Disease-transmitting vectors are living organisms that pass infectious agents from one animal/human to another. The epidemiologically important vectors are usually hematophagous arthropods, including mosquitoes, ticks, triatome bugs, sand flies, and tsetse flies. All of them harbor an endogenous microbiota that functionally complements their host's biology. Different arthropod vectors are ecologically and behaviorally distinct, and as such, their relationships with symbiotic microbes vary. In this review, we summarize the recent discoveries that reveal how bacterial metabolic activities influence development, nutrition, and pathogen defense in mosquitoes, ticks, triatome bugs, and sand flies. These studies provide a foundation for a systematic understanding of vector-microbiota interactions and for the development of integrated approaches to control vector-borne diseases.

Invasive tick species and the pathogens they transmit pose increasing threats to human and animal health around the world. Little attention has been paid to the characteristics enabling tick species to invade. Here we analyze examples of tick invasion events in North America to identify factors that facilitated the invasion. Commonalities among invasive ticks are that they thrive in anthropogenically modified habitats, feed on either domestic animals or wildlife occurring in high density, and can survive across a broad range of climatic conditions. Invasive tick species varied widely in life history and reproductive habits, suggesting that invasion occurs when multiple characteristics converge. The combination of potential characteristics leading to invasion, however, improves our ability to predict future invaders and inform surveillance.

Trypanosomes are global blood parasites that infect a wide range of vertebrate hosts. Several species of Trypanosoma cause disease in humans and domesticated animals, and the majority are transmitted between hosts by haematophagous invertebrate vectors. Ticks have long been speculated as vectors for Australian trypanosomes. Recent studies using advanced molecular techniques have refocused attention on these arthropods, and whilst they have renewed discussions about Trypanosoma species and their vectors, these reports have simultaneously led to premature conclusions concerning the role of ticks as vectors. Here the controversy surrounding ticks as trypanosome vectors is discussed. We highlight the unanswered questions concerning the role played by ticks in trypanosome transmission and suggest future approaches to resolving these key knowledge gaps.

From seminal publications in the early 1970s, the world learned that dendritic cells (DCs) are powerful and versatile antigen-presenting cells. It took a few years until the first studies expanded our understanding of the pivotal role of these immune 'soldiers' against ticks. Advances in biochemistry, molecular biology, and bioinformatics have shed light on the identification of key salivary molecules that modulate the biology of DCs in favor of tick parasitism. Here, we present a critical overview of the discoveries accumulated on the tick-host battlefield from a DC perspective. Moreover, the clinical significance of DC-targeted tick salivary components is discussed, not only as facilitators of the transmission of tick-borne pathogens or vaccine candidates, but also as potential immunobiologics to treat immune-mediated diseases.

Recent molecular and cellular studies have highlighted a potentially important role for tick exosomes in parasite transmission. Here we summarize evolving hypotheses about the largely unknown cellular events that may take place at the tick–host–pathogen interface, focusing on a potential role for arthropod exosomes in this tripartite interaction.

Tick-borne diseases are common occurrences in both the medical and veterinary clinical settings. In addition to the constraints related to their diagnosis and clinical management, the control and prevention of these diseases is often difficult, because it requires the disruption of a complex transmission chain, involving vertebrate hosts and ticks, which interact in a constantly changing environment. We provide a contemporary review of representative tick-borne diseases of humans and discuss aspects linked to their medical relevance worldwide. Finally, we emphasize the importance of a One Health approach to tick-borne diseases, calling physicians and veterinarians to unify their efforts in the management of these diseases, several of which are zoonoses.

There has been considerable debate as to whether global risk from vector-borne diseases will be impacted by climate change. This has focussed on important mosquito-borne diseases that are transmitted by the vectors from infected to uninfected humans. However, this debate has mostly ignored the biological diversity of vectors and vector-borne diseases. Here, we review how climate and climate change may impact those most divergent of arthropod disease vector groups: multivoltine insects and hard-bodied (ixodid) ticks. We contrast features of the life cycles and behaviour of these arthropods, and how weather, climate, and climate change may have very different impacts on the spatiotemporal occurrence and abundance of vectors, and the pathogens they transmit.

Tick saliva facilitates tick feeding and infection of the host. Gene expression analysis of tick salivary glands and other tissues involved in host-pathogen interactions has revealed a wide range of bioactive tick proteins. Transcriptomic analysis has been a milestone in the field and has recently been enhanced by next-generation sequencing (NGS). Furthermore, the application of quantitative proteomics to ticks with unknown genomes has provided deeper insights into the molecular mechanisms underlying tick hematophagy, pathogen transmission, and tick-host-pathogen interactions. We review current knowledge on the transcriptomics and proteomics of tick tissues from a systems-biology perspective and discuss future challenges in the field.

Tick- and tsetse-borne diseases cost Africa approximately US$4-5 billion per year in livestock production-associated losses. The use of pyrethroid-treated cattle to control ticks and tsetse promises to be an increasingly important tool to counter this loss. However, uncontrolled use of this technology might lead to environmental damage, acaricide resistance in tick populations and a possible exacerbation of tick-borne diseases. Recent research to identify, quantify and to develop strategies to avoid these effects are highlighted.

Dogs and people are exposed to and susceptible to infection by many of the same tick-borne bacterial pathogens in the order Rickettsiales, including Anaplasma phagocytophilum, Ehrlichia canis, E. chaffeensis, E. ewingii, Rickettsia rickettsii, R. conorii, and other spotted fever group rickettsiae. Recent findings include descriptions of novel Ehrlichia and Rickettsia species, recognition of the occurrence and clinical significance of co-infection, and increasing awareness of Rhipicephalus sanguineus-associated diseases. Newer molecular assays are available, although renewed efforts to encourage their use are needed. This review highlights the ecology and epidemiology of these diseases, and proposes avenues for future investigation.

Ticks and the diseases for which they are vectors engage in complex interactions with their mammalian hosts. These interactions involve the developmental processes of tick and pathogen, and interplay between the defensive responses and counter responses of host, tick and pathogen. Understanding these interactions has long been an intractable problem, but progress is now being made thanks to the flood of genomic information on host, tick and pathogen, and the attendant, novel experimental tools that have been generated. Over 40 cell lines are currently available from 13 ixodid and one argasid tick species. The successful isolation and propagation of several economically important tick-borne pathogens in tick cell lines has created a useful model to study interactions between tick cells and these viral and bacterial disease agents. Tick cell lines have already proved to be a useful tool in helping to define the complex nature of the host–vector–pathogen relationship. With the availability of genomics tools, tick cell lines will become increasingly important as a complement to tick and tick-borne disease research in vivo once genetic transformation and gene silencing using RNA interference become routine.

Ticks are ectoparasites of wild and domestic animals, and humans. A more comprehensive understanding of tick function and the tick–pathogen interface is needed to formulate improved tick-control methods. RNA interference (RNAi) is the most widely used gene-silencing technique in ticks where the use of other methods of genetic manipulations has been limited. In the short time that RNAi has been available, it has proved to be a valuable tool for studying tick gene function, the characterization of the tick–pathogen interface, and the screening and characterization of tick protective antigens. This review considers the applications of RNAi to tick research and the potential of this technique for tick functional studies, and to elucidate the tick–pathogen and tick–host interface. The link between parasites and eosinophilia has been known for more than a century, although the role of eosinophils in host protection is still an open issue. Much less appreciated, however, is the concurrent systemic induction of a related cell type, the basophil, in parasitized hosts. To date, little is known about the role of basophils in immunity against parasites, but recent evidence points to a possible crucial role in the initiation of T-helper type 2 responses in the host. In this article, we review the current understanding of parasitic infections and basophils and discuss their putative role in immunity.
