Grape downy mildew is one of the major diseases on grapes, which can occur from seedling to fruit maturity, often causing destructive losses in rainy years. To accurately predict the occurrence of grape downy mildew and minimize the hazards of grape downy mildew, based on the agrometeorological data and disease occurrence data during grape growth in 2020, a prediction model for the occurrence of grape downy mildew was developed by 4 machine learning algorithms ( binary logistic regression, support vector machine, decision tree and K-nearest neighbors), and the models were verified using the data in 2021. The results showed that the decision tree model had the best evaluation indicators in the disease prediction model, which the accuracy was 94%, and the precision, recall and F1-score was 91%, 90% and 91%, respectively. The decision tree model's performance was still better than the other models using validation data. Therefore, the decision tree model could be further used to develop early warning systems for grape downy mildew, which should provide technical support and decision-making guidance for controlling it in production. Sulfur is an essential macronutrient for plant growth and development, and sulfate transporter (SULTR) plays key roles in sulfate (SO42-) uptake and transport in higher plants. In this study, 10 SULTR genes (CsaSULTR1;1～CsaSULTR4;1) were identified in cucumber. Phylogenetic analysis showed that SULTRs from cucumber and Arabidopsis could be divided into four groups (GROUP 1～4). Most CsaSULTR genes contained 9～12 introns, and closely related members shared similar gene structures. Promoter analysis revealed that CsaSULTR genes may be involved in responses to hormones and environmental stress. Expression profiling analysis by RNA-seq indicated that CsaSULTR genes have distinct expression patterns in different tissues, and some of them had significantly changed expression in response to downy mildew infection. [Objectives] Brc2-Cys Prx is the subfamily of Prxs, which are members of the enzymatic antioxidant defense system of plants and plays an important role in scavenging reactive oxygen species(ROS) in plants. Cloning and characterization of Brc2-Cys Prx from non-heading Chinese cabbage are significant for research antioxidant defense genes expression patterns of non-heading Chinese cabbage. [Methods] In this study, with non-heading Chinese cabbage inbred resistance 'Suzhouqing' and susceptible inbred 'Aijiaohuang' as experimental materials, Brc2-Cys Prx gene was obtained using RACE technology. A series of bioinformatics software was used to speculate acid sequence and its nucleotide sequence of Brc2-Cys Prx. Under downy mildew infection and different abiotic stresses(NaCl, PEG6000, CdCl2 and ABA), gene expression patterns of Brc2-Cys Prx were investigated by using real-time quantitative RT-PCR in leaves. [Results] The full length CDS of Brc2-Cys Prx gene was 999 bp, containing an 810 bp open reading frame to encode a total of 270 amino acid residues. Meanwhile, the Brc2-Cys Prx protein from non-heading Chinese cabbage had no signal peptide sequence. Prediction of the relative molecular mass of Brc2-Cys Prx was 28.15×103 with the theoretical isoelectric point of 5.87. The two-dimension structure of Brc2-Cys Prx protein mainly consisted of α-helixes and random coils. Brc2-Cys Prx protein contained a PRX_Typ2cys domain and it belongs to thioredoxin superfamily. Homology analysis showed that the homologous species should be mainly belong to the cruciferae, and the Brc2-Cys Prx from non-heading Chinese cabbage had a highly identity with that from Brassica rapa and Brassica napus. Under downy mildew infection, gene expression of Brc2-Cys Prx in 'Aijiaohuang' was up-regulated gradually compared to that in 'Suzhouqing',which was almost always lower than that in 'Suzhouqing'. The mRNA levels of Brc2-Cys Prx in leaves were down-regulated under 0.2 mol·L-1 NaCl and 200 g·L-1 PEG6000 treatments. The mRNA levels of Brc2-Cys Prx in leaves were up-regulated under 150 μmol·L-1 CdCl2 treatment and peaked at 24 h. The mRNA levels of Brc2-Cys Prx in leaves were up-regulated at beginning and down-regulated after 12 h under 100 μmol·L-1 ABA treatment. [Conclusions] Brc2-Cys Prx gene had a different expression between resistant inbred line 'Suzhouqing' and susceptible inbred line 'Aijiaohuang' and it had a higher expression on the 'Suzhouqing' than the 'Aijiaohuang' under downy mildew infection. It can be speculated that the high transcription level of Brc2-Cys Prx is one of the reasons that resistant line has a higher resistance than the susceptible line under downy mildew infection. Brc2-Cys Prx gene has different expression pattern under abiotic stress, presumably due to reasons related to different substances or signaling pathways. [ABSTRACT FROM AUTHOR]

The cultivation and production of cucumber are seriously affected by downy mildew caused by Pseudoperonospora cubensis . Downy mildew damages leaves, stems and inflorescences, and then reduces the yield and quality of cucumber. This review summarized the research advances in cucumber downy mildew, including pathogen detection and defense pathways, regulatory factors, mining of pathogens-resistant candidate genes, proteomic and genomic analysis, and development of QTL remarks. This review may facilitate clarifying the resistance mechanisms of cucumber to downy mildew.
