Grape (Vitis vinifera L.) transcriptomes have been extensively profiled in different tissues, at various developmental stages, under biotic or abiotic stresses, and other conditions. However, there is still no user-friendly tool to explore these precious data. A total of 1 019 microarray sample data were downloaded from the National Center for Biotechnology Information Gene Expression Omnibus database (NCBI GEO). Weighted gene correlation network analysis (WGCNA) was conducted. Forty-one gene co-expression modules were identified. Module functional annotation suggested that these modules performed different functions and were associated with experimental conditions ／ phenotypes. Hub genes, which bear important functions, were screened based on intramodule connectivity. Module genes with unknown functions were annotated according to the "guilt-by-association" rationale. Objective: To evaluate the association between the expression of NF-kB inhibitor alpha (NFKBIA) gene and the prognosis and immune infiltration of the tumor microenvironment in patients with skin cutaneous melanoma (SKCM). Methods: GEPIA2 database was used to analyze the differential expression of NFKBIA in SKCM tissues and normal skin tissues. GEPIA2 and Ualcan databases were utilized to analyze the association between NFKBIA expression and SKCM prognosis. TIMER and TISIDB were used to investigate the correlation between NFKBIA and tumor-infiltrating lymphocytes (TIL) and immune regulator genes in SKCM. The association between NFKBIA and subsets of SKCM cells as well as their functional states were analyzed at single-cell level in TISCH and Cancer SEA databases. The paraffin embedded tissues from 14 SKCM patients preserved in Jingmen No.2 People's Hospital were obtained for this study, and immunohistochemical staining was used to detect the NFKBIA protein expression in SKCM tissues and para-cancerous tissues. Results: NFKBIA was lowly expressed in SKCM tissues, and SKCM patients with low NFKBIA expression had a poor prognosis (P<0.05). NFKBIA expression level was positively correlated with the infiltration of B cells, CD8+ T cells, CD4+ T cells, macrophages, neutrophils and dendritic cells (all P<0.01). What's more, the expression of NFKBIA was positively correlated with TIL abundance and immunoregulatory genes (all P<0.01). NFKBIA was expressed in SKCM immune cells and positively correlated with cell differentiation and inflammation in tumor microenvironment (R=0.28, 0.23, all P<0.05). Immunohistochemical staining results demonstrated that the protein expression of NFKBIA was significantly lower in SKCM tissues than that in para-cancerous tissues (35.71% vs 85.71%, P<0.05). Conclusions: NFKBIA has a low expression in SKCM tissues, and it is correlated with immune infiltration in SKCM, which can be used as a prognostic marker and treatment target for SKCM. There are few therapies for fibrosis and the efficacy is limited. Therefore, there is an urgent need to further understand how fibrosis may regress and to identify potential therapeutic targets. In this study, three representative mouse mRNA expression datasets on liver, kidney, and lung fibrosis samples were downloaded from the Gene Expression Omnibus (GEO). The differentially expressed genes (DEGs) were identified using GEO2R and Venn analysis. Gene functional enrichments were performed on the DEGs using Webgestalt online tool. Proteinprotein interaction (PPI) network was generated by STRING database. Hub genes were explored with the CytoHubba plugin, and the ten DEGs with the highest (maximal clique centrality) MCC score in common DEGs of three organs and liver specific DEGs were selected as hub genes. In the study, an integrated analysis ol liver-renal-lung fibrosis datasets based on mouse model was conducted. GSE36066 and GSE97546 were applied lor first round DEGs analysis. As the study not only discovered common DEGs ol fibrosis in three organs, but also investigated the liver fibrosis specific DEGs, another liver fibrosis dataset GSE55747 was involved lor validation. Results showed that 58 common DEGs were identified lor liver-lung-renal fibrosis, and 85 specific DEGs lor liver fibrosis. Functional enrichment demonstrated that the common DEGs were mainly enriched in the immune response and inflammatory response process. While the liver fibrosis specific DEGs were related with the lipid and laity acid metabolic process. Then 20 hub genes were selected with PPI networks. The druggability was further evaluated towards the hub genes so as to provide potential targets. The study provides following conclusions: TYRO BP, FCGR3B, ALOXS AP and CD 14 lend to be die fibrosis therapeutic targets, and CYP8B1 and IJGT2A3 are potentially correlated with non-alcoholic laity liver disease (NAFLD) or non-alcoholic slealo-hepalilis (NASH). While lor genes FCGR\B, CIQB, LY86, and CD53, there is no research evidence to show their direct correlation with fibrosis, and further validation on their functions in the fibrogenesis is needed. Previously 10 differentially expressed new microRNAs (miRNAs) on sunflower rust resistance were predicted and screened by high-throughput sequencing technique and bioinformatics analysis. This paper continiusly foucused on their expressions by qRT-PCR. Results showed that expression levels of Han-miR21, Han-miR25, and Han-miR42 in leaves inoculated Puccinia helianthi race 300 (resistant group B) were up-regulated compared with healthy leaves treated with water (control group A) and the expression levels of Han-miR11, Han-miR34, Han-miR43, Han-miR47, Han-miR67 showed down-regulated. Expression levels of Han-miR25 and Han-miR42 in leaves inoculated Puccinia helianthi race 737 (susceptible group C) were up-regulated, and expression levels of Han-miR11, Han-miR21, Han-miR34, Han-miR43, Han-miR47, and Han-miR67 were down-regulated. Results of real-time PCR were consistent with 80% of the previous high-throughput sequencing results. By software miRanda prediction, a total of 117 target genes were predicted, of which 86 had bioinformatic functional annotations. By miRbase database, these 10 new predicted miRNAs revealed high similarity (62-100%) to miRNAs in Arabidopsis, tobacco, rice and other crops, among which Han-miR42 and Han-miR67 were involved in disease stress response. Expression of 1 target genes of Han-miR21 (which was up-regulated in resisitant group) was down-regulated. While expression of 3 target genes of Han-miR43 (which was down-regulated in resistant group) were up-regulated. Objective To investigate the expression heterogeneity of transforming growth factor-β-induced protein (TGFBI) in glioblastoma (GBM) multiform and the mechanism of its promotion on malignancy of glioma stem cells (GSCs) in vitro. Methods Using The Cancer Genome Atlas (TCGA), Chinese Glioma Genome Atlas (CGGA), REMBRANDT and Oncomine database to analyse the potential associations between grades, subtypes, overall survival (OS) of GBM and the expression level of TGFBI and the correlation between TGFBI and tumor-associated macrophages (TAMs) marker CD11b and CD163. Treating human monocytes cell line U937 with sequential peripheral blood mononuclear cells and IL-4 to get Primed- and M2-TAMs, quantitative real-time polymerase chain reaction (qRT-PCR) were used to detect the expression level of markers for M1 -, M2 - TAMs and TGFBI. Performing immunofluorescent and immunohistochemical stain to reveal the distribution of TGFBI, M2-TAMs and GSCs in specimen of GBM patients, adding rhTGFBI to the culture medium to see its impacts on the sphere formation and invasion capacity of NCH-421K. Results According to the RNA-seq and microarray data of GBM patients in each database, expression level of TGFBI positively correlated to GBM grades and negatively to OS, Furthermore, expression level of TGFBI were strongly correlate with either pan TAMs marker CD11b or M2-TAMs marker CD163, and as the induction of U937-derived M2-TAMs, the expression level of M2-TAMs marker and TGFBI increased accompanied by decreasingly M1-TAMs marker. Also, we found an obvious co-location between TGFBI and CD163, SOX2 in frozen and paraffin sections of GBM specimens. Lastly, we demonstrate that rhTGF86BI could enhance the sphere formation and invasion capacity of NCH-421K in vitro. Objective To analyze the expression and significance of kinesin family member 23 (KIF23) in breast cancer based on databases mining. Methods In this study, the differential expression of KIF23 gene in breast cancer and normal tissues was compared using the Oncomine and GCBI databases, and then its expression level was overall analyzed. The relationship between KIF23 expression level and prognosis survival, including relapse-free survival (RFS), overall survival (OS), distant metastases-free survival (DMFS) and post-progression survival (PPS), in breast cancer patients were analyzed by the Kaplan-Meier Plotter database. Results It was discovered that KIF23 was more abundantly expressed in breast cancer and a total of 39 studies in the Oncomine database were involved in the expression level of KIF23 in breast cancer and normal tissues. The median rank of KIF23 gene was 900.0 in all differentially expressed genes of breast cancer (P＜0.05). The expression levels of KIF23 were higher in 23 invasive and 4 non-invasive samples of breast cancer, the median rank of differentially expressed genes was statistically significant (P＜0.05). KIF23 was highly expressed in breast cancer (587.27) compared with that of normal tissues (90.89) after integrated analysis (P＜0.05), which was also proved in the GCBI. In addition, Kaplan-Meier Plotter database analysis indicated that the high expression of KIF23 was closely associated with poor survival. Further analysis showed that the expression levels of survival indicators in ER (+) group were statistically significant. Conclusion KIF23 is highly expressed in breast cancer and is associated with poor prognosis in patients, which may be important for the diagnosis, treatment and prognosis assessment of breast cancer. Objective To investigate the expression and significance of NFE2L3 in hepatocellular carcinoma (HCC). Methods We selected 44 cases of HCC patients, and the expression of NFE2L3 protein in the HCC tissues and adjacent normal tissues was detected by two-step immunohistochemical method. The relationship between expression of NFE2L3 and clinicopathological features of HCC was analyzed. The differential expression of NFE2L3 in 371 HCC tissues and 51 adjacent tissues from the cancer genome atlas (TCGA) database was also analyzed. Results The positive expression rates of NFE2L3 in the HCC tissues was 70. 45% (31/44), which was higher than that of the adjacent tissues [15. 91% (7/44)], P<0.05. The expression of NFE2L3 in HCC tissues was related to tumor differentiation and hepatitis B virus infection (P<0.05), but was not related to age, sex, cirrhosis, liver function grade and clinical stage of tumor (all P<0.05). The expression of NFE2L3 in the HCC tissues was significantly higher than that in the adjacent tissues from TCGA (log,FC = 0. 750 121 651 , AveExpr = 2. 336 727 847, P<0.01).
