Cardiac stem cells (CSCs) transplantation has been recognized to be effective on the treatment of myocardial infarction (MI), but some techniques still need to be developed in the isolation and culture of CSCs, which is the key problem restricting the clinical application of CSCs. This study was focused on the isolation of Lin - (lineage-negative) Sca-1 + (stem cell antigen-1-positive) CSCs from newborn C57BL/6J mice (0-3 d) by mixed enzymatic-explant isolation in combination with immunomagnetic separation. The digesting time, digesting frequency, incubation temperature, stirring speed, centrifugation time and rotational speed were strictly controlled in the experiment. In order to increase the survival rate of CSCs, the medium changing time and manner were optimized in primary CSCs culture. The percentages of Sca-1 + cells in primary and passage cells were detected by flow cytometry and immunofluorescence staining. The results showed that: (1) the proportion of Lin - Sca-1 + cells within the collected cells could be as high as (85.03 ± 5.60)% after isolation and purification; (2) In vitro culture of Lin - Sca-1 + CSCs grew into spheres on the 5 th day, and over the whole bottom of the dish on the 7 th day. The growth curve showed that the cells were in logarithmic growth phase on the 3 rd day; (3) Immunofluorescence staining data showed that the expression of Sca-1, the CSCs membrane-specific marker, was decreased after subculture, and flow cytometry data showed that the percentages of Sca-1 + cells were (71.82 ± 2.63)%, (58.38 ± 3.70)% and (46.19 ± 4.72)% in passage 1 (P1), P3, and P5 CSCs, respectively. The above results suggest that high purity of Lin - Sca-1 + CSCs can be obtained by enzymolysis combined with immunomagnetic separation method. Moreover, the CSCs culture system is stable. In our experiment, the Sca-1 + CSCs isolation and culture method has been successfully established, and it is simple, stable, effective and reliable. The method can provide a stable methodological basis for the treatment of MI by Lin - Sca-1 + CSCs transplantation.

In order to explore the anti-aging effect of baicalein, female Drosophila melanogaster as a model organism was used to study the effects of baicalein on natural aging model and aging models induced by hydrogen peroxide（H2O2） and paraquat. The bioinformatics approach was used to predict the possible target for the anti-aging activity of baicalein, and the target pathways were identified. The oxidative stress pathway was a focus in experiment. Baicalein at concentrations of 0.04 mg·m L-1 and 0.2 mg·m L-1 extended the mean and maximum lifespans in the natural aging model, and effectively reduced the damages of oxidative stress by H2O2 and paraquat. 31 senescence-related targets together with the oxidative stress pathway were modulated by baicalein. The experiments revealed that baicalein might delay aging process through attenuation of the oxidative stress response by decreasing the reactive oxygen species（ROS）, malondialdehyde（MDA） and oxidized glutathione（GSSG） in Drosophila melanogaster.

Objective: To explore the perioperative airway management for transoral surgical lysis in patients with irreducible atlantoaxial dislocation and posterior atlantoaxial or occipitocervical fixation., Methods: Thirty patients undergoing surgical lysis through transoral approach with oral endotracheal intubation (OEI group) between September 2009 and June 2010 were recruited. And another 31 patients underwent transoral surgical lysis from December 2008 to August 2009 with percutaneous dilatational tracheostomy (PDT group). The time interval from artificial airway establishment to start of operation, the duration of nasogastric feeding, the incidence of respiratory complications and the postoperative length of hospital stay were compared between two groups., Results: The time interval from artificial airway establishment to the start of operation in the OEI group was significantly shorter than that in the PDT group [(39 ± 15) vs (58 ± 16) min, P < 0.01]. The duration for nasogastric feeding in the OEI group was significantly shorter than that in the PDT group [(6.0 ± 1.2) vs (7.9 ± 0.3) days, P < 0.01]. And the postoperative length of hospital stay was also shorter in the OEI group than that in the PDT group [(9.5 ± 1.7) vs (11.8 ± 3.4) days, P < 0.01]., Conclusion: The management of perioperative airway with oral endotracheal intubation may avoid the complications of PDT, improve the utilizing efficiency of operating rooms and shorten the postoperative length of hospital stay.
