Plant-associated microorganisms play an important role in controlling heavy metal uptake and accumulation in aerial parts. The microbial community and its interaction with Cd accumulation by willow were assessed to explore the association of phytoextraction efficiency and rhizospheric microbial populations. Therefore, the rhizosphere microbial compositions of three willow genotypes grown in two Cd polluted sites were investigated, focusing on their interactions with phytoremediation potential. Principal coordinate analysis revealed a significant effect of genotype on the rhizosphere microbial communities. Distinct beneficial microorganisms, such as plant growth promoting bacteria (PGPB) and mycorrhizal fungi, were assembled in the rhizosphere of different willow genotypes. Linear mixed models showed that the relative abundance of PGPB was positively associated (p 0.01) with Cd accumulation, since these microbes significantly increased willow growth. The higher abundance of arbuscular mycorrhizal fungi in the rhizosphere of Salix × aureo-pendula CL 'J1011' at the Kejing site, showed a negative correlation with the Cd content, but a positive correlation with biomass. Conversely, mycorrhizal fungi, were more abundant in the rhizosphere of S. × jiangsuensis CL. 'J2345' and positively correlated with the Cd content in willow tissues. This study provides new insights into the distinctive microbial communities in rhizosphere of different willow genotypes, which may be consistent with the phytoremediation potential.

A mechanistic understanding of perfluorooctanoic acid (PFOA) toxicity to plants is essential for future risk assessment of PFOA in agricultural soil. In this study, soil-grown cucumber (Cucumis sativus) was exposed to 0, 0.2, and 5 mg/kg of PFOA for 60 days. At harvest, contaminant accumulation, cucumber biomass, photosynthesis profiles and metabolites were measured. Results showed that PFOA depressed cucumber biomass and accumulated highest in leaves. Photosynthesis analysis revealed that PFOA at both doses reduced the chlorophyll contents and net photosynthesis rate of cucumber leaves. Gas chromatography-mass spectrometry-based non-targeted metabolomics revealed that PFOA induced metabolic reprogramming in cucumber leaves, including up-regulation of phenols (at 0.2 and 5 mg/kg) and down-regulation of amino acids (at 5 mg/kg), indicating disrupted nitrogen and carbon metabolism. Results revealed how PFOA represses plant growth by down-regulating photosynthetic pigments and disturbing the metabolism of carbohydroxides, phenols and amino acids. These findings provide valuable information for understanding the molecular mechanisms involved in plant responses to PFOA-induced stress.

Tetrabromobisphenol A (TBBPA) is one of the most widely used brominated flame retardants worldwide. The degradation and fate of this organic pollutant of soils is of great concern and can be strongly affected by geophagous earthworms through ingestion and burrowing activities. Using 14C-tracers, we studied the effects of the geophagous earthworm Metaphire guillelmi on the mineralization, metabolism, and bound-residue formation of TBBPA in a typical Chinese rice paddy soil during 30days of incubation in the laboratory. Earthworms significantly decreased both mineralization (from 3.9±0.3% of the initial amount to 2.6±0.2%) and dissipation (from 90.6±0.6% to 84.1±1.2%) of TBBPA in the soil, and stimulated the generation of O-methylation metabolites (TBBPA methyl ethers; from 1.4±0.4% to 15.4±0.6%). This resulted in a strong decrease in bound-residue formation of TBBPA and its metabolites in the soil (from 80.3±0.4% to 41.8±3.1%). Results from a first-order, two-compartment model that describes the fate of TBBPA in soil indicated that the TBBPA-derived bound residues were mainly attributed to the binding of metabolites to the soil matrix and not to the binding of TBBPA, and that earthworms reduced the kinetic rates of both polar metabolite generation and their bound-residue formation. Our results suggested that the geophagous earthworm Metaphire guillelmi strongly influenced the fate of TBBPA by altering the composition of metabolites and therefore bound-residue formation. The increased persistence of TBBPA and the formation of persistent O-methylation metabolites by M. guillelmi would increase the environmental risk of TBBPA.
