Objective To investigate the effects of simulated microgravity by rotary cell culture system (RCCS) on expression profiles of long non-coding RNA (lncRNA) in mouse fibroblasts L929 cell line. Methods L929 cells were cultured in vitro and randomly divided into simulated microgravity (SMG) group and normal gravity (NG) group. Each group had three samples, the rotator axis of SMG group was paralleled to the ground rotation, while the rotator axis of NG group was vertical to the ground rotation, and the speed of rotation was consistent for the two groups. The samples of two groups were collected on 7th day of culture and the total RNAs were extracted, labeled and hybridized in sequence. The lncRNA and mRNA were detected by Agilent Mouse lncRNA Chips respectively. Differentially expressed lncRNA were identified and then validated by RT-qPCR. GO and Pathway analysis were applied to determine the functional distribution of these target genes. The integration predictions of the lncRNA and mRNA co-expression had been proposed to refine the functional lncRNA-mRNA relationships. Results There were 238 differentially ex-pressed lncRNAs including 134 lncRNAs up-regulated and 104 lncRNAs down-regulated, and 237 differentially expressed mRNAs including 53 mRNAs up-regulated and 184 mRNAs down-regulated significantly in mouse fibroblasts L929 cell line under simulated microgravity by RCCS. The RT-qPCR showed a high concordance with chip microarray results in 4 differentially expressed lncRNA. GO analysis showed that the differentially expressed lncRNAs were related to the biological processes such as negative regulation of megakaryocyte differentiation and negative regulation of wound healing. Pathway analysis showed that these target genes were related to the signal pathways of systemic lupus erythematosus and TGF-β. The lncRNA-mRNA co-expression networks were also established. Conclusion The simulated microgravity by RCCS could significantly affect the expression profiles of lncRNA and mRNA in mouse fibroblasts L929. The lncRNA target genes prediction and functional enrichment analysis based on gene chip technology may provide the theoretical basis for illustrating the mechanism and management of weightlessness stress injury. Objective To study the effect of cell-mediated immune suppression effect of rocket kerosene (RK) through dermal application in mice. Methods Skin delayed type hypersensitivity (DTH) was used to observe the relation of the RK amount the skin exposed and the cellular immune inhibitory function. Different amount of the undiluted fuel was smeared directly onto the dorsal skin of mice. Mice in negative and positive control groups were treated with acetone. After the last exposure, all the mice except those in negative control group were allergized by evenly smearing with 1% dinitrofluorobenzene (DNFB) solution on their dorsum. Five days after allergy, 1% DNFB solution was smeared onto right ear of all mice to stimulate the allergic reaction. Twenty-four hours after attack, the auricle swelling, spleen index and thymus index in corresponding mice were determined. In the first series of experiments, different dosages of RK were applied once, and the ICR mice were randomly divided into negative control group, positive control group and experimental group (0.5ml/kg.BW×1, 1ml/kg.BW×1 and 2ml/kg.BW×1 group). In the second series of experiments, the certain and same dosage of RK was applied for different times, and the ICR mice were randomly divided into negative control group, positive control group and experimental group (0.5ml/kg.BW×1, 0.5mL/kg.BW×2, 0.5ml/kg.BW×3, 0.5ml/kg.BW×4 and 0.5mL/kg.BW×5 group). In the third series of experiments, the different dosages of RK were applied more than once, and the ICR mice were randomly divided into negative control group, positive control group and experimental group (0.5ml/kg.BW×5, 1ml/kg.BW×5 and 2ml/kg.BW×5 group). Lymphocyte proliferation experiment in vitrowas conducted to observe the persistent time of the cell-mediated immune suppression in mice by RK dermal exposure. The lymphocyte proliferation induced by concanavalin A (Con A) was analyzed by MTT assay, and T lymphocyte subsets (CD3+, CD4+ and CD8+) in peripheral blood and spleen lymphocyte cell cycle were analyzed by flow cytometry. Results RK dermal exposure (1ml/kg.BW×1) alleviated the ear edema, suppressed the spleen index elevation and thymus index reduction caused by DNFB sensitization in ICR mice, and the suppression effect increased with exposed dosage and time increasing. Objective To summarize the clinical and pathological features of pyogenic liver abscess (PLA) in patients with diabetes mellitus (DM). Methods A retrospective study was performed to analyze the clinical and pathological data of 42 DM patients with PLA in 306 Hospital of PLA from January 2001 to December 2010. Results All the DM patients with PLA were cured without relapse and mortality, and the hospital stay was 19-57 days. Thirty-three patients received minimally invasive surgical treatment, including 10 percutaneous needle aspiration (PNA), 23 percutaneous catheter drainage (PCD), and 3 patients undergoing surgical excision, with full and uneventful recovery. Among the 10 patients undergoing PNA, 6 patients were cured after one aspiration procedure, 3 patients after two, and 1 patient after three aspirations. The drainage tubes were removed from patients who received PCD after a mean of 8.6 days. 3-4 days after surgery, the temperature of patients returned to normal, and no complications occurred. The pathological examination revealed numerous fibrous septa in the abscess cavity, inflammatory cells infiltration in the liver tissues surrounding the abscess, and the presence of Mallory bodies in the cytoplasm. The examination also showed that the number of glycogen granule in the hepatocyte was decreased, and the expression of cytokeratin 8 increased (P＜0.05). Conclusion More attention should be paid to DM with the complication of PLA because of its particular clinical and pathological characteristics.

Objective To compare the effects of routine liquid resuscitation on hemorrhagic shock in uncontrolled and controlled states for exploring the strategy of liquid resuscitation. Methods Twenty-eight healthy male SD rats were randomly divided into three groups: control (n=8), controlled hemorrhagic shock (CHS, n=10), and uncontrolled hemorrhagic shock (UHS, n=10). In the CHS and UHS groups, the rats were made to bleed from the femoral artery till the blood pressure declined to 30 mmHg within 15 minutes. Thereafter, the roots of the rat tails in the three groups were cut. The trunks of the tails were ligated to stop the bleeding in the control and CHS groups, but it was not ligated in the UHS group, and no treatment was given. Imitating war condition, the animals were divided into three phases: pre-hospital period (30–90 minutes), hospital period (90–150 minutes), and recovery period (150 minutes to 72 hours). The blood pressure was maintained at 60mmHg in the pre-hospital period by transfusion. The bleeding point was ligated in the hospital period, and the blood pressure was maintained at 90mmHg by blood and fluid transfusions. In the recovery period, the observation time was maintained up to 72 hours. The mean arterial pressure (MAP), central venous pressure (CVP), heart function, blood gas analysis, hematocrit, and blood lactic acid were determined. The amount of bleeding, quantity of infusions, and survival time of animals were observed and recorded. Results Based on the design of the experiment, the MAP of rats in the CHS and UHS groups was maintained at 60mmHg and 90mmHg in the pre-hospital period and hospital period by liquid resuscitation, respectively. There was no significant difference in the MAP and CVP between the CHS and UHS groups. However, the hematocrit of the rats in the UHS group in the pre-hospital period was clearly lower than that in the CHS group. Starting from the pre-hospital period, blood lactic acid content increased continuously in the UHS group, while there was no obvious increase after the hospital period in the CHS group. The speed for increment of heart rate and maximum intraventricular pressure were clearly lower in the UHS group than in the CHS group starting from the hospital period. Acidosis and hypoxemia improved significantly in the CHS group after fluid resuscitation. However, the rats in UHS group were still in the state of acidosis and hypoxemia. The quantity of infusion fluid in the CHS group in the pre-hospital period (44.5±10.1ml/kg) was lower than that in the UHS group (74.5±11.4ml/ kg, PConclusion Compared with the controlled hemorrhagic shock group, rapid liquid resuscitation in rats with uncontrolled hemorrhage can cause an increment of blood loss, hemodilution, damage to cardiac function, and increased mortality.
