Magnetic beads (MBs) are often considered as an effective carrier in heterogeneous assays due to the simplicity of separation and washing, and the ability to increase and control the surface area. However, the effect of the MBs surface on the analytical parameters is poorly characterized and is often postulated from intuitive considerations. Herein, experimental evaluation through the comparison of MBs and microwell plate was carried out using the miRNA-141 (biomarker for cancer) as a target, the detection of which was performed by chemiluminescent assay with a homogeneous mismatched catalytic hairpin assembly (mCHA) reaction. The mCHA reaction produced double-stranded (ds) DNA labeled at one end with fluorescein (Flu) for capture with anti-Flu antibodies immobilized on a solid carrier, on the other end with biotin for recognition by streptavidin-polyperoxidase conjugate. The conditions of immobilization of anti-Flu antibody on MBs (a diameter of 440 nm) performed using a carbodiimide method were optimized by varying the antibody concentration in the reaction solution. It was shown that the dependence of chemiluminescent signal as a function of the concentration of anti-FluAb-MBs conjugates had a bell-shaped character. The maximum chemiluminescence was produced at the concentration of the conjugates of 2 × 10 9 particles/mL, with a surface area of 65 mm 2 . The identical surface area was used upon the assay performance with polystyrene microplates. Comparison of MBs- and microplate-assays for miRNA-141 determination showed that the obtained calibration curves and their detection limit values were the same and did not depend on the used carrier. We report the approach for the detection of Au@Pt core@shell nanoparticles (nanozymes) with peroxidase-mimicking activity (PMA) in samples with high endogenous peroxidase activity (EPA). Unlike the endogenous peroxidases in plant extracts that are inhibited by elevated H 2 O 2 (>20 mM), the PMA of nanozymes was stable in concentrated H 2 O 2 (up to 4 M). Such a different stability of enzymes and Au@Pt to the substrate allowed for eliminating EPA and detecting only nanozymes. The developed approach was used for reaching a lower limit of detection (LOD) and eliminating the background for the lateral flow immunoassay (LFIA) of the important plant pathogen potato virus X (PVX) in leaf and tuber extracts. Using the PMA of Au@Pt, the LOD was reduced to 4 and 8 pg/mL in tuber and leaf extracts, respectively. The LOD values are 250- and 500-times lower in comparison with LFIA with conventional gold nanoparticles. We propose nucleic acid lateral flow assay (LFA) coupled with reverse transcription recombinase polymerase amplification (RT-RPA) resulting from step-by-step multiparametric adjustments to both RT-RPA reactions and LFA interactions. The assay was realized for RNA virus detection using the example of potato virus X (PVX), a dangerous phytopathogen. The assay stages were adjusted for sensitive detection. (1) DNA target was designed and verified. A fragment (146 bp) of coat protein gene (gp5) and biotin-/fluorescein-labeled forward/reverse primers were chosen to produce target amplicons. (2) In a test strip, the construction advantage of the realization of the highest affinity interaction (biotin-streptavidin in our research) through gold nanoparticle conjugate (streptavidin immobilized on the GNP surface) was demonstrated. (3) RPA with reverse transcription was adjusted including primer concentration, order of components' mixing, and reaction temperature. Due to the adjustments, the assay was able to detect 0.14 ng PVX per g potato leaves at 30 min. The PVX assay was 260 times more sensitive than conventional lateral flow assay based on antibodies and demonstrated the same sensitivity as PCR detection. This study presents new type of the lateral flow immunoassay (LFIA) for multi-target analysis. A test, named alarm-LFIA, has an essentially new function that consists in notice (signaling the danger) about the presence at least one target from the controlled list without identification. The design of the alarm-LFIA assumes one test zone, which contains a mixture of antibodies, and multi-specific conjugate that binds the several targets. The alarm test is based on the novel conjugate with broaden specificity due to the immobilisation of a mix of antibodies, specific to several structurally different targets, on the surface of gold nanoparticles. For proof of concept, multi-specific conjugate to five important potato viruses (potato virus X, -M, -S, -Y and potato leaf roll virus) was fabricated using five antibodies with different specificity. The alarm-LFIA was developed for rapid detection of the total infection caused by up to five viruses. Detection limits of the viruses in potato leaf extracts are from 10 to 30 ng/mL. The alarm-LFIA was successfully used for viruses' detection in potato leaves; results were confirmed by enzyme-linked immunosorbent assay. The scheme of immunochromatographic competitive analysis with "external" specific antibodies is offered for the first time. The test strip includes all the necessary components for analysis (membranes, protein antigen conjugates, colored nanoparticles), except specific free unconjugated antibodies that are transferred to the buffer used for sample dilution. Thus, the concentrations of the antibodies and markers can be chosen independently, and dilution of the sample with buffer (a routine procedure for any immunochromatographic test) ensures pre-incubation of the antibodies with the sample. Both factors increase the sensitivity of the test. The proposed scheme has been proven to be effective for the determination of deoxynivalenol and T-2 toxin; detection limits of 500pg/mL and 50pg/mL, respectively, were achieved. This is an order of magnitude lower than the limits of detection of traditional tests using the same reagents. The analysis was conducted in water-organic extracts (20% methanol); the duration of the analysis is 10min (it's the same time as for a traditional test). A barcode lateral flow immunoassay (LFIA) based on magnetic nanoparticles with a controllable cut-off level was developed for the first time. The regulation of the cut-off levels was based on the changes in the position and concentration of three antibodies (two monoclonal and polyclonal) with different affinities (K D =4.22×10 -9 , 9.67×10 -9 , 1.05×10 -8 M), respectively. To obtain specific conjugates, monoclonal antibodies were covalently immobilized on the magnetic nanoparticles' surface. Potato virus X causing a reduction in the potato yield was used as a model polyvalent antigen. To detect potato virus X in the leaf extracts, a barcode LFIA with cut-off levels of 3, 30, and 150ng/mL was used for the analysis. The application of magnetic concentration leads to a six-fold reduction in the first cut-off level (0.5ng/mL) in comparison with magnetic LFIA without the concentration stage., (Copyright © 2016. Fluorescence polarisation immunoassays (FPIAs) based on monoclonal antibodies for detection of three strobilurin fungicides - kresoxim-methyl (KM), trifloxystrobin (TF) and picoxystrobin (PC), were developed and optimised. Fluorescein-labelled derivatives of target antigens (tracers) were synthesised and purified by thin-layer chromatography. Influence of tracer structures on the assay parameters was investigated. For KM and TF, the best assay performance was achieved with the homologous pairs of reagents. For the PC assay, the heterologous tracer, i.e. fluorescein-labelled derivative of TF, was used. The developed FPIAs were applied to the determination of KM, TF and PC in red wine. Most optimal sample preparation was achieved with cross-linked poly(vinylpyrrolidone) as a sorbent. This clean-up is simple, rapid and allows determination of all three strobilurin fungicides in one sample. Detection limits of the developed FPIAs in red wine were 28, 6 and 5ng/mL for KM, TF and PC, respectively. Recovery in spiked samples averaged between 80% and 104%. Intra- and inter-assay coefficients of variance were less than 12%. Ralstonia solanacearum is a dangerous and economically important pathogen of potatoes and other agricultural crops. Therefore, rapid and sensitive methods for its routine diagnostics are necessary. The aim of this study was to develop a rapid control method for R. solanacearum with a low limit of detection (LOD) based on a lateral flow immunoassay (LFIA) with silver enhancement. To minimize the LOD, the membrane type, antibody amount for conjugation with gold nanoparticles, conjugate concentration and antibody concentration in the analytical zone were optimized. Silver enhancement was used to decrease the LOD of the LFIA. For silver enhancement, release fiberglass membranes with pre-absorbed silver lactate and hydroquinone were placed on the analytical zone, and a drop of silver lactate was added. The LFIA with silver enhancement was found to be 10-fold more sensitive (LOD 2×10(2) CFU/mL; 20 min) in comparison with the common analysis (LOD 2×10(3) CFU/mL; 10 min). The specificity of the developed LFIA was studied using different strains of R. solanacearum (54 samples) and other widespread bacterial pathogens (18 samples). The LFIA detected all tested strains, whereas non-specific reactions were not observed. The developed tests were used for the control of bacteria in extracts of infected and non-infected potato tubers, and the quantitative analysis results (based on the densitometry of line colouration) were confirmed by ELISA with a correlation coefficient equal to 0.965., (Copyright © 2016 Elsevier B.V. An immunochromatographic assay was developed to detect fluoroquinolone antibiotic ofloxacin based on the competitive binding of ofloxacin and the membrane-immobilized ofloxacin-protein conjugate to colloidal gold-labeled antibodies in the course of the labeled antibodies, and to test sample flow through the membrane. The specific feature of labeling by colloidal gold is that native antiserum is used instead of purified immunoglobulins or specific antibodies. This makes the synthetic procedure easier, with no sacrifice in the detection limit. The proposed test makes it possible to detect down to 30 ng mL(-1) of ofloxacin, which corresponds to the demands of food safety assessment. The assay time is 10 min. The assay provides reliable information on the ofloxacin content in milk without the sample preparation and in chicken and pork meat with the minimum sample preparation (the separation of the insoluble fraction of the homogenate by centrifugation). The high degree of detection of ofloxacin in foodstuffs by the proposed assay (70-112%) was shown by a comparison with the data obtained with the use of a commercial immunoenzymatic kit., (© 2013 Published by Elsevier B.V.)

A pretreatment-free immunochromatographic assay for detection of chloramphenicol (CAP) in milk was developed. The assay is based on competition between CAP molecules in the sample and immobilized CAP-protein conjugate for binding to monoclonal anti-CAP antibodies conjugated with colloidal gold particles (average diameter 30nm). The assay is carried out in the course of sample flowing along test strip with immobilized reactants, and its results can be detected by the naked eye or by a photometric device. Effect of the concentration of immunoreactants on assay characteristics was studied. The assay protocol with maximal sensitivity and reliability was optimized using measured values of brightness of lines. Detection limit for CAP is 10ngmL(-1). Assay duration is 10min, and it can be carried out at room temperature without any additional devices and reactants. The developed test strip has been applied to CAP detection in dairy products.

Conventional immunoassays on microtiterplates are very useful analytical tools in environmental analysis, but the long assay times, usually in the range of hours, are a drawback. To overcome this disadvantage, the development of fast (express) assay formats is described, which use polyelectrolytes as carriers. Two semi-homogeneous immunochemical methods, namely the polyelectrolyte-ELISA (enzyme-linked immunosorbent assay) and the express-FIIAA (flow injection immunoaffinity analysis) for the analysis of the herbicide atrazine were set-up. Using polyclonal antisera for atrazine, the following results were obtained. Standard curves for atrazine showed a linear range from 3 to 100mugl(-1) in polyelectrolyte-ELISA and 0.3-100mugl(-1) in express-FIIAA. The test midpoints in polyelectrolyte-ELISA and express-FIIAA were 12 and 5mugl(-1), respectively. The duration time of the immunochemical reaction was in both assays 15min, but the total assay time differed (30min (polyelectrolyte-ELISA) and 18min (express-FIIAA)). A significant difference between the formats could be observed in the number of samples that can be determined per day. The polyelectrolyte-ELISA can handle samples in parallel on a microtiterplate (usually 20/plate), whereas in the express-FIIAA the samples are automatically analysed one after another. This first demonstration of these techniques shows the potential of these methods, but also their limitations.

Immunoenzyme assay (ELISA) is proposed and characterized for determination of alkylphenol ethoxylates, a primary class of manufactured non-ionic surfactants. The assay is based on the obtained polyclonal antibodies against nonylphenol (NP), the main stable intermediate of the decomposition of nonylphenol ethoxylates. A mixture of non-modified branched isomers of NP was applied as hapten coupled to protein carriers by Mannich reaction with the use of formaldehyde. The proposed ELISA format is based on immobilized NP-(soybean trypsin inhibitor) conjugate as a competitor of antigen molecules contained in the tested sample for binding with specific antibodies indirectly labeled via an anti-species immunoperoxidase conjugate. The developed ELISA allows to reveal NP with the limit of detection about 10ngml(-1) and NP-related compounds such as octylphenol, alkylphenoletoxylates, alkylphenolcarboxylates and their halogenated derivatives. The ELISA was applied for assaying polluted water samples, namely influents and effluents from different wastewater treatment plants (WWTP) and tap water. ELISA and chromatographic data demonstrate good correlation (r = 0.94), while ELISA gives higher values. Due to endocrine disrupting and other toxic activities of some metabolites of alkylphenolic non-ionic surfactants, the developed assay may be effectively used in ecological monitoring and sanitary control.
