Objective: To study the expression of insulin-like growth factor binding protein 3(IGFBP3) in chemosensitivity and chemosensitivity cells and tissues of gastric cancer, and to explore its function and mechanism of abnormal expression in the chemoresistance of gastric cancer. Methods: The expression of IGFBP3 in chemotherapeutic drug-sensitive gastric cancer cells AZ521 and SC-M1, expressions of cisplatin-resistant cells AZ521/cisplatin, SC-M1/cisplatin and gastric cancer were detected by real-time quantitative fluorescence polymerase chain reaction(qRT-PCR) and Western blot assay(WB). After decreasing and increasing the expression of IGFBP3, the sensitivity of gastric cancer cells to chemotherapeutic drugs was detected by cell counting kit-8(CCK-8)reagent and flow cytometry(FCM). The promoter methylation level of IGFBP3 was determined by the methylation specific PCR(MSP)assay. Results: The expression of IGFBP3 in chemo-tolerant gastric cancer cells and tissues was lower than that in chemo-sensitive gastric cancer cells and tissues(P <0.05). Interference of IGFBP3 expression in sensitive cells can promote drug resistance of gastric cancer cells. While recovery of IGFBP3 expression in drug-resistant cells can significantly reversed drug resistance. MSP results showed that the expression of IGFBP3 was regulated by DNA methylation. Higher methylation of IGFBP3 promoter in drug-resistant cells resulted in decreased expression of IGFBP3(P <0.05). The drug-resistant gastric cancer cells were treatment with methyltransferase inhibitor dicetabine(DAC), the expression of IGFBP3 was restored, its sensitivity to chemotherapeutic drugs were increased(P<0.05).Conclusion: DNA hypermethylation in the promoter region of IGFBP3 leads to a decrease in its expression, which leads to a decrease in the ability of chemotherapeutic drugs to induce apoptosis of gastric cancer cells, and ultimately leads to chemotherapeutic resistance of gastric cancer cells. Objective: To investigate the effect of Gukang capsule combined with Sodium Hyaluronate Injection treatment of osteoarthritis, and its influence on inflammatory reaction stress. Methods: A total of 110 cases of osteoarthritis patients were selected as the research object, based on the random data table they were divided into control group and observation group, 55 cases in each group, patients in the control group were treated with Sodium Hyaluronate Injection treatment, observation group of patients were treated with Gukang capsule combined with Sodium Hyaluronate Injection treatment, the total effective rate, the clinical symptom score, the inflammatory factor and the incidence of adverse reaction after treatment were compared. Results: The total effective rate of treatment in the observation group was 90.91%, which was significantly higher than that of the control group(70.91%), and the difference was statistically significant(x2=7.308, P<0.05). There was no significant difference in Lysholm score, VAS score, TNF-α and CRP level between the two groups before treatment(P>0.05). The Lysholm score of the observation group was significantly higher than that of the control group.The VAS score, TNF-α and CRP were significantly lower than those in the control group, and the difference was statistically significant(P<0.05). There was no significant difference in the incidence of adverse drug reactions between the two groups(x2=0.121, P=0.728). Conclusion: Gukang capsule combined with Sodium Hyaluronate Injection, can effectively improve the osteoarthritis of clinical therapeutic effect, improve the patients clinical symptoms, reducing the inflammatory response to stress, high safety, which has a certain clinical value. Objective: To analyze the surgical approach and method for patients with 4 or more levels cervical spondylopathy with intervertebral instability. Methods: 64 eligible patients admitted from Nov 2008 to Dec 2012 were enrolled for the retrospective analysis. 33 cases were chosen as the traditional anterior cervical discectomy and fusion with plate fixation surgery (group A), another 31 patients were chosen as improved single door verterbral canal pressure-reducing shaft grafts Arch titanium plate fiation surgery (group B). After treatment, the clinical outcomes were assessed using Japanese Orthopedic Association (JOA) score andvisual analogue scale (VAS) score.The length of operation time, blood loss, hospital stays, range of motion of cervical spine, cervical spine curvature were also compared. Results: All the patients were operated successfully. The mean operation time of group B (90.8±22.2 min) was significantly shorter than that of group A (150.7±30.3 min), (P<0.05). The average amount of blood loss was less in group B (180±45 m L) than that of group A (320±50 m L, P<0.05). The hospitalization day of group B was significantly shorter than that of group A (P<0.05). The surgery incision of two groups were all healed at stage I. The JOA and VAS score were both significantly improved when discharging discharge from hospital compared with the preoperative scores of 2 groups (P<0.05). At 6 months after the surgery, there were significant differences of Ishihara Index between group A (improved 2.2±1.6) and group B (no difference improve) (P<0.05). There was no significant difference in the Cobb angle between Group A ( reduced 4.2±3.3°) and group B(reduced 4.0±2.9°) (P>0.05). Conclusions: Both the anterior and posterior cervical discectomy could obtain ideal clinical efficacy. However, the posterior way showed shortened operation time, reduced blood loss, shortened hospitalization length, improved security and broaden the operation indications, and could be a better surgery manner for the patients wirh 4 or more levels cervical spondylopathy with intervertebral instability. Objective: To observe the effect of H2O2 on the expression of UII/UT system in C2C12. Methods: Cell injury was determined by Placenta blue staining and LDH kit, the content of UII in cell lysate and cell incubation fluid were assayed by RIA, mRNA levels were assayed by RT-PCR, Protein levels were assayed by Western Blot. Results: UII content in cell incubation fluid was increased after H2O2 of different concentration was administrated, the UII content was increased by 83.1 % (P<0.01) and 94.5 % (P<0.01) when the concentration of H2O2 was elevated to 10-4M and 10-3M. The simulation of different concentration of H2O2(0.5×10-6M, 10-6M, 10-5M and 10-4M) significantly increased the expression of UT mRNA. However, The expression of UT mRNA was decreased significantly when the concentration of H2O2 was elevated to 10-3M. The UT protein expression was increased by 200.1%, 200.1%, 255.6% and 200.1% when the concentration of H2O2 was elevated to 10-6M, 10-5M, 10-4M and 10-3M(P<0.05). Objective: To study the efficacy of vertebroplasty in the treatment of elderly osteoporotic spinal fracture. Methods: A total of 120 elderly patients with senile osteoporotic spinal fracture were selected, who were admitted to West China Hospital of Sichuan University from January 2012 to January 2014, and treated by vertebroplasty with the Sky bone expander system through percutaneous puncture. 74 elderly patients took a prone position (prone position group), 46, the lateral position (lateral position group), comparing the vertebral height, Cobb angle, and VAS score of the two groups before operation and 3d and 3 months after operation. Results: There was statistical significance in the vertebral height between the two groups before operation (F=4.231, P=0.042), and the vertebral height of the two groups 3d and 3 months after operation was significantly higher than that before operation, with statistical significance (F=4.03, P=0.047).There was no statistical significance in the Cobb angle changes between the two groups before operation (F=3.042, P=0.072), but the mean Cobb angle of the two groups 3d and 3 months after operation was significantly lower than that before operation,with statistical significance (F=9.86, P=0.021). The VAS score in the two groups had no significant difference before operation (F=2.035, P=0.089), but the VAS score of the two groups 3d and 3 months after operation was significantly lower than that before operation,with statistical significance (F=12.86, P=0.005). Conclusion: Vertebroplasty has an obvious curative effect in the treatment of elderly osteoporotic spinal fracture, which can effectively restore the vertebral height and Cobb angle, and quickly relieve the symptoms of pain.
