Situational Judgment Tests (SJTs) are criterion valid low fidelity measures that have gained much popularity as predictors of job performance. A broad variety of SJTs have been studied, but SJTs measuring personality are still rare. Personality traits such as Conscientiousness are valid predictors of many educational, work and life-related outcomes and SJTs are less prone to faking than classical self-report measurements. We developed an SJT measure of Dependability, a core facet of Conscientiousness, by gathering critical incidents in semi-structured interviews using the construct definition of Dependability as a prompt. We examined the psychometric properties of the newly developed SJTs across two studies (N = 546 general population; N = 440 sales professionals). The internal validity of the SJTs was examined by correlating the SJT scores with related self-report measures of Dependability and Conscientiousness, as well as testing the unidimensionality of the measure with CFA. Additionally, we specified a bi-factor model of SJT, self-report and behavioral checklist measures of Dependability accounting for common and specific measurement variance. External validity was examined by correlating the SJT scale and specific factor with work-related outcomes. The results show that the Dependability SJTs with an expert based scoring procedure were psychometrically sound and correlated moderately to highly with traditional self-report measures of Dependability and Conscientiousness. However, a large proportion of SJT variance cannot be accounted for by personality alone. This supports the notion that SJTs measure general domain knowledge about the effectiveness of personality-related behaviors. We conclude that SJT measures of personality can be a promising addition to classical self-report assessments and can be used in a wide variety of applications beyond measurement and selection, for instance as formative assessments of personality., Competing Interests: The authors have declared that no competing interests exist.

Purpose: To estimate the prevalence of off-label and unlicensed prescribing during 2008 at a major paediatric teaching hospital in Western Australia., Methods: A 12-month retrospective study was conducted at Princess Margaret Hospital using medication chart records randomly selected from 145,550 patient encounters from the Emergency Department, Inpatient Wards and Outpatient Clinics. Patient and prescribing data were collected. Drugs were classified as off-label or unlicensed based on Australian registration data. A hierarchical system of age, indication, route of administration and dosage was used. Drugs were classified according to the Anatomical Therapeutic Chemical Code., Results: A total of 1,037 paediatric patients were selected where 2,654 prescriptions for 330 different drugs were prescribed to 699 patients (67.4%). Most off-label drugs (n = 295; 43.3%) were from the nervous system; a majority of unlicensed drugs were systemic hormonal preparations excluding sex hormones (n = 22, 32.4%). Inpatients were prescribed more off-label drugs than outpatients or Emergency Department patients (p < 0.0001). Most off-label prescribing occurred in infants and children (31.7% and 35.9% respectively) and the highest percentage of unlicensed prescribing (7.2%) occurred in infants (p < 0.0001). There were 25.7% of off-label and 2.6% of unlicensed medications prescribed across all three settings. Common reasons for off-label prescribing were dosage (47.4%) and age (43.2%)., Conclusion: This study confirmed off-label and unlicensed use of drugs remains common. Further, that prevalence of both is influenced by the clinical setting, which has implications in regards to medication misadventure, and the need to have systems in place to minimise medication errors. Further, there remains a need for changes in the regulatory system in Australia to ensure that manufacturers incorporate, as it becomes available, evidence regarding efficacy and safety of their drugs in children in the official product information.

Background: M-CSF recruits mononuclear phagocytes which regulate processes such as angiogenesis and metastases in tumors. VEGF is a potent activator of angiogenesis as it promotes endothelial cell proliferation and new blood vessel formation. Previously, we reported that in vitro M-CSF induces the expression of biologically-active VEGF from human monocytes., Methodology and Results: In this study, we demonstrate the molecular mechanism of M-CSF-induced VEGF production. Using a construct containing the VEGF promoter linked to a luciferase reporter, we found that a mutation reducing HIF binding to the VEGF promoter had no significant effect on luciferase production induced by M-CSF stimulation. Further analysis revealed that M-CSF induced VEGF through the MAPK/ERK signaling pathway via the transcription factor, Sp1. Thus, inhibition of either ERK or Sp1 suppressed M-CSF-induced VEGF at the mRNA and protein level. M-CSF also induced the nuclear localization of Sp1, which was blocked by ERK inhibition. Finally, mutating the Sp1 binding sites within the VEGF promoter or inhibiting ERK decreased VEGF promoter activity in M-CSF-treated human monocytes. To evaluate the biological significance of M-CSF induced VEGF production, we used an in vivo angiogenesis model to illustrate the ability of M-CSF to recruit mononuclear phagocytes, increase VEGF levels, and enhance angiogenesis. Importantly, the addition of a neutralizing VEGF antibody abolished M-CSF-induced blood vessel formation., Conclusion: These data delineate an ERK- and Sp1-dependent mechanism of M-CSF induced VEGF production and demonstrate for the first time the ability of M-CSF to induce angiogenesis via VEGF in vivo.
