The haematological response of regular, repeated blood harvesting was investigated in 40 Thoroughbred and non-Thoroughbred horses that donate 8 litres of blood every 3 weeks for the purposes of commercial blood production. When this volume of blood was removed on five occasions over 12 weeks, no adverse effect on packed cell volume (PCV), haemoglobin (HB), and red blood cell count (RCC) was observed. Although PCV, RCC and Hb values decreased during the first week after blood collection, followed by a gradual increase in values until the next harvest time, all values remained within published reference ranges. Derived red cell indices [mean corpuscular volume (MCV), mean corpuscular haemoglobin (MCH), and mean corpuscular haemoglobin concentration (MCHC)] also remained within reference range. We conclude that the removal of approximately 8 litres of blood (approximately 16 ml kg(-1)or 20 per cent of blood volume for a 500 kg horse) from blood donor horses every 3 weeks allows time for adequate recovery of haematological variables and does not result in adverse haematological changes., (Copyright 2000 Harcourt Publishers Ltd.)

The aim of this study was to establish the normal patterns of breath hydrogen excretion in adult ponies following either voluntary consumption or administration per stomach tube of a test meal/carbohydrate substrate. After an overnight fast, the ponies (n = 7) received either no test meal (ie fasted), glucose, xylose, lactose, lactulose, wheat flour or oats. Exhaled breath samples were collected in duplicate at 0 minutes and at 30-minute intervals thereafter for eight hours. Analysis of duplicate breath hydrogen measurements (n = 714) indicated that the breath hydrogen collection/storage method was reliable. Fasting resulted in negligible levels of breath hydrogen excretion. Increases in breath hydrogen concentration greater than 10 ppm, sustained for at least 30 minutes, were regarded as being biologically significantly different than fasting data and were observed for all ponies following the ingestion of oats or the administration of wheat flour, for three ponies following the administration of glucose and xylose and for two ponies following the administration of lactulose and lactose. The pattern of breath hydrogen excretion was subject to variation between animals following the ingestion of identical test meals. This study provides evidence of incomplete glucose absorption in healthy ponies and indicates that ingestion of non-absorbable carbohydrates do not always result in hydrogen excretion.

Papillomaviral DNA has been identified in peripheral blood cells of both cattle and humans with and without associated disease and it has been suggested that such cells may act as sites of viral latency. In order to investigate the possibility of latent papillomaviral infection in the aetiopathogenesis of the equine sarcoid, peripheral blood derived DNA samples from 20 healthy and 34 sarcoid-affected donkeys were subject to polymerase chain reaction (PCR) using papillomaviral specific primers. Analysis of blood derived DNA samples failed to demonstrate the presence of papillomaviral DNA in any animal. Screening of 37 matched sarcoid derived DNA samples confirmed the presence of BPV in 34 diseased donkeys. This study supports the hypothesis of BPV as an aetiological agent in the equine sarcoid and suggests that latent virus in circulating peripheral blood cells does not play a role in the pathogenesis and epidemiology of the equine sarcoid.

Validation of an assay for measurement of fructosamine in equine serum and plasma utilised blood samples collected from 24 British native breed ponies. The results indicated that fructosamine can be measured easily using an assay which is precise and accurate. Paired plasma and serum fructosamine measurements were highly correlated, however, greater variations were observed within serum compared with those in plasma. A reference range for fructosamine in plasma was calculated to be 256.9 +/- 60.6 mumol litre-1 (mean +/- 2 SD). In order to assess the fructosamine response following experimental cyathostome infection, nine British native breed ponies were allocated to one of three groups: Group 1 (3.9 million third-stage cyathostome larvae (L3) over a nine-week-period), Group 2 (3.15 million L3 over seven weeks) and Group 3 (uninfected controls). From four weeks prior to infection, blood was obtained, once weekly, from all animals for measurement of plasma fructosamine, plasma albumin and serum globulin concentrations. Plasma fructosamine concentrations decreased in all infected ponies post-infection due to enteric protein loss and/or altered protein composition and/or increased protein turnover.
