2. The Antinociceptive Properties of the Corydalis yanhusuo Extract.

Abstract

Corydalis yanhusuo. W.T. extracts (YHS) are widely used for the treatment of pain and inflammation. There are a few studies that assessed the effects of YHS in pain assays; however, none of these studies has systematically compared its activities in the different pain animal modes namely: acute, inflammatory and chronic pain. Furthermore, little is known about the mechanism of YHS activity in these assays. The aim of this study was to systematically evaluate the antinociceptive properties of YHS by testing it in four standardized pain assays and to investigate its mechanism. YHS antinociceptive properties were analyzed in the tail flick, the formalin paw licking, the von Frey filament and the hot box assays after spinal nerve ligation, which monitors acute nociceptive, persistent inflammatory and chronic neuropathic pain, respectively. YHS pharmacological profile was determined by screening it against a battery of G-protein coupled receptors and its mechanism of action was studied using knock-out mice. Our study shows that YHS, at a non-sedative dose, increases the tail flick latency in the tail flick assay without resulting in development of tolerance. YHS also decreases paw licking time in the formalin assay. Further, YHS increases paw withdraw threshold and latency in the von Frey filament and the hot box assays, respectively. In vitro, YHS exhibits prominent dopamine receptor antagonistic properties. In dopamine D2 receptor knockout mice, its antinociceptive effects are attenuated in acute and neuropathic pain but not inflammatory pain assays. Our results therefore indicate that YHS effectively attenuates acute, inflammatory and neuropathic pain, without causing tolerance. The effects on acute and neuropathic pain, but not inflammatory pain, are at least partially mediated through dopamine D2 receptor antagonism. Since YHS is a dietary supplement commercially available in the United States, our data suggest that it might be a candidate for alternative pain treatment.

Abstract

By screening extracts of venom from the Asian scorpion Buthus martensii Karsch (BmK) for their abilities to activate opioid receptors, we have identified BmK-YA, an amidated peptide containing an enkephalin-like sequence. BmK-YA is encoded by a precursor that displays a signal sequence and contains four copies of BmK-YA sequences and four of His4-BmK-YA, all flanked by single amino acid residues. BmK-YA and His4-BmK-YA are amidated and thus fulfill the characteristics expected of bioactive peptides. BmK-YA can activate mammalian opioid receptors with selectivity for the δ subtype while His4-BmK-YA is inactive at opioid receptors. The discovery of BmK-YA suggests that scorpion venom may represent a novel source of bioactive molecules targeting G protein-coupled receptors (GPCRs) and reveal additional insights on the evolution of the opioid precursors.

5. Improving wheat seedling quality through deep ploughing and soil compaction at sowing in lime concretion black soil.

Abstract

The straw incorporation in lime concretion black soil compromises the emergence and quality of winter wheat seedlings in Huaibei Plain, China, lowering the potential of wheat productivity. To overcome the disadvantage, a two-year field experiment was conducted in 2017–18 and 2018–19 to investigate the effects of different tillage modes on seedling emergence and subsequent seedling growth, and final grain yield (GY) in winter wheat. The modes are rotary tillage with compaction after sowing (RCT), rotary tillage after deep ploughing (PT) and rotary tillage after deep ploughing with compaction after sowing (PCT), with the traditional rotary tillage (RT) method as the control. Compared to RT, greater soil moisture content (SMC) at the seedling stage was observed in deep ploughing or compaction treatment, and the highest SMC was achieved in PCT; the time of reaching the maximum number of seedlings was 1 d sooner in RCT or PT, and 3 d in PCT; the seedling number in RCT, PT and PCT was significantly increased by 32.6%, 34.5% and 61.5% respectively. The population size, shoot and root growth of winter wheat in ploughing mode was significantly enhanced than that of rotary treatment at the over-wintering stage; compared to no compaction after sowing, plant growth in compaction treatments was significantly promoted with greater plant population size and height of seedlings. At harvest, GY in RCT, PT and PCT was significantly improved by 5.87%, 10.8% and 16.4%, respectively, compared to RT and the highest GY was achieved in PCT by up to 8, 350.1 kg ha-1 due to the increased spike number. In conclusion, the seedling quality in the straw incorporation practice was improved through rotary after deep ploughing and compaction after sowing for lime concretion black soil in Huaibei Plain, China or a similar soil type. Abstract

In shale, liquid hydrocarbons are accumulated mainly in nanometer-scale pores or fractures, so the pore types and PSDs (pore size distributions) play a major role in the shale oil occurrence (free or absorbed state), amount of oil, and flow features. The pore types and PSDs of marine shale have been well studied; however, research on lacustrine shale is rare, especially for shale in the oil generation window, although lacustrine shale is deposited widely around the world. To investigate the relationship between nanometer-scale pores and oil occurrence in the lacustrine shale, 10 lacustrine shale core samples from Songliao Basin, NE China were analyzed. Analyses of these samples included geochemical measurements, SEM (scanning electron microscope) observations, low pressure CO2 and N2 adsorption, and high-pressure mercury injection experiments. Analysis results indicate that: (1) Pore types in the lacustrine shale include inter-matrix pores, intergranular pores, organic matter pores, and dissolution pores, and these pores are dominated by mesopores and micropores; (2) There is no apparent correlation between pore volumes and clay content, however, a weak negative correlation is present between total pore volume and carbonate content; (3) Pores in lacustrine shale are well developed when the organic matter maturity (Ro) is >1.0% and the pore volume is positively correlated with the TOC (total organic carbon) content. The statistical results suggest that oil in lacustrine shale mainly occurs in pores with diameters larger than 40 nm. However, more research is needed to determine whether this minimum pore diameter for oil occurrence in lacustrine shale is widely applicable.
