Amyotrophic lateral sclerosis (ALS) is a neurodegenerative disease characterized by progressive involvement of upper motor neuron (UMN) and lower motor neuron (LMN). With the rapid development of sequencing technique, genetic screening of ALS improved from a single locus site sequencing to whole exome sequencing (WES) or whole genome sequencing (WGS), more and more pathogenic genes affecting the occurrence and development of diseases have been discovered. At present, the pathophysiological mechanisms of ALS mainly include abnormal protein aggregation, abnormal DNA or RNA function, changes in cytoskeleton and axon dynamics, autophagy process, lysosome dysfunction, and mitochondrial dysfunction. Exploring the genetic mechanism of ALS has great significance to elucidate its pathophysiological mechanism. This paper reviews the main pathogenic genes, genetic research methods and related pathogenic mechanisms of ALS, reveals the pathogenesis and development of the disease and related pathophysiological mechanisms, and provides new strategies for the treatment of ALS. 7T ultra-high field MRI (7T MRI), with its high resolution, high signal-to-noise ratio (SNR) and high contrast, is superior to conventional field intensity MRI in clinical applications such as diagnosis of hippocampal sclerosis (HS) and focal cortical dysplasia (FCD), assisting in the identification of pathological subtypes of the both, locating the scope of surgical excision, and has made progress in the diagnosis and treatment of drug-resistant epilepsy (DRE). Functional movement disorder (FMD) is a common conversion disorder, and those who involve the face are called functional facial disorders. It seriously affects patients' social contact and work. Functional neurological disorder (FND) is a disorder of the autonomic motor or sensory system with well-defined symptoms, which covers functional movement disorder, psychogenic non-epileptic seizures, functional sensory disorder, functional cognitive disorder, persistent postural perceptual dizziness and so on. Its complex and diverse characteristics make the diagnosis and treatment of the disease and research difficult, so FND has long been a "gray area", but its high incidence, disability rate and high consumption of medical resources remind us that FND cannot be ignored. Vertigo and dizziness are common clinical symptoms. Although most patients have a good prognosis, vascular vertigo and dizziness can have catastrophic consequences if not recognized promptly. At present, vascular dizziness and vertigo are still very significant problems in clinical missed diagnosis, diagnosis generalization and over examination. The publication of the international "Vascular vertigo and dizziness: diagnostic criteria" provides a basis for clinicians' diagnosis and treatment behavior. However, in practical clinical work, clinicians might have to deal with more complicated situations. This paper reviews the progress of clinical diagnosis of vascular vertigo and dizziness from the aspects of clinical features, imaging and laboratory examination, so as to provide a basis for the clinical diagnosis and treatment of vascular vertigo and dizziness as well as the construction of a more accurate prediction model. Functional neurological disorder (FND) is a neuropsychiatrie disorder characterized by the presence of neurological symptoms in the absence of any neurological abnormality that can be linked to a known pathology. There is currently limited research available regarding diagnosis and treatment of FND in children and adolescents. Pediatricians and neurologists have insufficient understanding of the disease. Patients have often been misdiagnosed, correctly diagnosed after lengthy delays, or lead to inappropriate treatments. This paper reviews the pertinent clinical and historical features, diagnostic criteria, and multi- disciplinary management of FND in the pediatric population, in order to improve clinical understanding of the disease as well as its diagnosis and treatment. Paroxysmal kinesigenic dyskinesia (PKD) is a kind of nervous system disease characterized by paroxysmal involuntary movements induced by sudden movements. Neuroimaging studies have found that there are structural changes or abnormal functional connectivity in the basal ganglia, thalamus, cortex and other brain regions in PKD patients. The weakened inhibitory and regulatory function of the basal ganglia to the thalamus or the dysfunction of the thalamus itself may cause over-activation of the cortical region, resulting in involuntary movements. Recent studies have found that abnormal cerebellar function plays an important role in the pathogenesis of PKD, and abnormal cerebellar cortex can affect its normal inhibitory effect on deep cerebellar nuclei, also leading to excessive activation of thalamic - cortical pathway. This paper reviewed the mechanism and research progress of PKD in recent years, mainly focusing on the basal ganglia-thalamic-cortical circuit and the cerebellar-thalamic-cortical circuit, so as to improve clinicians' understanding of the pathogenesis of movement disorders. Epilepsy is a common neurological disease in clinic. Epilepsy caused by structural causes can be cured by surgery, which has been the focus of attention in the field of epilepsy surgery. With the development of gene sequencing technology, more and more studies have found that structural etiology may be caused by somatic mutations of genes. In this paper, somatic mutation-related pathogenic genes of four common pathological types of epilepsy surgery, including malformation of cortical development (MCD), long-term epilepsy associated tumor (LEAT), hippocampal sclerosis (HS) and hypothalamic hamartoma (HH), were reviewed, and their effects on surgical protocol formulation and prognosis were reviewed, in order to provide a new idea for the treatment of epilepsy. Neurointervention surgery is an important method for diagnosis and treatment of neurological disorders and related brain scientific research. Robot-assisted neurointervention surgery can improve the minimally invasive and accurate interventional surgery, which is an important research direction. At present, the navigation technology of neurointervention surgery has gradually developed from the traditional optical navigation to MRI navigation, the interventional materials have developed from rigid puncture needle to active flexible puncture needle, and the surgical path planning and navigation technology and the surgical robot control technology have also been developing. This paper summarizes the four key technologies mentioned above and reviews the research progress of robot-assisted neurointervention surgery to promote its clinical application. With the booming development of rehabilitation medicine engineering in recent years, the role of lower limb exoskeletal robotic systems in the treatment of walking function recovery of post- stroke has received much attention. This paper intends to classify and summarize the lower limb exoskeletal robotic systems used for walking function recovery at different periods of stroke, summarize the mechanism of improving walking function, and review the impact of lower limb exoskeletal robotic systems on the walking function of stroke patients at different periods, current challenges and future development direction, in order to promote the clinical application of lower limb exoskeletal robotic systems. With the rapid development of artificial intelligence (AI) technology, especially the application of deep learning (DL), the detection and quantitative evaluation of typical imaging markers of small cerebral vascular disease (CSVD) has been accelerated and the accuracy has been improved. In recent years, it has attracted much attention in the field of medical imaging. This paper intends to summarize the research progress and problems of deep learning in the imaging markers of CSVD such as cerebral microbleeds (CMBs), white matter hyperintensities (WMH), enlarged perivascular space (EPVS), lacunes, recent small subcortical infarcts (RSSI) and cerebral atrophy, so as to provide support for the precise treatment of CSVD. Medical artificial intelligence (AI) applies AI technology to the medical field to meet clinical needs, combines with the experience of doctors, and relies on the computing, analysis and decision-making ability of AI to provide accurate intelligent assistance for clinical diagnosis and treatment. As the number of obese and elderly population increases worldwide, sarcopenic obesity is becoming associated with a higher risk of adverse events and outcomes in multiple clinical situations, including cancer. However, there is a lack of unified definition and diagnostic criteria for sarcopenic obesity, and the interaction between sarcopenic obesity and cancer needs to be further clarified. This paper systematically and comprehensively summarizes the relevant definitions and diagnostic methods of sarcopenic obesity, discusses its clinical impact on cancer patients in detail, including the impact on patients undergoing surgery and chemotherapy, and briefly describes the main prevention and treatment strategies. This paper reviews literature and concludes that the incidence of sarcopenic obesity is high in cancer patients, but its definition and diagnostic criteria are still controversial. Sarcopenic obesity is an independent predictor of cancer prognosis with important clinical application value.

Primary liver cancer is one of the common cancers in the world and with extremely high morbidity and mortality. This paper details the current epidemiology of primary liver cancer, population attributable fractions and associated risk factors in China. In this paper, we found that hepatitis B virus and hepatitis C virus are still the main risk factors for the development of primary liver cancer by searching the databases such as PubMed, Web of Science, and CNKI. With hepatitis B virus vaccination and antiviral treatment, the incidence of primary liver cancer in China has slightly decreased, but the incidence of primary liver cancer caused by metabolic factors such as diabetes, obesity and non-alcoholic fatty liver disease is gradually increasing; smoking and alcohol consumption are also important risk factors. This article summarizes the epidemiological characteristics and risk factors of primary liver cancer, which can provide practical evidence-based medicine evidence for the development of preventive and control measures for primary liver cancer.

In recent years， with the continuous progress of DNA extraction and detection technology， cell-free DNA（cfDNA）has been widely used in the life science field， and its potential application value in forensic identification is becoming more and more obvious. This paper reviews the concept， formation mechanism， and classification of cfDNA，etc.， and describes the latest research progress of cfDNA in personal identification of crime scene touch DNA samples and non-invasive prenatal paternity testing （NIPPT）. Meanwhile， this paper summarizes the potential application of cfDNA in injury inference， and discusses the advantages and disadvantages of common cfDNA analysis methods and techniques， and its application prospects， to provide a new idea for the wide application of cfDNA in the field of forensic science.

Biological evidence is relatively common evidence in criminal cases, and it has strong probative power because it carries DNA information for individual identification. At the scene of fire-related cases, the complex thermal environment, the escape of trapped people, the firefighting and rescue operations, and the deliberate destruction of criminal suspects will all affect the biological evidence in the fire scene. Scholars at home and abroad have explored and studied the effectiveness of biological evidence identification in fire scenes, and found that the blood stains, semen stains, bones, etc. are the main biological evidence which can be easily recovered with DNA in fire scenes. In order to analyze the research status and development trend of biological evidence in fire scenes, this paper systematically sorts out the relevant research, mainly including the soot removal technology, appearance method of typical biological evidence, and possibility of identifying other biological evidence. This paper also prospects the next step of research direction, in order to provide reference for the identification of biological evidence and improve the value of biological evidence in fire scenes.

The patient-reported outcome measurement information system can help doctors make decisions, promote nursing-patient communication and improve patient compliance, which has been widely used in lung cancer clinical practice in Western countries and has become the most valuable tool for evaluating outcome indicators in lung cancer patients. This paper reviewed the current status of the application of patient-reported outcome measurement information system for lung cancer patients both domestically and internationally, aiming to provide reference for clinical nursing research and improve the quality of life of lung cancer patients. The patient-reported outcome measurement information system still has great development potential in the field of lung cancer in China, and it was expected that more practical explorations to promote its true implementation and development in China. Osteoporosis (OP) is one of the main problems affecting the health of middle-aged and elderly people, and its most serious complication is osteoporotic fracture (OPF), which also has a high disability and mortality rate. The accurate screening and diagnosis of OP mainly rely on imaging analysis, and the diagnosis result rely on the subjective judgment of doctors. Therefore, the traditional imaging method of OP have certain challenges to the accurate screening, diagnosis and differentiation of OP. The cross-fusion of machine learning (ML) and imaging images effectively improves the diagnostic efficiency of clinicians, while realizing the prediction of unknown diseases (such as OPF). Recently, significant progress has been made in the screening and diagnosis of OP and in the diagnosis and prediction of OPF. This paper will review the recent related studies on ML model-assisted screening and diagnosis of OP based on imaging data, so as to provide new ideas for accurate clinical diagnosis and treatment of OP. Gait disorder is one of common motor symptoms in patients with Parkinson's disease (PD), significantly affecting their quality of life and mobility. Robot - assisted gait training (RAGT) can improve the motor symptoms of PD patients, particularly in terms of lower limb motor function. It corrects abnormal spatiotemporal parameters of gait, such as gait velocity, step length, and step frequency, reduces the occurrence of freezing of gait, improves endurance, enhances balance ability, and ultimately improves quality of life. Postural disorder is a common disabling movement complication of Parkinson's disease (PD), in which camptocormia is the most common disabling postural disorder, which further aggravates the movement disorder of PD, and seriously reduces the activities of daily living and quality of life of patients. BACKGROUND: Currently, electrospun nanofibers, which are biomimetic materials of natural extracellular matrix and contain a three-dimensional network of interconnected pores, have been successfully used as scaffolds for various tissue regeneration, but are still faced with the challenge of extending the biomaterials into three-dimensional structures to reproduce the physiological, chemical as well as mechanical properties of the tissue microenvironment. OBJECTIVE: To summarize the process and principles of electrostatic spinning and to explore the applications of the resulting electrospun nanofibers in tissue regeneration of skin, blood vessels, nerves, bone, cartilage and tendons/ligaments. METHODS: With “electrospinning, electrospun nanofibers, electrospun nanofiber scaffolds, tissue regeneration” as the Chinese and English search terms, Google Academic Database, PubMed, and CNKI were searched, and finally 88 articles were included for review. RESULTS AND CONCLUSION: (1) The electrospun nanofibers are a natural fibrous extracellular matrix mimetic material and contain a three-dimensional network of interconnected pores that have been successfully used as scaffolds for a variety of tissue regeneration applications. (2) Several papers have described the great potential of electrospun nanofiber scaffolds applied to the regeneration of skin, blood vessels, nerves, bones, cartilage and tendons/ ligaments, providing a solid theoretical basis for its final application in clinical disease treatment, or for its transformation into practical products to enter the market. (3) However, the current research results are mostly based on cell experimental research results in vitro, and whether it can be finally applied to human body still needs clinical verification. (4) At present, many kinds of electrospun products for various clinical needs have been commercialized in and outside China, indicating that the research field of electrospun nanofiber scaffolds for soft and hard tissue regeneration has great research value and application potential. In recent years, the research of Ferroptosis and the killing of tumor cells by PD-1/PD-L1 inhibitors is a hot spot. This paper aims to review treatment delay in first-episode schizophrenia, depression, and bipolar disorder, and to compare related factors of treatment delay in the three first-episode mental disorders. It is found that increased patient responsibility, stigma, lack of disease-related knowledge, lack of access to resources, and insufficient medical support lead to delay treatment, and making patients to have longer course, heavier symptoms, and lower social functions. Petrosal vein (PV) is an important drainage vein of posterior cranial fossa, needs to be properly preserved during cerebellopontine angle (CPA) surgery. Whether it is safe to sacrifice PV is a topic of intense debate and worth discussing. On one hand, some systematic studies concluded that the complications after obliteration of PV were negligible, and on the other hand, fatal complications caused by the sacrifice of PV were reported intermittently. This paper intends to review the anatomy of PV, the complications after sacrifice of PV, the evidence and opinions on the safety of PV sacrifice, and preoperative or intraoperative methods for evaluating the safety of PV sacrifice, so as to refine the surgical management of PV by neurosurgeons.
