Objective To investigate the connotation and characteristics of curriculum ideology and politics, and to summarize the practice framework of curriculum ideology and politics in the teaching of Surgery in higher vocational students. Methods Students major in clinical medicine from 4 classes in academic year of 2022 were randomly divided into the study group and control group. The literature research method based on educational network and academic journals was adapted to in the teaching of curriculum ideology and politics in the control group. The action research method was used in the teaching of curriculum ideology and politics in the study group. The teaching efficacy was evaluated and measured between two groups. Results The students satisfaction with teaching was 93. 24%(69/74)in the control group and 98. 80%(82/83)in the study group. The number of questions answered was 1324 in the study group and 983 in the control group. Conclusion The curriculum ideology and politics by using action research method is effective to improve the independent learning of higher vocational students and teaching quality of Surgery. Objective To investigate the impact of long non-coding RNA (lncRNA) FGD5-AS1 on the osteogenic differentiation of human bone marrow-derived mesenchymal stem cells (hBM-MSCs) through regulation of the microRNA miR-93-5p/bone morphogenetic protein-2(BMP2) axis. Methods hBM-MSCs in logarithmic growth phase were taken, and the expression levels of lncRNA FGD5-AS1, miR-93-5p and BMP2 mRNA were detected before and after osteogenic differentiation; The cells were transfected or co-transfected with pcDNA FGD5-AS1, miR-93-5p inhibitor, miR-93-5p mimics and corresponding negative controls, respectively, then divided into pcDNA NC group, pcDNA FGD5-AS1 group, inhibitor NC group, miR-93-5p inhibitor group, pcDNA FGD5-AS1+mimics NC group, or pcDNA FGD5-AS1+miR-93-5p mimics group and non-transfected cells were taken as blank group. CCK-8 assay was applied to detect cell proliferation ability of each group; Alkaline phosphatase (ALP) kit was applied to detect its activity; Alizarin red staining was applied to identify cellular mineralized nodule formation; Western blot was applied to detect the levels of BMP2, osteogenesis-related markers-osteocalcin (OCN), osteopontin (OPN), and osterix(OSX); Dual-luciferase experiment was applied to verify the targeting relationship of miR-93-5p with FGD5-AS1 and BMP2, respectively. Results lncRNA FGD5-AS1 and BMP2 were found to be targets of miR-93-5p. After osteogenic differentiation, the expression of FGD5-AS1 and BMP2 was increased and the expression of miR-93-5p was decreased (P＜0.05). Compared with pcDNA NC group, the expression of FGD5-AS1 in pcDNA FGD5-AS1 group was significantly increased(P＜0.05), indicating successful transfection; Mineralized nodules, cell proliferation, ALP activity and the expression of BMP2, OCN, OPN and OSX were obviously higher (P＜0.05) in pcDNA FGD5-AS1 group. Compared with the inhibitor NC group, the expression of miR-93-5p in miR-93-5p inhibitor group was significantly decreased (P＜0.05), indicating successful transfection; Mineralized nodules, cell proliferation, ALP activity and the expression of BMP2, OCN, OPN and OSX were obviously improved (P＜0.05) in miR-93-5p inhibitor group. Over-expression of miR-93-5p inhibited the promoting effect of FGD5-AS1 on the osteogenic differentiation of hBM-MSCs(P＜0.05). Conclusions Up-regulation of FGD-AS1 promotes the osteogenic differentiation of hBM-MSCs, which might be related to the miR-93-5p/BMP2 axis.

The deuterium-tritium fusion reaction is the fastest commercially achievable artificially controlled fusion reaction. However, tritium, as a fuel and neutron radiation-activated material, poses radioactive safety challenges. To simulate the migration of tritium and radioactive dust in the environment under accident conditions, a dispersion simulation program called ACCTRI (ACCidental model for TRItium release) has been developed based on the modified Gaussian multi-puff model, considering dry and wet deposition and tritium re-emission effects. The concentration and dose results calculated using ACCTRI are very close to those calculated using UFOTRI and HotSpot, with a maximum difference of no more than one order of magnitude. A comparison with the experimental results reveals that the results of ACCTRI with tritium buoyancy correction are close to the experimental data. ACCTRI has good accuracy and can be used as a reference for environmental radioactivity safety in fusion reactor site selection and hypothetical accident analysis.

ObjectiveTo learn about the fertility level and gender changes of household registration in Qingpu District from 2012 to 2020， and to provide a theoretical basis for the population development planning.MethodsBirth information of newborns with household registration in Qingpu District from 2012 to 2020 was collected through the Shanghai birth medical certificate information system and the public security department. Access 2003 software was used to combine the data，and Excel 2010 software for statistical analysis.ResultsThere were 28 284 live births in household registered in Qingpu District from 2012 to 2020. The crude birth rate was 6.57‰，and the total fertility rate was 30.29‰. Comprehensive fertility rate was 1.12. The sex ratio at birth was 105.49.ConclusionThere is a low fertility level in Qingpu District. The proportion of the second child increased year by year but decreased after 2017. Negative population growth is intensified year by year.

During the development of oilfield, the produced water with high salinity is seriously corroded to pipeline equipment. Owing to the good corrosion resistance, amorphous Ni - W coating applied on the surface of the pipeline connector can improve the anti - corrosion performance of connector. In this work, the corrosion behaviors of amorphous Ni - W coating and L245 steel in sulfur - containing NaCl solution were studied through immersion and electrochemical methods. Additionally, the surface morphology, composition, and structure of Ni - W coating and L245 steel were analyzed by scanning electron microscope (SEM), energy spectrum (EDS) and X - ray diffractometer (XRD), and the corrosion principles of amorphous Ni - W coating and L245 steel were studied through electrochemical tests. Results showed that sulfide ions would accelerate the corrosion of the L245 steel, and the amorphous Ni - W coating could effectively improve the corrosion resistance of L245 steel in the sulfur - containing NaCl solution. When the sulfide ion concentration was 0.001 0 mol/L, a passivation film formed on the surface of amorphous Ni - W coating, which slowed down the corrosion. When the sulfide ion concentration was 0.005 0 mol/L, Cl- and S2- could work together, resulting in a pitting corrosion on the surface of the amorphous Ni - W coating.

Karst has an important impact on the safe construction and operation of tunnels in the intercity railway survey in limestone areas. Therefore targeted high-precision geological exploration is very important. In order to reduce the impact of complex urban environments and improve the accuracy and efficiency of intercity railway karst exploration, statistical analysis is conducted on the size of actual karst caves detected, and the karst detection effects of four methods including cross hole seismic CT, drilling, opposing coils transient electromagnetics method (OCTEM), and microtremor exploration in a 200 m long experimental area are compared and analyzed based on the comparison of theoretical and practical detection effects of a certain intercity railway. The detection effects of different hole spacing on cross hole seismic CT are studied, the resolution of cross hole seismic CT detecting karst caves, the principles for interpreting the velocity of cross hole seismic CT in specific areas, and the exploration effects of OCTEM and microtremor exploration in the absence of conditions for conducting cross hole seismic CT are studied. The research results indicate that in cross hole seismic CT work, the quality of the original data with a 15 m hole spacing is higher than that with a 30 m hole spacing, and the karst identification rate and resolution are higher. The determination of the scale and spatial location of karst development is more accurate, and the estimation of grouting and grouting dosage in later construction is more reliable. Cross hole seismic CT with a spacing of 15 m can accurately reveal karst caves above 2 m. In specific areas, the development of karst in limestone areas can be quantitatively explained as weathering layers, dissolution cracks, karst caves, and relatively complete limestone based on the magnitude of seismic wave velocity. When cross hole seismic CT working conditions are not available, conducting OCTEM and micro motion supplementary surveys can identify larger karst caves and qualitatively determine the degree of karst development in limestone areas. The survey method has the characteristics of high survey accuracy and is not easily affected by the environment along the intercity railway. Flame retardant expandable polystyrene foams were prepared by coating with melamine modified urea formaldehyde resin. The flame retardant system was based on ammonium polyphosphate, and the combination of three kinds of two-dimensional layered material and zinc borate was used as synergist. The influence of different flame retardants on fire resistance, smoke suppression and thermal stability was discussed. The experimental results show that the flame retardancy is significantly improved by the presence of expandable graphite and zinc borate with mass ratio of 2:1 and total additive amount of 24 phr. The limiting oxygen index of EPS foams is increased to 32.6%, with V-0 rating in UL-94 vertical burning test. The smoke density grade is decreased to 27.31. Compared with the sample containing only expandable graphite, the introduction of zinc borate enhances the strength of residue char from 14.3 to 86.1. Thermal analysis suggests that the thermal stability and char formation are improved by the presence of synergist. The residual char observation reveals that zinc borate is beneficial to form complete and integrated char layers with less crack and breakage.

Relying on the actual engineering case of Liucun Tunnel of Nanchong high-speed railway, this paper studies the key process of using 3D laser scanning technology in shield tunnel line and slope adjustment measurement. Firstly, the measurement principle of 3D laser scanning technology is introduced, and the analysis is carried out from four aspects: preparation for the operation, external scanning, data processing and comparison of results. In order to avoid cumulative errors when splicing point clouds, a control point-based station splicing method was adopted to splice the 18 stations into a complete 3D point cloud data of 3. 7 km in length. The tunnel section interception parameters were set according to the line design data, and the tunnel center coordinates were obtained by RANSAC method after denoising using a circle curve fitting method based on orthogonal distance regression. Research shows that the accuracy of the tunnel center coordinates detected by 3D laser scanning technology is better than 10 mm, which can meet the line and slope adjustment measurement of shield tunnels. The results of the study show that the 3D laser scanning technology can not only significantly improve the efficiency of work, but the high-density and high-precision laser point cloud obtained can also provide the basic data for a large amount of subsequent work, which has a broad application prospect. Emergency disposal of water in earthquake-induced dammed lakes must adequately utilize fundamental data for efficient decision making. This paper presents a comparative study of the rapid estimation of downstream flood risks induced by dammed lakes using the Muskingum and Hydrodynamics methods combined with remote sensing, DEM, hydrologic data, and GIS techniques. Both methods were used to calculate the downstream potential impact area from the Tangjiashan dammed lake when the dam-break water level reached 690m. The results from both methods showed that the downstream Beichuan Town would be submerged. The rapidity, accuracy, and complementarities of both methods were evaluated based on the potential impact area results, data requirements, and calculation times. [ABSTRACT FROM AUTHOR]

Objective: To study the effect of kaempferol on the BaP induced apoptosis in human chorionic trophoblast ceUs HTR8/SV neo. Methods: 10 µmol/L BaP were added 0.01, 0.1, 1 µmol/L kaempferol jointly deal with human HTR8/SV neo cells 48 h, observed cells growth under the inverted microscope, Gimesa staining and Hochest33342 fluorescent staining observed morphologic changes, determination of apoptosis rate changes by flow cytometry. Results: (1) Flow cytometry results showed that BaP alone cell viability was (72.58±0.29)%, with 0.01,0.1,1 µmol/L kaempferol the role of cell viability was (84.96±1.34)%, (89.54±1.64)% and (91.01 ±1.26)%, respectively, compared with BaP alone group, the differences were statistically significant (P<0.01). (2) BaP alone can cause cell chromatin condensation, apoptotic bodies and other typical apoptotic morphology, dense hyperchromatic nuclei, fragmentation shape. According to the problem of the endless flowing of data stream of Combat simulation and the affections of concept phenomenon on its classification model accuracy, an incremental learning simple Bayesian classification algorithm based on Weight window is proposed. The algorithm build a weight window on the data stream of equipment combat simulation in order to learn a new classification model with taking full advantage of the historical time data. The goal is to reduce the unlimited inflows of data stream of equipment combat simulation and mitigate the impact of the concept drift on the accuracy of the classification model, thus improving the accuracy of the simple Bayesian classifier model. Numerical experiments also prove that the algorithm is effective and outperforms other similar algorithm counterparts in terms of classification performance, classification accuracy rate and classification. Combat simulation data stream exists a variety of associated between data attributes, so analysis of data stream association rules is designed to find out the hidden relationship. In view of he combat simulation data stream characteristics of large volume data, variable length, this paper studies how to get he frequent itemsets in the the association rules, in order to obtain frequent itemsets from combat simulation data stream within the limited space quickly, in view of space usage shortages of association rules algorithms Lossy Counting, we proposed a kind of the improved frequent itemsets search method (FIBM)based on the matrix, the method use triangular itemsets matrix instead of the trie tree structure in Lossy Couting algorithm, thereby effectively reducing the Lossy Couting algorithm space occupancy rate. HIF-lα shRNA synthesized and empty vector pEZsiRNA6. 1 were ligated by gene engineering technology, and pEZsiRNA6. 1/hHif-lα recotnbinant expression vector was constructed, and PCR and sequencing confirmed the corrections of pEZsiRNA6. 1/hHif-lα recotn-binant expression vector. Further, pEZsiRNA 6. 1/hHif-lα vector was transfected into human esophageal squamous cell carcinoma EC-9706 cell line, and G418 was utilized to select the cell line with HIF-la stable interference, the detection result of fluorescence microscope demonstrated that EC-9706 cells stably transfection with pEZsiRNA6. 1/hHif-lα appeared the green fluorescence. After treatment with CoCl2 for 4h, the result of Western Blot revealed HIF-lα protein levels after chemical induction were markedly reduced in the cells after transfection, while it remained unchanged in the cells transfected with negative plasmid. The article presents a study which examines the ovarian morphologic changes in infertile cows. Researchers use B-mode ultrasound scanner to detect the images of unusual ovaries of infertile dairy cows, including ovary cyst, inactive ovaries and ovarian atrophy. Results offer a diagnostic reference to sterility diseases in cows.

[Introduction] Trenchless horizontal directional drilling is widely used in power cable line project to cross natural or artificial obstacles at present.Multiple cable protection tubes need to be laid successfully at one time in engineering applications. It is defined as multiple pipelines pulling crossing, and it is widely used because of it's excellent economic and environmental protection advantages. However, there is only the calculation method of single pipe pullback force of plastic pipe at present. This paper aims to study the calculation method of pullback force of multiple pipelines. [Method] This paper was based on a 220 kV cable line project in the Pearl River Delta. This project used horizontal directional drilling to cross the national first-class waterway for twice at the same place, and most of the crossing stratum was plastic clay layer. The calculation method of single pipe pullback force was used for analysis in the project, and the change curve of pull back force was measured on site during the implementation. [Result] The comparative analysis between theoretical calculation and measured results shows that: under the condition of plastic clay layer, the measured pullback force of multiple pipelines is 4～6 times that of single pipe with the same weight and envelope diameter. The straight-line pullback of pipelines is very important to reduce the pullback force and implementation risk. [Conclusion] In this paper, the concept of multiple pipelines pulling and the calculation reference value of pulling force in trenchless horizontal directional drilling crossing project can provide a reference for the crossing design and construction of similar projects in the future.

[Introduction] The paper aims to research the settlement regular and mechanism of the ash dam according to the similarities and differences of measured settlement and conventional settlement regular analysing results. [Method] Firstly, the conventional settlement regular and mechanism of shield construction were introduced in details. Secondly, the settlement regular of the ash dam was researched by the measured settlement curve of the tunnel through the ash dam, and the measured settlement, numerical simulation of settlement and conventional settlement regular were comprised and analysed. Finally, the causes of the sudden change of the settlement of the ash dam were analysed, and the settlement mechanism of the ash dam was obtained. [Result] The results show that the stone riprap of ash dam is large and uneven, its cohesive force is poor, and its gap is large, thus lead to shield excavate difficulty and easy to cause over-excavation and under-excavation, which is the reason for the difference between ash dam settlement and general settlement law. After the shield tunneling, all the measuring points are sliding towards the tunnel palm face rapidly, shows that there are some external factors causing formation deformation and mutation, the law of actual measurement is obviously inconsistent with the general law. The recombination of ash dam stone riprap structure causes the stratum to slip to the palm face, the surface subsidence mechanism of ash dam is obtained. The results of this paper also show that the ash dam reinforcement is very important to the shield construction safety. [Conclution] The research results can provide references for other similar projects.

To solve the problems associated with extracting coal underlying the slope between two adjacent open pits and its safety, authors proposed a coordination mining technology to extract the coal underlying the slope between two adjacent open pits by using theoretical calculation and analysis. Authors analyzed the implementation conditions and the bases of the technology, provided a conception and calculation method of stripping ratio of coal underlying the slope, and investigated key techniques such as smooth connecting of mining and stripping engineering positions, reasonable utilization and distribution of dumping space, optimal location of in-pit haulage system development. The technology was applied in two coal open pits of Guodian Mengdong Energy Holdings Co., Ltd. The results show that the technology makes coal underlying the slope between two adjacent open pits be mined economically and safely, enhances the recovery rate of coal resources, eventually maximizes the economic benefits of two adjacent open pits. Chinese patent medicine preparations containing Epimedii Folium and Psoraleae Fructus have been associated with the occurrence of idiosyncratic drug-induced liver injury(IDILI). However, the specific toxic biomarkers and mechanisms underlying these effects remain unclear. This study aimed to comprehensively assess the impact of bavachin and epimedin B, two principal consti-tuents found in Psoraleae Fructus and Epimedii Folium, on an IDILI model induced by tumor necrosis factor-α(TNF-α) treatment, both in vitro and in vivo. To evaluate the extent of liver injury, various parameters were assessed. Lactate dehydrogenase(LDH) release in the cell culture supernatant, as well as the levels of alanine aminotransferase(ALT) and aspartate transaminase(AST) in mouse plasma were measured. Additionally, histological analysis employing hematoxylin-eosin staining was performed to observe liver tissue changes indicative of the severity of liver injury. Furthermore, a pseudo-targeted metabolomics approach was employed, followed by multivariate analysis, to identify differential metabolites. These identified metabolites were subsequently subjected to Kyoto Encyclopedia of Genes and Genomes(KEGG) pathway enrichment analysis. The results showed that at the cellular level, after 2 hours of TNF-α stimulation, bavachin significantly increased the release of LDH in HepG2 cells compared to the normal group and the group treated alone; after the combination of bavachin and epimedin B, the release of LDH further significantly increased on the original basis. Similarly, although the individual or combination treatments of bavachin and epimedin B did not induce liver injury in normal mice, the combination of both drugs induced marked liver injury in TNF-α treated mice, leading to a significant elevation in plasma AST and ALT levels and substantial infiltration of inflammatory immune cells in the liver tissue. Pseudo-targeted metabolomics analysis identified seven common differential metabolites. Among these, D-glucosamine-6-phosphate, N1-methyl-2-pyridone-5-carboxamide, 17beta-nitro-5a-androstane, irisolidone-7-O-glucuronide, and N-(1-deoxy-1-fructosyl) valine emerged as potential biomarkers, with an area under the curve(AUC) exceeding 0.9. Furthermore, our results suggest that the metabolism of nicotinic acid and nicotinamide, as well as the linoleic acid metabolic pathway, may play pivotal roles in bavachin and epimedin B-induced IDILI. In conclusion, within an immune-stressed environment mediated by TNF-α, bavachin and epimedin B appear to induce IDILI through disruptions in metabolic processes.

Grass swale has been widely used in sponge city construction, which can effectively improve the urban ecological environment. To explore the regulation mechanism of runoff in grass swale, runoff scouring experiment was carried out to study the hydrodynamic characteristics of runoff and the distribution of cross-section velocity under the combined conditions of five slopes (1%, 2%, 3%, 4%, 5%) and five scour flows (20, 30, 40, 50, 60 L·min -1 ). With the increases of flow rate and slope, flow velocity, Reynolds number and Froude number all increased gradually, while the Manning roughness coefficient and Darcy-Weisbach friction coefficient decreased gradually. The velocity ( V ) could be expressed as a power function V =0.3387 Q 0.555 S 0.6601 of flow rate ( Q ) and slope ( S ). The variation ranges of Reynolds number and Froude number were 1160.95-6596.82 and 0.17-1.21, respectively. The runoff flow patterns were all turbulent. The flow pattern was greatly affected by the slope. When flow rate and slope were small, they had great influence on friction coefficient. Under the experimental conditions, the Darcy-Weisbach friction coefficient was negatively correlated with Reynolds number. The velocity distribution of cross-section showed symmetrical distribution on both sides of the center. The maximum velocity point was located at the center of water surface. With the increases of flow rate and slope, the velocity contours of cross section gradually became dense and the gradient of the velocity change increased. Our results provide a theoretical basis for the design, application and hydraulic calculation of grass swale in the construction of sponge cities in loess areas, and reveal the runoff regulation mechanism by analyzing the hydraulic characteristics of grass swale runoff.

AbstractObjective: To explore the effect and mechanism of curcumin on human T-cell acute lymphoblastic leukemia (T-ALL) cell apoptosis induced by Mcl-1 small molecule inhibitors UMI-77., Methods: T-ALL cell line Molt-4 was cultured, and the cells were treated with different concentrations of curcumin and Mcl-1 small molecule inhibitor UMI-77 for 24 h. The MTT method was used to detect the cell survival rate after different treatment; According to the results of curcumin and UMI-77, the experimental settings were divided into control group, curcumin group (20 μmol/L curcumin treated cells), UMI-77 group (15 μmol/L Mcl-1 small molecule inhibitor UMI-77 treated cells) and curcumin+ UMI-77 group (20 μmol/L curcumin and 15 μmol/L Mcl-1 small molecule inhibitor UMI-77 treated cells), MTT method was used to detect cell proliferation inhibition rate, Annexin V-FITC/PI double staining method and TUNEL staining were used to detect cell apoptosis, DCFH-DA probe was used to detect cell reactive oxygen species, JC-1 fluorescent probe was used to detect mitochondrial membrane potential, Western blot was used to detect the expression levels of apoptosis-related proteins and Notch1 signaling pathway-related proteins., Results: After the treatment of Molt-4 cells with different concentrations of curcumin and Mcl-1 small molecule inhibitor UMI-77, the cell survival rate was decreased (P<0.05); Compared with the control group, the cell proliferation inhibition rate of the curcumin group and the UMI-77 group were increased, the apoptosis rate of cell was increased, the level of ROS was increased, the protein expression of Bax, Caspase-3 and Caspase-9 in the cells were all increased, and the protein expression of Bcl-2 was reduced (P<0.05); Compared with the curcumin group or UMI-77 group, the cell proliferation inhibition rate and apoptosis rate of the curcumin+UMI-77 group were further increased, and the level of ROS was increased. At the same time, the protein expression of Bax, Caspase-3 and Caspase-9 in the cells were all increased, the protein expression of Bcl-2 was reduced (P<0.05); In addition, the mitochondrial membrane potential of the cells after curcumin treatment was decreased, and the proteins expression of Notch1 and HES1 were reduced (P<0.05)., Conclusion: Curcumin can enhance the apoptosis of T-ALL cells induced by Mcl-1 small molecule inhibitor UMI-77 by reducing the mitochondrial membrane potential, the mechanism may be related to the inhibition of Notch1 signaling pathway.

The aim of this study was to investigate the mechanism of luteolin regulating lipoxygenase pathway against oxygen-glucose deprivation/reperfusion(OGD/R) injury in H9 c2 cardiomyocytes. First, Discovery Studio 2019 was used for the molecular docking of luteolin with three key enzymes including lipoxygenase 5(ALOX5), lipoxygenase 12(ALOX12), and lipoxygenase 15(ALOX15) in lipoxygenase pathway. The docking results showed that luteolin had high docking score and similar functional groups with the original ligand. From this, H9 c2 cardiomyocytes were cultured in vitro, and then the injury model of H9 c2 cardiomyocytes was induced by deprivation of oxygen-glucose for 8 h, and rehabilitation of oxygen-glucose for 12 h. Cell viability was detected by tetrazolium(MTT) colorimetry. H9 c2 cardiomyocytes were observed with a fluorescence inverted microscope, and colorimetry was used to detect the level of lactate dehydrogenase(LDH) in cell supernatant. The results showed that luteolin could significantly protect the morphology of H9 c2 cells, significantly improve the survival rate of H9 c2 cardiomyocytes in OGD/R injury model, reduce the level of LDH in cell supernatant, inhibit cytotoxicity, and maintain the integrity of cell membrane. The inflammatory cytokines interleukin-6(IL-6) and tumor necrosis factor-α(TNF-α) were detected by enzyme-linked immunosorbent assay. Compared with the model group, luteolin can significantly reduce the release of IL-6 and TNF-α. Western blot was employed to detect the protein levels of ALOX5, ALOX12, and ALOX15 in lipoxygenase pathway. After luteolin intervention, the protein levels of ALOX5, ALOX12, and ALOX15 were significantly down-regulated compared with those in model group. These results indicate that luteolin can inhibit the release of IL-6 and TNF-α by restraining the activation of lipoxygenase pathway, thereby playing a protective role in the cardiomyocyte injury model induced by OGD/R.

The Yongding River basin is an important water conservation and ecological barrier area in the Northwest of Hebei Province. Reduced runoff and deterioration of the water environment in this area have become increasingly prominent under the effects of climate change and intensive human activities. Clarifying the chemical characteristics and factors of surface water and groundwater in the upper Yongding River basin can provide data and support for the sustainable use of water resources. Stable isotopes of hydrogen and oxygen ( δ 2 H and δ 18 O) were used to study the sources of surface water and groundwater. Mathematical statistics and hydrogeochemical methods were then used to analyze the regional hydrogeochemical processes and factors of surface water and groundwater. The results showed that precipitation was the main source of surface water and groundwater. Under the effects of natural factors and human activities, the Yang River and Sanggan River basins exhibited significant differences in surface water chemistry. The sub-basins were ranked by ion concentration as follows: Sanggan River>Yang River. The main cation and anions of the Sanggan River basin were Na + , Cl - , and SO 4 2- , while in the Yang River basin, Ca 2+ and HCO 3 - were the most common. The water chemistry of the Sanggan River exhibited greater variation than that of the Yang River. Surface water chemistry were mainly controlled by mineral dissolution and evaporation, but human activities were reflected in different sub-basins. Surface water in the Sanggan River basin was affected by industrial wastewater discharge, while that of the Yang River basin was affected by agricultural production and cities. However, the continuous increase of Cl - and SO 4 2- concentrations, caused by industrial wastewater discharge and acid rain, was the limiting factor for sustainable use of surface water resources. In future, surface water in Sanggan River basin should be used with consideration to the effects of both total salinity and chemical composition of the water, while in Yang River, a focus should be placed on total salinity. The use of surface water resources in accordance with local conditions is an effective measure for the sustainable use of water resources and the restoration of groundwater levels in this region.

Objective: To observe the efficacy differences between acupuncture combined with grain-moxibustion and acupuncture on acute urinary retention after epidural anaesthesia for anorectal diseases., Methods: A total of 60 patients were randomized into an acupuncture combined with grain-moxibustion group and an acupuncture group, 31 cases in each one. In the acupuncture group, acupuncture was applied at Zhongji (CV 3), Guanyuan (CV 4), Qihai (CV 6), Shuidao (ST 28), Pangguangshu (BL 28), Sanyinjiao (SP 6) and Yinlingquan (SP 9). In the acupuncture combined with grain-moxibustion group, grain-moxibustion was given at Zhongji (CV 3), Guanyuan (CV 4), Qihai (CV 6) and Shuidao (ST 28) on the basis of acupuncture. Those who failed to urinate 60 min after the first treatment received the second treatment. 30 min after the second treatment, the lower abdominal symptom scores before and after treatment as well as the bladder residual urine volume after the first urination after treatment were compared between the two groups. In addition, the clinical efficacy and security were evaluated., Results: Compared before treatment, the symptom scores after treatment were reduced in the two groups ( P <0.05), and the score in the acupuncture combined with grain-moxibustion group was lower than that in the acupuncture group after treatment ( P <0.05). The bladder residual urine volume in the acupuncture combined with grain-moxibustion group was (26.71±17.01) mL, which was lower than (35.32±20.76) mL in the acupuncture group ( P <0.05). The total effective rate was 93.5% (29/31) in the acupuncture combined with grain-moxibustion group, which was superior to 71.0% (22/31) in the acupuncture group ( P <0.05)., Conclusion: The efficacy of acupuncture combined with grain-moxibustion is superior to simple acupuncture on acute urinary retention after epidural anaesthesia for anorectal diseases, which is safe and reliable.

Xiaojin Pill, was firstly recorded in Life-saving Manual of Diagnosis and Treatment of External Diseases, with its primitive name of "Xiaojin Dan". Xiaojin Pill is a classic prescription for treating carbuncle and it is the first choice for Chinese medicine in the clinical treatment of hyperplasia of mammary glands. Objective: To evaluate the efficacy of adjuvant endocrine therapy (AET) in breast cancer patients with a positive-to-negative switch of hormone receptor status after neoadjuvant chemotherapy (NAC). Methods: One hundred and six patients who presented with hormone receptor (HR)-positive breast cancer at diagnosis and turned to HR-negative after NAC during December 2000 and December 2013 in Jiaxing Maternity and Child Health Care Hospital were retrospectively identified. Kaplan-Meier analysis and log-rank test were used for univariate analyses of factors related to disease free survival (DFS) and overall survival (OS). Multivariate analysis was carried out using the Cox proportional hazards model in patients with DFS and OS. Results: All the patients were categorized into two groups on the basis of the administration of AET:61 AET-administered patients (57.5%) and 45 AET-naïve patients (42.5%). After a median follow-up of 68 months (range 14-103 months), human epidermal growth factor receptor 2 (HER-2) status, initial clinical stage, pathological axillary lymph node status and the use of AET were identified as the variables affecting DFS and OS (all P <0.05). Patients treated with AET had a significantly improved 5-year DFS rate when compared with that without AET (77.1% vs 53.5%, P <0.05). The 5-year OS of AET-administered patients was also better than that of AET-naïve patients (80.9% vs 71.0%, P <0.05). Cox regression analysis showed that AET-administered or not was the independent predictor for 5-year DFS ( HR =2.096, 95% CI :1.081-4.065, P <0.05). Conclusion: Patients with HR altered from positive to negative after NAC may still gain benefit from AET.

Objective: To predict the suitable distribution of Cornus officinalis in planting regionalization., Methods: Based on early field surveys and the geographic information data of Cornus officinalis specimens in the Chinese Virtual Herbarium( CVH),the 27 climatic variables,6 soil variables and 1 topographic variable in Shaanxi province were downloaded in World Clim and Harmonized World Soil Database, respectively. The Maximum entropy( Max Ent) was used to predict the potential suitable distributions and areas in Shaanxi province using geographic information system( GIS) after all the variables were made correlational analyses., Results: The AUC of Max Ent( 0. 98) showed the modeling process had a very high precision in this running. The statistical analysis of environment variables in highly potential areas demonstrated that Cornus officinalis prefered to grow in areas with average monthly minimum temperature of March( tmin3,- 3. 0 ~ 9. 8 ℃),precipitation of March( prec3,7 ~ 185 mm),annual average temperature( bio1,6. 7 ~ 17. 6 ℃),annual precipitation( bio12,558 ~ 1 817 mm) and altitude( alt,98 ~ 1 620 m). The predicted results showed that the total most suitable planting area of Cornus officinalis was as high as 45 000 km2 and it was divided into four most suitable regions according to climate types and topography in Shaanxi province. The four high suitable planting regions were the humid climate planting regions in the northern slope of Qinling Mountain zone( Ⅰ-1,5 900 km2),the sub-humid climate planting regions in the basin and valley of Shangluo-Danjiang( Ⅰ-2,11700 km~2),the humid climate planting regions in the basin and valley of Hanzhong-Ankang( Ⅱ-1,15100 km~2),and the humid climate planting regions in mountainous regions of Micang-Bashan Mountain( Ⅱ-2,8 400 km~2),respectively., Conclusion: It is suggested that the planting base should be built in the Parts Ⅱ-1,Ⅰ-2 and Ⅰ-1 based on the original cultivation, the better varieties with high yield, good quality and disease resistance should be vigorously promoted the cultivation, and the mechanical picking tools should be researched to reduce the labor cost for fruit gathering. It will give a scientific method to build suitable planting regionalization to improve the quality of medicinal materials in the modernization of traditional Chinese medicine.

Objective: To study the effect of pseudolaric acid B (PLAB) on cell proliferation and cycle of human prostate carcinoma DU-145 cells. method: Its inhibitory effect on the cell growth was measured by MTT method. Characteristics of cell death were determined by Hoechest 33342 staining. The cell cycle was detected by flow cytometry. The expressions of cyclin B1, cyclin D1 and CDK1 were detected by Real time-PCR and Western blot, respectively., Result: PLAB notably inhibited DU-145 cell growth in a dose- and time dependent manner (P < 0.05). Its IC50 values of PLAB for DU-145 cells for 24, 48 and 72 h were 4.53, 2.39 and 2.08 micromol x L(-1), respectively. Having been treated with 5 micromol x L(-1) PLAB for 24 h, the cells showed such apoptosis characteristics as nuclei chromatin condensation and apoptotic body. With the increase in PLAB concentration, the proportion of G2/M phase cells strikingly increased in a dose- and time dependent manner (P < 0.05), meanwhile cyclin B1 and CDK1 showed over-expressions (P < 0.05), and the cyclin D1 showed under-expression (P < 0.05)., Conclusion: PLAB can inhibit the growth of DU-145 cells and induce the cell cycle G2/M arrest, accompanied with the over-expression of cyclin B1 and CDK1, which may be related with its regulation cycle-associated protein degradation.

This study is to investigate the antitumor activity of CIP-36 on multidrug resistant human oral squamous carcinoma cell line (KBV200 cells) in vitro and the possible anticancer mechanisms. MTT assay, Hoechst fluorescein stain, RT-PCR and immunohistochemistry were carried out on KBV200 and KB cells. The growth of many tumor cells was obviously inhibited by CIP-36, especially the multidrug resistant cells KBV200. Obvious apoptosis could be observed in the Hoechst 33342 staining experiments. The results of RT-PCR showed that the levels of p53, p21, caspase-3 and bax mRNA increased, and meanwhile the expression of mdr-1 and bcl-2 mRNA decreased in a dose-dependent manner. The data were significantly different from that of vehicle. The expression of P-gp significantly decreased with the increasing dosage of CIP-36 examined by immunohistochemistry. It can be concluded that CIP-36 could change resistance-related genes and proteins to overcome multidrug resistance in the KBV200 cell line.

Structural modifications were performed with natural product of oleanolic acid to search for novel anticancer drugs. Ten oleanolic acid derivatives were designed and obtained by the reaction of oxidation, acylation or hydrolyzation, etc. The cytotoxic activity of derivatives was evaluated against HeLa, HepG2 and BGC-823 cells in vitro by MTT assay, gefitinib and etoposide used as a positive control. The results showed that compound 5a was particularly active to inhibit HepG2 cells growth, and anti-tumor activity of compound 7 on HeLa cells was significantly stronger than oleanolic acid. They are worthy to be studied further.

Objective: To study protective effect and pathogenesis of complex salvia miltiorrhiza (DanShen) on acute mercury poisoning in rabbits., Methods: Models of acute mercury poisoning was made in rabbits. The effect of complex salvia miltiorrhiza on blood urea nitrogen (BUN), copper-protein (CP), lactate dehydrogenase (LDH), acid phosphatase (ACP) and the malondialdehyde (MDA) in plasma, superoxide dismutase (SOD) in erythrocyte and MDA, SOD in tissues homogenate were observed., Results: The administration of complex salvia miltiorrhiza after mercury injection 0.5 h and 9.5 h, decreased BUN, CP, MDA, LDH and ACP, and prevented the reduction of SOD. Compared with mercury poisoning group, the difference was statistical significant (P < 0.05, P < 0.01)., Conclusion: It is suggested that acute mercury poisoning may result in renal damage but also multiple organ tissues, and complex salvia miltiorrhiza possesses protective effect, through stabilized membranes.

Objective: To investigate the effect of fluoride concentration on the corrosion behavior of Ti-12Zr alloy and Ti-6Al-4V alloy in a simulated oral environment., Methods: Each specimen was put into the acidic artificial saliva (pH = 4.0) with different NaF concentration (0%, 0.05%, 0.1%, 0.2%) and electrochemical experiment was carried out. Then the surface microstructure and compositions of specimen were analyzed by scanning electron microscope (SEM) after immersed in the test solution with constant temperature., Results: The corrosion potential (E(corr)) and polarization resistance (R(p)) values of titanium alloy decreased with the increasing of NaF concentration, while the corrosion current density (I(corr)) values increased with the increasing of NaF concentration in acidic artificial saliva. The E(corr), R(p) and I(corr) values of titanium alloy changed significantly when the NaF concentration was over 0.1% (P < 0.05). The R, between Ti-12Zr alloy and Ti-6Al-4V alloy had a statistically significant difference, and the R(p) of Ti-12Zr alloy was higher than that of Ti-6Al-4V alloy when the NaF concentration was 0.2% (P < 0.05)., Conclusion: The fluoride ions have a negative influence on the corrosion resistance of Ti-12Zr alloy and Ti-6Al-4V alloy, especially in the acidic artificial saliva which contained over 0.1% NaF.

Objective: To evaluate the overall efficacy and transplant-related mortality (TRM) of related and unrelated allogeneic peripheral blood hematopoietic stem cell transplantation (allo-PBSCT) in chronic myeloid leukemia (CML) patients conditioned with fludarabine-busulfan (FB) reduced intensity regimen., Methods: Forty-four patients received FB (Flud 30 mgxm(-2)xd(-1) x 5 d, BU 4 mgxkg(-1)xd(-1) x 3 d) conditioning followed by allo-PBSCT. Of them, 29 patients were transplanted with related donor and 15 unrelated donor (URD). All patients received mycophenolate mofetil (MMF), CsA and MTX for acute GVHD (aGVHD) prophylaxis. 5 mg/kg rabbit-antithymocyte globulin (ATG-Fresenius) was incorporated in 15 URD recipients., Results: All patients were successfully engrafted. The median times to ANC above 0.5 x 10(9)/L in related (RG) and unrelated groups (URG) were 13.7 (9 - 18) d and 13.6 (12 - 17) d, and PLT above 20 x 10(9)/L were 15.3 (9 - 20) d and 14.7 (10 - 26) d, respectively. Two patients in RG. 1 in URG developed graft rejection 5 - 8 months after transplantation. One of the two patients in RG received second transplantation and engrafted. The cumulative incidence of aGVHD and cGVHD were 13.8% (4/29) and 46.4% (13/28) in RG, and were 33.3% (5/15) and 57.1% (8/14) in URG respectively. Two patients in RG relapsed after transplantation, and obtained CR again after donor stem cell infusion (DSI). Median time of follow-up was 34.7 (2 - 73) months. Thirty-four patients were alive and 10 died. The main causes of death were IP, GVHD, graft rejection and infection. The 5-year overall survival (OS) probability was 77.0%, and the disease-free-survival (DFS) was 73.9%, of which, 79.0% and 74.1% were in RG, and 73.3% and 73.3% in URG, respectively., Conclusions: Fludarabine-busulfan based reduced intensity conditioning for allo-PBSCT with either related or unrelated donors is a safe, less toxic and curative approach to CML.

Objective: To evaluate the biocompatibility of polylactic-co-glycolic acid (PLGA) for culturing bFGF gene-transfected bone marrow stromal cells (BMSCs) and assess the feasibility of this cell complex for repairing cartilage defect in rabbits using tissue engineering method., Methods: BMSCs transfected by bFGF gene were cultured on PLGA matrix to assess the biocompatibility of PLGA. The cell complex was then implanted into the cartilage defect in rabbits, and its effect in cartilage defect repair was evaluated by histological observation and immunohistochemical staining., Results: BMSCs transfected by bFGF gene grew normally on PLGA matrix. After implantation, the complex showed good effect for cartilage defect repair in rabbits., Conclusion: PLGA has good biocompatibility with the transfected BMSCs, and the cell complex can be used for repairing rabbit cartilage defect and may potentially serve as a substitute of cartilage autograft.

Objective: To construct a small interfering RNA (siRNA) targeting mouse vascular endothelial growth factor receptor-2 (VEGFR2) and study its serum stability and gene silencing efficiency in vitro., Methods: The synthesized siRNA targeting VEGFR2 (siVEGFR2) diluted in RNase-free water was mixed at a 1:1 ratio with fresh serum and incubated at 37 degrees celsius;. Gel electrophoresis was performed to determine the integrity of siVEGFR2 incubated for different time lengths. Oligofectamine 2000 was used to mediate siVEGFR2 transfection of MS1 cells, and semi-quantitative RT-PCR was used to evaluate VEGFR2 gene silencing effect induced by the siRNA., Results: The naked siRNA incubated in serum underwent gradual degradation with prolonged incubation time and became virtually undetectable after 24 h. Transfection of MS1 cells with siVEGFR2 significantly down-regulated the expression of VEGFR2 mRNA in comparison with the blank group and control siRNA transfection group (P<0.001), while no significant difference was found in VEGFR2 mRNA levels between the latter two groups (P=0.157)., Conclusion: The naked siRNA is unstable in serum and not suitable for direct administration in vivo. The designed siRNA can effectively silence the VEGFR2 gene expression in MSI cells in vitro.

Chromosomal aneuploidy was a notable cytogenetic character in cancer cells. However, the mechanism of aneuploidy aberration is not clear up to the present. In order to probe into the mechanism of aneuploidy aberration of cancer cells, Centromeric dots (Cd) variation on chromosomes of BGC823 and A549 cells were studied by a simultaneous silver staining of both NOR and Cd from centromeric dots variation. The results showed that: (1) Frequency of Cd loss in BGC823 cells was 1.75%, Cd duplication laggard was 0.28%, small Cd was 1.82%, and Cd-NOR amalgamation was 0.95%. Compared to normal embryonic villi cells, frequencies of Cd loss and Cd-NOR amalgamation of BGC823 cells increased significantly (P<0.0125=, and that frequencies of Cd replication laggard and small Cd showed no difference between BGC823 cells and normal embryonic villi cells. (2) Frequency of Cd loss in A549 cells was 2.73%, Cd duplication laggard was 0.49%, small Cd was 1.73%, and Cd-NOR amalgamation was 0.71%. Frequencies of Cd loss and Cd replication laggard in A549 cells were significantly higher than those of normal embryonic villi cells(P<0.0125), moreover frequencies of Cd-NOR amalgamation and small Cd showed no difference between A549 cells and normal embryonic villi cells. We suggested that Cd loss and Cd-NOR amalgamation might be related with aneuploidy formation of BGC823 cells, and that Cd loss and Cd replication laggard might be related with aneuploidy formation of A549 cells.

This paper introduces the technology of recognizing CD4 cell by a computer, which successfully segments the objects from the background through image processing and analyses the four features including geometric features, optical density, color and texture features. The method based on Principal Component Analysis algorithm is used for feature extraction, and a Back Propagation neural network classifier is constructed, from which the CD4 cell can be well recognized. CD4 cell, image segmentation, feature analysis, Principal Component Analysis (PCA), Back Propagation (BP) neural network classifier is constructed, from which the CD4 cell can be well recognized.

To investigate the relationship between interleukin-18 (IL-18) and human acute graft-versus-host disease (aGVHD), 26 patients undergoing allogeneic peripheral blood stem cell transplantation (allo-PBSCT) were included in this study. IL-18 secreted by peripheral blood mononuclear cells (MNCs) was measured by enzyme-linked immunosorbent assay (ELISA) before transplantation and during aGVHD. The results showed that grade I GVHD and grades III-IV GVHD developed in 10 and 5 cases, respectively. The levels in the supernatants of MNCs from patients with aGVHD were significantly higher than those in the cases without aGVHD. The levels of IL-18 were correlated with the severity of aGVHD. It is concluded that IL-18 plays an important role in the development of aGVHD.
