Medical procedures that produce aerosolized particles are under great scrutiny due to the recent concerns surrounding the COVID-19 virus and increased risk for nosocomial infections. For example, thoracostomies, tracheotomies and intubations/extubations produce aerosols that can linger in the air. The lingering time is dependent on particle size where, e.g., 500 μm (0.5 mm) particles may quickly fall to the floor, while 1 μm particles may float for extended lengths of time. Here, a method is presented to characterize the size of 600 μm particles resulting from surgery in an operating room (OR). The particles are measured in-situ (next to a patient on an operating table) through a 75mm aperture in a ∼400 mm rectangular enclosure with minimal flow restriction. The particles and gasses exiting a patient are vented through an enclosed laser sheet while a camera captures images of the side-scattered light from the entrained particles. A similar optical configuration was described by Anfinrud et al.; however, we present here an extended method which provides a calibration method for determining particle size. The use of a laser sheet with side-scattered light provides a large FOV and bright image of the particles; however, the particle image dilation caused by scattering does not allow direct measurement of particle size. The calibration routine presented here is accomplished by measuring fixed particle distribution ranges with a calibrated shadow imaging system and mapping these measurements to the in-situ imaging system. The technique used for generating and measuring these particles is described. The result is a three-part process where 1) particles of varying sizes are produced and measured using a calibrated, high-resolution shadow imaging method, 2) the same particle generators are measured with the in-situ imaging system, and 3) a correlation mapping is made between the (dilated) laser image size and the measured particle size. Additionally, experimental and operational details of the imaging system are described such as requirements for the enclosure volume, light management, air filtration and control of various laser reflections. Details related to the OR environment and requirements for achieving close proximity to a patient are discussed as well.

Brain aggregates of β amyloid (βA) protein plaques have been widely recognized as associated with many neurodegenerative diseases, and their identification can assist in the early diagnosis of Alzheimer's disease. We investigate the feasibility of using a spectral x-ray coherent scatter system with a silicon strip photon-counting detector for identifying brain βA protein plaques. This approach is based on differences in the structure of amyloid, white and grey matter in the brain. We simulated an energy- and angular-dispersive X-ray diffraction system with an x-ray pencil beam and Silicon strip sensor, energy-resolving detectors. The polychromatic beam is geometrically focused toward a region of interest in the brain. First, the open-source MC-GPU code for Monte Carlo transport was modified to accommodate the detector model. Second, brain phantoms with and without βA were simulated to assess the method and determine the radiation dose required to obtain acceptable statistical power. For βA targets of 3, 4 and 5 mm sizes in a 15-cm brain model, the required incident exposure was about 0.44 mR from a 60 kVp tungsten spectrum and 3.5 mm of added aluminum filtration. The results suggest that the proposed x-ray coherent scatter technique enables the use of high energy x-ray spectra and therefore has the potential to be used for accurate in vivo detection and quantification of βA in the brain within acceptable radiation dose levels.

Cargo containers constitute the most critical component of global trade: 108 million containers represent the movement of about 95% of the world's manufactured goods. The steady increase in cargo container shipments has had a profound effect on world security: the threat associated with smuggling of shielded special nuclear material is elevated every year. Containers reaching the borders of the U.S. are currently not radiographically inspected due to time and dose considerations stemming from the use of bremsstrahlung beams for imaging. Bremsstrahlung spectra are low-energy peaked, resulting in low penetration values, especially through dense cargoes. The use of monoenergetic radiography beams could alleviate many of these problems due to higher energy and low background continuum. Using Monte Carlo simulations of a realistic imaging scenario with support from previous experimental measurements, we demonstrate how the use of monoenergetic photon beams in radiography can simultaneously reduce the radiation dose imparted to the cargo and any potential stowaways while increasing image quality. Dual-energy methods are leveraged to calculate material atomic number. Image quality is evaluated by measuring the noise standard deviation, contrast-to-noise ratio, and the pixel error as the dose is decreased.

Terahertz (THz) imaging is a widely used technique in the study and detection of many chemicals and biomolecules in polycrystalline form because the spectral absorption signatures of these target materials often lie in the THz frequencies. When the size of dielectric grain boundaries are comparable to the THz wavelengths, spectral features can be obscured due to electromagnetic scattering. In this study, we first investigate this granular scattering effect in identification of chemicals with THz spectral absorption features. We then will propose a signal processing technique in the so-called "quefrency" domain to improve the ability to resolve these spectral features in the diffuse scattered THz images. We created a pellet with α-lactose monohydrate and riboflavin, two biologically significant materials with well-known vibrational spectral resonances, and buried the pellet in a highly scattering medium. THz transmission measurements were taken at all angles covering the half focal plane. We show that, while spectral features of lactose and riboflavin cannot be distinguished in the scattered image, application of cepstrum filtering can mitigate these scattering effects. By employing our quefrency-domain signal processing technique, we were able to unambiguously detect the dielectric resonance of lactose in the diffused scattering geometries. Finally we will discuss the limitation of the new proposed technique in spectral identification of chemicals.

In this paper, a novel perturbation approach for implementing the independently reconfigurable dual-mode dual-band substrate integrated waveguide (SIW) filter is proposed. Dual-frequency manipulation is achieved by adding perturbation via-holes (the first variable) and changing the lengths of the interference slot (the second variable) in each cavity. The independent control of the upper passband only depends on the second variable while the lower passband is independently tuned by combining the two variables. Using such a design method, a two-cavity dual-band SIW filter is designed and experimentally assessed with four via-holes and an interference slot in each cavity. The dual-band filter not only has a frequency ratio (fR) ranging from 1.14 to 1.58 but also can be considered as a single passband one with a tunable range of 40.5% from 1.26 GHz to 2.12 GHz. The scattering parameters |S11| and |S21| are in the range of -10.72 dB to -37.17 dB and -3.67 dB to -7.22 dB in the operating dual bands, respectively. All the simulated and measured results show an acceptable agreement with the predicted data.

Imaging of $^{95m}$Tc radioisotope was conducted using an electron tracking-Compton camera (ETCC). $^{95m}$Tc emits 204, 582, and 835 keV $\gamma$ rays, and was produced in the $^{95}$Mo(p,n)$^{95m}$Tc reaction with a $^{95}$Mo-enriched target. The recycling of the $^{95}$Mo-enriched molybdenum trioxide was investigated, and the recycled yield of $^{95}$Mo was achieved to be 70\% - 90\%. The images were obtained with each of the three $\gamma$ rays. Results showed that the spatial resolution increases with increasing $\gamma$-ray energy, and suggested that the ETCC with high-energy $\gamma$-ray emitters such as $^{95m}$Tc is useful for the medical imaging of deep tissue and organs in the human body., 電子飛跡検出型コンプトンカメラ(ETCC)を用いて放射性同位元素$^{95m}$Tcの画像撮像を実施した。$^{95m}$Tcは、204, 582, 835keVの3本の$\gamma$線を放出し、濃縮同位体$^{95}$Moを用いて$^{95}$Mo(p,n)$^{95m}$Tc反応で合成される。濃縮$^{95}$Mo同位体三酸化物の再利用について実験を実施し、再生率70$\sim$90\%を達成した。画像は3本の$\gamma$線それぞれを用いて解析し取得した。その結果、$\gamma$線エネルギーが高いほど空間分解能が向上することが判り、$^{95m}$Tcのような高エネルギー$\gamma$線放出核を利用することで、ETCCが人体の深部の組織や器官の医療画像撮像に有効であることを示唆する結果を得た。

The use of light diffraction for the microbiological diagnosis of bacterial colonies was a significant breakthrough with widespread implications for the food industry and clinical practice. We previously confirmed that optical sensors for bacterial colony light diffraction can be used for bacterial identification. This paper is focused on the novel perspectives of this method based on digital in-line holography (DIH), which is able to reconstruct the amplitude and phase properties of examined objects, as well as the amplitude and phase patterns of the optical field scattered/diffracted by the bacterial colony in any chosen observation plane behind the object from single digital hologram. Analysis of the amplitude and phase patterns inside a colony revealed its unique optical properties, which are associated with the internal structure and geometry of the bacterial colony. Moreover, on a computational level, it is possible to select the desired scattered/diffracted pattern within the entire observation volume that exhibits the largest amount of unique, differentiating bacterial features. These properties distinguish this method from the already proposed sensing techniques based on light diffraction/scattering of bacterial colonies. The reconstructed diffraction patterns have a similar spatial distribution as the recorded Fresnel patterns, previously applied for bacterial identification with over 98% accuracy, but they are characterized by both intensity and phase distributions. Our results using digital holography provide new optical discriminators of bacterial species revealed in one single step in form of new optical signatures of bacterial colonies: digital holograms, reconstructed amplitude and phase patterns, as well as diffraction patterns from all observation space, which exhibit species-dependent features. To the best of our knowledge, this is the first report on bacterial colony analysis via digital holography and our study represents an innovative approach to the subject.

Advances in light shaping techniques are leading to new tools for optical trapping and micromanipulation. For example, optical tweezers made from Laguerre-Gaussian or donut beams display an increased axial trap strength and can impart angular momentum to rotate a specimen. However, the application of donut beam optical tweezers to precision, biophysical measurements remains limited due to a lack of methods for calibrating such devices sufficiently. For instance, one notable complication, not present when trapping with a Gaussian beam, is that the polarization of the trap light can significantly affect the tweezers' strength as well as the location of the trap. In this article, we show how to precisely calibrate the axial trap strength as a function of height above the coverslip surface while accounting for focal shifts in the trap position arising from radiation pressure, mismatches in the index of refraction, and polarization induced intensity variations. This provides a foundation for implementing a donut beam optical tweezers capable of applying precise axial forces.

This study investigated the potential of a newly proposed scattering foil free (SFF) electron beam scanning technique for the treatment of skin cancer on the irregular patient surfaces using Monte Carlo (MC) simulation. After benchmarking of the MC simulations, we removed the scattering foil to generate SFF electron beams. Cylindrical and spherical phantoms with 1 cm boluses were generated and the target volume was defined from the surface to 5 mm depth. The SFF scanning technique with 6 MeV electrons was simulated using those phantoms. For comparison, volumetric modulated arc therapy (VMAT) plans were also generated with two full arcs and 6 MV photon beams. When the scanning resolution resulted in a larger separation between beams than the field size, the plan qualities were worsened. In the cylindrical phantom with a radius of 10 cm, the conformity indices, homogeneity indices and body mean doses of the SFF plans (scanning resolution = 1 degrees) vs. VMAT plans were 1.04 vs. 1.54, 1.10 vs. 1.12 and 5 Gy vs. 14 Gy, respectively. Those of the spherical phantom were 1.04 vs. 1.83, 1.08 vs. 1.09 and 7 Gy vs. 26 Gy, respectively. The proposed SFF plans showed superior dose distributions compared to the VMAT plans.

The possibility to observe microsecond dynamics at the sub-micron scale, opened by recent technological advances in fast camera sensors, will affect many biophysical studies based on particle tracking in optical microscopy. A main limiting factor for further development of fast video microscopy remains the illumination of the sample, which must deliver sufficient light to the camera to allow microsecond exposure times. Here we systematically compare the main illumination systems employed in holographic tracking microscopy, and we show that a superluminescent diode and a modulated laser diode perform the best in terms of image quality and acquisition speed, respectively. In particular, we show that the simple and inexpensive laser illumination enables less than 1 μs camera exposure time at high magnification on a large field of view without coherence image artifacts, together with a good hologram quality that allows nm-tracking of microscopic beads to be performed. This comparison of sources can guide in choosing the most efficient illumination system with respect to the specific application.

Optical imaging through biological samples is compromised by tissue scattering and currently various approaches aim to overcome this limitation. In this paper we demonstrate that an all optical technique, based on non-linear upconversion of infrared ultrashort laser pulses and on multiple view acquisition, allows the reduction of scattering effects in tomographic imaging. This technique, namely Time-Gated Optical Projection Tomography (TGOPT), is used to reconstruct three dimensionally the internal structure of adult zebrafish without staining or clearing agents. This method extends the use of Optical Projection Tomography to optically diffusive samples yielding reconstructions with reduced artifacts, increased contrast and improved resolution with respect to those obtained with non-gated techniques. The paper shows that TGOPT is particularly suited for imaging the skeletal system and nervous structures of adult zebrafish.

We report on the construction of a Fourier plane imaging system attached to a cell phone. By illuminating particle suspensions with a collimated beam from an inexpensive diode laser, angularly resolved scattering patterns are imaged by the phone's camera. Analyzing these patterns with Mie theory results in predictions of size distributions of the particles in suspension. Despite using consumer grade electronics, we extracted size distributions of sphere suspensions with better than 20 nm accuracy in determining the mean size. We also show results from milk, yeast, and blood cells. Performing these measurements on a portable device presents opportunities for field-testing of food quality, process monitoring, and medical diagnosis.

Pathogen reduction is a viable approach to ensure the continued safety of the blood supply against emerging pathogens. However, the currently licensed pathogen reduction techniques are ineffective against non-enveloped viruses such as hepatitis A virus, and they introduce chemicals with concerns of side effects which prevent their widespread use. In this report, we demonstrate the inactivation of both enveloped and non-enveloped viruses in human plasma using a novel chemical-free method, a visible ultrashort pulsed laser. We found that laser treatment resulted in 2-log, 1-log, and 3-log reductions in human immunodeficiency virus, hepatitis A virus, and murine cytomegalovirus in human plasma, respectively. Laser-treated plasma showed ≥70% retention for most coagulation factors tested. Furthermore, laser treatment did not alter the structure of a model coagulation factor, fibrinogen. Ultrashort pulsed lasers are a promising new method for chemical-free, broad-spectrum pathogen reduction in human plasma.

We theoretically and experimentally analyze the formation of thick Purple Membrane (PM) polyacrylamide (PA) films by means of optical spectroscopy by considering the absorption of bacteriorhodopsin and scattering. We have applied semiclassical quantum mechanical techniques for the calculation of absorption spectra by taking into account the Fano effects on the ground state of bacteriorhodopsin. A model of the formation of PM-polyacrylamide films has been proposed based on the growth of polymeric chains around purple membrane. Experimentally, the temporal evolution of the polymerization process of acrylamide has been studied as function of the pH solution, obtaining a good correspondence to the proposed model. Thus, due to the formation of intermediate bacteriorhodopsin-doped nanogel, by controlling the polymerization process, an alternative methodology for the synthesis of bacteriorhodopsin-doped nanogels can be provided.

Here we report label-free optical imaging of single particles of the influenza virus attached on a glass surface with a simple objective-type total internal reflection dark-field microscopy (TIRDFM). The capability of TIRDFM for the imaging of single viral particles was confirmed from fine correlation of the TIRDFM images with fluorescent immunostaining image and scanning electron microscopy image. The density of scattering spots in the TIRDFM images showed a good linearity against the virus concentration, giving the limit of detection as 1.2×10(4) plaque-forming units per milliliter. Our label-free optical imaging method does require neither elaborated sample preparation nor complex optical systems, offering a good platform for rapid and sensitive counting of viral particles.
