The impact of trace metal elements (TMEs) on plants is one current pollution problem, the severity of which is increasing with industrial development, population growth and inappropriate agricultural practices. The latter can have irreversible effects on ecosystems, including species extinction, trophic chain contamination and altered human health, particularly in the case of consumed plants such as zucchini squash ( Cucurbita pepo L.). This study aims to investigate the effects of nickel on various physiological and biochemical parameters of zucchini growth, with a particular focus on how this toxic metal impacts the quality of fruit that is consumed by humans. To achieve this, plants aged 45 days were grown for one month on solid media loaded with different concentrations of Ni (0, 100, 300 and 500 µM). The results showed that exposure of plants to Ni resulted in significantly altered growth and higher accumulation of Ni in the shoots (1314 µg·g -1 DW) than in roots and fruits. Concerning non-enzymatic antioxidants, the results showed that Ni toxicity significantly increased total polyphenols, especially in shoots at 300 µM Ni, while flavonoid content decreased in the roots and shoots in response to Ni treatment. Our results also show that nickel tolerance in C. pepo is ensured by a combination of several mechanisms such as an increase in the content of proline. This species can survive and tolerate, to different degrees, toxic cations at concentrations up to 500 µM but with visible symptoms of toxicity such as chlorosis of the leaves. Indeed, based on thresholds of hyperaccumulation, we can qualify Cucurbita pepo as a hyperaccumulator species of nickel.

Under stress or in optimum conditions, plants foster a specific guild of symbiotic microbes to strengthen pivotal functions including metabolic regulation. Despite that the role of the plant genotype in microbial selection is well documented, the potential of this genotype-specific microbial assembly in maintaining the host homeostasis remains insufficiently investigated. In this study, we aimed to assess the specificity of the foliar metabolic response of contrasting olive genotypes to microbial inoculation with wet-adapted consortia of plant-growth-promoting rhizobacteria (PGPR), to see if previously inoculated plants with indigenous or exogenous microbes would display any change in their leaf metabolome once being subjected to drought stress. Two Tunisian elite varieties, Chetoui (drought-sensitive) and Chemleli (drought-tolerant), were tested under controlled and stressed conditions. Leaf samples were analyzed by gas chromatography-mass spectrometry (GC-TOFMS) to identify untargeted metabolites. Root and soil samples were used to extract microbial genomic DNA destined for bacterial community profiling using 16S rRNA amplicon sequencing. Respectively, the score plot analysis, cluster analysis, heat map, Venn diagrams, and Krona charts were applied to metabolic and microbial data. Results demonstrated dynamic changes in the leaf metabolome of the Chetoui variety in both stress and inoculation conditions. Under the optimum state, the PGPR consortia induced noteworthy alterations in metabolic patterns of the sensitive variety, aligning with the phytochemistry observed in drought-tolerant cultivars. These variations involved fatty acids, tocopherols, phenols, methoxyphenols, stilbenoids, triterpenes, and sugars. On the other hand, the Chemleli variety displaying comparable metabolic profiles appeared unaffected by stress and inoculation probably owing to its tolerance capacity. The distribution of microbial species among treatments was distinctly uneven. The tested seedlings followed variety-specific strategies in selecting beneficial soil bacteria to alleviate stress. A highly abundant species of the wet-adapted inoculum was detected only under optimum conditions for both cultivars, which makes the moisture history of the plant genotype a selective driver shaping microbial community and thereby a useful tool to predict microbial activity in large ecosystems.

Date palms are a vital part of oasis ecosystems and are an important source of income in arid and semi-arid areas. Crossbreeding is limited due to the long juvenile stage of date palms and their dioecious nature. The aim of this study was to create triploid date palms to obtain larger and seedless fruits and to increase resilience to abiotic stresses. A tetraploid date palm mutant was crossed with a diploid male palm, yielding hundreds of seeds suspected of containing triploid embryos. Six years after planting, four palms with confirmed triploidy reached maturity. They are phenotypically distinct from diploids, with a thicker rachis, thinner spines, wider and longer midleaf spines, and a longer apical spine. They were classified as sterile bisexual, sterile male and fertile female. One of the latter produced very tasty dates with a very small seed, which is promising for the marketability and profitability of date palm fruits. This first report on triploid date palms provides a way in which to make a significant leap forward in date palm breeding. Given the vigor and fruit quality of female triploid date palms, compared to their diploid counterparts, they will be the target of breeding programs and may spearhead new oases.

The tomato as a raw material for processing is globally important and is pivotal in dietary and agronomic research due to its nutritional, economic, and health significance. This study explored the potential of machine learning (ML) for predicting tomato quality, utilizing data from 48 cultivars and 28 locations in Hungary over 5 seasons. It focused on °Brix, lycopene content, and colour (a/b ratio) using extreme gradient boosting (XGBoost) and artificial neural network (ANN) models. The results revealed that XGBoost consistently outperformed ANN, achieving high accuracy in predicting °Brix (R² = 0.98, RMSE = 0.07) and lycopene content (R² = 0.87, RMSE = 0.61), and excelling in colour prediction (a/b ratio) with a R² of 0.93 and RMSE of 0.03. ANN lagged behind particularly in colour prediction, showing a negative R² value of -0.35. Shapley additive explanation's (SHAP) summary plot analysis indicated that both models are effective in predicting °Brix and lycopene content in tomatoes, highlighting different aspects of the data. SHAP analysis highlighted the models' efficiency (especially in °Brix and lycopene predictions) and underscored the significant influence of cultivar choice and environmental factors like climate and soil. These findings emphasize the importance of selecting and fine-tuning the appropriate ML model for enhancing precision agriculture, underlining XGBoost's superiority in handling complex agronomic data for quality assessment.

Broomrapes ( Orobanche spp.) are root parasitic plants that threaten agricultural production in many parts of the world. In this study, the effect of two orobanche species, Orobanche crenata and O. foetida , on faba bean plants was studied in Tunisia. The two orobanche species inhibited both biomass production and pod formation, decreased the chlorophyll (Chl) content and total lipid (TL), and enhanced electrolyte leakage (EL) and lipid peroxidation. Concomitantly, orobanche parasitism induced a lower degree of fatty acid (FA) unsaturation due to a shift in the FA composition. On the other hand, with regard to orobanche seeds, oleic and linoleic acids were the predominant FA in the two orobanche species. After orobanche seed germination and penetration of host tissues, all the orobanche development stages showed a decrease in the TL content and changes in the FA composition in comparison to orobanche seeds. The level of TL was equal to or lower in all parasite development stages (except for S4) than that in the roots and leaves of healthy faba bean plants. These results suggest that the negative effect of orobanche infestation on faba bean development can be attributed to the reduced chlorophyll content and alteration in membrane stability attested by the reduced TL level and FA unsaturation.

Managing crop yields and optimizing water use is a global challenge, as fresh water supply decreases rapidly and demand remains high. Therefore, understanding how plants react to varying water levels is crucial for efficient water usage. This study evaluates how tomato plants adapt to varying water levels (100%, 50% of crop evapotranspiration, and non-irrigated control) over two growing seasons in 2020 and 2021. Root images were captured weekly during an 8-week monitoring period in 2020 and 6 weeks in 2021 using a non-destructive CI-600 in-situ root imager at depths between 10 and 70 cm. Under water stress, plants developed deeper, more extensive root systems to maximize water uptake, consistent with prior research. Root depth and architecture varied with soil depth and the severity of water stress. Year-to-year variations were also found, likely due to changes in irrigation levels and environmental conditions such as temperature. SPAD values were higher under control conditions, especially in the 2021 growing season, suggesting reduced chlorophyll degradation, while no significant differences were observed in chlorophyll fluorescence (Fv/Fm) between treatments, suggesting stable photosynthetic efficiency under varied water stress conditions. These findings contribute to our understanding of root zone optimization and drought-resilient cultivar breeding, contributing to more sustainable agricultural practices.

Gibberellic acid-stimulated Arabidopsis ( GASA ) gene family is a class of functional cysteine-rich proteins characterized by an N-terminal signal peptide and a C-terminal-conserved GASA domain with 12 invariant cysteine (Cys) residues. GASA proteins are widely distributed among plant species, and the majority of them are involved in the signal transmission of plant hormones, the regulation of plant development and growth, and the responses to different environmental constraints. To date, their action mechanisms are not completely elucidated. This review reports an overview of the diversity, structure, and subcellular localization of GASA proteins, their involvement in hormone crosstalk and redox regulation during development, and plant responses to abiotic and biotic stresses. Knowledge of this complex regulation can be a contribution to promoting multiple abiotic stress tolerance with potential agricultural applications through the engineering of genes encoding GASA proteins and the production of transgenic plants.

Growing conditions and seasonal fluctuations are critical factors affecting fruit and vegetable nutritional quality. The effects of two partially overlapping cropping seasons, early (ECS; January-May) and full (FCS; March-July), on the main carpometric traits and bioactive components of different watermelon fruits were investigated in the open field. Four watermelon genotypes, comprising of three commercial cultivars 'Crimson Sweet', 'Dumara', 'Giza', and the novel hybrid 'P503 F1', were compared. The carpometric traits varied significantly between genotypes. Soluble solids and yield were higher under FCS than ECS. The variation affecting colour indexes between the two growing seasons exhibited a genotype-dependent trend. The antioxidant components and radical scavenging activity of watermelon fruits were also significantly affected by differences in received solar energy and temperature fluctuations during the trial period. The average citrulline, total phenolics and flavonoid contents were 93%, 71% and 40% higher in FCS than in ECS. A genotype-dependent variation trend was also observed for lycopene and total vitamin C between cropping seasons. The hydrophilic and lipophilic radical scavenging activities of the pulp of ripe watermelon fruits of the different genotypes investigated varied between 243.16 and 425.31 µmol Trolox Equivalent (TE) of 100 g -1 of fresh weight (fw) and from 232.71 to 341.67 µmol TE of 100 g -1 fw in FCS and ECS, respectively. Our results, although preliminary, show that the functional quality of watermelon fruits is drastically altered depending on the environmental conditions that characterize the ECS and LCS.

Light-emitting diodes (LEDs) are popular as a light source for in vitro plants because they save energy and allow the morphology of the plant to be altered. The purpose of this study was to show that switching from classical fluorescent light (FL) to LED light can have both beneficial and adverse effects. Pistacia vera plantlets were exposed to FL, monochromatic Blue LED light (B), monochromatic Red LED light (R), and a 1:1 mixture of both B and R (BR). R increased the total weight, shoot length, number of shoots ≥ 1 cm, and proliferation. It also reduced hyperhydricity (HH), but also dramatically increased shoot tip necrosis (STN) and leaf necrosis (LN). B cured plants of HH and STN, but hardly enabled proliferation. It did not solve the problem of LN, but the plants were high in total chlorophyll and carotenoids. BR reduced HH but enabled limited proliferation, high STN, and LN. All three LED treatments reduced HH compared to FL. B induced both high total phenolic and flavonoid content and high DPPH-scavenging activity. These results show that switching from FL to LED can have a significant positive or negative effect on proliferation and quality. This suggests that finding an optimal lighting regimen will take a lot of trial and error.

Polyphenol has been used in treatment for some health disorders due to their diverse health promoting properties. These compounds can reduce the impacts of oxidation on the human body, prevent the organs and cell structure against deterioration and protect their functional integrity. The health promoting abilities are attributed to their high bioactivity imparting them high antioxidative, antihypertensive, immunomodulatory, antimicrobial, and antiviral activity, as well as anticancer properties. The application of polyphenols such as flavonoids, catechin, tannins, and phenolic acids in the food industry as bio-preservative substances for foods and beverages can exert a superb activity on the inhibition of oxidative stress via different types of mechanisms. In this review, the detailed classification of polyphenolic compunds and their important bioactivity with special focus on human health are addressed. Additionally, their ability to inhibit SARS-CoV-2 could be used as alternative therapy to treat COVID patients. Inclusions of polyphenolic compounds in various foods have demonstrated their ability to extend shelf life and they positive impacts on human health (antioxidative, antihypertensive, immunomodulatory, antimicrobial, anticancer). Additionally, their ability to inhibit the SARS-CoV-2 virus has been reported. Considering their natural occurrence and GRAS status they are highly recommended in food.

Many plants are able to synthesize essential oils (EOs), which play key roles in defense against weeds, fungi and pests. This study aims to analyze the chemical composition and to highlight the antioxidant, antimicrobial and phytotoxic properties of the EOs from Eucalyptus falcata , E. sideroxylon and E. citriodora growing in Tunisia. EOs were analyzed by gas chromatography coupled to mass spectrometry (GC/MS) and their antioxidant properties were determined by total antioxidant capacity (TAC), DPPH and ABTS assays. The phytotoxic potential was assessed against weeds ( Sinapis arvensis , Phalaris canariensis ) and durum wheat crop ( Triticum durum ) and compared to chemical herbicide glyphosate. The antifungal activity was investigated in vitro against eight target fungal strains. All EOs displayed a specific richness in oxygenated monoterpenes (51.3-90%) and oxygenated sesquiterpenes (4.8-29.4%), and 1,8-cineole, citronellal, citronellol, trans -pinocarveol, globulol, spathulenol and citronellyl acetate were the main constituents. Eucalyptus EOs exhibited remarkable antioxidant activity and E. citriodora oil exhibited significant activity when compared with E. falcata and E. sideroxylon EOs. The phytotoxic potential of the tested oils had different efficacy on seed germination and the growth of seedlings and varied among tested herbs and their chemical composition variability. Their effectiveness was better than that of glyphosate. At the post-emergence stage, symptoms of chlorosis and necrosis were observed. Furthermore, a decrease in chlorophyll and relative water content, electrolyte leakage and high levels of MDA and proline were indicators of the oxidative effects of EOs and their effectiveness as bioherbicides. Moreover, all the EOs exhibited moderate fungitoxic properties against all the tested fungal strains. Therefore, according to the obtained results, Eucalyptus EOs could have potential application as natural pesticides.

Proteomic analysis was performed to investigate the differentially abundant proteins (DAPs) in barley roots during the tillering stage. Bioinformatic tools were used to interpret the biological function, the pathway analysis and the visualisation of the network amongst the identified proteins. A total of 72 DAPs (33 upregulated and 39 downregulated) among a total of 2580 proteins were identified in response to compost treatment, suggesting multiple pathways of primary and secondary metabolism, such as carbohydrates and energy metabolism, phenylpropanoid pathway, glycolysis pathway, protein synthesis and degradation, redox homeostasis, RNA processing, stress response, cytoskeleton organisation, and phytohormone metabolic pathways. The expression of DAPs was further validated by qRT-PCR. The effects on barley plant development, such as the promotion of root growth and biomass increase, were associated with a change in energy metabolism and protein synthesis. The activation of enzymes involved in redox homeostasis and the regulation of stress response proteins suggest a protective effect of compost, consequently improving barley growth and stress acclimation through the reduction of the environmental impact of productive agriculture. Overall, these results may facilitate a better understanding of the molecular mechanism of compost-promoted plant growth and provide valuable information for the identification of critical genes/proteins in barley as potential targets of compost.

Trace metal element (TME) pollution is a major threat to plants, animals and humans. Agricultural products contaminated with metals may pose health risks for people; therefore, international standards have been established by the FAO/WHO to ensure food safety as well as the possibility of crop production in contaminated soils. This study aimed to assess the accumulating potential of aluminum and barium in the roots, shoots and fruits of Abelmoschus esculentus L., and their effect on growth and mineral nutrition. The content of proline and some secondary metabolites was also evaluated. After treating okra plants with aluminum/barium (0, 100, 200, 400 and 600 µM) for 45 days, the results showed that Al stimulated the dry biomass production, whereas Ba negatively affected the growth and the fructification yield. The okra plants retained both elements and exhibited a preferential accumulation in the roots following the sequence: roots > shoots > fruits, which is interesting for phytostabilization purposes. Al or Ba exposure induced a decline in mineral uptake (K, Ca, Mg, Zn and Fe), especially in roots and shoots. In order to cope with the stress conditions, the okra plants enhanced their proline and total phenol amounts, offering better adaptability to stress.

Quinoa constitutes among the tolerant plants to the challenging and harmful abiotic environmental factors. Quinoa was selected as among the model crops destined for bio-saline agriculture that could contribute to the staple food security for an ever-growing worldwide population under various climate change scenarios. The auxin response factors (ARFs) constitute the main contributors in the plant adaptation to severe environmental conditions. Thus, the determination of the ARF-binding sites represents the major step that could provide promising insights helping in plant breeding programs and improving agronomic traits. Hence, determining the ARF-binding sites is a challenging task, particularly in species with large genome sizes. In this report, we present a data fusion approach based on Dempster-Shafer evidence theory and fuzzy set theory to predict the ARF-binding sites. We then performed an "In-silico" identification of the ARF-binding sites in Chenopodium quinoa . The characterization of some known pathways implicated in the auxin signaling in other higher plants confirms our prediction reliability. Furthermore, several pathways with no or little available information about their functions were identified to play important roles in the adaptation of quinoa to environmental conditions. The predictive auxin response genes associated with the detected ARF-binding sites may certainly help to explore the biological roles of some unknown genes newly identified in quinoa.

The growing demand for native planting material in ecological restoration and rehabilitation for agro-silvo-pastoral ecosystems has resulted in a major global industry in their sourcing, multiplication, and sale. Plant tissue culture is used for producing high-quality, disease-free, and true-to-type plants at a fast rate. Micropropagation can help to meet the increasing demand for planting material and afforestation programs. However, in vitro plant propagation is an expensive technique compared to conventional methods using suckers, seeds, and cuttings. Therefore, adopting measures to lower production costs without compromising plant quality is essential. This can be achieved by improving the culture media composition. Incorporating organic growth additives can stimulate tissue growth and increase the number of shoots, leaves, and roots in culture media. Organic growth supplementation speeds up the formation and development of cultures and yields vigorous plants. Plant regeneration from meristems (shoot tips and axillary buds) is a reliable way to produce true-to-type plants compared with callus and somatic embryogenesis regeneration, but in vitro culture environments can be mutagenic. Therefore, detecting somaclonal variations at an early stage of development is considered crucial in propagating plants. The genetic stability of in vitro regenerated plants needs to be ascertained by using DNA-based molecular markers. This review aims to provide up-to-date research progress on incorporating organic growth additives to enhance in vitro tissue culture protocols and to emphasize the importance of using PCR-based molecular markers such as RAPD, ISSR, SSR, and SCoT. The review was assessed based on the peer-reviewed works published in scientific databases including Science Direct, Scopus, Springer, JSTOR, onlinelibrary, and Google Scholar.

Lead stands as a food contaminant through its accumulation in consumed plants. In this study, the effects of lead (II) chloride (PbCl2) and its levels of uptake on morphological and phytochemical responses of fenugreek were assessed to evaluate its tolerance and safety for human consumption. Results revealed that PbCl2 (50−2000 mg L−1) did not affect the germination rate, but it decreased the radicle length and amylase activity. After three months of Pb treatments, the elemental analysis showed that Pb accumulation was greater in roots than shoots, and it was not present in harvested seeds. The bioaccumulation factor > 1 and the translocation factor << 1 observed for 1000 mg L−1 PbCl2 suggested appropriateness of fenugreek as a phytostabilizer. Additionally, increased lipid peroxidation, hydrogen peroxide, flavonoid levels and catalase activity were observed in Pb-treated fenugreek. Meanwhile, decreased chlorophyll content was detected under these conditions. In turn, the total phenol was correlated with Pb treatment only in roots. HPLC analysis proved that under Pb stress, gallic acid was the most produced compound in treated roots compared to shoots, followed by quercetin. Syringic and chlorogenic acids were more produced in shoots. In conclusion, fenugreek can be used for Pb phytoremediation and is safe for consumption after Pb treatments in the traditional medicine system.

Xylem vessels are essential pivotal organs in bulk hydraulic flow through the whole woody plant. However, environmental constraints generate disagreements in xylem structures, which are characterized by air emboli and occlusions formations, compromising water conductivity in grapevines. The aim of this work was to explore xylem morphology dynamics through the xylem sap flow of five Tunisian grapevine cultivars during the natural bleeding sap periods of 2019, 2021, and 2022. In fact, Sakasly, Khamri, Hencha, Razegui1, and Razegui2 rain-fed grapevine cultivars revealed differential responses towards xylem sap movement. The results demonstrated that the xylem sap flow was significantly more abundant in 2019 than 2021 and 2022 bleeding sap campaigns. A variation was revealed between the cultivars regarding the xylem sap flow. In fact, Sakasly gave the best xylem flow during the three campaigns. Razegui1 and Razegui2 registered approximately similar xylem sap flow, while Hencha and Khamri present the lowest sap fluxes during the three campaigns. Moreover, several vascular occlusions forms were identified from stem cross sections using environmental scanning electron microscopy (ESEM), including tyloses, gels, starch, and gum deposits. The highest occlusion number was observed in Sakasly, Razegui1, and Razegui2 cultivars. Among different biogenic calcium shapes, several were observed for the first time in grapevine, including multi-faceted druse, cubic, crystalline sand, styloids, spherical, or drop-like structures. Considering their lower flow and totally blocked vessels, both Hencha and Khamri confirmed their susceptibility to environmental constraints. However, Sakasly, Razegui1, and Razegui2 cultivars presented higher tolerance according to their sap flow and xylem morphology.

Regulated deficit irrigation (RDI) strategies may greatly contribute to save irrigation water, especially in low water availability conditions. The effects of RDI on the growth process, photosynthesis, fruit yield, and some quality traits were assessed for two years on three plum ( Prunus salicina Lindl.) cultivars ('Black Diamond', 'Black Gold' and 'Black Star') grown in Midwest Tunisia. The experiment was conducted during two successive seasons (2011-2012). Two water regimes were applied per cultivar during the phase of fruit growth until fruit ripening. Stressed trees receive 50% of the amount of irrigation compared to controls (CI). Several eco-physiological parameters and some pomological criteria were measured, based on the climatic condition (ETo, ETc, and VDP). Results showed that the three cultivars had an aptitude for tolerance for moderate stress with varying degrees of response time to drought stress. Globally, a slight decrease in the gas exchange rate (AN and gs) and the water status (RWC, Ψmin, and Ψos) was registered. Tree yields, size and weight show a slight decrease under water stress. Nevertheless, there was an improvement in the total soluble solid content (SSC) and firmness in same cultivars. Our results proved that the Black Star cultivar was the most tolerant to deficit irrigation, in reason that it maintains a good water status and a high photosynthetic activity.

Devrra triradiata Hochst. ex Boiss is an occasional plant species in the Northern region of Saudi Arabia. The shrub is favored on sandy desert wadis, gypsaceous substrate, and sandy gravel desert. In folk medicine, the plant is used for many purposes; to relieve stomach pains, against intestinal parasites, and for the regulation of menstruation. The present study describes the chemical composition of the essential oils (EOs) of different plant parts of D. triradiata . In vivo and in vitro biological activities of plant extracts and essential oils were also studied. Phenylpropanoids, elemicin (flowers: 100%), dillapiole (Stems: 82.33%; and seeds: 82.61%), and apiol (roots: 72.16%) were identified as the major compounds. The highest antioxidant activity was recorded for the EOs of roots and stems (IC 50 = 0.282 µg/mL and 0.706 µg/mL, respectively). For plant extracts, ethyl acetate showed the highest antioxidant activities (IC 50 = 2.47 and 3.18 µg/mL). EOs showed high antifungal activity against yeasts with low azole susceptibilities (i.e., Malassezia spp. and Candida krusei). The MIC values of EOs ranged between 3.4 mg/mL and 56.4 mg/mL. The obtained results also showed phytotoxic potential of plant extracts both on the germination features of Triticum aestivum seeds and the vegetative growth of seedlings.

In the literature, little information is available on the effect of Selenium (Se) on durum wheat yield and grain quality performances. A field investigation was conducted to explore the effect of exogenous Se foliar supply on two types of durum wheat germplasm; i.e., 16 advanced lines and nine modern varieties. The Se effect was assessed on grain yield as well as on technological quality traits (moisture, protein and gluten contents, Zeleny sedimentation index, and deformation energy) in two contrasting environments in Tunisia, namely Kef-Boulifa (semi-arid region) and Beja (sub-humid region). The results displayed significant effects of environments, Se foliar application, and cultivars on grain yield and quality attributes. For grain yield performance, the beneficial effect of Se was more pronounced under the Kef-Boulifa environment, and conversely for the grain quality. A genetic variation was observed within and among the two environments under both Se treatments (with and without Se). Notably, the Se-treated advanced lines displayed the highest grain yield under Kef-Boulifa and Beja conditions. Although these cultivars showed better grain quality in both sites, the modern varieties valorized the Se foliar application better. Cultivars that recorded the highest values for the studies attributes were not necessarily those that valorized the Se supply better. Interestingly, some advanced lines have noted superiority compared to the modern varieties. In this study, cultivars that combine both good yield and good grain quality were determined for semi-arid (L11, L1, Dhahbi, and Maali) and sub-humid (L2, L14, L6, L3, Salim, and INRAT 100) zones. The screening results provide genetic material that could be exploited in breeding programs to improve Se use efficiency.

The current study aimed to evaluate the naturally occurring antimicrobial and antidiabetic potential of various Echium humile ( E. humile ) solvent extracts (hexane, dichloromethane, ethyl acetate, methanol and aqueous). The bioactive compounds were identified using HPLC-MS, revealing the presence of sixteen phytochemical compounds, with the most abundant being p -coumaric acid, followed by 4,5-di- O -caffeoylquinic acid, trans-ferulic acid and acacetin. Furthermore, E. humile extracts showed marked antimicrobial properties against human pathogen strains, with MIC values for the most relevant extracts (methanol and ethyl acetate) ranging from 0.19 to 6.25 mg/mL and 0.39 to 12.50 mg/mL, respectively. Likewise, methanol was found to be bactericidal towards S. aureus , B. cereus and M. luteus , fungicidal against P. catenulatum and F. oxysporum and have a bacteriostatic/fungicidal effect for the other strains. In addition, the E. humile methanolic extract had the greatest α-glucosidase inhibitory effect (IC 50 = 0.06 ± 0.29 mg/mL), which is higher than the standard drug, acarbose (IC 50 = 0.80 ± 1.81 mg/mL) and the aqueous extract (IC 50 = 0.70 ± 0.67 mg/mL). A correlation study between the major phytochemicals and the evaluated activities was investigated. Docking studies evidenced that most of the identified phenolic compounds showed strong interactions into the binding sites of S. aureus tyrosyl-tRNA synthetase and human lysosomal acid-α-glucosidase, confirming their suitable inhibitory effect. In summary, these results may provide rational support to explore the clinical efficacy of E. humile and its secondary metabolites in the treatment of dual diabetes and infections.

Rare earth elements (REEs) present a group of nonessential metals for the growth and development of plants. At high concentrations, they can induce internal stress and disturb the physiological and biochemical mechanisms in plants. The potential uptake of lanthanum (La) and cerium (Ce) by the horticultural plant Helianthus annuus and the effect of these elements on its growth, its absorption of macroelements, and the contents of phenolic compounds and flavonoids were assessed. The plants were exposed to 0, 1, 2.5, 5, and 10 µM of La and Ce for 14 days. The results showed a remarkable accumulation of the two REEs, especially in the roots, which was found to be positively correlated with the total phenolic compound and flavonoid content in the plant shoots and roots. The plant's growth parameter patterns (such as dry weight and water content); the levels of potassium, calcium, and magnesium; and the tolerance index varied with the concentrations of the two studied elements. According to the tolerance index values, H. annuus had more affinity to La than to Ce. Although these metals were accumulated in H. annuus tissues, this Asteraceae plant cannot be considered as a hyperaccumulator species of these two REEs, since the obtained REE content in the plant's upper parts was less than 1000 mg·Kg -1 DW.

In view of climate change, increasing soil salinity is expected worldwide. It is therefore important to improve prediction ability of plant salinity effects. For this purpose, brackish/saline irrigation water from two areas in central and coastal Tunisia was sampled. The water samples were classified as C3 (EC: 2.01-2.24 dS m -1 ) and C4 (EC: 3.46-7.00 dS m -1 ), indicating that the water was questionable and not suitable for irrigation, respectively. The water samples were tested for their genotoxic potential and growth effects on Vicia faba seedlings. Results showed a decrease in mitotic index (MI) and, consequently, growth parameters concomitant to the appearance of micronucleus (MCN) and chromosome aberrations when the water salinity increased. Salt ion concentration had striking influence on genome stability and growth parameters. Pearson correlation underlined the negative connection between most ions in the water inappropriate for irrigation (C4) and MI as well as growth parameters. MI was strongly influenced by Mg 2+ , Na + , Cl - , and to a less degree Ca 2+ , K + , and SO 4 2- . Growth parameters were moderately to weakly affected by K + and Ca 2+ , respectively. The purpose of this study was to evaluate for the first time the phytochemical constituents and biological properties of three (ethanol, acetone, and hexane) Arthrocnemum indicum (Willd.) Moq. ( A. indicum ) extracts. Quantitative analysis revealed the significantly ( p < 0.05) dominance of ethanolic extract on total polyphenol (TPC; 303.67 ± 4.16 mg GAE/g DR) and flavonoid (TFC; 55.33 ± 2.52 mg CE/g DR) contents than the other extracts, also displaying high and equipotent condensed tannin (TCTC) contents as the acetone extract. The qualitative HPLC-MS analysis elucidates 19 and 18 compounds in ethanolic and acetonic extracts, respectively, belonging to the phenolics and flavonoids chemical classes. The extracts were also screened for their in vitro antioxidant activities using 1,1-diphenyl-2-picrylhydrazyl, superoxide anion, and ferric ion (Fe 3+ ) reducing antioxidant power (FRAP), demonstrating the potent antioxidant activity of ethanolic extract, due to its stronger scavenging DPPH • (IC 50 = 7.17 ± 1.26 μg/mL) which is not significantly ( p > 0.05) different from the positive control, BHT (IC 50 = 10.70 ± 0.61 μg/mL), however moderate activity through FRAP and superoxide anion radicals have been observed. Four Gram-positive, four Gram-negative bacteria, and four pathogenic fungi were used for the antimicrobial activity. In addition, S. epidermidis, M. luteus, E. faecalis, C. glabrata, C. parapsilosis, C. krusei were found to be the most susceptible strains towards ethanolic extract. Cytotoxicity values against human colon adenocarcinoma cells (HT29) and human epidermoid cancer cells (Hep2), and one continuous cell lineage control (Vero) revealed that the HT29 cancer cell line was the most responsive to A. indicum shoot extract treatment and significantly ( p < 0.05) different from the other cancer cells. Moreover, when tested for their antidiabetic inhibitory effect, ethanol extract recorded the highest antidiabetic effect with IC 50 = 13.17 ± 1.04 mg/mL, which is 8.4-fold higher than acetone extract. Therefore, the present study provides new findings on the use of A. indicum shoot ethanolic extract to cure many incurable diseases.

The use of biostimulant (BS) holds a promising and environmental-friendly innovation to address current needs of sustainable agriculture. The aim of the present study is twofold: (i) assess the potential of durum wheat seed coating with microbial BS ('Panoramix', Koppert), a mix of Bacillus spp., Trichoderma spp., and endomycorrhiza, compared to two chemical products ('Spectro' and 'Mycoseeds') through germination bioassay, pot and field trials under semi-arid conditions, and (ii) identify the most effective method of BS supply ('seed coating', 'foliar spray', and 'seed coating + foliar spray') under field conditions. For this purpose, three modern durum wheat cultivars were tested. 'Panoramix' was the most efficient treatment and enhanced all germination (germination rate, and coleoptile and radicle length), physiological (relative water content, chlorophyll content, and leaf area), and agro-morphological (plant height, biomass, seed number per spike, thousand kernel weight, and grain yield) attributes. Unexpectedly, the individual application of 'Panoramix' showed better performance than the combined treatment 'Panoramix + Spectro'. Considering the physiological and agro-morphological traits, the combined method 'seed coating + foliar spray' displayed the best results. Principal component analysis confirmed the superiority of 'Panoramix' treatment or 'seed coating + foliar spray' method. Among tested durum wheat cultivars, 'Salim' performed better especially under 'Panoramix' treatment, but in some case 'Karim' valorized better this BS showing the highest increase rates. Based on these study outcomes, 'Panoramix' might be used as promising sustainable approach to stimulate durum wheat performance.

The Tunisian Ziziphus lotus plant was investigated to determine its phytoconstituents and evaluate its biological activities. In particular, the GC/MS technique was used to describe the chemical composition of Z. lotus active extracts and fractions. Among the obtained extracts, the yields of the dried root methanolic extract (29.80%) and the fruit aqueous extract (48.00%) were the highest ones. The dried root methanolic extract exhibited the highest amount in the total phenolics (186.44 ± 0.26 mg GAE/g DW), total flavonoids (102.50 ± 3.53 mg QE/g DW), and tannins (60.714 ± 2.2 mg catechin/g DW). The root aqueous extracts revealed the highest antioxidant activity with an IC 50 of 8.96 ± 0.38 mg/L and 16.46 ± 0.60 mg/L for the ABTS • and DPPH• assays, respectively. The total antioxidant capacity was accorded to the methanolic extract of the dried roots with a value of 304.07 ± 1.11 µg AAE/mg. The drying process was found to improve the qualitative and quantitative properties of the + extracts. The evaluation of the cytotoxic activity against the SH-SY5Y cell line was carried out using MTT assay. The petroleum ether and dichloromethane extracts of the dried roots showed relevant cytotoxic activities. The thin layer chromatography and the GC-MS/GC-FID analysis led to the identification of the 13-epimanool as a potent cytotoxic compound.Z. lotus extracts. The evaluation of the cytotoxic activity against the SH-SY5Y cell line was carried out using MTT assay. The petroleum ether and dichloromethane extracts of the dried roots showed relevant cytotoxic activities. The increasing popularity of pomegranate ( Punica granatum L.), driven by the awareness of its nutraceutical properties and excellent environmental adaptability, is promoting a global expansion of its production area. This investigation reports the variability in the weight, moisture, pH, total soluble solids, carbohydrates, organic acids, phenolic compounds, fatty acids, antioxidant activities, and element composition of different fruit parts (juices, peels, and kernels) from four (Ako, Emek, Kamel, and Wonderful One) of the most widely cultivated Israeli pomegranate varieties in Salento (South Italy). To the best of our knowledge, this is the first systematic characterization of different fruit parts from pomegranate cultivars grown simultaneously in the same orchard and subjected to identical agronomic and environmental conditions. Significant genotype-dependent variability was observed for many of the investigated parameters, though without any correlation among fruit parts. The levels of phenols, flavonoids, anthocyanins, and ascorbic and dehydroascorbic acids of all samples were higher than the literature-reported data, as was the antioxidant activity. This is likely due to positive interactions among genotypes, the environment, and good agricultural practices. This study also confirms that pomegranate kernels and peels are, respectively, rich sources of punicic acid and phenols together, with several other bioactive molecules. However, the variability in their levels emphasizes the need for further research to better exploit their agro-industrial potential and thereby increase juice-production chain sustainability. This study will help to assist breeders and growers to respond to consumer and industrial preferences and encourage the development of biorefinery strategies for the utilization of pomegranate by-products as nutraceuticals or value-added ingredients for custom-tailored supplemented foods.

In recent years, there has been an urgent need for local strategies to ensure food sustainability in Tunisia, recognized as a climate change hotspot region. In this context, adaptation measures, including the adoption of high-yielding durum wheat cultivars with adequate agronomical practices, are an important avenue to improving the productivity of the smallholders that represent 80% of Tunisian farmers. Thus, this study highlights the impact of (i) the adoption of the recently marketed durum wheat cultivar 'Salim' as compared to the common cultivar 'Karim' and the transfer of a technical package to 11 farmers in the Nebeur delegation/Kef-Tunisia (semi-arid region) during the 2013/2014 and 2014/2015 cropping seasons, and (ii) climate change on the expected mean grain yield and biomass by 2070, using the CropSyst agronomic cultivation model based on multi-year crop simulations run with a daily weather series (2020-2070). The adoption of 'Salim' with the recommended package, compared to 'Karim' with the farmer practices, significantly increased the grain yield (37.84%) and biomass (55.43%). Otherwise, the impact of the 0.8 °C temperature rise on the potential yields and biomass over the next 51 years was positive. Contrary to expectations, the yield increases for the two cultivars were very close, but the yield of 'Salim' (36.02 q ha -1 ) remains much higher than that of 'Karim' (23.34 q ha -1 ). On other hand, 'Salim' experienced a higher increase for biomass compared to that of 'Karim'. These results indicate that the adoption of the 'Salim' cultivar with its technical package might be considered as a strategy of adaptation to Nebeur conditions and to future climate change events.

In order to characterize and conserve the endemic pastoral species Medicago tunetana , many prospecting missions were carried out in mountainous regions of the Tunisian ridge. Twenty-seven eco-geographical and morphological traits were studied for six M. tunetana accessions and followed by molecular analysis using seven Simple Sequence Repeat (SSR). Only five markers were polymorphic and reproductible in the six M. tunetana populations. A total of 54 alleles were observed with an average of 10.8 bands/primer/genotype. Mean Polymorphism Information Content (PIC), Nei gene diversity (h) Shannon's information index (I) indicated the high level of polymorphism. The generated dendrogram with hierarchical UPGMA cluster analysis grouped accessions into two main groups with various degree of subclustring. All the studied accessions shared 57% of genetic similarity. Analysis of variance showed high significant difference between morphological traits among M. tunetana populations where MT3 from Kesra showed different morphological patterns regarding leaf, pod and seeds traits. Canonical correspondence analysis (CCA) showed two principal groups of M. tunetana populations based on potassium, total and active lime contents in soil. Our results suggest that SSR markers developed in M. truncatula could be a valuable tool to detect polymorphism in M. tunetana . Furthermore, the studied morphological markers showed a large genetic diversity among M. tunetana populations. This approach may be applicable for the analysis of intra specific variability in M. tunetana accessions. Our study could help in the implementation of an effective and integrated conservation programs of perennial endemic Medicago .

The genus Plantago is particularly interesting for studying the mechanisms of salt tolerance in plants, as it includes both halophytes and glycophytes, as well as species adapted to xeric environments. In this study, the salt stress responses of two halophytes, P. crassifolia and P. coronopus, were compared with those of two glycophytes, P. ovata and P. afra . Plants obtained by seed germination of the four species, collected in different regions of Tunisia, were subjected to increasing salinity treatments for one month under greenhouse conditions. Morphological traits and biochemical parameters, such as ion accumulation and the leaf contents of photosynthetic pigments, osmolytes, oxidative stress markers and antioxidant metabolites, were measured after the treatments. Salt-induced growth inhibition was more pronounced in P. afra , and only plants subjected to the lowest applied NaCl concentration (200 mM) survived until the end of the treatments. The biochemical responses were different in the two groups of plants; the halophytes accumulated higher Na + and proline concentrations, whereas MDA levels in their leaves decreased, indicating a lower level of oxidative stress. Overall, the results showed that P. coronopus and P. crassifolia are the most tolerant to salt stress, and P. afra is the most susceptible of the four species. Plantago ovata is also quite resistant, apparently by using specific mechanisms of tolerance that are more efficient than in the halophytes, such as a less pronounced inhibition of photosynthesis, the accumulation of higher levels of Cl - ions in the leaves, or the activation of K + uptake and transport to the aerial part under high salinity conditions.

Glebionis coronaria (L.) Cass. ex Spach is a troublesome weed in cereal cropping systems in northern Tunisia. Recently, failures in controlling this weed have been reported by farmers. Field surveys and farmers interviews were conducted to highlight the potential causes of G. coronaria occurrence and the associated yield losses in wheat. Survey results revealed a significant correlation between farmers' awareness of resistance occurrence and cultural practices, mainly sowing date and tillage, while G. coronaria abundance was related to the lack of herbicide rotation and the frequency of ALS-inhibiting herbicide use. High G. coronaria infestations (more than 20 plants/m 2 ) caused a significant decrease in wheat grain yield, reaching almost 75% at a density of 100 plants/m 2 . Field and pot experiments showed low efficacies of ALS-inhibiting herbicides to control G. coronaria populations. The application of field rates of tribenuron-methyl and mesosulfuron + iodosulfuron failed to control the tested populations, and generally, G. coronaria dry weight increased compared to nontreated ones (potential hormetic effect). These findings were further investigated in two selected resistant populations through tribenuron dose-response experiments, plants from both populations exhibited high resistance factors (greater than 300), surviving up to 16-fold the recommended field dose. This is the first report for G. coronaria resistance occurrence to ALS-inhibiting herbicides in Tunisia and the second case worldwide.
