Objective: To explore the antiviral activity of the small-molecule compound AM679 in hepatitis B virus (HBV) replication and infection cell models. Methods: The positive regulatory effect of AM679 on EFTUD2 expression was validated by qPCR and Western blotting. HepAD38 and HepG2-NTCP cells were treated with AM679 (0.5, 1, and 2 nmol/L). Negative control, positive control, and AM679 combined with the entecavir group were set up. HBV DNA intra-and extracellularly, as well as the expression levels of intracellular HBV total RNAs and 3.5kb-RNA changes, were detected with qPCR. Hepatitis B surface antigen (HBsAg) and hepatitis B e antigen (HBeAg) levels were measured in the cell supernatant by an enzyme-linked immunosorbent assay (ELISA). The t-test method was used for the statistical analysis of the mean difference between groups. Results: EFTUD2 mRNA and protein expression levels were significantly increased in HepAD38 and HepG2-NTCP cells following AM679 treatment, with a statistically significant difference ( P  < 0.001). Intra-and extracellular indicators such as HBV DNA, HBV RNAs, HBV 3.5kb-RNA, HBsAg, and HBeAg were decreased to varying degrees in both cell models, and the decrease in these indicators was more pronounced with the increase in AM679 concentration and prolonged treatment duration, while the combined use of AM679 and entecavir had a more significant antiviral effect. The HBV DNA inhibition rates in the supernatant of HepAD38 cells with the use of 2 nmol/L AM679 were 21% and 48% on days three and nine, respectively. The AM679 combined with the ETV treatment group had the most significant inhibitory effect (62%), with a P  < 0.01. More active HBV replication was observed after silencing EFTUD2, while the antiviral activity of AM679 was significantly weakened. Conclusion: AM679 exerts anti-HBV activity in vitro by targeting the regulation of EFTUD2 expression.

Objectives: To establish the GC-MS qualitative and quantitative analysis methods for the synthetic cannabinoids, its main matrix and additives in suspicious electronic cigarette (e-cigarette) oil samples., Methods: The e-cigarette oil samples were analyzed by GC-MS after diluted with methanol. Synthetic cannabinoids, its main matrix and additives in e-cigarette oil samples were qualitatively analyzed by the characteristic fragment ions and retention time. The synthetic cannabinoids were quantitatively analyzed by using the selective ion monitoring mode., Results: The linear range of each compound in GC-MS quantitative method was 0.025-1 mg/mL, the matrix recovery rate was 94%-103%, the intra-day precision relative standard deviations (RSD) was less than 2.5%, and inter-day precision RSD was less than 4.0%. Five indoles or indazole amide synthetic cannabinoids were detected in 25 e-cigarette samples. The main matrixes of e-cigarette samples were propylene glycol and glycerol. Additives such as N ,2,3-trimethyl-2-isopropyl butanamide (WS-23), glycerol triacetate and nicotine were detected in some samples. The content range of synthetic cannabinoids in 25 e-cigarette samples was 0.05%-2.74%., Conclusions: The GC-MS method for synthesizing cannabinoid, matrix and additive in e-cigarette oil samples has good selectivity, high resolution, low detection limit, and can be used for simultaneous qualitative and quantitative analysis of multiple components; The explored fragment ion fragmentation mechanism of the electron bombardment ion source of indole or indoxamide compounds helps to identify such substances or other compounds with similar structures in cases.

Indole, as a typical N-heterocyclic aromatic compound, is widespread in natural environment. A growing number of researches have proved that indole is a new interspecies and interkingdom signal molecule with certain biological activities. Indole could regulate virulence, biofilm formation, antibiotic tolerance and quorum sensing of bacteria. Indole not only modulates plant growth and defense system, but also affects intestinal inflammation, oxidative stress and hormone secretion in animals. Hence indole plays important roles in diverse aspects such as microbial metabolism, human health and plant growth, holding important significance both in biology and ecology. This review presents the history of indole from biological metabolism to signal transmission, the current knowledge on indole as an intercellular and interspecies signal of microorganisms, and interkingdom signal between bacteria and plants or animals. This review will help to explore the biological significant and ecological mechanism of indole metabolic and signal regulation in complex environment.

Poly(glycidyl methacrylate-ethyleneglycol dimethacrylate) (Poly(GMA-EDMA)) monolith functionalized with cobalt phthalocyanine tetracarboxylic acid (CoPcTc) was prepared. The monolith was used for transferrin (Tf) glycopeptide enrichment. By taking advantage of hydrogen bonds between isoindole subunits of phthalocyanine and glycans and coordination interaction between cobalt and glycopeptides, the monolithic material was efficient and selective. After enrichment of transferrin through the functionalized monolith, 17 glycopeptides were identified by electrospray ionization quadrupole time-of-flight mass spectrometry. When the concentration of transferrin was reduced to 8.8×10 -10 mol/L, three glycopeptides could still be obtained. The present method has great potential for trace sample analysis.

Objective To examine whether tripartite motif-containing protein 34 (TRIM34) is colocalized with micronuclei and investigate the influence on the movement of micronuclei chromosome in mitosis. Methods The eukaryotic expression vector TRIM34-pEGFP-N3 was constructed, identified and then transfected into HEK293T cells. With 4', 6-diamidino-2-phenylindole 2HCI (DAPI) staining, the colocalization between TRIM34 and micronuclei was observed under a fluorescence microscope. Moreover, MitoTracker(R)Deep Red was used to identify the colocalization between the complex of TRIM34-micronulei and mitochondria under a confocal microscope. Finally, the effect of TRIM34 on the movement of micronuclei chromosome in mitosis was examined. Results DNA sequencing confirmed that the vector TRIM34-pEGFP-N3 was constructed successfully. A fluorescence microscope revealed that TRIM34 could be colocalized with micronuclei in HEK293T cells transfected with TRIM34-pEGFP-N3. In the same manner, a confocal microscope distinctly showed that TRIM34 was colocalized with micronuclei similarly in appearance. However, there was no distinguished colocalization relationship between the complex of TRIM34-micronulei and mitochondria. Interestingly, the micronuclei chromosome conjugated with TRIM34 was hardly transferred to equatorial plate during the metaphase stage of mitosis. Conclusion TRIM34 is colocalized with micronuclei chromosome and hampers its movement to equatorial plate in mitosis.

Chuangxinmycin (CM) from Actinoplanes tsinanensis was an antibiotic discovered by Chinese scientists about 40 years ago. It contains a new heterocyclic system of indole fused with dihydrothiopyran, whose biosynthetic mechanism remains unclear. CM is used as an oral medicine in the treatment of bacterial infections in China. The simple structure makes CM as an attractive candidate of structure modification for improvement of antibacterial activity. Recently, we analyzed the secondary metabolites of Actinoplanes tsinanensis CPCC 200056, a CM producing strain, as a natural CM analogue. We discovered the first natural CM analogue 3-demethylchuangxinmycin (DCM) as a new natural product. Compared to CM, DCM exhibited a much weaker activity in the inhibition of the bacterial strains tested. The finding provides valuable information for the structure-activity relationship in the biosynthesis of CM.

Objective: To investigate the role of glycogen synthase kinase 3β (GSK-3β) in the maturation and function of murine bone marrow-derived dendritic cells (BMDCs)., Methods: Mature DCs (mDCs) induced by LPS were examined for GSK-3β phosphorylation level with Western blotting before and after LPS exposure. To explore the role of GSK-3β in maturation and function of DCs, we added SB216763, a selective inhibitor of GSK-3β, in the cell culture of immature DCs (iDCs), and examined CD40 and CD86 expressions in the cells by flow cytometry and the expression of IL-6, IL-12 and IL-10 mRNA by real-time PCR; the changes of the immunogenicity of the cells was evaluated by mixed lymphocyte reaction. The expression of GSK-3β and RelB was examined by Western blotting in DC2.4 cells transfected with a lentiviral vector over-expressing murine GSK-3β gene., Results: LPS exposure significantly lowered GSK-3β activity in iDCs as demonstrated by increased Ser9 phosphorylation and reduced Tyr216 phosphorylation. GSK-3β inhibition induced DC maturation by increasing the expression of surface costimulatory molecules CD40 and CD86, lowered the expressions of IL-6 and IL-12 while enhanced the expression of IL-10 in iDCs, and impaired mixed lymphocyte reaction of the cells. In DC2.4 cells, lentivirus-mediated over-expression of GSK-3β obviously down-regulated the expression of RelB., Conclusions: GSK-3β is a crucial enzyme involved in the differentiation and maintenance of an immature phenotype of DCs. GSK-3β is constitutively active in iDCs to inhibit their spontaneous maturation. DCs become phenotypically mature after inhibition of GSK-3β, which also executes a proinflammatory task in DC activation. The reduction of RelB protein levels as a result of GSK-3β overexpression supports GSK-3β as a new target for inducing tolerogenic DCs.

Three compounds were isolated from solid culture of endophyte Myrothecium roridum IFB-E091 in Artemisia annua. Their structures were determined as (S)-(-)-N-[2-(3-hydroxy-2-oxo-2,3-dihydro-1H-indol-3-yl)-ethyl]-acetamide (1), N-(4-hydroxyphenethyl)acetamide (2) and asperfumoid (3), in which compound 1 was a new indole derivative. In cytotoxicity assay, the compound 1 had no obvious inhibition activity in human hepatoma cell line SMMC-7721 and human cervical carcinoma cell line HeLa.

Objective: To study the chemical constituents of the whole plant of Sphagneticola trilobata., Methods: The compounds were isolated and purified by column chromatography and their structures were determined by spectroscopic techniques., Results: Three lignans, two indolics and two phenolic glycosides were isolated from the whole plant of Sphagneticola trilobata and identified as syringaresinol-4-O-β-D-glucopyranoside(1), pinoresinol-4-sulfate(2), pinoresinol-4-O-β-D-glucopyranoside(3), 1H-indole-3-carboxylic acid (4), 1H-indole-3-carbaldehyde(5), 2,6-dimethoxy-4-hydroxyphenol-1-O-β-D-glucopyranoside (6), and 3,5-dimethoxy-4-hydroxyphenol-1-O-β-D-glucopyranoside (7)., Conclusion: Compounds 1 - 7 are isolated from the genus Wedelia for the first time. Compound 4 demonstrates significant inhibitory activity against α-glucosidase.

The interactions of bovine serum albumin (BSA) with five novel silicon (N) phthalocyanines(SiPcl-5) axially modified by nucleosides (cytidine, 5-N-cytidine, methyl cytidine, uridine and methyl uridine) derivatives were studied by fluorescence spectroscopy. The results show that there are strong interactions between these silicon phthalocyanines and BSA with a binding constant of (4.90-83.18) x 10(5) mol(-1) x L. Therefore, the non-covalent BSA conjugate of bis(2', 3'-O-isopropyl-cytidine-oxy) phthalocyaninatosilicon(IV) (SiPc1) was further been prepared. The molar ratio of phthalocyanine to albumin was found to be 1:1 for the obtained SiPcl-BSA conjugate. The absorption spectra of SiPc1 and SiPc1-BSA in the visible region have no significant difference, both showing an Q-band maximum at about 686 nm. It indicates that the spectroscopic characteristics of SiPc1 are not affected by binding to albumin. The SiPcl-BSA conjugate exhibits high photodynamic activity against human hepatoma cell line HepG2 with an IC50 value of 3.0 x 10(-7) mol x L(-1). By comparsion, SiPc1-BSA has a higher photodynamic activity than SiPc1 (in PBS formation, IC50 = 7.0 x 10(-7) mol x L(-1)), which can be attributed to its higher cellular uptake.

Taking application of some isolation and purification technologies, such as solvent extraction, preliminary solvent isolation, column chromatographies over silica gel and Sephadex LH-20 gel and preparative HPLC, 10 compounds were obtained from Gynura nepalensis cultivated in the suburban area of Beijing. Their structures were identified by spectroscopic methods and comparison with literature as (3R) -3-hydroxy-β-ionone (1), (3S,5R, 6S, 7E) -5, 6-epoxy-3-hydroxy-7-megastigmen-9-one (2), (+) -boscialin (3), 3, 6-trans-3-hydroxy-α-ionone (4), 3, 6-cis-3-hydroxy-α-ionone (5), 3, 4-cis-3, 4-dihydroxy-β-ionone (6), ethyl caffeate (7), loliolide (8), 1H-indole-3-carbaldehyde (9), and 3-(hydroxyacetyl)indole (10), respectively. All compounds were isolated from the title plant for the first time, and with compounds 1, 2, 4-7, 9 and 10 being isolated from Gynura species for the first time. Structurally, the above compounds 1-6 belong to C13 nor-sesquiterpenoids, sharing the same carbon skeleton of megastigmane. According to this study, they are one of major kinds of chemical constituents of Gynura nepalensis and have important reference value for the investigation on phytotaxonomy of this species.

PARP [poly(ADP-ribose)polymerase] represents a novel potential target in cancer therapy. It is involved in a DNA repair process by catalyzing the transfer of ADP-ribose units from NAD to a number of its substrate proteins. In this work, a series of novel azaindole derivatives was designed and synthesized. Moreover, 16 target molecules were screened and 8 compounds displayed inhibitory activity against PARP-1. It has been demonstrated that these azaindoles bearing cycloamine substituents at 2-position were active to both PARP-1 and PARP-2.

Objective: To prepare (-)-Antofine liposomes with high activity and good water-soluble and study its anti-breast cancer activity both in vitro and in vivo., Methods: (-)-Antofine liposomes were prepared by film dispersion method, the encapsulation efficiency of (-)-Antofine liposomes were determined by UV spectrophotometer, the cytotoxic activity of (-)-Antofine liposomes on MCF-7 cell was measured by MTT assay, the antitumor activity and major organs indexes were evaluated by tumor-bearing nude mice experimental in vivo., Results: The average encapsulation efficiency of (-)-Antofine liposomes was (80.8 +/- 2.2)%, average drug loading was (26.7 +/- 1.4)%, it was relatively good water-soluble and had good anti-tumor activity in vitro and in vivo., Conclusion: (-)-Antofine liposomes prepared by film dispersion method has high encapsulation efficiency, the water-soluble and the anti-tumor activity are improved compared with (-)-Antofine.

Photophysical photochemical properties, in vitro photodynamic anticancer activity and interaction with albumin of a new axially modified silicon phthalocyanine, i. e. di (2-amino-6-trifluoromethyl-4-pyrimidinyloxy) silicon phthalocyanine (SiPcF), were studied in the present paper. The Q band maximum absorption of SiPcF located at 686 nm with the molar absorption coefficient of 2.3 x 10(5) mol(-1) * L * cm(-1). The fluorescence emission of SiPcF peaked at 694 nm with a fluorescence quantum yield of 0.46. Its singlet oxygen quantum yield produced by photosensitization is 0.38, suggesting that SiPcF is an effective 1O2 photosensitizer. There is strong interaction between SiPcF and bovine serum albumin (BSA). The binding constant is 4.33 x 10(5) mol X L(-1) and the number of binding sites is 1. In vitro experiments show that SiPcF had a high photodynamic anticancer activity against human hepatoma cells HepG2 with an IC50 value down to 5 x 10(-7) mol X L(-1).

Objective: To provide a reference evidence for choosing salt-resistent and drought-resistent varieties of Isatis indigotica Fort.., Methods: The tissue culture seedlings of different Isatis indigotica tetraploids were used as plant materials. The diploid was used as CK, which were cultured for 30 days. 2 g/L NaCl and 15% PEG-6000 were used as stress treatment respectively for 12 hours, then poured out the solution, the materials were continuously cultured for 7 days. The contents of indirubin and indigotin were determined by HPLC., Results: The contents of indigotin and indirubin of DB3, DB5 and DB12 with PEG treatment and DB2, DB3 and DB12 with NaCl treatment were increased., Conclusion: DB3 and DB12 can be further studied as reproduction materials.

The resonance Rayleigh scattering spectral detection system was designed based on the 2, 9, 16, 23-tetracarboxylate-phthalocyanine zinc and protein system. In the system, excitation light source is 405 nm wide band gap semiconductor lasers, and monochromator is 475 nm narrow-band band-pass filter, and the detector is low-noise and high-gain photoelectric amplifier based on blue-ray enhanced photodiode. Experiment shows that, the solution's strong absorption wavelength is near 420 nm. Under the action of incentive light, resonance Rayleigh scattering is generated at the resonant wavelength, and the scattering intensity is proportional to the protein content. The system uses 2, 9, 16, 23-tetracarboxylate as the spectrum probe to determine the concentration of serum proteins by resonance Rayleigh scattering method. Its linear detection range is 10 - 50 mg.mL-1, and its detection limit is 0. 001 mg.mL-1. The newly developed device for detecting concentration of the serum protein has the advantages of small size, low cost, low power consumption, and being easy to use.

Objective: To investigate the chemical constituents in water-soluble fraction of Carthamus tinctorius., Methods: Compounds were isolated and purified by macroporus resin, silica gel, Sephadex LH-20 column chromatography and preparative HPLC. The structures were identified by spectral analysis., Results: Twelve compounds were isolated and identified as 4-Hydroxybenzaldehyde (1); E-1-(4'-hydroxypheny) -but-1-en-3-one (2); 3-Formylindole (3); 2-Acetyl-5-hydroxymethylfuran (4); p-Hydroxycinnamic acid (5); (6R, 7E, 9R) -9-hydroxy-4,7-megastigmandien-3-one (6); 4-hydroxyacetophenone (7); 5-(hydroxymethyl) -2-furaldehyde (8); 4-Hydroxybenzoic acid (9); Stigmasterol-3-O-beta-D-glucopyranoside (10); Daucosterol (11); beta-sitosterol (12)., Conclusion: Compounds 1 - 4, 6, 7, 10 are isolated from this plant for the first time.

Farnesoid X receptor (FXR) belongs to the nuclear receptor superfamily. It is highly related to the formation of metabolic syndrome and the glucose homeostasis, and therefore represents an important drug target against metabolic diseases and diabetes. In recent years, great progress has been made in the agonists, antagonists, and crystal structures of FXR. The diverse FXR ligands and their structure-activity relationship are reviewed in this article. The advances in the crystal structures of FXR in complex with different ligands are also introduced.

A novel 206 nm excilamp generated by microwave-driven Kr/I2 mixtures was employed for nitrogen-heterocyclic compounds (NHCs) degradation in aqueous solution. The photodissociation efficiencies of indole and quinoline with 206 nm excilamp were estimated on the basis of removal efficiency of targeted compounds and the loss of total organic carbon (TOC). The results indicated that removal efficiency of 20 mg x L(-1) indole was as high as 62.0% after 80 min and TOC loss efficiency of 50.7% for 150 min. The irradiation time, initial concentration and pH value had some influences on quinoline degradation. Indole removal efficiency and TOC loss was markedly higher than that of quinoline under the same condition. The intermediates were identified qualitatively by gas chromatography/mass spectrum (GC/MS) with headspace sampling after they were extracted by rotary evaporator. GC/MS analysis indicated that indole and quinoline underwent ring-open dissociation under 206 nm irradiation, as a result, benzene, xylene, acetate, aldehyde, as well as ester compounds were formed, while indole aggregation reaction occurred during indole photodegradation. At last, degradation mechanisms of quinoline and indole in aqueous media with 206 nm excilamp were proposed on the basis of intermediates.

Objective: To analyze the components of essential oil from Lactuca sativa seeds growing in Xinjiang., Methods: The components of essential oil from Lactuca sativa seeds were analyzed by gas chromatography-mass spectrometry (GC-MS)., Results: 62 components were identified from 71 separated peaks,amounting to total mass fraction 95.07%. The dominant compounds were n-Hexanol (36.31%), n-Hexanal (13.71%), trans-2-Octen-l-ol (8.09%) and 2-n-Pentylfuran (4.41%)., Conclusion: The research provides a theoretical basis for the exploitation and use of Lactuca sativa seeds resource.

Based on the structure of 5-fluoroindol-2-one and fragments from thirteen multi-target tyrosine kinase inhibitors which have been marketed or in the phase of clinical research, eleven 3-aromatic Shiff base-5-fluoroindol-2-one derivatives were designed and synthesized. Their structures were identified by 1H NMR, MS and elemental analysis. In vitro antitumor bioactivities evaluation was done by MTT method. It was shown that most of synthesized compounds had antitumor activities and compounds 1b, 1g, 1i and 1h were better than or equal to the antitumor activity of positive control.

Four kinds of covalent serum albumin (BSA or HSA) conjugates of beta-tetrakis[(3,5-dicarboxy)phenoxy]-phthalocyaninatozinc (1) and beta-octakis [(3, 5-dicarboxy) phenoxy]-phthalocyaninatozinc (2) were synthesized by the approach of amide bond. The molar ratio of phthalocyanine to albumin in conjugates were found to be 6 - 7.1. Their absorption spectra were measured in PBS solution. When conjugated to albumin, compound 1 displays more distinct monomer absorption characteristics (with the maximum absorption at 677 nm) than its free form. Compound 2 in albumin framework exists mostly in monomer form, which is beneficial to photodynamic therapy in aqueous solution. Both 2-BSA and 2-HSA show a sharp and intense Q-band at 681 nm and 682 nm with the molar extinction coefficient of 2.01 x 10(5) and 2.05 x 10(5) mol(-1) L cm(-1) respectively. The Q-band absorption spectra and existent state of phthalocyanine 1 or 2 in conjugates were not affected by the pH value of aqueous solution, which are obviously different from that of the corresponding free phthalocyanine.

Interaction between 1-[4-(2-carboxyl-ethyl-)phenoxy] phthalocyanine Zinc(II) (ZnPcC1) and albumin (human serum albumin or bovine serum albumin) was studied. ZnPcC1 can be covalently bound to albumin through amide bond formation. The molar ratios of ZnPcC1 to albumins are found to be about 7 : 1 in the covalent bioconjugates. On the other hand, there are strong non-covalent interactions between ZnPcC1 and albumins with a binding constant of ca. 1.0 x 10(5) mol(-1) x L. Binding sites competition experiments suggest that the binding site locates in subdomain I B of human serum albumin. When conjugated to albumin, no matter covalent conjugation or non-covalent conjugation, the ZnPcC1 exhibit more distinctive characteristic monomer absorption than the free ZnPcC1, which is a property beneficial to photodynamic therapy. Covalent conjugation results in the Q-band of ZnPcC1 red-shifting about 5 nm, whereas non-covalent conjugation does not lead to red-shift.

Our work focuses on the quality control and structural identification of Photocyanine as a cancer therapeutic photosensitizer. Photocyanine is a mixture which contains four ZnPcS2P2 type substituted Phthalocyanine isomers. In order to obtain the single component from Photocyanine, the mixture of four isomers possessing the similar structures and chemical property had been isolated and purified. An HPLC method with a mixture of methanol-acetonitrile-ion-pair buffer as the mobile phase was applied to isolate the four isomers by means of a semi-preparative C18 column. To remove the salts which were mixed in the preparative product, a SPE C18 column was used to separate the salts by elution with water and then the marker component was eluted by methanol. Subsequently, a column of Sephadex LH-20 gel was applied to elute the crudes with methanol to desalination. The purity of the isolated compound was measured by TLC and four different isomers of phthalocyanine were obtained. The chemical structures of them were elucidated by 1H NMR spectra, gCOSY and NOE1D. An HPLC-DAD method was developed for simultaneously determination of four major isomers in Photocyanine with a C18 column (Grace Smart, 150 mm x 4.6 mm ID, 5 microm). The separation was carried out with a gradient program at a flow rate of 1.0 mL x min(-1). The mobile phase was a mixture of acetonitrile and ion-pair buffer (0.01 mol x L(-1) hexadecyl trimethyl ammonium bromide and 0.01 mol x L(-1) potassium dihydrogen phosphate, adjusted the pH value to 6.8 with potassium hydroxide solution). The resolution values of four isomers were 2.5, 1.20, 1.33, and 1.8. Linear regression analysis for four compounds was performed by the external standard method. Four constituents were linear in the concentration range of 0.005 to 10 microg. The values of relative standard deviation (RSD) of intra-day were 0.12%, 0.66%, 0.99%, and 1.21%, respectively. The limits of detection for four compounds were 15 ng, 20 ng, 12 ng, and 25 ng, respectively. This method was simple, accurate and reproducible. The developed method can be successfully applied to analyze isomers in Photocyanine.

Objective: To screen the factors that affect indirubin-generated significantly in the process of preparing indigo naturalis, optimize level combination and determine the optimum technology for indirubin-generated., Method: Using concentration of indirubin (mg x g(-1)) that generated by fresh leaf as an index, Plackett-Burman design, Box-Behnken design response surface analysis as the statistical method, we screened the significantly influencing factors and the optimal level combination., Result: The soaking and making indirubin process in preparing indigo naturalis was identified as the wax is not removed before immersion with immersion pH 7, solvent volume-leaf weight (mL: g)15, soaked not avoided light, soaking 48 h, temperature 60 degrees C, ventilation time of 180 min, and added ammonia water to adjust pH to 10.5., Conclusion: The soaking and making indirubin process in preparing indigo naturalis is optimized systematically. It clarify the various factors on the impact of the active ingredient indirubin which controlled by industrialized production become reality in the process of preparing indigo naturalis, at the same time, it lay the foundation for processing principle of indigo naturalis.

To study chemical constituents of the leaves of Nauclea officinalis, eight alkaloids were isolated from 95% ethanol extract by various chromatographic methods. The structures were elucidated on the basis of spectroscopic data (IR, UV, ESI-MS, 1D and 2D NMR) and identified as naucleactonin C (1), strictosamide (2), vincosamide (3), pumiloside (4), angustoline (5), angustine (6), 18, 19-dihydroangustine (7) and naucleofficine D (8). Compound 1 is a new indole alkaloid. Compounds 6 and 7 were isolated from this plant for the first time.

Accessed to literatures at home and abroad, we introduced the process of indigo naturalis transforming from dyestuff industry into pharmaceutical industry. It is affirmed that the precursors of indigo and indirubin are isatan A, isatan B, isatan C and indicant. Meanwhile, author clarified the mechanism of transformation for these precursors transforming into indigo and indirubin. And we summarized methods of determination for these precursors. In summary, these references provide us accordance of study on processing principle of Indigo naturalis, and lay the foundation for technics of making indigo and indirubin entering into modern industry.

Influenza is a major threat to millions of people worldwide. Vaccines and antiviral agents are two main options available to reduce the impact of the influenza virus, while anti-influenza agents are the most effective means to prevent the transmission of the highly contagious virus and to treat the epidemics of disease. At present, four anti-influenza agents have been approved by the FDA for the treatment of influenza, including two M2 protein ion channel inhibitors-amantadine and rimantadine and two neuraminidase inhibitors-zanamivir and oseltamivir. Arbidol hydrochloride, launched in Russia, is a potent inhibitor of influenza virus, too. Neuraminidase inhibitors could be classified generally by structure into six different kinds: sialic acid derivatives, benzoic acid derivatives, cyclohexene derivatives, cyclopentane derivatives, pyrrolidine derivatives and natural products. In this paper, recent progress in the research of the action mechanisms and structure-activity relationships of these anti-influenza virus agents were reviewed.

Objective: To investigate the chemical constituents with immunosuppressive function from Alisma orientalis., Method: The chemical constituents were isolated and purified by kinds of column chromatography and its structures were elucidated by NMR spectra and physicochemical properties. Its immunocompentence of lymphocytes taken from spleen of mouse were examined by MTT assay., Result: Twelve compounds were isolated and identified as clovandiol (1), orientalol E (2), alismoxide (3), alismol (4), 4alpha, l0alpha-dihydroxy-5beta-H-guaj-6-en (5), alismorientols A (6), alisol F (7), alisol A (8), 13beta,17beta-epoxy alisol A (9), alisol B 23-acetate (10), 1H-indole-3-carboxylic acid (11) and cuccinic acid (12). Compounds 9, 10 and alisol A 24-acetate showed immunosuppressive function., Conclusion: Compounds 1, 5, 11 and 12 were isolated firstly from this genus,and the NMR spectra data of 1 were corrected firstly, some protostan-type triterpenoids may be developed as new drug with immunosuppressive function.

In decades, metallo-phthalocyanines (MPcs) have undergone a renaissance because of their singular and unconventional physical properties. However, for the successful application of MPcs in practical devices, it is important to disperse MPc molecules into solid state matrix to fabricate MPc doped composite with desired properties. Inorganic glass is an ideal matrix because of its transparency and high environmental stability. One attractive approach to fabricating MPc/inorganic composite is sol-gel technique. In the present paper, silica gel glass matrix was prepared by hydrolysis and poly-condensation of tetraethyloxysilane. 1,2-dicyanobenzene and analytically pure soluble nickle salt were used as the nickle phthalocyanine (NiPc) reactants and chemical synthesis technique was used to prepare NiPc doped sol-gel materials at several temperatures. During the heat treatment, four 1, 2-dicyanobenzene molecules and one nickle ion collide to form a NiPc molecule. In-situ synthesizing process of NiPc in the pores of silica gel glass matrix was traced by UV/Vis absorption spectra. Owing to the remarkable absorption band of NiPc in visible region, quantity of in-situ synthesized NiPc was calculated by the absorbance at certain wavelength of 670 nm, using composites with physically doped NiPc as a reference. The in-situ synthesized kinetics was studied in detail and found to be consistent with Avrami-Erofeev equation The reaction grades were deduced to be 4.5, 4.5, 3.7, 3.2 and 1.9 respectively at temperatures of 180 degrees C, 185 degrees C, 190 degrees C, 195 degrees C and 200 degrees C, respectively.

A high performance liquid chromatography-electrospray ionization mass spectrometry (HPLC-ESI/MS) method for the simultaneous determination of five effective components in Qingrejiedu oral liquid was developed. The HPLC separation was performed on a Zorbax SB C18 column (250 mm x 3.0 mm, 5 microm) using 0.4 mmol/L sodium acetate solution containing 0.2% formic acid (A) and acetonitrile (B) as the mobile phase with gradient elution (0 min, 10% B; 0 --> 6 min, 10% B --> 30% B; 6 --> 15 min, 30% B --> 39% B; 15 --> 16 min, 39% B --> 80% B) at a flow rate of 0.35 mL/min. The analytes were detected by ESI(+)-MS under selected ion monitoring mode (0 --> 9.7 min, m/z 377; 9.7 --> 12 min, m/z 411; 12 --> 14.7 min, m/z 447; 14.7 --> 18 min, m/z 557; 18 --> 25 min, m/z 263). The linear ranges were 0.050-50 mg/L, 0.020-20 mg/L, 0.005-30 mg/L, 0.010-15 mg/L and 0.010-10 mg/L with detection limits of 0.010, 0.005, 0.001, 0.002 and 0.003 mg/L for chlorogenic acid, geniposide, baicalin, forsythin and indirubin, respectively. The average recoveries ranged from 97.0% to 101.7%. The relative standard deviations were less than 2.2%. This method is rapid, accurate, and suitable for the quality control of the five effective components in Qingrejiedu oral liquid.

The interaction of p-HPcZn and myoglobin was studied by fluorescence spectra and synchronous fluorescence spectra methods under the physiological condition. The p-HPcZn can quench the fluorescence of myoglobin effectively, and it is indicated that there is a strong interaction occurring between p-HPcZn and myoglobin. The results of the fluorescence spectra with changing temperature proved that the interaction can lead to the formation of complex of p-HPcZn with myoglobin, and quench the fluorescence of myoglobin through the static quenching mechanism. Dealing with the values of fluorescence spectra, the binding parameter and the binding site of the interaction can be obtained, whose values are 2.481 X 10(5) and 0.444 respectively. In addition, the interaction can change the conformation of myoglobin markedly also.

The tetra-sulfonated phathalocyanine cobalt(CoPcS4)was synthesized by using 4-sulfonic phthalic acid as raw material, CoCl2 as template agent and (NH4)2 MoO4 as catalyst. CoPcS4 was demonstrated to be C4h mono-isomer by elementary analysis, IR, NMR and HLPC. Depolymerization of CoPcS4 was found in DMSO. The depolymerization increased with the pH value raising. The interaction of tetra-sulfonated phathalocyanine cobalt with bovine serum albumin (BSA) was studied by UV-Vis and fluorescence spectra The number of binding sites and binding constant were determined at equal pace of the order of magnitude 10(5) L x mol(-1). CoPcS4 binds with BSA at Site I and Site II. In conclusion, COPcS4 binds with BSA commendably and albumin plays a role of storage and transport.

In order to find new indolin-2-one derivatives as antitumor agents, a series of 3-pyrrole substituted 1-(5-formyl-2-furanylmethyl) indolin-2-one derivatives were designed and synthesized. 5-Formyl-2 ,4-dimethyl-lH-pyrrole-3-carboxylic acid ethyl ester was condensed with 5-substituted indolin-2-one 2a-2d to afford 3-[(pyrrol-2-yl) -methylidenyl] indolin-2-ones 3a-3d. Through N-alkylation, 1-(5-formyl-furfuryl) -indolin-2-one 4a-4d were prepared. Compounds 4a-4d were then condensed with indolin-2-one to afford bis-indolin-2-one derivatives 5a-5d. The structures of the synthesized compounds were determined by 1H NMR, MS and element analysis. Antitumor activities of all the synthesized compounds in vitro were tested. All the 12 synthesized compounds possess antitumor activities against SPC-A1 strain. Especially the compounds 5a-5d possess potent antitumor activities better than sunitinib. Their IC50 are all below 5 micromol x L(-1).

Objective: To investigate the chemical constituents of the aerial parts of Plumbago zeylanica Linn., Methods: The constituents of the EtOAc-soluble portion in the 95% ethanol extract were isolated and purified by means of chromatography. Compounds were identified by their physical characteristics and spectral features., Results: Nine compounds were isolated as plumbagin (I), isoshinanolone (II), plumbagic acid (III), beta-sitosterol (IV), 4-hydroxybenzaldehyde (V), trans-cinnamic acid (VI), vanillic acid (VII), 2, 5-dimethyl-7-hydroxychromone (VIII), indole-3-carboxaldehyde (IX)., Conclusion: Compounds V, VII, VIII and IX were isolated for the first time from Plumbago Linn.

Objective: To study the chemical constituents in the root of Isatis indigotica., Method: The constituents root were separated through various chromatographic techniques and their structures were elucidated by means of physicochemical properties and the analysis of their spectral data., Result: Eleven compounds were isolated and identified as (+) -isolariciresinol (1), lariciresinol (2), lariciresinol-9-O-beta-D-glucopyranoside (3), lariciresinol-4'-O-beta-D-glucopyranoside (4), lariciresinol-4,4'-bis-O-beta-D-glucopyranoside (5), 3-formylindole (6), 1-methoxy-3-indolecarbaldehyde (7), 1-methoxy-3-indoleacetonitrile (8), deoxyvasicinone (9), epigoitrin (10), adenosine (11)., Conclusion: Compounds 4-8 were isolated from I. indigotica for the first time.

FTIR has been used broadly for its quick analytical rate, good reproducibility, low cost and no waste sample. The rule of infrared spectroscopic characteristics of nine 3-substituted phenyl-5-(3'-indolyl)-isoxazoline derivatives (containing phenyl and indolyl) was studied, and the influence of the substituted groups on the IR was indicated. All the 1H NMR chemical shifts of the nine novel compounds were discussed. The rule of change in chemical shifts is the same as that of IR. The study will provide a new way of elicitation for studies on the spectroscopy of this kind of compounds.

Octa-carboxylic phthalocyanine aluminium (AlPc(COOH)8) was used as fluorescent probe of infrared region. The interaction of octa-carboxylic phthalocyanine aluminium (AlPc(COOH)s) and bovine serum albumin(BSA) was studied by UV/ Vis and fluorescence spectra methods. The binding constant KA and n of phthalocyanine aluminium with BSA were determined. The results were K=5. 74 X 10(5) , n= 5. 7 and K= 3. 51 X 10(5) , for these two methods respectively. The same results by using two different analytical methods were obtained. Besides, hemin chloride(HE), ibuprofen(IB) and L-tryptophan(TRP) were used as probes, and the effects of these probes on the spectra of AlPc(COOH)8 )-BSA were studied by competitive binding method. The result indicated that, by adding HE into the AlPc(COOH)8)-BSA system, obvious spectral change of the system was observed, while adding TRP and IB caused no spectral changes. The binding site of octa-carboxylic phthalocyanine aluminium on the BSA was found to be at the I site by competitive binding method.

The interactions of the new photosensitizer {2, 3, 9, 10, 16, 17, 23, 24-octakis[(3,5-dicarboxy)phenoxy]-phthalocyaninato}zinc(II) [ZnPc(COOH)16] and serum albumins (BSA and HSA) were investigated by absorption and fluorescence spectroscopy. The binding constants were found to be 2.25-2.94 x 10(6) L x mol(-1). The 1 : 1 ZnPc (COOH)16-BSA and ZnPc (COOH)16-BSA conjugates were also prepared by incubating a mixture of ZnPc(COOH)16 and the corresponding serum albumin in 2 : 1 mole ratio followed by separation by gel permeation chromatography. The Q band absorption and fluorescence emission of ZnPc(COOH)16 are slightly red-shifted in the conjugates, and the spectral features suggest that the phthalocyanine exists predominantly in monomeric form in the conjugates.

The synergistic effects of ultra sound and zero-valent iron were studied by measuring the degradation of X-3B in zero-valent reduction, ultra sound degradation and synergistic systems. The effects of pH, ultra sound power and irradiation time on the synergistic system were explored. The results show that the X-3B is degraded difficultly by ultra sound and the total removal rate is less than 7.75% after 25min ultrasonic irradiation. When pH is 5.0, 7.0 and 9.0, the degradation rate of X-3B in synergistic system is about 1.68, 1.76 and 1.67 times, respectively, than the total removal rate of zero-valent iron reduction plus ultrasound degradation. After 25min ultrasonic irradiation in synergistic system, 99.42% of X-3B is removed at pH7.0. However, the degradation rate of X-3B in zero-valent iron reduction system is much higher than that of synergistic system at pH3.0. The degradation mechanism of X-3B in zero-valent iron reduction system is different from that of synergistic system. The X-3B is degraded into intermediate pollutants in the former, but is degraded completely in the latter. Accordingly, the X-3B can be degraded effectively in the synergistic system of zero-valent iron and ultra sound, when pH is 5.0 approximately 7.0 and the power of ultra sound is 400 approximately 600W.

Aim: To explore the anti-lymphoma efficacy of ZnPcS(2)P(2)-mediated photodynamic therapy(PDT) combined with mGM-CSF and mB7.1 gene-modified high-tumorigenicity lymphoma cells (EL-4 cells) in C57BL/6 mice., Methods: EL-4/pLXSN, EL-4/GM and EL-4/B7 were generated by consecutive infection of EL-4 cells with respective retroviruses, i.e pLXSN, pLXSNmGM-CSF and pLXSNmB7.1. The total of 40 C57BL/6 mice receiving the inoculation of EL-4 cells five days before the experiment, were randomly divided into five groups with 8 in each group: control group (without any treatment), ZnPc-PDT control group, ZnPc-PDT+EL-4/pLXSN control group, EL-4/B7+EL-4/GM control group and ZnPc-PDT+EL-4/B7+EL-4/GM group. The procedure of PDT was carried out as follows: the mice were injected with ZnPcS(2)P(2) via the lateral tail veins, and 4 h later the tumor was irradiated with 670 nm laser. The size of tumor node was measured every other day. The survival time and relative tumor volume(RTV) of each group were compared. The histological morphology of tumor was observed., Results: Compared with each control group, the survival time of experimental group was significantly prolonged (P<0.01). Moreover, difference of the survival time was measured between ZnPc-PDT control group and EL-4/B7+EL-4/GM control group (P=0.01)., Conclusion: Tumor-localized immune stimulation by mGM-CSF and mB7.1 remarkably enhanced ZnPc-PDT-elicited anti-lymphoma potency.

Aim: To design and synthesize new compounds of prandial glucose regulator with more simple structure., Methods: The target compounds were synthesized from diethyl succinate and benzaldehyde or 4-fluorobenzaldehyde by four-step reactions. Thus 18 compounds were synthesized. Their structures were comfirmed by NMR, MS and IR., Results: Seventeen compounds had different hypoglycemic activity in mice, among them, 9 compounds had higher hypoglycemic activity and 6 compounds had character of prandial glucose regulator., Conclusion: Part of the compounds have higher hypoglycemic activity deserve to be further investigated.

With metal phthalocyanine used as the photosensitizer, the cell membrane fluidity under the conditions of different irradiation time in photodynamic therapy was investigated by fluorescence polarization. Results show that with the photosensitizer phthalocyanine excited, the fluorescence polarization of the fluorophose was increased and the cancer cell membrane fluidity was decreased due to the photodynamic therapy. Cell proliferation was also studied by MTT. Results show that cell membrane fluidity and cell proliferation are influenced to the same extent, suggesting that cell membrane is one of the binding sites in PDT.

A new silicon phthalocyanine axially substituted by Nipagin, i.e. bis(4-methoxycarboxyl phenoxy)phthalocyaninatosilicon, has been synthesized and characterized by IR, NMR, HPLC and elemental analysis. Its electronic absorption spectra and fluorescence spectra in different solvents were investigated. It was found that the compound existed in the form of monomer in a saline solution containing 2% (phi) Cremophor EL and 20% (phi) propanediol. The electronic absorption spectra of the compound in the above saline solution were typical for non-aggregated phthalocyanines, showing an intense and sharp Q band at 683 nm with a molar absorption coefficient of 7.47 x 10(4) mol(-1) x L x cm(-1). The compound exhibited a relatively strong fluorescence emission at 690 nm with a quantum yield of 0.34 and with a fluorescence lifetime of 4.7 ns. The primary in vitro assay showed the compound had photodynamic killing activities against B16 melanoma cells with a LD50(half lethal dose) of 1.2 x 10(-4) mol x L(-1).

Objective: To study the chemical constituents of the leaf of Isatis indigotica., Method: Chromatography and spectral analysis were respectively used to isolate and identify the constituents., Result: Three compounds were isolated from the ethanol extracts of theleaf of I. indigotica, and identified as indirubin, tryptanthrin and L-pyroglutamic acid., Conclusion: L-pyroglutamic acid was isolated from the genus for the first time, and tryptanthrin was isolated from the leaf of this plant for the first time.

The binding between two surface-active substituted 3H-indole fluorescence probes, i. e., iodo-dihexadecyl methyl-2-(p-dodecyl amino phenyl)-3, 3-dimethyl-5-carboethoxy-3H-indole ammonium and iodo-dimethyloctadecyl-2-(p-dodecyl amino phenyl)-3,3-dimethyl-5-carboethoxy-3H-indole ammonium, and bovine serum albumin (BSA) in aqueous solution was studied using fluorescence. The binding constant and binding site number of molecule 1 and molecule 2 with BSA were obtained. It was confirmed that electrostatic interaction is the primary driving force for the combination of BSA with molecule 1 or molecule 2. According to the Förster resonance energy transfer theory, the distances between molecule 1, molecule 2 and tryptophan of BSA were calculated to be 2.90 nm and 4.02 nm, respectively.

A substituted 3H-indole quaternary ammonium molecule was designed and synthesized using hexamethylphosphoramide (HMPA) as a solvent. The products were purified and characterized by IR, 1H-NMR, MS and elemental analysis. The binding reaction of this compound with bovine serum albumin (BSA) in aqueous solution was studied using fluorescence. Their binding constant is Ka = 1.995 x 10(5) dm3 x mol(-1) and the binding site number is n = 1.12. It is confirmed that the combination is a single static quenching process.

For indole-3-butyric acid (IBA), the non-protected fluid room temperature phosphorescence (NP-RTP) and the fluid room temperature phosphorescence with polymeric dispersant polyethyleneglycol-200 (PEG-200), PEG-400 or non-ionic surfactant Tween-20, Tween-40, Tween-80, Tween-85, Brij35 and emulsifier OP as a medium have been studied in detail. When the surfactants or polymeric dispersants were added to the NP-RTP system, the profile of RTP spectrum was not changed, but the RTP intensity and the pre-irradiation time required to attain a stable RTP signal were increased. Whether the surfactants or polymeric dispersants were added to the system or not, no RTP signal was observed when TlNO3 was used as a heavy atom perturber (HAP), but intense RTP emissions were observed when KI was present as HAP. Recoveries of 95.2%-104% with relative standard deviations of 2.4%-4.0% were obtained in spiked water and soil samples.

The interactions between a fluorescent molecular probe, i. e., [2-(p-hexylamino) phenyl-3, 3-dimethyl-5-ethoxycarbonyl-3H-indole] methyldioctadecylammonium iodide (A) and Triton X-100/heptane/hexanol/water reverse micelle have been investigated by spectroscopy. Micropolarity of the environment, fluorescence anisotropy parameter and the pH effect of A in Triton X-100 reverse micelle were determined. Furthermore, the state of water in reverse micellar systems was studied by FTIR. According to the above experimental results, some information on the structure of Triton X-100 reverse micelle was obtained and the probable site of A in this system was discussed.
