Knowledge of leveraging biomass characteristics is essential for achieving a microbial community with a desired structure to optimize anaerobic bioreactor performance. This study investigates the successive granule transformations in a high-rate anaerobic system with intermittent gas sparging and sequential increases in organic loading rates (OLRs), by establishing the correlations between the granule microstructures and reactor operating parameters. Over the course of a 196-day lab-scale trial, the granules were visualized in various stages using scanning electron microscopy, and digital image processing was applied for further quantifying their surface properties. Correlation analyses revealed that irregularities of the granule microstructures (surface properties, specific surface area and pore volume) emerged at stage 4 when the OLR was 13.31 kg COD/m 3 ·day and in stage 5 in the absence of gas sparging. The loading ratio (substrate surface loading to upward velocity) was identified to be the main parameter controlling the granule transformations, and the surface structures were classified into three categories for further interpretation. Confocal laser scanning microscopy analyses showed that the granule core started to hollow out from stage 4. It is also found that a rough granule surface helped accelerate the growth of the granular diameter under gas sparging. Overall, this study not only establish quantitative correlations between the granules microstructures and reactor operating parameters, but also shed light on the use of intermittent gas sparging to control the surface properties of anaerobic granules in high-rate anaerobic bioreactors. The effect of chitosan on the development of granular sludge in upflow anaerobic sludge blanket reactors (UASB) when treating wastewater polluted with the organic solvents ethanol, ethyl acetate, and 1-ethoxy-2-propanol was evaluated. Three UASB reactors were operated for 219 days at ambient temperature with an organic loading rate (OLR) of between 0.3 kg COD m −3 d −1 and 20 kg COD m −3 d −1 . One reactor was operated without the addition of chitosan, while the other two were operated with the addition of chitosan doses of 2.4 mg gVSS −1 two times. The three reactors were all able to treat the OLR tested with COD removal efficiencies greater than 90%. However, the time required to reach stable operation was considerably reduced in the chitosan-assisted reactors. The development of granules in the reactors with chitosan was accelerated and granules larger than 2000 μm were only observed in these reactors. In addition, these granules exhibited better physicochemical characteristics: the mean particle diameter (540 and 613 μm) was approximately two times greater than in the control reactor (300 μm), and the settling velocities exceeded 35 m h −1 . The extracellular polymeric substances (EPS) in the reactors with the chitosan was found to be higher than in the control reactor. The protein-EPS content has been correlated with the granule size. The analyses of the microbial communities, performed through denaturing gradient gel electrophoresis and high-throughput sequencing, revealed that the syntrophic microorganisms belonging to genus Geobacter and the hydrogenotrophic methanogen Methanocorpusculum labreanum were predominant in the granules. It is commonly accepted that high salt concentrations negatively affect microbial activity in biological wastewater treatment reactors such as upflow anaerobic sludge blanket (UASB) reactors. Microbial aggregation in such reactors is equally important. It is well documented that anaerobic granules, when exposed to high salinity become weak and disintegrate, causing wash-out, operational problems and decreasing process performance. In this research, the possibility of microbial granule formation from dispersed biomass was investigated at salinity levels of 5 and 20 g Na + /L. High removal efficiencies of soluble influent organics were achieved at both salinity levels and this was accompanied by fast and robust formation of microbial granules. The process was found to be stable for the entire operational period of 217 days. As far as we know this is the first time it has been demonstrated that stable granule formation is possible at a salinity level as high as 20 g Na + /L. Methanosaeta was identified as the dominant methanogen at both salinity levels. Streptococcus spp. and bacteria belonging to the family Lachnospiraceae were identified as the dominant microbial population at 5 and 20 and g Na + /L, respectively. [ABSTRACT FROM AUTHOR]

For the anaerobic biological treatment of saline wastewater, Anaerobic Digestion (AD) is currently a possibility, even though elevated salt concentrations can be a major obstacle. Anaerobic consortia and especially methanogenic archaea are very sensitive to fluctuations in salinity. When working with Upflow Sludge Blanket Reactor (UASB) technology, in which the microorganisms are aggregated and retained in the system as a granular biofilm, high sodium concentration negatively affects aggregation and consequently process performances. In this research, we analysed the structure of the biofilm and granules formed during the anaerobic treatment of high salinity (at 10 and 20 g/L of sodium) synthetic wastewater at lab scale. The acclimated inoculum was able to accomplish high rates of organics removal at all the salinity levels tested. 16S rRNA gene clonal analysis and Fluorescence In Situ Hybridization (FISH) analyses identified the acetoclastic Methanosaeta harundinacea as the key player involved acetate degradation and microbial attachment/granulation. When additional calcium (1 g/L) was added to overcome the negative effect of sodium on microbial aggregation, during the biofilm formation process microbial attachment and acetate degradation decreased. The same result was observed on granules formation: while calcium had a positive effect on granules strength when added to UASB reactors, Methanosaeta filaments were not present and the degradation of the partially acidified substrate was negatively influenced. This research demonstrated the possibility to get granulation at high salinity, bringing to the forefront the importance of a selection towards Methanosaeta cells growing in filamentous form to obtain strong and healthy granules. Methanogenic granular sludge from anaerobic bioreactors plays a primary role in treatment of various high-strength industrial wastewaters. The common problem of sludge floatation can lead to washout of granules from the reactor and severely affect reactor performance. However, an understanding of the specific key trigger-factors and appropriate control strategies for granular sludge floatation remains elusive. In this study, the concentration of acetate, rather than that of other volatile fatty acids (VFAs, i.e. propionate and butyrate) and granular sludge properties, was identified to be positively, linearly correlated with the amount of floating granules. The number of floating granules on propionate (18 ± 6) or butyrate-containing (34 ± 13) wastewater was comparable with that of non-VFA control wastewater (30.5 ± 7.5), and much lower than that of acetate-containing wastewater (80.5 ± 10.5). A scenario of excessive acetate-triggered granular sludge floatation is proposed based on these results as well as on the microbial community profile and spatial distribution, porous structure of granules, and impacts of operational parameters. Two new strategies, acetate-depletion and co-substrate addition, effectively reduced the number of floating granules by 28.5% and 51.6%, respectively. The effect of temperature on selenium (Se) removal by upflow anaerobic sludge blanket (UASB) reactors treating selenate and nitrate containing wastewater was investigated by comparing the performance of a thermophilic (55 °C) versus a mesophilic (30 °C) UASB reactor. When only selenate (50 μM) was fed to the UASB reactors (pH 7.3; hydraulic retention time 8 h) with excess electron donor (lactate at 1.38 mM corresponding to an organic loading rate of 0.5 g COD L −1 d −1 ), the thermophilic UASB reactor achieved a higher total Se removal efficiency (94.4 ± 2.4%) than the mesophilic UASB reactor (82.0 ± 3.8%). When 5000 μM nitrate was further added to the influent, total Se removal was again better under thermophilic (70.1 ± 6.6%) when compared to mesophilic (43.6 ± 8.8%) conditions. The higher total effluent Se concentration in the mesophilic UASB reactor was due to the higher concentrations of biogenic elemental Se nanoparticles (BioSeNPs). The shape of the BioSeNPs observed in both UASB reactors was different: nanospheres and nanorods, respectively, in the mesophilic and thermophilic UASB reactors. Abstract: CuO nanoparticles (NPs) are released into wastewater due to the widespread use and generation as by-product in various applications (e.g. semiconductor manufacturing). However, information on the behavior and impact of CuO NPs on wastewater treatment processes is very limited. The objective of this study was to evaluate the fate and long-term effect of CuO NPs (average size = 37 nm) on high-rate anaerobic bioreactors. A laboratory-scale upflow anaerobic sludge blanket reactor was operated with a synthetic wastewater containing low concentrations of CuO NPs (1.4 mg Cu L−1) and a mixture of volatile fatty acids for 107 days. CuO NPs were largely removed during anaerobic treatment and on the average only 20–32% of the NPs fed to the reactor escaped with the effluent. Scanning electron microscopy and chemical analysis confirmed that CuO NPs were partitioned into the anaerobic sludge. While short-term exposure to CuO NPs (1.4 mg Cu L−1) only caused minor inhibition to methanogenesis, extended exposure caused severe toxicity and reduced the acetoclastic methanogenic activity by more than 85%. Moreover, the reactor performance was completely disrupted and the methane production decreased by more than 50%. Abstract: In contrast to the general aerobic detoxification of industrial effluents containing cyanide, anaerobic cyanide degradation is not well understood, including the microbial communities involved. To address this knowledge gap, this study measured anaerobic cyanide degradation and the rearrangements in bacterial and archaeal microbial communities in an upflow anaerobic sludge blanket (UASB) reactor biomass treating brewery waste water using bio-methane potential assays, molecular profiling, sequencing and microarray approaches. Successful biogas formation and cyanide removal without inhibition were observed at cyanide concentrations up to 5 mg l−1. At 8.5 mg l−1 cyanide, there was a 22 day lag phase in microbial activity, but subsequent methane production rates were equivalent to when 5 mg l−1 was used. The higher cumulative methane production in cyanide-amended samples indicated that part of the biogas was derived from cyanide degradation. Anaerobic degradation of cyanide using autoclaved UASB biomass proceeded at a rate more than two times lower than when UASB biomass was not autoclaved, indicating that anaerobic cyanide degradation was in fact a combination of simultaneous abiotic and biotic processes. Phylogenetic analyses of bacterial and archaeal 16S rRNA genes for the first time identified and linked the bacterial phylum Firmicutes and the archaeal genus Methanosarcina sp. as important microbial groups involved in cyanide degradation. Methanogenic activity of unadapted granulated biomass was detected at higher cyanide concentrations than reported previously for the unadapted suspended biomass, making the aggregated structure and predominantly hydrogenotrophic nature of methanogenic community important features in cyanide degradation. Abstract: Treatment and disposal of high volume of salty waste production in recirculating aquaculture systems (RASs) is a major challenge and the sludge is often a source of environmental pollution and salinization of receiving soils and water bodies. Anaerobic digestion is an efficient mean for the treatment of wastes of different origins and might serve a useful tool for the reduction of salty aquaculture discharge load. Use of an upflow anaerobic sludge blanket (UASB) reactor for digestion of brackish aquaculture sludge from RASs under different C:N ratios, temperatures, and hydraulic retention times demonstrated high removal efficiencies of over 92% as volatile solids (VS), 98% as chemical oxygen demand and 81% as total suspended solids in all reactors. Methane production topped 7.1 mL/gVS d and was limited by low C:N ratio but was not influenced by temperature fluctuations. The treated liquid effluent from all reactors was of sufficient quality for reuse in the RAS, leading to significant water recycling and saving rates. Abstract: Nowadays, carbon emission and therefore carbon footprint of water utilities is an important issue. In this respect, we should consider the opportunities to reduce carbon footprint for small and large wastewater treatment plants. The use of anaerobic rather than aerobic treatment processes would achieve this aim because no aeration is required and the generation of methane can be used within the plant. High-rate anaerobic digesters receive great interests due to their high loading capacity and low sludge production. Among them, the upflow anaerobic sludge blanket (UASB) reactors have been most widely used. However, there are still unresolved issues inhibiting the widespread of this technology in developing countries or countries with climate temperature fluctuations (such as subtropical regions). A large number of studies have been carried out in order to enhance the performance of UASB reactors but there is a lack of updated documentation. In face of the existing limitations and the increasing importance of this technology, the authors present an up-to-date review on the performance enhancements of UASB reactors over the last decade. The important aspects of this article are: (i) enhancing the start-up and granulation in UASB reactors, (ii) coupling with post-treatment unit to overcome the temperature constraint, and (iii) improving the removal efficiencies of the organic matter, nutrients and pathogens in the final effluent. Abstract: An aerobic granular sludge (AGS) reactor was run for 280 days to study the competition between Phosphate and Glycogen Accumulating Organisms (PAOs and GAOs) at high temperatures. Numerous researches have proven that in suspended sludge systems PAOs are outcompeted by GAOs at higher temperatures. In the following study a reactor was operated at 30 °C in which the P-removal efficiency declined from 79% to 32% after 69 days of operation when biomass removal for sludge retention time (SRT) control was established by effluent withdrawal. In a second attempt at 24 °C, efficiency of P-removal remained on average at 71 ± 5% for 76 days. Samples taken from different depths of the sludge bed analysed using Fluorescent in situ hybridization (FISH) microscopy techniques revealed a distinctive microbial community structure: bottom granules contained considerably more Accumulibacter (PAOs) compared to top granules that were dominated by Competibacter (GAOs). In a third phase the SRT was controlled by discharging biomass exclusively from the top of the sludge bed. The application of this method increased the P-removal efficiency up to 100% for 88 days at 30 °C. Granules selected near the bottom of the sludge bed increased in volume, density and overall ash content; resulting in significantly higher settling velocities. With the removal of exclusively bottom biomass in phase four, P-removal efficiency decreased to 36% within 3 weeks. This study shows that biomass segregation in aerobic granular sludge systems offers an extra possibility to influence microbial competition in order to obtain a desired population. Abstract: The objective of the current study was to expand the knowledge of the role of acetogenic Bacteria in high rate anaerobic digesters. To this end, acetogens were enriched by supplying a variety of acetogenic growth supportive substrates to two laboratory scale high rate upflow anaerobic sludge bed (UASB) reactors operated at 37 °C (R1) and 55 °C (R2). The reactors were initially fed a glucose/acetate influent. Having achieved high operational performance and granular sludge development and activity, both reactors were changed to homoacetogenic bacterial substrates on day 373 of the trial. The reactors were initially fed with sodium vanillate as a sole substrate. Although % COD removal indicated that the 55 °C reactor out performed the 37 °C reactor, effluent acetate levels from R2 were generally higher than from R1, reaching values as high as 5023 mg l−1. Homoacetogenic activity in both reactors was confirmed on day 419 by specific acetogenic activity (SAA) measurement, with higher values obtained for R2 than R1. Sodium formate was introduced as sole substrate to both reactors on day 464. It was found that formate supported acetogenic activity at both temperatures. By the end of the trial, no specific methanogenic activity (SMA) was observed against acetate and propionate indicating that the methane produced was solely by hydrogenotrophic Archaea. Higher SMA and SAA values against H2/CO2 suggested development of a formate utilising acetogenic population growing in syntrophy with hydrogenotrophic methanogens. Abstract: The effect of reactor process configuration on anaerobic production of useful energy (hydrogen and methane) from a complex substrate was investigated for the following reactor systems: suspended growth, two-phase mixed, two-stage mixed, upflow anaerobic sludge blanket (UASB) reactor, and two-phase UASB. The mixed two-phase and two-stage configurations yielded the highest specific energy productions of 13.3 and 13.4 kJ/g COD fed, respectively. Reactor process configuration influenced microbial pathways in acidogenic reactors in that butyrate was the predominant volatile acid in phased configurations, whereas acetate was predominant in the staged configuration. The UASB reactor achieved the highest average daily energy production per reactor volume of 101 kJ/L reactor-d. All reactor configurations achieved high COD removals on the order of 99%. However, hydrogen represented only 3% of the total energy produced by the two-phase mixed and two-phase UASB configurations. Theoretical analysis revealed that the maximum specific energy production by the two-phase suspended-growth configuration is only 9% higher than that for a single-stage mixed reactor. Consequently, the production of hydrogen from complex substrates in these process configurations does not seem to be justifiable solely from an energy point of view. Instead, it is suggested that phased anaerobic systems should be considered primarily for improved process stability whereas resultant hydrogen production is of secondary benefit. Abstract: Uranium has been responsible for extensive contamination of groundwater due to releases from mill tailings and other uranium processing waste. Past evidence has confirmed that certain bacteria can enzymatically reduce soluble hexavalent uranium (U(VI)) to insoluble tetravalent uranium (U(IV)) under anaerobic conditions in the presence of appropriate electron donors. This paper focuses on the evaluation of anaerobic granular sludge as a source of inoculum for the bioremediation of uranium in water. Batch experiments were performed with several methanogenic anaerobic granular sludge samples and different electron donors. Abiotic controls consisting of heat-killed inoculum and non-inoculated treatments confirmed the biological removal process. In this study, unadapted anaerobic granular sludge immediately reduced U(VI), suggesting an intrinsic capacity of the sludge to support this process. The high biodiversity of anaerobic granular sludge most likely accounts for the presence of specific microorganisms capable of reducing U(VI). Oxidation by O2 was shown to resolubilize the uranium. This observation combined with X-ray diffraction evidence of uraninite confirmed that the removal during anaerobic treatment was due to reductive precipitation. The anaerobic removal activity could be sustained after several respikes of U(VI). The U(VI) removal was feasible without addition of electron donors, indicating that the decay of endogenous biomass substrates was contributing electron equivalents to the process. Addition of electron donors, such as H2 stimulated the removal of U(VI) to varying degrees. The stimulation was greater in sludge samples with lower endogenous substrate levels. Abstract: A multi-compartment anaerobic bioreactor, designated the anaerobic migrating blanket reactor (AMBR), did not perform well in terms of chemical oxygen demand (COD) removal after an increase in sulfate load, compared to a conventional upflow anaerobic sludge blanket (UASB) reactor. The trophic structures of the bioreactors were characterized by analyzing the electron flows, formation and consumption of fermentation intermediates and terminal product (methane and hydrogen sulfide) formation. Critical performance parameters were linked to operational perturbations such as increase in sulfate load and changes in flow reversal schemes in the AMBR. Both of these manipulations affected the microbial communities, which were monitored by terminal restriction fragment length polymorphism (T-RFLP) analysis targeting the bacterial and archaeal domains. The less stable AMBR did not produce granular biomass, and in response to increased sulfate concentrations, experienced a reversal in the distribution of hydrogenotrophic methanogens that correlated with a shift in electron flow from butyrate to propionate. As this shift occurred, bacterial populations such as butyrate-producing clostridia, became predominant, thus leading to reactor imbalance. The stable UASB reactor developed and retained granules and maintained a relatively stable archaeal community. Sulfate perturbation led to the selection of a novel bacterial group (Thermotogaceae), which was most likely well adapted to the increasingly sulfidogenic conditions in the bioreactor. Abstract: This paper evaluates the use of upflow anaerobic sludge bed (UASB) bioreactors (30°C, pH=7.0) to remove selenium oxyanions from contaminated waters (790μgSeL−1) under methanogenic and sulfate-reducing conditions using lactate as electron donor. One UASB reactor received sulfate at different sulfate to selenate ratios, while another UASB was operated under methanogenic conditions for 132 days without sulfate in the influent. The selenate effluent concentrations in the sulfate-reducing and methanogenic reactor were 24 and 8μgSeL−1, corresponding to removal efficiencies of 97% and 99%, respectively. X-ray diffraction (XRD) analysis and sequential extractions showed that selenium was mainly retained as elemental selenium in the biomass. However, the total dissolved selenium effluent concentrations amounted to 73 and 80μgSeL−1, respectively, suggesting that selenate was partly converted to another selenium compound, most likely colloidally dispersed Se0 nanoparticles. Possible intermediates of selenium reduction (selenite, dimethylselenide, dimethyldiselenide, H2Se) could not be detected. Sulfate reducers removed selenate at molar excess of sulfate to selenate (up to a factor of 2600) and elevated dissolved sulfide concentrations (up to 168mgL−1), but selenium removal efficiencies were limited by the applied sulfate-loading rate. In the methanogenic bioreactor, selenate and dissolved selenium removal were independent of the sulfate load, but inhibited by sulfide (101mgL−1). The selenium removal efficiency of the methanogenic UASB abruptly improved after 58 days of operation, suggesting that a specialized selenium-converting population developed in the reactor. This paper demonstrates that both sulfate-reducing and methanogenic UASB reactors can be applied to remove selenate from contaminated natural waters and anthropogenic waste streams, e.g. agricultural drainage waters, acid mine drainage and flue gas desulfurization bleeds. Over 99% of dimethyl phthalate (DMP) and 93% of chemical oxygen demand (COD) were effectively removed in a continuous upflow anaerobic sludge blanket (UASB) reactor from a wastewater containing 600mg/L DMP at 8h of hydraulic retention time (HRT), corresponding to a loading rate of 3g-COD/(Ld). Each gram of sludge, expressed as volatile suspended solids (VSS), had a maximum specific methanogenic activity (SMA) of 24mg-CH4/(g-VSSd) using DMP as the sole carbon source. The sludge yield was estimated as 0.08g-VSS/g-COD. During anaerobic degradation, DMP was de-esterified, first to mono-methyl phthalate (MMP) and then to phthalate, before being de-aromatized and subsequently converted to CH4 and CO2. The maximum specific degradation rates of DMP, MMP and phthalate were 415, 88 and 36mg/(g-VSSd), respectively. Analysis based on polymerase chain reaction (PCR) and denaturing gradient gel electrophoresis (DGGE) showed a gradual shift of microbial population with the increase of DMP loading. This study compares the performance of a pilot-scale combination of UASB and DHS system to that of activated sludge process (ASP) for the treatment of municipal sewage. Both systems were operated in parallel with the same sewage as influent. The study was conducted for more than 300 days, which revealed that organic removal efficiency of system was comparable to that of ASP. Unfiltered BOD removal by both systems was more than 90%. However, system outperformed ASP for pathogen removal. In addition, volume of excess sludge production from was 15 times smaller than that from ASP. Moreover, unlike ASP, there is no requirement of aeration for the operation of system, which makes it an economical treatment system. Considering the above observations, it was concluded that system can be a cost-effective and viable option for the treatment of municipal sewage over ASP, especially for low-income countries. This study investigated the effect of the COD/SO4 2− ratio (4 and 1) and the sulfide concentration on the performance of thermophilic (55°C) acidifying (pH 6) upflow anaerobic sludge bed reactors fed with sucrose at an organic loading rate of 4.5gCODlreactor −1 day−1. Sulfate reduction efficiencies amounted to 65% and 25–35% for the COD/SO4 2− ratios of 4 and 1, respectively. Acidification was complete at all the tested conditions and the electron flow was similar at the two COD/SO4 2− ratios applied. The stepwise decrease of the sulfide concentrations in the reactors with a COD/SO4 2− ratio of 1 by N2 stripping caused an immediate stepwise increase in the sulfate reduction efficiencies, indicating a reversible inhibition by sulfide. The degree of reversibility was, however, affected by the growth conditions of the sludge. Acidifying sludge pre-grown at pH 6, at a COD/SO4 2− ratio of 9 and exposed for 150 days to 115mgl−1 sulfide, showed a slower recovery from the sulfide inhibition than a freshly harvested sludge from a full scale treatment plant (pH 7 and COD/SO4 2−=9.5) exposed for a 70 days to 200mgl−1 sulfide. In the latter case, the decrease of the sulfide concentration from 200 to 45mgl−1 (35mgl−1 undissociated sulfide) by N2 stripping caused an immediate increase of the sulfate reduction efficiency from 35% to 96%. Abstract: A consecutive-reaction kinetic model for the sucrose-fed upflow anaerobic sludge bed (UASB) reactor that accounts for a layered structure of the granule and the mass fraction of methanogens (f) is proposed. When the UASB reactor was maintained at the volumetric loading rates (VLR) of 7.9–13.8kg chemical oxygen demand (COD)/m3 d, the accumulated volatile fatty acids (VFAs) increased with increasing VLR, whereas the experimental f decreased with increasing VLR. This was primarily because methanogenesis was the rate-limiting step and the sucrose-fed granule was a layered structure. The calculated residual concentrations of sucrose and the intermediates VFAs using the layered-structure model are less deviated from the experimental measurements than those using the homogeneous-structure model. The calculated effectiveness factors for sucrose uptake and intermediates VFAs uptake (η 1; η 2) ranged from 0.18 to 0.35 and 0.65 to 0.96, respectively, indicating that the overall substrate (sucrose or intermediates VFAs) removal in the UASB reactor was diffusion-controlled, especially at the VLRs of 7.9–10.6.kgCOD/m3 d. This finding was also confirmed by the simulated concentration profiles of sucrose and VFAs in the UASB-granule. From the simulation results, the effect of internal mass transfer resistance on overall substrate (sucrose) removal should not be neglected, especially for a granule size of greater than 2.0mm. Abstract: The mechanism of faecal coliform removal in down-flow hanging sponge (DHS) system treating up-flow anaerobic sludge blanket (UASB) reactor effluent was the subject of this study. The results obtained revealed that the most important removal mechanism of faecal coliform in the DHS system is adsorption, followed by predation. Die-off is a relatively minor removal mechanism in the DHS system. The impact of physical and chemical factors such as contact time, effective sponge bulk volume and pH values on faecal coliform removal has been investigated. Increasing the contact time and sponge bulk volume exerted positive effect on the removal of faecal coliform. Changing the pH values between 6.5 and 9.0 did not show significant impact. Reducing sponge pore size from 1.92 to 0.56mm, increased faecal coliform removal by a value of 1.3log10. [Copyright &y& Elsevier]

Abstract: The aim of this study was to investigate variations in the occurrence and removal of enterovirus and norovirus genomes, Giardia cysts, Cryptosporidium oocysts and the most commonly used faecal indicators in a Swedish wastewater pilot plant. Paired samples were taken from the inlet and outlet of each treatment line: tertiary filtration, membrane bioreactor (MBR) and upflow anaerobic sludge blankets (UASB). (Oo)cysts and indicators were enumerated using standard methods and viruses using RT-PCR. Giardia cysts and enteroviruses were constantly detected, mean numbers 103.11 cysts and 104.0 PCR unitsL−1, respectively. Oocysts were found in 5/19 samples, mean number5 L−1. Noroviruses were found in 6/7 influent samples, with an average titre of 103.28 L−1, during winter, but only in 2/15 in the rest of the year (mean 200L−1). MBR treatment removed indicators more efficiently than did the other two lines, with 5log removal of E. coli. Human virus genome removal did not differ between the MBR and tertiary treatment line. Microorganism removal in UASB was significantly lower for all the organisms studied. E. coli, enterococci and Cl. perfringens removal was correlated () with enterovirus genome removal, with R-values around 0.4. However, values for removal of indicators were more strongly correlated to each other. Removal of viruses based on enumeration using RT-PCR probably underestimates infectious virion removal. Abstract: The effect of nitrophenolic shock loads on the performance of three bench-scale upflow anaerobic sludge blanket (UASB) reactors was studied using synthetic wastewater. Reactors R1, R2 and R3 were fed with 30mg/L concentration of 2-nitrophenol (2-NP), 4-nitrophenol (4-NP) and 2,4-dinitrophenol (2,4-DNP), respectively, along with methanol (COD=2000mg/L), sodium nitrate (NO3 −-N=200mg/L), and other nutrients. The reactors were in continuous operation for more than 2 years before the shock loading study was performed. Five nitrophenolic shock loadings of 45, 60, 75, 90 and 120mg/Ld were administrated by increasing the influent nitrophenolic concentration to 45, 60, 75, 90 and 120mg/L, respectively, while keeping hydraulic retention time as 24h. The shocks were given continuously for a period of 4 days before switching back to normal nitrophenolic loading (30mg/Ld). The reactors were allowed to recover to normal performance level before administrating the next nitrophenolic shock load. The study showed that the nitrophenolic shock load of as high as 120mg/Ld did not affect the reactors performance irreversibly. After resuming the normal nitrophenolic loading, it took almost 3–18 days for the reactors to recover from the shock effect. The study was further extended to assess the maximum possible mixed nitrophenolic loading (2NP:4NP:2,4:DNP=1:1:1) to which 2,4-DNP acclimated granular sludge containing reactor (R3) can be exposed without hampering the reactor (R3) performance irreversibly. The reactor was able to achieve pseudo-steady-state at a mixed nitrophenolic loading of 180mg/Ld with more than 90% removal of all the three nitrophenols, but failed at a mixed nitrophenolic loading of 225mg/Ld. Abstract: Mixing and transport phenomena affect the efficiency of all bioreactor configurations. An even mixing pattern at the macro-level is desirable to provide good conditions for substrate transport to, and from, the microbial aggregates. The state of segregation of particulate material in the reactor is also important. The production of biogas in anaerobic reactors is another factor that affects mixing intensity and hence the interactions between the liquid, solid and gaseous phases. The CSTR model with some degree of short-circuiting, dead zones and bypassing flows seems to describe the overall hydrodynamics of UASBs. However, few data are available in the literature for full-scale reactors that relate process performance to mixing characteristics. Dispersion studies using LiCl were done for four hydraulic loading rates on a full-scale UASB treating domestic wastewater in Ginebra, Valle del Cauca, southwest Colombia. COD, TSS, and Settleable Solids were used to evaluate the performance of organic matter removal. The UASB showed a complete mixing pattern for hydraulic loading rates close to the design value (i.e. Q=10–13ls−1 and HRT=8–6h). Gross mixing distortions and localised stagnant zones, short-circuiting and bypass flows were found in the sludge bed and blanket zones for both extreme conditions (underloading and overloading). The liquid volume contained below the gas–liquid–solid separator was found to contribute to the overall stagnant volume, particularly when the reactor was underloaded. Abstract: This work accesses the influence of cycle duration on the intermittent operation of mesophilic UASB reactors inoculated with flocculent sludge and used in dairy wastewater (DWW) treatment. Five cycle lengths ranging from 24 to 144h were compared for loads between 2.5 and 29.0gCOD/l/d. COD balances are presented which demonstrate the importance of a feedless period in the conversion to methane of the substrates removed during the feed period. The maximum applicable loads determined for the system were higher for the longer cycle times. The higher conversion to methane of the removed COD was obtained for the 96h cycle (48h feed+48h feedless) resulting in a more stable operation. The 96h cycle (48h feed+48h feedless) was considered as the optimum for the treatment of dairy effluents in intermittent UASB reactors. Compared to the maximum applicable loads reported in the literature for continuous systems (3–6gCOD/l/d) treating dairy effluents the stable operation loads attained with intermittent operation were considerably higher (22gCOD/l/d). Abstract: Sludge characteristics available inside the reactor are of vital importance to maximize advantages of UASB reactor. The organic loading rate and sludge loading rate applied during start-up are among the important parameters to govern the sludge characteristics. Effects of these loading rates on the characteristics of the sludge developed are evaluated in six laboratory scale UASB reactors. The sludge characteristics considered are VSS/SS ratio of the sludge, sludge volume index, specific gravity, settling velocity and metal contents of the sludge developed under different loading rates. The experimental results indicate that, for developing good characteristics sludge, during primary start-up from flocculent inoculum sludge, organic loading rate and sludge loading rate should be in the range of 2.0–4.5kg COD/m3 d and 0.1–0.25kg COD/kg VSSd, respectively (chemical oxygen demand, COD). Abstract: Upflow anaerobic sludge blanket (UASB) process has been successfully applied in the treatment of municipal and industrial wastewaters. Several researchers have investigated the suitability of the process for the treatment of phenols and phenolic wastewaters. The anaerobic treatment of phenols is still at an investigative stage. With increasing recognition of the UASB process, feasibility studies on the treatment of wastewater containing phenol and cresols (o-, m- and p- isomers) in UASB have been reviewed. It is reported that phenol concentration up to a range of 500–750mg/L is generally not inhibitory to the UASB process. Phenol concentrations greater than 500mg/L can be effectively treated with acclimatization of inocula, recirculation of the treated effluent and/or supplementing with co-substrates such as glucose, VFA and dilute molasses. The degradability of phenol is more than p-cresol, which in turn is more than m- and o-cresol. [Copyright &y& Elsevier]

An integrated chemical–physical–biological treatment concept for the low-cost treatment of domestic wastewater is proposed. Domestic wastewater was subjected to a chemically enhanced primary treatment (CEPT), followed by treatment in an upflow anaerobic sludge blanket (UASB) reactor. In addition, a regenerable zeolite was used to remove NH4+, either after CEPT pretreatment or after biological treatment in the UASB reactor. The CEPT pretreatment consisted of the addition of a coagulant (FeCl3) and an anionic organic flocculant and removed on average 73% of the total chemical oxygen demand (CODt), 85% of the total suspended solids, and 80% of PO43- present in the wastewater. The UASB system, which consequently received a low CODt input of approximately 140 mg/L, was operated using a volumetric loading rate of 0.4 g CODt/L. d (hydraulic retention time [HRT]=10 h) and 0.7 g CODt/L. d (HRT=5 h). For these conditions, the system removed about 55% of the CODt in its influent, thus producing an effluent with a low CODt of approximately 50 mg/L. The zeolite, when applied in batch mode before the UASB reactor, removed approximately 45% of the NH4+, whereas its application as a post-treatment cartridge resulted in almost 100% NH4+ removal. The simple design and relatively low operating costs, due to low costs of added chemicals and low energy input (estimated at € 0.07–0.1 per m3 wastewater treated), combined with excellent treatment performance, means that this system can be used as a novel domestic wastewater treatment system for developing countries. A central composite design was employed to methodically investigate anaerobic treatment of aircraft deicing fluid (ADF) in bench-scale Upflow Anaerobic Sludge Blanket (UASB) reactors. A total of 23 runs at 17 different operating conditions (0.8% 1.6% ADF (6000–12,000 mg/L COD), 12–56 h HRT, and 18–36 g VSS/L) were conducted in continuous mode. The development of four empirical models describing process responses (i.e. COD removal efficiency, biomass-specific acetoclastic activity, methane production rate, and methane production potential) as functions of ADF concentration, hydraulic retention time, and biomass concentration is presented. Model verification indicated that predicted responses (COD removal efficiencies, biomass-specific acetoclastic activity, and methane production rates and potential) were in good agreement with experimental results. Biomass-specific acetoclastic activity was improved two-fold from 0.23 g COD/g VSS/d for inoculum to a maximum of 0.55 g COD/g VSS/d during ADF treatment in UASB reactors. For the design window, COD removal efficiencies were higher than 90%. The predicted methane production potentials were close to theoretical values, and methane production rates increased as the organic loading rate is increased. ADF toxicity effects were evident for 1.6% ADF at medium organic loadings (SOLR above 0.5 g COD/g VSS/d). In contrast, good reactor stability and excellent COD removal efficiencies were achieved at 1.2% ADF for reactor loadings approaching that of highly loaded systems (0.73 g COD/g VSS/d). In the present study, the effects of a cationic polymer on reactor start-up and granule development were evaluated. A control reactor R1 was operated without adding polymer, while the other five reactors designated R2, R3, R4, R5 and R6 were operated with different polymer concentrations of 20, 40, 80, 160 and 320 mg/L, respectively. Experimental results demonstrated that adding the polymer at a concentration of 80 mg/L markedly accelerated the start-up time. The time required to reach stable treatment at an organic loading rate (OLR) of 4 g COD/L.d was reduced by approximately 50% in R4 as compared with the control reactor. The same reactor with 80 mg/L polymer was able to achieve an OLR of 12 g COD/L.d after 59 days of operation, while R1, R2, R3, R5 and R6 achieved the same loading rate at much longer period of 104, 80, 69, 63 and 69 days, respectively. Comparing with the control reactor, the start-up time of R4 was shortened markedly by about 43% at this OLR, while other reactors also recorded varying degree of shortening. Monitoring on granule development showed that the granule formation was accelerated by 30% from the use of the appropriate dosage of polymer. Subsequent granules characterization indicated that the granules developed in R4 with 80 mg/L polymer exhibited the best settleability, strength and methanogenic activity at all OLRs. The organic loading capacities of reactors were also increased by the polymer addition. The maximum organic loading of the control reactor was 24 g COD/L.d, while the polymer-assisted reactor added with 80 mg/L polymer attained a markedly increased organic loading of 40 g COD/L.d. The laboratory results obtained demonstrated that adding the cationic polymer could result in shortening of start-up time and enhancement of granulation, which in turn lead to improvement in organics removal efficiency and loading capacity of the UASB system. This paper reviews different theories on anaerobic sludge granulation in UASB-reactors that have been proposed during the past two decades. The initial stages of the formation of anaerobic granules follow the same principles as biofilm formation of bacteria on solid surfaces. There exist strong evidence that inert carriers play an important positive role in granulation. Most researchers conclude that Methanosaeta concilii is a key organism in granulation. Only the Cape Town Hypothesis presumes that an autotrophic hydrogenotrophic organism, i.e., Methanobacterium strain AZ, growing under conditions of high H2-pressures, is the key organism in granulation. Many authors focus on the initial stage of granulation, and only a few contributions discuss the latter stages in granulation: granule maturation and multiplication. Granule enhancing factors in the latter stages predominantly rely on manipulation of the selection pressure, through which selectively heavier sludge particles are retained in the UASB reactor. This paper reports on the performance of three units of a sub-surface horizontal flow constructed wetland (CW) pilot plant in polishing effluent from the upflow anaerobic sludge blanket (UASB) reactor plant. Studies on the use of UASB for on-site wastewater pre-treatment were initiated in Tanzania for the first time in 1993, while initial research works on the application of CW for polishing effluent from UASB, the results of which are reported in this paper began in 1999. In this study the UASB reactor plant received and pre-treated part of the wastewater from the student''s hostels at the University College of Lands and Architectural Studies in Tanzania. Out of the three units, unit B was planted with Phragmites mauritianus, unit C with Typha latifolia and A was used as a control. Both P.mauritianus and T.latifolia have been extensively researched in Europe and USA as suitable species of vegetation in CW. However, very limited studies on the suitability of these wetland plants have been reported under tropical climate. The studied parameters were chemical oxygen demand (COD), nitrate (NO3-N), nitrite (NO2-N), ammonium (NH4-N), faecal coliforms (FC), total coliforms (TC), pH, temperature and dissolved oxygen (DO). The study was carried out at an average hydraulic retention time of 1.93 days (1.85 in unit A, 1.96 in unit B and 1.99 in unit C) obtained as a ratio of the volume of wastewater in the wetland and the volumetric flow rate of wastewater through the wetland unit while taking into consideration the porosity of the media. Better performance for the vegetated units B and C were obtained compared to the control unit A. Nutrients were least removed in all units (NH4-N 11.2%, 25.2% and 23% in units A, B and C, respectively, NO3-N 32.2%, 40.3% and 44.3% for units A, B and C, respectively, and NO2-N 23.9%, 38.5% and 23.1% for units A, B and C, respectively). The COD removal rate was 33.6%, 56.3% and 60.7% for units A, B and C, respectively. The study also indicated that pH increased from the influent to the effluent and that DO increase was related to the decrease of temperature. The effect of COD/NO3−-N ratio on the biotransformation and removal of 2-nitrophenol (2-NP), 4-nitrophenol (4-NP), and 2,4-dinitrophenol (2,4-DNP) was studied in bench scale upflow anaerobic sludge blanket (UASB) reactors. Sodium acetate and sodium nitrate were used as electron donor (substrate) and electron acceptor, respectively. Nitrate nitrogen loading was increased from 0.098 to 0.6 kg/m3 d in order to keep COD/NO3−-N ratio as 20.8, 14.3, 9.8, 5.0, 4.0 and 3.33. Throughout the study, input nitrophenolic concentration and hydraulic retention time (HRT) were kept constant as 30 mg/l and 24 h, respectively. 2-Aminophenol (2-AP), 4-aminophenol (4-AP) and 2-amino,4-nitrophenol (2-A,4-NP) were found as the major intermediate metabolite of 2-NP, 4-NP and 2,4-DNP, respectively. Removal of all the three nitrophenols increased with lowering of COD/NO3−-N ratio. However, nitrophenols removal got adversely affected when COD/NO3−-N ratio was reduced below 5. Maximum removal achieved were 91.63%, 90.17% and 86.10% for 2-NP, 4-NP and 2,4-DNP, respectively at a COD/NO3−-N ratio of 5. Upflow anaerobic sludge blanket (UASB) reactor has been employed in industrial and municipal wastewater treatment for decades. However, the long start-up period required for the development of anaerobic granules seriously limits the application of this technology. In order to develop the strategy for rapid UASB start-up, the mechanisms for anaerobic granulation should be understood. This paper attempts to provide a up-to-date review on the existing mechanisms and models for anaerobic granulation in the UASB reactor, which include inert nuclei model, selection pressure model, multi-valence positive ion-bonding model, synthetic and natural polymer-bonding model, Capetown''s model, spaghetti theory, syntrophic microcolony model, multi-layer model, secondary minimum adhesion model, local dehydration and hydrophobic interaction model, surface tension model, proton translocation-dehydration theory, cellular automaton model and cell-to-cell communication model. Based on those previous works, a general model for anaerobic granulation is also proposed. It is expected that this paper would be helpful for researchers to further develop a unified theory for anaerobic granulation and technology for expediting the formation of the UASB granules. This study investigated the applicability of dynamic membrane technology in anaerobic membrane bioreactors for the treatment of high strength wastewaters. A monofilament woven fabric was used as support material for dynamic membrane formation. An anaerobic dynamic membrane bioreactor (AnDMBR) was operated under a variety of operational conditions, including different sludge retention times (SRTs) of 20 and 40 days in order to determine the effect of SRT on both biological performance and dynamic membrane filtration characteristics. High COD removal efficiencies exceeding 99% were achieved during the operation at both SRTs. Higher filtration resistances were measured during the operation at SRT of 40 days in comparison to SRT of 20 days, applying a stable flux of 2.6 L/m(2) h. The higher filtration resistances coincided with lower extracellular polymeric substances concentration in the bulk sludge at SRT of 40 days, likely resulting in a decreased particle flocculation. Results showed that dynamic membrane technology achieved a stable and high quality permeate and AnDMBRs can be used as a reliable and satisfactory technology for treatment of high strength wastewaters., (Copyright © 2013 Elsevier Ltd.
