Abstract

Sample preparation is key to the success of proteomics studies. In the present study, two sample preparation methods were tested for their suitability on the mature, recalcitrant leaves of six representative perennial plants (grape, plum, pear, peach, orange, and ramie). An improved sample preparation method was obtained: Tris and Triton X-100 were added together instead of CHAPS to the lysis buffer, and a 20% TCA-water solution and 100% precooled acetone were added after the protein extraction for the further purification of protein. This method effectively eliminates nonprotein impurities and obtains a clear two-dimensional gel electrophoresis array. The method facilitates the separation of high-molecular-weight proteins and increases the resolution of low-abundance proteins. This method provides a widely applicable and economically feasible technology for the proteomic study of the mature, recalcitrant leaves of perennial plants.

Abstract

The automatic generation of test cases oriented paths in an effective manner is a challenging problem for structural testing of software. The use of search-based optimization methods, such as genetic algorithms (GAs), has been proposed to handle this problem. This paper proposes an improved adaptive genetic algorithm (IAGA) for test cases generation by maintaining population diversity. It uses adaptive crossover rate and mutation rate in dynamic adjustment according to the differences between individual similarity and fitness values, which enhances the exploitation of searching global optimum. This novel approach is experimented and tested on a benchmark and six industrial programs. The experimental results confirm that the proposed method is efficient in generating test cases for path coverage.

Abstract

Leymus chinensis is a dominant, rhizomatous perennial C3 species in the grasslands of Songnen Plain of Northern China, and its productivity has decreased year by year. To determine how productivity of this species responds to different precipitation regimes, elevated CO2 and their interaction in future, we measured photosynthetic parameters, along with the accumulation and partitioning of biomass. Plants were subjected to combinations of three precipitation gradients (normal precipitation, versus normal ± 40%) and two CO2 levels (380±20 µmol mol-1,760±20 µmol mol-1) in controlled-environment chambers. The net photosynthetic rate, and above-ground and total biomass increased due to both elevated CO2 and increasing precipitation, but not significantly so when precipitation increased from the normal to high level under CO2 enrichment. Water use efficiency and the ratio of root: total biomass increased significantly when precipitation was low, but decreased when it was high under CO2 enrichment. Moreover, high precipitation at the elevated level of CO2 increased the ratio between stem biomass and total biomass. The effect of elevated CO2 on photosynthesis and biomass accumulation was higher at the low level of precipitation than with normal or high precipitation. The results suggest that at ambient CO2 levels, the net photosynthetic rate and biomass of L. chinensis increase with precipitation, but those measures are not further affected by additional precipitation when CO2 is elevated. Abstract

The negative air ion (NAI) concentration is an essential indicator of air quality and atmospheric pollution. The NAI concentration can be used to monitor air quality on a regional scale and is commonly determined using field measurements. However, obtaining these measurements is time-consuming. In this paper, the relationship between remotely sensed surface parameters (such as land surface temperature, normalized difference vegetation index (NDVI), and leaf area index) obtained from MODIS data products and the measured NAI concentration using a stepwise regression method was analyzed to estimate the spatial distribution of the NAI concentration and verify the precision. The results indicated that the NAI concentration had a negative correlation with temperature, leaf area index (LAI), and gross primary production while it exhibited a positive correlation with the NDVI. The relationship between land surface temperature and the NAI concentration in the Daxing'anling region is expressed by the regression equation of y = -35.51x1 + 11206.813 (R2 = 0.6123). Additionally, the NAI concentration in northwest regions with high forest coverage was higher than that in southeast regions with low forest coverage, suggesting that forests influence the air quality and reduce the impact of environmental pollution. The proposed inversion model is suitable for evaluating the air quality in Daxing'anling and provides a reference for air quality evaluation in other areas. In the future, we will expand the quantity and distribution range of sampling points, conduct continuous observations of NAI concentrations and environmental parameters in the research areas with different land-use types, and further improve the accuracy of inversion results to analyze the spatiotemporal dynamic changes in NAI concentration and explore the possibility of expanding the application areas of NAI monitoring.
