Emerging chemical contaminants (ECCs) are among the major environmental threats in present century. A variety of ECCs is released into aquatic environments with little knowledge about their long-term impacts to organisms. We examined the role of acclimation of the freshwater rotifer Brachionus calyciflorus to silver nanoparticles (Ag-NPs) and 5-fluorouracil (5-FU) for determining their ability to deal with these ECCs individually and in mixtures along multiple generations. Additionally, transgenerational effects were also assessed during the recovery phase. Rotifers acclimated at EC 10 of Ag-NPs along generations showed a higher ability to deal with higher concentrations of these nanoparticles or 5-FU along generations. Rotifers acclimated to EC 10 of 5-FU showed varied responses, as their population growth rates were affected at the initial generations once exposed to higher concentration (EC 50 ) of the same or a new contaminant; however, the rotifers acquired resistance in later generations. The exposure of generational Ag-NP-acclimated rotifers to the mixture of Ag-NPs and 5-FU at EC 50 led to a shift from no effects to negative effects along successive generations, suggesting a decrease in resistance, which remained even in the post-exposure recovery phase. Similar transgenerational adverse effects were also observed for the generational Ag-NP-acclimated rotifers released from 5-FU. Rotifers acclimated to 5-FU showed a decrease in population growth rate at the first generation of recovery phase, possibly shifting their optimal environmental conditions when released from contaminants. Overgrazing generally induces dwarfism in grassland plants, and these phenotypic traits could be transmitted to clonal offspring even when overgrazing is excluded. However, the dwarfism-transmitted mechanism remains largely unknown, despite generally thought to be enabled by epigenetic modification. To clarify the potential role of DNA methylation on clonal transgenerational effects, we conducted a greenhouse experiment with Leymus chinensis clonal offspring from different cattle/sheep overgrazing histories via the demethylating agent 5-azacytidine. The results showed that clonal offspring from overgrazed (by cattle or sheep) parents were dwarfed and the auxin content of leaves significantly decreased compared to offspring from no-grazed parents'. The 5-azaC application generally increased the auxin content and promoted the growth of overgrazed offspring while inhibited no-grazed offspring growth. Meanwhile, there were similar trends in the expression level of genes related to auxin-responsive target genes (ARF7, ARF19), and signal transduction gene (AZF2). In the context of global changes, fish are increasingly exposed to multiple stressors that have cascading effects from molecules to the whole individual, thereby affecting wild fish populations through selective processes. In this review, we synthetize recent advances in molecular biology and evolutionary biology to outline some potentially important effects of stressors on fish across biological levels. Given the burgeoning literature, we highlight four promising avenues of research. First, (1) the exposure to multiple stressors can lead to unexpected synergistic or antagonistic effects, which should be better taken into account to improve our predictions of the effects of actual and future human activities on aquatic organisms. Second, (2) we argue that such interactive effects might be due to switches in energy metabolism leading to threshold effects. Under multiple stress exposure, fish could switch from a "compensation" strategy, i.e. a reallocation of energy to defenses and repair to a "conservation" strategy, i.e. blocking of stress responses leading to strong deleterious effects and high mortality. Third, (3) this could have cascading effects on fish survival and population persistence but multiscale studies are still rare. We propose emerging tools merging different levels of biological organization to better predict population resilience under multiple stressors. Fourth (4), there are strong variations in sensitivity among populations, which might arise from transgenerational effects of stressors through plastic, genetic, and epigenetic mechanisms. This can lead to local adaptation or maladaptation, with strong impacts on the evolutionary trajectories of wild fish populations. With this review, we hope to encourage future research to bridge the gap between molecular ecology, ecotoxicology and evolutionary biology to better understand the evolution of responses of fishes to current and future multiple stressors in the context of global changes. There is a lack of information about the transgenerational effects of titanium dioxide nanoparticles (nano-TiO 2 ) in plants. This study aimed to evaluate the impacts of successive exposure of nano-TiO 2 with different surface properties to basil ( Ocimum basilicum ). Seeds from plants exposed or re-exposed to pristine, hydrophobic, or hydrophilic nano-TiO 2 were cultivated for 65 days in soil unamended or amended with 750 mg·kg −1 of the respective particles. Plant growth, concentration of titanium and essential elements, as well as content of carbohydrates and chlorophyll were evaluated. There were no differences on Ti concentration in roots of plants sequentially exposed to pristine or hydrophobic nano-TiO 2 , or in roots of plants exposed to the corresponding particle, only in the second cycle. However, sequential exposure to hydrophilic particles resulted in 65.2% less Ti in roots, compared to roots of plants exposed the same particles, only in the second cycle. The Ti concentrations in shoots were similar in all treatments. On the other hand, pristine and hydrophilic particles reduced Mg in root by 115% and 81%, respectively, while pristine and hydrophobic particles reduced Ni in shoot by 84% and 75%, respectively, compared to unexposed plants in both cycles. Sequential exposure to pristine nano-TiO 2 increased stomatal conductance (214%, p  ≤ 0.10), compared to plants that were never exposed. Hydrophobic and hydrophilic nano-TiO 2 reduced chlorophyll b (52%) and total chlorophyll (30%) but increased total sugar (186%) and reducing sugar (145%), compared to unexposed plants in both cycles. Sequential exposure to hydrophobic or hydrophilic nano-TiO 2 resulted in more adverse effects on photosynthesis but in positive effects on plant growth, compared to pristine nano-TiO 2 . Ocean acidification is likely to have profound impacts on marine bivalves, especially on their early life stages. Therefore, it is imperative to know whether and to what extent bivalves will be able to acclimate or adapt to an acidifying ocean over multiple generations. Here, we show that reduced seawater pH projected for the end of this century (i.e., pH 7.7) led to a significant decrease of shell production of newly settled juvenile Manila clams, Ruditapes philippinarum . However, juveniles from parents exposed to low pH grew significantly faster than those from parents grown at ambient pH, exhibiting a rapid transgenerational acclimation to an acidic environment. The sodium composition of the shells may shed new light on the mechanisms responsible for beneficial transgenerational acclimation. Irrespective of parental exposure, the amount of Na incorporated into shells increased with decreasing pH, implying active removal of excessive protons through the Na + /H + exchanger which is known to depend on the Na + gradient actively built up by the Na + /K + -ATPase as a driving force. However, the shells with a prior history of transgenerational exposure to low pH recorded significantly lower amounts of Na than those with no history of acidic exposure. It therefore seems very likely that the clams may implement less costly and more ATP-efficient ion regulatory mechanisms to maintain pH homeostasis in the calcifying fluid following transgenerational acclimation. DNA methylation is an epigenetic mechanism of particular importance in developmental biology, but methylation also varies along organisms' life cycle. Recent studies have deliberated copper (Cu) exposure induced epigenetic changes in Enchytraeus crypticus , a standard species belonging to one of the most common and important genera of soil invertebrates in many ecosystems. There is however no information on how DNA methylation levels change within the life cycle of this species. We here investigate the global DNA methylation profile along the life cycle of E. crypticus and compare this to the expression of target genes involved in methylation. Results showed that after the lowest DNA methylation level at day 3 (early embryonic stage, blastula) there was an increase by day 7 (organogenesis) after which levels were maintained at days 11, 18 and 25. DNA methyltransferase associated protein 1 (DMPA1) and Methyl Binding Domain 2 (MBD2) gene expression was highest during embryo stages (3 to 7 days), then decreasing (11, 18 days) and finally unregulated in adults (25 days). Hence, we here show that DNA methylation in E. crypticus changes among the different life stages, from cocoons to adults. Such information is a key knowledge to use this endpoint and tool in an ecotoxicology context. This means that it is almost implicit that gene expression levels are age specific for a given stressor. It seems logic to recommend to always compare individuals with the same age between treatments, and to be careful when extrapolating results among life stages. Once, we understand more of these effects we may even be able to predict which life stage is more sensitive to specific stressors. An experimental design that aims to cover epigenetics of stressors in a multigenerational exposure, including transgenerational effects, should ensure the synchronous age of organisms for sampling analysis purposes. Ocean acidification can negatively impact marine bivalves. Pivotal to projecting their fate is the ability to acclimate and adapt to shifts in seawater chemistry. Transgenerational plasticity enables marine bivalves to acclimate, yet the underlying mechanisms at different levels of biological organization remain poorly understood. Here, we performed a transgenerational experiment to understand biochemical responses of the Manila clam, Ruditapes philippinarum , following exposure to moderately reduced seawater pH (from 8.1 to 7.7). Activities of tissue calcification-relevant enzymes, such as carbonic anhydrase (CA), acid phosphatase (ACP) and alkaline phosphatase (ALP), energy-metabolizing enzymes, such as Na+/K+-ATPase (NKA) and Ca2+/Mg2+-ATPase (CMA), as well as tissue energy reserves (glycogen, lipid and protein) were assayed. With decreasing seawater pH, adult R. philippinarum exhibited significantly increased CA activity, and especially the clams with a history of transgenerational exposure displaying significantly higher CA activity than those spawned from parents exposed to ambient seawater pH. Yet, ACP and ALP activities remained unaffected. Transgenerational exposure to reduced seawater pH led to significant increases of NKA activity, while no transgenerational response of CMA activity was observed. Tissue glycogen and lipid contents were significantly depleted under acidified conditions regardless of transgenerational exposure. Yet, transgenerational alleviation in the net protein degradation was found. These findings suggest that our current understanding of transgenerational responses is still limited by the achievable time-window possible in the laboratory. While the energetic budget is lower under acidified conditions, there is no evidence of transgenerational recovery in term of energetic budget. Therefore, this work demonstrates that the critical basis of ocean acidification resilience can most likely be explained in energetic terms.
