Selfed and backcross progenies developed from tetraploid somatic hybrids between Solanum tuberosum (tbr) and S. commersonii (cmm) were characterized for nonacclimated freezing tolerance (NA) and acclimation capacity (ACC) (two independent genetic components of freezing tolerance) under controlled environments. The segregation covered 28% and 71% of the parental range for NA and ACC, respectively, with the distribution skewed toward the tbr parent. Therefore, ACC appeared to be relatively easier to recover in the segregating generation. Some first backcross progeny had greater freezing tolerance than the cultivated parent primarily through the increase in ACC. When grown in the field, the improved freezing tolerance observed in the selfed progeny under controlled conditions was confirmed. Among NA, ACC, and freezing tolerance after acclimation (AA, which is the cumulative performance of NA and ACC), AA exhibited the highest correlation coefficient with field frost tolerance. In addition to freezing tolerance, vine maturity and tuber traits including tuber yield, tuber number per plant, mean tuber weight, and specific gravity were also segregating. No significant correlation between undesirable tuber traits and freezing tolerance was detected. Vine maturity and freezing tolerance were significantly correlated, so more careful selection for earliness was necessary in incorporating freezing tolerance. Yield comparable or superior to the backcross parent Wis AG 231 and an early Canadian cultivar, ‘Sable’, was found in many backcross progeny and some selfed progeny. The observed high yield can be attributed to the increase in mean tuber weight as well as tuber number. Moreover, a high portion of progeny had a specific gravity higher than 1.085, and some greater than 1.1. The implications derived from this study in breeding for freezing tolerance and further use of these materials are discussed.

The expression of freezing tolerance was examined in interspecific F1 and somatic hybrids of potatoes using 20 species and 34 different combinations between hardy and sensitive species. In the field, the frost tolerance of hybrids resembled either that of the hardy parent, the sensitive parent, or the parental mean, depending on the species combination and the genomic ratio (ratio of the number of sets of chromosomes contributed from each parent). Similar phenomena were observed when the non-acclimated freezing tolerance (NA) and the acclimation capacity (ACC) (two independent genetic components of freezing tolerance) were evaluated separately under controlled environments. In general, the expression level of freezing tolerance was higher in hybrids with more genomes contributed from the hardy parent than from the sensitive parent. In addition, the effectiveness or combining ability of genes conferring freezing tolerance from the hardy species also showed some influence on the expression of freezing tolerance. All three parameters, namely NA, ACC and acclimated freezing tolerance (AA) (NA plus ACC), were significantly correlated to the frost tolerance exhibited in the field. This indicates that the controlled freezing test used in this study could provide a good estimate of field performance. The implications of these results in breeding for freezing tolerance in potatoes are discussed.

An important question in the conservation of potato germ plasm is whether germ plasm in the gene bank, although stable, still represents the in situ populations from which it was collected, sometimes many decades ago. The answer would direct objective decisions regarding the value of re-collections and in situ preservation. The present study was undertaken as a project of the Association of Potato Inter-gene-bank Collaborators (APIC). It measured genetic differentiation between potato germ plasm maintained in the US gene bank for many years and current in situ populations re-collected from the same original sites in the wild. Solanum jamesii and Solanum fendleri from the United States were used as representatives of potato germ plasm. Re-collections were carried out in 1992 at the same locations at which gene bank-conserved accessions had been collected in 1958 and 1978. RAPD markers revealed significant genetic differences between gene bank-conserved and re-collected in situ populations for all seven comparisons of S. jamesii (diploid outcrosser), and 12 of 16 comparisons within S. fendleri (tetraploid inbreeder). The average genetic similarities were 65.2% for S. jamesii and 80.4% for S. fendleri. Possible explanations and consequences of these unexpectedly large differences are discussed.

Effects of gene bank seed-increases on the genetic integrity of potato germ plasm is a major concern of gene bank managers. Thus the Association of Potato Inter-gene-bank Collaborators (APIC), a consortium of world potato gene bank leaders, initiated this joint research project using RAPD markers to determine genetic relationships between increased generations within accessions. Solanum jamesii (2n=2x=24) and S. fendleri (2n=4x=48), two wild potato species native to North America, were used as plant material. These species represented two major breeding systems found among Solanum species: outcrossing diploids and inbreeding disomic tetraploids, respectively. Comparisons were made between populations one generation apart and between sister populations generated from a common source. Fourteen such comparisons within S. jamesii accessions had an average similarity of 96.3%, and 21 such comparisons within S. fendleri accessions had an average similarity of 96.0%. No pairs of populations were significantly different, despite the fact that RAPD markers easily separated all of these very similar accessions within their respective species. Only one of six S. jamesii accessions analyzed showed a significant change in gene frequencies among generations. These findings indicate that there has been minimal loss or change of genetic diversity in ex situ germplasm using the gene bank techniques standard at NRSP-6 and other world potato gene banks.

The inheritance of endosperm balance number (EBN), a genetic, dose-dependent crossability system functioning in tuber-bearing Solanum (potato) species, was investigated for certain wild potato species having an EBN equal to one half of their ploidy. The EBN of Solanum acaule, a disomic 4(2EBN) South American species, was investigated by producing F1 and F2 hybrids with artificial 4x(2EBN) S. commersonii. This allowed assessment of recombination among the two genomes of disomic S. acaule and that of S. commersonii. When crossability of the hybrids with 1EBN, 2EBN and 4EBN standards was tested, no variation for EBN was detected. The apparent lack of recombination and segregation for EBN in these hybrids indicates that the genomes of S. acaule and S. commersonii carry EBN in a genetically-similar way. Combined with previous reports, these data indicate that the inheritance of EBN is similar in widely-separated taxa from South America and Mexico.

Endosperm Balance Number (EBN) is a genetic, dose-dependent crossability system functioning in tuber-bearing Solanum species. Each species has been assigned 1EBN, 2EBN, or 4EBN. Species thus designated cross only within their EBN group. Doubling of chromosome number also doubles the EBN. The ploidy: EBN ratio is not consistent among Solanum species. Some diploids are 2EBN while others are 1EBN. Some tetraploids are 4EBN while others are 2EBN. Species from Mexico typically have EBNs that are one-half of their ploidy [e.g. 2x(1EBN), 4x(2EBN)]. Hybrids of Mexican species and a South American species, 2x(1EBN) S. Commersonii, and its 4x(2EBN) colchicine derivative were made and crossed to 1, 2, and 4EBN standard testers to determine the relationship of the genetic organization of EBN among and within these species. Diploid hybrids crossed only to 1EBN standard testers. Hybrids of 4x(2EBN) S. commersonii and 4x(2EBN) Mexican species crossed almost exclusively to 2EBN standard testers. Complex tetraploid hybrids involving S. commersonii, S. stenophyllidium (a Mexican diploid), and Mexican tetraploids of series Longipedicellata also crossed only to 2EBN testers. The apparent lack of recombination and segregation for EBN in these hybrids indicates that the genomes of the Mexican diploid and tetraploid species carry EBN in a way genetically similar to that of the South American species S. Commersonii.
