Rice cultivars of Nanjing 9108 with good taste quality was adopted as material under the pure nitrogen condition of 270 kg hm-2 in this study. Rice was cultivated with two methods including mechanical transplanting with pothole seedlings (MT) and mechanical transplanting with carpet seedlings (MC). Controlled-release fertilizer and quick-acting nitrogen fertilizer were mixed at a ratio of 5:5. Controlled-release fertilizer was mixed from four different release periods (40, 80, 100, and 120 days) at a ratio of 1:4 to form three mixing modes (40+80, 40+100, and 40+120). Conventional urea split fertilization (CK) was used as a control, and the effects of different controlled-release fertilizer ratios on yield formation and nitrogen accumulation of high-quality japonica rice were studied. Under the same fertilizer treatment, compared with mechanical transplanting with carpet seedlings (MC), the two-year yield of mechanical transplanting with pothole seedlings (MT) was significantly increased by 3.9% and 4.9%, respectively. The reason was that the transplanting of pothole seedling machine could improve the accumulation of photosynthetic substances in the middle and late stages and obtain larger panicle type with higher grain number per panicle, seed setting rate, and 1000-grain weight. Under the cultivation mode of pothole seedling transplanting and carpet seedling transplanting, compared with CK, the two-year yield of 40+80 and 40+100 controlled-release fertilizer treatments were higher than CK, and the two-year yield of 40+100 controlled-release fertilizer treatments was the highest, which was significantly increased by 7.3% and 9.2%, respectively. The reason was that 40+100 controlled-release fertilizer treatment had higher effective panicle number and population glume amount. Compared with 40+80 and 40+120 controlled-release fertilizer treatments, the nutrient absorption law of 40+100 controlled-release fertilizer treatment was more consistent with Nanjing 9108, which not only ensured the occurrence of early tillering, but also ensured the stable production of photosynthetic substances at later stage, thus obtaining stable panicle number, grains per panicle, and 1000-grain weight. Compared with CK, the nitrogen recovery efficiency, nitrogen agronomic efficiency, nitrogen physiological efficiency, and nitrogen partial productivity of 40+100 controlled-release fertilizer treatments were significantly increased by 10.0%-12.5%, 24.6%-30.5%, 11.4%-18.6%, and 7.3%-9.1%, respectively. In conclusion, the mechanical transplanting with pothole seedlings (MT) had obvious advantages over mechanical transplanting with carpet seedlings (MC), which was a cultivation method that was conducive to high-quality japonica rice. At the same time, the 40+100 controlled-release fertilizer ratio could better meet the nutrient requirements of good-quality japonica rice at various stages, and could obtain high yield, which could be used as a simplified fertilization scheme for high-quality japonica rice. Objective To explore the efficacy and safety of TERVO stent in the treatment of acute ischemic stroke caused by brachial artery disease. Methods This retrospective study included 9 patients from March 2017 to September 2019 who suffered with acute ischemic stroke caused by brachial artery disease. Among them 5 cases suffered with middle cerebral artery (MCA) M2 occlusion, 2 cases with MCA M3 occlusion and 2 cases with posterior cerebral artery (PCA) P1 occlusion. All patients underwent thrombectomy by TREVO stent while 5 of them were also treated with intravenous thrombolysis in the Department of Vascular Intervention of Civil Aviation General Hospital. The vascular recanalization rate and postoperative bleeding rate were observed, meanwhile the National Institutes of Health Stroke Scale (NIHSS) was used before and 14 d after thrombectomy to assess the degree of neurological impairment. And the prognosis was assessed by the modified Rankin Scale (mRS) 90 d after the operation. Results Among the 9 patients, 8 cases successfully completed the operation of thrombectomy, with a success rate of 8/9. mTICI grading immediately after thrombectomy, showed that 6 patients had grade level 3, 2 had level 2b and one had level 2a, while the rate of vascular recanalization was 8/9. Intracranial hemorrhage occurred in one case, massive cerebral infarction in one case and the clinical symptoms of the other 7 patients were significantly improved after treatment. NIHSS score at admission of 9 patients was 9.00 (7.50, 11.00) while who scored 4.00 (2.50, 5.50) 14 d after thrombectomy. mRS ≤ 2 in 7 cases and > 2 in 2 cases were followed up in 90 d after thrombectomy, with a good prognosis in 7/9. Objective: To observe the occurrence time of neuralgia and the expression of purinergic ligand-gated ion channel 7 receptor (P2X7R) in the dorsal horn of the spinal cord after intraperitoneal injection of streptozotocin (STZ) in diabetic rats, and to explore the effect of electroacupuncture (EA) and pretreatment of EA on the heat pain threshold and expression of P2X7R in the spinal dorsal horn in rats with diabetic neuropathic pain (DNP), and to explore the possible mechanism of EA for DNP., Methods: PartⅠ: Thirty male SD rats were randomly selected from 64 male SD rats as the control group; the remaining rats were given intraperitoneal injection of STZ (10 mg/mL) at a dose of 65 mg/kg to establish the diabetes model, and 30 rats were successfully modeled as the model group. The control group and the model group were divided into three subgroups respectively at 7, 14 and 21 days, with 10 rats in each subgroup. Body mass, fasting blood glucose (FBG) and thermal pain threshold were recorded at 7, 14 and 21 days after injection; the expression of P2X7R in spinal dorsal horn was detected by Western blot. PartⅡ: Eight SD rats were randomly selected from 35 male SD rats as the blank group, and the remaining 27 rats were given intraperitoneal injection of STZ (10 mg/mL) at a dose of 65 mg/kg to establish the diabetes model. The 24 rats with successful diabetes model were randomly divided into a DNP group, an EA group and a pre-EA group, 8 rats in each group. Fifteen to 21 days after STZ injection, the EA group received EA at "Zusanli" (ST 36) and "Kunlun" (BL 60), continuous wave, frequency of 2 Hz, 30 min each time, once a day; the intervention method in the pre-EA group was the same as that in the EA group. The intervention time was 8 to 14 days after STZ injection. The body mass, FBG and thermal pain threshold were recorded before STZ injection and 7, 14 and 21 days after STZ injection; the expression of P2X7R in spinal dorsal horn was detected by Western blot 21 days after injection., Results: PartⅠ: Compared with the control group, in the model group, the body mass was decreased and FBG was increased 7, 14 and 21 days after STZ injection ( P <0.01), and the thermal pain threshold was decreased 14 and 21 days after STZ injection ( P <0.05), and the expression of P2X7R in spinal dorsal horn was increased 7, 14 and 21 days after STZ injection ( P <0.05, P <0.01). PartⅡ: Compared with the blank group, in the DNP group, the body mass was decreased and fasting blood glucose were increased 7, 14 and 21 days after STZ injection ( P <0.01). Compared with the DNP group, in the pre-EA group, the heat pain threshold was increased 14 and 21 days after STZ injection ( P <0.05), while in the EA group, the heat pain threshold was increased 21 days after STZ injection ( P <0.01), and the expression of P2X7R in the dorsal horn in the EA group and the pre-EA group was decreased ( P <0.01)., Conclusion: The diabetic neuropathic pain is observed 14 days after STZ injection. EA could not only treat but also prevent the occurrence of DNP, and its mechanism may be related to down-regulation of P2X7R expression in the dorsal horn of the spinal cord.

Objective: To report on a case with severe hemophilia A (HA) due to a large duplication of F8 gene., Methods: Inversion detection, Sanger sequencing, and multiplex ligation-dependent probe amplification (MLPA) were used to detect the mutation in the proband and his mother., Results: The patient, a 7-year-old boy, was diagnosed with severe HA at 8 months. No inhibitor was developed over 150 exposure days. Intronic inversion detection and Sanger sequencing have failed to identify pathogenic variants, while MLPA revealed a large duplication [Ex 1&#95;22 dup (2 copies)] in the proband, for which his mother was a carrier [Ex 1&#95;22 dup (3 copies)]. Large duplications of the F8 gene have so far been found in 24 HA patients, all of whom had a severe phenotype, only one had a history of inhibitors., Conclusion: Large duplications of F8 gene are associated with severe HA. The diagnostic rate for HA may be increased by MLPA.

In this study, the aerosol number size distribution in the range of 10 nm-10 μm was collected from August 16 to October 04, 2019 at Ordos using a wide-range particle spectrometer (WPS). Combined with PM (PM 2.5 and PM 10 ), pollution gases, meteorological data, and the HYSPLIT model, the characteristics and impact factors of new particle formation (NPF) were discussed. The results indicated that there were 19 NPF events during the observation period, which have different effects on diurnal variation in aerosol number concentration in different modes. The NPF events caused a sharp increase in the number concentration of nucleation and Aitken mode aerosols, but had little effect on the number concentration of accumulation and coarse mode aerosols. The temperature, wind speed, and total solar radiation during NPF days were usually higher than those in non-NPF days, and the RH during NPF days was lower. On NPF days, the mass concentrations of PM 2.5 , PM 10 , CO, and NO 2 were lower than those on non-NPF days, while the mass concentrations of O 3 and SO 2 were higher. NPF events were observed in 40.0% of northern air masses and 29.6% of southern air masses. There were significant differences in meteorological elements in different NPF event air mass types. The southern NPF event air mass type had the lowest wind speed and the highest RH, with averages of (2.4±1.5) m·s -1 and (48.8±10.8)%, respectively. The northern NPF event air mass type had the highest wind speed and total solar radiation, with averages of (4.2±1.9) m·s -1 and (664.5±255.6) W·m -2 , respectively. The western air mass type of NPF event had the lowest RH, with an average of (29.8±12.7)%. The formation rates of new particles in the different air mass types of NPF events were similar, ranging from 1.5 to 1.8 cm -3 ·s -1 . The largest growth rate was (12.7±13.6) nm·h -1 in the southern NPF event air mass type, which was 1.2 times and 1.4 times higher than the NPF events of northern air masses and western air masses.

This study aims to analyse the value of CODEHOP RT-PCR in the detection of Flavivirus. According to the amino acid sequences of polyproteins of different flaviviruses published in GenBank, a pair of primers was designed using the CODEHOP method. One-step RT-PCR was used to detect Japanese encephalitis virus strain JEV1201, Dengue virus strain JKD001, and yellow fever virus vaccine YV6161. BLAST analysis and phylogenetic analysis were performed after the RT-PCR products of nucleocapsid genes were sequenced. The results showed that this method could amplify Flavivirus specifically, and the size and sequence of the target fragment accorded with the anticipated result. JEV1201 had the highest homology to Japanese encephalitis virus strain YL2009-4/YC2009-3, belonging to the branch of the phylogenetic tree of Japanese encephalitis virus strains. JKD001 had the highest homology to Dengue virus strain DENV-2/ID/1022DN/1975, belonging to the branch of the phylogenetic tree of Dengue virus strains. YV6161 had the highest homology to Yellow fever virus strain 17D, belonging to the branch of the phylogenetic tree of Yellow fever virus strains. In conclusion, the method of CODEHOP RT-PCR can be effectively used to detect, identify, and phylogenetically analyse Flavivirus.

Objective: To verify the effect of bloodletting therapy at Jing-well points and semen coicis on patients with traumatic cerebral infarction., Methods: Ninety patients were randomized into a bloodletting therapy at Jing-well points group (bloodletting group), a semen coicis group and a comprehensive therapy group, 30 cases in each one. The conventional basic medication was applied in all of the three groups. In the bloodletting group, the bloodletting therapy was done at twelve Jing-well points with three-edged needle, 3 drops of blood required at each one, three times a day. In the semen coicis group, the semen coicis preparation was applied via nasal feeding or oral administration, 90 g each day, three times a day. In the comprehensive therapy group, the bloodletting therapy at twelve Jing-well points and semen coicis preparation were used in combination and the methods were same as the above two groups. After 4 weeks of treatment, the efficacy was assessed with nerve function defectscale (NDS). Fugl-Meyer scale of the upper and lower limb function was used to evaluate the motor function of the affected limbs of the patients before and after treatment., Results: The scores of Fugl-Meyer scale of the upper and lower limb function were increased apparently after treatment in the patients of every group (P < 0.01, P < 0.05). The score increase was much more obvious in the bloodletting group and the comprehensive therapy group as compared with the semen coicis group (all P < 0.01). The result in the comprehensive therapy group was superior to the bloodletting group (all P < 0.05). The total effective rates of NDS in the comprehensive therapy group, bloodletting group and semen coicis group were 96.7% (29/30), 83.3% (25/30) and 76.7% (23/30) separately. The result in the comprehensive therapy group was higher apparently than those in the bloodletting group and semen coicis group separately (both P < 0.05). The result in the bloodletting group was better than that in the semen coicis group (P < 0.05)., Conclusion: The bloodletting therapy at Jing-well points and semen coicis alleviate apparently nerve function defect, improve the motor function of the affected limbs and achieve the better efficacy.

Objective: To identify the genotype and clades of hantavirus (HV) in Zhejiang province., Methods: The partial S and M segment of the HV in Zhejiang province were amplified with RT-PCR using genotype-specific primers, and then were sequenced and compared with other known hantaviruses., Results: The genotype of 11 strains were HTNV and other 7 strains were SEOV by homology and phylogenesis analysis, yet the clade distribution was significantly different among foci of Zhejiang with 5 clades of HTNV and 3 clades of SEOV. There also existed special clade of HTNV named ZNB-1, ZNB-2, A3 and of SEOV named Gou3, ZJ5. The homology of M segments of ZNB-1 and ZNB-2 with other HTNV clades were 69.7%-74.0% except Nc167, A3 with other HTNV clades were 73.6%-76.3% except B78., Conclusion: Zhejiang province is co-circulating with HTN and SEO. Say the least of the clades are 5 of HTNV and 3 of SEOV and there also existed special clade of HTNV and SEOV.

Objective: To study the immunophenotype and its relationship with clinical characteristics in children with acute lymphoblastic leukemia (ALL)., Methods: Bone marrow or blood samples (2-3 mL) with heparin anticoagulation from 139 children with ALL were obtained, and immunophenotypes were identified by flow cytometry., Results: In 139 ALL children, there were 103 cases (74.1%) of B-ALL, 24 cases (17.3%) of T-ALL, 12 cases of T/B biphenotypic (8.6% of T/BALL). In the 103 children with B-ALL, CD19 (90.3%), CD10 (83.5%) and CD20 (27.2%) were expressed as major antigens. In the 24 children with T-ALL, the major antigens were CD3 (79.2%), CD7 (66.7%) and CD5 (33.3%). In the 12 children with B/T-ALL, T-lymphoid antigens included CD7 (50.0%) and CD5 (41.7%), while the B-lymphoid antigens included CD19 (50.0%) and CD10 (33.3%). Of the 139 children with ALL, 32 cases (23.0%) showed myeloid antigen expression (My+ ALL) and the main expression antigens were CD13, CD33, CD14 and MPO. CD34 was expressed in 31 cases. CD34-positive expression (15.6%) in My+ ALL children was significantly lower than in My-ALL children (24.3%). HLA-DR was expressed in 82 of the 139 ALL children. The expression of CD10, CD34 and HLA-DR in the standard-risk, medium risk, high-risk ALL children was significantly different. There were significant differences in gender and incidence of bleeding between the My+ ALL and My-ALL groups (P<0.05)., Conclusions: Immunetyping can differentiate the sources of leukemic cells. The expression of CD10, CD34 and HLA-DR antigen is related to the clinical classification of ALL.

Objective: To study the relationship of methylation of inhibitor of DNA binding 4 (ID4) gene core promoter region with childhood T line, B line and T/B acute lymphocytic leukemia (ALL)., Methods: Methylation-specific polymerase chain reaction (MS-PCR) was used to detect the methylation status of ID4 promoter region in 18 children with newly-diagnosed ALL (2 cases of T-ALL, 13 cases of B-ALL and 3 cases of T/B-ALL). Thirty-four hospitalized children with non-tumor disease served as the control group., Results: The complete methylation rate of ID4 gene promoter region (15/18, 83%) was significantly higher than the partial methylation rate (3/18, 17%) in the 18 ALL children (P<0.05). The complete methylation rate of ID4 gene promoter region in children with T-ALL, B-ALL and T/B-ALL (50%, 85% and 100% respectively) was significantly higher than that in the control group (18%; P<0.05). In contrast, the partial methylation rate and non-methylation rate in the three ALL groups were significantly lower than those in the control group (P<0.05). There were no statistically significant differences in the methylation patterns among the B-ALL, T-ALL and T/B-ALL cases., Conclusions: The methylation of ID4 promoter region may be related to the pathogenesis of childhood ALL. The methylation patterns of ID4 promoter region are identical in B-ALL, T-ALL and T/B-ALL.

Objective: To investigate the biological characteristics of childhood T-lineage acute lymphoblastic leukemia (T-ALL) and their clinical significance., Methods: Immunophenotyping was performed by three-color flow cytometry analysis using CD45 /SSC gating in 23 children with newly diagnosed T-ALL. Meanwhile cytogenetic analysis was performed., Results: CD3(+) expression of T-lineage antigens was apparently higher than CD7(+) and CD5(+) expression. CD19(+) expression of B-lineage antigens was apparently higher than CD22(+), CD10(+) and CD20(+) expression. Myeloid antigen was expressed in 4 cases (17%). CD34(+) and HLA-DR(+) were observed in 4 cases (17%) and 5 cases (22%), respectively. cCD3(+) and cCD79(+) were expressed in 23 cases (100%) and 22 cases (96%), respectively. The chromosome detection in 8 cases with T-ALL showed hyperdiploid or Ph(+) chromosome (one case each). The fusion gene detection in 5 cases showed MLL rearrangements in two cases and positive SIL/TAL1 fusion gene in one case. CD3 expression was related with the complete remission rate., Conclusions: Immunophenotyping is an important tool for diagnosis of T-ALL. However, the immunophenotype of T-ALL is heterogeneous. So, immunophenotyping along with cytogenetic and molecular genetic analysis is needed in the treatment and prognosis evaluation of T-ALL.

Objective: To study the relationship between human parvovirus B19 infection and childhood idiopathic thrombocytopenic purpura (ITP) by the principle of evidence based medicine., Methods: Papers related to the relationship between human parvovirus B19 infection and childhood ITP published between 1994 and 2008 were retrieved electronically from the Chinese Journals Full-text Database and the Wanfang Data. These relevant papers on case-control trials were statistically studied by meta analysis., Results: Eight papers that met the inclusion criteria were included for this meta analysis. Five hundred and sixteen cases of childhood ITP and 246 healthy controls were enrolled. The meta analysis showed that the incidence of human parvovirus B19 infection in the ITP group was significantly higher than that in the control group (OR=13.71, 95% CI=7.07-26.59, Z=7.75, p<0.01)., Conclusions: Human parvovirus B19 infection is closely associated with childhood ITP.

Objective: To investigate the expression of CD147 and matrix metalloproteinase-9 (MMP-9) in children with non-Hodgkin's lymphoma (NHL) and its correlation with clinical stage, tumor size, bone marrow invasion, immunological typing, serum lactate dehydrogenase (LDH) concentration, and prognosis., Methods: Specimens excised from NHL patients were prepared. Expression of CD147 and MMP-9 were tested by streptavidin-biotin complex (SABC) immunohistochemistry and its correlation with clinical results were analyzed., Results: The positive rate of CD147 expression was 73% (45/62), 17 cases were (-), 11 cases (+), 34 cases (++) and 21 cases (+++). The positive rate of MMP-9 expression was 81% (50/62), 12 cases were (-), 13 cases (+), 18 cases (++) and 19 cases (+++). The Spearman rank correlation analysis indicated that there was a positive correlation between CD147 and MMP-9 expressions in NHL (r(S) = 0.763, P = 0.034). Expression of CD147 was determined in relation to factors that included clinical bone marrow invasion, tumor size, LDH level as well as the clinical stage; expression of MMP-9 had a positive correlation with bone marrow invasion, tumor size and clinical phases. The 5-year survival rates (5YSR) were 78% (22/28) and 45% (15/34) in the cases whose CD147 expression was (-)-(+) and (++)-(+++), respectively, and 5YSR were 84% (21/25) and 43% (16/37) in the cases whose MMP-9 expression was (-)-(+) and (++)-(+++) respectively, the difference was significant. Cox multivariate analysis showed that both CD147 and MMP-9 were important prognostic factors., Conclusion: The increased expression of CD147 and/or MMP-9 correlates with a poor clinical outcome in patients with NHL.

Objective: To study the effects of CDDP (cis-dichlorodiamine platinum) on the telomerase of human choroidal melanoma cells and to investigate the toxic effects of CDDP on these cells. To study the relationship between these two effects and to explore the possibility of using CDDP in the chemotherapy of choroidal melanoma., Methods: It was an experimental research. CDDP was added to the culture medium of primary cultured human choroidal melanoma cells at different concentrations (0.01, 0.1, 1, 10 and 100 mg/L, 72 h) and times (12, 24, 48, 72 and 96 h, 10 mg/L) and the results were compared with that of the controls. Toxic effects of CDDP were evaluated by MTT test and the level of telomerase was measured by PCR-ELISA assay. The correlation between these two effects was analyzed., Results: The telomerase activity was inhibited by CDDP time dependently and dose dependently. Cell viability was decreased when the concentration of CDDP attained 0.1 mg/L and acted for 24 hours. The cell toxicity of CDDP was correlated negatively (r = -0.900, P = 0.037) with the inhibition of telomerase. The cell death was lagged behind the decrease of telomerase., Conclusions: CDDP is an effective telomerase inhibitor which can decrease the telomerase activity of cultured human choroidal melanoma cells significantly. This effect is dose and time dependent. CDDP can also cause the death of cultured melanoma cells.

Objective: To evaluate the effect and safety of BCG vaccine on initially treated pulmonary tuberculosis and its controlling effect on multidrug-resistant tuberculosis., Methods: All 360 volunteers with initially treated pulmonary tuberculosis of positive smear and culture were divided into immunotherapy group (180 cases, also BCG group) and control group (180 cases) at random pair. The patients in BCG group were treated with chemotherapy of a regimen of 2HRZ/2HR and immunotherapy with BCG for 4 months,and the first BCG vaccine was given a month after chemotherapy. Meanwhile, the patients in the control group were treated with chemotherapy of 2HRZ/4HR only., Results: (1) The negative conversion rate of sputum smear in BCG group was 98.3% (177/180), and it was 97.2% (175/180) in control group. There was no significant difference between the two groups both at the ends of 4 and 6 months after treatment (chi2 = 0.1278, P > 0.05). (2) The positive conversion rate of sputum smear in BCG group was 2.3% (4/177), and it was 6.9% (12/175) in control group followed up for 5 years. The successful rate was 96.1% (173/180) in BCG group, and it was significantly higher than that of 90.6% (163/180) in control group (chi2 = 4.4643, P < 0.05). (3) In the 5-year follow up, bacteriologic result was similar to that of X-ray. (4) The occurrence rate of multidrug-resistant tuberculosis was 2.3% (4/177) in BCG group,significantly lower than that of 7.3% (13/178) in the control group (chi2 = 4.9513, P < 0.05)., Conclusion: As an adjunct chemotherapy,immunotherapy with BCG vaccine should be helpful for patients with initially treated pulmonary tuberculosis. It would further strengthen the effects of chemotherapy and reduce the occurrence rate of multidrug-resistant tuberculosis.

This study was aimed to investigate the changes of silencer of death domains (SODD), survivin, caspase 3, caspase 8 and caspase 9 in the apoptotic process of human leukemia cells induced by chemotherapeutic drugs in order to explore the molecular mechanism of apoptotic modulatory genes and to search for the new target of chemotherapeutic drugs. After Jurkat cells were induced by chemotherapeutic drugs, the translocated phosphatidylserine was labeled with annexin V/PI, and the apoptosis incidence was measured by FCM; The expression changes of SODD, caspase 3, caspase 8 and caspase 9 were determined by Western blot; the changes of survivin mRNA and protein were determined by RT-PCR and immunohistochemistry SABC method respectively. The results indicated that high expressions of SODD and survivin could inhibit apoptotic signaling pathway; VCR down-regulated the function of SODD protein and effectively induced the apoptosis of Jurkat cells in a time-dependent manner and activates caspase 3 through the death receptor-mediated activation of caspase 8, in which caspase 9 and survivin were not degraded. It is concluded that SODD participates in the apoptotic process induced by VCR which induces the Jurkat cell apoptosis by downregulating expression of SODD protein and priming death receptor pathway. In the apoptotic process, the mitochondrion apoptotic pathway is not trigged.

Objective: To investigate Daxx expression and its clinical significance in children with acute leukemia., Methods: The expression of Daxx protein was detected by immunohistochemical assay in 50 children with newly diagnosed acute leukemia (34 cases of acute lymphocytic leukemia and 16 cases of acute non-lymphocytic leukemia). Twenty children with normal bone marrow were used as the control group., Results: Daxx protein was expressed in 38.0% of 50 children with acute leukemia, which was significantly higher than that of the control group (5.0%) (P < 0.05). The children with acute non-lymphocytic leukemia had significantly higher Daxx expression levels (62.5%) than those with acute lymphocytic leukemia (26.5%; P < 0.05) as well as the control group (P < 0.05). There were no significant differences in the Daxx expression between acute lymphocytic leukemia children and the control group. Daxx protein was expressed in 55.6% of high risk group of acute lymphocytic leukemia but it was not expressed in standard risk group of acute lymphocytic leukemia (P < 0.05)., Conclusions: Daxx expression is abnormal in children with acute leukemia and associated with some clinical features of acute leukemia, suggesting that it may play an important role in the genesis and development of acute leukemia.

Objective: Survivin, a unique member of the inhibitor of apoptosis protein (IAP) family, plays an important role in regulating both apoptosis and cell division. Overexpression of survivin is associated with increased risk of recurrence and poor outcome in cancer patients. This study aimed to investigate the expression of survivin and its location as well as the relationship between cellular location and expression of survivin and the therapeutic efficacy at the cellular level., Methods: The expression of survivin protein was detected by immunohistochemical assay in bone marrow cells from 62 children with acute leukemia and 40 hospitalized children who did not have leukemia (Control group), and in a human acute T lymphocytic leukemia cell line (Molt-4 cells) treated in vitro with daunorubicin (DNR). Cell apoptosis was detected using flow cytometry., Results: Survivin protein was expressed in 41.9% of the 62 children with acute leukemia but in only 5.0% of the Control group (chi(2)=16.66; P < 0.01). The expression rate of survivin was 46.2% in cytoplasm and 53.9% in nucleus in the children with acute leukemia (chi(2)0.3077; P> 0.05). However, the remission rate of patients in whom survivin expression was seen in the nucleus was significantly higher than that in patients in whom survivin was expressed in cytoplasm after chemotherapy. The survivin expression in Molt-4 cells decreased remarkably by DNR treatment in a time and dosage-dependent manner. DNR treatment also induced survivin transllocation from cytoplasm to nucleus and cell apoptosis in a time and dosage-dependent manner., Conclusions: Survivin may play an important role in the development and prognosis of childhood acute leukemia. The different expression pattern of survivin in the cytoplasm and the nucleus may be associated with therapeutic efficacy and prognosis in acute leukemia. DNR may reduce the survivin expression in leukemic cells and induce cell apoptosis.

Objective: To screen the FANCA gene mutation and explore the FANCA protein function in Fanconi anemia (FA) patients., Methods: FANCA protein expression and its interaction with FANCF were analyzed using Western blot and immunoprecipitation in 3 cases of FA-A. Genomic DNA was used for MLPA analysis followed by sequencing., Results: FANCA protein was undetectable and FANCA and FANCF protein interaction was impaired in these 3 cases of FA-A. Each case of FA-A contained biallelic pathogenic mutations in FANCA gene., Conclusions: No functional FANCA protein was found in these 3 cases of FA-A, and intragenic deletion, frame shift and splice site mutation were the major pathogenic mutations found in FANCA gene.

Objective: To better understand and measure the impact from immunization activities over the last 10-years, we conducted a sero-epidemiological study using the remaining blood samples of a nationwide survey on Nutrition and Health in Chinese residents, in 2002., Methods: HBsAg, anti-HBs and anti-HBc in blood samples were tested using enzyme-linked immunosorbent assay(ELISA) reagents and revised by solid phase radioimmunoassay and Abbott ELISA reagents., Results: Among population older than 3 years, the prevalence rates of HBsAg, anti-HBs and hepatitis B virus(HBV) infection were 9.09%, 37.48% and 50.04% but for children between 3 and 12 year olds, they were 5.03%, 45.33% and 29.10% respectively. Among population older than 3 years, the rates of HBsAg and HBV infection in urban area were 4.61% and 43.51%, but the rates in rural area were 9.41% and 56.77% respectively while for children from 3-years to 12-years old, they rates were 2.10%, 20.45% in urban area and 8.25%, 39.22% in rural areas, respectively., Conclusion: Results revealed that since the hepatitis B vaccine was introduced into the expanded program on immunization(EPI) from 1992, the prevalence rates of HBsAg and HBV infection, especially in the children from 3 to 12 years of age, had obviously declined, suggesting that some changes had happened in the epidemic characters of hepatitis B in China.

Objective: To understand the relationship between Demodex and bacteria infection in rosacea (brandy nose), and to find effective means for the treatment., Methods: Cellophane tape was used to detect Demodex on the nasolabial grooves and the face; sebum and tissue on face was scraped and cultured to examine bacteria under microscope. The hospital-made anti-rosacea lotion was used on the affected part two times a day for 7 days., Results: It was found that 193 (74.2%) of 260 cases with rosacea were infected by Demodex and 209 (80.4%) of the patients were infected by bacteria. The overall effective rate of the treatment for rosacea was 73.5%., Conclusion: Bacteria infection in rosacea is an important factor inducing rosacea. The curative effect of the anti-rosacea lotion is good.

Objective: MDM2 is considered a proto-oncogene due to its ability to inhibit P53 tumor-suppressor function. But, evidence showed that MDM2 might have a P53-independent role in tumorigenesis. MDM2 is over-expressed in human sarcoma and carcinoma. Recent studies showed that MDM2 might act as a transcriptional factor to modulate expressions of other genes involved in cell cycle regulation and transformation. In the present study, the investigators hypothesized that MDM2 directly affected NF-kappaB expression and function in a P53-independent manner., Methods: MDM2 was transfected to acute lymphoblastic leukemia (ALL) line EU-4 cells lacking P53 expression and expressing very low levels of MDM2. MDM2 and P65 expression in mRNA level and protein level were detected by Western blot and Northern blot after transfection. Since the expression of E-selectin is P65 dependent, E-selectin promoter-CAT construct and P65 and MDM2 expression plasmids were co-transfected to EU-4 cells. CAT activation was determined with ELISA. The effect of adriamycin (ADM) at the concentrations of 15 micro g/ml, 7.5 micro g/ml, 5 micro g/ml and 1 micro g/ml on MDM2-transfected EU-4 cells and the parent cells was detected by MTT assay., Results: The results showed that MDM2 up-regulated P65 expression at both mRNA and protein levels, and MDM2 increased P65-mediated transactivation of E-selectin promoter. Without P65, MDM2 had no effect on the transactivation of E-selectin. Moreover, MDM2 antisense could not change the transactivation of E-selectin. MTT results showed that the survival rate of MDM2 transfected EU-4 cells was higher than that of parental cells. The results suggested that MDM2 transfection increase drug resistance of EU-4 cells to ADM compared with parent cells., Conclusion: (1) MDM2 up-regulated transcriptionally P65 expression. (2) MDM2 increased drug resistance of leukemia cells to ADM. (3) MDM2 elevated NF-kappaB activity in a P53-independent manner in childhood lymphoblastic leukemia cell line.
