With increasingly improved analytical performances, miniature mass spectrometers are expandingly applied in more application scenarios. The capability of high throughput analysis of multi-targets is a crucial function of a miniature mass spectrometer, especially for in-situ applications. As a powerful method for target screening, multiple reaction monitoring (MRM) was conventionally realized on triple quadrupole (QQQ) mass spectrometers. Previously, pseudo-multiple reaction monitoring (pseudo-MRM) operation mode has been realized on a "brick" mass spectrometer equipped with a single ion trap. However, pseudo-MRM on an ion trap was realized through the tandem-in-time fashion, and the throughput of pseudo-MRM was still limited. As a continuous effort to boost performances of the "brick" miniature mass spectrometer, parallel pseudo-MRM mode was developed, and the throughput could be raised up to 10 times for multi-target screening. To achieve better isolation and collision induced dissociation (CID) efficiencies, the mass range of interest was divided into several segments based on optimized Mathieu q value. The fast screening of 10 different drugs was used as an illustrative example. Furthermore, parallel quantitative analysis was also demonstrated and verified using the internal standard calibration method.

The study focuses on the development of a new procedure for the preparation of reference samples with a given concentration for X-ray fluorescence analysis by adding certain volume of analyzed elements solutions with a known concentration to certified reference rock materials and further fusion with borate fluxes. The presented method of preparing emitters allows not only to obtain samples with the required concentrations for the elements to be determined, but also to preserve the influence that other elements have on the analytical signal. A set of 12 certified reference samples of rocks was used to construct calibration dependencies. The preparation of multicomponent modified reference samples (MRS) was carried out on the basis of certified reference materials (CRM) included in the set for constructing calibration curves. An electrokinetically pumped sheath-flow nanoelectrospray interface provides an efficient means of transferring ions from a capillary electrophoresis system into a mass spectrometer. To characterize its performance, we analyzed angiotensin solutions prepared in a background of 0.25 mg/mL of a BSA tryptic digest. Calibration curves were prepared from 10 zeptomole (10(−21) mol) to 10 femtomole (10(−14) mol) of angiotensin injected into the capillary. A parallel reaction monitoring approach was used; MS1 was set to m/z = 523.8 ± 2, and fragment ion signals at 263.1389 (y(2+)) and 784.4095 (b(6+)) were used to generate selected ion electropherograms. Calibration curves based on peak areas were linear (log-log slope of 0.94 for the y(2+) fragment and 0.98 for the b(6+) fragment). We then injected 1-zmol (600 copies) of angiotensin in the BSA background using a 10-μm ID separation capillary. Triplicate analyses consistently produced co-migrating peaks for the two fragment ions. The b(6+) electropherogram showed no background signal, whereas the y(2+) electropherogram showed a few noise spikes that were smaller than the peak maximum. Bienayme-Tchebycheff inequality was used to estimate detection limits of 230 ymol (140 ions) injected into the separation capillary. To the best of our knowledge, these electropherograms present the smallest number of molecules detected using mass spectrometry coupled with a separation.

In this work, the analytical performance of a third-order/four-way calibration is evaluated to model a highly overlapped system, where two spectral dimensions are extremely similar, and the results are then compared with the results of second-order/three-way calibration. The four-way data were obtained during the photodegradation of fluoroquinolones (ciprofloxacin, norfloxacin and flumequine) in the form of excitation-emission matrices and modeled by unfolded partial least squares coupled to residual trilinearization (U-PLS-RTL). According to the results, the model obtained with the second-order algorithm (unfolded partial least squares coupled to residual bilinearization: U-PLS-RBL) was unsatisfactory due to high spectral overlap. The third-order approach obtained a satisfactory fit and better figures of merit (LOD, REP, RMSEP, and sensitivity, among others) even in the presence of unexpected interferences due to third-order advantages. Finally, the analytical method based on third-order multivariate calibration was applied to quantify these fluoroquinolones in spiked fish-farming water samples. In this case, the third-order advantage allowed us to satisfactorily model the data and to quantify these compounds in this complex matrix, demonstrating the superior analytical performance of the high-order data that were evaluated.

Fabricating simple, accurate and user-friendly diagnostic device for “point of care testing” (POCT) applications is one of the most challenging objectives in the analytical field. Hemin detection is important for drugs monitoring, diagnosis, and forensic latent bloodstain imaging. Herein is developed, luminol chemiluminescence biosensor for hemin detection using artesunate as coreactant. A possible mechanism to account for the chemiluminescence reaction is discussed. Hemin was detected using both photomultiplier tube (PMT) and smartphone as detector. The detection limit for hemin using smartphone as detector is 20 nM, enabling the visual detection of hemin in blood sample with a dilution factor of blood up to 120,000. While PMT detector is used, the system is able to detect hemin down to 0.22 nM. In addition to high sensitivity, this sensing system exhibit high selectivity. It can successfully distinguish bloodstain from other stains while applying the system for point of care testing using smart phone as detector. Moreover, the system can detect artesunate with a linear range from 0.1 nM to 1.0 μM with a limit of detection of 0.078 nM.

In this study, we used nano-smectite (SMT) and multiwall carbon nanotube (MWCNT) as a recognition surface over the disposable pencil graphite electrode for the low-level detection of ascorbic acid (AA) and acetyl salicylic acid (ACA). Firstly, nano-semectit was obtained by the purification of natural bentonite clay. XRD patterns and Scanning electron microscopy (SEM) were employed to ensure the purity of the bentonite samples. Pencil graphite rodes were immersed into suspensions of SMT, MWCNT or the both.The surface morphology of the sensor was investigated by cyclic voltammery (CV), electrochemical impedance spectroscopy (EIS) and SEM techniques. The effect of pH, percentage composition of modifiers, immobilization time, accumulation time and accumulation potential was optimized for reaching the best electroanalytical response for AA and ACA. The sensor developed proved to give good recovery, notable electro-catalytic activity and appropriately distant signals for the simultaneous determination of the analytes. Common contaminates, uric acid, l-cysteine, dopamine, glucose, Na+, Cl- and citric acid were investigated for their interference effects. Uric acid, citric acid and dopamine were found to interfere to some extent. The electrode exhibited wide working ranges and a fairly satisfactory detection limit of 0.096 and 0.241 μM for AA and ACA, respectively. The electrode system proved to be practical for the analysis of pharmaceutical tablets and the recovery results are very satisfactory.

In this work, a simple and cost-effective method was developed for Na, Mg, Al, K, Ca, Ti, V, Cr, Mn, Ni, Cu, Zn, Rb, Sr, Ba and Pb determination in glass samples employing suspension sampling and total reflection X-ray fluorescence (TXRF). The experimental conditions were optimized using multivariate approach being the optimum conditions located by Derringer's desirability function. Proposed method was validated in accordance with the National Institute of Metrology, Quality and Technology (Inmetro, Brazil). Limit of detection (LD) and limit of quantification (LQ) were adequate for determination of trace elements in glass. The evaluation of the accuracy and precision was realized by analysis of standard reference materials of glass (NIST 612). For the majority of the elements, good agreement was achieved between the certified value and the value obtained in the NIST 612. The relative standard deviation (RSD%) was achieved between 3.6 and 10.3%. Moreover, no significant differences were observed between the proposed method compared to the ICP-MS. After the validation step, the method was applied to 31 glass samples and with aid of an exploratory principal component analysis (PCA), a perfect discrimination of the glass from originally manufactured smartphones was obtained. In addition, soda-lime glass could be reasonably distinguished from smartphone screens. The developed method is fast, consumes low amounts of reagents and allows the determination of many analytes simultaneously. Additionally, the method does not require calibration curves. Therefore, TXRF proved to be attractive and useful for routine analysis of glass.

In the past few years, microwave induced plasma optical emission spectrometry (MIP-OES) has generated great interest as an alternative technique to inductively coupled plasma-based techniques due to its lower operational cost. Since MIP-OES suffers from severe matrix effects due to easily ionizable elements (EIEs) (Na, Ca, etc.), it is unclear whether this technique could be employed for elemental bioavailability studies in soils and sediments since the main extractant solutions employed in such works may contain high levels of these elements. Thus, the aim of this work was to evaluate the feasibility of MIP-OES as a detector for such applications. To this end, the influence of different extractant solutions (0.25 mol L−1 MgCl2, 0.25 mol L−1 CaCl2, 0.10 mol L−1 acetic acid, 0.05 mol L−1 Na2EDTA, 0.25 mol L−1 NaNO3, 0.25 mol L−1 NaOAc/HOAc and 0.10 mol L−1 NH2OH·HCl) on the analyte emission of several elements (As, Cd, Co, Cr, Cu, Fe, Mg, Mn, Mo, Ni, Pb, Rh, Se, Sr and Zn) was investigated. Results were compared to those obtained using a reference solution made of 5% w w−1 HNO3 solution. For saline extractant solutions, both the optimum nebulizer gas flow rate (Qg) and analyte emission were modified with regard to the reference solution. In general, the optimum Qg was reduced by between 0.1 and 0.2 L min−1 for both ionic and atomic lines. Under optimum Qg conditions, analyte emission was supressed by saline solutions except for atomic lines with an upper electronic state below 4 eV, which were enhanced. The magnitude of matrix effects was strongly dependent on EIE ionization energy. The lower the ionization energy, the greater the matrix effects were registered. No measurable matrix effects were registered on both Qg and analyte emission within experimental uncertainties for NH2OH·HCl and acetic acid extractant solutions. Experimental data suggest that matrix effects were related to changes in plasma characteristics and the analyte excitation/ionization mechanism. To mitigate matrix effects and improve long-term MIP-OES performance, internal standardization using either Rh (343.489 nm and 369.236 nm) or OH molecular emission band (308.958 nm) was required. This calibration methodology was successfully applied to the study of the elemental bioavailability in soil samples from a vineyard affected by copper-based fungicides and sediment samples from an area affected by mining waste. This paper presents the Outer Plug Standardization - OPS, a novel technique as an alternative for the classic internal standardization using multiple injection in capillary zone electrophoresis is proposed herein. This technique was applied in a new method for the determination of chloride, nitrate and sulfate in rainwater samples. After the injection accuracy was tested and proved to be a minor error source (average 1.26% RSD), the OPS was applied and it improved the intra-day and inter-day precision of the analytical method by 32.5% and 24.7%, respectively. Using a capillary with effective length of 23.5 cm the electrophoretic separation of the three inorganic anions could be achieved in1 min, with detection limits of 0.05, 0.09 and 0.11 mg L

Ambient ionization in open environment brings a capability of a coupled mass spectrometry to detect target molecules in situ. However, it is limited to qualitative and semi-quantitative analysis. By coupling of an ambient ionization-based Direct Analysis in Real Time (DART) with high-resolution quadrupole time-of-flight mass spectrometry (QTOF/MS), we observe that, in one-chemical system, the target molecule displays a non-linear response in MS signal vs concentration, accompanying with large variation in MS signal, suggesting two obstacles for quantification to be overcome. Surprisingly, in a two-chemical system, we observe an apparent suppression effect. We prove that, due to this observed suppression effect, a fluctuant response in the MS signal of the stable isotope-labeled analogue can immediately reflect the change in the analyte concentration and ionization efficiency. For example, by taking advantage of this effect, even the analyte of different concentrations despairingly displayed similar signals would be accurately calibrated through the suppression of the internal stable isotope standard. This puts an important foundation on accurate and linear quantitation of analytes in complex matrix using DART-MS assay. Moreover, we for the first time demonstrate an application of in situ calibration of DART-MS for direct and accurate quantification of target molecule (creatinine) in highly complex samples (human urine) without any pre-separation. The quantification is also validated using HPLC-UV analysis (n = 38). At last, we show that stable isotope-labeled-creatinine (m/z 117.0850 amu) can be used for simultaneous in situ calibration of some other urinary metabolites with a mass/charge ratio varying from 120.069 amu to 333.125 amu.

This study describes, for the first time, the development of two platforms of competitive fluorescent immunoassays for bioanalysis of crizotinib (CZT), a potent drug used for the treatment of non-small cell lung cancer (NSCLC). These platforms were microwell-based heterogeneous fluoroimmunoassay (FIA) and a kinetic exclusion assay (KinExA) with KinExA™ 3200 immunosensor. Both FIA and KinExA were developed using same reagents; mouse anti-CZT antibody and a capturing reagent of CZT conjugated with bovine serum albumin (CZT-BSA). In the FIA, the CZT-BSA coated onto the microwells of the assay plate was present simultaneously in the assay mixture (CZT and its antibody). In the KinExA, the antibody was allowed to pre-equilibrate with CZT, and then the incubation mixture was rapidly passed through a microcolumn containing CZT-BSA coated onto polymethyl methacrylate (PMMA) beads. The analytical performances of both assays were comparatively evaluated in terms of assay working range, limit of detection, precision profile, and accuracy. The results revealed that KinExA yielded higher sensitivity and better precision than FIA; whereas, both assays had comparable accuracies. Both FIA and KinExA were superior to all the existing chromatographic methods for CZT in terms of the assay sensitivity, convenience, analysis throughputs. The proposed FIA and KinExA are anticipated to effectively contribute to the therapeutic drug monitoring (TDM) of CZT in clinical settings.

This article describes a new, simplified approach for performing quantitative colorimetric assays on paper-based analytical devices that uses calibrant-loaded paper devices to perform external calibration and standard addition calibration using one calibration point. Calibrant-loaded devices consist of sensing areas pre-loaded with a colored product which is produced from the reaction of a standard solution of the analyte with the appropriate colorimetric reagents. When the sample is added into the calibrant-loaded sensing zone the analytical signal (i.e. color intensity) increases proportionally to the concentration of the analyte in the tested sample. The total measured signal corresponds to the sum of the concentration of the analyte in the sample and the standard solution pre-stored in the device and is used to calculate the concentration of the analyte in the sample based on the principles of linear calibration. The applicability of this approach was benchmarked in three colorimetric assays (i.e., for the determination of iron, nickel, and amino acids) that use different reaction chemistries. This work demonstrates a simplified calibration-free approach for assays performed on paper-based analytical devices that requires minimum experimental and computational effort, it can be used for either external or standard addition calibration and can be used to identify and eliminate the presence of interferences in a sample.

This article presents a new method for the simpler and faster determination of tin in beverages using EDXRF. Absorption coefficients for aqueous calibration samples were calculated and shown to be nearly identical to those of the beverage samples, thus permitting the use of aqueous standard solutions for external calibration. Beverage samples could then be measured directly using the external calibration. Determination of tin using this method takes 4 min. The LOD and LOQ were 4 mg L−1 and 15 mg L−1 respectively, and the precision was 3.89%. Different canned beverages (cold coffee, various fruit juices) were measured and the results compared to the concentrations obtained using ICP-OES after digestion. The two methods showed good compatibility, thus establishing the newly developed method as a rapid, simple, and accurate method for the determination of tin in beverages.

Fluorescent probes have been valuable tools for bioanalytical multiplex assays. However, as a common phenomenon in multiplex fluorescence assays, spectral overlap usually leads to difficulty in spectral analysis for multiple analytes. Although multiway calibrations have provided mathematic approaches for complex spectral analysis, it remains a grand challenge for these methods in practical applications because of the problems such as prior rank estimation. Herein, we report a novel second-order calibration algorithm of alternating residual trilinearization (ART) for the decomposition of complex spectra generated from multiplex fluorescence assays. By alternating iterative convergence to the spectral profiles of each component in convergence process, ART enables automatic rank estimation for second-order calibration, thus able to avoid the risk of chemical meaningless fitting of component spectra. Combined with fluorescence excitation-emission matrix (EEM) spectroscopy, the performance of ART has been demonstrated by a simulated example and an analytical experiment performed using molecular beacons (MBs) for the simultaneous assay of three breast cancer related DNA targets. The results revealed that the proposed algorithm is capable of automatic estimating the number of underlying components during its convergence process to produce acceptable performance in spectral profile resolution and concentration estimation. Compared with other existing iterative trilinear decomposition strategies such as parallel factor analysis (PARAFAC) requiring a prior rank estimation, the proposed ART therefore provides a robust second-order calibration strategy for complex spectral analysis in multiplex fluorescence assays.

A novel non-enzymatic superoxide anion radical (O2•−) sensor was prepared by silver nanoparticles (AgNPs) decorated organic frameworks (MOFs) compound-derived metal oxide coated carbon material. Transmission electron microscopy (TEM) was employed to characterize morphologies and structures of the as-prepared samples AgNPs/Co3O4@C. Electrochemical studies indicated that the AgNPs/Co3O4@C/GCE exhibited excellent detection ability to O2•−. It offered a linear range of 8 orders of magnitude and a super low detection limits of 0.0564 pM. The sensor was successfully adapted to detection of O2•− released from living cells. Furthermore, the capacities of scavenging O2•− of food antioxidants were also studied by electrochemical method at cellular milieu. Accordingly, this work provided tremendous potential for reduction incidence of oxidative stress by intaking food antioxidants.

We report the colorimetric detection of dopamine (DA) on microfluidic paper-based analytical devices (μPADs) using an oxidation-reduction method. Here, dopamine reacts with ferric chloride forming reduced Fe2+ that subsequently reacts with phenanthroline to form the red tris(1,10-phenanthroline)iron(II) complex. The devices were fabricated by wax printing and changes in color intensity were recorded using a common cell phone. Subsequent analysis using Photoshop software, yielded a limit of detection (LOD) for DA of 0.37 μmol/L with a linear range of 0.527–4.75 μmol/L and relative standard deviation of 0.11% (inter-day) and 0.15% (intra-day) for n = 15 paper chips. The effects of detection conditions have been investigated and are discussed. Cow serum samples and human blood serum and plasma samples were detected. The work, herein, demonstrates the potential of this method as a low cost and rapid colorimetric technique to detect DA in real samples.

A comparison of the performance of two electrochemical immunosensors for the determination of the anorexigen biomarker peptide YY (PYY) is reported by using as scaffolds screen printed gold electrodes modified either by oxidative grafting of p-aminobenzoic acid (p-ABA) or by assembling of a 4-mercaptobenzoic acid (4-MBA) SAM. Covalent immobilization of capture antibodies on the surface-confined carboxyl groups was carried out by EDC/NHSS chemistry, and competitive immunoassays between target PYY and Biotin-PYY were implemented. Upon labeling with alkaline phosphatase (AP)-streptavidin conjugate and 1-naphtyl phosphate addition, differential pulse voltammograms recorded between −0.2 and +0.7 V were used as analytical readout. All the steps involved in the functionalization of the electrodes and the preparation of the immunosensors were monitored by electrochemical impedance spectroscopy. The calibration plot for PYY using the AP-Strept-Biotin-PYY(PYY)-anti-PYY-Phe-N-SPAuE immunosensor provided a linear current vs. log [PYY] plot extending between 10−6 and 103 ng/mL PYY with a detection limit of 3 × 10−7 ng/mL. These analytical characteristics are remarkably better than those obtained with the immunosensor prepared with 4-MBA SAM-SPAuEs. The AP-Strept-Biotin-PYY(PYY)-anti-PYY-Phe-N-SPAuE immunosensor was used to analyze human serum and saliva samples spiked with PYY at concentrations fitting with normal levels in these biological fluids.

N-acetylcysteine (N-AC) has widespread application such as pharmaceutical drug and nutritional supplement. Its adverse effects are rash, urticaria, and itchiness and large doses of N-AC could potentially cause damage to the heart and lungs. Therefore, in this work, a sensitive voltammetric sensor based on a carbon paste electrode modified with silica nano particles (i.e. Mobil Composition of Matter (No. 41) modified with Boron Trifluoride or BF3@MCM-41) with a combination of 4,4'-dihydroxybiphenyl (DHB) (BF3@MCM-41/DHB/CPE) was designed for determination of N-AC. The electrochemical oxidation of N-AC was examined using various techniques such as cyclic voltammetry (CV), chronoamperometry and differential pulse voltammetry (DPV). Under the optimum conditions, some parameters such as electron transfer coefficient (α) and heterogeneous rate constant (ks) were estimated for N-AC. Due to the use of N-AC for the treatment of acetaminophen (AC) overdose, the application of modified electrode was investigated for the simultaneous determination of N-AC and AC in blood serum and tablet samples. Since, the signals of these species overlap and due to the presence of interfering species in blood samples, the simultaneous determination of mentioned species is difficult or impossible. To overcome this challenge, parallel factor analysis (PARAFAC) was used for the analysis of the complex matrices to obtain the spectral profile of each component and interference. To achieve this goal, electrochemical second-order data were generated using a simple change in pulse height of differential pulse voltammetry. The results of the presently proposed strategy for the real samples analysis are similar to those obtained with HPLC. Thus, the proposed method has acceptable performance for simultaneous determination of the two species in real samples.

A non-invasive textile-based colorimetric sensor for the simultaneous detection of sweat pH and lactate was created by depositing of three different layers onto a cotton fabric: 1.) chitosan, 2.) sodium carboxymethyl cellulose, and 3.) indicator dye or lactate assay. This sensor was characterized using field emission scanning electron microscopy and fourier transform infrared spectroscopy. Then, this sensor was used to measure pH and lactate concentration using the same sweat sample. The sensing element for sweat pH was composed of a mixture of methyl orange and bromocresol green while a lactate enzymatic assay was chosen for the lactate sensor. The pH indicator gradually shifted from red to blue as the pH increased, whereas the purple color intensity increased with the concentration of lactate in the sweat. By comparing these colors with a standard calibration, this platform can be used to estimate the sweat pH (1−14) and the lactate level (0–25 mM). Fading of the colors of this sensor was prevented by using cetyltrimethylammonium bromide (CTAB). The flexibility of this textile based sensor allows it to be incorporated into sport apparels and accessories hence potentially enabling real-time and continuous monitoring of sweat pH and lactate. This non-invasive sensing platform might open a new avenue for personal health monitoring and medical diagnosis as well as for determining of the physiological conditions of endurance athletes.

There is substantial variation in levels of extractable phenolic compounds of red grapes (Vitis vinifera L.). Therefore, it could be desirable to known the aforesaid parameter at least for each vine. Nowadays, interest has shifted toward the development of portable vis/NIR systems, innovation in optical system design and miniaturization for its friendly use directly in the field. Spectra of intact grapes and grapes skins were recorded at harvest time in two different vintages (2016 and 2017 respectively) using a portable micro NIR spectrophotometer (908–1676 nm). A number of chemometric approaches have been used for spectral interrogation and evaluation of the aforesaid device. Spectral data have been correlated with red grape skin extractable polyphenols (total phenolic, anthocyanins and flavanols) by modified partial least squares regression (MPLS) using a number of spectral pretreatments. Moreover, different statistics strategies have been performed to develop a qualitative analysis of the data (linear discriminant analysis, discriminant partial least square analyses and Pearson's similarity index). After an exhaustive analysis of the obtained results in two different seasons, it can be concluded that the use of the portable micro NIR device for the “in vineyard” screening of extractable polyphenols in red grape skins is hampered by a number of factors. Environmental and physiological conditions should be considered to evaluate and remove factors that hamper a good sorting the berries according to their extractable polyphenol contents.

This study describes, for the first time, the development of an automated sensitive flow fluorescent noncompetitive immunoassay based on kinetic-exclusion analysis (KinExA) for the quantitative determination of human plasma levels of monoclonal antibodies (mAbs) used for cancer immunotherapy. The assay was adapted on KinExA™ 3200 biosensor and optimized and validated for bevacizumab (BEV) and cetuximab (CET), as representative examples of the mAbs, using their specific antigens. These antigens were the human vascular endothelial growth factor (VEGF) and epidermal growth factor receptor (EGFR) for BEV and CET, respectively. The limits of detection were 1.28 and 52.64 ng mL−1 for BEV and CET, respectively. The accuracy of the assay was demonstrated with analytical recovery of analytes from spiked plasma at 96.2–104.3 and 96.8–105.3% for BEV and CET, respectively. The precision of the assay was satisfactory as shown by relative standard deviation (RSD) at 2.2–5.7 and 2.5–6.1% for assay of BEV and CET, respectively. The high sensitivity of the assay allowed the use of very small volumes (~ 1 µL) of plasma sample for analysis. Automated analysis by the proposed KinExA-based assay facilitates the processing of large numbers of mAbs-containing specimens in studies of pharmacokinetics (PK), pharmacodynamics (PD), and therapeutic drug monitoring (TDM) of therapeutic mAbs. The proposed assay can be used to overcome the problems encountered in the existing conventional immunoassays for mAbs.

This paper proposes a closed inline system for decomposition of wine, aiming at the determination of lead using electrothermal atomization atomic absorption spectrometry (ETAAS). The system is built using a 0.8 mm diameter PTFE tube, which is wrapped around an 8 W UV lamp. The sample in the presence of 70% hydrogen peroxide is circulated on an 8 W UV lamp at the flow rate of 1 mL min−1 for 45 min. Under these conditions, the carbon content varied from 10% to 2% for a red wine sample before and after digestion, respectively. The system has allowed the determination of lead in wine samples using the analytical line 283.306 nm in the presence of aluminum as the chemical modifier and pyrolysis and atomization temperatures of 800 and 1800 °C, respectively. Then, lead can be quantified employing the external calibration technique with limits of detection 0.27 and quantification 0.89 µg L−1, and characteristics mass of 18 pg. The precision expressed by relative standard deviation (RSD%) was 2.13%, calculated using six replicates of a digested solution of a wine sample with the lead content of 16.35 µg L−1. For evaluation of the accuracy method, two wine samples were analyzed simultaneously by the method proposed and also by inductively coupled plasma mass spectrometry (ICP-MS). A statistical test demonstrated no significant difference between the means obtained by these two techniques. Also, experiments involving addition/recovery tests confirmed the method's accuracy. The system was employed for digestion and determination of lead in four Brazilian wine samples. The lead content varied from 2.19 to 43.48 µg L−1.

This work describes the use of virtual standards as calibration samples in an innovative multivariate calibration approach for the on-line monitoring of alkyl-esters content during biodiesel production process using a miniature near infrared (NIR) spectrometer. For comparison purposes, a partial least squares (PLS) model was built using synthetic blends prepared in laboratory with different concentrations of oil, glycerol, biodiesel, and ethanol and resulted in a satisfactory predictive ability (root mean square error of prediction, RMSEP, of 1.51% w/w). When compared to conventional methods, calibration with synthetic blends has the advantage of simplifying the experimental procedure and reducing the need for reference analysis. Nevertheless, it still requires the preparation of a considerable number of blends in laboratory. To overcome this limitation, this study proposed an innovative approach where a PLS model was constructed based on virtual standards: representative calibration spectra were created by mathematically mixing spectra from pure components and performing an adjustment using the Piecewise Direct Standardization (PDS) method. This significantly reduced the need for calibration synthetic blends and led to similar results (RMSEP of 1.75% w/w), compared to the previous approach. This work also demonstrates the use of the constructed models to predict the concentration profiles of alkyl-esters during the batch transesterification process.

Diabetes mellitus is a global endemic with a rapidly increasing prevalence in both developing and developed countries. Recently, hemoglobin A1c has been recommended by the American Diabetes Associations as a possible substitute for fasting blood glucose for the diagnosis of diabetes, because it is an indicator of long-term glycemic control. Also, centrifugal microfluidic systems have good potential for use in the point of care testing systems. In this study, a centrifugal microfluidic disc was designed and manufactured to measure hemoglobin A1c in whole blood using an immunoturbidimetry based method. Also, a new passive valve, named septum valve, was presented to precisely control the entry and exit of reagents. This design comprises three inputs for injection of reagents and a blood sample, three septum valves, a two-part mixing chamber and a chamber to measure the absorbance of the sample based on the immunoturbidimetry method. Fourteen blood samples were tested using the manufactured disc, and the results were very congruous with the clinical data. By using the designed centrifugal microfluidic disc, the hemoglobin A1c in whole blood was measured in 8 min, with a standard deviation of ± 0.36% HbA1c.

This work presents a second-to-none method for Taxol isolation from the Endophytic fungus Cladosporium sphaerospermum (AUMC 6896) and the Entomopathogenic fungus Metarizium anisopliae (AUMC 5130). The extracts were analyzed by high performance liquid chromatography (HPLC) and Liquid chromatography coupled to tandem mass spectrometry (LC-MS/MS) using positive electrospray ionization (ESI) in the multiple reaction monitoring (MRM) mode. This is rapid, consistent, reproducible, accurate, and sensitive for quantifying Taxol across multiple samples. The yield of crude Taxol product obtained from Potato Dextrose broth (PDB) medium inoculated with Cladosporium sphaerospermu and Metarizium anisopliae was found to be 3.732, and 0.0023 μg L−1 respectively. The yield can be improved by adding ammonium acetate or salicylic acid to the culture broth. Addition of ammonium acetate (AA) (20 mg L−1) to culture media resulted in an increase of Taxol yield to 30.365 and 27.289 μg L−1 respectively. Production of Taxol was 29.844 and 67.254 μg L−1 for the two fungus species when ammonium acetate was substituted by 90 mg L−1 salicylic acid (SA). Adding both AA (20 mg L−1) and SA (90 mg L−1) to the culture media resulted in an increase of the Taxol yield to 4.054 and 116.373 μg L−1 respectively. Our proposed analytical method offers very fast (3 min) quantitation of Taxol in comparison with other published methods. These findings represent a new bioprospecting of the endophytic fungi that may serve as a potential material for the production of Taxol for anticancer treatment.

The aim of this work was to develop and validate a NIR method for the quantification of three active ingredients from powder blends. Calibration set formulations were selected based on a D-optimal experimental design with three factors (ibuprofen, paracetamol, caffeine) and five variation levels (80–90–100–110–120%). NIR spectra were recorded in transmittance mode using a rotating sample configuration. Prior to model development the effect of spectral pre-processing was assessed by evaluating its impact over the transfer of orthogonality from concentration space to spectral space. NIR method was validated on the full calibration range with external prediction sets, using the accuracy profile approach. Robustness testing results showed that the accuracy of predictions for the analyte found in lower concentrations (caffeine) was influenced by relative humidity, while paracetamol/ibuprofen predictions were robust to all factors. Redefinition of interfering factor variation level was beneficial to reduce the bias in caffeine content predictions. Also, alternative solutions are provided for ensuring robustness and successful routine use.

Leaf analysis is a useful way of diagnosing the nutritional status of the plants and therefore fast methods of analysis are demanded to aid in fertilization management decisions. In this work, a strategy based on the combined use of near-infrared spectroscopy (NIR) and portable energy dispersive X-Ray Fluorescence (EDXRF) is proposed as a suitable cheap and rapid alternative to traditional wet analytical methodologies. The approach has the major benefit of minimal sample preparation since leaves need to be only dried and ground. The ability of both techniques individually and applying two strategies of data fusion for the prediction of the most important plant nutrients, namely N, P, K, Ca, Mg, Mn, Zn, and B was tested. Predictive models were constructed using Partial Least Squares (PLS) to correlate the spectra with the nutrient contents. Models of unequal prediction performance in terms of the ratio of predictive deviation (RPD) were obtained for the different parameters when considering both techniques separately. Low-level data fusion, which consists of a concatenation of the raw data from both techniques, showed little improvement and even decreased the predictive ability for some elements. Better results were obtained with mid-level data fusion, that is, merging data after a feature extraction step performed by means of Principal components analysis (PCA). The results show that a fair quantitative prediction is possible for Ca, K and Mn with RPDs ≥ 2 for external validation, whereas models for N and P allowed a semiquantitative estimation. Mg and B models were less satisfactory and can be used only for distinguish between low and high levels, while Zn content cannot be predicted. Finally, the potential of the fusion of FT-NIR and EDXRF spectroscopic data for the fast screening of olive crop nutritional status has been tested. Deficiencies in important elements like N and K has been successfully detected.

Interference of the Schlieren effect on sea water analysis by spectrophotometry is caused by the flow of solutions of different ionic strengths through a flow cell. A flow injection assay protocol programmed in a flow-batch format removes this interference and allows the use of a calibration line, obtained in deionized water, for determination of analytes in sea water samples of different salinity. This Single Line Calibration (SLC) technique is validated on the most frequently performed nutrient assays. Automated determinations, performed at rates ranging from 20 to 60 samples/hr, covered seawater sample ranges from nM to mM with limits of detection: 12 nM for nitrite, 94 nM for nitrate, 47 nM for phosphate, and 240 nM for silicate. Reproducibility of the determinations was equal to or better than, 3% r.s.d. and day to day calibration was within 10%. The programmable FI, uses about 1/5th volume of reagents compared to continuous flow techniques.

A simple method for measuring water concentration from 1 to 10000 mg L−1 is described. The approach involves adding CaO powder to an oil sample and measuring the amount of Ca(OH)2 produced by the reaction of CaO with water. Collection of the powder occurs by passing a fixed volume of the oil through an infrared transparent membrane and the amount of water is determined from the intensity of the OH stretching mode of Ca(OH)2 at 3645 cm−1. The approach is demonstrated with transmission, vegetable, and extreme pressure oils. These oils represent classes of oils that are problematic for measurement by transmission infrared spectroscopy, using a fixed pathlength cell, as described in ASTM method E2412. Values for the water levels equivalent to those measured by Karl Fischer titration are obtained with a linearity R2 value of > 0.996 and %RSD of 6.7% over a wide detection range of 1 mg L−1 to 10000 mg L−1. No calibration is required, as the amount of water is determined using the extinction coefficient for the band at 3645 cm−1.

In this work, a method for total sulphur determination in automotive gasoline using dried matrix spot sampling is proposed. The method is based on the deposition of the sample on a cellulose-based filter paper and subsequent sulphur quantification via CS diatomic molecule using high-resolution continuum source graphite furnace molecular absorption spectrometry (HR-CS GF MAS). The sample deposition was carried out, along with the chemical modifier, on a 20-mm filter paper disc previously adapted into a polytetrafluoroethylene (PTFE) mould. The liquid phase was removed by heating the PTFE mould, and then the gasoline sample-embedded filter paper was punched in smaller discs (procedure A) or pulverised (procedure B) before the analyses. The mixture of Pd and Mg was used as chemical modifier to stabilise the sulphur compounds on the filter paper and on the graphite furnace. All the calibration curves constructed using seven different sulphur-containing compounds had a coefficient of determination higher than 0.995 and a linear range from 2 to 150 mg kg−1 S. By using the optimised conditions, the best characteristic mass, limits of detection and quantitation were 6 ng, 0.6 and 1.8 mg kg−1, respectively. The two sampling procedures (A and B) were evaluated for real samples, and procedure B was chosen since it markedly improved the precision. Using this procedure, satisfactory recovery values from 95 to 106% were obtained in the spike-recovery tests. In addition, the S concentrations for the certified reference materials were not statistically different from the certified values at 95% confidence level. Sulphur concentrations from 20 to 46 mg kg−1 were found in the six analysed gasoline samples, and these values were statistically assessed using a reference method (ASTM 5453). Spectral interference caused by MgF and MgCl diatomic molecules was observed and investigated.

Paper-based analytical devices (PADs) with four new designs could be fabricated using commercially available home-based scan-and-cut printer. They serve for miniaturised platforms for chemical analysis. Replication analysis of a sample together with the calibration (using the analyte standards at different concentrations) can be completed in a single run, by utilising smartphone as the detector. Some new approaches for choosing detection zones were suggested. The four proposed PAD designs here were used as models in microliter scale operation to demonstrate the well-known chemistries of colorimetric determinations of iron, phosphate, and hardness using 1,10-phenanthroline and simple aqueous guava leaf extract; molybdate, and EBT-EDTA complexometric titration, respectively, through calibrations: where Blue (B) value = 88.2log [Fe3+] - 80.8, R2 = 0.989; B value = 1.75 [Fe3+] + 0.198, R2 = 0.999; Grey scale (I) value = 1.77 [Fe3+] – 1.22, R2 = 0.997; Red (R) value = 16.1log [PO43−] + 8.95, R2 = 0.999; Hue (H) value = 43.3log [Ca2+] + 233, R2 = 0.994, respectively. For the hardness, using one of the PAD designs, true titration was also possible. Applications of the proposed devices and procedures were demonstrated for real world samples with validation. Additionally, kinetic study of the molybdenum blue for phosphate was demonstrated using one of the PADs.

Methods to determine the release of biocides (e.g. copper) and substances of concern (e.g. zinc) from antifouling paints are required for both the development of efficient products and their environmental risk assessment. To date, there are only two standardized methods available to estimate such release rates, but their reliability has been put into question. An alternative method, allowing determination of environmental release rates in the field of metallic or organometallic biocides by X-Ray Fluorescence (XRF), has been developed and applied in recent years. In this study, the potential for standardization of the XRF method is investigated through evaluation of its accuracy, precision and transferability between instruments. Accurate quantification of copper (Cu) and zinc (Zn) in μg cm−2, despite differences in chemical composition, was demonstrated through comparison of calibration regression slopes for ten different antifouling paints and confirmed through the measurement of validation samples. Universal antifouling paint calibration curves are proposed for the determination of Cu and Zn in thin paint films, with a prediction uncertainty of around ±130 μg/cm2 for both metals. The transferability of the method to another instrument was also demonstrated. For both analyzers, concentrations of validation samples were within 5% of those determined through wet chemical analysis. Pre-requisites and recommendations for the application of the method as well as its applicability to both short- and long-term release rate studies in the field are also presented and discussed.

Quantification of dissolved organic carbon (DOC) and iron (Fe) in surface waters is critical for understanding the water quality dynamics, brownification and carbon balance in the northern hemisphere. Especially in the remote areas, sampling and laboratory analysis of DOC and Fe content at a sufficient temporal frequency is difficult. Ultraviolet-visible (UV-Vis) spectrophotometry is a promising tool for water quality monitoring to increase the sampling frequency and applications in remote regions. The aim of this study was (1) to investigate the performance of an in-situ UV-Vis spectrophotometer for detecting spectral absorbances in comparison with a laboratory benchtop instrument; (2) to analyse the stability of DOC and Fe estimates from UV-Vis spectrophotometers among different rivers using multivariate methods; (3) to compare site-specific calibration of models to pooled models and investigate the extrapolation of DOC and Fe predictions from one catchment to another. This study indicates that absorbances that were measured by UV-Vis sensor explained 96% of the absorbance data from the laboratory benchtop instrument. Among the three tested multivariate methods, multiple stepwise regression (MSR) was the best model for both DOC and Fe predictions. Accurate and unbiased models for multiple watersheds for DOC were built successfully, and these models could be extrapolated from one watershed to another even without site-specific calibration for DOC. However, for Fe the combination of different datasets was not possible.

Developing effective ways for controlling the quality of natural products that are sold in high volume worldwide is currently one of the most urgent goals of analytical chemistry. Black pepper (Piper nigrum) is a major spice that constitutes 34% of the global spice market. Black pepper is highly prone to quality variations, arising from alteration of the piperine content. Miniaturized NIR spectroscopy offers a high-throughput, cost-effective and laboratory independent technique for quality control of black pepper. Handheld NIR spectrometers differ in the implemented technology, with impact on their analytical performance and unit cost. In this work, the performance of three miniaturized NIR instruments in quantification of piperine in whole and milled seeds of black pepper was investigated. With HPLC method used as the reference analysis, partial least squares regression (PLSR) models were constructed for NIR spectra while the prediction accuracy with respect to the spectrometer used was monitored through root mean square error of prediction (RMSEP) determined for an independent test set validation. Prediction of piperine content in milled seeds was achieved with a RMSEP of 0.18% using the benchtop spectrometer Buchi NIRFlex N-500; while handheld instruments offered, respectively, 0.22% (microPHAZIR), 0.26% (MicroNIR 2200) and 0.42% (SCiO). In contrast, for the analysis of the whole pepper seeds, only the Buchi NIRFlex N-500 (RMSEP: 0.30%) and the Viavi MicroNIR 2200 (RMSEP: 0.37%) gained reliable results. The performance of Thermo Scientific microPHAZIR was satisfactory (RMSEP: 0.75%). The consumer-aimed spectrometer SCiO (Consumer Physics) was found adequate for milled samples, while the analysis of the whole seeds was unsuccessful. To gain better understanding of the features of PLSR models of piperine content in black pepper, quantum mechanical simulation of NIR absorption bands of piperine was performed. This step enabled elucidating the chemical information relevant to the calibration model, and interpreting the difference in the analytical performance of the compared spectrometers with statistical parameters of the calibration models below acceptable levels.

In pesticide residues analysis, the drift is the difference between the concentration of two bracketing calibrations in the same batch. According to the SANTE/12682/2019 guideline a criterion of ±30% must be met or positive samples should be reanalyzed. This study aimed to investigate, for the first time, the efficiency of using analyte protectants (Aps) and the sandwich injection approach (SIA) to eliminate the drift between bracketing matrix matched-calibrations taking strawberry as an example. The strawberry samples were prepared according to the citrate-buffered QuEChERS (quick, easy, cheap, effective, rugged, and safe) procedure, followed by solvent exchange from acetonitrile to n-hexane:actone (9:1). Two batches were injected with the same sequence on GC-MS/MS, the only difference was that the first batch was without Aps and the second was with Aps. The sequence of the batch was as follows: blank solvent injection, 5 strawberry matched-calibrations at 0.05 μg/ml, separated by 20 blank strawberry injections after each strawberry matched-calibration injection. The drift was measured by considering the results of the first calibration as 100% and comparing the rest 4 injections with it. After 20 injections, out of the studied 219 pesticides, more than half of the pesticides fell out of criteria when analyte protectants were not used, and by the end of the samples batch 95% of the analytes were out of criteria. Only 8% of the studied analytes were out of criteria for the Aps batch after 20 injections. In the end of the 80 samples batch, 17% were out of criteria. Furthermore, at the end of the protected matrix-matched calibration batch, 90% of the pesticides had an RSD less than 15% in comparison with only 5% of the analytes for the non-protected batch. Moreover, the non-protected batch had an obvious negative drift in comparison with the protected batch. For example, the number of pesticides that had a lower result in the second matrix matched-calibration for the non-protected batch was more than twice the number in the protected batch (194 compared to 91 out of 219 pesticides for both experiments).

In the paper results of Laser-Induced Breakdown Spectroscopy (LIBS) measurements of Al and Si content in aqueous solutions are presented. Calibration curves exhibit good linearity and satisfying R2 coefficients of determination (over 0.99). The calibration procedures were next used for the determination of Al and Si concentration in biological material transferred into water solutions of unknown Al and Si content. Obtained data were verified by the AAS method (atomic absorption spectrometry). Measurement results showed good conformity. The LIBS spectra were also processed using Factorial Analysis (FA). It was found for Si samples that instead of a classic approach based on the selection of a particular spectral line for the calibration the FA makes possible the use of entire spectra to obtain quantitative information on the investigated material.

Three intelligent chemometric multi-way calibration methods including alternating trilinear decomposition (ATLD), alternating trilinear decomposition assisted multivariate curve resolution (ATLD-MCR) and multivariate curve resolution-alternating least squares (MCR-ALS) combined with high performance liquid chromatography-diode array detection (HPLC-DAD) were used to quantify ten molecular targeted anti-tumor drugs in three complex biological matrices (plasma, urine and cell culture media matrices). All analytes can be successfully eluted in 6.5 min. In this experiment, various degrees of time shifts occurred in different samples. While slight time shifts exist in the chromatographic analysis, satisfactory results can be obtained by the three proposed methods. When the time shift was large (5.6 s), the average spiked recoveries obtained by ATLD analysis were in the range of 58.9%–116.5%, which was less than satisfactory. However, the average recoveries obtained by MCR-ALS and ATLD-MCR analysis were 89.8%–114.8% and 84.5%–106.1% respectively, and more satisfactory results were obtained. For further research, ATLD-MCR and MCR-ALS methods were compared, and the results were evaluated by statistical tests. Accuracies of concentrations obtained by them were considered to be no significant difference. In addition, compared with other methods currently published, the proposed chemometric methods combined with the HPLC-DAD can rapidly, simultaneously and accurately determine varieties of molecular targeted anti-tumor drugs in different complex biological matrices even in the presence of severe peak overlaps, severe time shifts, slight baseline drifts and different unknown background interferences.

Hyperspectral imaging was used for large-scale monitoring of the residual moisture in wide textile webs at the end of the drying process that follows their washing or finishing by impregnation in aqueous solutions or dispersions. Such data are essential for optimizing the energy efficiency and the precise control of the drying process. Quantitative analysis of the recorded spectral data was carried out with multivariate regression methods such as the partial least squares (PLS) algorithm. Reference data for calibration of the prediction models were determined by gravimetry. The drying of textile materials from both natural or synthetic fibers possessing different water absorption capacities (cotton, polyamide, polyester), which were partially finished with an optical brightener, was investigated. Moisture contents in the range from 0 to about 12 wt% were considered in the calibration models. For all systems, the root mean square error of prediction (RMSEP) for the residual moisture was found to be about 0.5 wt%, that is, about 1 g/m2. In addition to the quantitative determination of the water content, hyperspectral imaging provides detailed information about its spatial distribution across the textile web, which may help to improve the control of the drying process. In particular, it was demonstrated that the developed methods were capable of detecting and visualizing inhomogeneous moisture distributions. Averaging of the individual values of the moisture content predicted from all spectra across the surface of the textile samples resulted in a very close correlation with the corresponding gravimetric reference values. Due to the averaging process, the difference between both values is generally lower than RMSEP even in case of samples with inhomogeneous distribution of the moisture. The high precision and the broad capabilities of the developed analytic methods for in-line monitoring of the moisture content hold the potential for an efficient process control in technical textile converting processes.

Robust quantitative analysis methods are very attractive but challenging with surface-enhanced Raman scattering (SERS) technique till now. Quantitative analysis methods using absolute Raman scattering intensities tend to desire very critical reproducibility of SERS substrates and consistency of testing conditions, as batch differences and inhomogeneity of SERS substrates as well as the fluctuation of measuring parameters placed challenging obstacles. Relative Raman scattering intensities, on the other hand, can release the adverse interferences mentioned above and provide effective and robust information as it is independent of the reproducibility of SERS substrates. By establishing external calibration working curves, we achieved accurate molecule composition prediction of molecules in multi-component systems. Further, by choosing or adding a label molecule with known concentration as Raman internal standards, the concentration of target molecules can be easily predicted. This approach proved the effectiveness and robustness of quantitative analysis with the relative Raman scattering intensities, even carried out with a flexible inhomogeneous SERS substrate.

A pH colorimetric sensor array (CSA) with fast response time (

Herein a semi-quantitative and quantitative method for rapid determination of water hardness was introduced. The method was based on color change of silver nanoparticles (AgNPs) in the presence of real water samples. Carbon dots were prepared from mulberry in a hydrothermal procedure and used as reductant of silver ion for synthesis of AgNPs. A classification method based on the color change of AgNPs in the presence of different water samples was also founded. The analysis based of the proposed method was cheap and rapid. On site semi-quantitative determination of total hardness of water can be performed by the proposed method. A linear calibration model based on the color analysis of the images of AgNPs in the presence of water samples was constructed. The model was applicable for determination of total hardness of water in the range of 116-248 mg L−1 of calcium carbonate. A variety of real water samples were included in the calibration model. The calibration method can be used to predict total hardness of water in a critical range above the soft water and below the very hard water. The results were compared by the standard titrimetric method based on ethylenediaminetetraacetic acid. Prediction of total hardness of real water samples based on the color model was in most cases below 20%.

Colistimethate sodium (CMS) is a widely administrated old-generation prodrug for the treatment of the life-threatening infections caused by multi-resistant Gram-negative bacteria. Until now, the quality control procedure of the CMS commercial products is based on microbiological assays. The aim of the study is the development of a chemical analysis methodology based on liquid chromatography - mass spectrometry (LC-MS) that could be used for the quality control of CMS products. The careful optimization of the LC and QToF-MS parameters was deemed crucial, as CMS is known to be a very complex mixture. Thus, a two stage Design of Experiments (DoE) pipeline has been followed, aiming towards the separation of the mixture components. According to the DoE results, a baseline-resolved chromatogram revealing more than 20 compounds was achieved. The separation was performed using a Waters Acquity BEH C8 column employing gradient elution. The mobile phase consisted of aq. ammonium formate 0.005 M (pH 6) (solvent A) and methanol/acetonitrile 79/21 (v/v) (solvent B). A second optimization experiment for the MS signal was employed in order to achieve maximum sensitivity. The singly charged signals were monitored for the validation in the positive ion mode. The calibration curve range was 50 – 110 μg mL-1, corresponding to the 80-120 % of the nominal CMS amount in the commercial products. Due to the complexity of the CMS chromatograms and the corresponding spectrum of each chromatographic peak, untargeted and targeted approaches were performed employing the MZmine software. Furthermore, apart from the classical univariate statistical analysis, partial least square regression (PLS-R) model was also employed, as the variables were more than the observations. The developed methodology has been employed to analyze several batches and inconsistences have been discovered.

A novel ferricyanide/Prussian blue (PB) assay for total antioxidant capacity (TAC) determination was developed exploiting the formation of PB nanoparticles in the presence of polyvinylpyrrolidone (PVP) as stabilizer. This improved method, named as "nanoparticle-based ferricyanide/Prussian blue assay (PBNP)", was applied to the TAC measurement of Cynara Scolymus L. (globe artichoke). The calibration results of the novel (PBNP) method were compared with those of a similar nanoparticle PB method performed in the absence of PVP, and of a sodium dodecyl sulfate-modified and acid-optimized ferricyanide reference assay. Compared to similar common Fe(III)-based TAC assays, much higher molar absorptivities, pointing out higher response to different kinds of antioxidants, were obtained with PBNP for all tested antioxidants, and lower LOD and LOQ values were achieved for thiols. As an additional advantage, methionine, not responding to other electron-transfer based TAC reagents, could be measured. PBNP could detect various antioxidants with one-two orders-of-magnitude lower LOD values than those of widely used TAC assays like CUPRAC and Folin-Ciocalteau well correlating with the proposed assay.

Absolute quantitation is always a challenge for in situ analysis of solid samples with ambient liquid extraction mass spectrometry due to unknown sampling efficiencies of analytes from complex solid matrices such as commercial packaging materials. Standards were usually dropped onto the sample surface for signal calibration, but the mass transfer of standards would not be the same as analytes distributed in samples. In this work, an in situ quantitation method via liquid microjunction sampling (LMJSS) coupled with mass spectrometry (MS) for photoinitiators (PIs) in packaging was developed without standard spiking. For direct in situ quantitation, mass transfer kinetic model for LMJSS of solid surface was proposed and validated. Results showed that the detection data well fitted the mass transfer model with adjusted R2 mostly in the range of 0.8–0.9 for 12 PIs in both lab-made mimetic positive samples and commercial packaging samples. According to the mass transfer kinetic model, two-point kinetic calibration method was proposed for calculation of the absolute concentration of PIs in solid samples by LMJSS of the same sample area for two times. The conditions of LMJSS including extraction solvent composition and solvent flowrate were optimized. With the optimized LMJSS-MS method and new calibration method, in situ analysis was done within only 2 min, and detection limits and quantitation limits of 15 PIs in mimetic samples were in the range of 0.06–1.5 mg/m2 and 0.34–6 mg/m2 respectively with relative standard deviation (RSDs) in the range of 2%–26%. The quantitation accuracy by the new calibration method for 13 PIs was in the range of 61%–162%. The method was applied to the quantitation of PIs in commercial packaging samples. 6 PIs were detected and identified in four commercial packaging materials, and the quantitation results were comparable with that by traditional solvent extraction-LC-MS method (relative recovery, 63%–127%).

Compounding of monoclonal antibody (mAbs) constantly increases in hospital. Quality control (QC) of the compounded mAbs based on quantification and identification is required to prevent potential errors and fast method is needed to manage outpatient chemotherapy administration. A simple and ultra-fast (less than 30 s) method using flow injection analysis associated to least square matching method issued from the analyzer software was performed and evaluated for the routine hospital QC of three compounded mAbs: bevacizumab, infliximab and rituximab. The method was evaluated through qualitative and quantitative parameters. Preliminary analysis of the UV absorption and second derivative spectra of the mAbs allowed us to adapt analytical conditions according to the therapeutic range of the mAbs. In terms of quantitative QC, linearity, accuracy and precision were assessed as specified in ICH guidelines. Very satisfactory recovery was achieved and the RSD (%) of the intermediate precision were less than 1.1%. Qualitative analytical parameters were also evaluated in terms of specificity, sensitivity and global precision through a matrix of confusion. Results showed to be concentration and mAbs dependant and excellent (100%) specificity and sensitivity were reached within specific concentration range. Finally, routine application on "real life" samples (n = 209) from different batch of the three mAbs complied with the specifications of the quality control i.e. excellent identification (100%) and ± 15% of targeting concentration belonging to the calibration range. The successful use of the combination of second derivative spectroscopy and partial least square matching method demonstrated the interest of FIA for the ultra-fast QC of mAbs after compounding using matching method.

Seed-growth synthesis is a common strategy to prepare silver nanoplates, whose peculiar plasmonic features can be exploited for surface enhanced Raman scattering (SERS) applications. Here we describe the fabrication and characterization of SERS chips using a peculiar in situ seed growth method, yielding a dense layer of nano-objects directly on a glass slide. In this way, geometric features (i.e. shape and dimensions) of the nano-objects can be tuned by controlling the growth time, obtaining a high concentration of hot spots on the surface. In particular, the SERS response of four kinds of chips were investigated to define the best SERS configuration in terms of size of the silver nano-objects, excitation wavelength and homogeneity of the SERS response. Silver nano-plates with a seeded growth time of 60 min demonstrated remarkable results both in terms of plasmonic enhancement, with an enhancement factor (EF) of 2 × 105 using a 532 nm laser excitation, and good homogeneity of the SERS response with intra- and inter-maps RSD of 10% and 5%, respectively. In order to demonstrate application of these chips for real sample analysis, an analytical procedure for the detection of a model pesticide, i.e. thiram fungicide, was developed and applied to its detection on green apples peels. SERS measurements on 60 min seeded growth silver nano-plates chip coupled with a multivariate PLS approach demonstrated high accuracy and repeatability for thiram detection in food matrix within the European law limits.

Herein we propose, for the first time, a rapid method based on flow injection analysis, electrospray ionization-tandem mass spectrometry (FIA-ESI-MS/MS) and multivariate calibration for the determination of l -leucine, l -isoleucine and L-allo-isoleucine in saliva. As far as we know, multivariate calibration has never been applied to the data from this non-separative approach. The possibilities of its use were explored and the results obtained were compared with the corresponding ones when using univariate calibration. Partial least square regression (PLS1) multivariate calibration models were built for each analyte by analyzing different saliva samples, and were subsequently applied to the analysis of another set of samples which had not been used in any calibration step. For Leu, the model worked satisfactorily with root mean square errors in the prediction step of 17%. This error can be considered acceptable and is common in methodologies that do not include a separation step. Results were compared with those obtained when univariate calibration was used, using the m/z transition 132.1 → 43.0 as the quantitation variable. In this case, the obtained results were not acceptable, with RMSEP of 236%, due to the fact that saliva samples contained another compound, different to the target analytes, which also shared the same transition. Ile and aIle have the same fragmentation patterns, so quantification of the sum of both compounds was performed, with RMSEP of 14% using a PLS1 model. Similar results were obtained when a univariate calibration model using the m/z transition 132.1 → 69.0 was employed. However, the use of this transition should be carefully examined when other compounds present in the matrix contribute to the analytical signal. The method increases sample throughput more than one order of magnitude compared to the corresponding LC-ESI-MS/MS method and is especially suitable as screening. When abnormally high or low concentrations of the analytes studied are obtained, the use of the method that includes separation is recommended to confirm the results.

The objective of this study was to develop and validate a novel analytical procedure for determination of total chromium and Cr(III) and Cr(VI) species released from metal implants into whole blood and joint effusion. Firstly, the ion-pair chromatographic method employing reversed-phase high-performance liquid chromatography inductively coupled plasma mass spectrometry (ICP-MS) for analysis of species was developed. Secondly, all samples, protein and low molecular fractions were analyzed for their total chromium content using ICP-MS. An electronic eye (EE) for fast and easy evaluation of grape phenolic ripening has been developed. For this purpose, berries of different grape varieties were collected at different harvest times from veraison to maturity, then an amount of the derived must was deposited on a white sheet of absorbent paper to obtain a sort of paper chromatography. Thus, RGB images of the must spots were collected using a flatbed scanner and converted into one-dimensional signals, named colourgrams, which codify the colour properties of the images. The dataset of colourgrams was used to build calibration models to relate the colour of the images with the phenolic composition of the samples – determined by reference analytical methods – and therefore to follow the ripening trend. Satisfactory calibration models were obtained for the prediction of the most important parameters related to phenolic ripening of grapes, such as colour index, tonality, total anthocyanins content, malvidin-3-O-glucoside and petunidin-3-O-glucoside.

The program WinMLR has been developed for the quantification of mixtures of several components by making a multilinear regression treatment of experimental data to a linear combination of standard signals. Data may be obtained with any multichannel detector having more measuring channels than components to be resolved. For a correct resolution, a linear relationship between the signal of every channel and the component concentration is required. The data must be located into ASCII files, one file for each standard or sample measurement, which contain the X and Y values in separated columns. The program allows three types of calibration methods: single standard, multiple standards and multiple standards addition. In the first case, the program requires to measure one standard per component. In the second case, the user can employ standards of both pure compounds and known mixtures under the sole constraint that the set of standards must include all components in different proportions. Similarly, the multiple standard addition method (more known as general standard addition method) requires the data obtained by adding known amounts of one or several mixture components to the sample so that all of them are included in the assays. In the multiple standards and multiple standards addition procedures, once the calibration has been done, the program allows the mixture to be resolved by weighted or unweighted regression.
