To investigate the dynamic change rules of volatile components from Atractylodis Macrocephalae Rhizoma with different stir-baking degrees (from slight stir-baking, stir-baking to yellow, stir-baking to brown, to stir-baking to scorch). In the present experiment, the Atractylodis Macrocephalae Rhizoma samples with different stir-baking degrees were collected at different processing time points. The contents of volatile oil in various samples were determined by steam distillation method, and the volatile compounds were extracted by using static headspace sampling method. Gas chromatography-mass spectrography (GC-MS) and automated mass spectral deconrolution and identification system (AMDIS) were combined with Kováts retention index to analyze the chemical constituents of the volatile compounds. The results showed that with the deepening of the stir-baking degree, the content of volatile oil was decreased step by step in 4 phases, and both the compositions and contents of volatile components from Atractylodis Macrocephalae Rhizoma showed significant changes. The results showed that the dynamic change rules of volatile components from Atractylodis Macrocephalae Rhizoma in the process of stir-baking were closely related to the processing degree; in addition, Atractylodis Macrocephalae Rhizoma and honey bran had adsorption on each other. The method of terminal restriction fragment length polymorphism (T-RFLP) was used to study the genetic diversity of eukaryotic picoplankton (0.2-5.0 microm) in the pelagic and littoral zones in 8 lakes with different trophic status in Nanjing. The objectives of this study were to confirm the difference of the genetic diversity of eukaryotic picoplankton among lakes and the main factors affecting this difference. T-RFLP indicated that there were various fingerprints among lakes and zones. The average terminal restriction fragments (T-RFs) in the littoral and pelagic zones were 16.4 and 15.9, respectively. The littoral zone in Lake Nan and the pelagic zone in Lake Mochou had 30 T-RFs and 27 T-RFs, respectively. The T-RFs were the least abundant (10) in the pelagic zone in Lake Baijia with relatively low trophic status. The genetic diversity of eukaryotic picoplankton was higher in the littoral zone than that in the pelagic zone except Lake Pipa and Mochou. The cluster analysis indicated that the similarities of the littoral zones and the pelagic zones were very high except Lake Baijia, Qian and Nan. The canonical correspondence analysis between the genetic diversity of eukaryotic picoplankton and environmental factors revealed the concentration of chlorophyll a had the most important impact on the eukaryotic picoplankton communities (p = 0.004). The results indicated that the genetic diversity of eukaryotic picoplankton is affected by the trophic status and has the difference in the pelagic and littoral zones.

To investigate the effects of aquatic plants on ammonia-oxidizing bacteria (AOB) at their root zones, four species of aquatic plants were selected, Phragmites communis, Typha angustifolia L., Potamogeton crispus L., and Limnanthemun nymphoides, which were widely used in ecological restorations. AOB in the samples were enumerated by most-probable-number (MPN) method. Nested polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) procedures were performed with ammonia oxidizer-selective primers. Main DGGE bands were excised from the gel and sequenced for phylogenetic affiliation. Results indicate that AOB densities are always higher at the root zones of emergent plants (Phragmites communis 2.8 x 10(5) cells/g and Typha angustifolia L.4.3 x 10(5) cells/g) than those of submerged and floating-leaved plant (Potamogeton crispus L. 9.3 x 10(4) cells/g and Limnanthemun nymphoides 7.7 x 10(4) cells/g). At the root zones, the oxidation-reduction potential is above zero and NH4+ concentration is lower than it in the bare surface sediment. Fourteen major bands were recovered from the DGGE gel, re-amplified and sequenced. Although the identified bands have their respective similar sequences in GenBank, most of them are related to Nitrosomonas-like. This type of bacteria would play an important role of nitrogen cycle in lake sediment after ecological restoration.

Combined effects of estrogenic compounds to high risk life-form were demonstrated, and the study method on combined effects of environmental contaminant mixture and environmental risk assessment were also discussed. A nonlinear aggression was determined on the changes of plasma vitellogenin concentration in crucian carp via the environmental pollutant exposure concentration, and the concentration-response relationship was determined for the single chemical of estradiol, ethynylestradiol, bisphenol A, and octylphenol, and that of the mixed compounds at equipotent concentrations. The combined effects of the four xenoestrogens were tested using indices of mixture toxicity and the biomathematical models of concentration addition or independent action. Weibull function was indicated as the best-fit function (R2 > or = 0.92) with curve estimation, the 95% confidence limits of effect concentration was achieved using the bootstrap method, the media effective concentration (EC50) with 95% confidence interval was 0.007 9 (0.006 8-0.010 0), 0.098 7 (0.090 0-0.111 0), 63.50 (56.58-70.62) and 250.59 (228.46-271.99) Mg x L(-1) respectively. Results suggested that estrogenic chemicals acted together in an additive manner and the combined effects can be accurately predicted in whole range of exposure concentration by the concept of similar joint action or concentration addition.It is proved as a scientific and feasible method to apply the model of joint action whereas the outcome of indices with a great deal of indetermination.

The methods of DGGE and cloning/sequencing were used to study the diversity and community structures of small planktons (0.8 - 20 microm) in different areas of Lake Taihu. DGGE indicated that there were markly various fingerprints in different areas and the diversities were higher in areas with low trophic status than those with relatively high trophic status. There were 23, 24 DGGE bands in East Taihu and Gonghu Bay, respectively (Shannon index were 3.135 and 3.178) and 18 bands in both Meiliang Bay and Wuli Bay (Both shannon index were 2.890). The result of cloning/sequencing indicated that there was a high diversity of small planktons in Lake Taihu and most of them phototrophic flagellate, heterotrophic flagellate, ciliate and fungi. There were various community structures in the three different clone libraries. In Meilang Bay, 28.6% OTUs(operational taxonomic unit)belonged to heterotrophic flagellate, followed by Cryptophyta (22.9%) and Chrysophyta (14.3%). In Central Lake, 25.7% OTUs belonged to Chrysophyta, followed by heterotrophic flagellate (20.0%) and Cryptophyta (14.3 %). In East Taihu, ciliates were the dominant group and only a few heterotrophic flagellates (40.9%) were detected. In addition, fungi were relatively abundant (12.2%) in this area.

Aim: To study the effect of proanthocyanidins on the COX-2 enzyme activity and COX-2 protein expression in LPS-induced RAW264.7 cells., Methods: After being pretreated with different concentrations of proanthocyanidins for 30 min, and then 1 mg x L(-1) LPS for 9 h, the effect of proanthocyanidins on the activity of COX-2 enzyme in RAW264.7 cells was analysed by radioimmunoassay (RIA). After being pretreated with different concentrations of proanthocyanidins for 30 min, and then 1 mg x L(-1) LPS for 9 h, the effects of proanthocyanidins on the expressions of COX-2 mRNA and protein in RAW264.7 cells were analysed by RT-PCR and Western blotting., Results: The activity of COX-2 enzyme was not inhibited by proanthocyanidins (0. 8, 4 and 20 mg x L(-1), P > 0.05 vs LPS group), but the activity of COX-2 enzyme was significantly inhibited by 10 micromol x L(-01) NS-398 (P < 0.01 vs LPS group). The expression of COX-2 mRNA was inhibited by proanthocyanidins (0. 8, 4 and 20 mg x L(-1)). The expression of COX-2 protein was inhibited by proanthocyanidins (4 and 20 mg x L(-1))., Conclusion: Proanthocyanidins had no effect on the activity of COX-2 enzyme in LPS-induced RAW264. 7 cells. Proanthocyanidins inhibited significantly the expression of COX-2 mRNA and protein in LPS-induced RAW264.7 cells.

Background & Objective: Nasopharyngeal carcinoma (NPC) is a common malignant tumor in southern China. Radiotherapy is a major therapy for NPC in early phase,while proper chemotherapy is necessary for NPC in late phase. Some chemotherapeutic drugs have been used to treat tumors by inducing apoptosis of tumor cells. This study was to investigate effects of curcumin on proliferation,and apoptosis of NPC cell line CNE-2Z., Methods: CNE-2Z cells were treated with different concentrations of curcumin, inhibition rates of cell proliferation,and IC(50) of curcumin were detected by MTT method. Cell apoptosis was analyzed by flow cytometry (FCM), agarose gel electrophoresis,and Hoechst33258-PI fluorescence staining., Results: Proliferation of CNE-2Z cells was inhibited obviously by curcumin in dose- and time-dependent manners. IC(50) of curcumin to CNE-2Z cells at 24,48,and 72 h were (24.05+/-0.47),(19.20+/-0.17),and (7.35+/-0.50) micromol/L, respectively. After treated with 5,10,and 20 micromol/L of curcumin for 24 h, apoptotic rates of CNE-2Z cells were (4.9+/-3.2)%,(10.7+/-2.7)%, and (14.7+/-0.5)%, respectively. After treated with 10,20 micromol/L of curcumin for 24 h, morphologic changes, such as chromatin shrinkage,nuclear condensation,and chromatin fragmentation,were observed in CNE-2Z cells by fluorescent staining, a fragmented DNA ladder was detected by electrophoresis., Conclusion: Curcumin may induce apoptosis, and inhibit proliferation of CNE-2Z cells.

False smut is an increasingly important rice disease in recent years, caused by a pathogen, Ustilaginoidea virens Cooke, Takahashi. The pathogen fungus grew well in PD medium, and could produce toxins, ustiloxins, which strongly inhibited the growth of radicle and embryo of plants. The rough ustiloxins were extracted with 100% methanol from the culture filtrate in the study. The antiserum against Ustiloxin A, the main component of the toxins, was raised by immunizing New Zealand rabbit. Elisa assay showed that the titers of the two antiserums obtained were 1: 20 000 and 1: 6 000, respectively. The specificity of the antiserums was also confirmed by Immuno-gold labeling.
