The prevalence of type 2 diabetes mellitus (T2DM) is increasing dramatically worldwide. Dysregulation of microRNA (miRNA) as key regulators of gene expression, has been reported in numerous diseases including diabetes. The aim of this study was to investigate the expression levels of miRNA-122, miRNA-126-3p and miRNA-146a in diabetic and pre-diabetic patients and in healthy individuals, and to determine whether the changes in the level of these miRNAs are reliable biomarkers in diagnosis, prognosis, and pathogenesis of T2DM. Additionally, we examined the relationship between miRNA levels and plasma concentrations of inflammatory factors including tumor necrosis factor alpha (TNF-α) and interleukin 6 (Il-6) as well as insulin resistance. In this case-control study, participants (n = 90) were allocated to three groups (n = 30/group): T2DM, pre-diabetes and healthy individuals as control (males and females, age: 25-65, body mass index: 25-35). Expression of miRNA was determined by real-time polymerase chain reaction (RT-PCR). Furthermore, plasma concentrations of TNF-α, IL-6 and fasting insulin were measured by enzyme-linked immunosorbent assay. Homeostatic model assessment for insulin resistance (HOMA-IR) was calculated as an indicator of insulin resistance. MiRNA-122 levels were higher while miRNA-126-3p and miRNA-146a levels were lower in T2DM and pre-diabetic patients compared to control (p<0.05). Furthermore, a positive correlation was found between miRNA-122 expression and TNF-α (r = 0.82), IL-6 (r = 0.83) and insulin resistance (r = 0.8). Conversely, negative correlations were observed between miRNA-126-3p and miRNA-146a levels and TNF-α (r = -0.7 and r = -0.82 respectively), IL-6 (r = -0.65 and r = -0.78 respectively) as well as insulin resistance (r = -0.67 and r = -0.78 respectively) (all p<0.05). Findings of this study suggest the miRNAs can potentially contribute to the pathogenesis of T2DM. Since in most mapping models geometric mean of different criteria are used to determine the desertification intensity, one of the most important issues in desertification studies is understanding the similar areas, which require similar management after determining the desertification intensity map. Two similar classes of desertification intensity may require different management due to differences in the criteria that affect its desertification severity. Therefore, after determining the geomorphological facies as the working units in Sistan plain, we used hierarchical cluster analysis to identify the homogeneous environmental management units (HEMUs) based on indices of MEDALUS model. According to the MEDALUS model, the studied area was divided into two categories namely medium and high desertification classes. Working units (geomorphological facies) are classified into five clusters according to HEMUs analysis based on climate, soil, vegetation, and wind erosion criteria. The first cluster (C11) include six facies with moderate and severe desertification; in all of these units the main effective factor was wind erosion, so they need the same management decisions controlling wind erosion. Two working units (1 and 4) with the same desertification severity were placed in two different clusters due to the main factors affecting each other. The results of the Mann-Whitney test showed that the value of the test statistics was 79. Also, the value of Asymp.Sig was obtained to be 0.018, which is less than 0.025 (two-tailed test), and it can be concluded that the classification of work units in the two models, clustering and desertification, is not equal (P<0.05). Background and Aims: Many countries lack resources to identify patients at risk of developing Type 2 diabetes mellitus (diabetes). We aimed to develop and validate a diabetes risk score based on easily accessible clinical data., Methods: Prospective study including 5277 participants (55.0% women, 51.8±10.5 years) free of diabetes at baseline. Comparison with two other published diabetes risk scores (Balkau and Kahn clinical, respectively 5 and 8 variables) and validation on three cohorts (Europe, Iran and Mexico) was performed., Results: After a mean follow-up of 10.9 years, 405 participants (7.7%) developed diabetes. Our score was based on age, gender, waist circumference, diabetes family history, hypertension and physical activity. The area under the curve (AUC) was 0.772 for our score, vs. 0.748 (p<0.001) and 0.774 (p = 0.668) for the other two. Using a 13-point threshold, sensitivity, specificity, positive and negative predictive values (95% CI) of our score were 60.5 (55.5-65.3), 77.1 (75.8-78.2), 18.0 (16.0-20.1) and 95.9 (95.2-96.5) percent, respectively. Our score performed equally well or better than the other two in the Iranian [AUC 0.542 vs. 0.564 (p = 0.476) and 0.513 (p = 0.300)] and Mexican [AUC 0.791 vs. 0.672 (p<0.001) and 0.778 (p = 0.575)] cohorts. In the European cohort, it performed similarly to the Balkau score but worse than the Kahn clinical [AUC 0.788 vs. 0.793 (p = 0.091) and 0.816 (p<0.001)]. This study aims to capture the stance of YouTube video content creators regarding the use of generative AI for language learning. Opinion mining and sentiment analysis techniques were employed to analyse the content, comments, and transcriptions of 66 YouTube videos published from December 2022 to October 2023. The findings revealed that most videos focused on speaking (n = 40) and writing skills (n = 24), with fewer videos addressing listening (n = 3) and reading (n = 19) skills. Sentiment analysis showed that videos predominantly conveyed optimistic (n = 42) and analytical (n = 17) sentiments, indicating a generally positive stance towards generative AI for language learning. Clustering analysis identified four thematic clusters: "language development and practices" (n = 33), "basic expression skills" (n = 25), "intercultural communication skills" (n = 6), and "language structure and meaning" (n = 2), representing different approaches to language learning with generative AI. Cross-sectional analyses revealed fluctuations in video counts and sentiment scores over time, with higher expectations for generative AI in writing and speaking skills, and relatively lower interest in listening skills. The findings suggest that YouTube video creators generally view generative AI as a promising tool for language learning, with a focus on developing practical communication skills, fostering intercultural understanding, and facilitating language development. Educational Data Mining (EDM) holds promise in uncovering insights from educational data to predict and enhance students' performance. This paper presents an advanced EDM system tailored for classifying and improving tertiary students' programming skills. Our approach emphasizes effective feature engineering, appropriate classification techniques, and the integration of Explainable Artificial Intelligence (XAI) to elucidate model decisions. Through rigorous experimentation, including an ablation study and evaluation of six machine learning algorithms, we introduce a novel ensemble method, Stacking-SRDA, which outperforms others in accuracy, precision, recall, f1-score, ROC curve, and McNemar test. Leveraging XAI tools, we provide insights into model interpretability. This study seeks to assess the hydrogeochemical characteristics of groundwater in the southern part of Thal Desert of Pakistan. The primary focus lies in identifying potential sources of contamination and evaluating their impact on groundwater and the ecosystem. Groundwater samples were collected from diverse sources including shallow hand pumps, tubewells, and dug wells, with depths ranging from 11 to 28 m. A comprehensive analysis was performed to scrutinize the physical, chemical, and microbial attributes of the samples. Utilizing visual aids like the Piper, Durov, and Gibbs diagrams, as well as Pearson correlation, scatter plots, Schoeller diagrams, and pie charts, the study evaluated the groundwater quality and its suitability for consumption. Results indicate that mineral infiltration from rainfall, domestic waste, and industrial effluents significantly affects groundwater quality, leading to widespread salinity. Weathering processes and ion exchange were identified as key factors contributing to elevate levels of bicarbonates, sodium, magnesium, and chloride ions. Employing the Water Quality Index (WQI) on 40 groundwater samples, findings reveal that 52.5% of samples demonstrated poor to not suitable quality, with 27.5% categorized as poor, 2.5% as very poor, and 22.5% not suitable consumption. Conversely, 47.5% of samples showcased good to excellent quality, with 25% rated as good and 22.5% as excellent. Wind erosion resulting from soil degradation is a significant problem in Iran's Baluchistan region. This study evaluated the accuracy of remote sensing models in assessing degradation severity through field studies. Sentinel-2 Multispectral Imager's (MSI) Level-1C satellite data was used to map Rutak's degradation severity in Saravan. The relationship between surface albedo and spectral indices (NDVI, SAVI, MSAVI, BSI, TGSI) was assessed. Linear regression establishes correlations between the albedo and each index, producing a degradation severity map categorized into five classes based on albedo and spectral indices. Accuracy was tested with 100 ground control points and field observations. The Mann-Whitney U-Test compares remote sensing models with field data. Results showed no significant difference (P > 0.05) between NDVI, SAVI, and MSAVI models with field data, while BSI and TGSI models exhibited significant differences (P ≤ 0.001). The best model, BSI-NDVI, achieves a regression coefficient of 0.86. This study demonstrates the advantage of remote sensing technology for mapping and monitoring degraded areas, providing valuable insights into land degradation assessment in Baluchistan. Feature selection is an important solution for dealing with high-dimensional data in the fields of machine learning and data mining. In this paper, we present an improved mountain gazelle optimizer (IMGO) based on the newly proposed mountain gazelle optimizer (MGO) and design a binary version of IMGO (BIMGO) to solve the feature selection problem for medical data. First, the gazelle population is initialized using iterative chaotic map with infinite collapses (ICMIC) mapping, which increases the diversity of the population. Second, a nonlinear control factor is introduced to balance the exploration and exploitation components of the algorithm. Individuals in the population are perturbed using a spiral perturbation mechanism to enhance the local search capability of the algorithm. Finally, a neighborhood search strategy is used for the optimal individuals to enhance the exploitation and convergence capabilities of the algorithm. The superior ability of the IMGO algorithm to solve continuous problems is demonstrated on 23 benchmark datasets. Then, BIMGO is evaluated on 16 medical datasets of different dimensions and compared with 8 well-known metaheuristic algorithms. The experimental results indicate that BIMGO outperforms the competing algorithms in terms of the fitness value, number of selected features and sensitivity. This bioassay evaluated the bioavailability (RBV) of a novel nanoparticle of methionine (nano-Met) relative to DL-methionine (DL-Met), and estimated methionine requirements for both sources in starting broilers. Five supplemental levels (0.05, 0.10, 0.15, 0.20, and 0.25% of diet) of DL-Met or nano-Met were added to a basal diet containing 0.35% standardized ileal digestible (SID) methionine to create 11 experimental diets, including a basal diet and 10 experimental diets containing 0.40, 0.45, 0.50, 0.55, and 0.60% SID-Met, respectively. A total of 825 one-day-old male Ross 308 birds were randomly assigned to 11 treatments with 5 pen replicates and 15 birds each. Body weight gain (BWG), breast meat yield (BMY), and thigh meat yield (TMY) increased (P < 0.001) while feed conversion ratio (FCR) and malondialdehyde (MDA) concentration in meat samples decreased (P < 0.001) with increasing dietary methionine. Based on the slope-ratio method, the RBV of nano-Met relative to DL-Met for BWG, FCR, and TMY were 102 (48–155%; R2 = 0.71), 134 (68–201%; R2 = 0.77), and 110% (27–193%; R2 = 0.55), respectively. Considering the statistical accuracy of the spline models, the estimated values of DL-Met for maximum BWG and nano-Met for maximum TMY were 0.578% and 0.561%, respectively, which were statistically higher than those recommended for commercial settings. The highest effect size of supplemental methionine was on MDA (ƞ2p = 0.924), followed by FCR (ƞ2p = 0.578), BMY (ƞ2p = 0.575), BWG (ƞ2p = 0.430), and TMY (ƞ2p = 0.332), suggesting the potent antioxidant properties of methionine. Our findings suggest that reducing the particle size of DL-Met to nanoparticles could be a promising strategy to enhance the efficiency of methionine supplementation in broilers, an idea that requires further investigation in future research. The study highlights the potential characteristics of droughts under future climate change scenarios. For this purpose, the changes in Standardized Precipitation Evapotranspiration Index (SPEI) under the A1B, A2, and B1 climate change scenarios in Iran were assessed. The daily weather data of 30 synoptic stations from 1992 to 2010 were analyzed. The HadCM3 statistical model in the LARS-WG was used to predict the future weather conditions between 2011 and 2112, for three 34-year periods; 2011–2045, 2046–2079, and 2080–2112. In regard to the findings, the upward trend of the potential evapotranspiration in parallel with the downward trend of the precipitation in the next 102 years in three scenarios to the base timescale was transparent. The frequency of the SPEI in the base month indicated that 17.02% of the studied months faced the drought. Considering the scenarios of climate change for three 34-year periods (i.e., 2011–2045, 2046–2079, and 2080–2112) the average percentages of potential drought occurrences for all the stations in the next three periods will be 8.89, 16.58, and 27.27 respectively under the B1 scenario. While the predicted values under the A1B scenario are 7.63, 12.66, and 35.08%respectively. The relevant findings under the A2 scenario are 6.73, 10.16, 40.8%. As a consequence, water shortage would be more serious in the third period of study under all three scenarios. The percentage of drought occurrence in the future years under the A2, B1, and A1B will be 19.23%, 17.74%, and 18.84%, respectively which confirms the worst condition under the A2 scenario. For all stations, the number of months with moderate drought was substantially more than severe and extreme droughts. Considering the A2 scenario as a high emission scenario, the analysis of SPEI frequency illustrated that the proportion of dry periods in regions with humid and cool climate is more than hot and warm climates; however, the duration of dry periods in warmer climates is longer than colder climates. Moreover, the temporal distribution of precipitation and potential evapotranspiration indicated that in a large number of stations, there is a significant difference between them in the middle months of the year, which justifies the importance of prudent water management in warm months. A comprehensive literature review of self-balancing robot (SBR) provides an insight to the strengths and limitations of the available control techniques for different applications. Most of the researchers have not included the payload and its variations in their investigations. To address this problem comprehensively, it was realized that a rigorous mathematical model of the SBR will help to design an effective control for the targeted system. A robust control for a two-wheeled SBR with unknown payload parameters is considered in these investigations. Although, its mechanical design has the advantage of additional maneuverability, however, the robot's stability is affected by changes in the rider's mass and height, which affect the robot's center of gravity (COG). Conventionally, variations in these parameters impact the performance of the controller that are designed with the assumption to operate under nominal values of the rider's mass and height. The proposed solution includes an extended Kalman filter (EKF) based sliding mode controller (SMC) with an extensive mathematical model describing the dynamics of the robot itself and the payload. The rider's mass and height are estimated using EKF and this information is used to improve the control of SBR. Significance of the proposed method is demonstrated by comparing simulation results with the conventional SMC under different scenarios as well as with other techniques in literature. The proposed method shows zero steady state error and no overshoot. Performance of the conventional SMC is improved with controller parameter estimation. Moreover, the stability issue in the reaching phase of the controller is also solved with the availability of parameter estimates. The proposed method is suitable for a wide range of indoor applications with no disturbance. This investigation provides a comprehensive comparison of available techniques to contextualize the proposed method within the scope of self-balancing robots for indoor applications. Aims: This study aims to comprehensively review the existing evidence and conduct analysis of updated randomized controlled trials (RCTs) of turmeric (Curcuma longa, CL) and its related bioactive compounds on glycemic and metabolic parameters in patients with type 2 diabetes (T2DM), prediabetes, and metabolic syndrome (MetS) together with a sub-group analysis of different CL preparation forms. Methods: An umbrella review (UR) and updated systematic reviews and meta-analyses (SRMAs) were conducted to evaluate the effects of CL compared with a placebo/standard treatment in adult T2DM, prediabetes, and MetS. The MEDLINE, Embase, The Cochrane Central Register of Control Trials, and Scopus databases were searched from inception to September 2022. The primary efficacy outcomes were hemoglobin A1C (HbA1C) and fasting blood glucose (FBG). The corrected covered area (CCA) was used to assess overlap. Mean differences were pooled across individual RCTs using a random-effects model. Subgroup and sensitivity analyses were performed for various CL preparation forms. Results: Fourteen SRMAs of 61 individual RCTs were included in the UR. The updated SRMA included 28 studies. The CCA was 11.54%, indicating high overlap across SRMAs. The updated SRMA revealed significant reduction in FBG and HbA1C with CL supplementation, obtaining a mean difference (95% confidence interval [CI]) of –8.129 (–12.175, –4.084) mg/dL and –0.134 (–0.304, –0.037) %, respectively. FBG and HbA1C levels decreased with all CL preparation forms as did other metabolic parameters levels. The results of the sensitivity and subgroup analyses were consistent with those of the main analysis. Conclusion: CL supplementation can significantly reduce FBG and HbA1C levels and other metabolic parameters in T2DM and mitigate related conditions, including prediabetes and MetS. Research studies have observed associations of vitamin D with inflammation but data in representative older adult studies is lacking. We aimed to investigate the association of C-reactive protein (CRP) with vitamin D status in a representative sample of the older Irish population. The concentrations of 25-hydroxyvitamin D (25(OH)D) and CRP was measured in 5,381 community dwelling Irish adults aged ≥50 years from the Irish Longitudinal Study on Ageing (TILDA). Demographic, health and lifestyle variables were assessed by questionnaire and categorical proportions of CRP were generated by vitamin D status and age. Multi-nominal logistic regression was used to investigate the association of 25(OH)D and CRP status. The prevalence (mean; 95% confidence interval (95% CI)) of normal CRP status (0–5 mg/dL) was 83.9% (82.6–85.0%), elevated status (5–10 mg/dL) 11.0% (9.9–12.0%) and high status (>10 mg/dL) was 5.1% (4.5–5.8%). Mean (95% CI) CRP concentrations were lower in those with normal vs. deficient 25(OH)D status (2.02 mg/dL (1.95–2.08) vs. 2.60 mg/dL (2.41–2.82); p<0.0001). In a logistic regression analysis, those with insufficient or sufficient 25(OH)D status were less likely to have a high CRP status compared to those with deficient 25(OH)D status (insufficient: coefficient (CE) -0.732, 95% CI -1.12–0.33, p<0.0001; sufficient: CE -0.599, 95% CI -0.95–0.24, p = 0.001). In conclusion older adults with deficient vitamin D status had higher levels of inflammation as measured by CRP. Given that inflammation is an important pathological driver of chronic diseases of ageing, and that emerging evidence suggests that vitamin D therapy can reduce inflammation in some disease settings, optimising vitamin D status could represent an effective low risk/low-cost pathway to modulate inflammation in community dwelling older adults. Advancement of research in education has propelled the augmentation of theoretical and practical knowledge in learning-oriented feedback. In recent years, the channels, modes, and orientations of feedback became manifold. Copious empirical evidence from the body of literature supported the strength of feedback in enhancing learning outcomes and promoting the motivation of learners. However, compared to the popularity in implementation and fruitfulness of findings in other educational domains, the application of state-of-the-art technology-enhanced feedback in fostering students' L2 oral abilities remain few and far between. To address the knowledge gap, the present study endeavored to investigate the effect of Danmaku-based and synchronous peer feedback on L2 oral performance and the acceptance thereof among students. Adopting a mixed-method design, the study recruited 74 (n = 74) undergraduate English majors from a Chinese university for a 16-week 2x2 experiment. The collected data were analyzed through statistical and thematic analysis respectively. The findings revealed that Danmaku-based and synchronous peer feed-back was impactful on students' performance in L2 oral production. Furthermore, the impacts of peer feedback on subdomains of L2 competence were statistically analyzed. Regarding students' perceptions, the incorporation of peer feedback was generally favored by participants who were satisfied and motivated in the learning process but lacked confidence in their assessment literacy. Furthermore, students expressed their agreement with the benefit of reflective learning and the subsequent enrichment in knowledge and horizon. A series of model tests are carried out on flexible retaining walls such as cantilevered piles, continuous walls, and sheet pile walls in the foundation pit to study the deformation, failure surface, and earth pressure distribution of soils in a passive zone. The shape, displacement, and shear strain of slip failure surface of sand in a passive area are analyzed by Particle Image Velocimetry. The slip failure surface is a broken line, the upper end slides out from the top of the soil, and the lower end is close to the zero displacements of the retaining wall. With the increase of the flexural deformation and horizontal displacement of the wall, the shear strain of the soil increases, and the shear fracture zone in the upper part of the sliding surface is more prominent. Based on the broken line rupture surface in the test results, the passive area can be divided into two zones, the limit state zone and the non-limit state zone. Then the mechanical models are set up respectively. Considering soil displacement, the upper and lower soil layer's internal friction angle and wall-soil interface friction angle mobilize differently. The relationship between mechanical parameters along the retaining wall and horizontal displacement is estimated. Finally, the earth pressure distribution is obtained by using the horizontal differential layer method. The calculation results of this paper are consistent with the existing research results and the model test results in terms of earth pressure distribution. With the increase of depth, the unit earth pressure increases in the limit state zone. Traffic congestion in large urban networks may take different shapes and propagates non-uniformly variations from day to day. Given the fact that congestion on a road segment is spatially correlated to adjacent roads and propagates spatiotemporally with finite speed, it is essential to describe the main pockets of congestion in a city with a small number of clusters. For example, the perimeter control with macroscopic fundamental diagrams is one of the effective traffic management tools. Perimeter control adjusts the inflow to pre-specified regions of a city through signal timing on the border of a region in order to optimize the traffic condition within the region. The precision of macroscopic fundamental diagrams depends on the homogeneity of traffic condition on road segments of the region. Hence, previous studies have defined the boundaries of the region under perimeter control subjected to the regional homogeneity. In this study, a cost-effective method is proposed for the mentioned problem that simultaneously considers homogeneity, contiguity and compactness of clusters and has a shorter computational time. Since it is necessary to control the cost and complexity of perimeter control in terms of the number of traffic signals, sparse parts of the network could be potential candidates for boundaries. Therefore, a community detection method (Infomap) is initially adopted and then those clusters are improved by refining the communities in relation to roads with the highest heterogeneity. The proposed method is applied to Shenzhen, China and San Francisco, USA and the outcomes are compared to previous studies. The results of comparison reveal that the proposed method is as effective as the best previous methods in detecting homogenous communities, but it outperforms them in contiguity. Studies investigating the effects of tocotrienols on inflammation and oxidative stress have yielded inconsistent results. This systematic review and meta-analysis aimed to evaluate the effects of tocotrienols supplementation on inflammatory and oxidative stress biomarkers. We searched PubMed, Scopus, and Cochrane Central Register of Controlled Trials from inception until 13 July 2020 to identify randomized controlled trials supplementing tocotrienols and reporting circulating inflammatory or oxidative stress outcomes. Weighted mean difference (WMD) and corresponding 95% confidence interval (CI) were determined by pooling eligible studies. Nineteen studies were included for qualitative analysis, and 13 studies were included for the meta-analyses. A significant reduction in C-reactive protein levels (WMD: −0.52 mg/L, 95% CI: −0.73, −0.32, p < 0.001) following tocotrienols supplementation was observed, but this finding was attributed to a single study using δ-tocotrienols, not mixed tocotrienols. There were no effects on interleukin-6 (WMD: 0.03 pg/mL, 95% CI: −1.51, 1.58, p = 0.966), tumor necrosis factor-alpha (WMD: −0.28 pg/mL, 95% CI: −1.24, 0.68, p = 0.571), and malondialdehyde (WMD: −0.42 μmol/L, 95% CI: −1.05, 0.21, p = 0.189). A subgroup analysis suggested that tocotrienols at 400 mg/day might reduce malondialdehyde levels (WMD: −0.90 μmol/L, 95% CI: −1.20, −0.59, p < 0.001). Future well-designed studies are warranted to confirm the effects of tocotrienols on inflammatory and oxidative stress biomarkers, particularly on different types and dosages of supplementation. To examine changes in U.S. medical school basic science faculty over the last 20 years (1998–2018), we undertook an observational study utilizing data from the American Association of Medical Colleges Faculty Roster. Rank (Instructor, Assistant Professor, Associate Professor, and Professor), sex (Female), and race/ethnicity (Asian, Black or African American, Hispanic, Latino, Spanish Origin, or Multiple Race-Hispanic, and White) were analyzed; this reflected a population of 14,047 (1998) to 18,601 (2018) faculty. Summary percent of faculty in various gender, race/ethnicity origin categories were analyzed across years of the study using regression models. We found that females (24.47% to 35.32%) were underrepresented at all timepoints and a minority of faculty identified as Black or African American (1.57% to 1.99%), Hispanic, Latino, Spanish Origin, or Multiple Race-Hispanic (3.03% to 4.44%), or Asian (10.90% to 20.41%). The largest population at all time points was White Male Professors (30.53% to 20.85%), followed by White Male Associate Professors (15.67% to 9.34%), and White Male Assistant Professors (13.22% to 9.75%). Small statistically significant increases were observed among female faculty and faculty at multiple ranks who identified as Black or African American or Hispanic, Latino, Spanish Origin, or Multiple Race-Hispanic. We then completed secondary analyses looking at the interaction of race/ethnicity and Gender. We found: (1) a significant increase (p<0.0001) in both genders who identify as Asian although males had a higher rate of increase (6 point difference, p<0.0001); (2) a significant increase for Black or African American females (P<0.01) not found among males; (3) significant increases (p<0.0001) among both genders of faculty who identify as Hispanic, Latino, Spanish Origin, or Multiple Race-Hispanic although females had an approximately 1% higher rate of increase; and (4) among faculty who identify as White, males had a significant decrease (p<0.0001) while females demonstrated an increase (p<0.0001). Forecasting possible future relationships between people in a network requires a study of the evolution of their links. To capture network dynamics and temporal variations in link strengths between various types of nodes in a network, a dynamic weighted heterogeneous network is to be considered. Link strength prediction in such networks is still an open problem. Moreover, a study of variations in link strengths with respect to time has not yet been explored. The time granularity at which the weights of various links change remains to be delved into. To tackle these problems, we propose a neural network framework to predict dynamic variations in weighted heterogeneous social networks. Our link strength prediction model predicts future relationships between people, along with a measure of the strength of those relationships. Numerous studies have revealed the role of dietary acid load as a potential risk factor for cardiovascular events and blood pressure. However, its role in predicting the mortality rate in patients underwent coronary artery bypass grafting surgery (CABG) has not been reported. In the current study we aimed to evaluate the relationship of dietary acid load and cardio-metabolic risk factors with ten year survival among patients underwent CABG. The current prospective cohort study comprises 454 patients underwent CABG. Anthropometric, clinical and biochemical measurements were performed. Dietary acid load was calculated as either potential renal acid load (PRAL) or net endogenous acid production (NEPA) using the data obtained from a semi-quantitative food frequency questionnaire (FFQ). Survival analysis was performed using Kaplan-Meier method followed by log-rank test. The association between all-cause mortality and study parameters was performed with Cox-proportional hazard model. Patients in the higher PRAL and NEAP quartiles had lower BMI and lower ejection fraction rate (P <0.05). Moreover, lower hematocrit values were observed in patients of higher PRAL quartiles. Higher PRAL scores were associated with higher mortality rate and reduced survival days (adjusted hazard ratio: 1.023 (1.00–1.04; P-value = 0.01). However, there was no relationship between NEAP and survival. We revealed that high PRAL scores are positive predictors of 10-year mortality in patients underwent CABG. Background: In the current meta-analysis, we aimed to systematically review and summarize the eligible studies evaluating the association between dietary acid load in terms of potential renal acid load (PRAL) and net-endogenous acid production (NEAP) with anthropometric parameters and serum lipids in adult population. Methods: In a systematic search from PubMed, Scopus, Web of Sciences and Cochrane electronic databases up to December 2018, relevant studies were included. Cross-sectional, case control or cohort studies evaluating the association between PRAL and NEAP with the mean values of body mass index (BMI), waist circumference (WC), low and high density lipoprotein cholesterol (LDL, HDL), triglyceride (TG), total cholesterol (TC) and the prevalence of obesity were included. Results: According to our results, having higher dietary acid load content in terms of high PRAL scores was associated with higher triglyceride concentrations (weighted mean difference (WMD): 3.468; confidence interval (CI): -0.231, 7.166, P = 0.04) and higher obesity prevalence (30% and 27% in highest versus lowest categories). Accordingly, being in the highest category of NEAP was associated with higher prevalence of obesity (25% and 22% in highest versus lowest category). In subgroup analysis, higher PRAL scores was associated with higher BMI in women (WMD: 0.122; CI: -0.001, 0.245; P = 0.049) and higher NEAP in men (WMD: 0.890; CI: 0.430, 1.350; P < 0.001). There was no association between dietary acid load and other studied parameters. Conclusions: In the current meta-analysis, high dietary acid load content was associated with higher serum triglyceride concentrations and higher obesity prevalence. Background: Trials have examined on the benefits of vitamin D supplementation in pregnant women. Objective: This review aimed to evaluate whether oral vitamin D supplements, when given to pregnant women with gestational diabetes mellitus (GDM), would improve maternal and neonatal outcomes, compared with no treatment or placebo. Method: We performed a systematic review following Cochrane methodology, and randomized trials were included where pregnant women with GDM received vitamin D supplementation versus placebo/no treatment or vitamin D and calcium versus placebo/no treatment. Primary outcomes were preeclampsia, preterm birth, cesarean delivery, gestational hypertension, and adverse events related to vitamin D supplementation. The search strategies were applied to the following databases: MEDLINE, Embase, LILACS, and CENTRAL. Similar outcomes in at least two trials were plotted using Review Manager 5.3 software. The quality of evidence was generated according to the Grading of Recommendations Assessment, Development, and Evaluation (GRADE). Results: The total of 1224 references were identified, eleven trials were potentially eligible, and six were included in this review (totaling 456 women). The meta-analysis of frequency of cesarean deliveries did not show significant differences between groups, none of the trials evaluated the remaining primary outcomes. For secondary outcomes, our results suggest that vitamin D supplementation in pregnant women with GDM may reduce newborn complications such as hyperbilirubinemia, polyhydramnios (RR: 0.40, 95% CI: 0.23 to 0.68; RR: 0.17, 95% CI: 0.03 to 0.89; respectively), and the need for maternal or infant hospitalization (RR: 0.13; 95% CI: 0.02 to 0.98; RR: 0.40, 95% CI: 0.23 to 0.69). However, the evidence was of low or very low quality. Conclusion: We did not find moderate or high quality evidence indicating that vitamin D supplementation, when compared with placebo, improves glucose metabolism, adverse maternal and neonatal outcomes related to GDM in pregnant women. Objective: To evaluate trends in racial, ethnic, and sex representation at US medical schools across 16 specialties: internal medicine, pediatrics, surgery, psychiatry, radiology, anesthesiology, obstetrics and gynecology, neurology, family practice, pathology, emergency medicine, orthopedic surgery, ophthalmology, otolaryngology, physical medicine and rehabilitation, and dermatology. Using a novel, Census-derived statistical measure of diversity, the S-score, we quantified the degree of underrepresentation for racial minority groups and female faculty by rank for assistant, associate, and full professors from 1990–2016. Methods: This longitudinal study of faculty diversity uses data obtained from the American Association of Medical Colleges (AAMC) Faculty Roster from US allopathic medical schools. The proportion of professors of racial minority groups and female faculty by rank was compared to the US population based on data from the US Census Bureau. The Roster includes data on 52,939 clinical medical faculty in 1990, and 129,545 in 2016, at the assistant professor level or higher. The primary measure used in this study was the S-score, a measure of representation based on the probability of the observed frequency of faculty from a racial/ethnic group and sex, given the racial and ethnic distribution of the US. Pearson correlations and 95% confidence intervals for S-score with time were used to measure trends. Results: Blacks and Hispanics showed statistically significant trends (p<0.05) towards increasing underrepresentation in most specialties and are more underrepresented in 2016 than in 1990 across all ranks and specialties analyzed, except for Black females in obstetrics & gynecology. White females were also underrepresented in many specialties and in a subset of specialties trended toward greater underrepresentation. Conclusions: Current efforts to improve faculty diversity are inadequate in generating an academic physician workforce that represents the diversity of the US. Background: A large number of observational studies have reported harmful effects of low 25-hydroxyvitamin D (25OHD) levels on non-skeletal outcomes. We performed a systematic quantitative review on characteristics of randomized clinical trials (RCTs) included in meta-analyses (MAs) on non-skeletal effects of vitamin D supplementation. Methods and findings: We identified systematic reviews (SR) reporting summary data in terms of MAs of RCTs on selected non-skeletal outcomes. For each outcome, we summarized the results from available SRs and scrutinized included RCTs for a number of predefined characteristics. We identified 54 SRs including data from 210 RCTs. Most MAs as well as the individual RCTs reported null-findings on risk of cardiovascular diseases, type 2 diabetes, weight-loss, and malignant diseases. Beneficial effects of vitamin D supplementation was reported in 1 of 4 MAs on depression, 2 of 9 MAs on blood pressure, 3 of 7 MAs on respiratory tract infections, and 8 of 12 MAs on mortality. Most RCTs have primarily been performed to determine skeletal outcomes, whereas non-skeletal effects have been assessed as secondary outcomes. Only one-third of the RCTs had low level of 25OHD as a criterion for inclusion and a mean baseline 25OHD level below 50 nmol/L was only present in less than half of the analyses. Conclusions: Published RCTs have mostly been performed in populations without low 25OHD levels. The fact that most MAs on results from RCTs did not show a beneficial effect does not disprove the hypothesis suggested by observational findings on adverse health outcomes of low 25OHD levels. Background: Low levels of serum 25-hydroxy vitamin D are associated with increased arterial stiffness and hypertension. Supplementation with vitamin D precursors has been proposed as a treatment option for these conditions. We examined the effect of oral cholecalciferol on arterial stiffness and blood pressure in healthy normotensive adults. Methods: 40 healthy adults were randomised in this double-blinded study to either oral cholecalciferol 3000 IU/day or matching placebo and were followed for 16 weeks to examine any effects on pulse wave velocity (PWV), augmentation index (AIx), peripheral and central blood pressure and 24-hour ambulatory blood pressure. Results: 22 subjects in the cholecalciferol arm and 18 subjects in the placebo arm completed the 16 weeks of follow-up. There was no difference in changes in PWV, AIx corrected for heart rate or central or peripheral blood pressure between the two groups. There was no correlation between serum 25-hydroxy vitamin D and any of these parameters. Conclusions: Oral cholecalciferol 3000 IU/day does not affect arterial stiffness or blood pressure after 16 weeks of treatment in healthy normotensive adults. Objectives: Vitamin D (VitD) deficiency is a health problem prevalent not only in the elderly but also in young adults. The primary objective of our observational pilot study “MUVY” (Mood, UVR, Vitamin D in Young women) was to test both the short-term and long-term effects of a series of three suberythemal UV radiation (UVR) exposures on the VitD status and well-being of young healthy women during winter in a repeat measure design. Methods: 20 healthy young women (Fitzpatrick skin types I–III, aged 21–25 years) received three full body broad band UVR exposures with an escalating erythemally weighted dose schedule during one week in winter, and completed self-report questionnaires monitoring symptoms of depression (Beck Depression Inventory, BDI) and affective state/well-being (Profile of Mood States, POMS) at baseline and three days after the last UVR exposure. 25-hydroxyvitamin D (25(OH)D) and 1,25-dihydroxyvitamin D (1,25(OH)2D) were measured in serum at baseline, and at study days 8, 36 and 50. Results: Mean baseline 25(OH)D level was 54.3 nmol/L (standard deviation (s.d.) = 24.1), with seven women having VitD deficient status. Relevant symptoms of depression, as indicated by low BDI total scores (0–8), were absent. After the three UVR exposures the increment of 25(OH)D was an average of 13.9 nmol/L (95% confidence interval (CI) = 9.4–18.4) and 26.2 pmol/L (95%CI = 7.2–45.1) for 1,25(OH)2D. Δ25(OH)D, and corresponding baseline levels were significantly and inversely associated (rho = -0.493, p = 0.027). Only 25(OH)D remained significantly increased above baseline for at least six weeks after the last UVR exposure. A strong inverse correlation of the POMS subscale “Vigor/Activity” and the increment in 1,25(OH)2D was found (rho = -0.739, p<0.001) at day 8. Conclusions: Three suberythemal whole body UVR exposures during one week are a simple and suitable method for improving 25(OH)D levels during winter, for at least six weeks, and especially in young women with VitD deficient status. Epidemiological studies have shown that up to one third of adults have insufficient levels of vitamin D and there is an association between low vitamin D concentrations and adverse brain outcomes, such as depression. Vitamin D has been shown to be involved in processes associated with neurogenesis during development. Therefore, the aim of this study was to test the hypothesis that adult vitamin D (AVD) deficiency in BALB/c mice was associated with (a) adult hippocampal neurogenesis at baseline, b) following 6 weeks of voluntary wheel running and (c) a depressive-like phenotype on the forced swim test (FST), which may be linked to alterations in hippocampal neurogenesis. We assessed proliferation and survival of adult born hippocampal neurons by counting the number of cells positive for Ki67 and doublecortin (DCX), and incorporation of 5-Bromo-2’-Deoxyuridine (BrdU) within newly born mature neurons using immunohistochemistry. There were no significant effects of diet on number of Ki67+, DCX+ or BrdU+ cells in the dentate gyrus. All mice showed significantly increased number of Ki67+ cells and BrdU incorporation, and decreased immobility time in the FST, after voluntary wheel running. A significant correlation was found in control mice between immobility time in the FST and level of hippocampal neurogenesis, however, no such correlation was found for AVD-deficient mice. We conclude that AVD deficiency was not associated with impaired proliferation or survival of adult born neurons in BALB/c mice and that the impact on rodent behaviour may not be due to altered neurogenesis per se, but to altered function of new hippocampal neurons or processes independent of adult neurogenesis. Induction of GLUT4 translocation in the absence of insulin is considered a key concept to decrease elevated blood glucose levels in diabetics. Due to the lack of pharmaceuticals that specifically increase the uptake of glucose from the blood circuit, application of natural compounds might be an alternative strategy. However, the effects and mechanisms of action remain unknown for many of those substances. For this study we investigated extracts prepared from seven different plants, which have been reported to exhibit anti-diabetic effects, for their GLUT4 translocation inducing properties. Quantitation of GLUT4 translocation was determined by total internal reflection fluorescence (TIRF) microscopy in insulin sensitive CHO-K1 cells and adipocytes. Two extracts prepared from purslane (Portulaca oleracea) and tindora (Coccinia grandis) were found to induce GLUT4 translocation, accompanied by an increase of intracellular glucose concentrations. Our results indicate that the PI3K pathway is mainly responsible for the respective translocation process. Atomic force microscopy was used to prove complete plasma membrane insertion. Furthermore, this approach suggested a compound mediated distribution of GLUT4 molecules in the plasma membrane similar to insulin stimulated conditions. Utilizing a fluorescent actin marker, TIRF measurements indicated an impact of purslane and tindora on actin remodeling as observed in insulin treated cells. Finally, in-ovo experiments suggested a significant reduction of blood glucose levels under tindora and purslane treated conditions in a living organism. Parkinson’s disease (PD) is characterized by the presence of Lewy bodies and degeneration of dopaminergic neurons. 1-methyl-4-phenylpyridinium (MPP+), a metabolite of neurotoxin 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) and Lewy body component α-synuclein activates glia in PD pathogenesis. Mast cells and glia maturation factor (GMF) are implicated in neuroinflammatory conditions including Multiple Sclerosis. However, the role of mast cells in PD is not yet known. We have analyzed the effect of recombinant GMF, MPP+, α-synuclein and interleukin-33 (IL-33) on mouse bone marrow-derived cultured mast cells (BMMCs), human umbilical cord blood-derived cultured mast cells (hCBMCs) and mouse brain-derived cultured astrocytes by quantifying cytokines/chemokines released using ELISA or by detecting the expression of co-stimulatory molecules CD40 and CD40L by flow cytometry. GMF significantly released chemokine (C-C motif) ligand 2 (CCL2) from BMMCs but its release was reduced in BMMCs from GMF knockout mice. GMF, α-synuclein and MPP+ released IL-1β, β-hexosaminidase from BMMCs, and IL-8 from hCBMCs. GMF released CCL5, and IL-33- induced the expression of GMF from hCBMCs. Novel GMF expression was detected in hCBMCs and BMMCs by immunocytochemistry. GMF released tumor necrosis factor-alpha (TNF-α) from mouse astrocytes, and this release was greater in BMMC- astrocyte coculture than in individual cultures. Flow cytometry results showed increased IL-33 expression by GMF and MPP+, and GMF-induced CD40 expression in astrocytes. Proinflammatory mediator release by GMF, MPP+ and α-synuclein, as well as GMF expression by mast cells indicate a potential therapeutic target for neurodegenerative diseases including PD. Gestational Diabetes Mellitus (GDM) and vitamin D deficiency are related to insulin resistance and impaired beta cell function, with heightened risk for future development of diabetes. We evaluated the impact of vitamin D supplementation on markers of glucose metabolism and cardio metabolic risk in Asian women with former GDM and hypovitaminosis D. In this double blind, randomized controlled trial, 26 participants were randomized to receive either daily 4000 IU vitamin D3 or placebo capsules. 75g Oral Glucose Tolerance Test (OGTT) and biochemistry profiles were performed at baseline and 6 month visits. Mathematical models, using serial glucose, insulin and C peptide measurements from OGTT, were employed to calculate insulin sensitivity and beta cell function. Thirty three (76%) women with former GDM screened had vitamin D level of <50 nmol/L at baseline. Supplementation, when compared with placebo, resulted in increased vitamin D level (+51.1 nmol/L vs 0.2 nmol/L, p<0.001) and increased fasting insulin (+20% vs 18%, p = 0.034). The vitamin D group also demonstrated a 30% improvement in disposition index and an absolute 0.2% (2 mmol/mol) reduction in HbA1c. There was no clear change in insulin sensitivity or markers of cardio metabolic risk. This study highlighted high prevalence of vitamin D deficiency among Asian women with former GDM. Six months supplementation with 4000 IU of vitamin D3 safely restored the vitamin D level, improved basal pancreatic beta-cell function and ameliorated the metabolic state. There was no effect on markers of cardio metabolic risk. Further mechanistic studies exploring the role of vitamin D supplementation on glucose homeostasis among different ethnicities may be needed to better inform future recommendations for these women with former GDM at high risk of both hypovitaminosis D and future diabetes.
