) Objective To explore effective treatments and prognostic factors for diffuse intrinsic pontine glioma (DIPG). Methods Clinical and imaging information and survival data of 14 DIPG patients, treated with radiotherapy combined with temozolomide and nitolizumab or radiotherapy combined with ACT001, were retrospectively analysed at Beijing Tiantan Hospital, Capital Medical University from April 2021 to January 2024. The median progression free survival (PFS) and overall survival (OS) were calculated using Kaplan - Meier survival curves, and multifactorial Cox regression analysis was used to investigate the effects of different factors on PFS and OS. Results The objective response rate (ORR) was 10/14, and the median PFS and OS were 7.83 and 8.30 months, respectively. Multfactorial Cox regression analysis identified the absence of enhancement on baseline imaging as a good prognostic variable for both PFS (RR = 0.052, 95%CI: 0.006-0.416; P = 0.005) and OS (RR = 0.046, 95%CI: 0.005-0.413; P = 0.006), while male (RR = 0.085, 95%CI: 0.009-0.764; P = 0.028), older age (RR = 0.631, 95%CI: 0.423-0.942; P = 0.024), and the absence of symptoms of cranial nerve involvement at the onset (RR = 0.116, 95%CI: 0.017- 0.781; P = 0.027) were also good prognostic variables for OS. Conclusions Female, younger age at diagnosis, cranial nerve involvement at the onset, and enhancement on baseline imaging are risk factors for the survival of children with DIPG. Objective To observe the expression changes of micro RNA-23a(miR-23a) and apoptotic protease-activating factor-1 (APAF-1) in endometrial carcinoma tissues and analyze its clinical significance. Methods 123 specimens of endometrial carcinoma in our hospital from May 2018 to May 2020 were selected as endometrial carcinoma group, and the para cancerous tissue was selected as the normal group. Expression of miR-23a and APAF-1 were examined in endometrial tissues; the relationship between miR-23a and APAF-1 and the clinicopathology of patients and the correlation between the two were analyzed; Kaplan-Meier method was used to analyze the survival of patients with endometrial cancer. Results Compared with the normal group, the expression level of miR-23a in endometrial carcinoma group was significantly increased, the expressions of APAF-1 mRNA and protein decreased significantly (P < 0.05); miR-23a and APAF-1 were significantly correlated with clinical stage, histological differentiation and myometrial invasion (P < 0.05); Pearson analysis showed that miR-23a was negatively correlated with APAF-1 (P < 0.05); Kaplan-Meier curves showed that PFS and OS were significantly higher in the miR-23a low-expression group as well as the APAF-1-positive expression group than in the miR-23a high-expression group and the APAF-1-negative expression group(P < 0.05). Conclusion The expression level of miR-23a is increased and APAF-1 expression is decreased in endometrial carcinoma, both of which correlate with endometrial cancer progression and patient prognosis, are expected to be biomarkers for clinical assessment of disease progression and prognosis in early-stage patients. Background and objective Pulmonary sarcomatoid carcinoma (PSC) is a rare subtype of non-small cell lung cancer (NSCLC), which is featured by low incidence, high malignancy rate, robust aggressive behavior and inferior prognosis. To date, there is no standardized treatment. The aim of this study is to better understand and accumulate more clinical experience of the disease by summarizing the clinicopathological features, diagnosis methods, therapeutic regimen and prognostic factors of PSC. Methods A total of 39 patients with PSC who diagnosed and received treatment in Beijing Chest Hospital from December 2013 to December 2023 were retrospectively recruited, and information including demographic characteristics, clinicopathological features, tumor-node-metastasis (TNM) stage, diagnosis method and therapeutic regimen were carefully collected. Meanwhile, follow-up was conducted. Kaplan-Meier method was used to analyze the prognostic factors of the disease. Results The PSC patients in this study ranged in age from 45 to 76 years old, including 35 males and 4 females. There were no specific clinical manifestations of PSC at initial diagnosis. Among the 39 patients, 20 underwent surgical resection and 19 received palliative chemoradiation or symptomatic supportive treatment. The 1-year and 5-year survival rates were 61.90% and 35.20% respectively. Univariate analysis indicated that family history of carcinoma, primary tumor site, TNM stage, lymph node metastasis, distant metastasis, whether or not received surgical resection, surgical method, treatment regimens, tumor tissue programmed cell death ligand 1 (PD-L1) expression ≥1% and mesenchymal-epithelial transition factor (MET) pathway abnormalities were correlated with the overall survival (OS) of patients (P<0.05). In the subsequent multivariate analysis, lymph node metastasis emerged as the only independent prognosticator in predicting inferior OS (P=0.037). Conclusion PSC is rarely seen in clinical practice and commonly occurs in elder men with smoking history. Tumor tissue PD-L1 expression ≥1% and MET abnormalities may predict inferior prognosis of PSC and lymph node metastasis was determined as the independent prognosticator of PSC. Surgical resection along with adjuvant medical treatment is the cornerstone for early and locally advanced patients, and the clinical utility of molecular targeting therapy and immunotherapy in PSC needs to be further investigated. Objective: To explore the efficacy and safety of PD-1 antibody versus high-dose interferon in the adjuvant treatment of stage IIB-IIID melanoma patients after surgery. Methods: Clinical data of patients with stage IIB-IIID cutaneous and acral melanoma admitted into Nanjing Drum Tower Hospital, the affiliated hospital of Nanjing University Medical School between September 2013 and September 2022 were retrospectively collected. All patients received high-dose interferon (HDI) or PD-1 antibody as adjuvant therapy after surgery. Univariate survival analysis was performed and survival curves were plotted by the Kaplan-Meier method; Log-Rank method was used to analyze and assess whether the differences in RFS, DMFS, and OS between the groups were statistically significant, and prognostic factors were evaluated by univariate and multivariate Cox regression. Results: A total of 91 patients were enrolled in this study with a median follow-up of 31.0 months. The median RFS was 29.2 and 32.3 months in the HDI group and the PD-1 antibody group, respectively, and the difference was not statistically significant (HR=0.90, 95%CI [0.50, 1.64]; P=0.736). The median DMFS and the median OS in the HDI group were 36.0 and 109.2 months respectively, and neither was reached in the PD-1 antibody group (both P>0.05). The most common site of first distant metastasis in both groups was the lung, and there was no statistically significant difference in the incidence of distant metastasis at any site (P>0.05). By univariate Cox analysis, compared with PD-1, antibody HDI reduced the risk of distant metastasis in patients with BRAFV600E/K mutations (HR=10.03, 95% CI [1.10, 91.35]; P=0.041). Subgroup analyses showed that in patients with cutaneous and acral melanoma, the difference in RFS between the HDI group and the PD-1 antibody group was not statistically significant (both P>0.05). The incidence of adverse reactions in the HDI group and the PD-1 antibody group was 83.3% and 79.1%, respectively, most of which were grade 1 or 2. No deaths related to adverse reactions occurred in both groups. Conclusion: In this study, there was no statistically significant difference in clinical efficacy and safety between PD-1 antibody and HDI adjuvant therapy for malignant melanoma patients. Patients with BRAFV600E/K mutations may benefit more from HDI. Background and objective Invasive mucinous adenocarcinoma (IMA) was a rare and specific type of lung adenocarcinoma, which was often characterized by fewer lymphatic metastases. Therefore, it was difficult to evaluate the prognosis of these tumors based on the existing tumor-node-metastasis (TNM) staging. So, this study aimed to develop Nomograms to predict outcomes of patients with pathologic N0 in resected IMA. Methods According to the inclusion criteria and exclusion criteria, IMA patients with pathologic N0 in The Affiliated Lihuili Hospital of Ningbo University (training cohort, n=78) and Ningbo No.2 Hospital (validation cohort, n=66) were reviewed between July 2012 and May 2017. The prognostic value of the clinicopathological features in the training cohort was analyzed and prognostic prediction models were established, and the performances of models were evaluated. Finally, the validation cohort data was put in for external validation. Results Univariate analysis showed that pneumonic type, larger tumor size, mixed mucinous/non-mucinous component, and higher overall stage were significant influence factors of 5-year progression-free survival (PFS) and overall survival (OS). Multivariate analysis further indicated that type of imaging, tumor size, mucinous component were the independent prognostic factors for poor 5-year PFS and OS. Moreover, the 5-year PFS and OS rates were 62.82% and 75.64%, respectively. In subgroups, the survival analysis also showed that the pneumonic type and mixed mucinous/non-mucinous patients had significantly poorer 5-year PFS and OS compared with solitary type and pure mucinous patients, respectively. The C-index of Nomograms with 5-year PFS and OS were 0.815 (95%CI: 0.741-0.889) and 0.767 (95%CI: 0.669-0.865). The calibration curve and decision curve analysis (DCA) of both models showed good predictive performances in both cohorts. Conclusion The Nomograms based on clinicopathological characteristics in a certain extent, can be used as an effective prognostic tool for patients with pathologic N0 after IMA resection. Background and objective The relationship between nutritional status and prognosis of cancer patients has emerged as a hotspot for research. The aim of this study is to explore the application value of the geriatric nutritional risk index (GNRI) in assessing the prognosis of elderly patients with non-small cell lung cancer (NSCLC), and establish a Nomogram to predict the prognosis of elderly patients with NSCLC. Methods The data of patients with NSCLC aged >65 years who were initially treated in the First Affiliated Hospital of Zhengzhou University from January 2016 to December 2019 were retrospectively analyzed. To determine the optimal cut-off value for GNRI, receiver operating characteristic (ROC) curve was constructed, and the patients were divided into high and low GNRI groups. Kaplan-Meier curve and Log-rank test were used to compare overall survival (OS) of the two groups. Univariate and multivariate Cox regression was used to analyze the risk factors for poor prognosis in elderly patients with NSCLC. Nomogram predicting survival in elderly patients with NSCLC was constructed and validated by using R software. Results Statistically significant differences in age, gender, body mass index (BMI), histological type, albumin, treatment methods, neutrophil to lymphocyte ratio (NLR), prognostic nutritional index (PNI), systemic immune-inflammation index (SII) and cytokeratin 19 fragment (CYFRA.21-1) were observed between the high and low GNRI groups (P<0.05). The Kaplan-Meier curve showed a shorter OS in the low-GNRI group. Multivariate Cox regression analysis showed that CYF^A21-1>3.3 ng/mL was an independent risk factor for the development of OS in patients with NSCLC, and GNRI>97.09 was a protective factor [hazard ratio (HR)=0.52, 95% confidence interval (CI): 0.34-0.79, P<0.05]. Patients in the stage IV had a 1.98-fold increased risk of death compared with patients in the stage I (95%CI: 1.02-3.86, P<0.05). The risk of death was 3.58 times higher in patients receiving chemotherapy alone compared with those receiving combination therapy (95%CI: 2.03-6.32, P<0.05). A Nomogram constructed on the basis of GNRI, which predicted the OS of elderly patients with NSCLC with a concordance index (C-index) of 0.70 (95%CI: 0.65-0.76), and the area under the curve (AUC) for 1 and 2-year survival rates to be 0.93 (95%CI: 0.87-0.98) and 0.72 (95%CI: 0.63-0.80), respectively, and the calibration curve has a good coincidence of prediction. Conclusion High GNRI scores are significantly associated with improved survival in elderly patients with NSCLC, and reliance on cut-off values may provide the appropriate timing for nutritional support. Background and objective With the aging of the population and the increased importance of lung cancer screening, the number of early-stage lung cancer patients has been on the rise in recent years, which can be classified into operable early-stage lung cancer and inoperable early-stage lung cancer. The most common pathological type is non-small cell lung cancer (NSCLC). Stereotactic body radiation therapy (SBRT) is the optimal treatment for inoperable early-stage NSCLC. The aim of this study was to investigate the prognosis of early-stage NSCLC patients treated with SBRT and its influencing factors in order to reduce the side effects of radiotherapy and improve the survival and quality of life. Methods Clinical data and follow-up outcomes of early-stage NSCLC patients treated with SBRT in our hospital from August 2010 to August 2020 were collected. Kaplan-Meier method was used to assess the prognosis, and the Cox proportional risk model was used for multivariate prognostic analysis. Results A total of 165 patients were included with a median follow-up time of 43.2 (range: 4.8-132.1) mon. The local control (LC) rates at 1-yr, 2-yr and 5-yr were 98.1%, 94.8% and 86.5% respectively. Karnof- sky performance status (KPS) score greater than 80 was an independent prognostic factor for LC (P=0.02). The overall survival (OS) rates at 1-yr, 2-yr and 5-yr were 97.6%, 93.0% and 68.9% respectively. A biological equivalent dose when a/^=10 (BED10) greater than 132 Gy was an independent prognostic factor for OS (P=0.04). Progression-free survival (PFS) rates at 1-yr, 2-yr and 5-yr were 93.3%, 79.5% and 55.3% respectively. The distance metastasis free survival (DMFS) rates at 1-yr, 2-yr and 5-yr were 94.5%, 83.2% and 58.4% respectively. BED10 greater than 150 Gy was an independent prognostic factor for DMFS (P=0.02). The regional control (RC) rates at 1-yr, 2-yr and 5-yr were 98.8%, 95.4% and 87.9% respectively. Conclusion SBRT is effective in treating early-stage NSCLC. KPS greater than 80 is an independent prognostic factor for LC; BED10 greater than 132 Gy is an independent prognostic factor for OS; BED10 greater than 150 Gy is an independent prognostic factor for DMFS. Background and objective Primary mediastinal germ cell tumor (PMGCT) is a rare but occasionally highly invasive mediastinal tumor. At present, there are few related disease special survival (DSS) studies on PMGCT, rare large data analysis, and uncommon DSS prognostic models. This study was to investigate the prognostic factors of DSS of the PMGCT patients, and build a simple and effective nomogram to predict the DSS prognosis in patients with PMGCT. Methods Retrospective clinicopathological data of 325 patients with PMGCT from 1975 to 2019 were extracted from Surveillance, Epidemiology, and End Results (SEER) database. The Kaplan-Meier method along with the Log-rank test were utilized to estimate the DSS. Cox proportional hazard regression model was used to screen the independent risk factors affecting prognosis, from which an individualized nomogram was constructed to predict 3-yr, 5-yr and 8-yr DSS of patients with PMGCT. The prediction accuracy of the model is evaluated by receiver operating characteristic (ROC) curve, correction curve and decision curve analysis (DCA) curve. Results The 3-yr, 5-yr and 8-yr survival rates of PMGCT were 84.6%, 83.6% and 83.3%, respectively. Univariate analysis showed that histology, surgery, age, tumor size, metastasis and stage could affect the prognosis of PMGCT. Multivariate analysis showed that histology, surgery, age and tumor size were independent risk factors for the prognosis of PMGCT patients, and the nomogram was constructed using these independent risk factors. The area under the curve (AUC) of ROC curve was 0.824. The results of the correction curves of 3-yr, 5-yr and 8-yr survival time and DCA, indicated that there was a good consistency between the predicted results of the nomogram evaluation and the real results. Conclusion Patients with histological classification of seminoma in PMGCT have a better prognosis than patients with non-seminoma. The prognosis of patients with over the age of 40 yr, tumor size >15 cm and without surgical treatment was even worse. Objective: To investigate the expression of tumor necrosis factor receptor associated protein 1 (TRAP1) in colon cancer tissues and its relationship with clinicopathological features and patient prognosis, as well as the related molecular mechanisms. Methods: The expression of TRAP1 in colon cancer and its relationship with clinicopathological features and patient prognosis were comprehensively analyzed by TCGA and GEO databases. In addition, 10 pairs of colon cancer tissues and corresponding paracancerous tissues surgically resected at the First Hospital of Shanxi Medical University between October 2020 and March 2021 were selected, and the expression of TRAP1 in colon cancer tissues of Chinese was detected by IHC staining for validation. Kaplan-Meier survival analysis was performed by running R package (survivor and survminer); The signal peptide and membrane piercing domain of TRAP1 protein were analyzed by online software, and GO analysis and KEGG analysis were carried out by gene enrichment analysis software. Colon cancer SW480 and SW620 cells were cultured and transfected with si-NC or si-TRAP1, and divided into blank control group, si-NC group and si-TRAP1 group. The TRAP1 expression in colon cancer cells of each group after transfection was detected by qPCR, and the cell cycle and apoptosis of the transfected cells were detected by FCM technique. Results: TRAP1 was highly expressed in colon cancer tissues compared with paracancerous tissues (P<0.01). The expression level of TRAP1 was correlated with lymph node metastasis (P<0.05), and the 5-year overall survival (OS) rate of colon cancer patients with high TRAP1 expression was lower than those with low TRAP1 expression (P<0.05). TRAP1 protein is a cytoplasmic protein, and functional enrichment results showed that TRAP1 and its related molecules are associated with signaling pathways such as cell cycle and ribosome biogenesis (all P<0.01). GSEA enrichment results showed that the levels of colon cancer metabolic reprogramming gene cluster and mitochondrial protein input gene cluster were elevated in the high TRAP1 expression group (all P<0.01). Knockdown of TRAP1 resulted in cell cycle arrest in G1 phase with significantly elevated level of apoptosis of colon cancer cells (all P<0.01). Conclusions: TRAP1 is highly expressed in colon cancer tissues and correlated with lymph node metastasis and dismal OS rate in patients. Objective We analyzed the clinical characteristics, survival of alpha-fetoprotein-producing gastric carcinoma (AFPGC) patients, and traditional Chinese medicine ( TCM) treatment on them to improve TCM clinicians′ recognition of AFPGC. Methods We retrospectively collected the clinical and survival characteristics of patients with gastric cancer who were pathologically diagnosed and treated with TCM from January 2012 to December 2018, including 46 patients with AFPGC and 363 patients with common gastric cancer, according to the eligibility criteria. To compare and analyze the survival of patients with different alpha-fetoprotein (AFP) levels, we performed COX multivariate analysis of the prognostic factors of AFPGC patients and an analysis of TCM treatment principles and prescriptions. Results AFPGC patients accounted for 4. 1% of all gastric cancers patients. Compared with common gastric cancer, the proportion of AFPGC was found to be higher in Uyghur and Kazakh patients than in Han patients (P<0. 001). AFPGC patients also showed marked invasion depths (P = 0. 032), higher ratio of lymphatic metastasis or distant metastasis(P = 0. 016, P < 0. 001), and had lower chances of receiving chemotherapy and surgery (P = 0. 018, P = 0. 001, respectively). The proportion of liver and peritoneal metastases was also higher in AFPGC patients (P<0. 001, each). The medium overall survival (mOS) of AFPGC patients was 11 (95% CI:7-14)months, while that for common gastric cancer was 33 (95% CI:26-45) months. AFPGC patients had a three-fold increased hazard ratio for death in common gastric cancer patients(HR = 3. 00, P<0. 001). The survival rates of AFPGC patients at 1, 3, and 5 years were 46. 7%, 11. 1%, and 2. 3%, lower, respectively, than that of the common gastric cancer patients (P<0. 001). We conducted subgroup analysis according to the level of AFP, < 20 μg / L, 20- 300 μg / L, and > 300 μg / L. The mOS of the three groups of patients was 33. 0, 14. 0, and 6. 5 months, and the difference was significant ( P < 0. 001 ). Radical surgery and AFP level were independent prognostic factors in AFPGC patients, and radical surgery could reduce the risk of death in AFPGC patients by 61. 2%. Patients with AFP >300 μg / L had a 1. 98-fold higher risk of death than patients with AFP 20-300 μg / L (P = 0. 029, P = 0. 048). The principles of TCM treatment include strengthening the spleen, replenishing qi, and removing phlegm from the stomach. We analyzed the frequency of antitumor TCM in the prescriptions of the patients at the first visit and found that the main drugs were used to strengthen vital qi and tonify. Conclusion AFPGC has unique clinical characteristics: poor biological behavior; prone to the liver, lymph node, and peritoneal metastasis; late-stage diagnosis; and a lower chance of receiving surgery and chemotherapy than ordinary gastric cancer. Survival is significantly worse than that of common gastric cancer and is significantly negatively correlated with AFP level. Radical resection is the only method that can improve the prognosis of AFPGC. Objective We analyzed the clinical characteristics, survival of alpha-fetoprotein-producing gastric carcinoma (AFPGC) patients, and traditional Chinese medicine ( TCM) treatment on them to improve TCM clinicians′ recognition of AFPGC. Methods We retrospectively collected the clinical and survival characteristics of patients with gastric cancer who were pathologically diagnosed and treated with TCM from January 2012 to December 2018, including 46 patients with AFPGC and 363 patients with common gastric cancer, according to the eligibility criteria. To compare and analyze the survival of patients with different alpha-fetoprotein (AFP) levels, we performed COX multivariate analysis of the prognostic factors of AFPGC patients and an analysis of TCM treatment principles and prescriptions. Results AFPGC patients accounted for 4. 1% of all gastric cancers patients. Compared with common gastric cancer, the proportion of AFPGC was found to be higher in Uyghur and Kazakh patients than in Han patients (P<0. 001). AFPGC patients also showed marked invasion depths (P = 0. 032), higher ratio of lymphatic metastasis or distant metastasis(P = 0. 016, P < 0. 001), and had lower chances of receiving chemotherapy and surgery (P = 0. 018, P = 0. 001, respectively). The proportion of liver and peritoneal metastases was also higher in AFPGC patients (P<0. 001, each). The medium overall survival (mOS) of AFPGC patients was 11 (95% CI:7-14)months, while that for common gastric cancer was 33 (95% CI:26-45) months. AFPGC patients had a three-fold increased hazard ratio for death in common gastric cancer patients(HR = 3. 00, P<0. 001). The survival rates of AFPGC patients at 1, 3, and 5 years were 46. 7%, 11. 1%, and 2. 3%, lower, respectively, than that of the common gastric cancer patients (P<0. 001). We conducted subgroup analysis according to the level of AFP, < 20 μg / L, 20- 300 μg / L, and > 300 μg / L. The mOS of the three groups of patients was 33. 0, 14. 0, and 6. 5 months, and the difference was significant ( P < 0. 001 ). Radical surgery and AFP level were independent prognostic factors in AFPGC patients, and radical surgery could reduce the risk of death in AFPGC patients by 61. 2%. Patients with AFP >300 μg / L had a 1. 98-fold higher risk of death than patients with AFP 20-300 μg / L (P = 0. 029, P = 0. 048). The principles of TCM treatment include strengthening the spleen, replenishing qi, and removing phlegm from the stomach. We analyzed the frequency of antitumor TCM in the prescriptions of the patients at the first visit and found that the main drugs were used to strengthen vital qi and tonify. Conclusion AFPGC has unique clinical characteristics: poor biological behavior; prone to the liver, lymph node, and peritoneal metastasis; late-stage diagnosis; and a lower chance of receiving surgery and chemotherapy than ordinary gastric cancer. Objective: To analyze the gene expression of coiled-coil domain containing protein 137 (CCDC137) in hepatocellular carcinoma (HCC) tissues and its relationship with clinicopathological features and prognosis of HCC patients, and to explore the effect of CCDC137 knockdown on the proliferation, migration and invasion of MHCC97H cells. Methods: The HCC dataset was downloaded from The Cancer Genome Atlas (TCGA) to obtain the CCDC137 gene expression profile and clinical information of patients. The correlation between the expression level of CCDC137 gene in HCC tissues and the clinicopathological indices and its impact on patients' prognosis were analyzed by bioinformatics method. Gene Set Enrichment Analysis (GSEA) was used to predict the possible pathways regulated by CCDC137 gene in HCC. Kaplan-Meier method and Log-Rank test were used for survival analysis; Cox proportional hazards regression model was used to analyze the risk factors affecting the prognosis of patients. The expression of CCDC137 in MHCC97H cells was inhibited by small interfering RNA technology. The mRNA and protein expression levels of CCDC137 in transfected MHCC97H cells as well as its effect on cell proliferation, migration and invasion were observed by qPCR, WB, CCK-8 and Transwell assays, respectively. Results: According to the data of 371 HCC patients retrieved from TCGA database, the expression level of CCDC137 mRNA in tumor tissues was significantly higher than that in the para-cancerous tissues (P<0.01). The high expression of CCDC137 mRNA was significantly correlated with tumor histological grade (OR=0.014), cancer tissue stage (OR=0.007), and T stage (OR=0.047) (all P<0.05). The OS rate of patients with high CCDC137 expression was significantly lower than that of patients with low CCDC137 expression (P<0.05), and multivariate Cox regression analysis suggested that CCDC137 gene could be an independent prognostic factor for HCC patients. GSEA results showed that the samples with high expression of CCDC137 gene were enriched in multiple pathways/gene sets such as base excision repair and spliceosome (P<0.01, FDR<0.05). After knockdown of CCDC137 gene, the proliferation, migration and invasion of MHCC97H cells were significantly decreased (all P<0.01). Conclusion: CCDC137 gene is highly expressed in HCC tissues. The high expression of CCDC137 is related to the occurrence, development and poor prognosis of HCC. Objective: To explore the mechanism and clinical significance of carboxypeptidase A4 (CPA4) promoting the proliferation, migration, invasion and epithelial-mesenchymal transformation (EMT) progression of non-small cell lung cancer (NSCLC) cells. Methods: IHC and WB were employed to detect the expression of CPA4 in NSCLC tissues and adjacent tissues of 105 patients with advanced or metastatic NSCLC in Jinan Central Hospital from February 2012 to December 2015 and the expression of CPA4 in NSCLC cells. Chi-square test was used to analyze the association between CPA4 expression and the clinicopathological characteristics of the patients. The relationship between CPA4 expression and the overall survival (OS) of the patients was analyzed by Kaplan-Meier method. H1299 and A549 NSCLC cells with stable overexpression and downexpression of CPA4 were constructed by cell transfection. Changes in the cell proliferation, invasion and migration after overexpression and knockdown of CPA4 were detected by CCK-8 assay, cell colony formation assay, scratch healing assay and Transwell assay. Xenograft tumor models and tail vein-lung metastasis tumor models of nude mice were established to detect the role of CPA4 in tumor tumorigenesis and metastasis in vivo. Changes in the markers of EMT in the tissues of xenografts were detected by WB and IHC. Results: Compared with paracancer tissues and normal lung epithelial cells, CPA4 was highly expressed in cancer tissues and cancer cells (all P<0.05). The OS of NSCLC patients with high CPA4 expression was significantly shorter than that of patients with low CPA4 expression (P<0.05). High expression of CPA4 was more common in poor differentiation, N2-3 lymph node involvement and TNM stage IV (P<0.05 or P<0.01). Overexpression of CPA4 promoted H1299 cells, while knockdown of CPA4 inhibited the proliferation, migration and invasion of A549 cells. Overexpression of CPA4 promoted the growth of H1299 cell xenograft tumors and lung metastatic tumors in nude mice. Overexpression of CPA4 promoted the changes of EMT process-related molecules in H1299 cells. Conclusion: CPA4 is highly expressed in NSCLC tissues and cells. Overexpression of CPA4 can induce EMT, promote the proliferation, migration and invasion of NSCLC cells and is related to tumor progression and prognosis. The positive impact of such therapies on survival in patients with gastric cancer has become clearer over time. In recent years, new achievements have been made in the fields of neoadjuvant radiotherapy, adjuvant radiotherapy, advanced tumor radiotherapy and combined immunoradiotherapy for gastric cancer. Objective Patients with locally advanced prostate cancer have a worse prognosis than those with localized prostate cancer. For patients with locally advanced prostate cancer, especially those with positive clinical lymph nodes, the treatment is still controversial at present. We will conduct further research on it to guide clinical practice. Methods Clinical data of patients diagnosed as clinically positive lymph node adenocarcinoma of the prostate from 2004 to 2015 were collected from the U.S. Surveillance, Epidemiology, and End Results Database（SEER database）. The patients were divided into surgical treatment group and radiotherapy group according to their treatment methods. Overall survival rates were compared and stratified by age. Results Age was a significant factor influencing the survival of patients with clinically lymph node positive prostate cancer. Overall, surgery group had better survival than radiotherapy group, despite poorer baseline outcomes for radiotherapy group; But in the patients over 70, radiation group showed similar survival rates as surgery group. Conclusion For clinically lymph node positive prostate cancer patients, individualized treatment should be selected according to the patient’s age. For patients over 70 years of age, radiation seems to be an alternative to surgery due to the patient’s health and life expectancy. This conclusion needs to be confirmed by some prospective trials. Background and objective Lung cancer has the highest mortality in China. Different treatments are of great significance to the prognosis of patients. By comparing stage Ia non-small cell lung cancer (NSCLC) patients' survival rates for ablation and for sub-lobectomy, we studied the difference in the effects of the two treatments on patient prognosis. Methods Using the Surveillance, Epidemiology, and End Results (SEER) database, we screened eligible patients with stage Ia NSCLC from January 2004 to December 2015. Then, 228 patients treated with ablation and 228 patients treated with sub- lobotomy were then selected based on propensity score matching. After stratification, matching, and adjustment the Kaplan- Meier analysis was performed to compare the overall survival rates of patients treated with the two procedures. Results The Kaplan-Meier survival analysis showed that there is a significant difference between the ablation group and the sub-lobectomy group (P<0.05). In the univariable analysis, the hazard ratio (HR) of sub-lobotomy group was 0.571 (95%CI: 0.455-0.717) compared with the ablation group. Patients treated with sub-lobectomy had a 0.571 times greater risk of adverse outcomes than those treated with ablation. In the multivariable analysis, the HR for sub-lobectomy group was 0.605 (95%CI: 0.477-0.766) compared with the ablation group. Patients treated with sub-lobectomy had a 0.605 time greater risk of adverse outcomes than those treated with ablation. The results suggested that the overall survival rate of patients with stage Ia NSCLC treated with sub-lobotomy was higher than that of patients treated with ablation. Conclusion This study suggests that there is a significant difference in overall survival of stage Ia NSCLC patients treated with ablation and with sub-lobotomy. Background and objective Da Vinci robotic surgery system is widely used in department of thoracic surgery. The aim of this study is to investigate the treatment outcome of stage I non-small cell lung cancer (NSCLC) via da Vinci Surgical System. Methods Clinical date of 347 stage I NSCLC patients, who underwent lobectomy and systematic node dissection from Jan. 2012 to Dec. 2017, were reviewed. 134 patients underwent robot-assisted thoracic surgery (RATS) and 213 patients underwent video-assisted thoracic surgery (VATS). To compare perioperative outcome (blood lose, postoperative drainage, drainage time, postoperative hospital stay, number of the LN dissection) and analyze overall survival (OS), disease free survival (DFS) of the two groups and prognostic factors. Results The RATS group got less blood lose [(49±39 mL for RATS vs (202±239) mL for VATS, P<0.05] and postoperative drainage [Day 1: (248±123) mL for RATS vs (350±213) mL for VATS; Day 2: (288±189) mL for RATS vs (338±189) mL for VATS, P<0.05]. There were no significant difference for drainage time (10±5 for RATS vs 11±8 for VATS, P<0.05) and postoperative hospital stay (13±6 for RATS vs 14±9 for VATS, P<0.05) between the two groups. The RATS group harvested a more number of mean stations (5±2 for RATS vs 4±2 for VATS) and amounts (18±9 for RATS vs 11±8 for VATS) of the lymph nodes, P<0.05. There was no statistically significant difference of OS between RATS and VATS group [1-year OS: 97.3% vs96%; 3-year OS: 89.8% vs 83.1%; 5-year OS: 87.5 % vs 70.3%; overall survival time (mean): 61 months vs 59 months, P>0.05]; corresponding there had a statistically significant difference of DFS between the two groups [1-year DFS: 93.7% vs 91 .3%; 3-year DFS: 87.7% vs 68.4%; 5-year DFS: 87.7% vs 52.5%; disease free survival time (mean): 61 months vs 50 months, P<0.05]. The univariate analysis found that the amounts of the lymph nodes dissection was the prognostic factor for OS and tumor diameter, surgical approach, stations and amounts of the lymph nodes dissection were respectively the prognostic factors for DFS. However, multivariate analysis found that there was not independently factors for OS, but the tumor diameter and surgical approach were independently associated with DFS. Conclusion There was no significant difference about OS between the two groups, but the RATS got better DFS. RATS got more number of the LN dissection and less blood lose. [ABSTRACT FROM AUTHOR]
