This experiment is to produce the human mAAT(modified anti-trypsin) which cures the emphysema specifically through mammalian galactophore of transgenic goat. 56 goats were selected as donor for superovulation by FSF + LH microinjection in this experiment. The pronucleic embryos were injected with human mAAT gene after fertilization in vivo, and transferred to the donors or receptors directly. The superovulation was better in March and May than in December with the number of ovulation of 19.50, 21.70 and 16.06, and number of fertilized embryos of 4.31, 6.48 and 3.57 per-animal respectively. The pregnant rates were 18.18% and 25% respectively after transferred to donors and receptors with natural estrus. The donors also can be used as the embryo receptor with no remarkable decrease of pregnant rate. 29 lamb were labored. 4 positive transgenic lamb were checked by PCR, PCR-Southern and Southern analysis. The integrated efficiency of foreign DNA was 13.79% with microinjection of high copy number of foreign DNA fragment.

The pH values of the mobile phase which affect the chromatographic behavior of organic acids in RP-HPLC have been studied. It was considered that the problem of organic acids on an RP-HPLC column was dissociation. The equation for finding the optimum pH of mobile phase was pHlimit of colunm < or = pH < or = pKa -2. In addition the effects of contents of methanol and acetonitrile in mobile phase were studied. By using the formula the mobile phases used for the analysis of organic monobasic acids--formic, acetic, propionic and butyric acid and of organic dibasic acids--propane diacid, butane diacid, pentane diacid and hexane diacid were proposed. For the analysis of the monobasic acids, the wavelength of UV-detector was set at 210 nm and the mobile phase was a mixture of phosphate buffer solution(15 mmol/L, pH 2.1)-methanol(85:15, V/V) and at a rate of 1.5 mL/min and the column was Diamond C18, 150 mm x 4.6 mm i.d.. The average coefficient of correlation was 0.9997 and RSD was less than 0.90%. The mobile phase for organic dibasic acids was a mixture of phosphate buffer solution (20 mmol/L, pH 2.1)-methanol(75:25, V/V). The other conditions were the same as for organic monobasic acids. The average coefficient of correlation was 0.9998 and RSD was less than 0.70%.

The reverse phase suspension and embedment technique were adopted to prepare magnetic dextran microsphere (MDMS). The dispersion medium was mixture of some organic solvents. Span-80 was used as stabilizer. The aqueous dextran with magnetic fluid was suspended in dispersion medium with epichlorohydrin as cross-linking reagent. The mixture was stirred for 30 minutes at room temperature and then heated at 70 degrees C for 4 hours, MDMS was thus obtained. MDMS was activated by epichlorohydrin on which 6-aminohexanoic acid, glycine or ethylene diamine was bonded as spacers. Then it was coupled with p-aminobenzamide, L-arginine methyl ester or guanidohexanoic acid and five magnetic affinity adsorbents were prepared. The MDMS was polydisperse particles with the size of 50-300 meshes and the content of Fe3O4 was about 6.2 per cent in the MDMS. Influence of some parameters such as viscosity and density of organic phase, the volume ratio of organic and aqueous phase, the quantity of surfactant and stirring speed on preparing MDMS was studied. Magnetic affinity adsorbents were used to purify crude urokinase in a bath mode and the effect of coupling reagents and ligands on results of purification was discussed. The bioactivity recovery was 40.0 to 60.7 per cent, the purification-fold was between 14.9 and 32.8, and the adsorptive capacity varies from 89 mg to 121 mg per milliliter of adsorbent.

Two kinds of affinity chromatographic packings for the separation of urokinase were synthesized by coupling of p-amino benzamidine (p-ABZ) to commercially available sepharose and polyepoxypropyl methacrylate (PEPMA). Then they were applied for separating crude urokinase. It was found that the average recovery of bioactivity on sepharose was higher than that on PEPMA, resulting in 108.3% and 43.4% respectively. The high rigidity of PEPMA permits fast flow of protein solutions and operation by higher-pressure affinity chromatography. The average purification times were 36.9 folds for PEPMA column and nine-folds for Sepharose column. The purification of crude urokinase described in this paper demonstrates that PEPMA column is effective for purifying biological products in a large scale.

Objective: To ascertain the frequency of prothrombin(F II) gene 3'-untranslated region(3'-UT) G20210A variant and to explore whether this mutation is related to arterial thrombosis in Chinese people., Methods: We studied 49 patients with thrombosis and 46 healthy Chinese persons. The patients had a first ischemic stroke. The G20210A mutant allele of the prothrombin gene in all blood specimens was investigated by DNA extraction, polymerase chain reaction amplification, Hind III digestion and polyacrylamide gel electrophoresis (PAGE)., Results: The patients and normal control subjects were all homozygous for the normal G20210G allele, and there was no any F II G20210A variant., Conclusion: Factor II gene 3'-UT G20210A mutant allele is absent in Chinese patients with ischemic stroke and normal subjects; its mutation may not be a major risk factor for thrombogenesis in Chinese people.

There were 2,471 school children suffering from enterobiasis in six primary schools of Tainan city. The infected children in five primary schools, Po-Ai, Yung-Hua, Fu-Hsiao, Pao-Jeng and Hsin-Nan, were treated with one single dosage of 100 mg mebendazole. In Shih-Men primary school, the sixth, the students were given placebos as a control group. Negative conversion rates of infected children were examined after three weeks of chemotherapy and school children in the six primary schools were surveyed for enterobiasis two months after chemotherapy to obtain infection rates. The method of examination was two consecutive-day adhesive cellophane perianal swabs. With the purpose of evaluating the efficiency of treatment, positive reduction rates were used and calculated according to the infection rates of school children gained before and after chemotherapy. Those rates in Pao-Jeng, Fu-Hsiao, Yung-Hua, Po-Ai and Hsin-Nan were 62.1%, 47.8%, 41.8%, 37.1% and 3.3%, respectively, and in Shih-Men 3.9%. Judging the data obtained from each grade or each class of schools showed that the efficacy of chemotherapy in reducing the rate of infection was variable. Hence, one single dose of mebendazole and education on personal hygiene were not sufficient to reduce the prevalence of enterobiasis in primary schools. This was because the cycle of E. vermicularis was relatively short, cutting out the routes of transmission was very difficult, and the factors involved were very complex. Overall, though the positive reduction rate presented in Hsin-Nan primary school showed nearly no success in reducing the infection, the other four schools showed valuable rates. Whether continue a treatment, of about three-months in duration, can inhibit the prevalence of enterobiasis among children in primary schools or not will need further study.

A survey was conducted for enterobiasis, in six primary schools at Tainan city. The survey was conducted in 1986, involving 8,120 school children. The manner of testing, was two consecutive day, adhesive cellophane paper perianal swabs. The test resulted in general infection rate of 30.4%. The positive rates among those primary school children were significantly different (chi 2-test, p less than 0.001). The highest rate (40.3%), was in Shih-Men primary school and the lowest rate (18.7%) in Pao-Jeng primary school. Gender occurrence of enterobiasis was determined by making a statistic analysis between sex. We found significantly higher occurrence in males (32.4%) as opposed to females (28.3%) (chi 2-test, p less than 0.001). The infection rates were also significantly different by grade (chi 2-test, p less than 0.001). The highest rate was found in the lower grades (35.3%), the next in the middle grades (32.4%), and the lowest in the higher grades (21.6%). A questionnaire demonstrated that the family status and personal hygiene are the most important factors in transmission of pinworm infection, among the group surveyed. Moreover, the educational levels, occupations of parents, and facilities of bathrooms, (between both primary schools) were also found significantly associated with the transmission of pinworm infection. Therefore, we believe that these factors play a role in transmission of pinworm infection among school children.
