Abstract Alkaline phosphatase (ALP) is a reliable biomarker for various diseases, making the development of new detection methods highly significant. Herein, we report a simple and accurate dual-signal optical detection strategy for measuring ALP activity across the entire clinical range in adults. Based on this, we designed a smartphone app to eliminate the limitations of large instruments, enabling real-time field detection. In this work, ALP catalyzes the conversion of the phosphate monoester of 2-phospho-L-ascorbic acid (PAA) into ascorbic acid (AA), a compound with strong reducing properties. AA facilitates the in-situ reduction of Ag+ to form Au@Ag core-shell nanoparticles, accompanied by significant changes in the ultraviolet-visible absorption spectra and color. These changes allow for the qualitative and quantitative detection of ALP activity. In UV absorption spectroscopy and colorimetric detection, the signal shows good linearity with ALP activity concentrations, with detection ranges of 0 U/L to 200 U/L (R2=0.9961) and 25 U/L to 250 U/L (R2=0.9771), respectively. Subsequent selective analysis and serum sample analysis demonstrated the high selectivity and accuracy of the proposed method, highlighting its potential for field application.

Abstract Agile and adaptive maneuvers such as fall recovery, high-speed turning, and sprinting in the wild are challenging for legged systems. We propose a Curricular Hindsight Reinforcement Learning (CHRL) that learns an end-to-end tracking controller that achieves powerful agility and adaptation for the legged robot. The two key components are (i) a novel automatic curriculum strategy on task difficulty and (ii) a Hindsight Experience Replay strategy adapted to legged locomotion tasks. We demonstrated successful agile and adaptive locomotion on a real quadruped robot that performed fall recovery autonomously, coherent trotting, sustained outdoor running speeds up to 3.45 m/s, and a maximum yaw rate of 3.2 rad/s. This system produces adaptive behaviors responding to changing situations and unexpected disturbances on natural terrains like grass and dirt.

Abstract Malignant ovarian sex cord-stromal tumors are rare neoplasms that account for approximately 5–7% of all ovarian malignancies, and they are primarily treated with surgery. The prognosis of patients with different surgical extents remains controversial. Therefore, the effects of different surgical extents on the prognosis of patients were explored in this retrospective cohort study. Patients with malignant ovarian sex cord-stromal tumors who underwent surgical treatment from January 2000 to December 2019 were selected. Abstract Non-small cell lung cancer (NSCLC) constitutes a significant portion of lung cancers and cytotoxic drugs (e.g. cisplatin) are currently the first-line treatment. However, NSCLC has developed resistance to this drug, which limits the therapeutic effect and thus affects prognosis. NSCLC sc-RNA-seq data were downloaded from the GEO database and Ku Leuven Laboratory for Functional Epigenetics, and bulk RNA-seq data were obtained from the TCGA database. The “Seurat” package was employed for scRNA-seq data processing, and the uniform manifold approximation and projection (UMAP) were applied for downscaling and cluster identification. Use the FindAllMarkers function to find differential genes (DEGs) for tumor stem cells. Then, we performed univariate regression analyses on the DEGs to identify potential prognostic genes. We created a machine learning framework based on potential prognostic genes, which combines 10 machine learning methods and their 101 combinations to get the optimal prognostic risk model. The model was evaluated in the training set and validation set. A nomogram was developed to provide physicians with a quantitative tool for prognosis prediction. Finally, we evaluated the expression and functionality of SLC2A1. We discovered 22 cell clusters containing 218379 cells by examining single-cell RNA sequencing datasets (GSE148071, KU_lom, GSE131907, GSE136246, GSE127465). Tumour cells were isolated for subpopulation analysis and 162 differential genes from SOX2_cancer were obtained. After univariate Cox analysis, we found 23 genes with prognostic potential prognostic value and utilized them to develop 101‑combination machine learning computational framework. We eventually picked the best performing ‘StepCox[both] + RSF’, which includes 8 genes. The model has a relatively high prediction accuracy in both TCGA and GEO datasets. In in vitro investigations, targeted suppression of the SLC2A1 gene resulted in significant reductions in proliferation, invasion and migration in A549 cells. In addition, a significant reduction in cisplatin resistance was seen in A549/DDP cells. The outcomes demonstrated the precision and credibility of the prognostic model for NSCLC, highlighting its potential significance in the treatment and prognosis of individuals affected by this disease. SLC2A1 may become a promising prognostic marker and a potential therapeutic target, offering valuable insights to inform clinical treatment decisions.

Abstract This work introduces and discusses the impacts of the water bridge on gas adsorption and diffusion behaviors in a shale gas-bearing formation. The density distribution of the water bridge has been analyzed in micropores and meso-slit by molecular dynamics. Na+ and Cl− have been introduced into the system to mimic a practical encroachment environment and compared with pure water to probe the deviation in water bridge distribution. Additionally, practical subsurface scenarios, including pressure and temperature, are examined to reveal the effects on gas adsorption and diffusion properties, determining the shale gas transportation in realistic shale formation. The outcomes suggest carbon dioxide (CO2) usually has higher adsorption than methane (CH4) with a water bridge. Increasing temperature hinders gas adsorption, density distribution decreases in all directions. Increasing pressure facilitates gas adsorption, particularly as a bulk phase in the meso-slit, whereas it restricts gas diffusion by enhancing the interaction strength between gas and shale. Furthermore, ions make the water bridge distributes more unity and shifts to the slit center, impeding gas adsorption onto shale while encouraging gas diffusion. This study provides updated guidelines for gas adsorption and transportation characteristics and supports the fundamental understanding of industrial shale gas exploration and transportation.

Abstract Peanut is one of the important oil and economic crops, among which the variegated testa peanut is a unique member. The molecular mechanisms underlying the pigment synthesis in variegated testa are still unclear. Differentially expressed genes (DEGs) in the flavonoid metabolism pathway in pigmented areas indicated that there were 27 DEGs highly related to the synthesis of variegated testa color among 1,050 DEGs. Of these 27, 13 were up-regulated and 14 were down-regulated, including 3 PALs, 1 C4H, 2 CHSs, 1 F3H, 1 F3'H, 2 DFRs, 2 LARs, 2 IAAs, 4 bHLHs, and 9 MYBs. GO (Gene Ontology) analysis indicated that DEGs were similarly enriched in three branches. KEGG (Kyoto Encyclopedia of Genes and Genomes) analysis suggested flavonoid biosynthesis is the most direct metabolic pathway for the synthesis of testa variegation. The liquid chromatography–tandem mass spectrometry (LC–MS/MS) results showed that cyanidin and delphinidin were the primary metabolites that caused the color differences between the pigmented and the non-pigmented areas. Through the verification of 20 DEGs via qPCR, the results were consistent with transcriptome sequencing in four comparison groups. The results in this study lay the foundation for revealing the molecular regulation mechanisms of flavonoid synthesis in variegated testa peanut.

Abstract Detailed crustal structure of large earthquake source regions is of great significance for understanding the earthquake generation mechanism. Numerous large earthquakes have occurred in the NE Tibetan Plateau, including the 1920 Haiyuan M8.5 and 1927 Gulang M8 earthquakes. In this paper, we obtained a high-resolution three-dimensional crustal velocity model around the source regions of these two large earthquakes using an improved double-difference seismic tomography method. High-velocity anomalies encompassing the seismogenic faults are observed to extend to depths of 15 km, suggesting the asperity (high-velocity area) plays an important role in the preparation process of large earthquakes. Asperities are strong in mechanical strength and could accumulate tectonic stress more easily in long frictional locking periods, large earthquakes are therefore prone to generate in these areas. If the close relationship between the aperity and high-velocity bodies is valid for most of the large earthquakes, it can be used to predict potential large earthquakes and estimate the seismogenic capability of faults in light of structure studies.

Abstract Association between the MGP gene rs1800801, rs1800802, rs4236 polymorphisms and vascular calcification and atherosclerotic disease was inconsistent. To clarify precise association, we performed this meta-analysis. Medline, Embase and China Knowledge Resource Integrated Database were systematically searched through December 2016. A total of 23 case-control studies, consisting of 5280 cases and 5773 controls, were included. The overall results suggested that the -7A polymorphism was associated with an increased risk for vascular calcification and atherosclerotic disease in the recessive model (OR = 1.50, 95% CI 1.01–2.24, P = 0.045). Subgroup analyses of Caucasians showed significant associations in the allelic model, recessive model, and homozygote model: allelic model (OR = 1.19, 95% CI 1.06–1.34, P = 0.004), recessive model (OR = 1.60, 95% CI 1.26–2.03, P

Abstract The blood-spinal cord barrier (BSCB) plays significance roles in recovery following spinal cord injury (SCI), and diabetes mellitus (DM) impairs endothelial cell function and integrity of BSCS. Endoplasmic reticulum (ER) stress occurs in the early stages of SCI and affects prognosis and cell survival. However, the relationship between ER stress and the integrity of BSCB in diabetic rats after SCI remains unclear. Here we observed that diabetic rats showed increased extravasation of Evans Blue (EB) dye, and loss of endothelial cells and pericytes 1 day after SCI compared to non-diabetic rats. Diabetes was also shown to induce activation of ER stress. Similar effects were observed in human brain microvascular endothelial cells. 4-phenylbutyric acid (4-PBA), an ER stress inhibitor lowered the adverse effect of diabetes on SCI, reduced EB dye extravasation, and limited the loss of endothelial cells and pericytes. Moreover, 4-PBA treatment partially reversed the degradation of tight junction and adherens junction both in vivo and in vitro. In conclusion, diabetes exacerbates the disruption of BSCB after SCI via inducing ER stress, and inhibition of ER stress by 4-PBA may play a beneficial role on the integrity of BSCB in diabetic SCI rats, leading to improved prognosis.

Abstract Motion sickness (MS) is a common physiological response that often occurs when individuals are exposed to environments with repeated acceleration stimuli. MS results from a mismatch between the vestibular system and visual and proprioceptive inputs. As a crucial organ for sensing acceleration stimuli, the vestibular system is closely related to the onset of MS. However, the complex pathogenesis of MS has led to its diagnosis primarily relying on subjective questionnaires, with a lack of objective indicators for evaluation. To identify objective indicators for evaluating MS, we conducted rotating chair stop experiments based on the principle of vestibulo-ocular reflex with 65 volunteers, obtaining their nystagmus slow-phase velocity and related time constants. Additionally, we conducted detailed MS questionnaires with these volunteers to assess their MS susceptibility. Through correlation analysis, we explored whether the nystagmus slow-phase velocity and related time constants significantly correlated with the MS questionnaire scores. The results showed significant positive correlations between the maximal nystagmus slow-phase velocity, cupula time constant, velocity storage time constant, and the nystagmus duration with the MS questionnaire scores in the 65 volunteers. These results indicated that these nystagmus parameters could serve as objective indicators for assessing MS susceptibility. Using K-Means clustering analysis, we classified MS susceptibility into categories I, II, III, and IV, and conducted K-Means clustering analysis on the corresponding nystagmus slow-phase velocity, cupula time constant, velocity storage time constant, and nystagmus duration. The magnitude and range of these indicators at different levels was quantified, offering objective and quantitative indicators for the clinical diagnosis of MS.

Abstract In this study, We aim to explore the association between the neutrophil to lymphocyte ratio (NLR), platelet to lymphocyte ratio (PLR), systemic immune-inflammatory index (SII), lymphocyte to monocyte ratio (LMR) and prognostic nutritional index (PNI) and distant metastasis of gastric cancer and develop an efficient nomogram for screening patients with distant metastasis. A total of 1281 inpatients with gastric cancer were enrolled and divided into the training and validation set.Univariate, Lasso regression and Multivariate Logistic Regression Analysis was used to identify the risk factors of distant metastasis. The independent predictive factors were then enrolled in the nomogram model. The nomogram’s predictive perform and clinical practicality was evaluated by receiver operating characteristics (ROC) curves, calibration curves and decision curve analysis. Multivariate Logistic Regression Analysis identified d-dimer, CA199, CA125, NLR and PNI as independent predictive factors. The area under the curve of our nomogram based on these factors was 0.838 in the training cohort and 0.811 in the validation cohort. The calibration plots and decision curves demonstrated the nomogram’s good predictive performance and clinical practicality in both training and validation cohort. Therefore,our nomogram could be an important tool for clinicians in screening gastric cancer patients with distant metastasis.

Abstract To identify different patterns of neurogenic lower urinary tract dysfunction management among Chinese community-dwelling individuals with spinal cord injury and explore the factors associated with latent classes. This was a cross-sectional study conducted in communities throughout China Mainland. Participants were recruited through the China Association of Persons with Physical Disability and a total of 2582 participants was included in the analysis. The data were collected by a questionnaire consisting of socio-demographic factors, disease-related factors, and a list of 8 bladder management methods. Latent class analysis was used to identify different latent classes of neurogenic lower urinary tract dysfunction management. Then the multinomial logistic regression was applied to analyze the relationship between neurogenic lower urinary tract dysfunction management patterns and socio-demographic and disease-related factors. Neurogenic lower urinary tract dysfunction management pattern among community-dwelling individuals with spinal cord injury was divided into four latent classes: “urinal collecting apparatus dominated pattern” (40.3%), “bladder compression dominated pattern” (30.7%), “intermittent catheterization dominated pattern” (19.3%) and “urethral indwelling catheterization dominated pattern” (9.6%). Multinomial logistic regression analysis found that the employment status, residential region, nursing need, payment method for catheterization products, hand function, time since spinal cord injury, urinary incontinence and concerns about social interaction affected by urination problems were significantly associated with latent classes. Only 19.3% of people used the intermittent catheterization as their main neurogenic lower urinary tract dysfunction management method. More attention needs to be paid to the promotion of the standardization process of intermittent catheterization in community-dwelling individuals with spinal cord injury. The associated factors of the four classes can be used for tailored and targeted interventions to increase the use of intermittent catheterization.

Abstract Lysophosphoglycerides (LPLs) have been reported to accumulate in myocardium and serve as a cause of arrhythmias in acute myocardial ischemia. However, in this study we found that LPLs level in the ventricular myocardium was decreased by the onset of acute myocardial ischemia in vivo in rats. Decreasing of LPLs level in left ventricular myocardium, but not right, was observed within 26 min of left myocardial ischemia, regardless of whether arrhythmias were triggered. Abstract Urinary tract infection (UTI) caused by spinal cord injury (SCI) can have significant morbidity. There is currently a lack of relevant data in China. This study explores incidence and risk factors of UTI in hospitalized patients with SCI in China, and will help healthcare professionals to make informed clinical decisions to reduce the incidence of UTI. This retrospective study analyzed the medical records of patients with SCI who were hospitalized at three campuses of a hospital in central China between August 2014 and August 2023. The files of patients with SCI were reviewed for demographics and clinical characteristics. Logistic regression analysis was performed to identify risk factors associated with UTI. A total of 538 patients were included in this study. The incidence of UTI was 49.8%. Sex, hypoproteinemia, urinary incontinence, bladder irrigation, timing of rehabilitation, duration of indwelling urinary catheter were risk factors of UTI. The implementation of specific preventive measures is anticipated to result in a decrease in the occurrence of UTI among individuals with SCI, consequently enhancing their overall quality of life and prognosis.

Abstract In order to obtain the real material parameters of heterogeneous rock, the material parameters of red sandstone specimens under uniaxial compression tests are inverted based on the Digital Image Correlation (DIC) method and the Finite Element Model Updating (FEMU) method. The DIC method is employed to calculate the displacement field of red sandstone specimens during uniaxial compression loading. Concurrently, a uniaxial compression elastic–plastic finite element numerical model with non-uniform material parameters is developed based on the FEMU method. The model adopts the Mohr–Coulomb yield criterion and adjusts the boundary conditions in real-time to maintain consistency with the test. The vertical displacement field of the numerical model is juxtaposed with that of the test to construct the objective function. Optimization is achieved using the Artificial Fish Swarm algorithm, which enables the acquisition of the non-uniform distribution and evolution process of the material parameters of specimens at different loading moments. The results indicate that this method can spatially obtain the non-uniform distribution field of material parameters and temporally track the evolution of material parameters during the loading process. This research lays a solid foundation for enhancing the accuracy of intelligent coal mining and dynamic disaster monitoring and early warning in coal mines.

Abstract We present the first data-driven pediatric model that explains cranial sutural growth in the pediatric population. We segmented the cranial bones in the neurocranium from the cross-sectional CT images of 2068 normative subjects (age 0–10 years), and we used a 2D manifold-based cranial representation to establish local anatomical correspondences between subjects guided by the location of the cranial sutures. We designed a diffeomorphic spatiotemporal model of cranial bone development as a function of local sutural growth rates, and we inferred its parameters statistically from our cross-sectional dataset. We used the constructed model to predict growth for 51 independent normative patients who had longitudinal images. Moreover, we used our model to simulate the phenotypes of single suture craniosynostosis, which we compared to the observations from 212 patients. We also evaluated the accuracy predicting personalized cranial growth for 10 patients with craniosynostosis who had pre-surgical longitudinal images. Unlike existing statistical and simulation methods, our model was inferred from real image observations, explains cranial bone expansion and displacement as a consequence of sutural growth and it can simulate craniosynostosis. This pediatric cranial suture growth model constitutes a necessary tool to study abnormal development in the presence of cranial suture pathology.

Abstract We conducted a cross-sectional study using an online questionnaire to investigate the current status of urinary tract infections (UTIs) and the use of clean intermittent catheterization (CIC) in Chinese community-based SCI patients and to explore the risk factors for UTIs in patients using CIC. Our findings suggest that the prevalence of UTIS is higher in Chinese community-based SCI patients compared with patients in medically developed countries. In addition, we found that CIC had the lower incidence of UTIs than urinary indwelling catheter (UIC) and suprapubic catheter (SPC), and that SCI patients with CIC had low rates of use and poor compliance. Further analysis indicated that most of the risk factors for UTIs in CIC patients were associated with irregular use of CIC. Therefore, we call for not only the need to provide stronger caregiver support and financial assistance to improve CIC adherence in Chinese community SCI patients, but also the establishment of a database of Chinese SCI patients in order to enhance the management of bladder emptying methods and further standardize the CIC operation in such patients, thus reducing the risk of UTIs in Chinese community SCI patients.

Abstract Soybean agglutinin (SBA) is a main anti-nutritional factor in soybean. SBA exhibits its anti-nutritional functions by binding to intestinal epithelial cells. Keratin8 (KRT8), Keratin18 (KRT18) and Actin (ACTA) are the representative SBA-specific binding proteins. Such cytoskeletal proteins act a crucial role in different cell activities. However, limited reports reveal what the signal transduction pathway of apoptosis caused by SBA when binding to KRT8, KRT18 and ACTA. We aimed to evaluate the effects of SBA on cell apoptosis and the expression of the cytoskeletal protein (KRT8, KRT18 and ACTA), reveal the roles of these cytoskeletal proteins or their combinations on SBA-induced cell apoptosis in IPEC-J2 cell line, evaluate the influences of SBA on the mitochondria, endoplasmic reticulum stress and death receptor-mediated apoptosis signal pathway and to show the roles of KRT8, KRT18 and ACTA in different apoptosis signal pathways induced by SBA. The results showed that SBA induced the IPEC-J2 cell apoptosis and decreased the mRNA expression of KRT8, KRT18 and ACTA (p KRT8 > KRT18. The roles of these three cytoskeletal proteins on IPEC-J2 apoptotic rates had a certain accumulation effect. Abstract In cross-country skiing competitions, the choice of pacing strategy is of decisive significance to athletes' performance. A reasonable pacing strategy is essential for athletes to improve their performance. In this paper, the mathematical models of cross-country skiing simulation and pacing optimization are established, including motion model, athlete power output model, and optimization model. The actual competition data of a Chinese athlete in Guyangshu 1.5 km track was compared with the model simulation results. The whole process time error is less than 3%, which verifies the accuracy of the motion and power output model. Gauss pseudo-spectral method is applied to the optimization model. By changing the distribution of athletes' power output, the racing time is minimized under the condition that the total energy output remains unchanged. Compared with the pacing strategy before optimization, the optimized racing time was shortened by 12.6 s, which verifies the effectiveness of the optimization model. Optimized results show that in the first significant uphill section, a recommendation is to use a more conservative strategy, while in the latter half of uphill sections the power output should be increased.

Abstract The wastewater treatment efficiency is crucial to constructing a livable ecological environment and promoting the sustainable development of economy and society. The differences in natural conditions, economic development and local policies between the Yangtze River Basin (YRB) and the Non-Yangtze River Basin (NYRB) increase the difficulty of wastewater treatment in governance. This study uses a modified Dynamic Data Envelopment Analysis (DEA) model to assess the wastewater treatment from 2013 to 2020, and divides the study period into two stages: the first stage (2013–2017) assesses the wastewater treatment efficiency of 18 provinces and cities in YRB and 12 provinces and cities in NYRB; the second stage (2018–2020) conducts statistical analysis of wastewater discharge pollutants in YRB and NYRB. The results conclude that the total wastewater treatment efficiency is generally low, but polarization is quite prominent. Among total wastewater treatment efficiency, NYRB scored 0.504, or slightly higher than YRB (0.398). In terms of expense efficiency, both NYRB and YRB scored below 0.4. In terms of chemical oxygen demand (COD) output efficiency, YRB (0.488) is better than NYRB (0.420). The second stage of statistical analysis presents that pollutant emissions are still high; the regions need to increase wastewater treatment investment and improve wastewater treatment efficiency.

Abstract Preventing wound infection and retaining an appropriate level of moisture around wounds represent the most critical issues in wound treatment. Towards these ends, special focus has been placed on Bombyx mori cocoons because the protective function of the silkworm cocoon resembles the manner in which the skin protects the human body. We have designed a facile technique to develop a novel silkworm cocoon-based wound film (SCWF) wound dressing utilizing a CaCl2-ethanol-H2O solution. To improve the anti-bacterial performance of SCWF, we have incorporated the ability of silk sericin to act as a reducing agent for the conversion of Ag+ to Ag, yielding nanoparticles (AgNPs) linked together by peptide bonds of silkworm cocoon wound film (SCWF-AgNPs). SCWF-AgNP dressing exhibited excellent biocompatibility, anti-bacterial performance, and good extensibility. Furthermore, in vivo experiments indicated that SCWF-AgNP dressing was able to significantly accelerate the healing rate of infected wounds in New Zealand White rabbits and histological examination revealed that it aided in the successful reconstruction of intact and thickened epidermis during 14 days of healing of impaired wound tissue. These results demonstrate that the present approach might shed new light on the design of anti-bacterial materials such as SCWF-AgNPs with promising applications in wound dressing.
