To establish a rapid and accurate method for the determination of Clostridium butyricum in microbial feed additives, different counting media and identification methods were compared and the samples in market were determined. The results showed that Clostridium butyricum could grow in sulfite iron agar, TSC agar and TSN agar, while the colony counting result of ferric sulfite agar medium was the highest. Clostridium butyricum could be identified by microbiochemical identification tubes, 16S rRNA gene amplifying and sequencing, qPCR amplification and MALDI-TOF MS, while the qPCR amplification was the most rapid and accurate as well as widely applied method. Two samples from market were determind with results of 9.1×108, 1.2×109 CFU/g respectively. Typical colonies were identified by qPCR with positive results. The study indicates that Clostridium butyricum in microbial feed additives can be effectively determined by ferric sulfite agar plate counting along with biochemical and molecular biological identification. Chestnut is often used in cloudy beverage for its good nutritional value and health care function. Due to abundant starch in chestnut, the chestnut beverage is easy to precipitate or stratify. Therefore, in this study, microcrystalline cellulose (MCC) and agar were composited to improve the stability of the chestnut suspension. The determination of particle size, viscosity, rheology and Turbiscan were carried out to evaluate the effect of MCC concentration on the stability of chestnut suspension. The results showed that the chestnut suspension was relatively stable with smaller particle size and higher potential value when MCC concentration was 2.4 g/L and agar concentration was 0.4 g/L, but little change in particle size occurred during thermal acceleration process. When MCC concentration was 2.4 and 2.8 g/L, the chestnut suspension had higher viscosity, and could provide better suspension stability for suspended particles in the system. The rheological result showed that when the concentration of MCC was 2.4 g/L, a relatively stable network structure could be formed in the system, resulting in the stronger damage resistance. The Turbiscan analysis showed that the TSI value of the system was relatively small, leading to stable within a short time when MCC concentration was 2.4 and 2.8 g/L.

【Objective】The present paper aimed to screen antagonistic bacterial strains of mango anthracnose, so as to lay a theoretical basis for biological control of mango anthracnose and provide strain resources.【Method】Colletotrichum asianum,the dominant species of mango anthrax, was used as the target, and 37 strains of Bacillus spp. obtained in previous studies were used for confrontation culture and in vitro inoculation test. The antagonistic strains with obvious control effect were selected and their antagonistic mechanism was analyzed.【Result】The results of confrontation test showed that the inhibitory rates of 10 antagonists against C. asianum were more than 78.00%,among which the inhibitory rates of M12-5,L18-7,L19-3,L17-3 and M11-6 reached 96.24%,94.94%,94.10%,91.25% and 90.00%,respectively. Eight strains of Bacillus spp. with obvious control effect were obtained by in vitro inoculation, among which strains L18-7 and L16-4 had the best control effect, with the average control efficiency of 83.00% and 79.16%,respectively. There was no significant difference between them(P＞0.05). Combined with morphological characteristics, 16S rDNA sequence and phylogenetic tree analysis based on rpoB gene sequence, strain L18-7 could be identified as B. velezensis. Strain L18-7 could produce extracellular protease and cellulase, and could produce active antagonists in five media including Luria-bertani(LB),beef peptone(NA) and potato glucose agar(PDA) etc. Strain L18-7 carried genes related to antibiotic production such as mycB,ituA,yndJ,fenB and bacA.【Conclusion】The antagonistic bacterium strain L18-7 of mango anthracnose was identified as B. velezensis. The strain grew well on LB medium at 28 ℃.It could produce antagonistic substances, extracellular protease and cellulase and carried related genes of antibiotics such as mycB,ituA,yndJ,fenB and bacA. A bacterial strain exhibiting antimicrobial activity was identified in farmland soil and screened using the transparent circle and agar diffusion methods. The strain was identified through morphological observation, physical and chemical analysis, and 16S rDNA sequence alignment, and the antimicrobial substances produced by the strain were identified using extracellular enzyme activity assay, protease hydrolysis, ammonium sulfate precipitation, membrane analysis, and stability analysis. The results showed that the identified strain was Lysobacter enzymogenes L-43, which demonstrated good antibacterial activity and no hemolysis. The antibacterial substances produced by L. enzymogenes L-43 had a broad antimicrobial spectrum and maintained a good inhibitory effect on Gram-positive bacteria. The diameter of the inhibition zone exceeded 10 mm. The antibacterial substances had no lysozyme activity and did not contain hydrogen peroxide. They were sensitive to protease E, protease K, pepsin, and trypsin, and the antibacterial activity recovery rate was 73.43%~79.00%. After membrane analysis, when the molecular weight cut-off of the dialysis bag was 3 500 u and 10 000 u, the antibacterial activity recovery rates were 93.43% and 59.82%, respectively. It was preliminarily determined that the antibacterial substances were mainly polypeptides with molecular weights between 3 500~10 000 u. The substances showed good thermal, acid-base, metal ion, chemical reagent, and storage stability, and the antibacterial activity recovery rate was 85.66% after treatment at 121 °C for 30 min. The antibacterial recovery rate was higher than 80% at pH 2~10, further confirming that the antibacterial substance was a polypeptide. In conclusion, the antibacterial substances produced by L. enzymogenes L-43 are stable peptides with a broad antibacterial spectrum. To develop a kind of food packaging material with good hydrophobicity, agar/konjac glucomannan (AK) composite film incorporated with ethyl cellulose (EC) was developed. The effect of the addition of EC on the microstructure, mechanical properties and water resistance ability (swelling rate, dissolution rate and water vapor transmittance) of the composite film were explored by scanning electron microscopy (SEM), Fourier infrared spectroscopy (FTIR), X-ray diffraction (XRD), and texture analyzer. The results showed that the EC molecular chain in AKE composite film was not completely extended, and the addition of EC inhibited the crystallization of agar. Compared with AK composite film, the AKE-25% composite film had the highest elongation at break (15.08%) and water vapor transmittance (9.88×10−12·g·cm/(cm2·s·Pa)). The dissolution rate decreased from 40.70% to 25.64%. Therefore, the inclusion of EC improved the ductility and water resistance for Agar/KGM/EC composites.

In order to solve the problems such as poor stability and rapid precipitation of passion fruit juice, the influence of the different stabilizers on the stability of passion fruit juice was explored using the automatic stability analyzer based on the space and time resolved extinction profiles technology (STEP-Technology) in the current study. The sedimentation rate, the integral transmittance and transmission infrared spectrum were selected as the main indexes to determine the stabilizing effects of agar and/or sodium carboxymethyl cellulose (CMC-Na) on the passion fruit juice stability. Results showed that, addition of CMC-Na resulted smoother light extinction curves compared to agar, indicating CMC-Na might exert better stabilizing effects when single stabilizer was used. The optimal stabilizing effect being obtained by addition of CMC-Na at 0.30%~0.35% with the lowest settlement rates ((0.0209±0.0003) %/s and (0.0181±0.0002) %/s, respectively) being recorded. When these two stabilizers were applied simultaneously, it was found that increase in CMC-Na and decrease in agar led to the smoother light extinction curves and enhanced stability of juice. The optimal stability were reached by addition of 0.35% CMC-Na+0.15% agar and 0.30% CMC-Na+0.20% agar, which showed the lowest settlement rates ((0.0231±0.0004) %/s and (0.0209±0.0004) %/s, respectively). These findings could provide a scientific basis for the development of passion fruit juice with improved stability.

A composite antibacterial film was prepared using agar and carrageenan as film-forming substrates, glycerol as the plasticizer, and different concentrations of the antibacterial agent nisin, and its physical and chemical indexes were comprehensively evaluated. This study was aimed at preventing microbial growth on blueberries and ameliorating the environmental pollution caused by traditional petroleum-based packaging materials. When the concentration of nisin in the compound antibacterial film was 0.28%, the antibacterial widths of Staphylococcus aureus and Listeria sp. were 3.326 and 1.812 mm, respectively; the water solubility, light transmittance, and tensile strength of the film were 45.01%, 79.78%, and 16.09 MPa, respectively. Therefore, the antibacterial film exhibited good water solubility and antibacterial, optical, barrier, and mechanical properties. Blueberries packaged with this antibacterial film at 4 and 20 ° had 4.20 and 2.46 lg cfu/g, respectively, of microbial content at the end of storage, and exhibited lower nutrient loss. Allelochemical signaling substances released by host roots and rhizosphere microbes play an important role in the host-seeking, mate-searching, and danger escape of root-knot nematodes (RKNs). At present, in order to develop chemoattractants and repellents of plant and microbial sources, it is an international hotspot and frontier for nematologists to identify semiochemicals and related molecular targets in the pre-infection stage of RKNs. This review focused on the chemotaxis and allelochemicals of RKNs, including attractive and repellent compounds, and summarized the progress on Trojan horse mechanism and trapping formation of RKNs. Furthermore, the application of the chemotactic models, including the plate agar and its modified model, Pluronic gel model, and the sand/soil model, on the nematodes chemotaxis test were summarized. The experiment was to explore Chinese medicine effective component from the rest of use value of waste liquid. Using chemical analysis method and AGAR plate punching method to assess the nutritional value and antibacterial effect, the determination of benefit, resistance to cold qingrejiedu, promote digestion and promoting blood circulation to remove blood stasis four categories a total of 21 kinds of commonly used traditional Chinese medical conventional nutrients content and pH value changes, to evaluate the antibacterial effect of Chinese patent medicine dregs on Escherichia coli, Staphylococcus aureus and Salmonella typhimurium. The results showed that the dry matter content of Tongmai granule (sucrose added) was the highest (99.10%), and that of Pharyngitis tablet was the lowest (71.86%). The crude protein (14.83%) and crude fat (9.52%) of Shenantler Bushen tablet were the highest, the crude protein (5.13%) of Ganmaoling capsule was the lowest, and the crude fat (0.88%) of Xiaomi Shuan was the lowest. Wujiashen Guiqi essence crude fiber content was the highest (61.68%), Liqi Shuxin tablet crude fiber content was the lowest (18.23%). The content of neutral detergent fiber (83.12%) and acid detergent fiber (69.98%) in compound Dantong tablet was the highest, the content of neutral detergent fiber in Liqi Shuxin tablet was the lowest (47.58%), and the content of acid detergent fiber in Care-guan preparation was the lowest (27.61%). The ash content of Qinglin granule was the highest (35.90%), and Baobao Le was the lowest (3.55%). Ginseng antler Bushen tablet, Qingliang Buccal tablet, pharyngitis tablet, Tongqiaobiyan tablet, Lianhua Qingwen capsule, Tongmai granule (without sucrose) and Danning tablet have certain antibacterial effects on Escherichia coli, Staphylococcus aureus and Salmonella typhimurium, and the antibacterial effect of different Chinese medicinal dregs are different. The experiment indicates that 21 kinds of Chinese medicine dregs all have certain nutritional value and bacteriostatic effect, which can be used as substitutes for bulk feed materials. Objective: To establish and evaluate a microbial sensitivity test method for Neisseria gonorrhoeae based on resazurin coloration., Methods: Based on the broth microdilution method, resazurin was added as a live bacteria indicator. WHO G, a WHO gonococcal reference strain, was used to optimize the incubation time for resazurin-stained bacteria and the color change was visually observed to obtain the results. Agar dilution method (the gold standard) and resazurin-based microdilution assay were used to determine the minimum inhibitory concentration (MIC) of azithromycin, ceftriaxone, and spectinomycin for 3 reference strains and 32 isolates of Neisseria gonorrhoeae . The results were analyzed based on essential agreement (EA), which reflected the consistency of the MIC values, category agreement (CA), which reflected the consistency in the determination of drug resistance, intermediary, and sensitivity, very major error (VME), which reflected false sensitivity, and major error (ME), which reflected pseudo drug resistance, to evaluate the accuracy of resazurin-based microdilution assay as a microbial sensitivity test of of Neisseria gonorrhoeae . CA and EA rates≥90% and VME and ME rates≤3% were found to be the acceptable performance rates., Results: The results obtained 6 hours after resazurin was added were consistent with those of the agar dilution method and the resazurin-based microdilution assay was established accordingly based on this parameter. The EA of resazurin-based microdilution assay for measuring the MIC results of azithromycin, ceftriaxone, and spectinomycin was 97.1%, 91.5%, and 94.3%, respectively, and the CA was 88.6%, 94.3%, and 94.3%, respectively. The VME was 0% for all three antibiotics, while the ME was 11.4%, 5.7%, and 5.7%, respectively., Conclusion: The resazurin-based microdilution assay established in this study showed good agreement with agar dilution method for measuring the MIC of antibiotics against Neisseria gonorrhoeae . Moreover, the sensitivity results of this method were highly reliable and could be easily obtained through naked eye observation. Objective: To explore the relationship between extracellular enzymes activity and virulence of Candida glabrata clinical isolates based on the infection model of Galleria mellonella larvae. Methods: Using experimental research methods, 71 strains of non-repetitive Candida glabrata were collected from Qinghai Provincial People's Hospital from June 2021 to January 2022. Bovine serum protein agar medium, egg yolk agar medium, sheep blood agar medium, Tween-80 agar medium and triglyceride agar medium were used to detect the aspartyl protease activity, phospholipase activity, hemolysis activity, esterase activity and lipase activity of Candida glabrata. Median lethal concentration (LC 50 ) was calculated by using 1.25×10 8 CFU/ml,2.50×10 8 CFU/ml,3.75×10 8 CFU/ml,5.00×10 8 CFU/ml suspension of Candida glabrata ATCC2001 to infect Galleria mellonella larvae. Histopathological and etiological analysis was performed to determine whether the infection model was successfully established. The clinical isolates of Candida glabrata were configured to infect Galleria mellonella larvae with LC 50 concentration to detect the pathogenicity of Galleria mellonella larvae.Spearman test or Pearson test were used to analyze the correlation between the extracellular enzyme activity of Candida glabrata clinical isolates and the pathogenicity of Galleria mellonella larvae. Results: 71 strains of Candida glabrata isolated clinically were detected to have low hemolytic activity after 2 days of culture. Aspartyl protease was detected after 4 days of culture, among which 7 strains (9.86%), 19 strains (26.76%) and 45 strains (63.38%) showed low, medium and high aspartyl protease activity. After 7 days of culture, 71 strains did not detect phospholipase, esterase and lipase activities. Candida glabrata on Galleria mellonella larvae of LC 50 =2.5×10 8 CFU/ml Fungal spore were found in the intestinal tissue pathological section of Galleria mellonella larvae in the experimental group, and Candida glabrata was identified by the microbial Mass Spectrometry after culture, while no fungi were found in the pathological section and culture of the control group. Spearman test shows that, there was a linear positive correlation between aspartyl protease activity and the survival rate of Galleria mellonella larvae ( r = 0.73, P <0.01), the difference was statistically significant.Pearson test shows that, there was no significant linear relationship between hemolytic activity and survival rate of Galleria mellonella larvae ( r = 0.16, P = 0.34), the difference was not statistically significant. Conclusion: The clinical isolates of Candida glabrata in this study had aspartyl protease activity and low hemolytic activity, but no phospholipase, esterase and lipase activity. The activity of aspartyl aspartyl protease of Candida glabrata was positively correlated with the pathogenicity of Galleria mellonella larvae.

Purpose: Modified agar pre-embedded paraffin embedding method was proposed to evaluate the effects on tissue integrity, histological morphology, protein and DNA detection in small specimens of core needle biopsies., Methods: The core needle biopsy specimens of 10 patients with oral mucosal squamous cell carcinoma were subjected to modified agar pre-embedded paraffin embedding using molded embedding molds and conventional paraffin embedding respectively, the dehydration time of the former was 3.5 h and that of the latter was 12 h. After tissue treatment, H-E staining, histological morphology, immunohistochemistry (IHC), and DNA fluorescence in situ hybridization (FISH) were performed, respectively. The results were compared and analyzed using GraphPad Prism 9 software package., Results: The modified agar pre-embedding method was less difficult to perform than the agar pre-embedding method, and easier to be promoted. Compared with conventional paraffin embedding method, the tissue dehydration time was significantly reduced(P＜0.001), and the results of microscopic histological morphology and subsequent IHC and FISH assays were reliable., Conclusions: The modified agar pre-embedded paraffin embedding method meets the requirements of clinical pathological diagnosis for tissue processing, and is worthy of clinical application for core needle biopsy specimens.

Glioblastoma is the most common primary cranial malignancy, and chemotherapy remains an important tool for its treatment. Sanggenon C(San C), a class of natural flavonoids extracted from Morus plants, is a potential antitumor herbal monomer. In this study, the effect of San C on the growth and proliferation of glioblastoma cells was examined by methyl thiazolyl tetrazolium(MTT) assay and 5-bromodeoxyuridinc(BrdU) labeling assay. The effect of San C on the tumor cell cycle was examined by flow cytometry, and the effect of San C on clone formation and self-renewal ability of tumor cells was examined by soft agar assay. Western blot and bioinformatics analysis were used to investigate the mechanism of the antitumor activity of San C. In the presence of San C, the MTT assay showed that San C significantly inhibited the growth and proliferation of tumor cells in a dose and time-dependent manner. BrdU labeling assay showed that San C significantly attenuated the DNA replication activity in the nucleus of tumor cells. Flow cytometry confirmed that San C blocked the cell cycle of tumor cells in G&#95;0/G&#95;1 phase. The soft agar clone formation assay revealed that San C significantly attenuated the clone formation and self-renewal ability of tumor cells. The gene set enrichment analysis(GSEA) implied that San C inhibited the tumor cell division cycle by affecting the myelocytomatosis viral oncogene(MYC) signaling pathway. Western blot assay revealed that San C inhibited the expression of cyclin through the regulation of the MYC signaling pathway by lysine demethylase 4B(KDM4B), which ultimately inhibited the growth and proliferation of glioblastoma cells and self-renewal. In conclusion, San C exhibits the potential antitumor activity by targeting the KDM4B-MYC axis to inhibit glioblastoma cell growth, proliferation, and self-renewal.

Objective: To explore the carrier status of carbapenems-resistant Klebsiella pneumoniae (CRKP) plasmids in burn patients and analyze the correlation of these plasmids with the transmission of CRKP. Methods: A retrospective observational study was conducted. A total of 26 CRKP strains, which were isolated from the clinic-related samples of 22 burn patients (with 20 males and 2 females, aged (42±16) years) admitted to the First Affiliated Hospital of Army Medical University (the Third Military Medical University) from January to December 2017, were collected and individually numbered. The plasmids of the strains were extracted by alkali lysis. After determination of the plasmid concentration by a nucleic acid concentration detector, the agarose gel electrophoresis was used to visualize the bands, and rough plasmids typing was performed. The plasmid of the smallest numbered CRKP in each plasmid type was transformed into competent Escherichia coli ( E. coli ) strain Top10 (hereinafter referred to as TOP10 strain). The growth of each transformed strains and a Top10 strain cultivated in ampicillin containing Luria-Bertani (LB) agar medium overnight was observed, and the proportion of successful transformation was calculated. The plasmids from the smallest numbered plasmid carrying CRKP strain of successfully transformed Top10 strains (hereinafter referred to as the smallest successfully transformed strain) and correspondingly numbered CRKP were extracted, and then, the agarose gel electrophoresis was used to visualize the bands. Aforementioned successfully transformed strains and a TOP10 strain were used for the antimicrobial susceptibility testing with 17 antibiotics commonly used in clinic. The plasmid from the smallest successfully transformed strain was sequenced using the next-generation sequencing technology. Bioinformatics analyses such as protein-coding gene prediction and protein sequence alignment were performed successively. The sequence was subsequently named pKP03-NDM1 according to the carrying of drug resistance gene. According to the whole genome sequence of the plasmid carried by the smallest successfully transformed strain, the polymerase chain reaction, agarose gel electrophoresis, and gene sequencing were used to detect the New Delhi metallo-beta lactamase-1 ( bla NDM-1 Plasmids were successfully extracted from 26 CRKP, with mass concentrations ranging from 19.3 to 189.8 ng/μL. Each of the 26 CRKP carrying plasmids showed at least one band longer than 2 500 bp in the agarose gel electrophoresis, which were roughly divided into 6 patterns of A, B, C, D, E, and F. After overnight cultivation, no growth of strains was observed in LB agar medium containing ampicillin inoculated with the TOP10 strain or TOP10 strains transformed by the plasmid of CRKP patterning A, B, D, or E. In contrast, TOP10 strains transformed by the pattern C plasmid from NO.3 CRKP and the pattern F plasmid from NO.15 CRKP resulted in numerous colony growths, and those transformed strains were named as TOP10-pKP03 and TOP10-pKP15, respectively. The proportion of successful transformation was 1/3. The plasmid carried by TOP10-pKP03 showed a single band in the agarose gel electrophoresis, which was the same size as the largest band of the plasmid from NO.3 CRKP. The TOP10 strain was sensitive to the 17 antibiotics commonly used in clinic. TOP10-pKP03 and TOP10-pKP15 were resistant to penicillins, cephalosporins, and carbapenems but remained sensitive to monocyclic β-lactam, aminoglycosides, quinolones and tigecycline. In addition to bla NDM-1 , this plasmid carried ble MBL , T4SS , bleomycin resistance gene, conjugation transfer elements, and relaxase, etc. The plasmid showed 99% nucleotide identity similarity and the same length to the plasmid pJN24NDM1 extracted from an E. coli isolate JN24. Totally 16 (61.5%) CRKP were confirmed to carrying bla NDM-1 gene, among the ST typing of the 16 strains, 11 strains were ST11, while ST215, ST260, ST395, ST2230, and new ST had 1 strain each. Among the ST typing of 10 bla NDM-1 -negative CRKP, 8 strains were ST11, while ST395 and ST2230 had 1 strain each. Conclusions: A bla NDM-1 gene carrying plasmid pKP03-NDM1 was extracted and sequenced from CRKP isolated from burn patients, with a high plasmid carrying rate. Meanwhile, this plasmid may mediate inter-CRKP and CRKP- E. coli horizontal transfer of bla NDM-1 , leading to transmission of antimicrobial resistance.
