Lignin is associated with cell wall rigidity, water and solute transport, and resistance to diverse stresses in plants. Lignin consists of polymerized monolignols (p-coumaryl, coniferyl, and sinapyl alcohols), which are synthesized by cinnamyl alcohol dehydrogenase (CAD) in the phenylpropanoid pathway. We previously investigated cold-induced IbCAD1 expression by transcriptome profiling of cold-stored tuberous roots of sweetpotato (Ipomoea batatas [L.] Lam). In this study, we confirmed that IbCAD1 expression levels depended on the sweetpotato root type and were strongly induced by several abiotic stresses. We generated transgenic sweetpotato plants overexpressing IbCAD1 (TC plants) to investigate CAD1 physiological functions in sweetpotato. TC plants displayed lower root weights and lower ratios of tuberous roots to pencil roots than non-transgenic (NT) plants. The lignin contents in tuberous roots of NT and TC plants differed slightly, but these differences were not significant. By contrast, monolignol levels and syringyl (S)/guaiacyl (G) ratios were higher in TC plants than NT plants, primarily owing to syringyl unit accumulation. Tuberous roots of TC plants displayed enhanced low-temperature (4 °C) storage with lower malondialdehyde and H2O2 contents than NT plants. We propose that high monolignol levels in TC tuberous roots served as substrates for increased peroxidase activity, thereby enhancing antioxidation capacity against cold stress–induced reactive oxygen species. Increased monolignol contents and/or increased S/G ratios might contribute to pathogen-induced stress tolerance as a secondary chilling-damage response in sweetpotato. These results provide novel information about CAD1 function in cold stress tolerance and root formation mechanisms in sweetpotato.

Invertase inhibitor (INH) post-translationally regulates the activity of invertase, which hydrolyzes sucrose into glucose and fructose, and plays essential roles in plant growth and development. However, little is known about the role of INH in growth and responses to environmental challenges in sweetpotato. Here, we identified and characterized an INH-like gene (IbINH) from sweetpotato. IbINH belongs to the pectin methylesterase inhibitor super family. IbINH transcript was the most abundant in storage roots. IbINH mRNA levels were significantly up-regulated in response to drought, abscisic acid (ABA), salicyclic acid (SA) and jasmonic acid (JA) treatments. Overexpressing IbINH in sweetpotato (SI plants) led to the decrease of plant growth and the increase of drought tolerance, while down-regulation of IbINH (RI plants) by RNAi technology resulted in vigorous growth and drought sensitivity. Furthermore, sucrose was increased and hexoses was decreased in SI plants, but the opposite results were observed in RI plants. Moreover, higher levels of sugars were accumulated in SI plants in comparison to non-transgenic plants (NT plants) and RI plants during water deficit. In addition, ABA biosynthesis-involved and abiotic stress response-involved genes were prominently up-regulated in SI plants under drought stress. Taken together, these results indicate that IbINH mediates plant growth and drought stress tolerance in sweetpotato via induction of source-sink strength and ABA-regulated pathway.

Reactive oxygen species (ROS) and nitric oxide (NO) are key signaling molecules involved in various developmental and stress responses in plants. NO and ROS production, which is triggered by various stimuli, activates downstream signaling pathways to help plants cope with abiotic and biotic stresses. Recent evidence suggests that the interplay between NO and ROS signaling plays a critical role in regulating stress responses. However, the underlying molecular mechanism remains poorly understood. We previously reported that transgenic tobacco overexpressing the swpa4 peroxidase (POD) gene from sweetpotato exhibits increased tolerance to stress. Overexpression of swpa4 also induces the generation of H2O2 and activates the expression of various extracellular acidic pathogenesis-related (PR) genes. Here, we show that swpa4 positively regulates the expression of ROS- and NO-related genes in transgenic tobacco plants. Plants expressing swpa4 exhibited increased expression of ROS-related genes and increased ROS-related enzyme activity under normal conditions and H2O2 treatment, whereas the expression of NO associated 1 (NOA1) only increased under normal conditions. Moreover, plants overexpressing swpa4 showed increased NO levels under normal conditions and after treatment with the NO donor sodium nitroprusside (SNP). Interestingly, treatment with a POD inhibitor dramatically reduced NO levels in swpa4 transgenic plants. These findings suggest that swpa4 regulates H2O2 and NO homeostasis in plants under stress conditions, thereby establishing a possible molecular link between the NO and ROS signaling pathways.

Sweetpotato [Ipomoea batatas (L.) Lam] is a globally important root crop with high industrial value. However, because sweetpotato tuberous roots undergo chilling injuries that negatively affect their quality at temperatures below 10 °C, postharvest damage during the winter season is a major constraint for industrialization. To understand chilling injury response during postharvest low temperature storage, we used next-generation sequencing technology to comprehensive analyze the transcriptome of tuberous roots stored at optimal (13 °C) or low temperature (4 °C) for 6 weeks. From nine cDNA libraries, we produced 298,765,564 clean reads, which were de novo assembled into 58,392 unigenes with an average length of 1100 bp. A total of 3216 differentially expressed genes (DEGs) were detected and categorized into six clusters, of which clusters 2, 4, and 5 (1464 DEGs) were up-regulated under low temperature. The genes in these three clusters are involved in biosynthesis of unsaturated fatty acids, pathogen defense, and phenylalanine metabolism. By contrast, genes in clusters 1, 3, and 6 (1752 DEGs), which were generally down-regulated at low temperature, encode antioxidant enzymes or are involved in glycerophospholipid, carbohydrate, or energy metabolism. We confirmed the results of the transcriptome analysis by quantitative RT-PCR. Our transcriptome analysis will advance our understanding of the comprehensive mechanisms of chilling injury during low temperature storage and facilitate improvements in postharvest storage of sweetpotato tuberous roots.

Arabidopsis ABSCISIC ACID-RESPONSIVE ELEMENT-BINDING FACTOR 3 (ABF3), a bZIP transcription factor, plays an important role in regulating multiple stress responses in plants. Overexpressing AtABF3 increases tolerance to various stresses in several plant species. Alfalfa (Medicago sativa L.), one of the most important perennial forage crops worldwide, has high yields, high nutritional value, and good palatability and is widely distributed in irrigated and semi-arid regions throughout the world. However, drought and salt stress pose major constraints to alfalfa production. In this study, we developed transgenic alfalfa plants (cv. Xinjiang Daye) expressing AtABF3 under the control of the sweetpotato oxidative stress-inducible SWPA2 promoter (referred to as SAF plants) via Agrobacterium tumefaciens-mediated transformation. After drought stress treatment, we selected two transgenic lines with high expression of AtABF3, SAF5 and SAF6, for further characterization. Under normal conditions, SAF plants showed smaller leaf size compared to non-transgenic (NT) plants, while no other morphological changes were observed. Moreover, SAF plants exhibited enhanced drought stress tolerance and better growth under drought stress treatment, which was accompanied by a reduced transpiration rate and lower reactive oxygen species contents. In addition, SAF plants showed an increased tolerance to salt and oxidative stress. Therefore, these transgenic AtABF3 alfalfa plants might be useful for breeding forage crops with enhanced tolerance to environmental stress for use in sustainable agriculture on marginal lands.

Tocopherol (vitamin E) is a chloroplast lipid that is presumed to be involved in the plant response to oxidative stress. In this study, we isolated and characterized five tocopherol biosynthetic genes from sweetpotato (Ipomoea batatas [L.] Lam) plants, including genes encoding 4-hydroxyphenylpyruvate dioxygenase (IbHPPD), homogentisate phytyltransferase (IbHPT), 2-methyl-6-phytylbenzoquinol methyltransferase (IbMPBQ MT), tocopherol cyclase (IbTC) and γ-tocopherol methyltransferase (IbTMT). Fluorescence microscope analysis indicated that four proteins localized into the chloroplast, whereas IbHPPD observed in the nuclear. Quantitative RT-PCR analysis revealed that the expression patterns of the five tocopherol biosynthetic genes varied in different plant tissues and under different stress conditions. All five genes were highly expressed in leaf tissues, whereas IbHPPD and IbHPT were highly expressed in the thick roots. The expression patterns of these five genes significantly differed in response to PEG, NaCl and H2O2-mediated oxidative stress. IbHPPD was strongly induced following PEG and H2O2 treatment and IbHPT was strongly induced following PEG treatment, whereas IbMPBQ MT and IbTC were highly expressed following NaCl treatment. Upon infection of the bacterial pathogen Pectobacterium chrysanthemi, the expression of IbHPPD increased sharply in sweetpotato leaves, whereas the expression of the other genes was reduced or unchanged. Additionally, transient expression of the five tocopherol biosynthetic genes in tobacco (Nicotiana bentamiana) leaves resulted in increased transcript levels of the transgenes expressions and tocopherol production. Therefore, our results suggested that the five tocopherol biosynthetic genes of sweetpotato play roles in the stress defense response as transcriptional regulators of the tocopherol production.

YUCCA6, a member of the YUCCA family of flavin monooxygenase-like proteins, is involved in the tryptophan-dependent IAA biosynthesis pathway and responses to environmental cues in Arabidopsis. However, little is known about the role of the YUCCA pathway in auxin biosynthesis in poplar. Here, we generated transgenic poplar (Populus alba × P. glandulosa) expressing the Arabidopsis YUCCA6 gene under the control of the oxidative stress-inducible SWPA2 promoter (referred to as SY plants). Three SY lines (SY7, SY12 and SY20) were selected based on the levels of AtYUCCA6 transcript. SY plants displayed auxin-overproduction morphological phenotypes, such as rapid shoot growth and retarded main root development with increased root hair formation. In addition, SY plants had higher levels of free IAA and early auxin-response gene transcripts. SY plants exhibited tolerance to drought stress, which was associated with reduced levels of reactive oxygen species. Furthermore, SY plants showed delayed hormone- and dark-induced senescence in detached leaves due to higher photosystem II efficiency and less membrane permeability. These results suggest that the conserved IAA biosynthesis pathway mediated by YUCCA family members exists in poplar.

We generated transgenic alfalfa plants (Medicago sativa L. cv. Xinjiang Daye) expressing a bacterial codA gene in chloroplasts under the control of the SWPA2 promoter (referred to as SC plants) and evaluated the plants under various abiotic stress conditions. Three transgenic plants (SC7, SC8, and SC9) were selected for further characterization based on the strong expression levels of codA in response to methyl viologen (MV)-mediated oxidative stress. SC plants showed enhanced tolerance to NaCl and drought stress on the whole plant level due to induced expression of codA. When plants were subjected to 250 mM NaCl treatment for 2 weeks, SC7 and SC8 plants maintained higher chlorophyll contents and lower malondialdehyde levels than non-transgenic (NT) plants. Under drought stress conditions, all SC plants showed enhanced tolerance to drought stress through maintaining high relative water contents and increased levels of glycinebetaine and proline compared to NT plants. Under normal conditions, SC plants exhibited increased growth due to increased expression of auxin-related IAA genes compared to NT plants. These results suggest that the SC plants generated in this study will be useful for enhanced biomass production on global marginal lands, such as high salinity and arid lands, yielding a sustainable agricultural product.

Oxidative stress is one of the major causative factors for injury to plants exposed to environmental stresses. Plants have developed diverse defense mechanisms for scavenging oxidative stress-inducing molecules. The antioxidative enzyme 2-cysteine peroxiredoxin (2-Cys Prx) removes peroxides and protects the photosynthetic membrane from oxidative damage. In this study, transgenic potato (Solanum tuberosum L. cv. Atlantic) expressing At2-Cys Prx under control of the oxidative stress-inducible SWPA2 promoter or enhanced CaMV 35S promoter (referred to as SP and EP plants, respectively) was generated using Agrobacterium-mediated transformation. The transgenic plants were tested for tolerance to stress. Following treatment with 3 μM methyl viologen (MV), leaf discs from SP and EP plants showed approximately 33 and 15% less damage than non-transformed (NT) plants. When 300 μM MV was sprayed onto whole plants, the photosynthetic activity of SP plants decreased by 25%, whereas that of NT plants decreased by 60%. In addition, SP plants showed enhanced tolerance to high temperature at 42 °C. After treatment at high temperature, the photosynthetic activity of SP plants decreased by about 7% compared to plants grown at 25 °C, whereas it declined by 31% in NT plants. These results indicate that transgenic potato can efficiently regulate oxidative stress from various environmental stresses via overexpression of At2-Cys Prx under control of the stress-inducible SWPA2 promoter.

A new dehydration responsive element-binding (DREB) protein gene encoding for an AP2/EREBP-type transcription factor was isolated by screening of the cDNA library for dehydration-treated fibrous roots of sweetpotato (Ipomoea batatas). Its cDNA (referred to as swDREB1) fragment of 1206 bp was sequenced from, which a 257 amino acid residue protein was deduced with a predicted molecular weight of 28.17 kDa. A search of the protein BLAST database revealed that this protein can be classified as a typical member of a DREB subfamily. RT-PCR and northern analyses revealed diverse expression patterns of the swDREB1 gene in various tissues of intact sweetpotato plant, and in leaves and fibrous roots exposed to different stresses. The swDREB1 gene was highly expressed in stems and tuberous roots. In fibrous roots, its mRNA accumulation profiles clearly showed strong expression under various abiotic stress conditions such as dehydration, chilling, salt, methyl viologen (MV), and cadmium (Cd) treatment, whereas it did not respond to abscisic acid (ABA) or copper (Cu) treatment. The above results indicate that swDREB1 may be involved in the process of the plant response to diverse abiotic stresses through an ABA-independent pathway.

Superoxide dismutase (SOD) cDNA, mSOD2, encoding cytosolic copper/zinc SOD (CuZnSOD) cDNA was isolated from suspension-cultured cells of cassava (Manihot esculenta Crantz) by cDNA library screening, and its expression was investigated in relation to environmental stress. mSOD2 is 774 bp in length with an open reading frame (ORF) of 152 amino acids, corresponding to a protein of predicted molecular mass 15 kDa and a pI of 5.22. One copy of the mSOD2 gene was found to be present in the cassava genome by Southern analysis using an mSOD2 cDNA-specific probe. Reverse transcriptase-polymerase chain reaction (RT-PCR) analysis revealed diverse expression patterns for the mSOD2 gene in various tissues of intact cassava plants, at various stages of the growth in suspension cultures, and in the leaf tissues exposed to different stresses. The mSOD2 gene was highly expressed in suspension-cultured cells and in the stems of intact plants. However, it was expressed at low levels in leaves and roots. During suspension cell growth, the mSOD2 transcript progressively increased during culture. Moreover, the mSOD2 gene in excised cassava leaves responded to various stresses in different ways. In particular, it was highly induced in leaf tissue by several abiotic stresses, including high temperature (37 degrees C), chilling (4 degrees C), methyl viologen (MV) exposure, and wounding treatment. These results indicate that the mSOD2 gene is involved in the antioxidative process triggered by oxidative stress induced by environmental change.
