Alternaria is an important toxigenic fungal genus endangering the quality of cereal grains. No data are available in the literature regarding the composition of Alternaria species in wheat from the main wheat‐cultivating regions of China. In this study, a total of 282 strains were isolated from wheat kernels collected in various provinces. Phylogenetic characterization was performed using two informative loci, ASA10 and ASA19, demonstrating that all strains belonged to A. alternata. A low molecular diversity was observed, which did not appear to be influenced by geographic origin. The secondary metabolite profile of the strains was evaluated in rice culture. Altertoxin I was synthesized at a high concentration by the majority of the strains, while the accumulation of altenuene was lower in both frequency and level than that of alternariol and its derivative. A. alternata did not cause readily observable symptoms on wheat spikes after spraying of the conidia at anthesis, but a higher quantity of altertoxin I and Alternaria DNA were detected in inoculated grains. Artificial inoculations of apple fruit showed that all examined strains were capable of causing typical rot symptoms and producing all four toxins to a certain degree. The widespread occurrence of A. alternata on wheat grains and its ability to synthesize various toxic metabolites may pose a serious hazard for both growers and consumers. [ABSTRACT FROM AUTHOR]

Wheat yellow rust (YR) is a most devastating disease in wheat worldwide. In recent years, the epidemic in China has expanded from traditional areas to the middle and lower reaches of the Yangtze River due to the changes in climate and cropping systems. To facilitate YR resistance breeding, it is meaningful to explore resistance loci from local germplasm resources. In this study, a linkage mapping was performed in a recombinant inbred line (RIL) population derived from Ningmai 9 × Yangmai 158 using a high‐density genetic map and phenotypic identification for adult plant resistance (APR) to YR in four environments. Phenotypic analysis showed that Yangmai 158 had better APR than Ningmai 9. Disease severity in different environments was significantly correlated with each other, and it was found that disease severity was significantly influenced by genotype, environment and their interaction. A total of 19 quantitative trait loci (QTLs) were identified, of which four could be detected in multiple environments and four corresponding KASP markers were then developed. Lines with different QTLs from the population including 476 chromosome segment substitution lines (CSSLs) were evaluated for APR. The results revealed that Qyr‐4B.4 and Qyr‐5B.2 could significantly reduce the disease severity, with their combination showing a better effect. Fusarium head blight (FHB) of wheat is a destructive fungal disease worldwide and has become more severe over the last two decades. Development of FHB resistant wheat varieties is the most effective way to manage FHB. The middle and lower reaches of the Yangtze River are the traditional FHB epidemic areas in China. The landraces and germplasms with resistance to FHB originating from this region were used to identity FHB resistance quantitative trait loci (QTLs). Yangmai 158 and Ningmai 9 are the most popular varieties for commercial wheat production in this region and both have moderate FHB resistance. A high‐density genetic map was constructed using 282 recombinant inbred lines (RILs) from a cross between Ningmai 9 and Yangmai 158. Ten QTLs related to Type II FHB resistance were identified, and QFhb‐3B.1 and QFhb‐5A were stably detected across all environments. Based on position alignment, QFhb‐3B.1 from Ningmai 9 is likely to be Fhb1 and QFhb‐5A from Yangmai 158 is a novel QTL not previously described. A competitive allele‐specific PCR (KASP) marker closely linked to QFhb‐5A was developed and could be used for marker‐assisted selection. Distribution of QFhb‐5A was tested with numerous accessions from a widespread core collection. The results suggest that QFhb‐5A has undergone both natural and artificial selection. Some RILs with both Fhb1 and QFhb‐5A presented better FHB resistance than the parents and could be used in FHB resistance breeding. [ABSTRACT FROM AUTHOR]

Fusarium crown rot of wheat has been spreading in the Huanghuai wheat‐growing area in China since 2010, leading to a potential yield loss. To investigate the pathogens associated with this disease in Jiangsu and Shandong provinces in recent years, 617 Fusarium isolates were isolated from nine sites in these two provinces between 2014 and 2016. Of these isolates, 372 were identified as Fusarium pseudograminearum, and the remaining isolates were identified as F. asiaticum and F. graminearum, suggesting that F. pseudograminearum is becoming a predominant causative pathogen of crown rot of wheat in eastern China. Trichothecene gene detection and chemical analyses of trichothecenes indicated that the F. pseudograminearum isolates belonged to the 3‐ADON or 15‐ADON chemotype, and one isolate had the NIV genotype but produced no detectable NIV. 3‐ADON isolates were predominant in Jiangsu, whereas 15‐ADON isolates were prevalent in Shandong. The mating type of the F. pseudograminearum isolates were identified. MAT‐1 and MAT‐2 existed, but in most collections, particularly those in Jiangsu, the ratios of the two mating types deviated significantly from an expected 1:1 ratio. Combined analyses of the natural occurrence of fusarium head blight (FHB), mycotoxins and mycotoxin-producing isolates of Fusarium spp. in fields of wheat revealed FHB epidemics in 12 of 14 regions in Hubei in 2009. Mycotoxin contamination ranged from 0·59 to 15·28 μg g−1 in grains. Of the causal agents associated with symptoms of FHB, 84% were Fusarium asiaticum and 9·5% were Fusarium graminearum, while the remaining 6·5% were other Fusarium species. Genetic chemotyping demonstrated that F. asiaticum comprised deoxynivalenol (DON), 3-acetyldeoxynivalenol (3-AcDON), 15-acetyldeoxynivalenol (15-AcDON) and nivalenol (NIV) producers, whereas F. graminearum only included DON and 15-AcDON producers. Compared with the chemotype patterns in 1999, there appeared to be a modest shift towards 3-AcDON chemotypes in field populations during the following decade. However, isolates genetically chemotyped as 3-AcDON were present in all regions, whereas the chemical 3-AcDON was only detected in three of the 14 regions where 3-AcDON accounted for 15-20% of the DON and acetylated forms. NIV mycotoxins were detected in seven regions, six of which also yielded NIV chemotypes. The number of genetic 3-AcDON producers was positively correlated with amounts of total mycotoxins (DON, NIV and acetylated forms) or DON in wheat grains. Chemical analyses of wheat grains and rice cultures inoculated with different isolates from the fields confirmed their genetic chemotypes and revealed a preferential biosynthesis of 3-AcDON and 4-AcNIV in rice. The genetic diversity and pathogenicity of isolates of Fusarium graminearum and F. asiaticum isolated from wheat heads in China were examined and compared with those of isolates of F. graminearum, F. asiaticum and F. meridionale from Europe, USA and Nepal. Genetic diversity was assessed by SSCP (single strand conformation polymorphism) and AFLP (amplified fragment length polymorphism) analysis and by molecular chemotyping. SSCP analysis of the Fg16F/Fg16R PCR amplicon differentiated F. graminearum, F. asiaticum and F. meridionale and revealed three haplotypes among sequence-characterized amplified region (SCAR) type 1 F. graminearum isolates. AFLP analysis showed a high level of genetic diversity and clustered the majority of Chinese isolates in one group along with other isolates of Asian origin. The second cluster contained F. graminearum isolates from China, Europe and the USA. Of the Chinese isolates, 79% were F. asiaticum and 81% of these were of the 3-AcDON chemotype, with only 9·5% of either chemotype 15-AcDON or NIV. All the Chinese and USA isolates of F. graminearum were 15-AcDON, whereas among the isolates from Europe, 21% were NIV and 8% were 3-AcDON chemotype. No evidence was found for possible differences in aggressiveness between F. graminearum and F. asiaticum. Highly aggressive isolates were present in each region and no evidence was found for any association between aggressiveness and geographical origin or chemotype among the isolates examined. Wangshuibai is a Chinese landrace wheat with a high level of resistance to fusarium head blight (FHB) and deoxynivalenol (DON) accumulation. Using an F7 population of recombinant inbred lines (RILs) derived from the cross between Wangshuibai and Annong 8455 for molecular mapping of quantitative trait loci (QTL) for FHB resistance, the proportion of scabbed spikelets (PSS) and DON content were assessed under field conditions. Composite interval mapping revealed that two and three QTL were significantly associated with low PSS and low DON content, respectively, over 2 years. QTL on chromosomes 3B and 2A explained 17 and 11·5%, respectively, of the phenotypic variance for low PSS, whereas QTL on chromosomes 5A, 2A and 3B explained 12·4, 8·5 and 6·2%, respectively, of the phenotypic variance for low DON content. The 3B QTL appeared to be associated mainly with low PSS, and the 5A QTL primarily with low DON content in Wangshuibai. The 2A QTL had minor effects on both low PSS and DON content. Microsatellite and AFLP markers linked to these QTL should be useful for marker-assisted selection of QTL for low PSS and low DNA content from Wangshuibai. [ABSTRACT FROM AUTHOR]

A soil-borne wheat virus causing severe mosaic and stunting symptoms on wheat in China has been characterized. It had been considered to be soil-borne wheat mosaic virus (SBWMV) because of its rod-shaped virions and similarities to epidemiology and host range. In this study, the virions purified from infected wheat tissue were approximately 20 nm in diameter and of two lengths (140–160 nm and 280–300 nm), with a coat protein of 19 kDa and two RNA components of approximately 7 and 3·5 kb. A rabbit antiserum was produced against the virus and a serological relationship to SBWMV from the USA (Oklahoma) was demonstrated. However, the coat protein was not recognized by most monoclonal antibodies against Oklahoma SBWMV in either ELISA, ISEM or Western blot analysis, indicating epitope differences. In RT-PCR experiments the viral nucleotide sequences were significantly different from those of SBWMV, and this was confirmed by partial sequencing of the cloned PCR fragments generated from RNA1 (c. 1100 nt) and RNA2 (c. 1400 nt), which showed homologies of about 79 and 63%, respectively, to corresponding regions of SBWMV. Because of these significant differences in serology and nucleotide sequence it is suggested that it is a new furovirus for which the name Chinese wheat mosaic virus (CWMV) is proposed. The pandemic of yellow rust on wheat in China, including cultivar-race interactions at the population level, is described by a prototype model (PANCRIN). As the framework of this model, the main epidemic system of wheat yellow rust in China was divided into 11 epidemiological regions. The major parameters are R, the rate of epidemic progress of each region in each month, determined by regional climatological factors and cultivar-race interactions; DIL, the dilution coefficient of inter-regional long-distance dissemination of this disease, determined mainly by distance; and F. fitness values of the pathogen on different cultivar-race combinations. With cultivar deployment, initial disease amount, and initial race frequency as inputs, this model can output the disease amount on each cultivar and the race frequencies of each region. Results of preliminary simulation experiments tallied with some historical situations such as the breakdown of disease resistance of some important cultivars Weak points of this model are also discussed.
