Transcription factor GATA4 regulates cardiac and osteoblast differentiation. However, its role in tooth development is not clear. Therefore, we generated Wnt1-Cre;GATA4 fl/fl mice, with conditional inactivation of the GATA4 gene in the dental papilla mesenchymal cells. Phenotypic analysis showed short root deformity along with reduced expressions of odonto/osteogenic markers. Proliferation (but not apoptosis) of cells around the apical area of the root was attenuated. In vitro, we knocked down GATA4 expression in stem cells of dental apical papilla (SCAPs). Proliferation, migration and odonto/osteogenic differentiation of SCAPs were affected in the shGATA4 group. Overexpression of GATA4 in SCAPs increased mineralization. Based on our previous iTRAQ results, guanine nucleotide binding proteins 3 (GNAI3) is one of the distinct proteins after GATA4 deletion. G protein signaling is involved in bone development, remodeling, and disease. In this study, both GATA4 deletion in the mouse root and knock-down in human SCAPs decreased the expression of GNAI3. Dual-luciferase and ChIP assay confirmed the direct binding of GATA4 to the GNAI3 promoter, both in vitro and in vivo. GNAI3 knock-down significantly decreased the odonto/osteogenic differentiation ability of SCAPs. We thus establish the role of GATA4 as a novel regulator of root development and elucidate its downstream molecular events.

Using a novel magnetic nanocomposite as adsorbent, a convenient and effective magnetic solid-phase extraction (MSPE) procedure was established for selective separation and concentration of nine perfluorinated compounds (PFCs) in surface water sample. Then an ultra high-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) system was employed for detection of PFCs. Good linearity of the developed analytical method was in the range of 0.5–100 ng L−1 with R2 > 0.9917, and the limits of detection (LODs) ranged from 0.029 to 0.099 ng L−1. At three fortified concentrations of 0.5, 5 and 50 ng L−1, the spiked recoveries of PFCs were in the range of 90.05–106.67% with RSDs −1, with perfluorooctane sulfonate and perfluoroctanoic acid as the most prevalent PFCs, and the greatest concentrations of PFCs were observed in Niuchao lakelet. The concentrations of the PFCs (C ≥ 11) were mostly less than the limits of quantification (LOQs), attributed to the possibility that the more hydrophobic long-chain PFCs are potential to accumulate in sediment and aquatic biota.

Ginsenoside Rg1 (Rg1) is one of the major bioactive ingredients in Panax ginseng, and it attenuates inflammation and apoptosis. The aims of our study were to explore the potential of Rg1 for the treatment of intestinal I/R injury and to determine whether the protective effects of Rg1 were exerted through the Wnt/β-catenin signaling pathway. In this study, Rg1 treatment ameliorated inflammatory factors, ROS and apoptosis that were induced by intestinal I/R injury. Cell viability was increased and cell apoptosis was decreased with Rg1 pretreatment following hypoxia/reoxygenation (H/R) in the in vitro study. Rg1 activated the Wnt/β-catenin signaling pathway in both the in vivo and in vitro models, and in the in vitro study, the activation was blocked by DKK1. Our study provides evidence that pretreatment with Rg1 significantly reduces ROS and apoptosis induced by intestinal I/R injury via activation of the Wnt/β-catenin pathway. Taken together, our results suggest that Rg1 could exert its therapeutic effects on intestinal I/R injury through the Wnt/β-catenin signaling pathway and provide a novel treatment modality for intestinal I/R injury.

Nickel-based hydroxide hierarchical nanoarrays (NiyM(OH)x HNAs M = Fe or Zn) are doped with non-noble transition metals to create nanostructures and regulate their activities for the oxygen evolution reaction. Catalytic performance in these materials depends on their chemical composition and the presence of nanostructures. These novel hierarchical nanostructures contain small secondary nanosheets that are grown on the primary nanowire arrays, providing a higher surface area and more efficient mass transport for electrochemical reactions. The activities of the NiyM(OH)x HNAs for the oxygen evolution reaction (OER) followed the order of Ni2.2Fe(OH)x > Ni(OH)2 > Ni2.1Zn(OH)x, and these trends are supported by density functional theory (DFT) calculations. The Fe-doped nickel hydroxide hierarchical nanoarrays (Ni2.2Fe(OH)x HNAs), which had an appropriate elemental composition and hierarchical nanostructures, achieve the lowest onset overpotential of 234 mV and the smallest Tafel slope of 64.3 mV dec−1. The specific activity, which is normalized to the Brunauer–Emmett–Teller (BET) surface area of the catalyst, of the Ni2.2Fe(OH)x HNAs is 1.15 mA cm−2BET at an overpotential of 350 mV. This is ~4-times higher than that of Ni(OH)2. These values are also superior to those of a commercial IrOx electrocatalyst.

Growing evidence indicates that exposure to organochlorine pesticides (OCPs) could increase non-Hodgkin lymphoma (NHL) risk. However, results from epidemiological studies investigating this association remain controversial. We thus conducted a meta-analysis to quantitatively evaluate the association between OCP exposure and NHL risk. Relevant publications were searched in PubMed, Web of Science and Embase and identified according to the inclusion criteria. Thirteen studies (6 nested case-control, 1 case-cohort and 6 case-control) were selected for this meta-analysis. We used odds ratios (ORs) with 95% confidence intervals (CIs) to estimate the relationship between OCPs exposure and NHL risk. The summary OR for included studies was 1.40 (95% CI 1.27 to 1.56). No overall significant heterogeneity in the OR was observed (Ph = 0.253, I2 = 12.6%). Furthermore, OR estimates in subgroup analyses were discussed and strong associations were observed for dichlorodiphenyldichloroethylene (DDE, OR = 1.38, 95% CI 1.14 to 1.66), hexachlorocyclohexane (HCH, OR = 1.42, 95% CI 1.08 to 1.87), chlordane (OR = 1.93, 95% CI 1.51 to 2.48) and hexachlorobenzene (HCB, OR = 1.54, 95% CI 1.20 to 1.99). This meta-analysis had suggested that total OCPs of interest was significantly positively associated with NHL risk.

Anthrax is a highly lethal infectious disease caused by the bacterium Bacillus anthracis and the associated shock is closely related to the lethal toxin (LeTx) produced by the bacterium. The central role played by the 63 kDa protective antigen (PA63) region of LeTx in the pathophysiology of anthrax makes it an excellent therapeutic target. In the present study, a human/murine chimeric IgG mAb, hmPA6, was developed by inserting murine antibody variable regions into human constant regions using antibody engineering technology. hmPA6 expressed in 293F cells could neutralize LeTx both in vitro and in vivo. At a dose of 0.3 mg/kg, it could protect all tested rats from a lethal dose of LeTx. Even administration of 0.6 mg/kg hmPA6 48 h before LeTx challenge protected all tested rats. The results indicate that hmPA6 is a potential candidate for clinical application in anthrax treatment.

New iron selenide superconductors by intercalating smaller-sized alkali metals (Li, Na) and alkaline earths using high-temperature routes have been pursued ever since the discovery of superconductivity at about 30 K in KFe2Se2, but all have failed so far. Here we demonstrate that a series of superconductors with enhanced Tc=30~46 K can be obtained by intercalating metals, Li, Na, Ba, Sr, Ca, Yb, and Eu in between FeSe layers by the ammonothermal method at room temperature. Analysis on their powder X-ray diffraction patterns reveals that all the main phases can be indexed based on body-centered tetragonal lattices with a~3.755-3.831 {\AA} while c~15.99-20.54 {\AA}. Resistivities show the corresponding sharp transitions at 45 K and 39 K for NaFe2Se2 and Ba0.8Fe2Se2, respectively, confirming their bulk superconductivity. These findings provide a new starting point for studying the properties of these superconductors and an effective synthetic route for the exploration of new superconductors as well., Comment: 22 pages, 5 figures
