ObjectiveTo investigate the genetic polymorphism of InDel loci in SifalnDel 45plex system in the Han population in Jiangsu Province and the Mongolian population in Inner Mongolia, and to evaluate the effectiveness of the system in forensic medicine.MethodsSifaInDel 45plex system was used for genotyping in blood samples of 398 unrelated individuals from the above two populations, and allele frequencies and population genetic parameters of the two populations were calculated respectively. Eight intercontinental populations in the gnomAD database were used as reference populations. The genetic distances between the two studied populations and eight reference populations were calculated based on the allele frequencies of 27 autosomal-InDels （A-InDels）. The phylogenetic trees and multidimensional scaling （MDS） analysis diagrams were constructed accordingly.ResultsAmong two studied populations, the 27 A-InDels and 16 X-InDels showed no linkage disequilibrium between each other and the allele frequency distributions were in Hardy-Weinberg equilibrium. The CDP of the 27 A-InDels in two studied populations were all higher than 0.999 999 999 9, and the CPEtrio were all less than 0.999 9. The CDP of the 16 X-InDels in Han in Jiangsu and Mongolian in Inner Mongolia female and male samples were 0.999 997 962, 0.999 998 389, and 0.999 818 940, 0.999 856 063, respectively. The CMECtrio were all less than 0.999 9. The results of population genetics showed that the Jiangsu Han nationality, Inner Mongolia Mongolian nationality and East Asian population clustered into one branch, showing closer genetic relationship. The other 7 intercontinental populations clustered into another group. And the above 3 populations displayed distant genetic relationships with the other 7 intercontinental populations.ConclusionThe InDels in the SifaInDel 45plex system have good genetic polymorphism in the two studied populations, which can be used for forensic individual identification or as an effective complement for paternity identification, and to distinguish different intercontinental populations.

All solid state lithium-ion batteries have the advantages of high safety, long cycle life and high energy density, as compared with the conventional lithium-ion batteries, and have been attracting more and more research interest. Solid electrolyte is a crucial component of all solid state lithium-ion batteries, which largely determine the performance of the batteries. In this work, a polymer-lithium-ceramics complex was designed and synthesized. The structure and dynamics of the polymer-ceramic interface was studied with solid-state NMR techniques.

Automatic program repair is a research hotspot in recent years and has made some progress.Most of the existing automatic program repair methods use the test suite to validate patch correctness.However,using the test suite to validate a large number of candidate patches will not only bring huge costs,but also lead to the overfitting problem of patches.Therefore,how to improve the efficiency of patch validation and effectively validate patch correctness has become an urgent problem.In order to reduce the cost and improve the patch accuracy,this paper proposes an approach combining two embedding techniques to validate patch correctness.Firstly,this approach uses Doc2Vec model to calculate the similarity between the patch and the error code,then it uses the classifier based on BERT model to filter out the error patches from the patches screened by the similarity.To evaluate the effectiveness of this approach,experiments are carried out based on five open source Java benchmarks.Experimental results show that this approach can effectively validate patch correctness and improve the efficiency of patch validation.

Considering the variable mass pipe flow of the main and branch boreholes, the CBM pinnate horizontal well productivity prediction model was proposed based on 3D gas-water two-phase flow characteristics in the coal seam. The corresponding calculation program is written based on C++ programming language. At the same time, an actual CBM well is simulated as an example. The simulated results are verified by the monitoring data from the actual site. This paper studies the effects of branch symmetry, the locations of branch points, the angle between main and branch bore-holes, branch length, and branch number on gas production per unit length of the CBM well. The comprehensive results demonstrate the high accuracy of the computation model, and its ability to meet the actual needs of the project. The characteristics of gas production per unit length along the main borehole is high on both ends and low in the middle, with local decline at the branch point locations, while the characteristic of gas production per unit length along the branch borehole is high at heel, low at toe and increases gradually in the middle. Gas production per unit length at branch locations along main borehole declines faster when branches are symmetrical than asymmetrical. As branch node moves from heel to toe along the main borehole, peak gas production per unit length also transforms from toe to heel. The angles of the main and branch boreholes have no effect on gas production. At the same time, gas production per unit length reduces as the branch length along main borehole increases. Gas production per unit length increases with the distance from branch node to heel end. Gas production per unit length increases with the increase of branch length and the angle between main and branch boreholes. However, it declines as the number of branches increase. A 77-year-old man was admitted at our hospital due to "generalized increase in the number of masses and enlargement of the masses observed for one month". Combined assessment of the imaging (computed tomography and magnetic resonance imaging) findings and results of lung centesis biopsy and liquid biopsy suggest that the patient had small cell lung cancer of the left upper lobe, with right hilar, mediastinal, bilateral axillary, abdominal and retroperitoneal lymph node metastases, as well as widespread subcutaneous soft tissue, liver, bilateral adrenal, bilateral kidneys and multiple brain metastases (extensive stage). In order to obtain an evaluation of the development of the disease as soon as possible, the circulating tumor cells (CTCs) and circulating tumor microemboli (CTM) in 6 mL peripheral blood were examined by subtraction enrichment-immunostaining fluorescence in situ hybridization (SE-iFISH) technology. A total of 919 epithelial cell adhesion molecule (EpCAM)-positive CTCs and 61 EpCAM-positive CTM were identified. Among them, there were 14 haploid CTCs (1.52%), 788 diploid CTCs (85.75%), 44 triploid CTCs (4.79%), 70 tetraploid CTCs (7.62%) and 3 pentaploid or higher-fold polyploid CTCs (0.33%). Herein, we reported a rare case with extremely high accounts of CTCs and CTM and positive findings for tumor markers, which was identified for the first time. The examination of CTCs by SE-iFISH contributed to the diagnosis, prognosis and treatment evaluation of cancer and facilitated the formulation of precise and individualized therapeutic regime., (Copyright© by Editorial Board of Journal of Sichuan University (Medical Sciences).)

Circadian rhythms widely exist in living organisms, and they are regulated by the biological clock. Growing evidence has shown that circadian rhythms are tightly related to the physiological function of the cardiovascular system, including blood pressure, heart rate, metabolism of cardiomyocytes, function of endothelial cells, and vasoconstriction and vasodilation. In addition, disruption of circadian rhythms has been considered as one of the important risk factors for cardiovascular diseases, such as myocardial infarction. This review summarizes the recent research advances in the relationship between circadian clock and cardiovascular diseases, hoping to improve treatment strategies for patients with cardiovascular diseases according to the theory of biological clock.

Objective: To comprehensively evaluate the clinical effect, safety and cost of Qianlieshutong Capsules (QC) in the treatment of chronic prostatitis., Methods: We searched Cochrane Library, PubMed, Springer, ProQuest, CNKI, Wanfang Data and VIP for randomized controlled trials (RCT) on the treatment of chorionic prostatitis with QC published from January 2000 to May 2018. According to the inclusion and exclusion criteria, two researchers independently completed the screening and evaluation of the articles, extraction of information, and meta-analysis of the included RCTs using the RevMan 5.3 software., Results: Totally 10 RCTs involving 1 796 cases were included in this study, in which the chronic prostatitis patients treated by the combination of QC and quinolones all showed a significantly better response than the controls (P < 0.05). QC combined with quinolones cost an average of ¥23 more than quinolones alone with a 1% increase of therapeutic effectiveness, ¥38.39 more with a 1-unit reduction of WBCs, and ¥38.84 more with a 1-point decrease in the NIH-CPSI score., Conclusions: The combination of QC with quinolones has a better therapeutic efficacy but a higher cost than quinolones alone in the treatment of chronic prostatitis.

Objective: To investigate the effect of lithium chloride (LiCl) on cell cycle of HK-2 cells and explore the possible pathways involved., Methods: HK-2 cells were treated with LiCl at different concentrations (5, 12.5, 20, and 25 mmol/L) for 12, 24, 48, or 72 h, and the changes in cell cycle and viability were detected using flow cytometry and CCK-8 assay, respectively. Western blotting was used to analyze the changes in the expressions of cyclin B1 and CDK1 (the two G2 phase-related proteins) and those of AKT/GSK-3β signaling pathway-related proteins in the treated cells., Results: LiCl treatment time- and concentration-dependently increased HK-2 cell percentage in G2 phase and decreased the cell vitality. The expressions of cyclin B1, CDK1, p-GSK-3β, and β-catenin increased and the expression of p-AKT decreased significantly in the cells as LiCl treatment time and concentration increased., Conclusion: LiCl may cause HK-2 cell cycle arrest in G2 phase through activation of the AKT/GSK-3β signaling pathway.

Objective: To investigate whether the p38 mitogen-activated protein kinase (MAPK) signaling pathway mediates advanced oxidation protein products (AOPPs)-induced epithelial-to-mesenchymal transition (EMT) in tubular cells., Methods: Human proximal tubular cells (HK-2 cells) exposed to AOPP-bovine serum albumin (BSA) were examined for expressions of p38 MAPK and phosphorylated p38 MAPK using Western blotting. Western blotting and quantitative RT-PCR were used to examine the protein and mRNA expressions of EMT markers E-cadherin and vimentin and endoplasmic reticulum stress marker glucose-regulated protein (GRP) 78 in cells treated with SB203580 (an inhibitor of the p38 MAPK signaling pathway) prior to AOPP exposure. The cells treated with AOPPs following pretreatment with salubrinal (an inhibitor of endoplasmic reticulum stress) were also examined for expressions of p38 MAPK and phosphorylated p38 MAPK., Results: AOPP treatment induced the phosphorylation of p38 MAPK in HK-2 cells. AOPP-induced decrease in E-cadherin expression and overexpression of vimentin and GRP78 were partly inhibited by pretreatment of the cells with SB203580. Salubrina partly suppressed AOPP-induced phosphorylation of p38 MAPK in the cells., Conclusion: p38 MAPK signaling pathway, which is regulated by endoplasmic reticulum stress, might mediate AOPP-induced EMT in HK-2 cells.

Objective: To investigate the effects of transforming growth factor-β (TGF-β) and Smad in chronic pancreatitis (CP) induced by dibutyltin dichloride (DBTC) combined with ethanol, and the effect for prevention and treatment of pancreatic fibrosis by DaChaiHu Decoction (tradeiional Chinese medicine)., Methods: Ninety-six healthy male Kunming mice were randomly divided into control group (Con group), chronic pancreatitis (CP group) group, Dachai Hu decoction treatment group (DCHD group) ( n =32). Then the mice were treated with DBTC 8 mg/kg by tail vein injection and fed with 10% ethanol replacing the normal drinking water to replicate mouse CP model. After three days of modeling the mice were randomly divided into CP group and DCHD group. The mice in DCHD group were administered intragastrically with DaChaiHu Decoction (1 g/ml, 6 g/kg·d) at the following 8 weeks. After modeling for 1 week, 2 weeks, 4 weeks and 8 weeks, the mice were anesthetized and sacrificed( n =8). The morphology and the degree of fibrosischanges of pancreatic tissue were detected by HE staining. The protein expressions of type I collagen and TGF beta R I, p-Smad 2/3 and Smad 7 in pancreatic tissue were detected by Western blot assay. The expressions of MMP-1/TIMP-1 mRNA inpancreatic tissue were detected by RT-PCR., Results: Compared with control group, DBTC combined with ethanol induced CP with increased activity of serum amylase and hyaluronic acid level. While in DCHD group, the activity of amylase and hyaluronic acid level were decreased significantly. Compared with control group, the protein expressions of COLA1, TGF beta R I, p-Smad 2/3 in CP Group were elevated,but the expression of Smad 7 was significantly reduced; In DCHD treatment group,the expression of TGF beta R I, p-Smad 2/3 and COLA1 were reduced, and the protein expression of Smad 7 was increased. In 2w and 4w, the level of MMP-1mRNA continued to decrease, while TIMP-1mRNA expression was increased significantly ( P <0.01). At each time point of DCHD group, the expression of MMP-1 were markedly increased and TIMP-1 were reduced, compared with those of the CP group ( P <0.05)., Conclusions: DaChaiHu Decoction play a role in the prevention and treatment of chronic pancreatitis and the development of fibrosis, the mechanism may related to inhibit the activation of TGF beta/Smad signaling pathway, and regulate the balance between synthesis and degradation of extracellular matrix.

At present, genome-wide association study on coronary heart disease (CHD) has been carried out in several major medical research centers worldwide. Most studies of CHD susceptibility loci or regions focused on chromosome 1, 3, 9, 11 and 16, while studies on chromosome 8 are rare. To the best of our knowledge, the genome study on chromosome 8 about CHD in Chinese Han population has never been reported before. We aimed to identify CHD susceptibility loci or regions in the Chinese Han population. First, two separated DNA pooling samples were prepared from 156 CHD cases and 1000 normal controls. Then, a total of 13 microsatellite markers at an interval of 10 cM on chromosome 8 were selected for genetic scanning. Finally, the difference of allele frequency at each locus between two pooled samples was analyzed by Chi-square test. Significant differences were found between cases and controls at D8S264(8p23.3-p23.2) and D8S285(8q12.1) (both P<0.05). Therefore, 8p23.3-p23.2 and 8q12.1 are possible to be associated with CHD and further study is needed to screen susceptible genes around these regions.
