The emergence of the Middle Palaeolithic, and its variability over time and space are key questions in the field of prehistoric archaeology. Many sites have been documented in the south-eastern margins of the Massif central and the middle Rhône valley, a migration path that connects Northern Europe with the Mediterranean. Well-dated, long stratigraphic sequences are essential to understand Neanderthals dynamics and demise, and potential interactions with Homo sapiens in the area, such as the one displayed at the Maras rock shelter ("Abri du Maras"). The site is characterised by exceptional preservation of archaeological remains, including bones dated using radiocarbon (14C) and teeth using electron spin resonance combined with uranium series (ESR/U-series). Optically stimulated luminescence was used to date the sedimentary deposits. By combining the new ages with previous ones using Bayesian modelling, we are able to clarify the occupation time over a period spanning 200,000 years. Between ca. 250 and 40 ka, the site has been used as a long-term residence by Neanderthals, specifically during three interglacial periods: first during marine isotopic stage (MIS) 7, between 247 ± 34 and 223 ± 33 ka, and then recurrently during MIS 5 (between 127 ± 17 and 90 ± 9 ka) and MIS 3 (up to 39,280 cal BP). In the last few decades, the field of ancient DNA has taken a new direction towards using sedimentary ancient DNA (sedaDNA) for studying human and mammalian population dynamics as well as past ecosystems. However, the screening of numerous sediment samples from archaeological sites remains a time-consuming and costly endeavor, particularly when targeting hominin DNA. Here, we present a novel high-throughput method that facilitates the fast and efficient analysis of sediment samples by applying a pooled testing approach. This method combines multiple extracts, enabling early parallelization of laboratory procedures and effective aDNA screening. Pooled samples with detectable aDNA signals undergo detailed analysis, while empty pools are discarded. We have successfully applied our method to multiple sediment samples from Middle and Upper Paleolithic sites in Europe, Asia, and Africa. Notably, our results reveal that an aDNA signal remains discernible even when pooled with four negative samples. We also demonstrate that the DNA yield of double-stranded libraries increases significantly when reducing the extract input, potentially mitigating the effects of inhibition. By embracing this innovative approach, researchers can analyze large numbers of sediment samples for aDNA preservation, achieving significant cost reductions of up to 70% and reducing hands-on laboratory time to one-fifth. A stratigraphic sequence from Ghar-e Boof, a cave site in Iran, covering a period of c. 80,000–30,000 BP and containing more than 20,000 seed and chaff remains, allows a detailed study of the use of annual seed species of Palaeolithic hunter-gatherer groups and its evolution under the influence of changing environmental conditions. Taxonomic changes in the archaeobotanical assemblage and the stable carbon isotope data of pistachio support a considerable change in environmental conditions over the sequence from MIS 5a to MIS 3. The exceptional dominance of wild ancestors of modern crop species, including glume wheat and large-seeded legumes from Middle Palaeolithic layers AH VI (OSL ranges 72–81 ka BP), coincides broadly with the transition from MIS 5a to MIS 4. With the beginning of MIS 4 these taxa are strongly reduced, corresponding with a strong decrease in global CO2 concentrations and in the Δ13C values of Pistacia khinjuk/atlantica from the site. Wild glume wheat completely disappears after Middle Palaeolithic AH Vb and never reappears at the site. We hypothesize that the Middle Palaeolithic niche that allowed the harvesting and consumption of wild cereals and legumes ended with a destabilization of the vegetation in early MIS 4. During the Upper Paleolithic, lions become an important theme in Paleolithic art and are more frequent in anthropogenic faunal assemblages. However, the relationship between hominins and lions in earlier periods is poorly known and primarily interpreted as interspecies competition. Here we present new evidence for Neanderthal-cave lion interactions during the Middle Paleolithic. We report new evidence of hunting lesions on the 48,000 old cave lion skeleton found at Siegsdorf (Germany) that attest to the earliest direct instance of a large predator kill in human history. A comparative analysis of a partial puncture to a rib suggests that the fatal stab was delivered with a wooden thrusting spear. We also present the discovery of distal lion phalanges at least 190,000 old from Einhornhöhle (Germany), representing the earliest example of the use of cave lion skin by Neanderthals in Central Europe. Here we present stable carbon, nitrogen and sulfur isotope ratios of collagen extracted from Rangifer, Equus and Bison bone (n = 128) from different stratigraphic levels at the chronologically well-constrained Middle and Upper Palaeolithic site of Les Cottés, France. Samples were taken from five phases of site use (US08, US06, US04 [upper and lower], and US02; ~ 45.8–35.3 ka cal BP) to explore the dietary and spatial palaeoecology of these ungulate species during MIS 3, and the contemporary climate. Temporal trends in δ15N values of all species broadly align with other climatic indicators at the site and the lowest values in US04 correspond to the Heinrich 4 cooling event, reflecting changes in the composition of soil/plant nitrogen at this time. Rangifer collagen is 13C-enriched compared to the other species throughout, consistent with lichen consumption. However, this isotopic niche partitioning between Rangifer and Equus/Bison is most extensive during US04, indicating plasticity in reindeer feeding behaviour, and potentially overall increased lichen biomass during this cooler/more arid phase. Rangifer δ34S values are consistently lower than Equus and Bison, which could be indicative of their more extensive spatial ranges incorporating greater inland areas. The evolution of the hominin hand has been widely linked to the use and production of flaked stone tool technologies. After the earliest handheld flake tools emerged, shifts in hominin hand anatomy allowing for greater force during precision gripping and ease when manipulating objects in-hand are observed in the fossil record. Previous research has demonstrated how biometric traits, such as hand and digit lengths and precision grip strength, impact functional performance and ergonomic relationships when using flake and core technologies. These studies are consistent with the idea that evolutionary selective pressures would have favoured individuals better able to efficiently and effectively produce and use flaked stone tools. After the advent of composite technologies during the Middle Stone Age and Middle Palaeolithic, fossil evidence reveals differences in hand anatomy between populations, but there is minimal evidence for an increase in precision gripping capabilities. Furthermore, there is little research investigating the selective pressures, if any, impacting manual anatomy after the introduction of hafted composite stone technologies ('handles'). Here we investigated the possible influence of tool-user biometric variation on the functional performance of 420 hafted Clovis knife replicas. Our results suggest there to be no statistical relationships between biometric variables and cutting performance. Therefore, we argue that the advent of hafted stone technologies may have acted as a 'performance equaliser' within populations and removed (or reduced) selective pressures favouring forceful precision gripping capabilities, which in turn could have increased the relative importance of cultural evolutionary selective pressures in the determination of a stone tool's performance. Although Middle Paleolithic (MP) hominin diets consisted mainly of ungulates, increasing evidence demonstrates that hominins at least occasionally consumed tortoises, birds, leporids, fish, and carnivores. Until now, the MP zooarchaeological record in the Zagros Mountains has been almost exclusively restricted to ungulates. The narrow range of hominin prey may reflect socioeconomic decisions and/or environmental constraints, but could also result from a research bias favoring the study of large prey, since archaeologists have undertaken no systematic taphonomic analyses of small game or carnivores in the region. Here, we report on the first comprehensive taphonomic analysis of an MP faunal assemblage from Ghar-e Boof (∼ 81–45 kyr), a Late Pleistocene site in the southern Zagros of Iran. Anthropogenic bone surface modifications point to hominins as the main agent of accumulation. Hominins preyed primarily on ungulates, particularly wild goat. However, we also found evidence for MP hominin exploitation of carnivores and tortoises at the site. Although small game represents only a minor portion of the diet, our results suggest that the hunting behavior of MP hominins in the Zagros was more diverse than previously thought, similar to what we find elsewhere in Eurasia. Observation of high-resolution terrestrial palaeoecological series can decipher relationships between past climatic transitions, their effects on ecosystems and wildfire cyclicity. Here we present a new radiocarbon dated record from Lake Fimon (NE-Italy) covering the 60–27 ka interval. Palynological, charcoal fragments and sediment lithology analysis were carried out at centennial to sub-centennial resolutions. Identification of the best modern analogues for MIS 3 ecosystems further enabled to thoroughly reconstruct structural changes in the vegetation through time. This series also represents an "off-site" reference record for chronologically well-constrained Palaeolithic sites documenting Neanderthal and Homo sapiens occupations within the same region. Neanderthals lived in a mosaic of grasslands and woodlands, composed of a mixture of boreal and broad-leaved temperate trees analogous to those of the modern Central-Eastern Europe, the Southern Urals and central-southern Siberia. Dry and other grassland types expanded steadily from 44 to 43 ka and peaked between 42 and 39 ka, i.e., about the same time when Sapiens reached this region. This vegetation, which finds very few reliable modern analogues in the adopted Eurasian calibration set, led to the expansion of ecosystems able to sustain large herds of herbivores. During 39–27 ka, the landscape was covered by steppe, desert-steppe and open dry boreal forests similar to those of the modern Altai-Sayan region. Much of what is known about human behavior and subsistence strategies in the Levantine Middle Paleolithic comes from long sequences from caves and rock shelters. In this context, studies of stone tool function have traditionally focused on determining the use of Levallois points and triangular elements, either as projectiles or, more rarely, multipurpose knives. Little is known about such tool use and hafting in Middle Paleolithic open-air sites in the Levant through the systematic application of micro-wear analysis. Here we report the results of a low and high-power study performed on the lithic assemblage of the Late Middle Paleolithic open-air site of Nahal Mahanayeem Outlet (NMO, Israel). Most pointed items, including Levallois and non-Levallois points, were used as butchering knives, many of them while hafted; to a much lesser extent they were also used for hide, bone, and wood/plant processing activities. Blades and flakes were mostly handheld and used as butchering knives, with hide, bone, antler, and wood/plant-processing tasks being rare. Hafted artifacts include morphologies and activities for which hafting is not required, indicating that NMO inhabitants possessed varied hafting expertise. Wood/plant processing tools, some of which were hafted, attest that manufacture and maintenance tasks were planned well in advance of game procurement at the site. These results attest to early evidence of hafted butchering knives and hafted plant processing tools for a Late Middle Paleolithic open-air site in the Levant, and support previous interpretations of NMO as a short-term task-specific location focused on animal processing activities, mostly butchery. Whether ethnoarcheological models of hunter-gatherer mobility, landscape use, and structuration of the inhabited space are relevant to the archeology of Neandertals and the Middle Paleolithic remains controversial. The thin lenses of hearth-associated stone tools and faunal remains excavated in sub-complex AS5 of Cueva Antón (Murcia, Spain) significantly advance these debates. Dated to 77.8–85.1 ka, these living floors are interstratified in river-accumulated sands and were buried shortly after abandonment by low-energy inundation events, with minimal disturbance and negligible palimpsest formation. Stone tools were made and ergonomically modified to fit tasks; their spatial distributions and use-wear reveal hearth-focused activities and a division of the inhabited space into resting and working areas. Site function varied with season of the year: units III-i/j1 and III-i/j2-3 record winter visits focused on filleting and hide processing, while woodworking predominated in unit III-b/d, which subsumes visits to the site over the course of at least one winter, one spring, and one summer. These snapshots of Neandertal behavior match expectations derived from the ethnographic and Upper Paleolithic records for the lifeways of hunter-gatherers inhabiting temperate regions with a markedly seasonal climate. The authors discuss functional characterization of Mousterian tools on the basis of their use-wear and residue analysis of five lithic tools from Mezmaiskaya cave and Saradj-Chuko grotto in the North Caucasus. The results represent the first comprehensive use-wear and residue analysis carried out on Mousterian stone artefacts in the Caucasus. During the last century, Grotta Romanelli (Southern Italy) has been a reference site for the European Late Pleistocene stratigraphy, due to its geomorphological setting and archaeological and palaeontological content. The beginning of the sedimentation inside the cave was attributed to the Last Interglacial (MISs 5e) and the oldest unearthed evidence of human occupation, including remains of hearths, was therefore referred to the Middle Palaeolithic. Recent surveys and excavations produced new U/Th dates, palaeoenvironmental interpretation and a litho-, morpho- and chrono-stratigraphical reassessment, placing the oldest human frequentation of the cave between MIS 9 and MIS 7, therefore embracing Glacial and Interglacial cycles. These new data provide evidence that the sea reached the cave during the Middle Pleistocene and human occupation occurred long before MISs 5e and persisted beyond the Pleistocene- Holocene boundary. The Mezmaiskaya cave is located on the North Caucasus near the border that divides Europe and Asia. Previously, fossil remains for two Neanderthals were reported from Mezmaiskaya Cave. A tooth from the third archaic hominin specimen (Mezmaiskaya 3) was retrieved from layer 3 in Mezmaiskaya Cave. We performed genome sequencing of Mezmaiskaya 3. Analysis of partial nuclear genome sequence revealed that it belongs to a Homo sapiens neanderthalensis female. Based on a high-coverage mitochondrial genome sequence, we demonstrated that the relationships of Mezmaiskaya 3 to Mezmaiskaya 1 and Stajnia S5000 individuals were closer than those to other Neanderthals. Our data demonstrate the close genetic connections between the early Middle Palaeolithic Neanderthals that were replaced by genetically distant later group in the same geographic areas. Based on mitochondrial DNA (mtDNA) data, we suggest that Mezmaiskaya 3 was the latest Neanderthal individual from the early Neanderthal's branches. We proposed a hierarchical nomenclature for the mtDNA haplogroups of Neanderthals. In addition, we retrieved ancestral mtDNA mutations in presumably functional sites fixed in the Neanderthal clades, and also provided the first data showing mtDNA heteroplasmy in Neanderthal specimen. Variations in the cross-sectional properties of long bones are used to reconstruct the activity of human groups and differences in their respective habitual behaviors. Knowledge of what factors influence bone structure in Homo sapiens and Neandertals is still insufficient thus, this study investigated which biological and environmental variables influence variations in the femoral robusticity indicator of these two species. The sample consisted of 13 adult Neandertals from the Middle Paleolithic and 1959 adult individuals of H. sapiens ranging chronologically from the Upper Paleolithic to recent times. The femoral biomechanical properties were derived from the European data set, the subject literature, and new CT scans. The material was tested using a Mantel test and statistical models. In the models, the polar moment of area (J) was the dependent variable; sex, age, chronological period, type of lifestyle, percentage of the cortical area (%CA), the ratio of second moment areas of inertia about the X and Y axes (Ix/Iy), and maximum slope of the terrain were independent covariates. The Mantel tests revealed spatial autocorrelation of the femoral index in H. sapiens but not in Neandertals. A generalized additive mixed model showed that sex, %CA, Ix/Iy, chronological period, and terrain significantly influenced variation in the robusticity indicator of H. sapiens femora. A linear mixed model revealed that none of the analyzed variables correlated with the femoral robusticity indicator of Neandertals. We did not confirm that the gradual decline in the femoral robusticity indicator of H. sapiens from the Middle Paleolithic to recent times is related to the type of lifestyle; however, it may be associated with lower levels of mechanical loading during adolescence. The lack of correlation between the analysed variables and the indicator of femoral robusticity in Neandertals may suggest that they needed a different level of mechanical stimulus to produce a morphological response in the long bone than H. sapiens. Marine Isotopic Stage 5 is associated with wetter climatic conditions in the Saharo-Arabian deserts. This stage also corresponds to the establishment of Middle Paleolithic hominins and their associated material culture in two geographical provinces in southwest Asia—the Eastern Mediterranean woodland and the Arabian Peninsula desert. The lithic industry of the Eastern Mediterranean is characterized by the centripetal Levallois method, whereas the Nubian Levallois method characterizes the populations of the Arabian desert. The Negev Desert, situated between these regions is a key area to comprehend population movement in correlation to climatic zones. This investigation addresses the nature of the Middle Paleolithic settlement in the Negev Desert during MIS 5 by studying the site of Nahal Aqev. High resolution chronological results based on luminescence dating and cryptotephra show the site was occupied from MIS 5e to MIS 5d. The lithic industries at Nahal Aqev are dominated by centripetal Levallois core method. These data demonstrate that Nahal Aqev is much closer in its cultural attributes to the Eastern Mediterranean Middle Paleolithic than to the Arabian Desert entity. We conclude that Nahal Aqev represents an expansion of Middle Paleolithic groups from the Mediterranean woodland into the desert, triggered by better climatic conditions. In the context of the late Levantine Middle Palaeolithic record, which is characterised by a wide range of Levallois strategies[29]-[31], a more detailed presentation of the data and specific attributes is required to assess the presence of Nubian technology as distinct from other Levallois methods at Shukbah. In the absence of direct refits, Usik et al. do not consider Nubian points to be distinguishable from the end-products of centripetal Levallois reduction; they observe only that "the Nubian Levallois method is based on the production of elongated Levallois points or pointed flakes"[20]. No convincing data are presented to justify the identification of Nubian Levallois artefacts Nubian Levallois technology is regarded as a distinct method of Levallois reduction; Levallois cores with distal and lateral preparation are not necessarily Nubian cores and can equally be attributed to the Levallois bidirectional and centripetal modes of core preparation[17]-[19]. Given that the analysed artefacts are not shown to fulfil the criteria for Nubian Levallois technology and instead represent other Levallois methods, the conclusion that "Nubian Levallois reduction strategies form part of a wider Levallois point production strategies, rather than a discrete technological approach" [ref.[1] SI7: 20] is contained in the methodological premises and, thus, inevitable. As a result, the use of Nubian Levallois technology by other populations that employ Levallois reduction methods and focus on the production of points is, perhaps, unsurprising. Increasing sample sizes within and between sites will undoubtedly enhance precision of comparative studies, yet our results provide the means to test key hypotheses - whether Nubian Levallois is discrete from other Levallois technologies. Conclusion Amongst the plurality of definitions of Nubian Levallois technology in the literature, the basis upon which Usik and colleagues[11] determined the criteria they set out is opaque, prohibiting clear differentiation from other Levallois methods. The use of birch tar can be traced back to the European Middle Palaeolithic and is relevant for our understanding of the technical skills and cognitive abilities of Neanderthals. Due to the lack of archaeological evidence, it remains unknown what techniques were used for birch tar making. Efficiency was recently used as a proxy to determine the method most likely used in the Middle Palaeolithic. Todtenhaupt et al. have proposed a technique employing a groove-like structure that is comparable with the recently presented condensation method. The groove method resulted in higher tar yields compared to other experimental aceramic production processes. However, the implications for Palaeolithic tar making remain unclear because some of the materials used in the experiment were not available then (polished granite slabs). To approach this problem, we replicated the groove with river cobbles and, in a second experiment with flint fragments, to evaluate whether similar results can be obtained. We were successful in producing birch tar in multiple runs with the cobble- and flint-grooves, which, in addition, proved to be more efficient than the condensation method in terms of tar yield per bark input. South Asia hosts the world's youngest Acheulean sites, with dated records typically restricted to sub-humid landscapes. The Thar Desert marks a major adaptive boundary between monsoonal Asia to the east and the Saharo-Arabian desert belt to the west, making it a key threshold to examine patterns of hominin ecological adaptation and its impacts on patterns of behaviour, demography and dispersal. Here, we investigate Palaeolithic occupations at the western margin of the South Asian monsoon at Singi Talav, undertaking new chronometric, sedimentological and palaeoecological studies of Acheulean and Middle Palaeolithic occupation horizons. We constrain occupations of the site between 248 and 65 thousand years ago. This presents the first direct palaeoecological evidence for landscapes occupied by South Asian Acheulean-producing populations, most notably in the main occupation horizon dating to 177 thousand years ago. Middle Paleolithic Neanderthal populations occupied Eurasia for at least 250,000 years prior to the arrival of anatomically modern humans. While a considerable body of archaeological research has focused on Neanderthal material culture and subsistence strategies, little attention has been paid to the relationship between regionally specific cultural trajectories and their associated existing fundamental ecological niches, nor to how the latter varied across periods of climatic variability. We examine the Middle Paleolithic archaeological record of a naturally constrained region of Western Europe between 82,000 and 60,000 years ago using ecological niche modeling methods. Evaluations of ecological niche estimations, in both geographic and environmental dimensions, indicate that 70,000 years ago the range of suitable habitats exploited by these Neanderthal populations contracted and shifted. These ecological niche dynamics are the result of groups continuing to occupy habitual territories that were characterized by new environmental conditions during Marine Isotope Stage 4. The Level VII of Amalda I cave (Gipuzkoa, Spain) represents one of the latest Middle Palaeolithic occupations in the Cantabrian Region. It is characterized by the presence of Middle Palaeolithic lithic industry and animal remains, with clear evidences of anthropic and carnivore manipulation. At this site, the Neanderthal presence has been questioned in relation to the role of carnivores in the accumulation of large, medium-sized and small mammals. It has also been proposed that the Neanderthal occupation could have consisted of short-term occupations, where different activities took place in a structured space within the cave. However, all hypotheses lacked any integrative analysis of the site formation processes. With the aim of understanding these processes, a combination of spatial techniques, based on GIS and inferential statistics (density analysis, hotspots tools and palaeotopographic reconstruction), along with the taphonomic study of identifiable and non-identifiable macromammals remains, were employed. This study has revealed distinct use of the cave space by Neanderthals and carnivores. The major concentrations of lithics and medium-size mammal remains were clearly accumulated by humans at the cave entrance, while the small-size mammals were gathered by carnivores in an inner zone. There is a relatively low amount of Middle Paleolithic sites in Europe dating to MIS 4. Of the few that exist, several of them lack evidence for anthropogenic fire, raising the question of how this period of global cooling may have affected the Neanderthal population. The Iberian Peninsula is a key area to explore this issue, as it has been considered as a glacial refugium during critical periods of the Neanderthal timeline and might therefore yield archaeological contexts in which we can explore possible changes in the behaviour and settlement patterns of Neanderthal groups during MIS 4. Here we report recent data from Abric del Pastor, a small rock shelter in Alcoy (Alicante, Spain) with a stratified deposit containing Middle Palaeolithic remains. We present absolute dates that frame the sequence within MIS 4 and multi-proxy geoarchaeological evidence of in situ anthropogenic fire, including microscopic evidence of in situ combustion residues and thermally altered sediment. We also present archaeostratigraphic evidence of recurrent, functionally diverse, brief human occupation of the rock shelter.
