Abstract The genus Erebia comprises numerous species in Europe. Due to preference of cold environments, most species have disjunct distributions in the European mountain systems. However, their biogeographical patterns may differ significantly. The Bright-eyed ringlet Erebia oeme is widespread in high-altitude grasslands of the European high mountains, hence showing a disjunct distribution pattern. Over its distribution, E. oeme shows high morphological variability indicating pronounced intraspecific differentiation. We analysed two mitochondrial (COI, Cytb) and eight nuclear markers as well as the Wolbachia surface protein-coding gene (WSP). A total of four lineages were identified: two Balkan lineages, one Slovenian lineage as well as one lineage containing all other individuals (Alps, Massif Central, Pyrenees). COI data only indicate a fifth lineage in the southern Carpathians. The region of origin of E. oeme is most likely the western Balkans. From here, E. oeme spread to the eastern Balkan area and further to the southern Carpathians as well as to Slovenia, from where it rapidly crossed the Alps to Massif Central and Pyrenees. Wolbachia was found to be highly prevalent with multiple strains, partly within one population. However, recent Wolbachia impact on the mitochondrial DNA and consequently influence on the intraspecific genetic structure was not detected.

Abstract In a variety of species, individuals appear to be consistent in the daily timing of their activity onset. Such consistent among-individual differences can result from both intrinsic factors, as individuals may e.g. differ genetically, and extrinsic factors, as the environment may vary on spatial and temporal scales. However, previous studies typically did not differentiate between their respective contributions on individual variation in the timing of activities. Here, we repeatedly measured the onset of activity in female great tits (Parus major) on consecutive days during the egg laying phase of the breeding season in four consecutive years. Subsequently, we used a variance partitioning analysis in order to determine which part of the total variation could be attributed to intrinsic (female identity) and extrinsic (nest box identity) factors. Overall, 27% of the total variation could be attributed to female identity. In addition, we found temporal variation in the activity onset, indicating that individuals can plastically adjust their timing. Yet despite their general ability to change the timing of activities over time, spatial environmental factors did not contribute significantly to the observed variation. Individuals may choose a habitat that matches the preferred timing of activities, or might not benefit from adjusting their timing to environmental factors that might vary on spatial scales.

Abstract For many decades, silicone elastomers with oil incorporated have served as fouling-release coating for marine applications. In a comprehensive study involving a series of laboratory-based marine fouling assays and extensive global field studies of up to 2-year duration, we compare polydimethylsiloxane (PDMS) coatings of the same composition loaded with oil via two different methods. One method used a traditional, one-pot pre-cure oil addition approach (o-PDMS) and another method used a newer post-cure infusion approach (i-PDMS). The latter displays a substantial improvement in biofouling prevention performance that exceeds established commercial silicone-based fouling-release coating standards. We interpret the differences in performance between one-pot and infused PDMS by developing a mechanistic model based on the Flory–Rehner theory of swollen polymer networks. Using this model, we propose that the chemical potential of the incorporated oil is a key consideration for the design of future fouling-release coatings, as the improved performance is driven by the formation and stabilization of an anti-adhesion oil overlayer on the polymer surface.

Abstract Determine the prevalence of pelvic floor disorders (PFD) stratified by age, race, body mass index (BMI), and parity in adult women attending family medicine and general internal medicine clinics at an academic health system. The medical records of 25,425 adult women attending primary care clinics were queried using International Classification of Diseases-10th Revision codes (ICD-10 codes) for PFD [urinary incontinence (UI), pelvic organ prolapse (POP), and bowel dysfunction (anal incontinence (AI) and difficult defecation)]. Prevalence and odds ratios were calculated using univariate and multivariate analysis for age, race, BMI, and parity when available. Multivariate logistic regression models were used to assess the impact of age, race, BMI, and parity on the likelihood of being diagnosed with a PFD. A separate model was constructed for each of the three PFD categories (UI, POP, and bowel dysfunction) as well as a model assessing the likelihood of occurrence for any type of PFD. The percentage of women with at least one PFD was 32.0% with bowel dysfunction the most common (24.6%), followed by UI (11.1%) and POP (4.4%). 5.5% had exactly two PFD and 1.1% had all 3 categories of PFD. Older age and higher BMI were strongly and significantly associated with each of the three PFD categories, except for BMI and prolapse. Relative to White patients, Asian patients were at significantly lower risk for each category of PFD, while Black patients were at significantly lower risk for UI and POP, but at significantly greater risk for bowel dysfunction and the presence of any PFD. Higher parity was also significantly associated with pelvic organ prolapse. Using multivariate analyses, age, race, and BMI were all independently associated with PFD. PFD are highly prevalent in the primary care setting and should be screened for, especially in older and obese women. BMI may represent a modifiable risk factor.

Abstract Several morphological and mitochondrial lineages of the alpine ringlet butterfly species Erebia pronoe have been described, indicating a complex phylogenetic structure. However, the existing data were insufficient and allow neither a reconstruction of the biogeographic history, nor an assessment of the genetic lineages. Therefore, we analysed mitochondrial (COI, NDI) and nuclear (EF1α, RPS5) gene sequences and compared them with sequences from the sister species Erebia melas. Additionally, we combined this information with morphometric data of the male genitalia and the infection patterns with Wolbachia strains, based on a WSP analysis. We obtained a distinct phylogeographic structure within the E. pronoe-melas complex with eight well-distinguishable geographic groups, but also a remarkable mito-nuclear discordance. The mito-nuclear discordance in E. melas and E. pronoe glottis can be explained by different ages of Wolbachia infections with different Wolbachia strains, associated selective sweeps, and hybridisation inhibition. Additionally, we found indications for incipient speciation of E. pronoe glottis in the Pyrenees and a pronounced range dynamic within and among the other high mountain systems of Europe. Our results emphasize the importance of combined approaches in reconstructing biogeographic patterns and evaluating phylogeographic splits.

Abstract Microplastics are ubiquitous in standing freshwater bodies, consequently lakes and reservoirs may be important sinks for these contaminants. However, the mechanisms governing the deposition of microplastics and their interactions with the sediments are understudied. We demonstrate how aggregation-based transport facilitates the sinking and infiltration of buoyant microplastics into freshwater reservoir sediments by employing experiments with intact sediment cores. Buoyant polyethylene microplastics were rapidly (1–4 h) incorporated into sinking iron-organic aggregates, followed by swift deposition into sediments. Ingression of microplastic bearing flocs into sediments was completed within 6 days and led to stable deposition of the incorporated particles for at least 2 months. Most microplastics were deposited in the top 2 cm of the sediments and few particles (5–15%) were re-released into the water. Our results show at least 85% burial of microplastics, indicating the significant role of freshwaters with low flow velocities in reducing microplastic loads to the oceans.

Abstract Sexual conflict arises when two individuals invest in their common offspring because both individuals benefit when their partner invests more. Conditional cooperation is a theoretical concept that could resolve this conflict. Here, parents are thought to motivate each other to contribute to provisioning visits by following the rules of turn taking, which results in equal and efficient investment. However, parents have other tasks besides provisioning, which might hinder taking turns. To investigate restrictions by other care tasks and whether turn taking can be used to match investment, we manipulated brooding duration in female blue tits (Cyanistes caeruleus) during the early nestling phase by changing nest box temperature. As expected, females subjected to cold conditions brooded longer than females under warm conditions. Yet, contrary to our prediction, females had similar visit rates in both treatments, which suggests that females in the cold treatment invested more overall. In addition, the females’ turn taking level was higher in the more demanding cold condition (and the calculated randomised turn taking levels of females did not differ), hence females don’t seem to be restricted in their turn taking strategy by other care tasks. However, males did not seem to match the females’ turn taking levels because they did not adjust their visit rates. Thus, level of turn taking was not restricted by an other sex-specific task in females and did not facilitate a greater investment by their male partners.

Abstract Parents allocate resources to offspring to increase their survival and to maximize their own fitness, while this investment implies costs to their condition and future reproduction. Parents are hence expected to optimally allocate their resources. They should invest equally in all their offspring under good conditions, but when parental capacity is limited, parents should invest in the offspring with the highest probability of survival. Such parental favouritism is facilitated by the fact that offspring have evolved condition-dependent traits to signal their quality to parents. In this study we explore whether the parental response to an offspring quality signal depends on the intrinsic capacity of the parents, here the female. We first manipulated the intrinsic capacity of blue tit (Cyanistes caeruleus) females through lutein-supplementation during egg laying, and we subsequently blocked the UV/yellow reflectance of breast feathers on half of the nestlings in each brood. We did not find evidence that the female intrinsic capacity shaped parental feeding or sibling competition according to offspring UV/yellow colouration. However, nestling UV/yellow colour affected costly behavioural interactions in the form of prey-testings (when a parent places a prey item into a nestling’s gape but removes it again). In lutein-supplemented nests, fathers but not mothers favoured UV-blocked chicks by testing them less often, supporting previous results. Accordingly, in lutein-supplemented nests, UV-blocked nestlings gained more mass than their siblings, while in control nests we found the opposite effect and UV-blocked nestlings gained less. Our results emphasize that the prenatal environment shaped the role of offspring UV/yellow colour during certain family interactions and are indicative for sex-specific parental care strategies.

Abstract The risk of breast cancer associated with CHEK2:c.1100delC is 2–threefold but higher in carriers with a family history of breast cancer than without, suggesting that other genetic loci in combination with CHEK2:c.1100delC confer an increased risk in a polygenic model. Part of the excess familial risk has been associated with common low-penetrance variants. This study aimed to identify genetic loci that modify CHEK2:c.1100delC-associated breast cancer risk by searching for candidate risk alleles that are overrepresented in CHEK2:c.1100delC carriers with breast cancer compared with controls. We performed whole-exome sequencing in 28 breast cancer cases with germline CHEK2:c.1100delC, 28 familial breast cancer cases and 70 controls. Candidate alleles were selected for validation in larger cohorts. One recessive synonymous variant, rs16897117, was suggested, but no overrepresentation of homozygous CHEK2:c.1100delC carriers was found in the following validation. Furthermore, 11 non-synonymous candidate alleles were suggested for further testing, but no significant difference in allele frequency could be detected in the validation in CHEK2:c.1100delC cases compared with familial breast cancer, sporadic breast cancer and controls. With this method, we found no support for a CHEK2:c.1100delC-specific genetic modifier. Further studies of CHEK2:c.1100delC genetic modifiers are warranted to improve risk assessment in clinical practice.

Abstract Merkel cell carcinoma (MCC) is a rare, but aggressive skin cancer the incidence of which has increased significantly in recent years. The majority of MCCs have incorporated Merkel cell polyomavirus (VP-MCC) while the remainder are virus-negative (VN-MCC). Although a variety of therapeutic options have shown promise in treating MCC, there remains a need for additional therapeutics as well as probes for better understanding MCC. A high-throughput screening campaign was used to assess the ability of > 25,000 synthetic and natural product compounds as well as > 20,000 natural product extracts to affect growth and survival of VN-MCC and VP-MCC cell lines. Sixteen active compounds were identified that have mechanisms of action reported in the literature along with a number of compounds with unknown mechanisms. Screening results with pure compounds suggest a range of potential targets for MCC including DNA damage, inhibition of DNA or protein synthesis, reactive oxygen species, and proteasome inhibition as well as NFκB inhibition while also suggesting the importance of zinc and/or copper binding. Many of the active compounds, particularly some of the natural products, have multiple reported targets suggesting that this strategy might be a particularly fruitful approach. Processing of several active natural product extracts resulted in the identification of additional MCC-active compounds. Based on these results, further investigations focused on natural products sources, particularly of fungal origin, are expected to yield further potentially useful modulators of MCC.

Abstract Soil property and class maps for the continent of Africa were so far only available at very generalised scales, with many countries not mapped at all. Thanks to an increasing quantity and availability of soil samples collected at field point locations by various government and/or NGO funded projects, it is now possible to produce detailed pan-African maps of soil nutrients, including micro-nutrients at fine spatial resolutions. In this paper we describe production of a 30 m resolution Soil Information System of the African continent using, to date, the most comprehensive compilation of soil samples ( $$N \approx 150,000$$ N ≈ 150 , 000 ) and Earth Observation data. We produced predictions for soil pH, organic carbon (C) and total nitrogen (N), total carbon, effective Cation Exchange Capacity (eCEC), extractable—phosphorus (P), potassium (K), calcium (Ca), magnesium (Mg), sulfur (S), sodium (Na), iron (Fe), zinc (Zn)—silt, clay and sand, stone content, bulk density and depth to bedrock, at three depths (0, 20 and 50 cm) and using 2-scale 3D Ensemble Machine Learning framework implemented in the mlr (Machine Learning in R) package. As covariate layers we used 250 m resolution (MODIS, PROBA-V and SM2RAIN products), and 30 m resolution (Sentinel-2, Landsat and DTM derivatives) images. Our fivefold spatial Cross-Validation results showed varying accuracy levels ranging from the best performing soil pH (CCC = 0.900) to more poorly predictable extractable phosphorus (CCC = 0.654) and sulphur (CCC = 0.708) and depth to bedrock. Sentinel-2 bands SWIR (B11, B12), NIR (B09, B8A), Landsat SWIR bands, and vertical depth derived from 30 m resolution DTM, were the overall most important 30 m resolution covariates. Climatic data images—SM2RAIN, bioclimatic variables and MODIS Land Surface Temperature—however, remained as the overall most important variables for predicting soil chemical variables at continental scale. This publicly available 30-m Soil Information System of Africa aims at supporting numerous applications, including soil and fertilizer policies and investments, agronomic advice to close yield gaps, environmental programs, or targeting of nutrition interventions.

Abstract Glacial refugia are centers of high biodiversity. Therefore, knowledge on their locations and reactions of associated populations and landscapes to climatic changes is crucial for conservation management. We here investigated the biogeography of a butterfly species linked to open forest habitats. Using mitochondrial and nuclear markers in combination with Bayesian simulations, we analyzed the location and age of potential glacial refugia of the species. We identified five putative refugia in Europe. Considering the ecological needs of our study species, tree density within these refugial areas, in contrast to earlier assumptions, must have exceeded the level of individually scattered trees. Our results also provide evidence that especially the refuge areas in the Carpathians were previously underestimated regarding their age: the refugia in the Southern Carpathians presented suitable conditions throughout several glacial cycles, probably since the Mindel or Riss cycles. Additionally, our analyses provided support for a forest refugium near the Tatra Mountains persisting the last glacial maximum. Our results underline the usefulness of this and probably other butterfly species as indicators of forest refugia.

Abstract The decomposition of beta-diversity (β-diversity) into its replacement (βrepl) and richness (βrich) components in combination with a taxonomic and functional approach, may help to identify processes driving community composition along environmental gradients. We aimed to understand which abiotic and spatial variables influence ant β-diversity and identify which processes may drive ant β-diversity patterns in Mediterranean drylands by measuring the percentage of variation in ant taxonomic and functional β-diversity explained by local environmental, regional climatic and spatial variables. We found that taxonomic and functional replacement (βrepl) primarily drove patterns in overall β-diversity (βtot). Variation partitioning analysis showed that respectively 16.8%, 12.9% and 21.6% of taxonomic βtot, βrepl and βrich variation were mainly explained by local environmental variables. Local environmental variables were also the main determinants of functional β-diversity, explaining 20.4%, 17.9% and 23.2% of βtot, βrepl and βrich variation, respectively. Findings suggest that niche-based processes drive changes in ant β-diversity, as local environmental variables may act as environmental filters on species and trait composition. While we found that local environmental variables were important predictors of ant β-diversity, further analysis should address the contribution of other mechanisms, e.g. competitive exclusion and resource partitioning, on ant β-diversity.

Abstract Acute respiratory distress syndrome (ARDS) is associated with high mortality. We sought to identify biological pathways in ARDS that differentiate survivors from non-survivors. We studied bronchoalveolar lavage fluid (BALF) from 36 patients with ARDS (20 survivors, 16 non-survivors). Each sample, obtained within seven days of ARDS onset, was depleted of high abundance proteins and labeled for iTRAQ LC-MS/MS separately. Protein identification and relative quantification was performed employing a target-decoy strategy. A variance weighted t-test was used to identify differential expression. Ingenuity Pathway Analysis was used to determine the canonical pathways that differentiated survivors from non-survivors. We identified 1115 high confidence proteins in the BALF out of which 142 were differentially expressed between survivors and non-survivors. These proteins mapped to multiple pathways distinguishing survivors from non-survivors, including several implicated in lung injury and repair such as coagulation/thrombosis, acute phase response signaling and complement activation. We also identified proteins assigned to fibrosis and ones involved in detoxification of lipid peroxide-mediated oxidative stress to be different in survivors and non-survivors. These results support our previous findings demonstrating early differences in the BALF protein expression in ARDS survivors vs. non-survivors, including proteins that counter oxidative stress and canonical pathways associated with fibrosis.

Abstract How much to invest in parental care and by who remain puzzling questions fomented by a sexual conflict between parents. Negotiation that facilitates coordinated parental behaviour may be key to ease this costly conflict. However, understanding cooperation requires that the temporal and sex-specific variation in parental care, as well as its multivariate nature is considered. Using a biparental bird species and repeated sampling of behavioural activities throughout a major part of reproduction, we show a clear division of tasks between males and females in provisioning, brooding and foraging. Such behavioural specializations fade with increasing nestling age, which stimulates the degree of alternated feeding visits, as a recently promoted form of conditional cooperation. However, such cooperation is thought to benefit offspring development, which is not supported by our data. Thus, from a proximate point of view, conditional cooperation via alternation critically depends on the division of parental tasks, while the ultimate benefits have yet to be shown.
