Niphades castanea Chao (Coleoptera: Curculionidae), an important fruit insect pest of chestnuts (Castanea spp.), could cause chestnut involucre abscission ahead of time through larvae boring and feeding basal involucres, eventually causing huge economic losses. In this research, mitochondrial (COI and COII) and nuclear (ITS1) markers were used to investigate genetic variation among 15 different geographical populations of chestnut pest N castanea. The molecular diversity of N. castanea populations revealing three main phylogenetic clusters, with cluster I specifically distributed at high elevations in the western sampling points. Mitochondrial genes indicated population expansion events, and the ITS1 marker suggested a history of population expansion. Genetic diversity differentiation was significant among populations, indicating that geographical isolation impacts genetic differentiation among these places. AMOVA analyses confirmed substantial genetic differentiation between populations. Mantel correlogram analyses revealed a significant positive correlation between genetic differentiation and altitude/geographical distance at lower elevations and ranges, which reversed to a negative correlation at higher altitudes and ranges for all markers, indicating the role of altitude and geographical distance in shaping genetic diversity in N. castanea. This study contributes to a comprehensive understanding of the distribution, genetic diversity, and evolutionary history of N. castanea in the central of China, underscoring the impact of geographical factors on its genetic structure.

BackgroundCopper (Cu) homeostasis are important processes in the cause of metabolic diseases, but the association between Cu and obesity remains unclear.MethodsParticipants were drawn from the 2011-2016 National Health and Nutrition Examination Survey (NHANES). Weighted logistic regression assessed the associations of serum Cu concentrations (tertiles) with obesity and central obesity in individuals without comorbidities. Obesity was defined as a BMI ≥30.0 kg/m2, and central obesity was defined as a waist circumference ≥80 cm for women and ≥95 cm for men.ResultsThis cross-sectional study included 1,665 adults without comorbidities, representing 24,744,034 people (mean age 35.1 years, 48.5% female). High serum Cu levels (tertile 3: ≥19.19 μmol/L) were associated with higher odds of obesity (adjusted odds ratio [OR]: 4.48, 95% CI[confidence interval]: 2.44-8.32) and central obesity (OR: 2.36, 95% CI: 1.19-4.66) compared to low serum Cu levels (tertile 1: ≤15.64 μmol/L). The dose-response curve showed a nonlinear association between Cu levels and obesity (P-nonlinear = 0.02) and a linear association with central obesity (P-nonlinear = 0.21).ConclusionThis study suggests that higher serum Cu levels are associated with increased odds of obesity in healthy American adults.

Can the information technology revolution lead to carbon emission reduction for firms? This study extends the limited evidence in the literature and investigate the role and mechanism of digital inclusive finance on enterprises' carbon emissions using panel data of 247 prefectural-level cities and 6019 industrial enterprises in China. Our findings indicate that digital inclusive finance can promote enterprise carbon emission reduction, and this effect remains significant after the instrumental variable estimation test. The effect has regional heterogeneity and the development of digital inclusive finance in the area east of Hu Huanyong line has a significant impact on reducing enterprise carbon emission. The role of digital inclusive finance is heterogeneous in enterprise ownership, with a remarkable effect in non-state-owned enterprises. Sub-dimension analysis indicates that the breadth of coverage, depth of use, and degree of digitalization of digital inclusive finance have differential effects on reducing enterprise carbon emissions. The stepwise regression method shows that the impact of digital inclusive finance on enterprise carbon emissions can be passed through effect of technological progress, environmental protection investment and financing constrain. This study has significant reference value for evaluating the impact of financial inclusion and policy implications in formulating differentiated strategies for achieving carbon emission reduction efficiency in enterprises. BACKGROUND:The relationship between serum hemoglobin A1c (HbA1c) and atrial fibrillation (AF) or postoperative AF (POAF) in coronary artery bypass (CABG) patients is still under debate. It is also unclear whether there is a dose-response relationship between circulating HbA1c and the risk of AF or POAF. METHODS AND RESULTS:The Cochrane Library, PubMed, and EMBASE databases were searched. A robust-error meta-regression method was used to summarize the shape of the dose-response relationship. The RR and 95%CI were using a random-effects model. In total, 14 studies were included, totaling 17,914 AF cases among 352,325 participants. The summary RR per 1% increase in HbA1c was 1.16 (95% CI: 1.07-1.27). In the subgroup analysis, the summary RR was 1.13 (95% CI: 1.08-1.19) or 1.12 (95% CI: 1.05-1.20) for patients with diabetes or without known diabetes, respectively. The nonlinear analysis showed a nonlinear (Pnonlinear = 0.04) relationship between HbA1c and AF, with a significantly increased risk of AF if HbA1c was over 6.3%. However, HbA1c (per 1% increase) was not associated with POAF in patients with diabetes (RR: 1.13, P = 0.34) or without known diabetes (RR: 0.91, P = 0.37) among patients undergoing CABG. CONCLUSION:Our results suggest that higher HbA1c was associated with an increased risk of AF, both in diabetes and in without diabetes or with unknown diabetes. However, no association was found between HbA1c and POAF in patients undergoing CABG. Further prospective studies with larger population sizes are needed to explore the association between serum HbA1c level and the risk of POAF.

Poor visual conditions in tunnels can easily cause traffic accidents, and it is difficult for emergency services to reach these areas. As an economical and effective visual guiding device, delineators have attracted wide attention. Based on the actual alignment of the Qinling Mountain No.1, No.2 and No.3 tunnels of the G5 Expressway in Xi'an City (Shaanxi Province, China), this paper designs a simulation experiment. Through a simulator study and a questionnaire survey, this paper discusses how delineators affect drivers' visual characteristics (including fixation area and pupil size) in different settings and with different road alignments. Twenty-five subjects participated in this research. The results show that setting delineators in tunnels can continuously guide drivers' vision and attract their attention to focus on the pavement. Compared with setting only pavement delineators, setting wall delineators and pavement delineators together can provide better guiding effects and ensure driving safety in both straight and curved sections. In addition, when driving in tunnels equipped with delineators, especially tunnels with both wall delineators and pavement delineators, the participants exhibited a smaller pupil diameter and lower pupil diameter change rate. In terms of the relationship between pupil size and road alignment, the results indicated that regardless of what type of delineator was used, the drivers exhibited the smallest pupil size and lowest pupil change rate when driving on the straight section compared with the curved sections.

The overnight polysomnographic (PSG) recordings of patients were scored by an expert to diagnose sleep disorders. Visual sleep scoring is a time-consuming and subjective process. Automatic sleep staging methods can help; however, the mechanism and reliability of these methods are not fully understood. Therefore, experts often need to rescore the recordings to obtain reliable results. Here, we propose a human-computer collaborative sleep scoring system. It is a rule-based automatic sleep scoring method that follows the American Academy of Sleep Medicine (AASM) guidelines to perform an initial scoring. Then, the reliability level of each epoch is analyzed based on physiological patterns during sleep and the characteristics of various stage changes. Finally, experts would only need to rescore epochs with a low-reliability level. The experimental results show that the average agreement rate between our system and fully manual scorings can reach 90.42% with a kappa coefficient of 0.85. Over 50% of the manual scoring time can be reduced. Due to the demonstrated robustness and applicability, the proposed approach can be integrated with various PSG systems or automatic sleep scoring methods for sleep monitoring in clinical or homecare applications in the future.

This study aimed to study the diagnostic value of targeted next-generation sequencing (NGS) in limb-girdle muscular dystrophies (LGMDs), and investigate the mutational spectrum of Chinese LGMD patients. We performed targeted NGS covering 420 genes in 180 patients who were consecutively suspected of LGMDs and underwent muscle biopsies from January 2013 to May 2015. The association between genotype and myopathological profiles was analyzed in the genetically confirmed LGMD patients. With targeted NGS, one or more rare variants were detected in 138 patients, of whom 113 had causative mutations, 10 sporadic patients had one pathogenic heterozygous mutation related to a recessive pattern of LGMDs, and 15 had variants of uncertain significance. No disease-causing mutation was found in the remaining 42 patients. Combined with the myopathological findings, we achieved a positive genetic diagnostic rate as 68.3% (123/180). Totally 105 patients were diagnosed as LGMDs with genetic basis. Among these 105 patients, the most common subtypes were LGMD2B in 52 (49.5%), LGMD2A in 26 (24.8%) and LGMD 2D in eight (7.6%), followed by LGMD1B in seven (6.7%), LGMD1E in four (3.8%), LGMD2I in three (2.9%), and LGMD2E, 2F, 2H, 2K, 2L in one patient (1.0%), respectively. Although some characteristic pathological changes may suggest certain LGMD subtypes, both heterogeneous findings in a certain subtype and overlapping presentations among different subtypes were not uncommon. The application of NGS, together with thorough clinical and myopathological evaluation, can substantially improve the molecular diagnostic rate in LGMDs. Confirming the genetic diagnosis in LGMD patients can help improve our understanding of their myopathological changes.

The effects of cultivar mixture cropping on yield, biomass, and water use efficiency (WUE) in winter wheat (Triticum aestivum L.) were investigated under non-irrigation (W0, no irrigation during growth stage), one time irrigation (W1, irrigation applied at stem elongation) and two times irrigation (W2, irrigation applied at stem elongation and anthesis) conditions. Nearly 90% of cultivar mixture cropping treatments experienced an increase in grain yield as compared with the mean of the pure stands under W0, those for W1 and W2 were 80% and 85%, respectively. Over 75% of cultivar mixture cropping treatments got greater biomass than the mean of the pure stands under the three irrigation conditions. Cultivar mixture cropping cost more water than pure stands under W0 and W1, whereas the water consumption under W2 decreased by 5.9%-6.8% as compared with pure stands. Approximately 90% of cultivar mixtures showed an increase of 5.4%-34.5% in WUE as compared with the mean of the pure stands, and about 75% of cultivar mixtures had 0.8%-28.5% higher WUE than the better pure stands under W0. Similarly, there were a majority of mixture cropping treatments with higher WUE than the mean and the better one of the pure stands under W1 and W2. On the whole, proper cultivar mixture cropping could increase yield and WUE, and a higher increase in WUE occurred under limited irrigation condition.

Excessive N fertilization results in low N-use efficiency (NUE) without any yield benefits and can have profound, long-term environmental consequences including soil acidification, N leaching and increased production of greenhouse gases. Improving NUE in crop production has been a longstanding, worldwide challenge. A crucial strategy to improve NUE is to enhance N uptake by roots. Taking maize as a model crop, we have compared root dry weight (RDW), root/shoot biomass ratio (R/S), and NUE of maize grown in the field in China and in western countries using data from 106 studies published since 1959. Detailed analysis revealed that the differences in the RDW and R/S of maize at silking in China and the western countries were not derived from variations in climate, geography, and stress factors. Instead, NUE was positively correlated with R/S and RDW; R/S and NUE of maize varieties grown in western countries were significantly greater than those grown in China. We then testified this conclusion by conducting field trials with representative maize hybrids in China (ZD958 and XY335) and the US (P32D79). We found that US P32D79 had a better root architecture for increased N uptake and removed more mineral N than Chinese cultivars from the 0-60 cm soil profile. Reported data and our field results demonstrate that a large and deep root, with an appropriate architecture and higher stress tolerance (higher plant density, drought and N deficiency), underlies high NUE in maize production. We recommend breeding for these traits to reduce the N-fertilizer use and thus N-leaching in maize production and paying more attention to increase tolerance to stresses in China.

Myocardial infarction leads to heart failure. Autophagy is excessively activated in myocardial ischemia/reperfusion (I/R) in rats. The aim of this study is to investigate whether the protection of sevoflurane postconditioning (SPC) in myocardial I/R is through restored impaired autophagic flux.Except for the sham control (SHAM) group, each rat underwent 30 min occlusion of the left anterior descending coronary (LAD) followed by 2 h reperfusion. Cardiac infarction was determined by 2,3,5-triphenyltetrazolium chloride triazole (TTC) staining. Cardiac function was examined by hemodynamics and echocardiography. The activation of autophagy was evaluated by autophagosome accumulation, LC3 conversion and p62 degradation. Potential molecular mechanisms were investigated by immunoblotting, real-time PCR and immunofluorescence staining.SPC improved the hemodynamic parameters, cardiac dysfunction, histopathological and ultrastructural damages, and decreased myocardial infarction size after myocardial I/R injury (P < 0.05 vs. I/R group). Compared with the cases in I/R group, myocardial ATP and NAD+ content, mitochondrial function related genes and proteins, and the expressions of SOD2 and HO-1 were increased, while the expressions of ROS and Vimentin were decreased in the SPC group (P < 0.05 vs. I/R group). SPC significantly activated Akt/mTOR signaling, and inhibited the formation of Vps34/Beclin1 complex via increasing expression of Bcl2 protein (P < 0.05 vs. I/R group). SPC suppressed elevated expressions of LC3 II/I ratio, Beclin1, Atg5 and Atg7 in I/R rat, which indicated that SPC inhibited over-activation of autophagy, and promoted autophagosome clearance. Meanwhile, SPC significantly suppressed the decline of Opa1 and increases of Drp1 and Parkin induced by I/R injury (P < 0.05 vs. I/R group). Moreover, SPC maintained the contents of ATP by reducing impaired mitochondria.SPC protects rat hearts against I/R injury via ameliorating mitochondrial impairment, oxidative stress and rescuing autophagic clearance.

Oxidative stress plays an important role in neuronal injuries caused by cerebral ischemia. It is well established that free iron increases significantly during ischemia and is responsible for oxidative damage in the brain. However, the mechanism of this ischemia-induced increase in iron is not completely understood. In this report, the middle cerebral artery occlusion (MCAO) rat model was performed and the mechanism of iron accumulation in cerebral ischemia-reperfusion was studied. The expression of L-ferritin was significantly increased in the cerebral cortex, hippocampus, and striatum on the ischemic side, whereas H-ferritin was reduced in the striatum and increased in the cerebral cortex and hippocampus. The expression level of the iron-export protein ferroportin1 (FPN1) significantly decreased, while the expression of transferrin receptor 1 (TfR1) was increased. In order to elucidate the mechanisms of FPN1 regulation, we studied the expression of the key regulator of FPN1, hepcidin. We observed that the hepcidin level was significantly elevated in the ischemic side of the brain. Knockdown hepcidin repressed the increasing of L-ferritin and decreasing of FPN1 invoked by ischemia-reperfusion. The results indicate that hepcidin is an important contributor to iron overload in cerebral ischemia. Furthermore, our results demonstrated that the levels of hypoxia-inducible factor-1α (HIF-1α) were significantly higher in the cerebral cortex, hippocampus and striatum on the ischemic side; therefore, the HIF-1α-mediated TfR1 expression may be another contributor to the iron overload in the ischemia-reperfusion brain.
