Objective To observe and detect the changes of interleukin-10（IL-10）and miR-513c-5p in peripheral blood of patients with unexplained recurrent spontaneous abortion（URSA），and to discuss their roles in the pathogenesis of URSA. Methods Thirty cases of normal pregnant women and 30 cases of URSA patients were included in this study. Six mL fasting peripheral venous blood was extracted. Serum IL-10 protein was detected by ELISA，and miR-513c-5p in peripheral blood mononuclear cells（PBMCS）was detected by qPCR. Target Scan 7. 1 software was used to predict the binding site between miR-513c-5p and IL-10；besides，luciferase reporter assay was used to confirm the binding between them. In addition，wild-type and mutated IL-10 mRNA 3´UTR plasmids containing binding sites were constructed，respectively. 293T cells were divided into the overexpressed group，inhibited group，and control group，which were transfected with miR-513c-5p mimics，inhibitor and negative control，respectively. Then，IL-10 mRNA in 293T cells of each group was detected by qPCR，and IL-10 protein in the supernatant of cell culture was detected by ELISA. Results Serum IL- 10 protein of URSA group was lower than that of the normal pregnancy group（P<0. 05），while the level of miR-513c-5p was higher than that of the normal pregnancy group（P<0. 05）. Target Scan 7. 1 and luciferase reporter assay showed that miR-513c-5p bound with IL-10 3´UTR via the complementary bases. The expression of IL-10 mRNA in the cells and the level of IL-10 protein in the cell supernatant were as follows：inhibited group>control group>overexpressed group（all P< 0. 05）. Conclusions In URSA patients，IL-10 protein level decreases and the expression of miR-513c-5p increases. MiR-513c-5p inhibits the expression of IL-10 by binding to its 3´UTR directly. Thereby，it breaks the maternal-fetal immunotolerance and leads to URSA；miR-513c-5p/IL-10 axis may become a novel therapeutic target for URSA clinically. [ABSTRACT FROM AUTHOR]
