The viscosity of the mucus, its DNA concentration and the size range of the DNA were determined on tracheobronchial samples from 11 horses with lower airway diseases before and after incubation with recombinant human deoxyribonuclease (rhDNase). The horses were divided into two groups on the basis of the cytology of the samples: group A (five horses) with more than 60 per cent neutrophils and group B (six horses) with fewer than 50 per cent neutrophils. The mean mucus viscosity and DNA concentration in the preincubation samples were significantly higher in group A than in group B, and there was a correlation between DNA concentration and mucus viscosity in the preincubation samples from group A. Incubation with rhDNase significantly reduced the viscosity of the samples only in group A.

The pathological changes observed in the lungs of 197 freshly dead to moderately decomposed harbour porpoises (Phocoenaphocoena) stranded in England and Wales between October 1990 and December 1996 were reviewed. In 135 (69 per cent of the cases) macroscopic nematode infections of the bronchial tract with Pseudalius inflexus and Torynurus convolutus, either singly or in combination, were recorded, and 106 (54 per cent) also had P inflexus within the pulmonary blood vessels. All the macroscopically parasitised porpoises were adults or juveniles although two neonates had histological evidence of nematode infection. There were 62 cases of mild to severe, subacute to chronic bronchitis and bronchiolitis, 113 cases of mild to severe chronic granulomatous interstitial pneumonia, and 34 cases of mild to severe vasculitis or thrombovasculitis of pulmonary blood vessels attributable to these nematode infections. In 35 cases necropurulent or purulent (broncho)pneumonias were attributed either to secondary bacterial infections of the lungs or to septicaemias associated in seven cases with Streptococcus canis, in two cases with group B Salmonella species, in one case with Escherichia coli and in one case with Streptococcus lactis. The pulmonary lesions in 67 animals known or diagnosed to have been entrapped in fishing gear were non-specific and included persistent foam in the airways in 45 cases, diffuse congestion in 53, oedema in 50, and multifocal intra-alveolar haemorrhage in 33 cases. Seven cases of acute fibrinous or chronic fibrous pleuritis, seven cases of chronic necropurulent pneumonia associated with mycotic infections, four porpoises with traumatic lesions of the thorax and other parts of the body consistent with fatal attack from bottlenose dolphins (Tursiops truncatus), and one case of diffuse bronchointerstitial pneumonia associated with generalised morbillivirus infection were also recorded.

The effect of three intraruminal sustained-release devices (SRD) against Dictyocaulus viviparus infection was tested in five groups of six calves. Group 1 served as untreated controls, and groups 2, 3 and 4 were dosed with a levamisole SRD, a fenbendazole SRD, and an ivermectin SRD, respectively. Group 5 was vaccinated against lungworm and received a levamisole SRD. The calves were turned out on May 28 and the devices given seven days later. All the calves received trickle infections with a total of 200 lungworm larvae between 9 and 34 days after turnout. They were housed on October 28, challenged with 5000 lungworm larvae and slaughtered three weeks later. No clinical signs of parasitic bronchitis were observed during the study. The treated groups gained significantly more weight (P < 0.05) than the controls, but did not differ among themselves. Larvae were first detected in the faeces of the control group between 25 and 32 days after the first infection, and had a group mean of 21 larvae per gram (lpg) after 60 to 80 days, after which the lpg gradually decreased. In group 2, larvae were detected near the end of the grazing season and never exceeded a group mean of 1.5 lpg. In group 3, a very low larval output was observed after housing (group mean 0.1 lpg). Groups 4 and 5 never became patent. The results of an ELISA followed the pattern of larval output; optical densities above the cut-off value were recorded in groups 1, 2 and 3. On the basis of worm recoveries after challenge, group 1 was immune. Group 4 had significantly more lungworms than group 2. There were no significant differences in worm numbers between groups 2, 3 and 5, but the worms in group 5 were retarded in growth (P < 0.05).

Forty-five calves were assigned randomly to three groups of 15 at turnout (day 0) in the spring of their first grazing season on to separate paddocks on a pasture known to carry infective larvae of Dictyocaulus viviparus. Group 1 calves were left as untreated controls, group 2 were injected with doramectin at 200 micrograms/kg at turnout and again on day 56, and group 3 were treated with an intraruminal bolus containing ivermectin. From day 42 onwards group 1 calves developed parasitic bronchitis which required repeated treatment with levamisole. The two endectocide regimes controlled lungworm infection, although some calves in group 2 developed some coughing during the week before the second dose of doramectin. After the end of the grazing season and again in May of the following year, five cattle from each group were infected experimentally with lungworm larvae and slaughtered 28 days later for lungworm counts and measurements of length to be made. At both times group 1 calves were found to be largely resistant to reinfection; group 2 were slightly more susceptible although the differences from group 1 were not statistically significant. Group 3 calves were more susceptible with no significant difference in worm counts from naive infection controls.

Each of 10 set-stocked calves was given a fenbendazole sustained-release intraruminal bolus at turnout for the control of parasitic bronchitis while a group of 10 similar calves was left untreated. The respiratory rates of the control calves were not greatly increased during the grazing season, but persistent coughing was evident from early July when they all had patent lungworm infections. Only occasional coughing was reported from the bolus-treated calves except for a transient increase in its frequency in late September. In mid-August, one of the treated calves was passing lungworm larvae and when they were housed six of the 10 had patent infections. In August tracer calves picked up an average of 23.5 lungworms per day from the control paddock but only 2.1 from the paddock grazed by the treated calves. In September the corresponding figures were 7.6 and 19.2 lungworms per day, respectively, from the control and 'treated' paddocks. After housing, the post mortem worm counts after an experimental challenge with Dictyocaulus viviparus larvae were reduced by 99.2 and 98.7 per cent (P < 0.0001), respectively, for the control and bolus-treated calves in comparison with weight-matched parasite-naive calves. Thus, despite a relatively low level of challenge during the grazing season, the treated animals had developed a considerable degree of protective immunity.

Two groups of 16 set-stocked calves were used to evaluate a new strategy for the prevention of parasitic bronchitis and parasitic gastroenteritis. One group was left untreated while the calves in the other were treated with abamectin at 0.2 mg/kg at turnout and again six weeks later. The treatment prevented the output of nematode eggs and lungworm larvae in faeces for at least 70 days. The number of infective larvae subsequently appearing on the pasture was reduced by 90.2 per cent and the infectivity of the pasture (as monitored by tracer calves) by 96.0 to 99.8 per cent in the case of Dictyocaulus viviparus, 88.2 to 99.2 per cent for Ostertagia ostertagi and 69.3 to 98.1 per cent for Cooperia oncophora. Parasitic bronchitis occurred in the control calves and both bronchitis and gastroenteritis in the tracer calves grazing the paddock grazed by the control calves, but no disease occurred either in any of the calves treated with abamectin or in the tracer calves grazing the paddock grazed by these calves.

Chronic tracheobronchial syndrome was diagnosed in eight dogs which had had a chronic mild cough for two to 13 months but were otherwise clinically normal. In five of them there was evidence of contact with other coughing dogs before the appearance of clinical signs and the majority at first responded favourably to antibacterial therapy. The coughing occurred mainly when the dogs pulled on their leads or became excited. Three cases had bronchoscopic evidence of active airway inflammation, and five had varying numbers of inflammatory cells (neutrophils) in bronchial aspirates. There was no evidence of excessive production of mucus in the airways of any of the dogs. Seven had an increased bronchial pattern on radiographs but only one of the dogs has developed chronic bronchitis.

The enzyme-linked immunosorbent assay (ELISA) for the serodiagnosis of parasitic bronchitis in cattle was evaluated in naturally injected, experimentally infected and vaccinated animals. Significant antibody titres could be demonstrated from five weeks after animals had been exposed to the parasite, so that infected animals could be identified during the summer. The test did not give false positive results in vaccinated animals. The technique proved particularly useful in revealing latent infections in milking herds in the autumn when heavy burdens of worms in the lungs do not generate any larvae in the faeces.

Two outbreaks of parasitic gastroenteritis were observed in a group of 10 first-season grazing calves, one in mid-July and one in mid-September. In both cases emergency anthelmintic treatment was needed to prevent further damage. Severe clinical signs were observed together with high faecal egg counts and high serum pepsinogen and gastrin concentrations. Low total protein and albumin concentrations were also observed, especially during the second outbreak. The ostertagia antibody levels followed a similar pattern to the serum pepsinogen and gastrin concentrations. At the end of the housing period a mild type II ostertagiasis was observed. In the second grazing season the heifers did not show any signs of parasitic gastroenteritis, but there was a serious outbreak of husk which required treatment.

A 16-day infection of Dictyocaulus viviparus in two groups of calves was treated with levamisole and fenbendazole respectively. Five days afterwards the calves were reinfected with 4000 larvae and necropsied 21 days later. Although the lungworm burdens of the two groups of calves were reduced by about 70 per cent compared to a control group the clinical signs of dyspnoea, tachypnoea and coughing were indistinguishable from a primary infection. This was due to pulmonary emphysema, oedema and an acute epithelialising pneumonia apparently associated with the death and disintegration of lungworms in situ, the result of an incompletely developed immune response. The results are compared with those obtained with the lungworm vaccine. It was concluded that the outcome of any system of "control" which depends on drug therapy and reinfection is unpredictable and that vaccination offers the only effective method of prophylaxis.

Fifteen red deer calves were put to pasture in two groups, at the same set stocking rates, one group of 10 and one of five, in separate but adjoining enclosures likely to be carrying infective stage larvae of dictyocaulus naturally parasitising the red deer of Scotland. The group of 10 had been vaccinated with a live, bovine lungworm oral vaccine; the group of five had not. Results did not indicate any advantage to the vaccinated deer in weight gain or general health but they excreted fewer dictyocaulus larvae. The findings, although based on only a few animals, support the conclusion that vaccination of red deer would not give them increased protection against the establishment of naturally occurring dictyocaulus infection although it would enhance the suppression of larval production.

Studies on the epidemiology of Dictyocaulus viviparus infections in Denmark have suggested that the adult lungworms present in calves around the sixth week after turnout play an important role in determining the subsequent pattern of disease. This trial was designed to test whether prophylactic treatment at this time would control disease in calves kept under British conditions. Thirty autumn-born Friesian or Friesian-cross bull calves were allowed to graze the whole of a 5 hectare field for six weeks after turnout. The field was then divided into two and the calves split into matching groups, one group being put into each of the paddocks. One group was treated with levamisole at this time and again two weeks later while the other was kept as an untreated control. Anthelmintic treatment resulted in a marked reduction in larval excretion and considerably delayed the build-up of infection on pasture. This in turn delayed the onset and reduced the severity of clinical signs in the treated group. However, as disease was not eliminated completely this prophylactic programme cannot be recommended to the British farmer in its present form. These findings are discussed in the context of the yet incomplete knowledge of the epidemiology of parasitic bronchitis.

Seventy-one worm-free Friesian calves were allocated by weight to three trial groups (1, 3 and 4) of 18 and a control group (2) of 17 animals. Calves in group 1 were vaccinated with a bovine lungworm oral vaccine on days 0 and 28, and on day 42 all groups were turned out to graze together on pasture known to be infected with Dictyocaulus viviparus larvae. Twenty-eight days after first exposure to infection one control calf died of parasitic bronchitis. Anthelmintic medication consisting of two doses of levamisole (7 . 5 mg/kg) at 14 day intervals was promptly administered to group 3 calves and three doses at the same intervals to group 4 calves. All calves were challenged with 20,000 infective D viviparus larvae on day 147. Calves were weighed every 14 days throughout the trial which ended 42 days after challenge. Pasture contamination and infectivity were monitored by pasture larval counts and tracer calves. Statistically there was no significant difference between the performances of treated and vaccinated groups before challenge but all were significantly superior to the control group. After challenge the productivity of all experimental groups was temporarily depressed but the levamisole treated cattle recovered more rapidly becoming significantly heavier than the vaccinates at the end of the trial. The mean group weight gains over the trial period were 89 . 92, 63 . 87, 88 . 67 and 98 . 70 kg in groups 1, 2, 3 and 4 respectively.

A group of 12 winter-born calves was divided into two groups of six. During the following summer one group grazed on pasture infected with Dictyocaulus viviparus, and was treated with ivermectin injections at three, eight and 13 weeks after turn out. The other group remained housed. Both groups were housed during the winter and then together with a group of younger calves were challenged with a trickle infection of D viviparus larvae at the rate of 25 third stage larvae/kg bodyweight for one month and then slaughtered. The group which had been exposed to previous infection was least affected by parasitic bronchitis and on the basis of serological titres and worm burdens had developed resistance to the challenge infection. The other older group was also more resistant than the younger calves.

A comparison was made of the material costs and effectiveness of three methods of early season suppression by anthelmintic medication of Ostertagia species and two of Dictyocaulus viviparus in calves, each method suppressing faecal egg output for different lengths of time from the start of spring grazing. The anthelmintics used were: Morantel bolus administered five days before going to grazing; oxfendazole given three times at three, six and nine weeks after the start of grazing and ivermectin injected three, eight and 13 weeks after going to pasture. The effectiveness of each was estimated by comparison with worm numbers in untreated control calves. Oxfendazole, which was active for the shortest time (about 65 days) from the start of grazing (May 1), produced a 78.1 per cent reduction in Ostertagia species and an 84.4 per cent reduction in D viviparus. The morantel bolus was estimated to be active for 90 days and resulted in a 94.3 per cent reduction of Ostertagia species. The ivermectin treatment, which, because of the prolonged excretion of the chemical and different sensitivity of worm species, was estimated to suppress Ostertagia species for 105 days and D viviparus for 119 days, caused reductions of 98.7 per cent of the former and 97.4 per cent of the latter species. Material costs per calf were estimated at pounds 1.25 for oxfendazole, pounds 2.00 for ivermectin and pounds 10 for the morantel bolus.

Lungworm-infected seeder calves were used on two 1.41 ha paddocks to ensure that groups of 11 susceptible trial calves would be exposed to heavy early season challenge with Dictyocaulus viviparus. This produced conditions for an artificially severe test of two control strategies. The first employed a front-loaded oxfendazole pulse release bolus, ie, an intraruminal device which released one therapeutic anthelmintic dose immediately and five subsequent pulses at approximately three-weekly intervals. These front-loaded boluses were given to five of 11 calves on one paddock as soon as parasitic bronchitis had become clinically obvious (34 days after turnout) while the remaining six calves were kept as untreated controls. Clinical signs quickly subsided in the treated animals and no further respiratory problems occurred despite continued exposure to reinfection. The other control strategy involved the administration at turnout of an oxfendazole pulse release device which released the first of five anthelmintic doses approximately three weeks after administration, to all 11 calves on the other paddock. This strategy was almost completely successful in preventing patent infections from establishing and reduced the infectivity of the pasture in August and September by 94.1 per cent as shown by tracer calf studies. The calves treated at turnout performed better than the calves treated with the front-loaded boluses for most of the season and had an average weight-gain advantage of 20.4 kg at housing (P less than 0.01).(ABSTRACT TRUNCATED AT 250 WORDS)

Sixty cattle (12 first season and 48 second season grazing animals) were allocated to three groups according to age and bodyweight. Each group was divided into "control" and "treated" subgroups. Before turnout, a morantel sustained release bolus (MSRB) was administered to each animal in the "treated" category. The groups were moved from field to field according to the farmer's normal rotational grazing policy. Each field was, however, divided into two equal halves, one of which was reserved for the MSRB treated cattle, while the other was used exclusively for the controls. Severe parasitic gastroenteritis occurred in the first season controls during early September, while milder disease affected the untreated animals in the smaller of the second season groups. No gastrointestinal disease was apparent in the corresponding MSRB treated cattle. A mild outbreak of parasitic bronchitis occurred in the first year controls during October; there was evidence of less sever lungworm infection in the matching MSRB treated animals. The larger second season group showed no signs of parasitic disease.

The study of cattle respiratory diseases during the last decade show that several observations made in the late 1960s are still relevant today. Many of these diseases are shared with other countries in Europe, but the developments discussed will be examined with the respiratory disease problems that occur in Britain in mind. The new information is dealth with as it applies to (i) structure, function and immunology of the lungs (ii) diseases of calves indoors (iii) diseases of young animals grazing, (iv) diseases of adult animals indoors and (v) diseases of adult animals grazing.

Broken wind is a syndrome characterised by chronic bronchitis and alveolar emphysema. Clinical signs include nasal catarrh, persistent coughing, dyspnoea and poor exercise tolerance. In racehorses, lung haemorrhages may result in epistaxis. Broken wind is a disease of domestication ascribed to pollution of the stable air with fungal spores from hay and straw. Treatment and prevention are based on the provision of fresh air and, if housing is unavoidable, the adoption of a permanent regime of dust-free stable management. If an early diagnosis is made and appropriate treatment instituted, the prognosis is considered to be reasonably good. The disease exemplifies the validity of the Royal Veterinary College motto Venienti occurrite morbo (treat the disease at its first appearance).

A virus isolated from a respiratory disease in chickens was identified as infectious bronchitis virus (IBV). By serum neutralisation tests the virus was shown to be serologically distinct from the Massachusetts type but to possess a one-way relationship with the Connecticut type. Experimentally, it caused a relatively mild respiratory disease in baby chicks. Vaccination of young chicks with H-120 Massachusetts IBV vaccine afforded a high degree of protection against challenge with the isolate. Some problems relating to the isolation and classification of field strains of IBV are briefly discussed.

The conventional lungworm vaccination schedule, requiring a minimum age of eight weeks, a four-week interval between doses and a further interval of not less than two weeks before turnout, was compared with a modified schedule in which the vaccine was given to calves at six and eight weeks old followed by a turnout immediately after the second dose. The control group consisted of unvaccinated calves, half of which corresponded in age to the conventional and half to the modified vaccination group. All groups were turned out to the same pasture area on May 13 with five calves which were artificially infected with Dictyocaulus viviparus so as to ensure adequate pasture challenge. Challenge caused only mild clinical signs in the vaccinated calves but caused severe husk with two fatalities in the controls. Respiratory rates and faeces larval counts for the vaccinated groups were significantly lower than for the control group and, in the main, not significantly affected by vaccination schedule. The vaccinated groups gained more weight than their respective controls of corresponding age and significantly so for both groups during the period when dictyocaulus infection exerted maximum effect. In this experiment, the modified schedule was as effective as the conventional although delayed challenge precluded a satisfactory test for the effect of turnout immediately after the second dose of vaccine. In a repeat experiment involving apparently a very severe pasture challenge, calves vaccinated at six and eight weeks old showed markedly less resistance than those vaccinated according to the conventional schedule. Probable reasons for the difference in outcome are considered.

Two groups of parasite-free calves, one of which had been treated with four doses of a homoeopathic oral vaccine for parasitic bronchitis due to Dictyocaulus viviparus and the other with a placebo, were infected at the rate of 25 infective larvae/kg bodyweight 18 days after the final dose. Both groups became severely affected by parasitic bronchitis, with clinical signs starting 13 days after infection. There were no discernible differences between the treated and control groups in their manifestations of resistance to D viviparus or their clinical responses to the disease produced.

The efficacy of the oxfendazole pulse release bolus system for the control of parasitic gastroenteritis and parasitic bronchitis in first-season grazing calves was evaluated in Belgium. Twenty-two calves were allocated to two groups. The calves in one group received a bolus at the time of turn out, while the other group remained untreated. The efficacy of the bolus was assessed by comparison of faecal worm egg counts, plasma pepsinogen concentrations, the antibody response to Ostertagia, Cooperia and Dictyocaulus species total plasma protein and albumin concentrations, and weight gains throughout the grazing season and the housing period. The oxfendazole pulse release bolus provided good control of parasitic gastroenteritis dominated by ostertagia. The effects of parasitic gastritis were greatly reduced as shown by the significantly lower values of serum pepsinogen and ostertagia antibody titres. The use of the bolus further reduced the adverse effects of parasitism as indicated by better liveweight gains and normal total plasma protein and albumin concentrations whereas in the untreated control group hypoproteinaemia and hypoalbuminaemia were observed. Most animals exhibited clinical signs of parasitic bronchitis at the end of the grazing season, and the bolus may not adequately control parasitic bronchitis in all cases at all times.

Four groups, each of six male Friesian calves, were set-stocked on separate 0.66 ha paddocks from May 7 until October 23 1986. Each of the animals in groups 1 and 4 was dosed with an oxfendazole pulse release bolus at turn out whereas the animals in groups 2 and 3 were left untreated. Parasite-free naive tracer calves were introduced into each paddock for a limited period 12 days after turn out and again at the end of the trial. No adverse reactions or clinical signs were observed in either of the groups of calves which received boluses. The development of clinical parasitic gastroenteritis in both the untreated groups necessitated the humane slaughter of two animals and emergency anthelmintic treatment of the remainder. The lower plasma pepsinogen concentrations, and lower faecal egg and larval counts and worm burdens post mortem, together with the absence of clinical signs of parasitic gastroenteritis and bronchitis in the treated calves, confirmed the high efficacy of the bolus treatment.

The term "fog fever" was originally used identify a disease of adult cattle grazing lush pastures in the autumn. Unfortunately, the name has subsequently been applied to other respiratory disorders which occurred under different epidemiological circumstances, so that the name "fog fever" has lost much of its original specificity and become almost synonymous with "acute respiratory distress". The pulmonary lesions in 151 cattle, of all ages, with acute respiratory distress are described in this report. While most of the animals were referred as examples of "fog fever", in only 43 of the 151 cases were the clinical signs, epidemiology and post mortem findings consistent with that disease. Twelve other pulmonary disturbances were encountered in the other animals and the pathology of these conditions has bee described.

In the West of Scotland the epidemiology of parasitic bronchitis in grazing calves was studied over a two year period with the aid of tracer calves and herbage examinations for Dictyocaulus viviparus larvae. The observations of both years emphasised the importance of overwintered lungworm larvae as a source of disease. In the first year it was shown that the ingestion and development of these overwintered larvae were, by themselves, directly responsible for severe morbidity, high faecal larval counts and deaths. In the second year it was shown that pasture ungrazed during the winter and spring and from which a hay crop was removed in mid-summer was still capable of producing clinical parasitic bronchitis in susceptible calves within three to four weeks of their introduction in later summer. In both years there was some evidence that the outbreaks appeared to be associated with the sudden availability of infective larvae on the herbage. The possibility that such larvae may have survived for many months in the soil is discussed. Despite the heavy challenge with lungworm larvae experienced by the grazing calves in the first year those vaccinated with lungworm vaccine survived, their clinical signs were mild and of short duration and their faecal larval output was greatly reduced.

Of 36 dogs with a history of chronic coughing or dysponea examined by bronchoscopy, eight were found to have parasitic tracheobronchitis. Oral treatment with the anthelmintic levamisole at a rate of 7.5 mg per kg bodyweight was found to be successful in every case.

Lungworm-infected seeder calves were used on four 1.41 ha paddocks to ensure that groups of 11 calves would be exposed to a heavy challenge with Dictyocaulus viviparus. By the 39th day after turnout there was a serious episode of respiratory disease and a diagnosis of parasitic bronchitis was confirmed by post mortem and faecal examination. One group of trial calves was treated with netobimin administered in the drinking water at 2.8 mg/kg/day for seven consecutive days; another group received the same treatment supplemented with flunixin meglumine at 2.2 mg/kg/day for three days; a third group was given a single oral dose of 7.5 mg netobimin/kg; only emergency treatments were given to calves in the control group. The clinical response to the drinking water treatments was highly satisfactory and better than the response to the single oral treatment.

In May 1985 four groups of 10 calves, aged between four and five months, were turned out on to separate, permanent pastures of equal area which had been seeded during the previous few days with larvae of Dictyocaulus viviparus. One group acted as a control, the second was vaccinated with lungworm vaccine before turnout and treated with thiabendazole three, eight and 13 weeks after turnout, while the third and fourth groups were given ivermectin three times (three, eight and 13 weeks after turnout) and twice (three and eight weeks after turnout), respectively. A severe outbreak of parasitic bronchitis resulted in the death of three control calves within five weeks of turnout and parasitic bronchitis and gastroenteritis affected the second group of calves after approximately four months at pasture. The calves given ivermectin excreted no lungworm larvae and remained free of clinical parasitism throughout the trial.
