Objective To observe the expression changes of micro RNA-23a(miR-23a) and apoptotic protease-activating factor-1 (APAF-1) in endometrial carcinoma tissues and analyze its clinical significance. Methods 123 specimens of endometrial carcinoma in our hospital from May 2018 to May 2020 were selected as endometrial carcinoma group, and the para cancerous tissue was selected as the normal group. Expression of miR-23a and APAF-1 were examined in endometrial tissues; the relationship between miR-23a and APAF-1 and the clinicopathology of patients and the correlation between the two were analyzed; Kaplan-Meier method was used to analyze the survival of patients with endometrial cancer. Results Compared with the normal group, the expression level of miR-23a in endometrial carcinoma group was significantly increased, the expressions of APAF-1 mRNA and protein decreased significantly (P < 0.05); miR-23a and APAF-1 were significantly correlated with clinical stage, histological differentiation and myometrial invasion (P < 0.05); Pearson analysis showed that miR-23a was negatively correlated with APAF-1 (P < 0.05); Kaplan-Meier curves showed that PFS and OS were significantly higher in the miR-23a low-expression group as well as the APAF-1-positive expression group than in the miR-23a high-expression group and the APAF-1-negative expression group(P < 0.05). Conclusion The expression level of miR-23a is increased and APAF-1 expression is decreased in endometrial carcinoma, both of which correlate with endometrial cancer progression and patient prognosis, are expected to be biomarkers for clinical assessment of disease progression and prognosis in early-stage patients. AIM: To explore the expression level of vitamin D receptor (VDR) in lung adenocarcinoma and its impact on the biological function of lung adenocarcinoma cells. METHODS: The mRNA expression level of VDR in lung adenocarcinoma and its relationship with prognosis were analyzed through 6 lung adenocarcinoma datasets (comprising a total of 792 lung adenocarcinoma tissues and 230 adjacent non-tumor tissues). Immunohistochemistry was used to detect VDR protein expression in 30 lung cancer patients. Lung adenocarcinoma cell lines A549 and H1650 were studied, with transfection of negative control (NC) or two VDR short hairpin RNAs (VDR-shRNAs). CCK-8 assay compared the cell viability of cells in each experimental group. Transwell and wound-healing assays compared the invasion and migration capabilities. Gene set enrichment analysis identified pathways enriched in lung adenocarcinoma tissues with high VDR expression. RESULTS: The mRNA expression level of VDR was significantly increased in lung adenocarcinoma tissues (P<0. 01). Immunohistochemistry further confirmed the high expression of VDR in lung adenocarcinoma (P< 0.01). The survival analysis showed that the expression of VDR had no significant effect on the overall survival of lung adenocarcinoma patients (P>0. 05). Knockdown of VDR significantly inhibited the cell viability, invasion, and migration ca⁃ pacity of lung adenocarcinoma cells (P<0. 05). Gene set enrichment analysis showed that lung adenocarcinoma tissues with high VDR expression were enriched in signaling pathways such as epithelial-mesenchymal transition (P<0. 05). Objective: To investigate diagnostic and prognostic value of actin-binding protein ANLN in clear cell renal cell carcinoma （ccRCC） and its relationship with tumor microenvironment. Methods: Gene expression data and clinical data for ccRCC were downloaded from The Cancer Genome Atlas （TCGA）. Relationship between ANLN expression and clinicopathological features was assessed by Wilcoxon rank sum test and Logistic regression. Receiver operating characteristic （ROC） curve was used to assess diagnostic value of ANLN expression in ccRCC. Kaplan-Meier and Cox regression analysis were used to investigate effect of ANLN expression on overall survival. Gene set enrichment analysis （GSEA） was used to identify signaling pathways associated with ANLN in ccRCC. Relationship between ANLN expression and immune infiltration was analyzed by ESTIMATE algorithm, tumor immune estimation resource （TIMER） and CIBERSORT algorithms. Relationship between ANLN and drug sensitivity was calculated using CellMiner database. Results: ANLN expression was significantly upregulated in ccRCC tissues. ANLN expression in ccRCC was correlated with clinicopathological features. ROC analysis showed that ANLN had a high diagnostic value in ccRCC. High ANLN expression was significantly associated with poor prognosis. Multivariate Cox regression analysis showed that high ANLN expression was an independent risk factor for overall survival in ccRCC patients. GSEA showed that ANLN was associated with multiple signaling pathways. In terms of immunity, ANLN was closely associated with tumor microenvironment, immune infiltration and immune checkpoint molecules in ccRCC. ANLN expression was negatively correlated with sensitivity of most antitumor drugs. Conclusion: ANLN is a potential diagnostic and prognostic biomarker and immunotherapeutic target for ccRCC. Leptomeningeal metastasis (LM) is a serious late complication in patients with solid tumors, which is common in patients with lung cancer, breast cancer and melanoma. In recent years, due to the progress of diagnosis, the diagnosis rate of LM has gradually increased. The main goal of the therapy is to maintain neurological function, improve the quality of life and the overall survival rate, and prolong the progression-free survival time of patients. Intrathecal therapy is one of the main treatments for LM, which can deliver drugs directly to the subarachnoid space. The traditional intrathecal drugs are methotrexate, cytarabine and thiotepa. With the development of new research, a variety of chemotherapeutic drugs, targeted drugs and immune checkpoint inhibitors have also been used in intrathecal therapy. In addition, different ways of intrathecal administration also bring new hope to patients. This article reviewed the clinical research progress of intrathecal therapy in the treatment of LM. Objective: To study the relationship between serum long chain non coding RNA-human plasmacytoma transformation and migration gene 1 (LncRNA-PVT1), microribonucleic acid-497-5p (miR-497-5p) expression and lung function and prognosis in patients with chronic obstructive pulmonary disease (COPD) with lung cancer. Methods: 116 patients with COPD combined with lung cancer underwent concurrent surgical treatment admitted to Linyi Cancer Hospital from March 2018 to March 2020 were selected as the observation group, while 50 patients with simple COPD during the same period were included in the control group. The relative expression levels of serum LncRNA-PVT1 and miR-497-5p and lung function indexes [forced expiratory volume in the first second (FEV1), forced vital capacity (FVC), FEV1/FVC, percentage of forced expiratory volume in the first second to predicted value (FEV1%pred)] were compared between two groups. The correlation between serum LncRNA-PVT1, miR-497-5p expression and lung function indexes in patients with COPD with lung cancer were analyzed by Pearson test. Patients with COPD with lung cancer were followed up for 3 years after operation, the prognostic factors of patients with COPD with lung cancer were analyzed by multivariate Cox regression model. 3 years overall survival rate of patients with low expression and high expression of LncRNA-PVT1 and miR-497-5p were analyzed by drawn Kaplan-Meier survival curve. Results: Compared with control group, the expression level of serum LncRNA-PVT1 in observation group was significantly increased (P<0.05), and the expression level of miR-497-5p was significantly decreased (P<0.05). Compared with control group, FEV1, FEV1% pred and FEV1/FVC in observation group were significantly lower(P<0.05). Pearson test results showed that, serum LncRNA-PVT1 was negatively correlated with FEV1, FEV1% pred and FEV1/FVC in patients with COPD with lung cancer (P<0.05); serum miR-497-5p was positively correlated with FEV1, FEV1% pred and FEV1/FVC(P<0.05). The Kplan-Meier survival curve showed that, the 3 years overall survival rate in LncRNA-PVT1 high expression group was 29.23%, which was significantly lower than 47.73% in LncRNA-PVT1 low expression group (P<0.05). 3 years overall survival rate in miR-497-5p high expression group was 46.77%, which was significantly higher than 23.40% in miR-497-5p low expression group (P<0.05). Multivariate Cox regression analysis showed that, lymph node metastasis, TNM stage IIIa, poor differentiation of tumor tissue, high expression of LncRNA-PVT1 and low expression of miR-497-5p were risk factors for prognosis of patients with COPD with lung cancer (P<0.05). Conclusion: LncRNA-PVT1 is highly expresse and miR-497-5p is lowly expresse in the serum of patients with COPD with lung cancer, the expression level is relate to lung function indexes, and the higher the expression level of LncRNA-PVT1 and the lower the expression level of miR-497-5p, the lower the 3 years overall survival rate. Background and objective Pulmonary sarcomatoid carcinoma (PSC) is a rare subtype of non-small cell lung cancer (NSCLC), which is featured by low incidence, high malignancy rate, robust aggressive behavior and inferior prognosis. To date, there is no standardized treatment. The aim of this study is to better understand and accumulate more clinical experience of the disease by summarizing the clinicopathological features, diagnosis methods, therapeutic regimen and prognostic factors of PSC. Methods A total of 39 patients with PSC who diagnosed and received treatment in Beijing Chest Hospital from December 2013 to December 2023 were retrospectively recruited, and information including demographic characteristics, clinicopathological features, tumor-node-metastasis (TNM) stage, diagnosis method and therapeutic regimen were carefully collected. Meanwhile, follow-up was conducted. Kaplan-Meier method was used to analyze the prognostic factors of the disease. Results The PSC patients in this study ranged in age from 45 to 76 years old, including 35 males and 4 females. There were no specific clinical manifestations of PSC at initial diagnosis. Among the 39 patients, 20 underwent surgical resection and 19 received palliative chemoradiation or symptomatic supportive treatment. The 1-year and 5-year survival rates were 61.90% and 35.20% respectively. Univariate analysis indicated that family history of carcinoma, primary tumor site, TNM stage, lymph node metastasis, distant metastasis, whether or not received surgical resection, surgical method, treatment regimens, tumor tissue programmed cell death ligand 1 (PD-L1) expression ≥1% and mesenchymal-epithelial transition factor (MET) pathway abnormalities were correlated with the overall survival (OS) of patients (P<0.05). In the subsequent multivariate analysis, lymph node metastasis emerged as the only independent prognosticator in predicting inferior OS (P=0.037). Conclusion PSC is rarely seen in clinical practice and commonly occurs in elder men with smoking history. Tumor tissue PD-L1 expression ≥1% and MET abnormalities may predict inferior prognosis of PSC and lymph node metastasis was determined as the independent prognosticator of PSC. Surgical resection along with adjuvant medical treatment is the cornerstone for early and locally advanced patients, and the clinical utility of molecular targeting therapy and immunotherapy in PSC needs to be further investigated. Objective To investigate the relationship between circulating tumor cells (CTCs) and clinical characteristics and prognosis of elderly patients with esophagogastric junction adenocarcinoma (AEG) by detecting CTCs in their peripheral blood. Methods A retrospective analysis was conducted on 95 elderly AEG patients at Liaocheng People's Hospital from August 2016 to May 2020. Negative enrichment-immuno fluorescence in situ hybridization (NE-FISH) was used to detect CTCs one week before treatment. The relationship between positive CTCs count and clinical characteristics of patients was analyzed. Progression free survival (PFS) and overall survival (OS) of elderly AEG patients were obtained through follow-up, and survival analysis was performed using Kaplan-Meier method. Cox regression analysis was used to analyze the relationship between CTCs count and prognostic indicators (PFS, OS) in elderly AEG patients. Results The positive rate of CTCs (≥2) in 95 elderly AEG patients was 72.6% (69/95). The positivity of CTCs in elderly AEG patients was closely related to TNM pathological staging, T staging, lymph node metastasis, and surgery (P＜0.05). Kaplan Meier survival analysis showed that, compared with the CTCs＜2 group, the median PFS (12 months vs 50 months) and median OS (20 months vs 50 months) of elderly AEG patients in the CTCs≥2 group were shorter and the differences were statistically significant (P＜0.01). Conclusion The use of NE-FISH technology for peripheral blood CTCs detection in elderly AEG patients is feasible. Positive CTCs are closely related to tumor metastasis and disease progression in elderly AEG patients, and CTCs≥2 may be an indicator of poor prognosis in elderly AEG patients.

Objective To explore circular RNAs(circRNAs) that regulate ferroptosis in cervical cancer,in the development of new therapeutic targets. Methods The differentially expressed circRNAs(DEcircRNAs),differentially expressed microRNAs(DEmiRNAs) and differentially expressed mRNAs(DEmRNAs) between cervical cancer and control groups were analyzed based on GSE102686,TCGA,GSE9750.circRNAs and mRNAs with targeted regulatory relationships with DEmiRNAs were predicted by the Starbase database and compared with differential analysis results.Enrichment analysis of mRNAs was performed to identify mRNAs associated with cell death(cell cycle,apoptosis,and iron death) and to construct relevant competitive endogenous RNAs(ceRNAs) regulatory networks.Univariate Cox regression analysis was performed to identify miRNAs and mRNAs that significantly affect patient outcomes.Cervical cancer tissues and distant control cervical tissue samples from 12 patients with cervical cancer were collected to detect gene expression using quantitative reverse transcription polymerase chain reaction(qRT-PCR) and Western blot,and targeted binding of genes was detected using a dual-luciferase reporter. Results A total of 179 DEcircRNAs,93 DEmiRNAs,and 739 DEmRNAs were identified between cervical cancer and controls,and 160 DEcircRNAs were found,and 565 DEmRNAs were targeted to DEmiRNAs.The ceRNAs regulatory network of circ_0001461/miR-145-5p/transferrin receptor(TFRC) was finally identified to be associated with ferroptosis.circ_0001461 and TFRC were significantly upregulated and miR-145-5p was downregulated in cervical cancer tissues(P

Background and purpose: According to current consensus, adverse high-risk pathological features are only associated with adjuvant therapy for stage II colorectal cancer (CRC). As important prognostic factors, we further explored the possibility of identifying patients with potential recurrence and poor prognosis based on these incorporating high-risk pathological features. Methods: This is a cohort study. A retrospective analysis was conducted on clinical data of CRC patients who underwent surgical treatment at the Second Department of Colorectal Surgery, Fudan University Affiliated Shanghai Cancer Center from 2008 to 2018. This study was approved by the Ethics Committee of the Fudan University Shanghai Cancer Center (approval No.: 050432-4-2108*), and the study complies with the Declaration of Helsinki. A total of 9 875 patients were enrolled, including 5 859 males and 4 016 females, aged [M (IQR)] 60 (16) years (range: 16 to 94). Median follow-up time was 1 779.0 days [95% CI: 1 750.1-1 807.9]. We used the Kaplan-Meier method to plot survival curves for different groups. Cox multivariate analysis was used to identify independent risk factors for 5-year overall survival (OS), disease-free survival (DFS) and recurrence-free survival (RFS). Finally, a column chart model was constructed to evaluate and stratify patient prognosis. The Strengthening the Reporting of Observational Studies in Epidemiology (STROBE) checklist was followed for this cohort study. Results: According to the number of incorporating high-risk pathological features, patients were divided into five groups: Hr_0 group (0 incorporating high-risk pathological feature), Hr_1 group (1 incorporating high-risk pathological feature), Hr_2 group (2 incorporating high-risk pathological features), Hr_3 group (3 incorporating high-risk pathological features), and Hr_4 group (4 or more incorporating high-risk pathological features). The Kaplan-Meier survival curve results indicated significant differences in OS, DFS and RFS among different groups (all P<0.001). Subgroup analysis was conducted on stage II colorectal cancer, and the survival curves of OS, DFS and RFS in different Hr groups overlapped with each other. Compared to the overall population, the survival differences in different groups were significantly reduced, indicating that stage II colon cancer patients with incorporating high-risk pathological features may benefit from adjuvant chemotherapy. The independent prognostic factors for RFS included age, pT stage, pN stage and Hr group. The survival curves of OS, DFS and RFS indicated that the prognosis of Hr_4 group was significantly worse than that of stage IIIc patients; 5.2% and 14.1% of stage I and II patients had two or more incorporating high-risk pathological features (Hr group≥2), respectively. Finally, a column chart model was constructed by incorporating the independent prognostic risk factors for CRC mentioned above. The calibration curve showed a good consistency between the actual observations and the predictions made by the nomogram, and the decision curve analysis (DCA) indicated that the model constructed in this study had good efficacy in stratifying recurrence. Conclusion: The number of incorporating high-risk pathological features is an independent prognostic factor for RFS in patients with stage I-III CRC. Combining it as a multiclass variable with age, pT and pN stage has good prognostic stratification and recurrence stratification efficacy, which is expected to guide clinical treatment. Objective To explore the clinical and molecular pathological features of primary glioblastoma (PGBM), along with the factors influencing its prognosis. Methods The clinical data and molecular pathology results of 49 PGBM patients who underwent surgical resection in the Affiliated Provincial Hospital of Anhui Medical University from January 2021 to July 2022 were analyzed retrospectively. Variables including gender, age, extent of tumor resection, postoperative treatment modalities, as well as genetic expressions such as MGMT promoter methylation, TP53 mutation, CDK4/6 amplification, and NF1 mutation were scrutinized. Statistical analyses were employed to correlate these clinical features and genetic mutations with patients'progression-free survival and overall survival. Results Male patients(65. 30％)outnumbered female patients(34. 70％). Gross total resection accounted for 81. 60％, while subtotal resection or partial resection constituted 18. 4％. Postoperatively, standard adjuvant chemoradiotherapy was predominantly administered(63. 30％). Molecular pathology assessments revealed MGMT promoter methylation positivity in 55. 10％, TP53 mutation in 38. 78％, CDK4/6 amplification in 48. 98％, and NF1 mutation in 28. 57％of cases. Correlation analysis indicated that extent of resection, postoperative adjuvant chemoradiotherapy, MGMT promoter methylation, TP53 mutation, and CDK4/6 amplification were independent prognostic factors (P＜0. 05), while NF1 mutation showed no significant correlation with prognosis (P＞0. 05). Conclusions The clinical characteristics and some gene mutations of PGBM patients are closely related to prognosis. Age, extent of tumor resection, postoperative treatment modalities, as well as MGMT promoter methylation, TP53 mutation, and CDK4/6 amplification, emerge as crucial indicators for predicting prognosis in PGBM patients, while further research is warranted to elucidate the impact of NF1 mutation on prognosis. Objective To investigate the clinical characteristics of survival in patients with high-risk myelodysplastic syndromes (HR-MDS) and provide a reference for the clinical prognosis of patients with HR-MDS. Methods General data, blood routine test, bone marrow smear With histopathology, cytogenetics, and other clinical data of 200 patients diagnosed with HR-MDS at Xiyuan Hospital of China Academy of Chinese Medical Sciences, during the period of January 2016-September 2022, were retrospectively analyzed. The included patients were categorized into the arsenic-containing herbal combination combined with demethylating agents (HMAs) treatment group and the arsenic-containing Chinese medicine compound combined with androgen treatment group. The influence of clinical indices on the survival characteristics of each group was analyzed. Results Comparison of the impact of clinical indicators on survival in 200 patients with HR-MDS who were treated with arsenic-containing herbal compounds in combination with HMAs or androgens showed that high-risk vs. very high-risk (P=0.018), hemoglobin (Hb) < 80 g/L vs. Hb ≥80 g/L (P=0.035), platelet (PLT) counts < 50 x 109 L-1 vs. PLT counts ≥50 x 109 L-1 (P < 0.001), and the difference in median progression-free survival (PFS) time between myelodysplastic syndromes converted to leukemia (MDS-AML) and non-MDS-AML (P=0.003) were statistically significant. Comparison of survival effects of clinical indicators in 68 patients with HR-MDS who were treated with arsenic-containing Chinese medicine compound combined with HMAs showed that the difference in median PFS between PLT count < 50 x 109 L-1 and PLT count ≥50 x 109 L-1 (P < 0.001) and the difference in median PFS between < 5 and ≥5 courses of chemotherapy (P=0.018) were statistically significant. Comparison of survival effects of clinical indicators in 132 patients with HR-MDS who were treated with arsenic-containing Chinese medicine compound combined with androgens showed that Hb < 80 g/L and Hg ≥80 g/L (P=0.028), PLT count < 50 x 109 L-1 and PLT count ≥50 x 109 L-1 (P=0.002), and the mean differences in PFS between MDS-AML and non-MDS-AML (P=0.024) were statistically significant. Conclusion The clinical characteristics of long-surviving patients treated with arsenic-containing herbal combination in combination with HMAs included PLT counts ≥50 x 109 L-1 and ≥5 courses of chemotherapy. The clinical characteristics of long-surviving patients treated with arsenic-containing herbal combination in combination with androgens included Hg ≥80 g/L, PLT count ≥50 x 109 L-1, and non-MDS-AML. Objective: To explore potential biomarkers that can predict the clinical efficacy of PD-1/PD-L1 inhibitors in malignancies. Methods: The PubMed, Web of Science, CNKI, Wanfang and VIP databases were searched from the establishment of the database to September 20, 2022. After literature screening, data extraction and the risk of bias were evaluated independently by two evaluators, the Meta-analysis was performed using RevMan5.4 and STATA16.0 software. Results: This paper included 18 studies with a total of 4 018 patients. Tumor patients with a high tumor mutational burden (TMB) were found to have higher overall survival (OS) (P=0.003,P=0.01) and progression-free survival (PFS)(P=0.000 2, P=0.04) with PD-1/PD-L1 inhibitors within 1 year and 2 years of follow-up. At different follow-up times, with 1% as the critical value, there was no statistical significance in the level of PD-L1 expression as a biomarker for predicting OS and PFS of PD-1/PD-L1 inhibitors (P>0.05). Conclusion: TMB can be used as a biological indicator to predict the clinical efficacy of PD-1/PD-L1 inhibitors in patients with malignant tumors within 2 years after treatment, but whether its efficacy can last longer remains to be further studied. Objective: To explore the effect of SQLE on the biological characteristics of pan-cancer, using bioinformatics methods to predict and analyze the correlation between SQLE and the clinical prognosis and immunity of pan-cancer. Methods: The expression level of SQLE in tumor and normal tissues and its correlation with different clinical stages of pan-cancer were analyzed, and the relationship between SQLE expression and overall survival and tumor microenvironment were evaluated. Gene enrichment analysis was conducted to study the biological functions and signaling pathways related to SQLE gene, then, calculate the correlation between SQLE gene and tumor mutation load and microsatellite instability, and analyze the correlation between SQLE gene and immunomodulatory genes. Results: SQLE gene was highly expressed in variety of tumors, and differentially expressed in different tumor stages. High expression of SQLE was a risk factor in variety of tumors. Patients with high expression of SQLE hade a worse prognosis. SQLE was correlated with TMB and MSI, and involved in tumor development through different pathways. Conclusion: SQLE, as a valuable prognostic biomarker and new therapeutic target for variety of cancers, has certain application value in basic research and clinical treatment of various cancers. Objective: To investigate whether endoplasmic reticulum transmembrane protein IRE1 (encoded by Ern1) can modulate the immunogenicity and tumorigenicity of cancer cells and explore the underlying mechanism. Methods: Correlation between ERN1 expression and the overall survival of cancer patients was explored with public cancer databases. CRISPR-Cas9 technology was used to delete Ern1 in mouse tumor cell lines MCA205 and TC-1. By CCK-8, flow cytometry, ELISA, luciferase reporter systems, subcutaneous tumor models, prime-boost regimens, we analyzed impact of Ern1 on tumor cell proliferation in vitro and tumor growth in vivo, intratumoral immune cell composition, tunicamycin-induced immunogenic cell death and activation of anti-tumor effector T cells. The growth kinetics of Ern1-/- tumors was also monitored in Ifnar-/- mice. Results: ERN1 expression was found to be negatively correlated with the overall survival of cancer patients across multiple cancer types. Although Ern1 deficiency didn't affect tumor cell proliferation in vitro, it largely delayed tumor growth or caused spontaneous tumor regression in immune-competent mice. As compared to wild type counterparts, Ern1 tumors harbored much more neutrophils but significantly less CD4T cells. Upon tunicamycin-induced endoplasmic reticulum stress, Ern1-/- cells were more vulnerable to cell death. Although Ern1-deficiency reduced HMGB1 release and calreticulin exposure, IFN-α/β secretion was increased, and strongly elevated type I IFN response in tumor cells and augmented IFN-γ production by anti-tumor effector T cells. Ern1-/- tumor cells restored the tumorigenic capacity in Ifnar-/- mice. Background and objective With further understanding and research into non-small cell lung cancer with tumours ≤2 cm in maximum diameter, segmental lung resection is able to achieve the same long-term prognosis as lobectomy. However, there are few studies on the prognostic effect of wedge resection on small volume invasive lung adenocarcinoma with an invasion depth of 0.5 to 1.0 cm. Therefore, this study focuses on the clinical efficacy and prognosis of wedge resection in patients with small-volume invasive lung adenocarcinoma. Methods A retrospective analysis of the medical records of 208 patients who underwent surgery in the Department of Thoracic Surgery of the Affiliated Provincial Hospital of Anhui Medical University from February 2016 to December 2017 was made, and the postoperative pathological results confirmed small volume invasive lung adenocarcinoma. According to their surgical methods, they were divided into lobectomy group (n=115), segmentectomy group (n=48) and wedge resection group (n=45). Kaplan-Meier survival curve estimation and Cox proportional risk regression model were used to explore the influence of different surgical methods on the prognosis of patients with small volume invasive lung adenocarcinoma. Results The wedge resection group had better perioperative outcomes compared with the segmentectomy group and lobectomy group, with statistically significant differences in intraoperative bleeding (P=0.036), postoperative drainage (P<0.001), operative time (P=0.018), postoperative time with tubes (P=0.001), and postoperative complication rate (P=0.006). There were no significant differences when comparing the three groups in terms of survival rate (lobectomy group vs segmentectomy group, P=0.303; lobectomy group vs wedge resection group, P=0.742; and segmentectomy group vs wedge resection group, P=0.278) and recurrence-free survival rate (lobectomy group vs segmentectomy group, P=0.495; lobectomy group vs wedge resection group, P=0.362; segmentectomy group vs wedge resection group, P=0.775). Univariate and multivariate survival analyses showed that consolidation tumor ratio (CTR) was the prognostic factor of overall survival and revurrence-free survival for patients with small-volume invasive lung adenocarcinoma (P<0.05). Conclusion Wedge resection in patients with small volume invasive lung adenocarcinoma can achieve long-term outcomes similar to segmentectomy and lobectomy. Objective: To explore the efficacy and safety of PD-1 antibody versus high-dose interferon in the adjuvant treatment of stage IIB-IIID melanoma patients after surgery. Methods: Clinical data of patients with stage IIB-IIID cutaneous and acral melanoma admitted into Nanjing Drum Tower Hospital, the affiliated hospital of Nanjing University Medical School between September 2013 and September 2022 were retrospectively collected. All patients received high-dose interferon (HDI) or PD-1 antibody as adjuvant therapy after surgery. Univariate survival analysis was performed and survival curves were plotted by the Kaplan-Meier method; Log-Rank method was used to analyze and assess whether the differences in RFS, DMFS, and OS between the groups were statistically significant, and prognostic factors were evaluated by univariate and multivariate Cox regression. Results: A total of 91 patients were enrolled in this study with a median follow-up of 31.0 months. The median RFS was 29.2 and 32.3 months in the HDI group and the PD-1 antibody group, respectively, and the difference was not statistically significant (HR=0.90, 95%CI [0.50, 1.64]; P=0.736). The median DMFS and the median OS in the HDI group were 36.0 and 109.2 months respectively, and neither was reached in the PD-1 antibody group (both P>0.05). The most common site of first distant metastasis in both groups was the lung, and there was no statistically significant difference in the incidence of distant metastasis at any site (P>0.05). By univariate Cox analysis, compared with PD-1, antibody HDI reduced the risk of distant metastasis in patients with BRAFV600E/K mutations (HR=10.03, 95% CI [1.10, 91.35]; P=0.041). Subgroup analyses showed that in patients with cutaneous and acral melanoma, the difference in RFS between the HDI group and the PD-1 antibody group was not statistically significant (both P>0.05). The incidence of adverse reactions in the HDI group and the PD-1 antibody group was 83.3% and 79.1%, respectively, most of which were grade 1 or 2. No deaths related to adverse reactions occurred in both groups. Conclusion: In this study, there was no statistically significant difference in clinical efficacy and safety between PD-1 antibody and HDI adjuvant therapy for malignant melanoma patients. Patients with BRAFV600E/K mutations may benefit more from HDI. Objective: To investigate the effect of ginsenoside Rb1 (Gn-Rb1) on oxeiptosis in hepatocellular carcinoma (HCC) HepG2 cells and its possible molecular mechanism. Methods: Bioinformatic was used to analyse the effect of the expression of PGAM5, a key gene of oxeiptosis, on the survival time of HCC patients. HCC tissues and paracancerous tissues of 8 HCC patients admitted into Liaoning Provincial Tumor Hospital were selected, and the expressions of oxeiptosis-related gene proteins and mRNAs were detected by WB assay and qPCR. HepG2 cells were randomly divided into the control group and the Gn-Rb1 group (intervened with 200 µmol/L Gn-Rb1). The effects of Gn-Rb1 on the colony formation ability and the migration ability of HepG2 cells were detected by the cell clone formation assay and the cell scratch assay, respectively. ELISA was used to detect the effect on the level of ROS. Microplate was used to detect the effect on the level of LDH; the effects of Gn-Rb1 on the expressions of key gene proteins and mRNAs were detected by WB assay and qPCR. Results: Bioinformatic analysis revealed that, compared with low-expression patients, patients with PGAM5 high-expression hepatocellular carcinoma had longer overall survival time (P<0.05). In clinical hepatocellular carcinoma and paracancerous tissue samples, it was found that compared with paracancerous tissues, at protein and mRNA levels, the expressions of KEAP1 and PGAM5 in tumor tissues were significantly lower; the expression of NRF2 was significantly higher (all P<0.01); and the expression of p-AIFM1 was significantly higher (P<0.05). After HepG2 cells were intervened with 200 µmol/L Gn-Rb1, compared with those in the control group, the migratory and colony-forming abilities of HepG2 cells in the Gn-Rb1 group reduced significantly (all P<0.01), and the level of LDH increased significantly (P<0.05). Compared with the control group, at mRNA and protein levels, the expressions of KEAP1 and PGAM5 in the Gn-Rb1 group were significantly higher (all P<0.05); the expression of NRF2 was significantly lower (all P<0.05); and the expression of p-AIFM1 protein was significantly lower (P<0.01). Conclusion: Oxeiptosis is inhibited in hepatocellular carcinoma tissues, and Gn-Rb1 promotes oxeiptosis in HCC HepG2 cells by regulating the KEAP1/PGAM5/AIFM1 pathway, thus suppressing the proliferation and migration abilities of HepG2 cells. Objective To summarize the clinical characteristics, treatment outcomes, and prognostic factors of children with non-metastatic Ewing's sarcoma (ES). Methods A retrospective analysis was conducted on the clinical data of 41 children with non-metastatic ES diagnosed and treated at the Shanghai Children's Medical Center, Shanghai Jiao Tong University School of Medicine from January 2010 to December 2018. All patients underwent chemotherapy based on the RMS-2009 protocol of the center, and local treatment such as surgery and/or radiotherapy was performed according to risk grouping. The Kaplan-Meier method was used to calculate the overall survival (OS) and event-free survival (EFS) rates. Univariate prognostic analysis was performed using the log-rank test, and multivariate analysis was conducted with Cox regression. Results Of the 41 children, 21 were male and 20 were female. The median age at diagnosis was 7.7 years (range: 1.2-14.6 years). The median follow-up time for patients with event-free survival was 68.1 months (range: 8.1-151.7 months). As of the last follow-up, 33 patients were in complete remission, and the overall 5-year EFS and OS rates were (78±6)% and (82±6)%, respectively. Univariate analysis by the log-rank test showed that a tumor diameter ≥8 cm, time from diagnosis to start of local treatment 516 weeks, and incomplete surgical resection were associated with poor prognosis (P<0.05). Multivariate Cox regression analysis indicated that incomplete surgical resection (HR=8.381, 95%CI: 1.681-41.801, P=0.010) was an independent risk factor for poor prognosis in children with ES. Secondary tumors occurred in 2 cases. Conclusions A comprehensive treatment strategy incorporating chemotherapy, surgery, and radiotherapy can improve the prognosis of children with ES. Poor prognosis is associated with an initial tumor diameter ≥8 cm, while complete surgical resection and early initiation of local treatment can improve outcomes. [ABSTRACT FROM AUTHOR]

Objective: To investigate the value of allograft inflammatory factor 1 (AIF1) in immunity and prognosis of acute myeloid leukemia (AML).Methods: AIF1 expression in AML and its relationship with survival and prognosis were analyzed by bioinformatics methods. The Cancer Genome Atlas (TCGA) and genotype-tissue expression (GTEx) data analysis showed that AIF1 may be a potential oncogene in AML. The function of AIF1 was further analyzed by functional enrichment analysis, gene set enrichment analysis (GSEA),and protein interaction network (PPI) analysis. Wilcoxon signed rank test and Logistic regression analysis were used to determine the relationship between AIF1 and the clinicopathological characteristics of AML patients. The relationship between AIF1 expression and overall survival (OS) of AML patients was evaluated by Cox regression and Kaplan-Meier survival curve. Finally, the expression of AIF1 in bone marrow samples of AML patients was verified by qRT-PCR and Western blot.Results: AIF1 is highly expressed in AML and is associated with poor prognosis. Overall survival of POSTN high expression group was shorter than that of POSTN low expression group. Conclusion: AIF1 is highly expressed in AML, whose expression is correlated with overall survival of AML patients. Objective: To investigate the expression of Ficolin-3(FCN3) in hepatocellular carcinoma (HCC) and its effects on cell proliferation, migration and epithelial-mesenchymal transformation (EMT).Methods: Firstly, bioinformatics was used to ana- lyze the expression of FCN3 in HCC and its relationship with the prognosis of patients. Then, the expressions of FCN3 in HCC cells and tissues were further detected by qRT-PCR and immunohistochemistry. Huh7 stable cell lines overexpressing FCN3 were constructed, and the effects of FCN3 on the proliferation, migration and invasion of HCC cells were detected by CCK-8, scratch and Transwell assays. The expression levels of E-cadherin, N-cadherin and Vimentin were detected by Western Blot assay. The effect of FCN3 on HCC proliferation was investigated in vivo by subcutaneous tumor formation in nude mice.Results: Bioinformatics analysis showed that the expression level of FCN3 in HCC tissues was significantly lower than that in paracancer tissues, and HCC patients with high expression of FCN3 had longer overall survival time. qRT-PCR showed that mRNA expression levels of FCN3 in HCC cell lines Huh-7, Hep3B and SNU-449 were significantly decreased compared with normal hepatocyte. Immunohistochemistry showed that the proportion of FCN3 staining positive cells in HCC tissues was significantly lower than that in para-cancer tissues. The Huh7 cell line with the relatively lowest expression of FCN3 was selected for lentivirus stable FCN3 gene, and stable OE-Huh7 and no-load lentivirus stable NC-Huh7 were obtained. Transwell, scratch and CCK8 tests showed that the migration, proliferation and invasion ability were significantly reduced in OE-Huh7 group compared with NC-Huh7 group. Western Blot results showed that the expression levels of Vimentin and N-cadherin protein decreased while the expression levels of E-cadherin protein increased in OE-Huh7 group compared with NC-Huh7 group. Conclusions: The expression of FCN3 is low in HCC, and FCN3 affects the prognosis of patients. Overexpression of FCN3 can inhibit the proliferation, migration and EMT of HCC cells. Objective To evaluate the impact of peripheral nerve aggression (PNI) on prognosis in patients with stage Ⅰ-Ⅲ early-onset colorectal cancer (EO-CRC) and to create a PNI-based nomogram prediction model. Methods This study analyzed data on 5 920 EO-CRC patients aged 50 years and younger enrolled in the U.S. Surveillance, Epidemiology, and End Results (SEER) database from 2010 to 2015. The patients were divided into the training group (n=4 164) and the validation group (n=1 756) by a ratio of 7∶3. Kaplan-Meier survival curve was drawn and Cox proportional risk model was used for univariate and multivariate prognosis analysis. Based on the significant variables of the multivariate model, a nomogram was constructed, and the predictive ability was evaluated by receiver operating characteristic curve (ROC), calibration curve and decision curve analysis (DCA). Results Compared with PNI negative patients, PNI positive patients tended to be young, lesions located in the left colon, with poorer differentiation, higher TNM stage, chemotherapy intervention, and higher CEA level (P＜0.05). Kaplan-Meier curve suggested that PNI positive group had a worse prognosis than PNI negative group. Cox univariate analysis suggested that age, sex, degree of tumor differentiation, TNM stage, T stage, N stage, whether to receive radiation therapy, whether to receive chemotherapy, CEA level, maximum tumor diameter and PNI status were variables affecting the survival and prognosis of patients. Multivariate analysis showed that PNI was an independent prognostic indicator for patients with stage Ⅰ to Ⅲ EO-CRC (HR=1.73, 95%CI: 1.44-2.08, P＜0.01). The area under ROC curve of nomogram model based on PNI was more than 0.7 in both 3-year and 5-year lifetime. Conclusion PNI can be used as an independent prognostic marker in patients with stage Ⅰ-Ⅲ EO-CRC, and PNI-based nomogram model can be used as an effective prognostic tool.

Objective To investigate the relationship between the expression of LDHA, SLC16A1 and SLC16A3 genes and pathologic features and prognosis in breast cancer. Methods Tissue samples from 1 060 breast cancer patients in The Cancer Genome Atlas (TCGA) were obtained. The association of LDHA, SLC16A1 and SLC16A3 gene expressions with clinicopathological features and prognosis of breast cancer were analyzed. Survival curve were drawn by Kaplan-Meier survival analysis, and univariable and multivariable survival prognosis were analyzed by Cox proportional hazard regression model. Results LDHA expression was associated with distant metastasis (M stage) (χ2=5.560, P=0.018), estrogen receptor (ER) expression (χ2=8.532, P=0.003), and human epidermal growth factor receptor 2 (HER-2) expression (χ2=4.418, P=0.036); SLC16A1 expression correlated with age (χ2=8.040, P=0.005), ER expression (χ2=17.428, P＜0.01), and progesterone receptor (PR) expression (χ2=5.486, P=0.019). SLC16A3 expression correlated with ER expression (χ2=22.447, P＜0.01), PR expression (χ2=20.590, P＜0.01). Patients with high expression of LDHA (χ2=3.856, P=0.049), SLC16A1 (χ2=3.978, P=0.046) and SLC16A3 (χ2=5.008, P=0.025) had lower cumulative survival rates. SLC16A1(HR=1.894, 95%CI: 1.246-2.878, P=0.003) and SLC16A3(HR=1.769, 95%CI: 1.009-2.847, P=0.019) were the independent risk factors for overall survival in breast cancer patients. Conclusion LDHA, SLC16A1 and SLC16A3 are associated with certain pathologic features and poorer prognosis of breast cancer, which may provide new prognostic indicators and therapeutic targets for breast cancer treatment.

Background and purpose: Follow-up data of 6 737 patients undergoing surgery for gastric cancer were collected based on hospital registration, and the 1-, 3- and 5-years observed overall survival (OS) rates and disease-free survival (DFS) rates were analyzed to provide real-world research evidence for the prevention and control of gastric cancer and policy making in China. Methods: A total of 6 737 gastric cancer patients who underwent surgical treatment at Fudan University Shanghai Cancer center from 2015 to 2020 were included in this study. Clinical information and the follow-up endpoint data were collected through medical records review, telephone visits and death registry data linkage. The last follow-up date was November 30, 2023. Kaplan-Meier method was applied in evaluating the 1-, 3- and 5-year OS rate and DFS rate, and survival data were described by different subgroups including age group, gender, treatment period, tumor staging, and pathological characteristics. Results: With a median follow-up time of 50.99 months, the 5-year OS rate of surgically resected gastric cancer patients was 70.37%, and 5-year DFS rate in Ⅰ-Ⅲ stage cases was 69.46%. The 5-year OS rates of stage Ⅰ, Ⅱ, Ⅲ and Ⅳ were 94.32%, 82.56%, 51.01% and 23.97%, respectively. The differences in survival among patients with different age, tumor location, gross classification, Borrmann classification and Laurence classification were significant. Conclusion: Staging is an important factor directly affecting the survival of gastric cancer patients. Screening and early diagnosis and treatment in large population, especially high-risk group, should be strengthened to further improve the patients’ survival.

Background Multiple myeloma is a highly heterogeneous disease. Super-aged patients with multiple myeloma are a special group. There is a lack of evidence-based medical basis in the treatment decision and general evaluation of them, and there is also some controversy in the assessment of frailty. Objective To explore the clinical features and the influencing factors of survival and prognosis in newly diagnosed super-aged multiple myeloma (MM) patients with the bortezomib based regimen and to evaluate the best assessment model for the general status of patients with MM. Methods The clinical data of newly diagnosed super-aged patients with MM in Beijing Chao-yang Hospital, Capital Medical University from November 2013 to January 2023 were retrospectively analyzed. Survival follow-up was conducted through the medical record system of Beijing Chao-Yang Hospital, Capital Medical University until April 1, 2023, with the end points of this study of overall survival (OS) and progression-free survival (PFS) . The patients were divided into the two-drug treatment group (n=18) and three-drug treatment group (n=11) , and the clinical and genetic characteristics of the two groups were compared. The Geriatric Assessment System (GA) score, UK Myeloma Research Alliance Risk Profie (MRP) score, and Mayo score were used to evaluate the frailty status and treatment efficacy. Survival curves of OS and PFS in MM patients were plotted using the Kaplan-Meier method. Log-rank test was used to compare the survival curves of different influencing factors. Multivariate Cox hazard risk regression analysis was used to explore the influencing factors of OS and PFS in MM patients. Results The median PFS was 8.70 (1.90-43.87) months and the median OS was 17.23 (2.00-72.83) months. A total of 21 (72.41%) patients experienced disease progression (PD) or relapse, and 12 (41.38%) patients died by the final follow-up. The objective remission rate (ORR) for first-line treatment was 82.76% (24/29) , the partial remission (PR) rate was 51.72% (15/29) , the very good partial remission (VGPR) rate was 24.14% (7/29) , and the complete remission (CR) rate was 6.90% (2/29) . There was no significant difference in CR rate, VGPR rate, PR rate and ORR between the two groups of newly diagnosed super-aged MM patients (P>0.05) . Multivariate Cox hazard risk regression analysis showed that MRP debilitation (HR=0.213, 95%CI=0.049-0.924, P=0.039) , elevated serum corrected calcium (HR=0.153, 95%CI=0.041-0.570, P=0.005) and maintenance therapy (HR=4.301, 95%CI=1.219-15.169, P=0.023) were independent influencing factors of PFS in newly diagnosed super-aged MM patients, while maintenance treatment (HR=4.372, 95%CI=1.049-18.221, P=0.043) was an independent influencing factor for OS in newly diagnosed super-aged MM patients. Conclusion There is no significant difference in the efficacy and prognostic impact of two-drug or three-drug Bortezomib based treatment. Serum corrected calcium elevation and maintenance therapy are independent prognostic factors for survival. MRP score can be used to assess the prognosis of newly diagnosed super-aged MM patients.

Background Malignant melanoma (MM) is caused by the malignant transformation of epidermal melanocytes with extremely high malignant degree, and its prevention and treatment is a key issue in the field of dermatology. Objective To investigate the prognosis of patients with MM, and influencing factors of the prognosis of patients with MM. Methods A total of 205 patients with malignant melanoma from the First Affiliated Hospital of Shanxi Medical University from January 2006 to December 2021 were selected as the subjects. The clinical data were obtained by collecting information from electronic medical records of patients, the overall survival (OS) was confirmed by telephone follow-up, and the last follow-up visit until 2022-12-31. The survival curve was plotted by the Kaplan-Meier method, and Log-rank test was used to compare the survival curves of different influencing factors. Multivariate Cox proportional hazard regression model was used to analyze the influencing factors of survival outcome in MM patients. Results The 1-year overall survival, 3-year overall survival, and 5-year overall survival rates of MM patients were 94.6% (194/205) , 81.9% (168/205) and 72.6% (149/205) , respectively, the mean OS was 71.77 months and the median OS was 66.83 months. The results of survival curve analysis showed statistically significant differences when comparing the OS of MM patients with different genders, ages, tumor thicknesses, tumor ulcers, lymph node metastases, and neutrophil-to-lymphocyte ratios (NLR) (P1.94 mm (HR=2.466, 95%CI=1.419-4.284) , tumor ulcer (HR=1.821, 95%CI=1.225-2.708) , lymph node metastasis (HR=1.999, 95%CI=1.305-2.892) , and NLR≥3 (HR=1.873, 95%CI=1.108-3.166) were the influencing factors of survival outcome in MM patients (P1.94 mm, tumor ulcer, lymph node metastasis and NLR≥3 are poor prognostic factors for MM patients. Medical workers can focus on MM patients with the above poor prognostic factors and enhance care to improve their survival rate, thus better guiding the clinic treatment.

Objective To investigate the association between clinical features and survival in distant metastatic endometrial carcinoma (EC) patients, and to construct a prognostic risk model to guide the clinical practice. Methods The clinical data of EC patients with distant metastases were retrospectively collected from the SEER database between 2010 and 2017. Univariate and multivariate Cox regression analyses were applied to determine the independent prognostic factors of metastatic EC patients, and then a nomogram model was constructed based on these predictive factors. The predictive performance of the model was evaluated and internally validated using C-index, receiver operating characteristic (ROC) curve, calibration curve and decision curve analysis (DCA). In addition, the model was externally validated using the data collected from the EC patients with distant metastases admitted to the First Affiliated Hospital of Chongqing Medical University between October 2013 and December 2019. Results A total of 1 807 EC patients with distant metastases who met the inclusion and exclusion criteria in the SEER database were enrolled in this study. Multivariate Cox analysis revealed that tumor size, grade, and histological type were independent risk factors for distant metastatic EC; and race, surgery, and chemotherapy were independent protective factors for metastatic distant EC. In the training cohort, the C-index of the constructed nomogram model was 0.711 (95%CI: 0.695~0.727); in the internal validation cohort, the C-index of the model was 0.744 (95%CI: 0.721~0.768); ROC curve analysis indicated that the model had good prediction and discrimination; calibration curve showed that there was a high consistency between the predicted survival probability and the actual survival probability; meanwhile, the results of DCA curves revealed that the model had great potential for clinical utility. Furthermore, based on the 21 EC patients with distant metastases included as an external validation dataset, the results showed that the model had good predictive ability. Conclusion A survival prediction model for distant metastatic EC is established, and it can assist clinicians in evaluating the prognosis of patients and performing personalized clinical decisions for this specific population.

Objective To explore the relationship between p-FGFR1Y654 expression and clinical pathological characteristics of esophageal squamous cell carcinoma patients and its prognostic value. Methods Tumor tissue samples from 103 cases of esophageal squamous cell carcinoma and 58 normal esophageal tissues were surgically collected in the General Hospital of Western Theater between January 2017 and July 2020. Objective·To establish a prognostic model for the overall survival (OS) of hepatocellular carcinoma (HCC) based on mitochondrial genes and clinical information.Methods·The gene expression and the clinical data of 369 HCC patients and 50 controls with normal liver were downloaded from The Cancer Genome Atlas (TCGA) database. The nuclear-encoded mitochondrial genes (NEMGs) were obtained from the MitoCarta3.0 database. The "DESeq2" R package and univariate Cox analysis were used to select NEMGs [ubiquinol cytochrome C reductase hinge protein (UQCRH), ATP citrate lyase (ACLY), phosphoenolpyruvate carboxykinase 2 (PCK2), Bcl-2 homologous antagonist/killer1 (BAK1), Bcl-2-associated X protein (BAX) and Bcl-2/adenovirus E1B interacting protein 3-like (BNIP3L)] in HCC that were associated with OS of HCC and participated in dysregulation of oxidative phosphorylation, tricarboxylic acid cycle and cell apoptosis. Multivariate Cox analysis was applied to select independent risk factors for OS of HCC. A comprehensive prognostic model and a prognostic nomogram with 6-NEMG risk characteristics and TNM staging were established. By using the median of prognostic scores as a cut-off, HCC patients were classified into low-risk and high-risk group. Kaplan-Meier survival curve analysis was conducted and log-rank test was performed to evaluate the survival rates between the low-risk and high-risk group. The area under the curve (AUC) values of receiver operating characteristic (ROC) curve were calculated via using the "timeROC" package. The prognostic model for HCC was validated by using the GEO HCC cohort (GSE14520) for 1, 3 and 5 years. Finally, the relative expression level of 6-NEMG was validated in 34 clinical samples of HCC from Xinhua Hospital, Shanghai Jiao Tong University School of Medicine by using real-time quantitative polymerase chain reaction (qPCR) method.Results·Compared to 6-NEMG risk signature only (AUCs for 1, 3 and 5 years were 0.77, 0.66 and 0.65, respectively) or TNM stage only (AUCs for 1, 3 and 5 years were 0.66, 0.67 and 0.63, respectively), ROC curve analysis showed that this integrated prognostic model displayed better predictive performance for 1-year (AUC, 0.78), 3-year (AUC, 0.73) and 5-year (AUC, 0.69) OS of HCC. The Kaplan-Meier survival curve analysis showed that the OS of HCC patients in the high-risk group was significantly worse than that in the low-risk group (P=0.001). In addition, predictive performance of the prognostic model (AUC for 1, 3 and 5 years is 0.67, 0.66 and 0.74, respectively) and prognostic differences between the high-risk and low-risk group (P=0.001) were further validated in GEO (GSE14520) external cohort, and these results were consistent with the TCGA data. In addition to BNIP3L, dysregulation of five other NEMGs in the clinical HCC cohort was validated. The correlation analysis in GSE14520 and HCC clinical cohort showed a positive correlation between prognosis score and the size and number of tumors.Conclusion·A new prognostic model that combines 6-NEMG risk characteristics with TNM staging for predicting OS in HCC patients was constructed and validated. Gastric cancer, a malignant tumor with high morbidity and mortality in China, has characteristics such as high heterogeneity and poor prognosis. With the advent of the 21st century, significant progress has been made in gastric cancer diagnosis and treatment due to the rapid development of genomics, laparoscopic minimally invasive techniques, targeted therapy and immunotherapy. This article summarized the important research progress in the field of gastric cancer prevention and treatment since the 21st century, and looked forward to the future. We hope to make greater progress and breakthroughs in early screening, diagnosis and precise treatment of gastric cancer, further improve the overall survival rate of patients, and transform gastric cancer into a controllable "chronic disease". BACKGROUND: Haploidentical hematopoietic stem cell transplantation is associated with a higher rate of graft rejection and therefore often requires a higher CD34+ cell dose, but the findings reported in existing studies regarding the relationship between CD34+ cell dose and study endpoints after allogeneic hematopoietic stem cell transplantation are controversial. OBJECTIVE: To investigate the effect of CD34+ cell dose on clinical outcomes of haploidentical hematopoietic stem cell transplantation for malignant hematological diseases. METHODS: 135 patients who underwent haploidentical hematopoietic stem cell transplantation at Hematopoietic Stem Cell Transplantation Center, Department of Hematology, First Affiliated Hospital of Zhengzhou University between January 2019 and December 2021 were included. Combining the results of previous studies and our center’s experience, the cohort was divided into two groups using a CD34+ cell count of 5.0×106 /kg as the cut-off point. Clinical outcomes related to graft implantation, relapse incidence, non-relapse mortality, overall survival and progression-free survival were evaluated in both groups. RESULTS AND CONCLUSION: (1) CD34+ cell dose correlated with platelet engraftment, with platelets implanted earlier in the high-dose group than in the lowdose group (14 days vs. 16 days, P=0.013). (2) There was no significant difference in 3-year overall survival between the two groups (67.5% vs. 53.8%, P=0.257); nor was there a significant difference in progression-free survival between the two groups (65.6% vs. 44.2%, P=0.106), but stratified analysis based on disease risk index revealed an association with elevated 3-year progression-free survival in the high-dose group among low-risk patients (72.0% vs. 49.3%, P=0.036). (3) The cumulative 3-year relapse incidence was smaller in the high-dose group than in the low-dose group (16.0% vs. 33.5%, P=0.05). (4) The rate of non-relapse mortality within 100 days was greater in the high-dose group than in the low-dose group, but there was no significant difference (17.3% vs. 6.7%, P=0.070); stratified analysis revealed that non-relapse mortality within 100 days was significantly higher in the high-dose group than in the low-dose group (20.0% vs. 3.3%, P=0.046). (5) In conclusion, CD34+ cell doses >5.0×106 /kg promote early platelet implantation, improve 3-year progression-free survival in low-risk patients at transplantation and reduce the cumulative relapse incidence. However, in high-risk patients, high-dose CD34+ cells result in increased non-relapse mortality within 100 days after transplantation, which is considered to be possibly associated with an increased occurrence of severe acute graft versus host disease in the early post-transplantation period. Objective Malignant lymphomas occurring in the prostate are relatively rare. Previously, data on prostate malignant lymphoma in the Chinese population were limited to case reports, leading to an incomplete understanding of the clinical characteristics of this disease. Methods This study conducted a systematic search and review of articles published from 1983 to 2019. The aim was to clarify the clinical features and treatment outcomes of prostate malignant lymphoma in the Chinese population. Results In this cohort, primary prostate lymphoma accounted for 75%, most patients presented with stage I disease (67% ), and aggressive lymphoma (mainly diffuse large B-cell lymphoma) accounted for 73%. Among them, 23% of patients chose R-CHOP ( Rituximab-Cyclophosphamide, Hydroxydaunomycin, Oncovin, Prednisone) chemotherapy, 26% of patients chose CHOP chemotherapy, 6% of patients chose radiotherapy or a combination of radiotherapy and chemotherapy, 23% of patients chose other chemotherapy; in whom 60% of patients chose surgery to relieve the obstructive symptoms. The half-year and one-year cumulative survival rates of the patients in this cohort were 70.7% and 54.8%, respectively, and the half-year and one-year cumulative survival rates of the treated patients were 76.5% and 56.5%, respectively. Conclusion In recent years, with the ongoing standardization of lymphoma classification and the application of new drugs, there has been a significant extension in the survival of patients with lymphoma. Lymphomas that originate in or spread to the prostate remain a form of malignancy within the lymphohematopoietic system. It is essential to alleviate symptoms such as lower urinary tract obstructions while obtaining sufficient tissue to confirm the pathological subtype. Background and objective Invasive mucinous adenocarcinoma (IMA) was a rare and specific type of lung adenocarcinoma, which was often characterized by fewer lymphatic metastases. Therefore, it was difficult to evaluate the prognosis of these tumors based on the existing tumor-node-metastasis (TNM) staging. So, this study aimed to develop Nomograms to predict outcomes of patients with pathologic N0 in resected IMA. Methods According to the inclusion criteria and exclusion criteria, IMA patients with pathologic N0 in The Affiliated Lihuili Hospital of Ningbo University (training cohort, n=78) and Ningbo No.2 Hospital (validation cohort, n=66) were reviewed between July 2012 and May 2017. The prognostic value of the clinicopathological features in the training cohort was analyzed and prognostic prediction models were established, and the performances of models were evaluated. Finally, the validation cohort data was put in for external validation. Results Univariate analysis showed that pneumonic type, larger tumor size, mixed mucinous/non-mucinous component, and higher overall stage were significant influence factors of 5-year progression-free survival (PFS) and overall survival (OS). Multivariate analysis further indicated that type of imaging, tumor size, mucinous component were the independent prognostic factors for poor 5-year PFS and OS. Moreover, the 5-year PFS and OS rates were 62.82% and 75.64%, respectively. In subgroups, the survival analysis also showed that the pneumonic type and mixed mucinous/non-mucinous patients had significantly poorer 5-year PFS and OS compared with solitary type and pure mucinous patients, respectively. The C-index of Nomograms with 5-year PFS and OS were 0.815 (95%CI: 0.741-0.889) and 0.767 (95%CI: 0.669-0.865). The calibration curve and decision curve analysis (DCA) of both models showed good predictive performances in both cohorts. Conclusion The Nomograms based on clinicopathological characteristics in a certain extent, can be used as an effective prognostic tool for patients with pathologic N0 after IMA resection. Objective To evaluate the efficacy and safety of anlotinib monotherapy and combined therapy in patients with advanced pheochromocytoma/paraganglioma. Methods Nine patients with advanced pheochromocytoma/paraganglioma (PPGL) who were admitted to the Department of Urology, Sun Yat-sen University Cancer Center from January 2018 to June 2023 were collected. Patients were divided into four groups according to different treatments: anlotinib monotherapy group (3 patients), anlotinib combined with PD-1 monoclonal antibody immunotherapy group (3 patients), anlotinib combined with immunotherapy and chemotherapy group (2 patients), and anlotinib combined with chemotherapy group (1 patients). The effectiveness and safety of different treatment regiments of anlotinib were analyzed. Results Objective response rate (ORR) : (44%), Partial response(PR): (44%), Stable disease(SD): (44%), Progressive disease(PD): (11%), Disease control rate (DCR): (89%). The ORR of 2 patients with SDH gene mutation, SDHB and SDHD respectively, was 100%. Median overall survival time(OS) was 16.3 months (IQR: 11.3 ~ 21.8 months). Median progression-free survival (PFS) was 16.3 months (IQR: 9.8 ~ 20.8 months). There were 2 patients with adverse events grade ≥ 3/4, all of which were hypertension. Conclusions Anlotinib monotherapy and combined therapy have preliminary efficacy and manageable safety in the treatment of advanced pheochromocytoma/paraganglioma. [ABSTRACT FROM AUTHOR]

Background and objective For resectable non-small cell lung cancer (NSCLC), the CheckMate-816 study demonstrated that neoadjuvant chemoimmunotherapy increased the rate of complete pathologic response (pCR) by 21.8% compared with chemotherapy alone and resulted in a significant benefit in event-free survival (EFS). This study aimed to investigate the safety and feasibility of this approach in the real world. Methods Clinical data from patients with NSCLC who underwent surgery after neoadjuvant chemoimmunotherapy or chemotherapy alone in two centers were analyzed retrospectively, and subgroup analyses were performed for the chemoimmunotherapy group according to treatment cycle. The primary study endpoints were EFS and major pathologic response (MPR), and the secondary study endpoints were pCR, overall survival (OS), treatment-related adverse events (TRA.Es), and surgery-related metrics. Results As of April 2023, 89 patients had been enrolled, including 54 in the chemoimmunotherapy group and 35 in the chemotherapy alone group. MPR was achieved in 31 (57.4%) and 5 (14.3%) patients in the chemoimmunotherapy group and chemotherapy alone group, respectively (OR=8.09, 95%CI: 2.72-24.04, P<0.001); pCR was achieved in 25 (46.3%) patients in the chemoimmunotherapy group and no patient in the chemotherapy alone group (P<0.001). The median follow-up time was 22.1 months. At 24 months, the EFS rates of the chemoimmunotherapy group and the chemotherapy alone group were 77.0% and 56.7%, respectively (P=0.026), and the OS rates were 87.1% and 67.7%, respectively (P=0.020). In the neoadjuvant chemoimmunotherapy group, there was no significant difference between the 1-2 cycles and 3-5 cycles groups in terms of operation time, intraoperative blood loss, and postoperative complications. Conclusion Neoadjuvant chemoimmunotherapy was more effective than chemotherapy alone and did not increase the risk associated with surgery. Objective: To investigate the expression characteristics of low-density lipoprotein receptor-related protein 11 (LRP11) and sex determining region Y-BOX12 (SOX12) in cervical cancer tissue, and to analyze their relationship with clinicopathological characteristics and prognosis. Methods: 90 patients with cervical cancer who were admitted to the Affiliated Hospital of Xuzhou Medical University from July 2015 to October 2017 were selected. The surgically removed cervical cancer tissue and normal paracancer tissue were collected, and the positive expression rates of LRP11 and SOX12 were detected by immunohistochemical method. The expression difference of LRP11 and SOX12 in cervical cancer tissue with different clinicopathological characteristics were compared. Postoperative follow-up was conducted until October 2022, Kaplan-Meier survival curve was used to analyze the differences in progression-free survival (PFS) and overall survival (OS) of patients with cervical cancer with different LRP11 and SOX12 expression levels. Results: The positive expression rates of LRP11 and SOX12 in cervical cancer tissue were higher than those in paracancer tissue (P＜0.05). The positive expression rates of LRP11 and SOX12 in cancer tissue of patients with low differentiation, FIGO stage Ⅱ stage and pelvic lymph node metastasis were higher than those of patients with moderate and highly differentiation, FIGO stage I stage and no pelvic lymph node metastasis (P＜0.05). At a median follow-up of 35 (15-45) months. A total of 4 cases were lost and 86 cases were successfully followed-up. The 5-year PFS survival rates of patients with cervical cancer with LRP11 and SOX12 positive expression were 67.19% and 63.79%, respectively, significantly lower than 86.36% and 89.29% of patients with cervical cancer with LRP11 and SOX12 negative expression (P＜0.05). The 5-year OS survival rates of patients with cervical cancer with LRP11 and SOX12 positive expression were 85.94% and 84.48%, while the 5-year OS survival rates of patients with cervical cancer with LRP11 and SOX12 negative expression were 100% (P＞0.05). Conclusion: The positive expression rate of LRP11 and SOX12 in cervical cancer tissue significantly increased, which are related to malignant clinicopathological characteristics of cervical cancer and low survival rate of PFS. Background and objective The SMARCA4 mutation has been shown to account for at least 10% of non-small cell lung cancer (NSCLC). In the present, conventional radiotherapy and targeted therapy are difficult to improve outcomes due to the highly aggressive and refractory nature of SMARCA4-deficient NSCLC (SMARCA4-DNSCLC) and the absence of sensitive site mutations for targeted drug therapy, and chemotherapy combined with or without immunotherapy is the main treatment. Effective SMARCA4-DNSCLC therapeutic options, however, are still debatable. Our study aimed to investigate the efficacy and prognosis of programmed cell death 1 (PD-1) immune checkpoint inhibitors (ICIs) in combination with chemotherapy and chemotherapy in patients with stage III-IV SMARCA4-DNSCLC. Methods 46 patients with stage III-IV SMARCA4-DNSCLC were divided into two groups based on their treatment regimen: the chemotherapy group and the PD-1 ICIs plus chemotherapy group, and their clinical data were retrospectively analyzed. Efficacy assessment and survival analysis were performed in both groups, and the influencing factors for prognosis were explored for patients with SMARCA4-DNSCLC. Results Male smokers are more likely to develop SMARCA4-DNSCLC. There was no significant difference in the objective response rate (76.5% vs 69.0%, P=0.836) between chemotherapy and the PD-1 ICIs plus chemotherapy or the disease control rate (100.0% vs 89.7%, P=0.286). The one-year overall survival rate in the group with PD-1 ICIs plus chemotherapy was 62.7%, and that of the chemotherapy group was 46.0%. The difference in median progression-free survival (PFS) between the PD-1 ICIs plus chemotherapy group and the chemotherapy group was statistically significant (9.3 mon vs 6.1 mon, P=0.048). The results of Cox regression analysis showed that treatment regimen and smoking history were independent influencing factors of PFS in patients with stage III-IV SMARCA4-DNSCLC, and family history was an individual influencing factor of overall survival in patients with stage III-IV SMARCA4-DNSCLC. BACKGROUND: In recent years, the application of new drugs such as monoclonal antibodies, small molecule targeted drugs and immunotherapy has significantly improved the short-term efficacy and long-term survival of patients with malignant lymphoma, but autologous hematopoietic stem cell transplantation still plays an important role in the overall treatment of malignant lymphoma. Preconditioning is a very important step in the whole process of autologous hematopoietic stem cell transplantation, but there is no standard preconditioning program at present. OBJECTIVE: To investigate the safety and effectiveness of Bendamustine, Etoposide, Cytarabine (Melphalan) BeEAM as preconditioning in autologous hematopoietic cell transplantation for patients with lymphomas. METHODS: From July 5, 2021 to May 31, 2022, 11 patients with lymphoma who underwent autologous hematopoietic stem cell transplantation with BeEAM preconditioning regimen in Department of Hematology of Henan Provincial People’s Hospital were enrolled in the study. The clinical characteristics, pretreatingrelated non-hematologic toxicity, and hematopoietic reconstitution of the patients were analyzed. RESULTS AND CONCLUSION: (1) The non-hematological toxicity of BeEAM pretreatment regimen mainly included oral mucositis, diarrhea and fever at granulosa stage, with the incidence of 72.7%, 63.6%, 90.9%, respectively. (2) The median time of neutrophil implantation was 9(8-11) days, and that of platelet implantation was 10(7-16) days. (3) BeEAM pretreatment regimen for autologous hematopoietic stem cell transplantation of lymphoma has controllable nonhematologic toxicity and rapid hematopoietic reconstruction, with transplant-related mortality of 0% at 100 days and transplant-related mortality of 0% at the end of follow-up period, showing good efficacy and safety.
