Chilling sensitivity is one of the greatest challenges affecting the marketability and profitability of sweet basil (Ocimum basilicum L.) in the US and worldwide. Currently, there are no sweet basils commercially available with significant chilling tolerance and traditional aroma profiles. This study was conducted to identify quantitative trait loci (QTLs) responsible for chilling tolerance and aroma compounds in a biparental mapping population, including the Rutgers advanced breeding line that served as a chilling tolerant parent, 'CB15', the chilling sensitive parent, 'Rutgers Obsession DMR' and 200 F2 individuals. Chilling tolerance was assessed by percent necrosis using machine learning and aroma profiling was evaluated using gas chromatography (GC) mass spectrometry (MS). Single nucleotide polymorphism (SNP) markers were generated from genomic sequences derived from double digestion restriction-site associated DNA sequencing (ddRADseq) and converted to genotype data using a reference genome alignment. A genetic linkage map was constructed and five statistically significant QTLs were identified in response to chilling temperatures with possible interactions between QTLs. The QTL on LG24 (qCH24) demonstrated the largest effect for chilling response and was significant in all three replicates. No QTLs were identified for linalool, as the population did not segregate sufficiently to detect this trait. Two significant QTLs were identified for estragole (also known as methyl chavicol) with only qEST1 on LG1 being significant in the multiple-QTL model (MQM). QEUC26 was identified as a significant QTL for eucalyptol (also known as 1,8-cineole) on LG26. These QTLs may represent key mechanisms for chilling tolerance and aroma in basil, providing critical knowledge for future investigation of these phenotypic traits and molecular breeding.

To counterbalance the growing human population and its increasing demands from the ecosystem, and the impacts on it, new strategies are needed. Use of organic fertilizers boosted the agricultural production, but further increased the ecological burden posed by this indispensable activity. One possible solution to this conundrum is the development and application of more environmentally neutral biofertilizers. The aim of this study was to compare the effectiveness of two doses of Hermetia illucens frass (HI frass) with the commercial cattle manure in the cultivation of basil under drought. Soil without the addition of any organic fertilizer was used as a baseline control substrate for basil cultivation. Plants were grown with cattle manure (10 g/L of the pot volume) or HI frass at two doses (10 and 12.5 g/L). The health and physiological condition of plants were assessed based on the photosynthetic activity and the efficiency of photosystem II (chlorophyll fluorescence). Gas exchange between soil and the atmosphere were also assessed to verify the effect of fertilizer on soil condition. In addition, the mineral profile of basil and its antioxidant activity were assessed, along with the determination of the main polyphenolic compounds content. Biofertilizers improved the fresh mass yield and physiological condition of plants, both under optimal watering and drought, in comparison with the non-fertilized controls. Use of cattle manure in both water regimes resulted in a comparably lower yield and a stronger physiological response to drought. As a result, using HI frass is a superior strategy to boost output and reduce the effects of drought on basil production.

Salinity is among the most important abiotic stresses, which negatively affect growth, nutrient uptake and yield of crop plants. Application of different micronutrients, particularly zinc (Zn) have the potential to ameliorate the negative impacts of salinity stress. However, the role of Zn in improving salinity tolerance of basil (Ocimum basilicum L.) is poorly understood. This study evaluated the impact of different Zn levels (0, 5 and 10 mg kg-1) on growth and nutrient acquisition traits of basil under different salinity levels (0, 0.5, 1.0 and 1.5% NaCl). Data relating to biomass production, chlorophyll index, sodium (Na), potassium (K) uptake, K/Na ratio, Zn, copper (Cu), manganese (Mn) and iron (Fe) uptake were recorded. Increasing salinity level reduced biomass production, chlorophyll index and nutrient uptake traits (except for Na and Fe accumulation) of basil. Zinc application (10 mg kg-1) improved biomass production, chlorophyll index and nutrient acquisition traits under normal as well as saline conditions. The reduction in chlorophyll index and biomass production was higher under 0 and 5 mg kg-1 than 10 mg kg-1 Zn application. The K concentration decreased under increasing salinity; however, Zn application improved K uptake under normal as well as saline conditions. Different growth and nutrient acquisition traits had negative correlations with Na accumulation; however, no positive correlation was recorded among growth and nutrient uptake traits. The results revealed that Zn application could improve the salinity tolerance of basil. However, actual biochemical and genetic mechanisms involved in Zn-induced salinity tolerance warrant further investigation.

Iodine biofortification has been gaining interest in recent years as a sustainable and innovative approach to eradicate iodine deficiency disorders. Studying the impact of iodine biofortification on plant phenotype, biochemical and physiological parameters is crucial to leverage the expertise and best practices for the agro-food industry and human health. The aim of this study was to evaluate iodine biofortification on the main quantitative and qualitative traits of basil (Ocimum basilicum L.) plants cultivated both in open field and in growth chamber. The impact of KI and KIO3 treatments was evaluated on biomass production, as well as on the synthesis of phenolic compounds, especially rosmarinic acid and other caffeic acid derivatives, and on the essential oil (EO) composition. These compounds are typically accumulated in basil leaves and strongly contribute to the plant nutraceutical value and aroma. In open field, the use of increasing concentrations of both iodine salts gradually enhanced iodine accumulation in leaves, also determining an increase of the antioxidant power, total phenolics, rosmarinic acid and cinnamic acid accumulation. The composition of EO was only slightly affected by the treatments, as all the samples were characterized by a linalool chemotype and a minor alteration in their relative content was observed. A growth chamber experiment was performed to test EO variation in controlled conditions, broadening the range of iodine concentrations. In this case, plant chemotype was significantly affected by the treatments and large EO variability was observed, suggesting that iodine form and concentration can potentially influence the EO composition but that in open field this effect is overcome by environmental factors.

There is an increasing interest in alternatives to peat in growing media due to environmental constraints. However, plants grown in peat substitutes often show impaired growth compared to plants grown in peat-based media. Hence, it would be interesting to know whether these deficiencies can be compensated by supplementing other growth factors, e.g. light. The present study aims to investigate the interactive nature between growing media and supplemental lighting on plant growth and morphology, and to examine whether supplemental light emitting diode (LED) lighting may compensate adverse growing media effects. Basil (Ocimum basilicum L.) and Chinese cabbage (Brassica rapa subsp. pekinensis) were grown in different growing media consisting of peat, green compost, coconut pulp, wood fibre, perlite and sphagnum moss under blue, red and far-red supplemental LED lighting. We found significant interactions between growing media and supplemental photosynthetically active radiation (PAR) on plant growth, morphology and development. At low light intensities, peat-based and substituted growing media performed similarly, whereas with increasing light intensities the peat-based growing media significantly outperformed their alternatives. The substrate choice determines the required amount of supplemental light to compensate for adverse growing media effects and the amount varies depending on plant species and season. Thereby, it was indicated that red light alleviates adverse growing media effects best. We also found that far-red light is not effective when background PAR is low and becomes more effective under high background PAR. The implications and prospects of the results are discussed. With the increasing demand for non-chemical weed control methods, phenolic acids have shown promise due to their natural weed inhibitory potential. In this study, the inhibitory effect of ferulic acid, vanillic acid and p-coumaric acid was investigated on Ambrosia artemisiifolia L. and the selectivity of Zea mays L. against these phenolic acids was tested. The seeds of A. artemisiifolia and Z. mays were treated in vitro with three phenolic acids at doses of 200–600 × 10−7 mol and in vivo foliar on A. artemisiifolia and Z. mays plants. While all phenolic acids had effects on the early growth of A. artemisiifolia, p-coumaric acid significantly reduced the length of radicle and hypocotyl by more than 60% while the effects on Z. mays were minimal. In vivo assessments using chlorophyll fluorescence and multispectral imaging showed selective stress responses in A. artemisiifolia but not in Z. mays after foliar application. The in vitro results show that p-coumaric acid is a promising compound for the control of A. artemisiifolia. However, these phenolic acids at these doses led to an insufficient reduction in photochemical efficiency. Therefore, these natural compounds need to be combined with other methods of weed control. Holy basil (Ocimum sanctum L.) and sweet basil (Ocimum basilicum L.) are the most commonly grown basil species in India for essential oil production and biosynthesis of potentially volatile and non-volatile phytomolecules with commercial significance. The aroma, flavor and pharmaceutical value of Ocimum species is a significance of its essential oil, which contains most of the monoterpenes and sesquiterpenes. A large number of plants have been studied for characterization and identification of terpene synthase genes, involved in terpenoids biosynthesis. The goal of this study is to discover and identify the putative functional terpene synthase genes in O. sanctum. HMMER search was performed by using a set of 13 well sequenced and annotated plant genomes including the newly sequenced genome of O. sanctum with Pfam-A database locally, using HMMER 3.0 hmmsearch for the two Pfam domains (PF01397 and PF03936). Using this search method 81 putative terpene synthases genes (OsaTPS) were identified in O. sanctum; the study further reveals 47 OsaTPS were putatively functional genes, 19 partial OsaTPS, and 15 OsaTPS as probably pseudogenes. All these identified OsaTPS genes were compared with other plant species, and phylogenetic analysis reveals the subfamily classification of OsaTPS in TPS-a, -b, -c, -e, -f and TPS-g subfamilies clusters. This genome-wide identification of OsaTPS genes, their phylogenetic analysis and secondary metabolite pathway mapping predictions together provide a comprehensive understanding of the TPS gene family in Ocimum sanctum and offer opportunities for the characterization and functional validation of numbers of terpene synthase genes. Haemaphysalis longicornis is a common Ixodida tick species found in temperate areas of Asian countries. An anti-tick assay was conducted on adult female H. longicornis ticks. Plant extract solutions were prepared at concentrations of 50, 25, and 10 mg/mL. Tick survival and mortality were assessed by counting the number of dead and live ticks at 24 h, 48 h, 72 h, and 96 h posttreatment. Out of 11 plant extracts screened, Artemisia judaica extract exhibited the highest potency with 100% mortality (5/5) at 48 h when applied at high and moderate concentrations (50 and 25 mg/mL). Similar results were observed at 96 h for the 10 mg/mL group compared to the untreated ticks. Cleome droserifolia extract demonstrated partial activity with 60% (3/5) and 20% (1/5) mortality at 96 h posttreatment at concentrations of 50 and 25 mg/mL, respectively. Forsskaolea tenacissima extract showed a weak effect with 100% tick mortality (5/5) only at the highest treatment concentration after 96 h. To confirm the activity of A. judaica, trial 2 was conducted. A. judaica demonstrated potency within 48 h in high dose and 72 h in moderate dose, with 100% mortality (15/15) at 96 h posttreatment compared to untreated ticks. The median lethal time 50 (LT50) values were 30.37 h for the high and 55.08 h for the moderate doses. Liquid chromatography‒mass spectrometry was performed on the most potent candidate (A. judaica) to identify its phytochemical components. The results revealed the presence of 9 compounds identified through manual annotation and 74 compounds from the Global Natural Products Social library. These compounds included terpenoids, steroids, phenylpropanoids, flavonoid glycosides, flavonoids, and benzenoids. Camphor was identified as the major component via both approaches. This study investigated the mitigating effects of spermidine on salinity-stressed yarrow plants (Achillea millefolium L.), an economically important medicinal crop. Plants were treated with four salinity levels (0, 30, 60, 90 mM NaCl) and three spermidine concentrations (0, 1.5, 3 μM). Salinity induced electrolyte leakage in a dose-dependent manner, increasing from 22% at 30 mM to 56% at 90 mM NaCl without spermidine. However, 1.5 μM spermidine significantly reduced leakage across salinities by 1.35–11.2% relative to untreated stressed plants. Photosynthetic pigments (chlorophyll a, b, carotenoids) also exhibited salinity- and spermidine-modulated responses. While salinity decreased chlorophyll a, both spermidine concentrations increased chlorophyll b and carotenoids under most saline conditions. Salinity and spermidine synergistically elevated osmoprotectants proline and total carbohydrates, with 3 μM spermidine augmenting proline and carbohydrates up to 14.4% and 13.1% at 90 mM NaCl, respectively. Antioxidant enzymes CAT, POD and APX displayed complex regulation influenced by treatment factors. Moreover, salinity stress and spermidine also influenced the expression of linalool and pinene synthetase genes, with the highest expression levels observed under 90 mM salt stress and the application of 3 μM spermidine. The findings provide valuable insights into the responses of yarrow plants to salinity stress and highlight the potential of spermidine in mitigating the adverse effects of salinity stress. The present study recorded indigenous knowledge of medicinal plants in Shahrbabak, Iran. We described a method using data mining algorithms to predict medicinal plants' mode of application. Twenty-oneindividuals aged 28 to 81 were interviewed. Firstly, data were collected and analyzed based on quantitative indices such as the informant consensus factor (ICF), the cultural importance index (CI), and the relative frequency of citation (RFC). Secondly, the data was classified by support vector machines, J48 decision trees, neural networks, and logistic regression. So, 141 medicinal plants from 43 botanical families were documented. Lamiaceae, with 18 species, was the dominant family among plants, and plant leaves were most frequently used for medicinal purposes. The decoction was the most commonly used preparation method (56%), and therophytes were the most dominant (48.93%) among plants. Regarding the RFC index, the most important species are Adiantum capillus-veneris L. and Plantago ovata Forssk., while Artemisia auseri Boiss. ranked first based on the CI index. The ICF index demonstrated that metabolic disorders are the most common problems among plants in the Shahrbabak region. Finally, the J48 decision tree algorithm consistently outperforms other methods, achieving 95% accuracy in 10-fold cross-validation and 70–30 data split scenarios. Exploration of edible insects as sustainable alternative nutrient-dense sources such as nutraceuticals have attracted more and more global attention recently. However, research on wood borer beetles have largely been overlooked. This study assessed the entomo-chemical properties of Titoceres jaspideus (Cerambycidae) and Passalus punctiger (Passalidae), which are widely consumed in many African countries, including Kenya. The crude protein content of the beetle larvae ranged between 27.5–39.8 mg BSA/g. In comparison with those of cereals, amino acids such as lysine (7.9–9.9 mg/g), methionine (0.48–0.64 mg/g) and threonine (2.31–2.55 mg/g) were considerably high in the larvae. Methyl-5Z,8Z,11Z,14Z-eicosatetraenoate and methyl-9Z-octadecenoate were the predominant polyunsaturated and monounsaturated fatty acids, respectively. High total phenols (>4.4 mg GAE/g), flavonoids (>3.6 mg QE/g) and anti-oxidative activities (>67%) were recorded for both larvae. This implies that increasing the consumption of wood-borer beetle larvae would positively impact the state of the natural environment and reduce the problem of malnutrition in the society. Thus, applying these strategies to develop insect food in a more familiar form can help to make insect-enriched foods more appealing to consumers, facilitating their widespread adoption as a sustainable and nutritious food source. Limited understanding of sweet basil (Ocimum basilicum L.) genetics and genome structure has reduced efficiency of breeding strategies. This is evidenced by the rapid, worldwide dissemination of basil downy mildew (Peronospora belbahrii) in the absence of resistant cultivars. In an effort to improve available genetic resources, expressed sequence tag simple sequence repeat (EST-SSR) and single nucleotide polymorphism (SNP) markers were developed and used to genotype the MRI x SB22 F2 mapping population, which segregates for response to downy mildew. SNP markers were generated from genomic sequences derived from double digestion restriction site associated DNA sequencing (ddRADseq). Disomic segregation was observed in both SNP and EST-SSR markers providing evidence of an O. basilicum allotetraploid genome structure and allowing for subsequent analysis of the mapping population as a diploid intercross. A dense linkage map was constructed using 42 EST-SSR and 1,847 SNP markers spanning 3,030.9 cM. Multiple quantitative trait loci (QTL) model (MQM) analysis identified three QTL that explained 37–55% of phenotypic variance associated with downy mildew response across three environments. A single major QTL, dm11.1 explained 21–28% of phenotypic variance and demonstrated dominant gene action. Two minor QTL dm9.1 and dm14.1 explained 5–16% and 4–18% of phenotypic variance, respectively. Evidence is provided for an additive effect between the two minor QTL and the major QTL dm11.1 increasing downy mildew susceptibility. The commercial-scale production of Caralluma tuberculata faces significant challenges due to lower seed viability and sluggish rate of root growth in natural conditions. To overcome these obstacles, using phyto-mediated selenium nanomaterials as an in vitro rooting agent in plant in vitro cultures is a promising approach to facilitate rapid propagation and enhance the production of valuable therapeutic compounds. This study aimed to investigate the impact of phytosynthesized selenium nanoparticles (SeNPs) on the morphological growth attributes, physiological status, and secondary metabolite fabrication in in vitro propagated Caralluma tuberculata. The results demonstrated that a lower dose of SeNPs (100 μg/L) along with plant growth regulators (IBA 1 mg/L) had an affirmative effect on growth parameters and promoted earliest root initiation (4.6±0.98 days), highest rooting frequency (68.21±5.12%), number of roots (6.3±1.8), maximum fresh weight (710±6.01 mg) and dry weight (549.89±6.77 mg). However, higher levels of SeNPs (200 and 400 μg/L) in the growth media proved detrimental to growth and development. Further, stress caused by SeNPs at 100 μg/L along with PGRs (IBA 1 mg/L) produced a higher level of total chlorophyll contents (32.66± 4.36 μg/ml), while cultures exposed to 200 μg/L SeNPs alone exhibited the maximum amount of proline contents (10.5± 1.32 μg/ml). Interestingly, exposure to 400 μg/L SeNPs induced a stress response in the cultures, leading to increased levels of total phenolic content (3.4 ± 0.052), total flavonoid content (1.8 ± 0.034), and antioxidant activity 82 ± 4.8%). Furthermore, the combination of 100 μg/L SeNPs and plant growth regulators (1 mg/L IBA) led to accelerated enzymatic antioxidant activities, including superoxide dismutase (SOD = 4.4 ± 0.067 U/mg), peroxidase dismutase (POD = 3.3 ± 0.043 U/mg), catalase (CAT = 2.8 ± 0.048 U/mg), and ascorbate peroxidase (APx = 1.6 ± 0.082 U/mg). Essential oil (EO) distillation units, which are commonly installed on farms, have difficultly accessing knowledge centers. The apparent simplicity of the process hides unwanted losses and deviations that go undetected and, therefore, not corrected. This article proposes improvements to the process that are based on "4.0" technologies in order to detect and correct two important deficiencies, with an impact on the yield, quality and environmental performance. The first deficiency comprises the steam channels that are formed through green mass (channeling), are well known and are considered normal by EO producers. Without detection and correction, this negatively affects the extraction results. The second is the lack of technology that is able to automatically determine the extraction endpoint. Smart sensing, control, self-configuration and the dynamic determination of improved process parameters make up a set of actions undertaken by a smart extraction plant (50-liter capacity). Nineteen experiments using lemongrass (Cymbopogon citratus) exhibited remarkable 24% and 10% improvements in the yield and quality, respectively. Energy consumption and a more than 50% reduction in the processing complete the set of results achieved. Mean daily temperature (MDT) and daily light integral (DLI) can interact to influence growth and development of plants. Our objectives were to determine 1) the extent DLI and MDT influence growth and development of purple basil 'Dark Opal' (Ocimum basilicum), sage 'Extrakta' (Salvia officinalis), spearmint 'Spanish' (Mentha spicata), and sweet basil 'Nufar' (Ocimum basilicum) and 2) the influence on purple basil color. Young plants were transplanted into hydroponic systems in five greenhouse compartments with MDT set points of 23, 26, 29, 32, or 35°C and DLIs from 5 to 19 mol·m‒2·d‒1, respectively. At harvest, growth, development, and leaf color was measured. Branch number of all genera increased as MDT increased from ~23 to 35°C. Sweet basil branch number increased as DLI increased from 5.5 to 13.2 mol·m‒2·d‒1, but the effect of DLI was attenuated as MDT decreased. In contrast, increasing DLI from ~5–6 to ~18–19 mol·m‒2·d‒1 increased sage and spearmint branch number more when MDT was lower (~23°C) compared to ~35°C, while branch number of purple basil was not influenced by DLI. The optimal MDT (MDTopt) for sage and spearmint fresh mass decreased from 27.5 to 23.5°C and from 30.4 to 27.8°C, respectively, as DLI increased from 6 to 18 mol·m‒2·d‒1, while sweet basil fresh mass MDTopt increased from 32.6 to 35.5°C as DLI increased from 6 to 11 mol·m‒2·d‒1. Purple basil was greener [hue angle (h°) = 99° to 138°] when MDT was ~35°C regardless of DLI, but when MDT was lower (~25°C), basil was more purple (h° = 335°) at a DLI of 18.7 compared to 5.0 mol·m‒2·d‒1 (h° = 98°). In sugarcane breeding, dense experiments have been considered in the initial phase (T1), such as the Simplified System (SS) of genotype selection. In this method, the seedlings of each family are transplanted directly from the seed box to the field, forming a kind of carpet. Despite the practical aspect of the method, selection problems are common, as stalks from the same individual within the family are subject to being taken to later evaluation stages, to the detriment of stalks from different individuals. To facilitate the discrimination of stalks of the same family in SS, we evaluated using RGB images (red:green:blue) and NIR (near infrared) spectra. We applied Euclidean distance (D) and Mahalanobis distance (D2) dissimilarity measures to the image and spectral data to distinguish stalks with different genotypes. RGB and NIR data were taken from type +1 leaf samples collected from two experimental blocks, totaling 31 evaluated families. The analyzes were carried out in two stages. In the first stage, we sought to evaluate the classification capacity using RGB images and NIR spectra, using D as a measure of dissimilarity. In the second step, we developed and validated a protocol using RGB images to classify clones, with D2 as a dissimilarity measure. Preliminary results, with distance D, allowed to discriminate clones based on the distance of the evaluated attributes and their combinations. In addition, with the analyzes using the D distance, it was identified that only the use of the R attribute (red band) would give satisfactory results for the second stage, which was the proposed analysis protocol, applying the D2 distance. Green synthesis of nanoparticles is becoming a method of choice for biological research due to its environmentally benign outcomes, stability and ease of synthesis. In this study, silver nanoparticles (AgNPs) were synthesized using stem (S-AgNPs), root (R-AgNPs) and mixture of stem and root (RS-AgNPs) of Delphinium uncinatum. The synthesized nanoparticles were characterized by standardized techniques and evaluated for their antioxidant, enzyme inhibition, cytotoxic and antimicrobial potentials. The AgNPs exhibited efficient antioxidant activities and considerable enzyme inhibition potential against alpha amylase, acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) enzymes. S-AgNPs showed strong cytotoxicity against human hepato-cellular carcinoma cells (HepG2) and high enzyme inhibitory effect (IC50 values 27.5μg/ml for AChE and 22.60 μg/ml for BChE) compared to R-AgNPs and RS-AgNPs. RS-AgNPs showed significant inhibition of Klebsiella pneumoniae and Aspergillus flavus and exhibited higher biocompatibility (<2% hemolysis) in human red blood cells hemolytic assays. Medicinal plants contain valuable compounds that have attracted worldwide interest for their use in the production of natural drugs. The presence of compounds such as rosmarinic acid, carnosic acid, and carnosol in Rosmarinus officinalis has made it a plant with unique therapeutic effects. The identification and regulation of the biosynthetic pathways and genes will enable the large-scale production of these compounds. Hence, we studied the correlation between the genes involved in biosynthesis of the secondary metabolites in R. officinalis using proteomics and metabolomics data by WGCNA. We identified three modules as having the highest potential for the metabolite engineering. Moreover, the hub genes highly connected to particular modules, TFs, PKs, and transporters were identified. The TFs of MYB, C3H, HB, and C2H2 were the most likely candidates associated with the target metabolic pathways. The results indicated that the hub genes including Copalyl diphosphate synthase (CDS), Phenylalanine ammonia lyase (PAL), Cineole synthase (CIN), Rosmarinic acid synthase (RAS), Tyrosine aminotransferase (TAT), Cinnamate 4-hydroxylase (C4H), and MYB58 are responsible for biosynthesis of important secondary metabolites. Thus, we confirmed these results using qRT-PCR after treating R. officinalis seedlings with methyl jasmonate. Background: Due to interactions with variety of environmental and physicochemical factors, the composition and diversity of bacteria and fungi in manure ectopic fermentation are constantly changing. The purpose of this study was to investigated bacterial and fungal changes in dairy manure ectopic fermentation, as well as the relationships between keystone species and physicochemical characteristics. Methods: Ectopic fermentation was carried out for 93 days using mattress materials, which was combined with rice husk and rice chaff (6:4, v/v), and dairy waste mixed with manure and sewage. Physicochemical characteristics (moisture content, pH, NH4+-N (NN), total organic carbon (TO), total nitrogen (TN) and the C/N ratio) of ectopic fermentation samples were measured, as well as enzymatic activity (cellulose, urease, dehydrogenase and alkaline phosphatase). Furthermore, the bacterial and fungal communities were studied using 16S rRNA and 18S rRNA gene sequencing, as well as network properties and keystone species were analyzed. Results: During the ectopic fermentation, the main pathogenic bacteria reduced while fecal coliform increased. The C/N ratio gradually decreased, whereas cellulase and dehydrogenase remained at lower levels beyond day 65, indicating fermentation maturity and stability. During fermentation, the dominant phyla were Chloroflexi, Firmicutes, Proteobacteria, Bacteroidetes, and Actinobacteria of bacteria, and Ascomycota of fungi, while bacterial and fungal community diversity changed dramatically and inversely. The association between physicochemical characteristics and community keystone taxon was examined, and C/N ratio was negative associated to keystone genus. Conclusion: These data indicated that microbial composition and diversity interacted with fermentation environment and parameters, while regulation of keystone species management of physicochemical factors might lead to improved maturation rate and quality during dairy manure ectopic fermentation. In recent years, the interest on baby-leaf lettuce has grown steadily, because it is richer in bioactive compounds than other traditional vegetables. However, the quality of lettuce is being increasingly affected by climate change. It is very rare for a climatic effect to occur in isolation. Even then, a large body of work has only focused on the effect of isolated heat stress, fertilization, and elevated CO2, on morphological, physiological and biochemical parameters. Thus, very few works have focused on how the combination of several of these factors can affect these parameters. For first time, the present work studied the combined effect derived from the application of two different levels of CO2 (400 and 1000 ppm of CO2), four different NO3-/ NH4+ ratios (100/0 (T-I), 100/0 before the short-term heat stress and finally without NO3- (T-II), 80/20 (T-III) and 50/50 (T-IV)), and a short-term heat stress (25 and 43°C), on some physiological and quality parameters (dry biomass, photosynthetic parameters, pigments content, lipid peroxidation and total soluble proteins content) of baby-leaf lettuce cv Derbi. Additionally, a comparison of that combined effect of all these parameters between inner and outer leaves was also performed. The results obtained indicated that the interaction between the nutrient solution containing a 50/50 NO3-/ NH4+ ratio and a high concentration of CO2 (1000 ppm) improved the biomass, photosynthesis, intercellular/external CO2 concentration ratio (ci/ca), stomatal conductance (gs), evapotranspiration (E) and lipid peroxidation, and protein content in this baby-leaf lettuce. The results obtained in this work lead us to conclude that this existing interaction between the NO3-/ NH4+ ratio and the elevated CO2 concentration may be considered as a new strategy for making baby-leaf lettuce more resistant to heat stress, in other words, stronger against the ever more frequent heat waves. There is an increasing interest in alternatives to peat in growing media due to environmental constraints. However, plants grown in peat substitutes often show impaired growth compared to plants grown in peat-based media. Hence, it would be interesting to know whether these deficiencies can be compensated by supplementing other growth factors, e.g. light. The present study aims to investigate the interactive nature between growing media and supplemental lighting on plant growth and morphology, and to examine whether supplemental light emitting diode (LED) lighting may compensate adverse growing media effects. Basil (Ocimum basilicum L.) and Chinese cabbage (Brassica rapa subsp. pekinensis) were grown in different growing media consisting of peat, green compost, coconut pulp, wood fibre, perlite and sphagnum moss under blue, red and far-red supplemental LED lighting. We found significant interactions between growing media and supplemental photosynthetically active radiation (PAR) on plant growth, morphology and development. At low light intensities, peat-based and substituted growing media performed similarly, whereas with increasing light intensities the peat-based growing media significantly outperformed their alternatives. The substrate choice determines the required amount of supplemental light to compensate for adverse growing media effects and the amount varies depending on plant species and season. Thereby, it was indicated that red light alleviates adverse growing media effects best. We also found that far-red light is not effective when background PAR is low and becomes more effective under high background PAR. The implications and prospects of the results are discussed.

Radiation intensity and carbon dioxide (CO2) concentration can be precisely controlled to manipulate plant yield and quality. Due to increased plant densities during seedling production, fewer inputs per plant are required, creating the potential to increase production efficiency. Therefore, the objectives of this research were to: 1) quantify the extent radiation intensity and CO2 concentration under sole-source lighting influence morphology and yield of sweet basil (Ocimum basilicum) seedlings, and 2) determine if differences in morphology, yield, and volatile organic compound (VOC) concentration persist after transplant in a common environment. Sweet basil 'Nufar' seedlings were grown in growth chambers with target CO2 concentrations of 500 or 1,000 μmol·mol‒1 under light-emitting diodes (LEDs) providing target photosynthetic photon flux densities (PPFD) of 100, 200, 400, or 600 μmol·m‒2·s‒1 for 16 h per day. After two weeks, seedlings were transplanted into a common greenhouse environment and grown until harvest. At transplant and three weeks after transplant (harvest), growth and developmental differences were quantified along with key terpenoid and phenylpropanoid concentrations at harvest. Radiation intensity and CO2 interacted influencing many aspects of plant morphology, though CO2 concentration effects were less pronounced than those of radiation intensity. As radiation intensity during seedling production increased from 100 to 600 μmol·m‒2·s‒1, basil seedlings were 38% taller, had a 713% larger leaf area, and had 65% thicker stems; at harvest, plants were 24% taller, had 56% more branches, 28% more nodes, 22% thicker stems, and weighed 80% more when fresh and dry. Additionally, after growing in a common environment for three weeks, eugenol concentration was greater in plants grown under a PPFD of 600 μmol·m‒2·s‒1 as seedlings compared to lower intensities. Therefore, increasing radiation intensity during seedling production under sole-source lighting can carry over to increase subsequent yield and eugenol concentration during finished production. The preservative effect of the addition of different essential oils (copaiba and oregano) on meat quality parameters and sensorial acceptability was analyzed for fresh ground beef patties over 21 days of display. Five treatments were assessed: control (CON) without antioxidants; addition of the synthetic additive butylated hydroxytoluene (BHT); addition 0.05% of copaiba essential oil (CEO); 0.05% of oregano essential oil (OEO); or blend of 0.025% copaiba and 0.025% oregano essential oils (BEO). The lowest cooking losses and greatest tenderness (P <0.05) were reached with the blend (BEO). The inclusion of oregano essential oil presented a more intense chroma (P <0.05), with the best color retained during display. Oregano essential oil (OEO) and the blend (BEO) showed the highest antioxidant activity, reducing the lipid oxidation of beef patties during display (P < 0.05). Consumers preferred the odor of beef patties with essential oils (OEO and BEO) to the CON; however, the flavor from OEO had the lowest acceptability and the worst scores for overall acceptability (P < 0.05). Patties with the blend addition (BEO) were the best scored on overall acceptability assessments. In conclusion, the oregano and copaiba essential oils blend had a good preservative effect on fresh beef patties during display and increased sensory acceptability of the product, thus being a possible alternative for replacing synthetic compounds in processed foods. Researchers are increasingly looking to plants as sources of novel ingredients active against vector-borne diseases. Medicinal plant extracts and their metabolites are an attractive source for such products. This study investigated the insecticidal activity of five essential oils extracted from the most common medicinal herbs in Jazan province, Kingdom of Saudi Arabia. Extracted oils and nanoemulsions synthesized from those oils were characterized before application at different concentrations to laboratory-reared fourth-stage larvae of Culex pipens. Basil (Ocimum bascilicum) and cumin (Cuminum cyminum) essential oils showed moderate larvicidal effect with LC50 81.07 ug/mL and 96.29 ug/mL, respectively. That activity was improved in their nanoemulsion forms, as evidenced by a reduction in the LC50 to 65.19 ug/mL for basil and 64.50 ug/mL for cumin. Clove (Syzygium aromaticum), henna (Lawsonia inermis) and ginger (Zingiber officinalis) oils showed weaker insecticidal activity, with LC50 values of 394 ug/mL, 306 ug/mL, and 494 ug/mL, respectively. Moreover, the nanoemulsion forms of those essential oils did not show any improvement in their insecticidal activity. Sweetpotato (Ipomoea batatas [L.] Lam) is a widely cultivated food crop with generally good adaptability. However, drought stress can cause a significant decline in yield. To reveal the response mechanism of sweetpotato to drought stress, an integrated physiological, proteomic and metabolomic investigation was conducted in leaves of two sweetpotato varieties with differing responses to drought stress, drought-resistant Wanzishu56 (WZ56) and a more sensitive variety, Ningzishu2(NZ2). Physiological analysis showed that the variety with better drought tolerance had superior performance in water retention capacity and photosynthetic efficiency under drought stress. A total of 1140 proteins were identified within the two varieties. Among them, 192 differentially expressed proteins were detected under drought conditions, including 97 that were up-regulated. Functional analysis showed that these up-regulated proteins were primarily involved in photosynthesis, reactive oxygen species metabolism, organonitrogen compound metabolism, and precursor metabolite catabolism and energy generation. All differentially expressed proteins in WZ56 that were involved in photosynthetic and glutathione metabolic processes were up-regulated. Enzyme activity assays were carried out to validate the proteomics data. Moreover, 75 metabolites were found to have a higher expression level in WZ56 than NZ2 under drought stress. The higher concentration of carbohydrates, amino acids, flavonoids and organic acids found in drought-stressed leaves of WZ56 suggested that these metabolites may improve the drought resistance of sweetpotato. This study uncovered specific-proteins and metabolites associated with drought resistance, providing new insights into the molecular mechanisms of drought tolerance in sweetpotato. Environmental abiotic stresses limit plant growth, development, and reproduction. This study aims to reveal the response of Brassica napus to salt stress. Here, transcriptomics, metabolomics, and proteomics analysis were performed on 15 Brassica napus leave samples treated with salt at different times. Through functional enrichment analyzing the differentially expressed genes (DEGs), differential metabolites (DMs) and differentially expressed proteins (DEPs), the key factors that dominate Brassica napus response to salt stress were identified. The results showed that the two key hormones responding to salt stress were Abscisic acid (ABA) and jasmonic acid (JA). Salt stress for 24h is an important milestone. Brassica napus adjusted multiple pathways at 24h to avoid over-response to salt stress and cause energy consumption. The increased expression in BnPP2C is tangible evidence. In response to salt stress, JA and ABA work together to reduce the damage caused by salt stress in Brassica napus. The increased expression of all BnJAZs after salt stress highlighted the function of JA that cannot be ignored responding to salt stress. In addition, some metabolites, such as N-acetyl-5-hydroxytryptamine, L-Cysteine and L-(+)-Arginine, play a critical role in maintaining the balance of ROS. Proteins like catalase-3, cysteine desulfurase, HSP90 and P450_97A3 were the most critical differential proteins in response to salt stress. Zingiber ottensii, is widely used in Asian traditional remedies for the treatment of many diseases. The present study explores anticancer activity of Z. ottensii essential oil (ZOEO) and its nanoformulations. ZOEO obtained from hydrodistillation of Z. ottensii fresh rhizomes was analysis using gas chromatography mass spectroscopy. Zerumbone (25.21%) was the major compound of ZOEO followed by sabinene (23.35%) and terpene-4-ol (15.97%). Four types of ZOEO loaded nanoformulations; nanoemulsion, microemulsion, nanoemulgels, and microemulgel, were developed. The average droplet size of the nanoemulsion and microemulsion was significantly smaller than that of the nanoemulgel and microemulgel. Comparison with other essential oils of plants of the same family on anticancer activity against A549, MCF-7, HeLa, and K562, ZOEO showed the highest cytotoxicity with IC50 of 43.37±6.69, 9.77±1.61, 23.25±7.73, and 60.49±9.41 μg/mL, respectively. Investigation using flow cytometry showed that ZOEO significantly increased the sub-G1 populations (cell death) in cell cycle analysis and induced cell apoptosis by apoptotic analysis. The developed nanoformulations significantly enhanced cytotoxicity of ZOEO, particularly against MCF-7 with the IC50 of 3.08±2.58, 0.74±0.45, 2.31±0.91, and 6.45±5.84 μg/mL, respectively. Among the four nanoformulations developed in the present study, nanoemulsion and microemulsion were superior to nanoemulgel and microemulgel in delivering ZOEO into cancer cells. Emergence of multidrug resistant pathogens is increasing globally at an alarming rate with a need to discover novel and effective methods to cope infections due to these pathogens. Green nanoparticles have gained attention to be used as efficient therapeutic agents because of their safety and reliability. In the present study, we prepared zinc oxide nanoparticles (ZnO NPs) from aqueous leaf extract of Acacia arabica. The nanoparticles produced were characterized through UV-Visible spectroscopy, scanning electron microscopy, and X-ray diffraction. In vitro antibacterial susceptibility testing against foodborne pathogens was done by agar well diffusion, growth kinetics and broth microdilution assays. Effect of ZnO NPs on biofilm formation (both qualitatively and quantitatively) and exopolysaccharide (EPS) production was also determined. Antioxidant potential of green synthesized nanoparticles was detected by DPPH radical scavenging assay. The cytotoxicity studies of nanoparticles were also performed against HeLa cell lines. The results revealed that diameter of zones of inhibition against foodborne pathogens was found to be 16–30 nm, whereas the values of MIC and MBC ranged between 31.25–62.5 μg/ml. Growth kinetics revealed nanoparticles bactericidal potential after 3 hours incubation at 2 × MIC for E. coli while for S. aureus and S. enterica reached after 2 hours of incubation at 2 × MIC, 4 × MIC, and 8 × MIC. 32.5–71.0% inhibition was observed for biofilm formation. Almost 50.6–65.1% (wet weight) and 44.6–57.8% (dry weight) of EPS production was decreased after treatment with sub-inhibitory concentrations of nanoparticles. Radical scavenging potential of nanoparticles increased in a dose dependent manner and value ranged from 19.25 to 73.15%. Whereas cytotoxicity studies revealed non-toxic nature of nanoparticles at the concentrations tested. Medicinal plants represent a great source of antimicrobial and phytochemical constituents which are increasingly used to treat microbial infections and other ailments such as tuberculosis, anemia, and trachoma. Despite the use of antibiotics, antimicrobial resistance continues to be a world issue, in as much as nutrition. This study investigated the presence of phytochemicals, proximate compositions, and antimicrobial activity of methanolic extract of Carapa procera bark. The bark of Carapa procera was collected, cleaned and air dried for 72 h. The powder obtained was treated with diethyl ether and soaked in methanol (99%) for 72 h to obtain crude extract. The extract was used to test for the presence of phytochemicals and antimicrobial activities. The raw bark was used for proximate analysis. The result showed presence of steroids, tannins and saponins, but no alkaloids present. The 100 mg/mL extract had the highest inhibition zone on all tested organisms from 24.00 ± 0.94 to 26.67 ± 1.18, and 50 mg/mL showed the least (16.67 ± 1.24) on Candida albicans. Staphylococcus aureus showed the lowest minimum inhibition concentration (MIC) of 3.12 mg/mL, whereas the Gram-negative bacteria exhibited variations in their sensitivity with E. coli having the highest MIC of 25 mg/mL. The extract had high MIC (6.25 mg/mL) on Candida albicans than clotrimazole (50 mg/mL). The proximate compositions of Carapa procera were moisture (6.07 ± 0.07%), ash (12.46 ± 0.46%), crude protein (9.54 ± 0.12%), crude fat (1.42 ± 0.06%) and carbohydrate (70.50 ± 0.35%). The energy value was 1413.17 kj. Thus, Carapa procera possesses both antimicrobial and nutritional potentials worth exploring and domesticating for sustainable management and conservation. Background: Cystic echinococcosis caused by the cestode Echinococcus granulosus remains a serious helminthic zoonosis affecting humans and animals in many endemic developing countries. Surgical intervention is the best management choice, although it is associated with high recurrence rates and serious complications. Also, the commonly used chemotherapeutics exhibited serious side effects. This study aimed to evaluate the protoscolicidal effects of eugenol (Eug) essential oil and its nanoemulsion (Eug-NE) against protoscoleces (PCs) of hydatid cysts in vitro. Methods: Eug-NE was prepared and characterized. Their cytotoxicity on macrophages was assessed by the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. E. granulosus PCs were treated with various concentrations of Eug and Eug-NE at different exposure times. The viability of protoscoleces was evaluated by the eosin exclusion test, and the changes in the morphology of protoscoleces were assessed. Albendazole (ABZ) was used as a positive control. Results: The cellular cytotoxicity of Eug and Eug-NE on macrophage cells, in minimum and maximum concentrations (0.2 and 1 μl/mL), were nearly negligible ranging from 4.7% to 8.3% and 3.7% to 7.2%, respectively. The results showed highly significant activity of Eug-NE and Eug against hydatid PCs compared to ABZ (P < 0.05). Eug and Eug-NE have similar protoscolicidal effects at all used concentrations. Their highest scolicidal activity (100% mortality rate) was recorded at 1 μl/ml after 30 min incubation (LC50 = 0.298—LC90 = 0.521 and LC50 = 0.309—LC90 = 0.646, respectively). Both formulations showed time- and dose-dependent effects. Conclusions: This study suggested the potent scolicidal activities of Eug and Eug-NE as promising alternative scolicidal agents. Conventional chemotherapeutic agents for colorectal cancer (CRC) cause systemic side effects and eventually become less efficacious owing to the development of drug resistance in cancer cells. Therefore, new therapeutic regimens have focused on the use of natural products. The anticancer activity of several parts of Calotropis gigantea has been reported; however, the effects of its stem bark extract on inhibition of cancer cell proliferation have not yet been examined. In this study, the anticancer activity of C. gigantea stem bark extract, both alone and in combination with 5-fluorouracil (5-FU), was evaluated. A crude ethanolic extract was prepared from dry, powdered C. gigantea barks using 95% ethanol. This was then partitioned to obtain dichloromethane (CGDCM), ethyl acetate, and water fractions. Quantitative analysis of the constituent secondary metabolites and calotropin was performed. These fractions exhibited cytotoxicity in HCT116 and HT-29 cells, with CGDCM showing the highest potency in both the cell lines. A combination of CGDCM and 5-FU significantly enhanced the cytotoxic effect. Moreover, the resistance of normal fibroblast, HFF-1, cells to this combination demonstrated its safety in normal cells. The combination significantly enhanced apoptosis through the mitochondria-dependent pathway. Additionally, the combination reduced adenosine triphosphate production and increased the production of reactive oxygen species, demonstrating the mechanisms involved in the induction of apoptosis. Our results suggest that CGDCM is a promising anti-cancer agent and may enhance apoptosis induction by 5-FU in the treatment of CRC, while minimizing toxicity toward healthy cells. Passiflora quadrangularis L. belongs to the family Passifloraceae which bears larger fruit with edible juicy mesocarp and pulp known as a good source of phytochemicals. Cultivation and plant management practices are known to influence the phytochemical compositions of agricultural produce. This study aimed to examine the influence of the cultivation practices on the antioxidant activities and secondary metabolites of the organically and conventionally grown P. quadrangularis. Findings revealed organically treated P. quadrangularis plants showed enhancement in their antioxidant properties and secondary metabolites profiles. Among the plant parts, leaves of P. quadrangularis grown organically possessed higher antioxidant activities compared to the conventional in all assays evaluated. The antioxidant activities in the edible parts of the P. quadrangularis fruit have also been enhanced through organic cultivation with significantly higher total phenolic content and DPPH in mesocarp, and the pulp showed higher total flavonoid content, DPPH and FRAP. This observation is supported by a higher level of vitamins and secondary metabolites in the samples. The secondary metabolites profile showed mesocarps were phenolic rich, the pulps were flavonoids rich while leaves showed good composition of phenolics, flavonoids and terpenoids with outstanding antioxidant activities. The common secondary metabolites for organically produced P. quadrangularis in different plant parts include 2-isopropyl-3-methoxycinnamic acid (mesocarp and pulp), myricetin isomers (pulp and leaves), and malvidin-3-O-arabinoside isomers (pulp and leaves). Symbiotic associations with endophytic fungi are ecologically important for medicinal and aromatic plants. Endophytic fungi highly affect the quantity and quality of herbal products. In this study, a pot experiment was carried out in the greenhouse to investigate the interactive effects of Piriformospora indica and arbuscular mycorrhizal (AMF) inoculation on the chlorophyll fluorescence, essential oil composition, and antioxidant enzymes of peppermint under saline condition. The results showed that Fo, YNPQ, YNO, and NPQ values were obviously increased under salinity conditions, while essential oil content, chlorophyll a and b, gs, Fm, Fv, ETR, ФPSII and Fv/Fm ratio decreased by increasing salinity. In addition, salt induced the excess Na+ uptake, whereas the opposite trend was observed for P and K+. The synergistic association of P. indica and AMF caused a considerable increase in the antioxidant ability, essential oil content, Fv/Fm ratio, ФPSII, and amount of P and K+ uptake in salt-stressed plants. The main peppermint oil constituents, menthol, menthone, and 1,8-cineole increased considerably in inoculated plants. Sweet basil (Ocimum basilicum) is an economically important allotetraploid (2n = 4x = 48) herb whose global production is threatened by downy mildew disease caused by the obligate biotrophic oomycete, Peronospora belbahrii. Generation of disease resistant cultivars by mutagenesis of susceptibility (S) genes via CRISPR/Cas9 is currently one of the most promising strategies to maintain favored traits while improving disease resistance. Previous studies have identified Arabidopsis DMR6 (Downy Mildew Resistance 6) as an S gene required for pathogenesis of the downy mildew-causing oomycete pathogen Hyaloperonospora arabidopsidis. In this study, a sweet basil homolog of DMR6, designated ObDMR6, was identified in the popular sweet basil cultivar Genoveser and found to exist with a high copy number in the genome with polymorphisms among the variants. Two CRISPR/Cas9 constructs expressing one or two single guide RNAs (sgRNAs) targeting the conserved regions of ObDMR6 variants were generated and used to transform Genoveser via Agrobacterium-mediated transformation. 56 T0 lines were generated, and mutations of ObDMR6 were detected by analyzing the Sanger sequencing chromatograms of an ObDMR6 fragment using the Interference of CRISPR Edits (ICE) software. Among 54 lines containing mutations in the targeted sites, 13 had an indel percentage greater than 96% suggesting a near-complete knockout (KO) of ObDMR6. Three representative transgene-free lines with near-complete KO of ObDMR6 determined by ICE were identified in the T1 segregating populations derived from three independent T0 lines. The mutations were further confirmed using amplicon deep sequencing. Disease assays conducted on T2 seedlings of the above T1 lines showed a reduction in production of sporangia by 61–68% compared to the wild-type plants and 69–93% reduction in relative pathogen biomass determined by quantitative PCR (qPCR). This study not only has generated transgene-free sweet basil varieties with improved downy mildew resistance, but also contributed to our understanding of the molecular interactions of sweet basil-P. belbahrii. Vehicular emissions cause heavy metal pollution and exert negative impacts on environment and roadside vegetation. Wild plants growing along roadsides are capable of absorbing considerable amounts of heavy metals; thus, could be helpful in reducing heavy metal pollution. Therefore, current study inferred heavy metal absorbance capacity of some wild plant species growing along roadside. Four different wild plant species, i.e., Acacia nilotica L., Calotropis procera L., Ricinus communis L., and Ziziphus mauritiana L. were selected for the study. Leaf samples of these species were collected from four different sites, i.e., Control, New Lahore, Nawababad and Fatehabad. Leaf samples were analyzed to determine Pb2+, Zn2+, Ni2+, Mn2+ and Fe3+ accumulation. The A. nilotica, Z. mauritiana and C. procera accumulated significant amount of Pb at New Lahore site. Similarly, R. communis and A. nilotica accumulated higher amounts of Mn, Zn and Fe at Nawababad and New Lahore sites compared to the rest of the species. Nonetheless, Z. mauritiana accumulated higher amounts of Ni at all sites compared with the other species included in the study. Soil surface contributed towards the uptake of heavy metals in leaves; therefore, wild plant species should be grown near the roadsides to control heavy metals pollution. Results revealed that wild plants growing along roadsides accumulate significant amounts of heavy metals. The calcineurin inhibitors (CNI) cyclosporine A and tacrolimus comprise the basis of immunosuppressive regimes in all solid organ transplantation. However, long-term or high exposure to CNI leads to histological and functional renal damage (CNI-associated nephrotoxicity). In the kidney, proximal tubule cells are the only cells that metabolize CNI and these cells are believed to play a central role in the origin of the toxicity for this class of drugs, although the underlying mechanisms are not clear. Several studies have reported oxidative stress as an important mediator of CNI-associated nephrotoxicity in response to CNI exposure in different available proximal tubule cell models. However, former models often made use of supra-therapeutic levels of tissue drug exposure. In addition, they were not shown to express the relevant enzymes (e.g., CYP3A5) and transporters (e.g., P-glycoprotein) for the metabolism of CNI in human proximal tubule cells. Moreover, the used methods for detecting ROS were potentially prone to false positive results. In this study, we used a novel proximal tubule cell model established from human allograft biopsies that demonstrated functional expression of relevant enzymes and transporters for the disposition of CNI. We exposed these cells to CNI concentrations as found in tissue of stable solid organ transplant recipients with therapeutic blood concentrations. We measured the glutathione redox balance in this cell model by using organelle-targeted variants of roGFP2, a highly sensitive green fluorescent reporter protein that dynamically equilibrates with the glutathione redox couple through the action of endogenous glutaredoxins. Our findings provide evidence that CNI, at concentrations commonly found in allograft biopsies, do not alter the glutathione redox balance in mitochondria, peroxisomes, and the cytosol. However, at supra-therapeutic concentrations, cyclosporine A but not tacrolimus increases the ratio of oxidized/reduced glutathione in the mitochondria, suggestive of imbalances in the redox environment. Damask rose (Rosa damascena Mill.) is an aromatic medicinal plant rich in bioactive compounds with high value in the food, pharmaceutical and cosmetic industries. Knowledge of the factors affecting the quantitative and qualitative properties of the compounds in its essential oil (EO) and the bioactivity of this EO is important in optimizing Damask rose cultivation and production. This research studied, for the first time, the effects of irrigation water and soil chemical and physical characteristics on the EO yield of this important commercial species and on it chemical composition and antimicrobial activity. The results showed the significant effect of crop cultivation site on yield, chemical composition and inhibition zone diameter (IZD) at the 1% significance level. The highest EO yield (~0.0266%), which belonged to the Noushabad site (EON), resulted from the increased soil electrical conductivity (EC) and the higher sand, gypsum and lime contents and irrigation water salinity. Analysis of the chemical composition of the EOs showed that their main compounds at all three crop sites were citronellol, nonadecane, heneicosane and geraniol. The EO obtained from the Yazdel site (EOY) had the highest contents of citronellol and geraniol (~29.05% and ~6.85%) that were directly correlated with soil potassium and phosphorus contents and inversely correlated with soil acidity and EC and its lime, nitrogen, and organic carbon contents. Antimicrobial assays indicated that the EO extracted from the Sefidshahr site (EOS), which had the largest inhibition zone diameter (~14.67 mm) for Aspergillus brasiliensis (IZD~14.67 mm) and the lowest MIC (~31.25 μg/mL) for Staphylococcus aureus and Pseudomonas aeruginosa, exhibited efficacy similar to that of rifampin, probably due to the dominance of the alkanes in it. The EOY and EOS also exhibited the strongest inhibitory and lethal activity against Candida albicans (MIC and MBC <15.63 μg/mL for EOY and MIC and MBC = 62.5 μg/mL for EOS), which were six and four times stronger than those of nystatin, respectively. Renewed consumer demand motivates the nutritional and sensory quality improvement of fruits and vegetables. Specialized metabolites being largely involved in nutritional and sensory quality of carrot, a better knowledge of their phenotypic variability is required. A metabolomic approach was used to evaluate phenotypic plasticity level of carrot commercial varieties, over three years and a wide range of cropping environments spread over several geographical areas in France. Seven groups of metabolites have been quantified by HPLC or GC methods: sugars, carotenoids, terpenes, phenolic compounds, phenylpropanoids and polyacetylenes. A large variation in root metabolic profiles was observed, in relation with environment, variety and variety by environment interaction effects in decreasing order of importance. Our results show a clear diversity structuration based on metabolite content. Polyacetylenes, β-pinene and α-carotene were identified mostly as relatively stable varietal markers, exhibiting static stability. Nevertheless, environment effect was substantial for a large part of carrot metabolic profile and various levels of phenotypic plasticity were observed depending on metabolites and varieties. A strong difference of environmental sensitivity between varieties was observed for several compounds, particularly myristicin, 6MM and D-germacrene, known to be involved in responses to biotic and abiotic stress. This work provides useful information about plasticity in the perspective of carrot breeding and production. Cannabis sativa has been cultivated since antiquity as a source of fibre, food and medicine. The recent resurgence of C. sativa as a cash crop is mainly driven by the medicinal and therapeutic properties of its resin, which contains compounds that interact with the human endocannabinoid system. Compared to other medicinal crops of similar value, however, little is known about the biology of C. sativa. Glandular trichomes are small hair-like projections made up of stalk and head tissue and are responsible for the production of the resin in C. sativa. Trichome productivity, as determined by C. sativa resin yield and composition, is only beginning to be understood at the molecular level. In this study the proteomes of glandular trichome stalks and heads, were investigated and compared to the proteome of the whole flower tissue, to help further elucidate C. sativa glandular trichome biochemistry. The data suggested that the floral tissue acts as a major source of carbon and energy to the glandular trichome head sink tissue, supplying sugars which drive secondary metabolite biosynthesis. In controlled environments, crop models that incorporate environmental factors can be developed to optimize growth and development as well as conduct cost and/or resource use benefit analyses. The overall objective of this study was to model growth and development of dill 'Bouquet' (Anethum graveolens), parsley 'Giant of Italy' (Petroselinum crispum), and watercress (Nasturtium officinale) in response to photosynthetic daily light integral (DLI) and mean daily temperature (MDT). Plants were grown hydroponically in five greenhouse compartments with MDTs ranging from 9.7 to 27.2 °C under 0%, 30%, or 50% shade cloth to create DLIs ranging from 6.2 to 16.9 mol·m‒2·d‒1. MDT and DLI interacted to influence dill fresh mass and height, and watercress maximum quantum yield of dark adapted leaves (Fv/Fm), height, and branch number while only MDT affected dill leaf number and watercress fresh mass and branch length. Besides dry matter concentration (DMC), parsley was influenced by MDT and not DLI. Increasing MDT from ≈10 to 22.4 °C (parsley) or 27.2 °C (dill and watercress), linearly or near-linearly increased fresh mass. For dill, increasing DLI decreased fresh mass when MDT was low (9.7 to 13.9 °C) and increased fresh mass when MDT was high (18.4 to 27.2 °C). DMC of dill, parsley, and watercress increased as MDT decreased or DLI increased, indicating a higher proportion of plant fresh mass is water at higher MDTs or lower DLIs. We evaluated the effects of the inclusion of copaíba (Copaifera officinalis L.) essential oil at 0.05 and 0.1% as a possible replacement of synthetic additive butylated hydroxytoluene (BHT) in sheep burgers during 14 days of storage in a refrigerated display case (4°C). During the shelf life days, analyzes of antioxidant activity, lipid oxidation, pH, color, cooking loss, texture and consumer acceptability were carried out on refrigerated burgers. The addition of copaíba essential oil showed an antioxidant effect in sheep burgers (P > 0.05), reducing lipid oxidation. Copaiba essential oil added at 0.05% showed the highest antioxidant activity, decreased cooking losses and delaying discoloration (loss of redness) during storage; it further improved, the tenderness of sheep burgers (P < 0.05). The treatments had no effect (P > 0.05) on consumer acceptability. Copaiba essential oil is a promising natural antioxidant to increase the shelf life of meat products, as well as being a viable solution to replace synthetic antioxidant BHT, thereby promoting the fresh‐like quality appeal of sheep burgers. Anopheles stephensi with three different biotypes is a major vector of malaria in Asia. It breeds in a wide range of habitats. Therefore, safer and more sustainable methods are needed to control its immature stages rather than chemical pesticides. The larvicidal and antibacterial properties of the Pelargonium roseum essential oil (PREO) formulations were investigated against mysorensis and intermediate forms of An. stephensi in laboratory conditions. A series of nanoemulsions containing different amounts of PREO, equivalent to the calculated LC50 values for each An. stephensi form, and various quantities of surfactants and co-surfactants were developed. The physical and morphological properties of the most lethal formulations were also determined. PREO and its major components, i.e. citronellol (21.34%), L-menthone (6.41%), linalool (4.214%), and geraniol (2.19%), showed potent larvicidal activity against the studied mosquitoes. The LC50/90 values for mysorensis and intermediate forms were computed as 11.44/42.42 ppm and 12.55/47.69 ppm, respectively. The F48/F44 nanoformulations with 94% and 88% lethality for the mysorensis and intermediate forms were designated as optimized formulations. The droplet size, polydispersity index, and zeta-potential for F48/F44 were determined as 172.8/90.95 nm, 0.123/0.183, and -1.08/-2.08 mV, respectively. These results were also confirmed by TEM analysis. Prepared formulations displayed antibacterial activity against larval gut bacteria in the following order of decreasing inhibitory: LC90, optimized nanoemulsions, and LC50. PREO-based formulations were more effective against mysorensis than intermediate. Compared to the crude PREO, the overall larvicidal activity of all nanoformulations boosted by 20% and the optimized formulations by 50%. The sensitivity of insect gut bacteria may be a crucial factor in determining the outcome of the effect of toxins on target insects. The use of bio-fertilizers in agro-ecosystems is considered to have the potential to improve plant growth in extreme environments featuring water shortages. However, while arbuscular mycorrhizal fungi (AMF) and bacteria bio-fertilizers have been used in other plants to enhance stress tolerance, little is known about their symbiotic effect on sorghum (Sorghum bicolor L.) growth under drought stress conditions. Therefore the aim of this study was to investigate the inoculation of sorghum with Nitroxin and Glomus mosseae and their interaction effects on the agro-physiological characteristics and grain yield of sorghum under drought stress conditions. Nitroxin is a bio-fertilizer that consists of a mixture of Azospirillum and Azotobacter bacteria. The results showed that co-inoculation of sorghum seeds with Nitroxin and AMF improved the chlorophyll (a, b and total) content, soluble proteins, water use efficiency) WUE(, relative water content (RWC), nitrogen (N) content in the plant, AMF spore density, proline content, grain yield, panicle length, the number of panicles per plant, grain number per panicle, 1000-grain weight and decreased the electrolyte leakage and water saturation deficit (WSD) in drought stress and non-stress conditions. Under drought stress conditions, there was a 27% increase in grain yield under the synergistic effects of bacteria and fungi compared to the non-application of these microorganisms. The results of this experiment show that Nitroxin and AMF bio-fertilizers can mitigate the negative effects of stress on plants in drought stress conditions by increasing the amount of photosynthetic pigments, soluble proteins and osmotic regulation and decreasing electrolyte leakage. Asarum heterotropoides Fr. var. mandshuricum (Maxim) Kitag (Chinese wild ginger) is an important medicinal herb. Essential oil extracted from its roots is the key ingredient and is mainly composed of phenylpropanoid compounds. As a skiophyte plant, light is a crucial factor for A. heterotropoides var. mandshuricum growth and metabolism. To investigate the effects of light irradiation on the essential oil biosynthesis in A. heterotropoides var. mandshuricum, the plants were cultivated in four light irradiation treatments (100, 50, 24 and 12% full sunlight). The photosynthetic capacity, essential oil content and composition, activities of several enzymes and levels of some secondary metabolites involved in the shikimic acid and cinnamic acid pathways were analyzed. The leaf mass per area, average diurnal net photosynthetic rate, and the essential oil content increased significantly with increasing light intensity. Phenylalanine, cinnamic acid, and p-coumaric acid in the cinnamic acid pathway were at their highest levels in plants cultivated in 100% full sunlight. The highest content of shikimic acid in the shikimic acid pathway was obtained in plants grown in 50% sunlight transmittance. The activity of the enzymes 3-Deoxy-D-arabino-heptulosonate-7-phosphate synthase, phenylalanine ammonia lyase, cinnamate-4-hydroxylase and 4-coumarate:CoA ligase increased proportionally with light intensity. Overall, we conclude that high light irradiation promotes high net photosynthetic rate, high activity of enzymes and high amounts of phenylpropanoid precursor metabolites leading to significant biosynthesis of essential oil in A. heterotropoides var. mandshuricum.
