The measurement of fecal progesterone metabolites (fPM) by enzyme immunoassay analysis is a non-invasive technique that permits gathering reproductive information from wildlife without the stress associated with restraint. In the collared peccary (Pecari tajacu), a high correlation between serum progesterone and fPM levels (r 2  = 0.783) suggests that fPM can be used to monitor their reproductive function. We monitored fPM during the estrous cycle of 15 collared peccary females. Estrous cycles averaged 27.9 ± 4.5 days (n = 28), ranging from 21 to 36 days. The luteal phase was 22.2 ± 4.8 days and the inter-luteal phase was 4.3 ± 1.4 days. Mean concentration of fPM across pregnancy were not different from those found during the luteal phase (1230 ± 718 and 1265 ± 584 ng/100 mg dried feces, respectively), however, significant differences were found when luteal phase concentrations were compared only against fPM concentrations during late pregnancy. In addition, late pregnancy fPM concentrations (1893 ± 551 ng/100 mg) were also significantly higher than those in the early (639 ± 339 ng/100 mg) and mid (1134 ± 449 ng/100 mg) pregnancy. For females during the early post-partum period, fPM concentrations were significantly increased (243 ± 118 ng/100 mg) than those of non-cycling females (103 ± 89 ng/100 mg). The analysis of fPM is a simple, non-invasive methodology to detect the ovarian activity in the collared peccary; moreover, it provides a husbandry tool, which may be used to help understand how social structure may impact reproduction., (Copyright © 2019. Cryopreservation may lead bovine oocytes to undergo morphological changes and functional damage due to the high-lipid content in the cytoplasm and the formation of reactive oxygen species. Against this background, the present study aimed to improve the cryotolerance and developmental competence of prepubertal calf oocytes by adding L-carnitine (LC) and/or resveratrol (R) to the IVM medium, as the former is involved in lipid metabolism and both are able to scavenge reactive oxygen species. With this purpose, different quality and functional oocyte parameters, such as spindle and chromosome configuration, DNA integrity, caspase activity, and the profile of genes involved in lipid metabolism and oxidative stress were evaluated in IVM bovine oocytes before or after vitrification/warming. Oocytes were matured in the absence (control) or presence of LC (3.03 mM) and/or R (1 μM) and then vitrified/warmed before IVF and embryo culture. All treatment groups (control, LC, R, and LC + R) of nonvitrified IVM oocytes showed similar rates (P > 0.05) of a normal spindle and chromosome configuration to oocytes vitrified/warmed after maturation in the presence of LC + R. When oocytes in all treatment groups were compared before and after vitrification, no significant differences were detected in DNA fragmentation as measured using the TUNEL method. However, the proportion of early apoptotic oocytes increased after vitrification/warming, except when previously matured with R. Vitrified/warmed oocytes matured in the presence of LC did not differ with nonvitrified oocytes in terms of the expression of ACACA, SLC2A1, PLIN2, HSPA1A, GPX1, and SOD1 genes. Similarly, expression of ACACA, SLC2A1, PLIN2, HSPA1A, and SOD1 genes in vitrified/warmed oocytes was similar to that of their fresh counterparts when matured in the presence of R. Finally, while the addition of LC and/or R to IVM medium had no effect on both cleavage and blastocyst rates either in fresh or vitrified oocytes. To conclude, the results of the present study report that the addition of LC and/or R to the IVM medium used for prepubertal bovine oocytes did not increase the embryo development potential of both fresh and vitrified oocytes. However, LC + R supplementation before vitrification decreased spindle damage, R addition-modulated apoptosis, and LC or R addition before vitrification positively affected the gene expression of vitrified/warmed oocytes., (Copyright © 2016 Elsevier Inc. Heat stress (HS) in mammals is a determining factor in the deterioration of spermatogenesis and can cause infertility. The aim of this study was to evaluate the effect of continuous summer circadian cycles on semen production, sperm cell features, fertility, prolificacy, and fecal cortisol metabolites from rabbits kept under an in vivo HS model. We split randomly 60 New Zealand White rabbits into two temperature-controlled rooms: The control group was maintained at comfort temperature (18 °C-22 °C) and an HS group, where the environmental temperature was programmed to increase from 22 °C to 31 °C and be maintained for 3 hours to this temperature at the central part of the day. Fecal cortisol metabolites were assessed to evaluate the stress conditions. Seminal parameters were analyzed. Although animals exposed to HS showed higher values of fecal cortisol metabolites (P = 0.0003), no differences were detected in fertility or prolificacy. Semen samples from HS males showed a significant decrease (P < 0.05) with respect to the controls in the percentage of viable spermatozoa (80.71% vs. 74.21%), and a significant (P ≤ 0.01) increase in the percentage of acrosomic abnormalities (22.57% vs. 36.96%) and tailless spermatozoa (7.91% vs. 12.83). Among motility parameters, no differences were found. This study describes a model of HS simulating a continuous summer daily cycle that allows periods of time to recover as it occurs under natural conditions. Although negative effects have been detected in several sperm parameters, fertility and prolificacy were not affected, suggesting a recovery of the reproductive function when normal conditions are reestablished., (Copyright © 2015 Elsevier Inc. The onset of sexual cycle postpartum was described in the collared peccary (Tayassu tajacu). Serum progesterone and 17beta-estradiol profiles, vaginal smears and external genitalia were analyzed in 20 animals housed with their piglets during the first postpartum month. The appearance of external genitalia showed no variation in any of the females: a shallow, reddish vulva, and vaginal mucus were constant features throughout the study. Based on hormonal profiles and vaginal smear cell patterns, 16 (80%) of the 20 peccaries showed signs of estrus and were considered cycling. The remaining four females (20%) did no show signs of estrus confirmed by low levels of progesterone (0.9+/-0.4 ng/mL) during the first postpartum month. In the cycling peccaries, a serum 17beta-estradiol peak (53.4+/-8.1 pg/mL) was observed on Day 7+/-1 postpartum, along with a linear increase in progesterone concentration from 3 (4.3+/-2.6 ng/mL) to 11 (30.8+/-4.9 ng/mL) days after this estradiol peak. Proportions of the different cells of the vaginal epithelium also changed in these females: superficial plus intermediate cells amounted to 76% of the cell total between Days 6 and 9 postpartum, corresponding to the estradiol peak. Nine (56%) of the 16 cycling females mated, indicated by the presence of sperm cells in their vaginal smears, and 6 (67%) became pregnant, reaching term. Non-pregnant cycling females (n=10) showed a steady decrease in serum progesterone concentration from 11 to 23 days after the estradiol peak, when basal levels were attained and a new estradiol peak registered, indicating the resumption of cyclicity in these females. The time interval between the two estradiol peaks was 23.5+/-2.1 days in these females. In pregnant females, progesterone concentrations continued to rise to levels of 60 ng/mL (n=6) 23 days after mating. These findings indicate that the lactating collared peccary female can become cycling and fertile during the early postpartum period, and that a predominance of superficial plus intermediate vaginal cells can be taken as the first sign of estrus.
