Studies have shown that clock gene Cry1 may have important roles in the endocrine process of seasonal reproduction in mammals. In this study, Duolang sheep (non-seasonal reproduction sheep breed) and Chinese Merino (seasonal reproduction sheep breed) were used to determine the expression change of Cry1 in hypothalamus-pituitary-ovary axis in different stage of estrous cycle by quantitative real-time PCR. The results showed that the Cry1 mRNA was expressed in all tested tissues, in which the expression levels in pineal gland and thyroid gland were higher than in other tissues. As far as different sheep breeds were concerned, the tissue expression profiles of Cry1 at different stage of estrous cycle were broadly similar. Besides hypothalamus, the expression levels of Cry1 in ovary, uterus, pineal gland, pituitary gland, and thyroid gland were all reached to peak in proestrus. The differences of expression change extent for Cry1 in vary, uterus, pineal gland, and pituitary gland in proestrus and oestrus were significant between different sheep breeds. The results suggested that Cry1 may play roles in switching on the estrus and seasonal reproduction.

Integrin linked kinase (ILK) is a scaffold protein, which plays important roles in hair follicle development. The cDNA sequence of novel ILK gene in sheep was cloned by PCR method and analyzed by bioinformatics. Tissue expression profiling in eight tissues and temporal profiling at different wool follicle anagen stages in skin was analyzed. The results showed that the whole open reading frame (ORF) of ILK gene was 1 359 bp in length, which encoded 452 amino acids. Bioinformatic analysis indicated that the secondary structure of ILK gene was mainly made up of three ankyrin repeats and a kinase domain, and there were multiple phosphorylation and Protein Kinase C sites in this gene. The RT-PCR result confirmed that ILK mRNA was expressed in heart, liver, spleen, lung, skeletal muscle, skin, and small intestine, and the expression level was much higher in skin, spleen, and liver than others. The q-PCR analysis demonstrated that the ex-pression level of ILK was significantly increased from March to May (early follicle anagen initiation) in both sheep breeds, Chinese Merino and Kazakh sheep, and there were certain differences from June to October between the two breeds. The above results indicated that ILK gene may play key roles in regulating secondary follicle growth.

Three sets of data for the P1, P2, F1, and F2 populations derived from three crosses between the normal fertility wheat (Triticum aestivum L.) cultivars with different ecotypes and the female sterile line (XND126) were used to investigate the inheritance of female fertility in wheat using mixed major gene plus polygenes inheritance model in 2005 and 2006. The results from the joint segregation analysis of the four generations showed that female fertility in wheat is controlled by two major genes plus polygenes, and the interaction between the two major genes is also detected.

Epigenetic phenomena, heritable changes in phenotype, and hence in gene regulation, but not in DNA sequence, occur in the process of polyploidization in many plants. These phenomena include gene silencing, DNA methylation, nucleolar dominance, transposable elements activation and genomic imprinting. Epigenetic phenomena might be caused by gene silencing or alternative gene expression from intergenomic interaction. These could also be induced from the changes of histone coding, without DNA methylation. Furthermore, hypomethylation, chromatin reconstitution or transposable element activation could contribute to epigenetic phenomena. These modifications may give rise to the diversification of gene expressing, to the genetic and cytological diploidization, as well as to intergenomic coordination. This paper reviews epigenetic phenomena in many plants and their influence on evolution of plant genome, thus hint the likely pathway of research in this field.

Biochip technology which had emerged from the fusion of biotechnology and micro/nanofabrication technology at the end of 1980s has been widely used in life science, medicine, clinical diagnosis, drug development, agriculture, environmental protection and strategies. DNA microarray (also call gene chip, DNA chip), one kind of biochips, is small chip containing many oligonucleotide probes. It can hybridize with labelled sample, making it possible to detect large numbers of oligonucleotides at one time. So DNA microarray can overcome the disadvantage of traditional hybridization technology such as complexity, low automation, poor efficiency and quantity of molecules detected. This paper describes a new method to detect transgenic plant with gene chip. We have developed a novel arrayed-primer extension technique. It combines hybridization and PCR in one step, while ordinary DNA microarray needs two separate steps. Therefore our method provide a feasibility to detect long DNA fragment .

As the third generation of genetic markers SNPs (single nucleotide polymorphisms) has been used extensively in gene mapping,disease-correlativity analysis ,population genetics and drug research. Here methods for detection are reviewed. Most SNP genotyping are a combination of method for interrogating SNPs and analysis technique.It described both parts and give a outlook for detection.

In this paper we introduce a new method of PCR and RAPD reaction by using a small quantity of wheat leaf tissue with alkali treatment. It can be used either to detect target gene or to analyze polymorphism of wheat varieties. This method is simple and reliable, especially for a large-scale detection.

In this paper we introduce a new storage method of PCR ingredient. PCR mixture except DNA template has been frozen to dry powder by the DNA-Plus system. This kind of powder was stored at room temperature or 4 degrees. PCR has been run in different period of storage. It was discovered that the samples of lyophilization could keep activity for a long time.
