[Objective] The strains that have antagonistic effects on the pathogens of Anoectochilus stem rot screened from biological samples from marine sources, and their biological control capabilities were studied to provide a theoretical basis for the development of corresponding biocontrol bacteria preparations. [Method] A variety of test media were used to isolate and purify strains from coral samples in Dongshan Coral Nature Reserve, Fujian Province through the dilution coating method. The colony confrontation test was used to screen out the strains with obvious inhibitory effect on the pathogen of Anoectochilus stem rot. According to the morphological characteristics of the strain and the sequence analysis of the rDNA-ITS region, its classification status was clarified. Then the plate confrontation test, GC-MS and growth rate method were used to verify and determine the biocontrol ability of the strain against the pathogen of Anoectochilus stem rot. [Result] A strain of Penicillium oxalicum HY181-2 with antagonistic effect on the pathogen Fusarium o-rysporum JXL of Anoectochilus stem rot was isolated. In order to eliminate the competitive influence of colony growth, a small amount of fermentation test was performed on HY181-2 to obtain fermentation extracts. GC-MS analysis results showed that the fermentation extract of strain HY181-2 has antioxidant and antibacterial effects. Meanwhile, the mycelial growth rate method was used to compare and analyze the inhibitory effect of strain HY181-2 fermentation extract and four fungicides on F. oxysporum JXL The laboratory virulence test results showed that, compared with the other four chemical fungicides, the fermentation extract of strain HY181 -2 had not the best inhibitory effect on the hyphae of strain JXL at the same concentration. But with the increase of the concentration, the fermentation extract of strain HY181-2 still had a significant inhibitory effect on the mycelium of strain JXL When the concentration was 100 mg/L, the fermentation extract of strain HY181-2 couldinhibit the hyphae of strain JXL as high as 85. 03 % . [Conclusion] Taking the virulence index as an indicator, the effect of the fermentation extract of strain HY181-2 in the indoor virulence determination test was second only to 10% difenoconazole, with an EC50 value of 2. 72 mg/L However, the fermentation extract of strain HY181-2 was derived from the growth and metabolism of microorganisms. Compared with chemical fungicides, it was safe, healthy and free of pesticide residues, and it can be continuously obtained with the growth of microorganisms. And the strain HY181-2 also had the unique characteristics of rapid growth of biocontrol bacteria If the strain HY181-2 was developed into a cor- responding microbial preparation and applied to actual production, it will not only reduce the use and frequency of chemical pesticides, further reduce pesticide residues, but also reduce the production costs of growers.
