Background Traditional Chinese medicine (TCM) is effective in the treatment of sepsis-induced acute lung injury (ALI) /acute respiratory distress syndrome (ARDS). However, the lack of standard and uniform outcome indicators in the design of most published clinical trials makes it difficult to combine and compare the results of similar studies, causing the impossibility of generating high-quality evidence to guide clinical decision-making. Objective To analyze the use of outcome indicators in randomized controlled trials on TCM for sepsis-induced ALI/ARDS from 2017 to 2022. Methods CNKI, Wanfang Data, SinoMed, PubMed, Embase, Web of Science and Cochrane Library were searched for RCTs on sepsis-induced ALI/ARDS, with the intervention group treated with TCM combined with other drugs or conventional treatment, and no restrictions in the control group. The ROB.2 tool of the Cochrane Review Group was used to assess the quality of literature and the classification of outcome indicators was summarized. Results 39 papers were included after screening, including 1 (2.56%) with low risk of bias, 3 (7.69%) with high risk of bias and 35 (89.74%) with concerns of bias. The outcome indicators in 5 RCTs were reported with a quality score of ≥5, indicating the complete reporting of outcome indicators. A total of 106 outcome indicators were reported and used for 443 times, including physicochemical testing outcome (55 types, 51.89%), TCM syndrome/sign outcome (2 types, 1.89%), symptom and syndrome outcome (13 types, 12.26%), long-term prognosis outcome (19 types, 17.92%), quality of life outcome (7 types, 6.60%), safety event outcome (8 types, 7.55%) and economic evaluation outcome (2 types, 1.89%). There were four outcome indicators with utilization rate>50%, including oxygenation index (82.05%), Acute Physiology and Chronic Health Evaluation Scoring System (58.97%), mechanical ventilation duration (51.28%) and interleukin-6 (51.28%). The composite outcome indicators of effective rate or TCM syndrome efficacy were used in 21 papers, involving a total of 12 times of effective rate, TCM syndrome score of 8 times and TCM symptom efficacy of 6 times. The total 106 outcome indicators were summarized and categorized sequentially including overall evaluation indicators (36 types, 33.96%), common indicators (36 types, 33.96%) and different focus indicators (34 types, 32.08%) . Conclusion At present, the selection of outcome indicators in RCTs on TCM for sepsis-induced ALI/ARDS is still nonstandard, mainly manifests in unclear differentiation between primary and secondary outcome indicators, trivialization of endpoint indicators, lack of follow-up data and characteristic indicators of TCM. A core outcome set of TCM outcome indicators for septic induced ALI/ARDS should be constructed to promote the high-quality development of TCM in the future.

Under atmospheric pressure, KOH or HCl solution was added to the butylated hydroxytoluene (BHT) oxidation system to adjust the pH, and the oxidative consumption of BHT was monitored by high performance liquid chromatography (HPLC). To investigate the oxidation properties of BHT under pressurized conditions, the temperature and pressure changes during the oxidation process were monitored using the mini-closed pressure vessel test (MCPVT). The oxidation products of BHT were characterized after crystallization and purification and the possible oxidation pathway was speculated. The results showed that under atmospheric pressure, the degradation rate of BHT was less than 5% when the pH was 4 or 6, indicating good stability. At pH 10, the degradation rate of BHT was up to 59% and the oxidation reaction was rapid. KOH and HCl decreased the initial oxidation temperature of BHT from 133.1 ℃ to 53.0 ℃ and 49.1 ℃ in a pressurized closed container, respectively. 2, 6-Di-tert-butyl-4-hydroperoxy-4-methyl-2, 5-cyclohexanedione and 2, 6-di-tert-butyl-4-methyl-4-(3, 5-di-tert-butyl-4-hydroxyphenyl)-2, 5-cyclohexadien-1-one were produced during the oxidation of BHT in both atmospheric alkaline and pressurized environments. The experimental results suggested that BHT was susceptible to oxidative degradation in alkaline and pressurized environments, producing toxic products, posing safety risks.

The change of illumination have a serious effect on pedestrian detection algorithms based on image difference, general adaptive background updating methods is difficult to eliminate the effects of illumination change. This paper proposed a new method of illumination changes elimination based on similarity of histogram distribution to solve this problem. It analyzed similarity of histogram distribution on local area between illumination changed image and normal image. It presented standard deviation of image which could use to measure similarity of histogram distribution. Through image difference experiment between two adjacent frames acquired under light frequency change environment, it shows that the method has a outstanding performance in robustness which can segment the moving targets accurately and restrain the changes of illumination significantly. Objective The purpose of this study was to investigate the correlation of serum oxidative stress with MMPs in rheumatoid patients with carotid atherosclerosis. Methods Ninety-two cases of rheumatoid patients were divided into non-carotid atherosclerosis group (group A, 45 cases) and carotid artery (group B,47 cases),while 40 cases of healthy person were selected as the control group (group C) . MMP-3, MMP-9, TIMP-1, MDA, SOD and GSH were detected. Results There were significant differences in MMP-9 and TIMP-1 between groupsA and C(P < 0.05),and in MMP-3,MMP-9 and TIMP-1 between group B and the other two groups (P < 0.05) . Compared with group C,SOD and GSH in group A have decreased significantly(P < 0.05) . There were significant differences in SOD and GSH between group B and groups A and C,and MDA in group B increased significantly compared with group A(P < 0.05) . SOD showed significant correlations with MMP-9 and TIMP-1(P < 0.05), while no correlation with MMP-3 (P > 0.05) . There were negative correlations of GSH with MMP-3 and MMP-9(P < 0.05),while was significant positive correlation with TIMP-1(P < 0.05) . Conclusion MMPs were closely related to the oxidative stress disorder in rheumatoid patients with carotid atherosclerosis, which promote the incidence of carotid artery atherosclerosis. Objective To investigate the correlation between carotid atherosclerosis and inflammatory factors in patients with rheumatoid and its clinical significance. Methods 92 patients with rheumatoid were divided into non- carotid atherosclerosis group (group A 45 cases), and carotid artery (group B 47 cases), while 40 cases of healthy people were selected as a control group ( group C) . Serum levels of TNF- a, IL-17, CRP, IMT and ICAM-1, VCAM-1 were detected. Results Serum levels of TNF-α, IL-17 of patients in A group were significantly higher than those in group C (P< 0.05), Serum levels of TNF-α, IL-17, CRP of of patients in group B were significantly different to group A and group C (P < 0.05). Serum levels of IMT, ICAM-1, VCAM-1 of A group were significantly higher than those in group C (P< 0.05), Serum levels of IMT, ICAM-1, VCAM-1 of group B were significantly higher than those in group A and group C (P<0.05) . Serum levels of TNF-α and IMT, ICAM-1, VCAM-1 showed a significant positive correlation (P<0.05), Serum levels of IL-17 and ICAM-1, VCAM-1 showed significant positive correlation (P< 0.05), Serum levels of CRP and IMT, ICAM-1, VCAM-1 showed no significant correlation (P>0.05). Conclusion Inflammatory cytokines and carotid atherosclerosis are closely related, and synergically promote the incidence of carotid artery atherosclerosis. [ABSTRACT FROM AUTHOR]

Taking No. 3 coal in Qinshui Basin as research object, factors such as solid phase of drilling fluid, water sensitivity, stress sensitivity, scaling, capillarity and polymer plugging which maybe damage coal reservoir were researched during coalbed methane drilling. Sample results analyzed show that No. 3 coal reservoir has low porosity and low permeability, high content of clay mineral. Lab evaluation results show water sensitivity, scaling, solid phase invasion of drilling fluid and polymer plugging are factors which induce the coal reservoir damage. Drilling fluid which has suitable salinity, suitable density, anti-scaling, and removable drilling fluid agents can protect coal reservoir from damage during drilling for pinnate horizontal wells. Orthotopic xenograft model of human brain cancer cells is a good preclinical model for evaluation of antitumor compounds. In the present study, an orthotopic xenograft model of U87MG-m Cherry-luc was established in Balb/c nude mice and the tumor growth was monitored via imaging technology including magnetic resonance imaging (MRI), in vivo imaging (IVI) and micro-CT. Furthermore, the model was evaluated with a positive drug temozolomide. Our data suggest that integrated imaging technology including MRI, IVI and micro-CT in orthotopic xenograft model can be used to facilitate monitoring of cancer progression and evaluate antitumor activity of drugs against glioma.

Poly (ADP-ribose) polymerase 1/2 (PARP1/2) can catalyze the poly (ADP ribose) (PAR) substrate protein modification and play an important role in the regulation of DNA damage repair, cell death and transcriptional activity. The PARP inhibitor olaparib (AZD2281) can be used as a sensitizer of radiotherapy and chemotherapy in the cancer treatment. Through establishment of biological fluorescent labeled 4T1 ectopic breast tumor model, we found that olaparib exhibited a poor effect on 4T1 breast cancer alone. However, in the combination with Taxol, olaparib significantly increased the anti-tumor effect of Taxol, and reduced the PAR levels of the tumor tissues. Importantly, olaparib did not amplify the toxicity of chemotherapy drugs. This study suggests that olaparib is a representative of the PARP inhibitor that can enhance Taxol’s antitumor effect in the 4T1 ectopic breast tumor model, which sets the foundation for future study of the mechanism of olaparib action.

Hypoxia occurs in chronic and acute vascular diseases and tumor formation. The ability of tumor cells to maintain a balance between an adaptation to hypoxia and cell death is regulated by a family of transcription factors called hypoxia-inducible factor 1 (HIF-1). Tumor hypoxia mediated by HIF-1 would facilitate the likelihood of resistance to chemotherapy and radiotherapy, proliferation, metastasis and the invasive potential; all of which culminate in a decrease in patient survival. And HIF-1 alpha subunit decides the activity of HIF-1, which is regulated by oxygen. So understanding the role of HIF in signal pathway, drug resistance mechanism and its feature is crucial for developing novel anticancer therapies. In recent years, more attentions have focused on HIF-1 alpha inhibitors. It is expected that development of more potent and selective HIF inhibitors will provide an effective treatment of cancer and other HIF-related diseases. So we will focus on the biological characteristics and mechanism of HIF-1 to review currently studied HIF-1 inhibitors.

Hypoxia is a general characteristic of most solid malignancies and intimately related to cancer progression. Homeostatic response to hypoxia is primarily mediated by hypoxia inducible factor-1alpha (HIF-1alpha) that elicits transcriptional activity through recruitment P300 coactivator. Targeting the interaction of HIF- alpha and P300 would thus constitute a novel approach for cancer treatment by suppressing tumor angiogenesis and metastasis. Here, a screening assay was developed for inhibitors targeting the interaction between HIF-1alpha and P300. The nucleotide sequence of human HIF-1alpha and P300 were cloned into pBIND and pACT vectors, named pBIND-HIF1alpha and pACT-P300. The interaction of HIF-1alpha and P300 was identified in HEK293 cell using mammalian two-hybrid system. And compound chetomin decreased their interaction in this mammalian two-hybrid system. We further verified HIF-1 inhibition effect of chetomin in U251-HRE cells. Therefore, we established a screening assay combined HIF-1alpha and P300 mammalian two-hybrid system and U251-HRE reporter assay for HIF-1 selective inhibitors.

Hypoxia-inducible factor-1 (HIF-1) is a key transcription factor on hypoxia responses in mammalian tissues. HIF-1 plays as a positive factor in solid tumor and leads to hypoxia-driven responses that enhance its downstream gene expression for tumor growth and survival. LXY6099 was obtained by the structural modification and optimization of manassantin A (MA) as a high potent HIF-1 inhibitor. Antitumor activity of LXY6099 was observed in this study. LXY6099 with an IC50 value of 2.46 x 10(-10) mol x L(-1) showed more sensitive inhibition activity to HIF-1 than that of MA detected by reporter gene assay (> 100 folds). It showed strong inhibition on the growth of human solid tumor cell lines. Furthermore, LXY6099 exhibited significant antitumor activity against established human tumor xenografts in nu/nu mice with treatment of MX-1 breast cancer. Thus, LXY6099 as a novel HIF-1 inhibitor could be further developed into anti-cancer agents.

ATP-binding cassette transporter A1 (ABCA1) promotes cholesterol and phospholipid efflux from cells to lipid-poor apolipoprotein A-I (apoA-I), and plays a key role in the initial steps of the whole process of reverse cholesterol transport (RCT). Upregulation of ABCA1 is beneficial for atherosclerosis (AS) prevention and/or therapy, which indicated that ABCA1 was a target for anti-AS drug development. In the previous study, a high-throughput screening method was established using ABCA1p-LUC HepG2 cell line to find the upregulators of ABCA1. In the present study, compound 2030421B was found using this method, with EC50 of 0.50 microg x mL(-1). The compound was further identified as an upregulator of ABCA1 expression by real-time quantitative reverse transcription-polymerase chain reaction (RT-PCR) and Western blotting analysis. Studies also showed that the 2030421B could induce apoA-I-mediated cholesterol efflux and inhibit lipids uptake into mouse peritoneal macrophages RAW264.7.

Objective: To investigate the differences of clinical manifestation and therapy of organophosphorus pesticide poisoning (OPP) between oral exposure and occupational exposure in field work., Methods: From July 2007 to July 2010, 85 patients with acute severe OPP were treated in a hospital, which were divided into oral poisoning group (51 cases) and non-oral poisoning group (34 cases). The differences of clinical manifestations, curative effects and prognosis between two groups were compared., Results: The rates of myoclonus and ataxia in cases with moderate poisoning of oral poisoning group were 86.4% and 90.9%, which were significantly higher than those (50.0% and 55.0%) of non-oral poisoning group (P<0.05 or P< 0.01). The rates of myoclonus, lung fluid and coma in cases with severe poisoning of oral poisoning group were 100.0%, 89.7% and 93.1%, respectively, which were significantly higher than those (71.4%, 64.3% and 50.0%) of non-oral poisoning group (P<0.05). The mean detoxification hours in cases with moderate poisoning and cases with severe poisoning of non-oral poisoning group were (35.0 +/- 6.2) and (45.0 +/- 11.1) hours which were significantly lower than those [(49.0 +/- 7.7) and (77.0 +/- 10.3) hours] in cases with moderate poisoning and cases with severe poisoning of oral poisoning group (P<0.05). In 24, 48 and 72 h after treatment, the cholinesterase (ChE) activities of non-oral poisoning group were higher than those of oral poisoning group (P< 0.05 or P<0.01). The used doses of pyraloxime methylchloride (PAM-Cl) or atropine and the used total dose of atropine in non-oral poisoning group were lower than those in oral poisoning group (P<0.05 or P<0.01)., Conclusions: The clinical manifestation of non-oral poisoning group is different from the clinical manifestation of oral poisoning group due to the high morbidity of OPP occurred at field site in summer. The used doses of atropine and PAM-Cl are less and the ChE activity recovers quickly for non-oral poisoning group.

Objective: To observe the dynamic changes of CD3+, CD4+, and CD8+ T lymphocytes in the peripheral blood of patients with sepsis and discuss the clinical significance., Methods: Sixty-four patients admitted in the Emergency Center and Emergency Intensive Care Unit of the Second Hospital of Wenzhou Medical University between August, 2007 and July, 2009 were enrolled in this study. CD3+, CD4+, and CD8+ T lymphocytes in the peripheral blood were detected by flow cytometry on days 1, 7 and 14 after admission, and the results were compared between the patients with improvement of the condition and those without improvement, with 20 healthy subjects as the control group., Results: On day 1 after admission, CD3+ and CD4+ T lymphocytes and CD4+/CD8+ T cell ratio were obviously lower in the 2 groups of patients with sepsis than in the control group (P<0.05), but no significant difference was found in CD8+ T lymphocytes. The sepsis patients with clinical improvement showed significant higher CD3+ and CD4+ T lymphocyte percentages and CD4+/CD8+ T cell ratio than those without improvement on day 1. In the patients with clinical improvement, CD3+ and CD4+T lymphocytes and CD4+/CD8+ T cell ratio increased gradually with time and till day 14, they were comparable with the control levels; in the patients without improvement, CD3+ and CD4+ T lymphocytes and CD4+/CD8+ T cell ratio showed no obvious alterations in the course of observation., Conclusion: Immune imbalance occurs in patients with sepsis represented by lowered CD3+ and CD4+T lymphocyte percentages and CD4+/CD8+ T cell ratio in relation to the severity of the condition. CD3+ and CD4+ T lymphocytes and CD4+/CD8+ T cell ratio can be used as the indicators for assessing the severity of sepsis.

Objective: To explore the gene expressions of LTC4 synthase homologs in concanavalin A (Con A)-induced mouse hepatitis and regulation role of cyclosporine A (Cs A) treatment., Methods: Male Balb/c mouse liver injury model was developed by iv injection of Con A (20 mg/kg) and protected by Cs A pretreatment (150 mg/kg) before Con A administration. Blood samples were collected at indicated times after Con A treatment with or without Cs A pretreatment. Liver damage was assessed by serum transaminase ALT and AST measurement and histological evaluation. Meantime, three LTC4 synthase homolog gene expressions were determined by RT-PCR., Results: Serum ALT and AST upregulation were accompanied with histological damage at 2 h after Con A administration, and further aggravated at 8 h. mGST2 gene expression increased 1.7 fold at 2 h and 1.9 fold at 8 h, while the expression of LTC4 S and mGST3 changed little. Pretreatment with Cs A prevented mouse liver from injury by Con A and partly inhibited the mGST2 gene expression upregulation., Conclusions: Administration of Con A in mouse lead to a significant increase of mGST2 gene expression without any significant effect on LTC4 S and mGST3 mRNA levels. Cs A pretreatment results in protection of liver damage, whereas fails to fully inhibit the increase of mGST2 gene expression.
