employed to detect the phagocytic activity of macrophages. NO release was detected by using NO kit, and macrophage activation associated protein expression levels (TNF-α, IL-6, IKKβ, p-IKKβ, IκBα, p-IκBα, NF-κB, and p-NF-κB) were detected by Western blot. Results showed that LMSC had an activation effect on macrophages; it can significantly increase the release of NO in RAW264.7 cells and promote the expression of cytokines TNF-α and IL-6. Moreover, LMSC significantly increased the phosphorylation of IKKβ, IκBα, and NF-κB p65. Furthermore, mannitol's one of the main constituents in LMSC significantly enhanced the phagocytic activity of macrophages. This study aims to investigate the targets and targets-involved mechanism for the macrophage activation of low molecular weight saccharides from Cistanche deserticola (LMSC). The phagocytic activity and NO release of RAW264.7 cells were detected, and results showed that LMSC exerts immune activation effect by significantly increasing the phagocytic activity and NO release. LMSC-conjugated epoxy-activated sepharose beads were prepared as affinity reagent to capture the target proteins. Twenty-four proteins such as Eef2 were identified by LC-MS/MS analysis. Targets group identification in complex Chinese medicine system is a key step for revealing the potential mechanism of Chinese medicine. The solid beads with magnetic core and benzophenone-modified surface were made in our study, and then benzophenone was activated and cross-linked with the C-H bonds of chemical compositions in Chinese medicines under UV excitation. Thus the chemical compositions of modified Wuzi Yanzong pill(MWP) were linked to the solid bead surface, and enriched the neuroprotective targets group of MWP after being co-incubated with nerve cell lysate. We performed proteomics analysis on these targets and discovereda total of 32 potential binding targets. KEGG analysis revealed that these targets were mainly associated with Hippo and Cell cycle signaling pathways, suggesting that MWP might be involved in regulating the proliferation and differentiation of neural stem cells. Our findings elucidate the potential targets and mechanism of MWP on anti-dementia and neuroprotection, and further providean approach for investigating the targets group in complex Chinese medicine system. Traditional Chinese medicine(TCM) is a complex system with multiple chemical compositions. The most significant character of TCM is that the chemical compositions interact with each other by multi-target synergism to treat diseases. Previous reports mainly focused on the investigation of single signaling pathway detection or the phenotypic analysis of proteomics difference; however, no studies have been conducted on the identification of direct targets of TCM. Therefore, it is difficult to analyze the molecular mechanism of traditional Chinese medicine from the target source, and it is difficult to explain its traditional efficacy scientifically, thus seriously affecting its clinical application and internationalization. In this article, we discussed the methodology for the identification of direct TCM targets(groups), and presented the strategy for preparation of TCM chemical composition solid coupling beads, as well as of enrichment and identification strategy of target proteins based on photosensitive coupling technique. We also discussed the advantages and limitations of this strategy, and put forward some new ideas for the future developments. Baoyuan decoction (BYD) is a well-known traditional Chinese medicine formula for coronary heart disease with Qi deficiency. However, the detailed pharmacological mechanism of BYD is still unknown because of its complicated chemical compositions. In this study, we synthesized a kind of solid beads with benzophenone groups on its surface. Benzophenone can be activated and chemically cross-linked with the C-H bonds of the chemical compositions in BYD (BYD beads) under UV activation. We thus captured all the target proteins from mouse heart tissue lysates by using BYD beads. Based on proteomics analysis, we discovered totally 46 potential binding target proteins, most of which were located in mitochondria. KEGG analysis revealed that these target proteins were mainly associated with TCA cycle and amino acid metabolism signaling pathways, suggesting that the cardioprotection of BYD might be associated with regulating mitochondrial function and energy production. Moreover, JC-1 staining analysis also confirmed the protective effect of BYD on mitochondrial damage. In summary, our findings elucidated the potential mechanism of BYD on cardioprotection through "target fishing" strategy, and further explained its traditional efficacy in the molecular level. In addition, we also provide an approach for investigating the target group of complicated compositions in Chinese herbal formula. Dried stem bark from Albizia julibrissin(AJ) is a common traditional Chinese herb with several therapy effects including insomnia, anxiety and anti-tumor. Recently, the anti-tumor effect and mechanism studies of AJ have drawn much attention; however, there are still some troubles in chemical composition separation, which leads to the difficulties in pharmacological research of AJ. In this study, we firstly confirmed the proliferation inhibitory effect of total saponins from AJ(TSAJ)on human hepatocarcinoma(HepG2) cells, and also tested the apoptosis induction effect of TSAJ. Then, we successfully captured the potential target proteins from HepG2 lysates by using TSAJ-modified solid beads, and identified the target proteins by LC-MS/MS. Finally, we confirmed 5 target proteins including Exportin-2, Beta-actin-like protein 2, Myosin-9, Protein transport protein Sec61 subunit beta,and Cytochrome c oxidase copper chaperone, which are responsible forcell apoptosis, proliferation, differentiation andmigration. Diabetes mellitus, a kind of chronic metabolic disease, has become one of major threats to human health with an increasing incidence. As a basic pathology of chronic complications related to diabetes mellitus, cerebral microangiopathy is mainly found in patients with Alzheimer's disease and lacunar infarction, and becomes one of the main reasons of death and disability in diabetic patients. The pathogenesis of cerebral microangiopathy is complicated, involving such signal pathways as metabolic abnormalities of polyol, saccharification hyperactivity, oxidative stress, abnormal transport of amyloid-β across blood-brain barrier and protein kinase C activation. Treatment targeting at related pathogenesis may bring a new hope to prevention and delay of the occurrence and development of cerebral microangiopathy of diabetes mellitus. To investigate the inhibitory effects of acteoside (ACT) on BV-2 microglial cells and the potential mechanism,LPS was used to treat BV-2 cells with or without ACT (12.5,25,50 μmol•L ⁻¹). Then, the expressions of inflammatory factors (NO,TNF-α,IL-6) and inflammation related proteins (iNOS,COX-2,p-IKKβ,IKKβ,p-ⅠκB,ⅠκB) were detected. In addition,the nuclear translocation of NF-κB was explored. The results showed that ACT could significantly suppress the inflammatory response against LPS stimulation by decreasing the expressions of NO,IL-6,TNF-α,iNOS,COX-2 and the phosphorylations of IKKβ and IκB. Moreover,the nuclear translocation of NF-κB p65 was inhibited by ACT. Hydrophidae, one of the precious traditional Chinese medicines, is generally drily preserved to prevent corruption, but it is hard to identify the species of Hydrophidae through the appearance because of the change due to the drying process. The identification through analysis on gene barcode, a new technique in species identification, can avoid this problem. The gene barcodes of the 5 species of Hydrophidae, Lapemis hardwickii, Hydrophis fasciatus, Aipysurus eydouxii, Hydrophis belcher and Hydrophis lamberti, were acquired through DNA extraction and gene sequencing. These barcodes were then in sequence alignment and test the identification efficiency by BLAST. Acousto-optic tunable filter (AOTF) has the advantages of small size, good stability, wide range of wavelength scanning, quick modulation speed and so on. So AOTF is widely used in spectral imaging. But the detection of spectral polarization imaging is seldom found by using AOTF individually., Anew method of spectral polarization imaging based on dual-AOTFs is proposed. This method shows that the incident light is firstly divided into two beams by splitting mirror, and then two beams of light are through two AOTFs respectively, and the polarization direction of +1 diffraction order formed by AOTF is at a 45° against the other. The intensity of 0°, 45°, 90°and 135° can be obtained by ±1 diffraction orders of dual-AOTFs. Finally, the Stokes parameters are obtained by ±1 diffraction orders imaging of dual-AOTFs. They are S0 (the sum of light intensity between 0° and 90°), S1(the difference of light intensity between 0° and 90°) and S2 (the difference of light intensity between 45° and 135°). The target degree of linear polarization (DoLP) and angle of linear polarization (AoLP) is realized by corresponding theoretical formula. Furthermore, the polarization imaging of different wavelengths is realized with frequency sweep of dual-AOTFs so as to ultimately detect the spectral polarization imaging. The experiment has been verified through polarization imaging of three different lights, such as λ=500 nm, λ=550 nm and λ=600 nm.

Baoyuan decoction (BYD) is a classical Chinese formula for coronary heart disease with Qi deficiency, blood stasis tonifying Qi and Yang deficiency. However, the chemical material basis and underlying action mechanisms of BYD still lack systemic study. In order to clarify the active compounds and the potential action mechanisms of BYD, the oxygen-glucose deprivation/recovery (OGD/R)-induced H9c2 cells injury models was used to screen the monomeric compounds of BYD with myocardial protection activity. PubChem's BioAssay database was then used to analyze the potential targets of active monomeric compounds, classify the predicted biological targets, and analyze the internal relation between active compounds of BYD and biological targets. The screening results showed that BYD aqueous extract and 17 monomeric compounds could significantly increase the survival rate of OGD/R-induced H9c2 myocardial cells. The results of virtual targets screening study showed that 15 monomeric compounds and the potential mechanisms for myocardial protection were related to oxidative stress pathway, calcium ion pathway, mitochondrial protection, anti-apoptosis, etc. These results verified that BYD had myocardial protection effect, which was obtained by network regulation of multi-components and multi-targets. Drug targets are special molecules that can interact with drugs and exert pharmacological functions in human body. The natural active small molecules are the bioactive basis of traditional Chinese medicine, and the mechanism study is a hot topic now, especially for the identification of their target proteins. However, little progress has been made in this field until now. Here, we summarized the recent technologies and methods for the identification of target proteins of natural bioactive small molecules, and introduced the main research methods, principles and successful cases in this field. We also explored the applicability and discussed the advantages and disadvantages among different methods. In view of the existing relatively cumbersome polarization measurement method of using liquid crystal phase variable retarder (LCVR) and acousto-optic tunable filter (AOTF), a new strategy of spectrum and polarization measurement is put forward. This method removed mechanical movement and reduced the amount of phase delay from four groups to two groups. This method realizes the spectrum and polarization measurements by measuring the plus and minus 1 level diffraction light of the system with two LCVR, one AOTF and two same version detectors. For the polarization measurement of a particular wavelength, the computer controlled LCVR driving voltage to achieve the required phase delay in accordance with the requirements of the wavelength, it also controlled the driving frequency to implement the required wavelength selection of AOTF. Because of the different driving frequencies of AOTF corresponding to the different wavelengths, so it can get the spectral information by scanning the whole frequency bands, In this paper detailed principles are deduced and the polarization model of AOTF is analyzed, it also deduced the measurement formula of I, Q, U in the corresponding Stokes parameter through the theoretical calculation Muller matrix of the LCVR and AOTF. Finally it analyzed and simulated the measurement error of the whole system which was caused by the tiny deviation of phase delay, the simulation results show that relative error <3% when the phase delay amount within the range of ±π/100. This experiment proved the feasibility and accuracy of the measurement system, the results show that the error of the measuring system overall <6%. This study provides a new method of polarization measurement which is simple, feasible and precise, it has important application value.

Background: The Ministries of Justice and Education have implemented adolescent substance abuse prevention programs for many years. Rates of substance abuse among high school students continue to rise in spite of such programs., Purpose: This study investigates adolescents' substance abuse-related knowledge and attitudes and self-confidence to resist substance use in order to identify predictive factors of poor adolescent resistance to substance use., Methods: A cross-sectional study survey gathered data on substance abuse-related knowledge and attitudes and self-confidence to resist substance use from 243 second-year senior high school students studying at two schools in northern Taiwan., Results: Participants were most knowledgeable about tobacco (80.2%), followed by alcohol (72.0%), ecstasy (56.0%), and marijuana (30.0%). Only 19.3% demonstrated an understanding of the harmful effects of using Ketamine. A 10-point Likert scale measured participant substance use attitudes. Alcohol was the substance participants were most willing to use (2.18 ± 3.27), followed by tobacco (0.66 ± 2.19), ecstasy (0.45 ± 1.88), Ketamine (0.43 ± 1.93), and marijuana (0.38 ± 1.83). Participants had higher awareness of the harmful effects of Ketamine (t = -2.37, p = .018), marijuana (t = -2.33, p = .021), and tobacco (t = -2.02, p = .044), with participants reporting greater self-confidence to resist using these three substances. Multiple regression analysis found the three most important factors affecting participant self-confidence to resist substance use to be gender (β= .26, p < .001), knowledge about the substance (β= .15, p = .028), and attitude toward substance use (β= -.20, p < .001)., Conclusions / Implications for Practice: Level of harmful effects knowledge and attitudes toward use varied among the various substances considered in this paper. Being female, having strong knowledge about the substance, and negative attitude toward substance use correlated with higher levels of self-confidence to resist substance use. Study results will be used in ongoing research designed to establish an empirical basis for adolescent substance use prevention and research, the results of which may be used directly by school nurses and military training education nurses.

Objective: To summary the efficacy and safety of aerosolized iloprost in patients with pulmonary hypertensive crisis., Methods: On the basis of conventional therapy, aerosolized iloprost (10 µg per time for 10 - 15 min in 2 hours interval, 8 times per day) was administered to four patients with idiopathic pulmonary arterial hypertension and pulmonary hypertensive crisis. Blood pressure, heart rate, systemic artery oxygen saturation, systolic pulmonary arterial pressure (sPAP) measured by echocardiography and the adverse events were analyzed., Results: After aerosolized iloprost therapy, sPAP was significantly decreased and systemic artery oxygen saturation was improved. Adverse events (nausea, vomiting, diarrhea, dry cough) were observed in two patients, and the iloprost use was stopped in one patient due to severe vomiting and diarrhea., Conclusion: Aerosolized iloprost could significantly reduce the sPAP and improve the systemic artery oxygen saturation in patients with pulmonary hypertension crisis.

Purpose: To evaluate the bond strength of 3 resin cements on ceramic-dentin., Methods: Thirty sound human posterior tooth were selected for this study and the flat dentin surfaces were prepared. The specimens were divided into 3 groups(n=10) randomly, which were bonded to the CAD/CAM ceramics with RelyX ARC(etch-and-rinse adhesive), Clearfil DC Bond(self-etching resin cement) and RelyX Unicem(self-adhesive resin cement). The bond strengths were tested and the bond surfaces were observed under stereoscopic microscope.SPSS 11.0 software package was used for statistical analysis., Results: The shear bond strength of RelyX ARC and Clearfil DC Bond was (15.90±6.15)MPa and (14.41±5.07)MPa, respectively. The shear strength of RelyX Unicem was(23.29±7.39)MPa, significantly higher than the others.The shear strength between the three resin cements was significantly different (P<0.05)., Conclusion: Clearfil DC Bond and RelyX Unicem can achieve a successful bond on CAD/CAM ceramic crowns.

A vertical transect investigation on Picea schrenkiana var. tianschanica forests was conducted at five different longitudinal sites (Zhaosu, Gongliu, Wusu, Urumqi, and Hami) in Tianshan Mountains, and the distribution pattern of P. schrenkiana var. tianschanica population at each site was analyzed based on theoretical distribution model and aggregation intensity index. On the whole, the P. schrenkiana var. tianschanica population in Tianshan Mountains presented a clumped distribution, and the distribution pattern and clustering intensity were affected by the developmental stages of stem and the ranges of altitude to some degree. The clustering intensity increased with the increasing size (DBH) or developmental stage of stem, and had the highest values at high altitudes.

Objective: To investigate the protective effects of cerebrospinal fluid (CSF)-containing Jiawei Wuzi Yanzong formula (CSF-C) on beta-amyloid protein-induced injury of hippocampal neurons., Methods: Primary hippocampal neurons were isolated and cultured for 7 days in vitro. CSF-C was prepared by cerebrospinal fluid extracted from SD rats fed continuously with various components of Jiawei Wuzi Yanzong formula (total formula, total flavonoids or total polysaccharides) respectively. The viability, morphological change, apoptotic rate and apoptosis-related proteins (caspase-3, PARP, Bcl-2, Bcl-XL, JNK1/2 and p38 MAPK) of neurons were detected in different groups: the untreated normal group, the Abeta(25-35)-treated group, and the CSF-C protected groups (co-acted by CSF-C and 20 micromol/L of Abeta(25-35)), respectively., Results: CSF-C showed significant neuro-protective effect, and the protection of CSF-C contained total flavonoids or total polysaccharides was significantly greater than that contained total formula (P < 0.05 or P < 0.01). Moreover, the effects of CSF-C contained various components on apoptosis-related proteins were different., Conclusion: Some flavonoid and polysaccharide components in Jiawei Wuzi Yanzong formula can pass through the blood brain barrier and protect neurons from beta-amyloid protein-induced neuron injury to some extents.

Objective: To investigate the impact of intervention program upon improvement of utilization quality of carbapenems and glycopeptides., Methods: Multifaceted intervention program of carbapenems and glycopeptides was conducted at our hospital. It involved written justification forms, expert consultation committee meetings, audit, feedback and re-feedback. From November 1, 2007 until October 31, 2008, retrospective audit and feedback were performed on all patients dosed with carbapenems or glycopeptides. Case reports were reviewed and data of ratio of appropriate antibiotic use, length of hospital stay, total cost, mortality and rate of vancomycin-resistant Enterococci (VRE) were collected and compared between the first and second half year of antibiotic intervention program., Results: A total of 397 cases were reviewed, 75 cases discussed at expert committee meetings and 58 feedback letters delivered to responsible doctors. The consumption of both carbapenems and glycopeptides decreased. The appropriate use of carbapenems and glycopeptides increased from 37.8% (45/119) to 53.5% (48/127, P < 0.05) and from 45.6% (36/79) to 63.9% (46/72, P < 0.05) respectively. The total cost and mortality of patients dosed with glycopeptides decreased from a median of RMB 65,700 (30,300 - 146,900) yuan to 55,700 (36,700 - 90,900) yuan, and from 39.2% to 26.4% respectively. The rate of VRE decreased from 5.63% in 2007 to 3.80% during the second half year of 2008., Conclusion: Antibiotic intervention program of carbapenems and glycopeptides is effective in decreasing the inappropriate antibiotic use.

Objective: To observe the airway inflammation and peripheral airway function in asthmatic patients with different control levels, and to investigate whether the airway inflammation profile detected by induced sputum reflects the peripheral airway dysfunction., Methods: The recruited asthmatic subjects (n = 66) were divided into 3 groups: asthma controlled (8 male and 13 female), asthma partly controlled (12 male and 16 female), asthma uncontrolled (6 male and 11 female). Twenty healthy subjects served as the control group (9 male and 11 female). On the 1(st) day, all the subjects were required to take asthma control test (ACT), and to receive measurement of lung function by oscillometry and spirometry as well as inflammatory cell profile of induced sputum and the concentration of eosinophil cationic protein (ECP). Exhaled nitric oxide (FE(NO)) was measured on the 2(nd) day, and oscillometry methacholine provocation was conducted for patients whose baseline FEV(1) was > or = 70% predicted. The provocation process was terminated when airway resistance was increased by twice of the basic value, or when the methacholine reached the highest concentration. Then airway resistance and lung function were examined after 3 minutes. Finally, airway resistance and lung function were measured again after the subjects had 5 consecutive deep inspirations (DI). Correlation analysis was conducted between ACT scores and inflammatory cells count, ECP concentrations of induced sputum and FE(NO) among different groups. The correlations were also made between the change of peripheral airway resistance triggered by provocation or DI and ACT scores, total eosinophil, ECP level of induced sputum, FE(NO) respectively., Results: The total eosinophil count and ECP level in induced sputum and FE(NO) in asthmatic patients increased with the decline of control level. Negative correlations between ACT scores and total eosinophil count as well as the ECP level were observed (r = -0.43, -0.56, P < 0.01). In the healthy control group, the percentage of increase in peripheral airway resistance (R(5)-R(20)) and central airway resistance (R(20)) did not show significant difference (F = 3.472, P > 0.05) with methacholine provocation, while the percentage of increase in R(5)-R(20) was greater than in R(20) in both controlled and partly controlled asthmatic patients with provocation (F = 18.09 and 14.14, P < 0.01), though the change of R(5)-R(20) showed no correlations with ACT scores, eosinophil count of induced sputum, ECP level and FE(NO). After DI, R(5)-R(20) decreased from (0.13 +/- 0.14) kPa x L(-1) x s(-1) to (0.08 +/- 0.09) kPa x L(-1) x s(-1) (t = 2.84, P < 0.05) in the healthy control group, while R(5)-R(20) increased from (0.24 +/- 0.15) kPa x L(-1) x s(-1) to (0.30 +/- 0.16) kPa x L(-1) x s(-1) in the controlled asthma group, from (0.31 +/- 0.18) kPa x L(-1) x s(-1) to (0.39 +/- 0.17) kPa x L(-1) x s(-1) in the partly controlled asthma group (t = 3.90 and 4.68, P < 0.01, respectively). No correlations were observed between the change of R(5)-R(20) after DI and ACT scores, total eosinophil counts, ECP level as well as FE(NO) (r = -0.07, 0.28, -0.14, 0.14, P > 0.05)., Conclusions: Even in asthma patients with controlled disease, eosinophilic inflammation in the airway was still present, and the eosinophilic inflammation became more severe with the decline of control level. Bronchodilatory effect caused by DI disappeared in asthmatic patients. The inflammation profile detected by induced sputum did not reflect the dysfunction of peripheral airways.
